Inborn errors of metabolism (IEM) are a heterogeneous group of rare genetic disorders that are generally transmitted as autosomal or X-linked recessive disorders. These defects arise due to mutations associated with specific gene(s), especially the ones associated with key metabolic enzymes. These enzymes or their product(s) are involved in various metabolic pathways, leading to the accumulation of intermediary metabolite(s), reflecting their toxic effects upon mutations. The diagnosis of these metabolic disorders is based on the biochemical analysis of the clinical manifestations produced and their molecular mechanism. Therefore, it is imperative to devise diagnostic tests with high sensitivity and specificity for early detection of IEM. Recent advances in biochemical and polymerase chain reaction-based genetic analysis along with pedigree and prenatal diagnosis can be life-saving in nature. The latest development in exome sequencing for rapid diagnosis and enzyme replacement therapy would facilitate the successful treatment of these metabolic disorders in the future. However, the longterm clinical implications of these genetic manipulations is still a matter of debate among intellectuals and requires further research.

Background: Lablab purpureus L. is a legume appropriate to grow in tropical environment as it is adaptable to a broad temperature spectrum, rainfall and altitudes. It develops well under humid and warm conditions at temperatures ranging between 18oC and 30oC. It is a drought hardy crop grown in humid and semi-arid regions. The species is propagated by seed and can be sown alone or intercropped; planted near hedges or near other less leafy plants to climb on. Lablab purpureus L. has a flowering response of short-day, with early and late flowering types. Some landraces flower after sowing as early as 55 days. It is predominantly self pollinated although some out crossing is known to occur. Methods: In this study, Lablab purpureus L. genetic diversity were analyzed utilizing AFLP markers on ten Indian Lablab purpureus L. accessions at Department of Botany, Bundelkhand University, Jhansi during 2019-20. Five primers set were used for checking the diversity in Lablab purpureus L. and the marker performance were measured by using 4 parameters i.e. PIC, Mean resolving Power, Resolving Power and MI. Result: A total of 52 bands were generated, of which 45 bands (88.46%) were polymorphic which varies from 6 (M-CAT/E-ACT ) to 13 (M-CAC/E-ACC, M-CAC/E-ACT) with mean of 9.00 bands per primer. The PIC value ranges between 0.55 and 0.76 with a mean value of 0.64. The average value of matrix index is 3.04. Higher the MI, better the marker is. The value of RP ranges between 2.60 and 7.80 with a mean value of 4.68. In the UPGMA dendrogram, the 10 accessions were separated into two main clusters with all the primers used. In this study, we have shown that molecular markers can be used successfully to understand the genetic structure of the accessions of Lablab purpureus L. and identified a series of sequence-specific markers that potentially could be used in Lablab purpureus L. genetic mapping analyses. Since Lablab purpureus L. has been considered as a promising crop because of its wide spectrum of adaptability to different ecological conditions than other legumes, participatory evaluation at an early breeding stage could shape the variety being developed to increase the productivity and other traits as well as for utilization as food.

Type 2 diabetes (T2D) is a complex disease with an elusive link between its molecular aetiology and clinical presentation. Although, the role of visceral adipose tissue in insulin-resistance and T2D is known, limited information is available on the role of peripheral-subcutaneous adipose tissue especially in Asian Indians. In this microarray-based study of diabetic and normal glucose tolerant Asian Indians, we generated the transcriptome of their thigh adipose tissue and analyzed differentially expressed genes (DEGs) using weighted gene co-expression network analysis; further we identified perturbed pathways implicated by these DEGs in relevant co-expression modules. We also attempted to link these pathways with known aspects of T2D pathophysiology in terms of their association with some of their intermediate traits, namely; adipocyte size, HOMA-B, HOMA-R, Hb1Ac, insulin, glucose-level, TNF-α, IL-6, VLDLs, LDLs, HDLs, and NEFAs. It was observed that several modules of co-expressed genes show an association with diabetes and some of its intermediate phenotypic traits mentioned above. Therefore, these findings suggest a role of peripheral subcutaneous adipose tissue in the pathophsiology of T2D in Asian Indians. Additionally, our study indicated that the peripheral subcutaneous adipose tissue in diabetics shows pathologic changes characterized by adipocyte hypertrophy and up-regulation of inflammation-related pathways., Graphical Abstract

The roles of abdominal visceral (VAT) and subcutaneous adipose tissue (SAT) in the molecular pathogenesis type-2 diabetics (T2D) among Asian Indians showing a &ldquo, thin fat&rdquo, phenotype largely remains obscure. In this study, we generated transcription profiles in biopsies of these adipose depots obtained during surgery in 19 diabetics (M: F ratio, 8:11) and 16 (M: F ratio 5:11) age- and BMI-matched non-diabetics. Gene set enrichment analysis (GSEA) was used for comparing transcription profile and showed that 19 gene sets, enriching inflammation and immune system-related pathways, were upregulated in diabetics with F.D.R. &lt, 25% and &gt, 25%, respectively, in VAT and SAT. Moreover, 13 out of the 19 significantly enriched pathways in VAT were among the top 20 pathways in SAT. On comparison of VAT vs. SAT among diabetics, none of the gene sets were found significant at F.D.R. &lt, 25%. The Weighted Gene Correlation Analysis (WGCNA) analysis of the correlation between measures of average gene expression and overall connectivity between VAT and SAT was significantly positive. Several modules of co-expressed genes in both the depots showed a bidirectional correlation with various diabetes-related intermediate phenotypic traits. They enriched several diabetes pathogenicity marker pathways, such as inflammation, adipogenesis, etc. It is concluded that, in Asian Indians, diabetes pathology inflicts similar molecular alternations in VAT and SAT, which are more intense in the former. Both adipose depots possibly play a role in the pathophysiology of T2D, and whether it is protective or pathogenic also depends on the nature of modules of co-expressed genes contained in them.

2 Abstract: Shortage of supply of good quality protein for meeting the requirements of increasing animal and human population has necessitated search for additional sources. Leaf protein concentrate (LPC), a concentrated form of proteins derived from the foliage of plants, is an inexpensive and most abundant source of available protein. Their protein value equals that of most animal products. Trees have been suggested as a potential source of LPC and the production of protein from tree leaves is advantageous over crops as they do not involve the recurring cost of cultivation. Shorea robusta is planted on large scale in India. The potential of its leaves as protein source has not been investigated. The present communication describes the isolation of LPCs, mineral elements from the leaves of Shorea robusta and its chemical evaluation. Fresh and matured leaves of Shorea robusta yield fairly good amount of LPC (5.96 g) per 100 g leaves. It was also found to contain very high amount of ash (9.24%), which consisted of calcium, Iron, Phosphorus, Potassium, Sulphur micronutriets. Comparing all the biochemical analysis, the LPCs recovered from Shorea robusta shows fairly good amount of protein-37.25 %, fat-7.41%, nitrogen free extract-37.85%, total carbohydrates-45.5%, total soluble sugar-1.94% along with low amount of anti-nutritio nal factors such as total phenolics and total saponins. Thus it was inferred that the leaves of the Shorea robusta have potential for its use in production of LPCs. However, detailed studies regarding minerals, vitamins, in-vitro digestibility, toxicity testing and amino acid compositions are needed to be carried out to standardize the use of leaves as a source of LPCs.

A total forty samples of eight different medicinal plants were taken for the detection of toxigenic fungi and their mycotoxins. The fungal mycroflora comprises of six different fungal species belonging to three genera. A. niger, Mucor species, A. flavus and Rhizopus species dominate other fungal species isolated. Among ten samples of different medicinal plants which were contaminated with A. flavus was further analysed for mycotoxins potential. Four of them shows positive results for mytoxins potential. Although the presence of toxigenic fungi in a product did not imply the presence of mycotoxins in the product, their presence represents a potential risks of contamination with mycotoxins. Therefore, these medicinal plants should be carefully stored and the growth of the naturally found toxic fungi should be inhibited. Plant extracts are very cost effective and eco-friendly, thus, can be an economic and efficient alternative for the large-scale synthesis of nanoparticles. The preparation of stable, uniform silver nanoparticles by reduction of silver ions with Emblica officinalis, Terminalia catappa and Eucalyptus hybrida extract is reported in the present paper. It is a simple process of global research interest for obtaining silver nanoparticles in least amount of time. These nanoparticles were characterized with UV-Vis spectroscopy, X-ray diffraction (XRD), transmission electron microscopy (TEM) and energy diffraction X- ray (EDX analysis which revealed that the silver nanoparticles are polydisperse and of different morphologies ranging from 20 to 80 nm in size. XRD results reveal that these nanostructures exhibit a face-centered cubic crystal structure. The UV/Vis spectra absorption peak confirms their production. Pioneering of reliable and eco-friendly process for synthesis of metallic nanoparticles biologically is an important step in the field of application of nanobiotechnology. Thus, these silver nanoparticles (Ag-NPs) may prove as a better candidate for drugs and can potentially eliminate the problem of chemical agents because of their biogenic nature. The indiscriminate use of antibiotics has fuelled the development of drug resistance at an alarming rate. To overpower this burning problem, the Ag-NPs may prove to be a universal solution.
