Aluminium and its composites are widely used in production to enhance the strength of lightweight objects. In this study, an AA7075/SiC composite was fabricated using a stir casting route. Multi-objective optimization and finite element analysis were performed with various process parameters on a manufactured aluminium composite (AA7075 + SiC) undergoing a laser beam welding process. Four welding parameters, i.e., pulse frequency, power, welding speed (transverse), and wire size were taken for laser welding as per the L-9 orthogonal array for experimental study. Tensile strength, deflection, temperature distribution, Rockwell hardness (fusion zone), and Rockwell hardness (heat affected zone) were taken as output parameters after welding. The standard deviation objective weighting–grey relational optimization method optimized the process parameter. ANSYS APDL 23 software was utilized to simulate the entire laser welding method with a cylindrical heat source to predict the temperature distribution in the butt-welded plates. This software uses finite element analysis and gives a deviation of only 5.85% for temperature distribution with experimental results. This study helps to understand the effect of various parameters on the welding strength of the aluminium composite.

Abstract Stevia rebaudiana (Bertoni), commonly known as stevia, is a sought-after natural sweetener, but its conventional propagation methods are slow and inefficient. This study aims to enhance the in vitro culture for stevia by investigating the impact of different Murashige and Skoog (MS) medium salt strengths and plant growth hormones on growth and rebaudioside A content. Apical bud-containing shoot segments were used as explants and cultured on various semi-solid and liquid MS media formulations, incorporating cytokinins (BAP and Kin), auxins (NAA and IAA), and different MS major salt concentrations (MS full, ½ MS, and ¼ MS). Assessments of shoot growth parameters, root formation, and HPLC analysis for rebaudioside A content were conducted. The optimal conditions for in vitro growth was found to be in the ¼ MS + Kin 3 mg/L + NAA 0.1 mg/L (semi-solid) medium, resulting in significantly improved shoot growth and enhanced 30.04% rebaudioside A content. Genetic fidelity of regenerated plants was confirmed using RAPD and ISSR markers. These findings offer valuable insights for optimizing in vitro propagation of stevia and potentially enhancing rebaudioside A content.

Although technical causes of endoscopic sinus surgery (ESS) failure have long been reported, we were curious about the quality of contemporary sinus surgery. The electronic health database of Mayo Clinic, Arizona was scrutinized to identify adult patients with diffuse chronic rhinosinusitis (CRS) who underwent revision ESS between January 2019 and September 2023 with a history of bilateral full-house ESS. Anatomical features on preoperative radiology and intraoperative endoscopy were cataloged: residual uncinate tissue, maxillary antrostomy with a non-incorporated natural os, residual ethmoidal septations (three septations > 3 mm), and inadequate sphenoid osteotomy (os size < 6 mm). Sixty-nine subjects were identified. A deviated nasal septum was present in 53.6%. Residual uncinate tissue was noted in 50.7% of patients associated with missed natural maxillary os in 39.13% of antrostomies. An inadequate os diameter was noted in 63.8% of sphenoidotomies. Significant residual septations were seen in posterior ethmoidal cells in 66.7% and anterior ethmoidal cells in 62.3% of patients. Residual frontoethmoidal cells were noted in 72.4% of patients. Although the extent of ESS must be individualized, a high prevalence of features reflecting suboptimal surgery that could limit the success of ESS was identified. These findings merit further consideration for focused training during residency and continuous professional development activities.

This research aims to forecast maximum temperatures and the frequency of heatwave days across four different temperature zones (Zone 1, 2, 3 and 4) in India. These four zones are categorized based on the 30-year average maximum temperatures (T30AMT) during the summer months of April, May, and June (AMJ). Two Machine Learning (ML) algorithms eXtreme Gradient Boosting (XGBoost) and Support Vector Regression (SVR) are employed to achieve this goal. The study utilizes nine key atmospheric variables namely air temperature, geopotential height, relative humidity, U-wind, V-wind, soil moisture, solar radiation, sea surface temperature, and mean sea level pressure at a daily scale spanning from 1991 to 2020 for the months of March, April, May, and June as predictors. The India Meteorological Department daily maximum temperature data spanning from 1991 to 2020 for the months of AMJ serves as the predictands. ML models are developed using spatially averaged atmospheric variables and daily maximum temperature across the grids falling within each temperature zone. Results indicate that for a 7-day lead time, SVR outperforms XGBoost in Zone-1 (T30AMT > 38 °C) and Zone-2 (T30AMT: 35.01 °C–38 °C) by more accurately capturing peak temperatures during training and testing. Abstract High-altitude (HA) environment presents immense physiological adversities for humans that have been overcome by supplementing bio-active phytochemicals from functional foods that support and accelerate acclimatization under these extreme environmental conditions. Several agricultural interventions have been investigated to enhance the phytochemical content in vegetables however; these studies have been limited to low-altitude (LA) regions only. In view of an existing knowledge gap, current work is designed to compare the phytochemical compositions of HA and LA-grown Brassicaceae vegetables (cabbage, cauliflower, knol-khol, and radish) using organic treatments via farm yard manure (FYM) and Azotobacter. The open field study was conducted as a two-factorial randomized block design. The first factor was treatment (T1-FYM, T2-Azotobacter, T3-FYM + Azotobacter, and T4-control) while the second was locations (HA and LA). Among all these treatments, the application of treatment T3 in HA-grown cabbage showed the highest total phenolic content (TPC; 9.56 μg/mg), total flavonoids content (TFC; 14.48 μg/mg), and antioxidant potential using 2,2-diphenyl-1-picrylhydrazyl (DPPH; 85.97%) and ferric reducing antioxidant power (FRAP; 30.77 μg/mg) compared to LA grown samples. Reverse Phase high performance liquid chromatography (RP-HPLC) analysis showed that treatment T3 at HA led to significantly high kaempferol (0.92 μg/mg) and sulforaphane (8.94 μg/mg) contents in cabbage whereas, indole-3-carbinol (1.31 μg/mg) was higher in HA grown cauliflower. The present study provides scientific evidence for the enrichment of health-promoting phytochemical compounds in Brassicaceae vegetables grown with T3 treatment specifically at HA.

Background: Improper use of over-the-counter (OTC) steroid medication has been linked to recalcitrant dermatophytosis. There is proven evidence of HPA axis suppression by the use of long-term oral steroids. This study aims to determine the prevalence and pattern of inappropriate OTC steroid use and its effects on the hypothalamus-pituitary-adrenal (HPA) axis in adults with recalcitrant dermatophytosis. Materials and Methods: This cross-sectional study of 2 months was conducted in a hospital setting and included patients of recalcitrant dermatophytosis with a history of OTC steroid use. Clinico-demographic details and basal serum cortisol levels were recorded in all and analyzed. Result: Of a total of 103 patients, 59.22% (n = 61/103) were males, and the mean duration of steroid abuse was 17.78 months. About 48.54% (n = 50/103), 3.88% (n = 4/103), and 47.57% (n = 49/103) patients reported the use of topical steroids, oral steroids, and both oral and topical steroids, respectively. Among all the topical steroid users (n = 99), clobetasol propionate 48.48% (n = 48/99), while among oral steroid users (n = 53), prednisolone 45.28% (n = 24/53) were the most commonly used agents, respectively. The morning serum cortisol levels (8–9 AM) were found to be decreased in 42.7% (n = 44/103), with a mean value of 44.28 ± 17.34 μg/dL. Conclusion: Improper OTC steroid use in recalcitrant dermatophytosis leads to HPA axis suppression. This highlights the need for intervention from apex health officials.

The work presents the design and performance evaluation of an all-optical AND logic gate exploiting the light beam interference effect. It showcases a novel configuration of oriented air-holes meticulously drilled into a silicon substrate, forming an easy-to-fabricate two-dimensional photonic crystal (2D-PC) arrangement. The comprehensive examination of the proposed gate has been carried out using the finite-difference time-domain (FDTD) method. Results demonstrate the gate’s capability to accurately satisfy its truth tables at a calculated wavelength of 1550 nm, which is imperative for C-band fiber-optic communication systems. The device’s structure boasts a unique 60-degree bend geometry to obtain a compact structure with minimum device area of 40.8μm2 and a substantially high contrast ratio (CR) of 8.15. The response time of 0.12 ps is the lowest among the state-of-the-art 2D-PC AND gates, as per the authors’ knowledge.

Various thermochemical and biochemical processes are resorted to transform agri-wastes into diverse green fuels. Current investigation encompassed three different types of biomass viz., gingelly, kodo millet and horse grams, whose desirability as biofuel feedstock have been largely unexplored till date. The existence of significant amount of cellulose (38.07 %), volatiles (75.19 %), calorific value (avg. 16.98 MJ/kg) in the gingelly biomass, demonstrates the effectiveness of the concerned biomass for utilization as feedstock in diverse industrial applications. The mean estimates of Eα were lower for kodo millet (approx. 150 kJ/mole), followed by gingelly (approx. 178 kJ/mole) and horse gram (approx. 180 kJ/mole). The mean estimates of ΔHα were 174.81 (FWO), 170.22 (KAS), 169.17 (S) and 170.40 (T) kJ/mol for the gingelly biomass. The mean estimates of ΔHα were 147.83 (FWO), 148.81 (KAS), 147.93 (S) and 149.04 (T) kJ/mol for kodo millet biomass, while for horse gram biomass, mean estimates of ΔHα were 178.91 (FWO), 169.61 (KAS), 168.56 (S) and 168.81 (T) kJ/mol. The minor difference of 3–4 kJ/mole between Aα and Hα, signifies the viability of the thermal disintegration process. From master plot, it's evident that the experimental curve intersects multiple theoretical curves, highlighting the intricate characteristics of the thermal disintegration process. The overall ethanol recovery was highest in gingelly as compared to both the biomasses. Gingelly biomass yielded an ethanol titer of 24.8 g/L after 24 h, resulting in a volumetric ethanol productivity of 1.03 g/L/h and an ethanol yield of 0.36 g/g.

IntroductionWe compared the efficacy of intralesional sclerotherapy using 3% sodium tetradecyl sulfate with non-sclerotherapy-based treatments for Hereditary Hemorrhagic Telangiectasia-associated epistaxis management.MethodologyThis is a retrospective study of patients who underwent surgical intervention for HHT-associated epistaxis management from 01/2010–02/2024. Patients undergoing sclerotherapy with intralesional 3% sodium tetradecyl sulfate were included in the sclerotherapy group and others undergoing conventional non-sclerotherapy-based procedures in the non-sclerotherapy group. Outcomes like breakthrough epistaxis, emergency visits, intra-op blood loss, blood transfusions, and procedure complications in the 3-month perioperative period were compared.ResultsTwenty-three patients who underwent 74 intranasal procedures were identified. In the sclerotherapy group, 17 patients underwent 47 procedures. In the non-sclerotherapy group, 10 patients underwent 27 procedures. Till the 3rd post-treatment month, fewer breakthrough epistaxis episodes were observed after sclerotherapy procedures (13/47) vs. non-sclerotherapy procedures (14/27); (p = 0.037). Most neurons are not replaced after injury and thus possess robust intrinsic mechanisms for repair after damage. Axon injury triggers a calcium wave, and calcium and cAMP can augment axon regeneration. In comparison to axon regeneration, dendrite regeneration is poorly understood. To test whether calcium and cAMP might also be involved in dendrite injury signaling, we tracked the responses of Drosophila dendritic arborization neurons to laser severing of axons and dendrites. We found that calcium and subsequently cAMP accumulate in the cell body after both dendrite and axon injury. Two voltage-gated calcium channels (VGCCs), L-Type and T-Type, are required for the calcium influx in response to dendrite injury and play a role in rapid initiation of dendrite regeneration. The AC8 family adenylyl cyclase, Ac78C, is required for cAMP production after dendrite injury and timely initiation of regeneration. Injury-induced cAMP production is sensitive to VGCC reduction, placing calcium upstream of cAMP generation. We propose that two VGCCs initiate global calcium influx in response to dendrite injury followed by production of cAMP by Ac78C. This signaling pathway promotes timely initiation of dendrite regrowth several hours after dendrite damage.

Concrete is a fundamental construction material, widely used due to its durability and versatility. However, enhancing its mechanical properties, such as strength, while simultaneously addressing sustainability concerns remains a significant challenge. This study presents a novel approach to optimize concrete mix designs by incorporating waste glass particles, using a Hybrid Swarm-Intelligent Gravitational Search Optimized Random Forest (SIGSORF) model. The primary objective is to improve the strength assessment of concrete while reducing environmental impact through waste glass utilization. The first step in the study is to examine the physical and chemical characteristics of waste glass to see if it may somewhat substitute traditional pebbles in the manufacturing of mortar. Then clean the data and preprocess for use in training and validating the SIGSORF model. The SIGSORF model is designed to intelligently select proportions of waste glass and other concrete components to maximize compressive strength and flexural strength while minimizing environmental impact. The experimental results are then compared with predictions made by the SIGSORF model, demonstrating its effectiveness in optimizing concrete mix designs for improved strength. Ultimately, this study promotes the utilization of waste materials in construction, fostering a more environmentally responsible and economically viable concrete production approach.

Spatiotemporal forecasting of traffic flow data represents a typical problem for urban traffic management, involving complex interactions, nonlinearities, and long-range dependencies due to the interwoven nature of the temporal and spatial dimensions. Traditional statistical and machine learning methods struggle to handle both temporal and spatial dependencies in such datasets. While graph convolutional networks and multi-head attention mechanisms have been widely adopted in this field, they often fail to accurately model dynamic temporal patterns and effectively differentiate noise from signals in traffic datasets, leading to potential overfitting. This paper proposes a wavelet-based temporal attention model, namely a wavelet dynamic spatiotemporal aware graph neural network (W-DSTAGNN), for tackling the traffic forecasting problem. A key feature of W-DSTAGNN is the use of wavelets that can effectively separate noise and capture multi-resolution temporal patterns. Wavelet decomposition can help by decomposing the signal into components that can be analyzed independently, reducing the impact of non-stationarity and handling long-range dependencies of traffic flow datasets. These enable our proposal to generate more robust and accurate forecasts of complex and dynamic traffic dependencies than commonly used spatiotemporal deep learning models. Benchmark experiments using three popularly used statistical metrics confirm that our proposal efficiently captures spatiotemporal correlations and outperforms eleven state-of-the-art models (including both temporal and spatiotemporal benchmarks) on three publicly available traffic datasets. Our proposed approach can better handle dynamic temporal and spatial dependencies, delivering reliable long-term forecasts, and generating interval forecasts to enhance probabilistic forecasting of traffic datasets.

Ascertainment of photonic stopband absolute topological character requires information regarding the Bloch eigenfunction spatial distribution. Consequently, the experimental investigations predominantly restrict themselves to the bulk‐boundary correspondence principle and the ensuing emergence of topological surface state. Although capable of establishing the equivalence/inequivalence of bandgaps, the determination of their absolute topological identity remains out of its purview. The alternate method of reflection phase‐based identification also provides only contentious improvements owing to the measurement complexities pertaining to the interferometric setups. To circumvent these limitations, the Kramers–Kronig amplitude‐phase causality considerations are resorted to and an experimentally conducive method is proposed for bandgap topological character determination directly from the parametric reflectance measurements. Particularly, it is demonstrated that in case of 1D photonic crystals, polarization‐resolved dispersion measurements suffice in qualitatively determining bandgaps’ absolute topological identities. By invoking the translational invariance of the investigated samples, a parameter “differential effective mass” is also defined, that encapsulates bandgaps’ topological identities and engenders an experimentally discernible bandgap classifier.

Background: Chronic Myeloid Leukemia is characterized by the presence of the Philadelphia Chromosome (Ph) which contains the BCR::ABL1 fusion gene that occurs due to a reciprocal translocation between chromosomes 9 and 22. This accounts for up to 15 % of all adult leukemias [1]. Most patients treated with first line tyrosine kinase inhibitor (TKI) imatinib achieve durable response but may undergo relapse at some stage [2]. The most important mechanism that may confer imatinib resistance is point mutation within BCR::ABL kinase domain. Other generation ABL tyrosine kinase inhibitors such as dasatinib, nilotinib, bosutinib and ponatinib help to overcome imatinib resistance [3]. Sensitivity of the patient to each of the above TKIs depends upon the individual candidate mutation present. Thus, it is important to perform mutation analysis for effective therapeutic management of CML patients once they show imatinib resistance. We used direct sequencing to identify the different types of mutations responsible for resistance of imatinib treatment from north India. Methods: In this study, the patient resistance for the imatinib were analyzed for BCR::ABL kinase domain mutation by direct sequencing and the detected mutations along with their percentage prevalence were reported. Results: 329 patients with CML-CP were analyzed for BCR::ABL kinase domain mutation. Total 66 (20.06 %) patients out of 329 had mutation in at least one of the domains of BCR::ABL conferring resistance to different generations of TKI. Mutations in BCR::ABL kinase domain was observed in different domain of BCR::ABL. ATP binding P-Loop (42.42 %), Direct binding site (36.36 %), C-Loop (10.60 %), A-Loop (6.06 %), SH2 contact (3.03 %), SH3 contact (1.51 %). Conclusion: Total 20.06 % patients (66/329) show mutation in at least one of the structural motifs of BCR-ABL kinase domain, which further confer the resistance to a particular generation of TKI.

BackgroundUnlike acute rhinosinusitis (ARS) which is mostly viral in etiology, the role of viruses in chronic rhinosinusitis (CRS) remains unclear. Viruses may play a role in initiation, exacerbations or perpetuate chronic inflammatory responses in the sinonasal mucosa. Research needs to characterize whether viruses are part of the normal sinonasal microbiome, colonizers or pathogenic.MethodsSystematic review of the English literature was conducted. Following databases were searched with an initial search conducted in November 2021 and then updated through June 2023: Ovid Medline (1946 to present), Ovid Embase (1988 to present), Scopus (2004 to present) and Web of Science (1975 to present). MeSH (Medical Subject Headings) terms included: viruses, virus diseases, sinusitis, and rhinovirus. Keywords: virus, viral infection*, sinusitis, rhinovirus, chronic rhinosinusitis, CRS, respiratory virus, respiratory infection*, and exacerbat*. A supplementary search was conducted through September 2023: Ovid Medline (1946 to present), Epub Ahead of Print, In-Process & Other Non-Indexed Citations and Ovid MEDLINE(R) Daily. Keywords used were: virus, viral infection*, sinusitis, chronic rhinosinusitis, CRS, respiratory virus, respiratory infection*, and exacerbat*.ResultsThirty studies on viruses in CRS met inclusion criteria for full review. These included 17 studies on prevalence of virus in CRS, 5 examining probable causes of host susceptibility to viral infections in CRS, and 8 studies examining pathological pathways in viral association of CRS. The prevalence of viruses in nasal specimens of CRS subjects was higher as compared to controls in most studies, though a few studies showed otherwise. Rhinovirus was the most common virus detected. Studies showed that viruses may be associated with persistent hyper-responsiveness in the sinonasal mucosa, susceptibility to bacterial infections, upregulation of genes involved in the immune response and airway remodeling as well as CRS exacerbations. Presence of viruses was also associated with worse symptom severity scores in CRS subjects.ConclusionMost data show higher presence of viruses in nasal and serum samples of CRS subjects as compared to controls but their exact role in CRS pathophysiology in unclear. Large studies with longitudinal sampling at all disease phases (i.e., prior to disease initiation, during disease initiation, during disease persistence, and during exacerbations) using standardized sampling techniques are needed to definitively elucidate the role of virus in CRS.

Abstract The toxicity of ammonia surged with arsenic pollution and high temperature (34 °C). As climate change enhances the pollution in water bodies, however, the aquatic animals are drastically affected and extinct from nature. The present investigation aims to mitigate arsenic and ammonia toxicity and high-temperature stress (As + NH3 + T) using zinc nanoparticles (Zn-NPs) in Pangasianodon hypophthalmus. Zn-NPs were synthesized using fisheries waste to developing Zn-NPs diets. The four isonitrogenous and isocaloric diets were formulated and prepared. The diets containing Zn-NPs at 0 (control), 2, 4 and 6 mg kg−1 diets were included. Superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione-s-transferase (GST) were noticeably improved using Zn-NPs diets in fish reared under with or without stressors. Interestingly, lipid peroxidation was significantly reduced, whereas vitamin C and acetylcholine esterase were enhanced with supplementation of Zn-NPs diets. Immune-related attributes such as total protein, globulin, albumin, myeloperoxidase (MPO), A:G ratio, and NBT were also improved with Zn-NPs at 4 mg kg−1 diet. The immune-related genes such as immunoglobulin (Ig), tumor necrosis factor (TNFα), and interleukin (IL1b) were strengthening in the fish using Zn-NPs diets. Indeed, the gene regulations of growth hormone (GH), growth hormone regulator (GHR1), myostatin (MYST) and somatostatin (SMT) were significantly improved with Zn-NPs diets. Blood glucose, cortisol and HSP 70 gene expressions were significantly upregulated by stressors, whereas the dietary Zn-NPs downregulated the gene expression. Blood profiling (RBC, WBC and Hb) was reduced considerably with stressors (As + NH3 + T), whereas Zn-NPs enhanced the RBC, WBC, and Hb count in fish reread in control or stress conditions. DNA damage-inducible protein gene and DNA damage were significantly reduced using Zn-NPs at 4 mg kg−1 diet. Moreover, the Zn-NPs also enhanced the arsenic detoxification in different fish tissues. The present investigation revealed that Zn-NPs diets mitigate ammonia and arsenic toxicity, and high-temperature stress in P. hypophthalmus.

Abstract Medicinal plants possess range of phytochemicals accountable for their diverse biological activities. Presently, such compounds have been isolated from medicinal plants, characterized and evaluated for their pharmacological potential. In the present study, the efforts have been made to isolate the compound(s) from Grewia tiliaefolia Vahl., plant known for its ameliorative effect on brain related diseases such as anxiety, depression, cognitive disorders and Parkinson’s disease. Plant extract was subjected to isolation of compound(s) using column chromatography and isolated compound was characterized by NMR FTIR and LCMS. The isolated compound was novel with the IUPAC name of the compound is propyl 3-hydroxy-10,13-dimethyl-6,7,8,9,10,11,12,13,14,15,16,17-dodecahydro-3H-cyclopenta[a]phenanthrene-17-carboxylate, designated as A-1 and has not been reported before. A-1 was further evaluated for its antioxidant potential using in vitro antioxidant assays (2,2-diphenyl-1-picryl-hydrazyl-hydrate, DPPH assay and reducing power assay, RPA). Also, Acetylcholinesterase (AChE) inhibitory potential of A-1 and extract was analysed. Results showed that A-1 exhibited significantly higher antioxidant activity in both DPPH and RPA assay as compared to plant extract. In case of AChE inhibitory activity again, A-1 has shown significantly higher activity as compared to plant extract. In silico study was conducted to predict its action on proteins playing crucial role in neurological and neurodegenerative disorders such as gamma amino butyric acid (GABA) receptor and glutamate α amino-3-hydroxyl-5-methyl-4-isoxazolepropionic acid (Glu AMPA) receptor in epilepsy and AChE enzyme in Alzheimer’s diseases. The compound has shown interaction in following order: AChE > GABA receptor > Glu AMPA receptor. Further, molecular dynamic simulations and ADME studies of A-1 and AChE enzyme revealed that A-1 yielded good results in all parameters and hence can relieve Alzheimer’s like symptoms.

Abstract East Kolkata Wetlands (EKW) is an important site for fish culture in sewage-fed areas, which are major receivers of pollutants and wastages from Kolkata. EKW is internationally important as the Ramsar site was declared on Aug 2002 with an area of 125 km2. EKW is a natural water body where wastewater-fed natural aquaculture has been practiced for more than 70 years. It is ecologically vulnerable due to the discharge of toxic waste through sewage canals from cities. Assessing the EKW to understand the inflow and load of the toxic metal (s) in fish, water, and sediments samples is essential. The field (samples collection from 13 sites) and lab (determination of toxic level of metals) based research were carried out to assess metal toxicity and health risk assessment in EKW. The levels of eighteen metals (18), namely Chromium, Vanadium, Cobalt, Manganese, Copper, Nickel, Zinc, Silver, Molybdenum, Arsenic, Selenium, Tin, Gallium, Germanium, Strontium, Cadmium, Mercury, and Lead, were determined using Inductively coupled plasma mass spectrometry (ICP-MS) in five fish tissues viz. muscle, liver, kidney, gill and brain, along with the water samples and soil sediments in 13 sampling sites. The bioaccumulation and concentration of metals in fish tissues, soil sediments, and water samples were well within the safe level concerning the recommendation of different national and international agencies except for a few metals in a few sampling sites like Cd, As, and Pb. The geoaccumulation index (Igeo) was also determined in the soil sediments, indicating moderate arsenic, selenium, and mercury contamination in a few sites. The contamination index in water was also determined in 13 sampling sites. The estimated daily intake (EDI), reference dose (RfD), target hazard quotient (THQ), slope factor and cancer risk of Cr, Mn, Co, Ni, Cu, Zn, As, Se, Cd, Pb and Hg from fish muscle were determined. Based on the results of the present investigation, it is concluded that fish consumption in the East Kolkata Wetland (EKW) is safe. The effects of bioaccumulation of metals in muscle tissue were well within the safe level for consumption as recommended by WHO/FAO.

Background: Disorders of sexual development (DSD) encompass a group of congenital conditions characterized by diverse genotypic and phenotypic variations. Ovotesticular (OT) DSD is a distinctive subtype within this spectrum. Among the array of DSD, OT-DSD stands as one of the most infrequent anomalies, with reported prevalence rates as scarce as 1 in 83,000. This study aims to elucidate the clinical, hormonal, cytogenetic, surgical, and histopathological characteristics of OT disorder of sex development (OT-DSD) within a tertiary center in North India. Methodology: A retrospective analysis was conducted, involving a comprehensive review of records pertaining to OT-DSD patients from the years 2018 to 2022, all of whom were incorporated into the study. Results: The mean age of presentation in this study was 10 years, spanning from 6 to 15 years. Predominantly, the affected individuals were male, with a solitary patient representing the female category. Clinical manifestations displayed a spectrum encompassing genital ambiguity, inguinal swelling, and primary amenorrhea. The karyotypes observed were 46,XX in four patients and 46,XY in one patient. A holistic assessment, inclusive of clinical evaluation, hormonal assays, pelvic ultrasonography, and surgical intervention when necessary, was administered to all patients. Among these cases, three patients were reared as males, their gender assignment driven by external genital appearance and sociocultural influences. Notably, none of the patients manifested gonadal tumors during the course of the study. Conclusion: In cases of ambiguous genitalia, the consideration of OT-DSD should be integral to the differential diagnosis, underscoring the significance of heightened clinical awareness and informed decision-making.

Background: Acute lymphoblastic leukemia (ALL) is a common pediatric malignancy, typically manifesting with symptoms of bone marrow and hematolymphoid organ infiltration. Although hepatic involvement in the form of hepatosplenomegaly is seen in nearly two-thirds of the patients, primary presentation as acute liver failure is rare. Clinical Description: A 12-year-old boy presented with complaints of abdominal distention with jaundice for the past 15–20 days, associated with transient fever of 6 days. Examination revealed pallor, icterus, and hepatosplenomegaly. A possibility of acute infectious or autoimmune hepatitis or hemolytic anemia was considered. Management: Liver function tests were deranged with hyperbilirubinemia with modest increase in transaminases and serum alkaline phosphatase along with deranged prothrombin time. The complete blood count showed pancytopenia with normal peripheral smear. Investigations for common infectious causes were noncontributory, as also those for autoimmune hepatitis. The direct Coombs test was also negative. In view of persistent pancytopenia with hyperbilirubinemia and hepatosplenomegaly, a bone marrow evaluation was done, which revealed B-cell ALL (B-ALL). Thus, the child was diagnosed with a case of ALL presenting with acute liver failure. The child was initially managed as a case of acute liver failure and on diagnostic confirmation, he was started on the B-ALL chemotherapy protocol as per his risk stratification with hepatic dose modification of chemotherapy. The child showed complete normalization of the liver functions by about 4 weeks of induction chemotherapy and achieved remission. Conclusions: A child presenting with acute liver failure may be having an underlying undiagnosed, clinically silent ALL. After ruling out common causes of acute liver failure, one must not forget the possibility of leukemic blast infiltration of the liver as a cause of liver failure. With early initiation of advanced chemotherapeutic induction regimens, there can be dramatic resolution of liver injury and remission of ALL.

ObjectivesIndia’s Covid-19 vaccination campaign engaged frontline workers (FLWs) to encourage vaccination among vulnerable segments of society. The FLWs report encountering a variety of barriers to vaccination and are often unsuccessful despite multiple visits to the same person. This cross-sectional study aims to pinpoint which of these barriers drive vaccine hesitancy among these segments, to help streamline vaccine communication, including FLW training, to better safeguard the population.MethodsTrained field enumerators contacted 893 individuals from five states across India and collected self-reported assessments of fifteen vaccination barriers (identified through discussions with FLWs), current vaccination status and future vaccination intentions, and covariates (demographics/comorbidities). Factor analysis of the fifteen barriers yielded two factors, one relating to fear of vaccine adverse effects and a second focused on peripheral concerns regarding the vaccine. The covariates significantly associated with current vaccination status were combined under a latent class regime to yield three cluster types (health access, financial strength, and demographics). The primary analysis examined the effect of the two barrier factors, the covariate clusters, and comorbidity, on current vaccination status and future vaccine intentions.ResultsFear of vaccine adverse effects was the primary driver of vaccine hesitancy; peripheral concerns frequently mentioned by the FLWs had no impact. Although cluster membership and the presence of comorbidities predicted vaccine uptake, neither of them materially altered the effect of fear of vaccine adverse effects with the following exception: fear of adverse effects was not associated with vaccination status among young Muslim men.ConclusionSubject to limitations, these results indicate that interventions to decrease vaccine hesitancy should focus primarily on fear associated with vaccines rather than spend resources trying to address peripheral concerns.

Introduction: New bioresources for catalytic application and fine chemical synthesis are the need of the hour. In an effort to find out new biocatalyst for oxidation-reduction reaction, leading to the synthesis of chiral intermediates, novel yeast were isolated from unique niche and employed for the synthesis of value added compounds.Methods: To determine the genetic relatedness of the isolated strain, HSB-15T, sequence analysis of the internal transcribed spacer (ITS) and D1/D2 domains of the 26S rRNA gene sequence was carried out. The distinctive features of the strain HSB-15T were also identified by phenotypic characterization. The isolated strain HSB-15T was employed for the reduction of selected naphthyl ketones to their corresponding alcohols and a biosurfactant was isolated from its culture broth.Results: The analysis of the ITS and D1/D2 domains of the 26S rRNA gene revealed that strain HSB-15T is closely related to the type strain of Starmerella vitae (CBS 15147T) with 96.3% and 97.7% sequence similarity, respectively. However, concatenated sequences of the ITS gene and D1/D2 domain showed 94.6% sequence similarity. Phenotypic characterization indicated significant differences between strain HSB-15T and its closely related species and consequently, it was identified as a novel species, leading to the proposal of the name Starmerella cerana sp. nov. The strain was able to reduce selected naphthyl ketones to their corresponding alcohols with remarkable efficiency, within a 12-hours. The strain HSB-15T also produced a surfactant in its culture broth, identified as sophorolipid upon analysis.Discussion: The study explored the potential of the novel strain, HSB-15T, as a whole-cell biocatalyst for the reduction of naphthyl ketones to their corresponding alcohols and also reports its capability to produce sophorolipid, a biosurfactant, in its culture broth. This dual functionality of HSB-15T both as biocatalyst and biosurfactant producer enhances its applicability in biotechnology and environmental science.

Loss of biodiversity, which has an impact on both plant development and genetic advancement, disrupts the fundamental services that ecosystems provide to humanity. Variability assessment is a challenging topic. Multivariate statistics can be useful for comparing and evaluating genetic variability. The data of 16 different morpho-physiological variables were subjected to several multivariate approaches, including principal component (PC) and correlation coefficient analysis, in order to assess the diversity of the twenty Turkey berry genotypes. The correlation coefficients discovered through this analysis were used to gauge the strength of the association between the traits. In this study, Pearson correlation analysis revealed a significant correlation between the observed phenotypic traits. Among these traits, number of branches per plant , number of leaves, leaf area, number of flowers per cluster, number of flower cluster per plant, number of fruits per cluster, number of fruit cluster per plant, and fruit diameter exhibited a positive and significant correlation with fresh fruit yield per plant, whereas days of first flowering exhibited a negative and significant correlation with fresh fruit yield per plant. Principal component (PC) analysis revealed that the first three PCs had Eigen values greater than 1, accounting for 81% of the overall variation. For morphological features, PC 1 accounted for the largest variability of 57% of the overall variation, and the lowest contribution (10%) was recorded by PC3. The genotypes St007, St006, St008, St010, St011, St001, St009, St018, St020, St003, St004, and St005 were shown to be more varied and better performers in terms of fresh fruit yield and yield contributing features based on the primary factor scores. According to research findings, the genotypes of Turkey berries identified in this work would serve as useful genetic tools for boosting the productivity of the fruits for upcoming breeding endeavors, especially in light of the unpredictability of climate change.

The Maillard reaction products (Melanoidin) are major environmental pollutants of the distillery and fermentation industries. Manganese peroxidase (MnP) was found to be an efficient ligninolytic enzyme capable of degrading Maillard products. MnP can convert Mn2 ions in wood and soils to the more reactive Mn2+ ions. The current study aims to isolate MnP producing bacteria from Similipal Biosphere Reserve (SBR), Odisha, and determine its biodegradation ability towards millards products. In total 10 ligninolytic bacterial colonies were isolated from the soil sample of SBR and their MnP activity was screened using phenol red dye as an indicator. The isolate, SMB4 was found to be the most efficient MnP-producing bacterium showing the highest MnP activity of 3.89 U/ml/min in un-optimized conditions. The bacterium was identified as Pseudoduganella violacea based on morphological, biochemical and molecular characterization. MnP activity of SMB4 was optimized using response surface methodology. Under optimized conditions SMB4 showed MnP activity of 6.12 U/ml/min which is 1.57 times from un-optimized condition. The isolate SMB4 was found to show significant decolorization of maillards products up to 83.68 % at temperature 37 °C within 192 h of incubation. The degraded product was analyzed using UV–vis and FTIR analyses. The current investigation suggests that MnP produced by Pseudoduganella violacea may be successfully exploited for bioremediation purposes to degrade Maillard products.

Abstract Explorations into the photonic analogs of topological materials have garnered significant research interest due to their application potential. Particularly in planar systems, the prospects of engendering extinguishable topological states can have wide-ranging implications. With an objective of employing these concepts for thermal emission engineering, here, we design and numerically investigate a quasi-monochromatic highly directional mid-infrared source elicited from inversion symmetry-protected topological interface states. Notably, by relying on the architecture of electro-optic effect-induced topological phase transitions, we introduce the possibility of ultrafast switching of thermal radiation. These reversible phase transitions, being free from carrier transport are inherently fast and evoke thermal emission modulation with a modulation depth upto 0.99. Specifically, our platform exhibits a near-perfect extinguishable spectral emission peak at $$4~\mu$$ 4 μ m with a quality factor of over 18500, displaying negligible parasitic emissions. Furthermore, the optimized interface state manifests itself for only one of the polarization modes, resulting in polarized emission under resonance conditions. To establish a methodical approach to parameter optimization, we also model our platform as a leaky mode resonator using the framework of temporal coupled-mode theory. We believe, our findings can provide a way forward in establishing complete control over the optical characteristics of the infrared thermal emitters.

Physical exercise enhances heat generation in human skin and subcutaneous tissues causing thermal stress. This thermal stress causes discomfort in the human body and deteriorates the physical performance of the human subjects. Appropriate choice of cloth for outer covering of human body is necessary to prevent this to provide comfort during the thermal stress. A mathematical model is proposed in this study to analyze the thermal stresses in human skin and subcutaneous tissues covered with various different cloth fabrics. The model considers four compartments namely cloth, epidermis, dermis, subcutaneous tissues. The bioheat equation along with appropriate boundary condition is employed in this model for one dimensional unsteady state case. The numerical simulation is performed using forward and central finite difference formulas. The findings have been utilized to examine how different types of fabric impact thermal stress experienced by the layers of human skin during physical activity. The polyester is found to be superior in comparison to cotton fabric as it shows less thermal stress and thus can be concluded to be more comfortable as a dress material.

Introduction There is some data suggesting vasodilatory effect of sympathetic blockade in patients with aneurysmal subarachnoid hemorrhage but the effect on medium‐sized intracranial arteries remains unclear. Methods Serial ipsilateral intracranial angiograms were obtained before and after percutaneous inferior sympathetic ganglion block. The angiogram files were imported to a DICOM reader, Horos Open‐Source Medical Image Viewer (Version 3.3.6).The percent change of arterial diameter (compared with pre‐blockage image) was calculated in intracranial internal carotid (ICA), middle (MCA) and anterior cerebral (ACA) arteries. We also assessed the inter‐observer reliability of arterial diameter change using the Bland‐Altman method after two raters independently measured the arterial diameter in 2 images with a total of 12 arterial measurements. Results In Patient 1, there was an increase in diameter of intracranial ICA, MCA and ACA at 1 minute (9.62%, 10.34%, 8.64%), 5 minutes (11.38%, 12.22%, 14.42%) and 10 minutes (8.66%, 11.30%, 5.50%), respectively.In Patient 2, there was an increase in diameter at 1 minute (0.72%, 2.08%, ‐1.75%), 5 minutes (12.04%, 0.64%, 4.50%), 10 minutes (6.86%, 7.23%, 10.38%), 20 minutes (9.45%, ‐2.16%, 13.32%), and 30 minutes (7.31%, 12.02%, 6.50%), respectively. The Bland‐Altman coefficient of variation was 1.32 (range 0.01 ‐5.57) with majority (5 out of 6, 83%) of the data points within the upper and lower limits of acceptance. Conclusions We observed vasodilation in medium‐sized intracranial arteries ipsilateral to the percutaneous inferior sympathetic ganglion block as early as one minute post treatment and persisting for at least until 30 minutes post blockade.

The COVID-19 pandemic was a watershed event for wastewater-based epidemiology (WBE). It highlighted the inability of existing disease surveillance systems to provide sufficient forewarning to governments on the existing stage and scale of disease spread and underscored the need for an effective early warning signaling system. Recognizing the potentiality of environmental surveillance (ES), in May 2021, COVIDActionCollaborative launched the Precision Health platform. The idea was to leverage ES for equitable mapping of the disease spread in Bengaluru, India and provide early information regarding any inflection in the epidemiological curve of COVID-19. By sampling both networked and non-networked sewage systems in the city, the platform used ES for both equitable and comprehensive surveillance of the population to derive precise information on the existing stage of disease maturity across communities and estimate the scale of the approaching threat. This was in contrast to clinical surveillance, which during the peak of the COVID-19 pandemic in Bengaluru excluded a significant proportion of poor and vulnerable communities from its ambit of representation. The article presents the findings of a sense-making tool which the platform developed for interpreting emerging signals from wastewater data to map disease progression and identifying the inflection points in the epidemiological curve. Thus, the platform accurately generated early warning signals on disease escalation and disseminated it to the government and the general public. This information enabled concerned audiences to implement preventive measures in advance and effectively plan their next steps for improved disease management.

Objective:To determine the benefits of the combination of bacillus Calmette-Guerin (BCG) and mitomycin C (MMC) in comparison to BCG alone in the treatment of patients with non-muscle invasive (NMI) bladder cancer.Materials and Methods:The randomized comparative study was conducted on 54 patients with NMI bladder cancer. Following the transurethral resection, patients were randomly grouped into two: Group A (n=27) included patients who received postoperative MMC at 40 mg diluted in 50 mL of normal saline on postoperative day 1, followed by intravesical BCG at 60 mg per week for 6 weeks and BCG monthly for 1 year, and group B (n=27) included patients who received intravesical BCG at 60 mg postoperatively per week for 6 weeks followed by BCG monthly for 1 year. The outcome measures were time to recurrence, progression of the disease to muscles or other organs, overall survival, and treatment-related side effects.Results:Compared to BCG alone, perioperative MMC in combination with BCG had comparable disease-free survival (85.18% vs. 66.66%, p=0.202), recurrence of disease (14.81% vs. 33.33%, p=0.202), and progression rate (11.1% vs. 25.9%, p=0.293). Side effects were minor and comparable between the study groups, which included dysuria, bacterial cystitis drug-induced cystitis, macroscopic hematuria prostatitis epididymitis fever, influenza-like symptoms, and fatigue.Conclusion:Overall, both protocols were found comparable in safety and efficacy in reducing the progression and recurrence of NMI bladder cancers without any significant superiority of MMC in combination with BCG in comparison to BCG alone.

The contamination of soils with heavy metals and its associated hazardous effects are a thrust area of today’s research. Rapid industrialization, emissions from automobiles, agricultural inputs, improper disposal of waste, etc., are the major causes of soil contamination with heavy metals. These contaminants not only contaminate soil but also groundwater, reducing agricultural land and hence food quality. These contaminants enter the food chain and have a severe effect on human health. It is important to remove these contaminants from the soil. Various economic and ecological strategies are required to restore the soils contaminated with heavy metals. Phytoremediation is an emerging technology that is non-invasive, cost-effective, and aesthetically pleasing. Many metal-binding proteins (MBPs) of the plants are significantly involved in the phytoremediation of heavy metals; the MBPs include metallothioneins; phytochelatins; metalloenzymes; metal-activated enzymes; and many metal storage proteins, carrier proteins, and channel proteins. Plants are genetically modified to enhance their phytoremediation capacity. In Arabidopsis, the expression of the mercuric ion-binding protein in Bacillus megaterium improves the metal accumulation capacity. The phytoremediation efficiency of plants is also enhanced when assisted with microorganisms, biochar, and/or chemicals. Removing heavy metals from agricultural land without challenging food security is almost impossible. As a result, crop selections with the ability to sequester heavy metals and provide food security are in high demand. This paper summarizes the role of plant proteins and plant–microbe interaction in remediating soils contaminated with heavy metals. Biotechnological approaches or genetic engineering can also be used to tackle the problem of heavy metal contamination.

Purpose – Despite the trend, managing and maximizing the effectiveness of blended workforce is not well-understood. The purpose of this paper is to institutionalize a blended workforce model in the post-COVID era, that is, a movement from homogenous workforce to heterogenous workforce of full-time employees working in tandem with gig talents connected via digital platforms. Design/methodology/approach – The evolution of gig economy is presented for contextualizing the development of prospective business models in the post-COVID era to establish clarity on the relationship between the employers and the blended workforce. To achieve this conceptual switch, a framework is proposed to support this type of workforce for creating a fair balance. Findings – By drawing on the concepts of various talent management functions, propositions were made predicting that the alignment of the multilateral activities of the gig workers with permanent workforce will be leveraged in the future to address the needs of short-term specialized skill-sets and scalable operations while creating a fair balance through a flexible and agile workforce. Originality/value – First, the paper explores how bridging the gap between the traditional and gig workforce can impact the key antecedents of a blended workforce ensuring a fair trial. Second, on an economical level, the COOKIE framework proposed in the paper is expected to play a crucial role in creating new job opportunities, boosting employee morale while minimizing costs and increasing productivity of the organizations.

Longevity in life span of city dwellers depends upon the quality of ambient air, which is often compromised due to urbanization. In this context, since independence, Bengaluru has rampantly grown with elevated air and automobile pollution levels. Ambient Carbon Monoxide [CO], which is considered to be the most dangerous criteria air pollutant, however has insufficient research/literature for the city of Bengaluru. To plug this gap, the pre-sent study engages a detailed examination for Temporal and Spatial Map-ping variation of Ambient ‘CO’ concentration in different wards of Bengaluru using calibrated Digital ‘CO’ sensor. The methodology involved primarily an intensive literature review followed by reconnaissance survey for fixation of monitoring stations about each ‘ward’. Further at these observatories, extensive primary data collection was ensued at periodic and regular intervals. The objective serves to delineate the most critical and non-critical places in Bengaluru to incorporate remedial measures. Startling 400+ violations with potential of triggering congestive heart failure, impaired performance in time discrimination, shortened time to angina response and vigilance decrement were observed. Sustainability measures conclude the study.

Cervical cancer (CC), the fourth worldwide incessant malignant neoplasia particularly in women, emerged as a serious public health problem. Metabolic syndrome (MetS) is an accumulation of different threats aspects, which could eventually cause diabetes and cardiovascular disease as co-morbidity, further contributing to mortality that is detected by central obesity, dyslipidemia, hyperglycemia, and hypertension. Accumulatively, endemic as well as research data supports the notion of MetS increasing general malignancies occurrences, further contributing to mortality rate. Nano-scale drug delivery system, provide targeted drug delivery, increases drug absorption rates, while drug decomposition and systemic adverse effects get decreased. This article has described the different metabolic syndromes playing role in cervical cancer, alongside cervical cancer nanocarriers modern therapy.

Gram pod borer (Helicoverpa armigera) is a major insect pest of chickpea. The gram pod borer begins to infest at vegetative stage and later feeds on flowers and developing pods. A field investigation was conducted to evaluate the bioefficacy of certain new molecule insecticides against Helicoverpa armigera (Hübner) on chickpea during Rabi 2020-21 in randomized block design with three replications. The outcomes revealed that the application of Chlorantraniliprole 18.5% SC @ 25g a.i./ha and Cyantraniliprole 10.26% OD @ 60g a.i./ha were established to be most effective treatments and application of Fipronil 5% SC @ 50g a.i./ha was least effective in respect of reduction of H. armigera larval population. The maximum yield was recorded in Chlorantraniliprole 18.5% SC @ 25g a.i./ha (14.00 q/ha) followed by Cyantraniliprole 10.26% OD @ 60g a.i./ha (13.73 q/ha) and lowest yield was recorded from Novaluron 75g a.i./ha (10.15 q/ha) treated plot. The economics of different new molecule insecticides indicated that higher benefit cost ratio (BCR) was observed from Lambda Cyhalothrin 30g a.i./ha (7.86:1) followed by Emamectin benzoate 12g a.i/ha (6.75:1) and the lower BCR was recorded from Cyantraniliprole 60g a.i./ha (1.64:1) and Novaluron 75g a.i./ha (1.58:1). Chlorantraniliprole and Cyantraniliprole are newer group of insecticides, which are relatively safer and more effective against gram pod borer as comparison to conventional insecticides and can be used in successful management of this key pest of chickpea.

Lakes are an essential component of biogeochemical processes, and variations in lake level are regarded as indicators of climate change. For more than a decade, satellite altimetry has successfully monitored variation in water levels over inland seas, lakes, rivers, and wetlands. Through altimetry, the surface water levels are measured at varying temporal scales depending on the orbit cycle of the satellite. The futuristic mission of Surface Water and Ocean Topography (SWOT) scheduled to be launched in year 2022 shall offer the spatial coverage and resolution suitable for water level estimation and volume calculation in small water bodies like lakes worldwide. With a radar interferometer in Ka-band, SWOT proposes to provide two-dimensional maps of water heights 21 days repeat orbit configuration. Cycle average SWOT datasets for land will be developed with higher temporal resolution, with temporal resolution varying geographically. This work assesses the potential of SWOT for monitoring water volumes over a case study lake by analyzing SWOT like synthetic data produced using the SWOT simulator developed by the Centre National d'Etudes Spatiales (CNES). With SWOT relying on a novel technology, the initial 90 days of this mission after launch shall focus on an extensive calibration and validation. Firsthand results of SWOT-simulated water levels and volumes are presented over a case study region in the tropical band, namely, Pookode Lake, in the ecologically fragile district of Wayanad, Kerala, India. It is the second-largest freshwater lake in Kerala that is being affected by anthropogenic activities, causing huge depletion in lake water storage in the last four decades. Our analysis indicated that the lake region is subjected to a rise in temperature of 0.018°C per year. We further assess the potential of remote sensing and SWOT data to monitor water storage of Pookode Lake, which is undergoing a rapid change. Results show that the proxy water surface elevations have immense potential in scientific studies pertaining to lake monitoring across the world. Overall, the study shows the potential of SWOT for monitoring the variability of water levels and volumes in this region.

Background: Lack of treatment of novel Coronavirus disease led to the search of specific antivirals that are capable to inhibit the replication of the virus. The plant kingdom has demonstrated to be an important source of new molecules with antiviral potential. Purpose: The present study aims to utilize various computational tools to identify the most eligible drug candidate that have capabilities to halt the replication of SARS-COV-2 virus by inhibiting Main protease (Mpro) enzyme Methods: We have selected plants whose extracts have inhibitory potential against previously discovered coronaviruses. Their phytoconstituents were surveyed and a library of 100 molecules was prepared. Then, computational tools such as molecular docking, ADMET and molecular dynamic simulations were utilized to screen the compounds and evaluate them against Mpro enzyme. Results: All the phytoconstituents showed good binding affinities towards Mpro enzyme. Among them laurolitsine possesses the highest binding affinity i.e. -294.1533 kcal/mol. On ADMET analysis of best three ligands were simulated for 1.2 ns, then the stable ligand among them was further simulated for 20 ns. Results revealed that no conformational changes were observed in the laurolitsine w.r.t. protein residues and low RMSD value suggested that the Laurolitsine-protein complex was stable for 20 ns. Conclusion: Laurolitsine, an active constituent of roots of Lindera aggregata, was found to be having good ADMET profile and have capabilities to halt the activity of the enzyme. Therefore, this makes laurolitsine a good drug candidate for the treatment of COVID-19.

The main aim of the project is to develop a system to charge the electric vehicle battery continuously and to control the three phase grid system. The perturb and observe method is used to get the maximum power from the solar PV array. The electric vehicle battery is connected at bi directional converter at the DC link and the Dc link is also connected at to the voltage source converter, this electric vehicle battery gets charged at low load demand and discharged at high load demand. This converter usually maintains the maximum power at the DC link by this the electric vehicle battery can be charged by taking a low rated battery to store energy of extra power. An adaptive recursive digital filter is used in better way to the maximum results with grid-connected solar PV-Electric vehicle battery system. The system with recursive filter control is controllable when the solar insolation is changing and load demand change. VSC works efficiently without any disturbance under no solar power generation and transfers the reactive power to the grid. The project is done with the help of simulation in MATLAB and test results on a hardware prototype is done through both in steady state conditions and dynamic conditions.

In recent years, graphene as a biomaterial has received considerable attention due to its outstanding physio-mechanical properties. In the present work, we found superior tensile strength, hydrophobic and antibacterial activities in graphene incorporated polyvinyl alcohol (PVA) based nanofibers, prepared by electrospinning. A series of ultrafine graphene-based ink (G-Ink) incorporated PVA nanofiber (GPN) with different concentrations of graphene (0, 0.008, 0.02, 0.04, 0.06, and 0.08% w/v) was fabricated. To overcome difficulty in direct dispersing graphene into the PVA solution, we have prepared graphene-based ink for dispersing into polymer solution. The morphology, composition, mechanical behaviour, and surface wettability of these membranes was investigated. The Fourier transform infrared spectra and the Raman spectra confirmed the successful incorporation of graphene into the GPN. Graphene when used as a nanofiller in polymers, provides excellent mechanical properties (814%), hydrophobicity (40%), and antibacterial properties. In the current study we tested GPN membranes for preserving two fast rotting foods like coriander leaves and tomato. We found that GPN membrane can be used safely for food packaging applications to increase the shelf life of perishing food items, such as up to 3 days for coriander leaves and 8 days for tomatoes.

A 34-year-old female presented with unilateral, localized, linear, and brown lesion over the left upper eyelid causing cosmetic blemish. On clinical examination, upturning of the eyelash under the involved skin was seen and diagnosed as a case of cilium incarnatum externum. A simple excision followed by electrolysis for this unusual abnormality provides satisfactory results for both patient and the treating physician. Only 9 cases have been reported till date in world literature.

Small-group learning activities (SGLAs) offer varied active learning opportunities and student benefits, but higher education instructors do not universally adopt SGLAs, in part owing to management burdens. We designed and deployed the SmartGroup system, a tool-based approach to minimize instructor burdens while facilitating SGLAs and associated benefits by managing peer group formation and peer group work assessment. SmartGroup was deployed in one course over 10 weeks; iterations of SmartGroup were provided continuously to meet the instructor’s needs. After deployment, the instructor and teaching assistant were interviewed, and 20 anonymous post-study survey responses were collected. The system exposed students to new perspectives, fostered meta-cognitive opportunities, and improved weaker students’ performances while being predominantly well-received in terms of usability and satisfaction. Our work contributes to the literature an exploration of tool-assisted peer group work assessment in higher education and how to promote wider SGLA adoption.

Parkinson’s disease (PD) is characterised by dopaminergic neuronal loss in the brain area. PD is a complex disease that deteriorates patients’ motor and non-motor functions. In experimental animals, the neurotoxin 6-OHDA induces neuropathological, behavioural, neurochemical and mitochondrial abnormalities and the formation of free radicals, which is related to Parkinson-like symptoms after inter-striatal 6-OHDA injection. Pathological manifestations of PD disrupt the cAMP/ATP-mediated activity of the transcription factor CREB, resulting in Parkinson’s-like symptoms. Forskolin (FSK) is a direct AC/cAMP/CREB activator isolated from Coleus forskohlii with various neuroprotective properties. FSK has already been proven in our laboratory to directly activate the enzyme adenylcyclase (AC) and reverse the neurodegeneration associated with the progression of Autism, Multiple Sclerosis, ALS, and Huntington’s disease. Several behavioural paradigms were used to confirm the post-lesion effects, including the rotarod, open field, grip strength, narrow beam walk (NBW) and Morris water maze (MWM) tasks. Our results were supported by examining brain cellular, molecular, mitochondrial and histopathological alterations. The FSK treatment (15, 30 and 45 mg/kg, orally) was found to be effective in restoring behavioural and neurochemical defects in a 6-OHDA-induced experimental rat model of PD. As a result, the current study successfully contributes to the investigation of FSK’s neuroprotective role in PD prevention via the activation of the AC/cAMP/PKA-driven CREB pathway and the restoration of mitochondrial ETC-complex enzymes.

Multiple sclerosis (MS) is a chronic neurodegenerative disease marked by oligodendrocyte loss, which results in central neuronal demyelination. AC/cAMP/CREB signaling dysregulation is involved in the progression of MS, including mitochondrial dysfunctions, reduction in nerve growth factors, neuronal inflammation, apoptosis, and white matter degeneration. Our previous research has shown that Forskolin (FSK), a naturally occurring direct adenylyl cyclase (AC)/cAMP/CREB activator, has neuroprotective potential to alleviate pathogenic factors linked with numerous neurological abnormalities. The current study intends to explore the neuroprotective potential of FSK at doses of 40 mg/kg and 60 mg/kg alone, as well as in combination with conventional medicines, such as Fingolimod (FNG), Donepezil (DON), Memantine (MEM), and Simvastatin (SIM) in EB-induced demyelinated experimental MS rats. Adult Wistar rats were divided into nine groups, and EB was infused stereotaxically in the rat brain’s intracerebropeduncle (ICP) area. Chronic gliotoxin EB treatment results in demyelination as well as motor and cognitive dysfunctions. FSK, combined with standard medications, improves behavioral dysfunctions, such as neuromuscular and motor deficits and memory and cognitive abnormalities. Following pharmacological treatments improved remyelination by enhancing myelin basic protein and increasing AC, cAMP, and CREB levels in brain homogenates. Furthermore, FSK therapy restored brain mitochondrial-ETC complex enzymes and neurotransmitter levels while decreasing inflammatory cytokines and oxidative stress markers. The Luxol fast blue (LFB) stain results further indicate FSK’s neuroprotective potential in preventing oligodendrocyte death. Therefore, the results of these studies contribute to a better understanding of the possible role that natural phytochemicals FSK could have in preventing motor neuron diseases, such as multiple sclerosis.

Conflict between patient and health care professionals is increasing over time. Traditionally, the medical profession has been considered the most honourable profession and the professionals are held with great esteem as they are considered next to God. Unfortunately, there seems to have been developed a distinct discord between the patient and the health care professional relationship with disturbing incidents catching the headlines. These conflicts range from simple disagreements to major violence against the very people who are trying to undertake patient’s care. Ultimately, it is in the best interest of both the patient and the health care professional to have a harmonious relationship based on utmost trust which is the very keynote of mutual understanding resulting in the benefit both of them. Authors are now trying with deep sincerity for mitigation of distrust and exploring possible resolution strategies to apply in clinical practice in order to overcome them.

BackgroundCholangiocarcinoma (CCA) is a rare disease, but it is amongst the most lethal cancers with a median survival under 1 year. Variations in DNA methylation and gene expression have been extensively studied in other cancers for their role in pathogenesis and disease prognosis, but these studies are very limited in CCA. This study focusses on the identification of DNA methylation and gene expression prognostic biomarkers using multi-omics data of CCA tumors from The Cancer Genome Atlas (TCGA).MethodWe have conducted a genome-wide analysis of differential DNA methylation and gene/miRNA expression using data from 36 CCA tumor and 9 normal samples from TCGA. The impact of DNA methylation in promoters and long-range distal enhancers on the regulation and expression of CCA-associated genes was examined using linear regression. Next, we conducted network analyses on genes which are regulated by DNA methylation as well as by miRNA. Finally, we performed Kaplan–Meier and Cox proportional hazards regression analyses in order to identify the role of selected methylation sites and specific genes and miRNAs in patient survival. We also performed real-time quantitative PCR (qPCR) to confirm the change in gene expression in CCA patients’ tumor and adjacent normal samples.ResultsAltered DNA methylation was observed on 12,259 CpGs across all chromosomes, of which 78% were hypermethylated. We observed a strong negative relationship between promoter hypermethylation and corresponding gene expression in 92% of the CpGs. Differential expression analyses revealed altered expression patterns in 3,305 genes and 101 miRNAs. Finally, we identified 17 differentially methylated promoter CpGs, 72 differentially expressed genes, and two miRNAs that are likely associated with patient survival. Pathway analysis suggested that cell division, bile secretion, amino acid metabolism, PPAR signaling, hippo signaling were highly affected by gene expression and DNA methylation alterations. The qPCR analysis further confirmed that MDK, HNF1B, PACS1, and GLUD1 are differentially expressed in CCA.ConclusionBased on the survival analysis, we conclude that DEPDC1, FUT4, MDK, PACS1, PIWIL4 genes, miR-22, miR-551b microRNAs, and cg27362525 and cg26597242 CpGs can strongly support their use as prognostic markers of CCA.

Purpose: Early onset opacification of the lens (cataract) has been observed to be on the rise globally. This study was conducted to determine the various types of presenile cataract and to determine the probable risk factors associated with the occurrence of presenile cataract. Materials and Methods: Patients in the age group of 18–40 years attending the Out-Patient Clinic of the Ophthalmology Department who were found to have presenile cataract were recruited as cases. An equal number of consecutive patients of the same age group were included in the control group. Those who gave consent to participate in the study were interviewed through a preformed questionnaire and underwent a complete ocular examination and set of blood investigations. The type of cataract was noted, data obtained were compiled, and examination and investigations done were documented and analyzed using frequency distribution and Chi-squared test. Results: In total, 90 cases and 90 controls that fulfilled the inclusion criteria were recruited. Most common type of cataract was found to be posterior subcapsular cataract. Presenile cataract was observed to be significantly associated with tobacco intake (P = 0.035), hypercholesterolemia (P = 0.002), fuel exposure (P = 0.004), and lower socioeconomic status (P =

In middle Gangetic plain, high arsenic concentration is present in water, which causes a significant health risk. Total 48 morphologically distinct arsenite resistant bacteria were isolated from middle Gangetic plain. The minimum inhibitory concentration (MIC) values of arsenite varied widely in the range 1–15 mM of the isolates. On the basis of their MIC, two isolates, AK1 (KY569423) and AK9 (KY569424) were selected. The analysis of the 16S rRNA gene sequence of selected isolates revealed that they are belong to the genus Pseudomonas. The AgNO3 test based microplate method revealed that isolates, AK1 and AK9, have potential in transformation of arsenic species. Further, the presence of aoxR, aoxB and aoxC genes in the both isolated strain AK1 and AK9 was confirmed, which play an important role in arsenic bioremediation by arsenite oxidation. Isolated strains also showed heavy metal resistance against Cr(IV), Ni(II), Co(II), Pb(II), Cu(II), Hg(II), Ag(I) and Cd(II).

Pancreatic ductal adenocarcinoma (PDAC) is the most common and among the deadliest of pancreatic cancers. Its 5-year survival is only ∼8%. Pancreatic cancers are a heterogeneous group of diseases, of which PDAC is particularly aggressive. Like many other cancers, PDAC also starts as a pre-invasive precursor lesion (known as pancreatic intraepithelial neoplasia, PanIN), which offers an opportunity for both early detection and early treatment. Even advanced PDAC can benefit from prognostic biomarkers. However, reliable biomarkers for early diagnosis or those for prognosis of therapy remain an unfulfilled goal for PDAC. In this study, we selected 153 PDAC patients from the TCGA database and used their clinical, DNA methylation, gene expression, and micro-RNA (miRNA) and long non-coding RNA (lncRNA) expression data for multi-omics analysis. Differential methylations at about 12,000 CpG sites were observed in PDAC tumor genomes, with about 61% of them hypermethylated, predominantly in the promoter regions and in CpG-islands. We correlated promoter methylation and gene expression for mRNAs and identified 17 genes that were previously recognized as PDAC biomarkers. Similarly, several genes (B3GNT3, DMBT1, DEPDC1B) and lncRNAs (PVT1, and GATA6-AS) are strongly correlated with survival, which have not been reported in PDAC before. Other genes such as EFR3B, whose biological roles are not well known in mammals are also found to strongly associated with survival. We further identified 406 promoter methylation target loci associated with patients survival, including known esophageal squamous cell carcinoma biomarkers, cg03234186 (ZNF154), and cg02587316, cg18630667, and cg05020604 (ZNF382). Overall, this is one of the first studies that identified survival associated genes using multi-omics data from PDAC patients.
