Abstract The evolution of fault friction during the interseismic period affects the mechanics of a future earthquake on the same fault patch. Frictional aging has been previously tied to time‐dependent contact area growth through observations made on rock analogs. However, our understanding of the processes that control frictional aging is limited and is dependent on experiments that explore only numerous mechanisms. We conduct slide‐hold‐slide experiments with a dual‐axis nanoindenter on single‐crystal surfaces of quartz and calcite. Our results show that frictional aging in diamond‐quartz contacts is independent of time and contact area, in stark contradiction to past experiments done on quartz‐quartz contacts in rocks. Diamond‐calcite contacts show modest frictional aging, but still well below previous reported values from calcite‐calcite contacts. These results suggest that frictional aging of like‐on‐like minerals may be of chemical origin, as suggested in recent studies with atomic force microscopy and molecular dynamics simulations.

Aging is important in the earthquake cycle where it affects the mechanical response of a future earthquake. Frictional aging has been studied extensively in the past through slide‐hold‐slide (SHS) experiments in rocks where the contact is slid at a constant velocity, then held stationary for a predetermined 'hold time', after which sliding resumes. These experiments show that the static friction measured at the end of the hold, as sliding commences, scales with the logarithm of hold time. This result is commonly interpreted as being due to contact area growth as observed in rock‐analogue materials (such as polymers). We conducted SHS experiments with a nanoindenter capable of inducing frictional sliding of a diamond tip over a substrate while continuously monitoring contact area. Experiments performed on single crystals of quartz and calcite show no or minimal frictional aging even though contact area grows with the logarithm of hold time. Our results suggest that frictional aging of these mineral contacts is due to a chemical mechanism and not explicitly due to contact area growth. Autonomous Underwater Vehicles (AUVs) operate independently using onboard batteries and data storage, necessitating periodic recovery for battery recharging and data transfer. Traditional surface-based launch and recovery (L&R) operations pose significant risks to personnel and equipment, particularly in adverse weather conditions. Subsurface docking stations provide a safer alternative but often involve complex fixed installations and costly acoustic positioning systems. This work introduces a comprehensive docking solution featuring the following two key innovations: (1) a novel deployable docking station (DDS) designed for rapid deployment from vessels of opportunity, operating without active acoustic transmitters; and (2) an innovative sensor fusion approach that combines the AUV’s onboard forward-looking sonar and camera data. The DDS comprises a semi-submersible protective frame and a subsurface, heave-compensated docking component equipped with backlit visual markers, an electromagnetic (EM) beacon, and an EM lifting device. This adaptable design is suitable for temporary installations and in acoustically sensitive or covert operations. The positioning and guidance system employs a multi-sensor approach, integrating range and azimuth data from the sonar with elevation data from the vision camera to achieve precise 3D positioning and robust navigation in varying underwater conditions. This paper details the design considerations and integration of the AUV system and the docking station, highlighting their innovative features. The proposed method was validated through software-in-the-loop simulations, controlled seawater pool experiments, and preliminary open-sea trials, including several docking attempts. While further sea trials are planned, current results demonstrate the potential of this solution to enhance AUV operational capabilities in challenging underwater environments while reducing deployment complexity and operational costs.

Abstract Background Plasmodium falciparum (Pf) is the leading protozoan causing malaria, the most devastating parasitic disease. To ensure transmission, a small subset of Pf parasites differentiate into the sexual forms (gametocytes). Since the abundance of these essential parasitic forms is extremely low within the human host, little is currently known about the molecular regulation of their sexual differentiation, highlighting the need to develop tools to investigate Pf gene expression during this fundamental mechanism. Methods We developed a high-throughput quantitative Reverse-Transcription PCR (RT-qPCR) platform to robustly monitor Pf transcriptional patterns, in particular, systematically profiling the transcriptional pattern of a large panel of gametocyte-related genes (GRG). Initially, we evaluated the technical performance of the systematic RT-qPCR platform to ensure it complies with the accepted quality standards for: (i) RNA extraction, (ii) cDNA synthesis and (iii) evaluation of gene expression through RT-qPCR. We then used this approach to monitor alterations in gene expression of a panel of GRG upon treatment with gametocytogenesis regulators. Results We thoroughly elucidated GRG expression profiles under treatment with the antimalarial drug dihydroartemisinin (DHA) or the metabolite choline over the course of a Pf blood cycle (48 h). We demonstrate that both significantly alter the expression pattern of PfAP2-G, the gametocytogenesis master regulator. However, they also markedly modify the developmental rate of the parasites and thus might bias the mRNA expression. Additionally, we screened the effect of the metabolites lactate and kynurenic acid, abundant in severe malaria, as potential regulators of gametocytogenesis. Conclusions Our data demonstrate that the high-throughput RT-qPCR method enables studying the immediate transcriptional response initiating gametocytogenesis of the parasites from a very low volume of malaria-infected RBC samples. The obtained data expand the current knowledge of the initial alterations in mRNA profiles of GRG upon treatment with reported regulators. In addition, using this method emphasizes that asexual parasite stage composition is a crucial element that must be considered when interpreting changes in GRG expression by RT-qPCR, specifically when screening for novel compounds that could regulate Pf sexual differentiation. In in vitro fertilization (IVF) treatments, early identification of embryos with high implantation potential is required for shortening time to pregnancy while avoiding clinical complications to the newborn and the mother caused by multiple pregnancies. Current classification tools are based on morphological and morphokinetic parameters that are manually annotated using time‐lapse video files. However, manual annotation introduces interobserver and intraobserver variability and provides a discrete representation of preimplantation development while ignoring dynamic features that are associated with embryo quality. A fully automated and standardized classifiers are developed by training deep neural networks directly on the raw video files of >6200 blastulation‐labeled and >5500 implantation‐labeled embryos. Prediction of embryo implantation is more accurate than the current state‐of‐the‐art morphokientic classifier. Embryo classification improves with video length where the most predictive images show only partial association with morphological features. Deep learning substitute to human evaluation of embryo developmental competence thus contributes to implementing single embryo transfer methodology.

OBJECTIVE: The goal of this study was to examine the effect of a structured, comprehensive nursing intervention on the course of rehabilitation over the first 6 mos after a first-ever stroke. DESIGN: Intervention (experimental) study. The participants were 155 elderly stroke survivors who were admitted to a geriatric rehabilitation department. Stratified random sampling generated intervention and control groups. The intervention was designed to work together with the routine rehabilitation program and consisted of 12 consecutive weekly meetings. The control group received only the routine rehabilitation program. Study variables were measured at entry to study and after 3- and 6-mo intervals. RESULTS: Although there were no significant differences between the two groups at entry to study, at 3 and 6 mos after stroke, the intervention group showed better scores than the control group in functional status (FIM instrument), depression (Geriatric Depression Scale), self-perceived health, self-esteem, and dietary adherence. CONCLUSION: The nursing intervention had both short- and long-term effects on functional, psychological, and emotional variables. Serious consideration should be given to implementing comprehensive nursing interventions during rehabilitation tailored to meet the specific needs of stroke survivors and their caregivers. The present study was carried out to examine the chemopreventive effects of carotenoids such as fucoxanthin, lycopene and lutein as well as curcumin and its derivative, tetrahydrocurcumin (THC), on development of putative preneoplastic aberrant crypt foci (ACF) in colons of mice initiated with 1,2-dimethylhydrazine dihydrochloride (DMH). Influence on proliferation of colonic crypt epithelial cells was also assessed in terms of 5-bromo-2'-deoxyuridine (BrdU) incorporation. five-week-old B6C3F1 male mice were divided into three groups, groups 1 and 2 being given DMH (20 mg/kg body wt, s.c.) twice a week for 3 weeks. Animals of group 1 were then treated with one of the test compounds, lycopene (0.005% and 0.0025%) or fucoxanthin (0.01%) in the drinking water and lutein (0.05T), curcumin (0.5%) or THC (0.5% and 0.2%) in the diet from weeks 5-12. Group 2 served as a carcinogen alone control and group 3 mice were given test compounds alone. All animals were killed at week 12. Numbers of ACF/mouse in the group 1 treated with fucoxanthin (47.1 ± 13.7), lutein (42.6 ± 19.6) or 0.5% THC (46.6 ± 17.7) were significantly decreased as compared to the control group 2 value (63.3 ± 19.4) (P <0.01). Numbers of aberrant crypts (ACs)/mouse were also significantly lower after treatment with lutein (79.9 ± 34.7) or 0.5% THC (81.8 ± 32.5) than in the control group (115.1 ± 37.1) (P <0.01). BrdU labeling indices (LI) in mice treated with lutein and 0.5% THC were significantly decreased in both upper and lower half compartments of colonic crypts as compared to the controls (P <0.05 and 0.01, respectively), especially the upper half data corresponding to reduction of ACs/mouse. The results thus suggest that fucoxanthin, lutein, and THC may have potential as chemopreventive agents against colon carcinogenesis. [ABSTRACT FROM PUBLISHER]

Craniofacial tissues undergo hard tissue development through mineralization and changes in physicochemical properties. This study investigates the mechanical and chemical properties of developing enamel, dentin, and bone in the mouse mandible. We employ a multi-modal, multi-scale analysis of the developing incisor and first molar at postnatal day 12 by integrating micro-computed tomography (microCT), nanoindentation (NI), energy dispersive spectroscopy (EDS), and Raman spectroscopy. Our findings demonstrate distinct patterns of mechanical, elemental, and chemical changes across mineralized tissues. These results suggest that mineral composition drives mechanical properties across different craniofacial hard tissues. Integrating multi-modal characterization of mineralized tissues opens new opportunities for investigating structure-function relationships in craniofacial biology and genetics., Competing Interests: Competing Interest Statement: No competing interests to disclose. Background: The blood-brain barrier (BBB), which separates the intravascular and neuropil compartments, characterizes the vascular bed of the brain and is essential for its proper function. Recent advances in imaging techniques have driven the development of methods for quantitative assessment of BBB permeability., Hypothesis/objectives: Permeability of the BBB can be assessed quantitatively in dogs with meningoencephalitis of unknown origin (MUO) and its status is associated with the occurrence of seizures., Animals: Forty dogs with MUO and 12 dogs without MUO., Methods: Retrospective, prospective cohort study. Both dynamic contrast enhancement (DCE) and subtraction enhancement analysis (SEA) methods were used to evaluate of BBB permeability in affected (DCE, n = 8; SEA, n = 32) and control dogs (DCE, n = 6; SEA, n = 6). Association between BBB dysfunction (BBBD) score and clinical characteristics was examined. In brain regions where BBBD was identified by DCE or SEA magnetic resonance imaging (MRI) analysis, immunofluorescent staining for albumin, glial fibrillary acidic protein, ionized calcium binding adaptor molecule, and phosphorylated mothers against decapentaplegic homolog 2 were performed to detect albumin extravasation, reactive astrocytes, activated microglia, and transforming growth factor beta signaling, respectively., Results: Dogs with BBBD had significantly higher seizure prevalence (72% vs 19%; P = .01) when compared to MUO dogs with no BBBD. Unlabelled: Selective mutism (SM) is a relatively rare childhood disorder and is underdiagnosed and undertreated. The purpose of the retrospective naturalistic study was to examine the long-term outcome of children with SM who were treated with specifically designed modular cognitive behavioral therapy (MCBT). Parents of 36 children who met diagnostic criteria of SM that received MCBT treatment were invited for a follow-up evaluation. Parents were interviewed using structured scales and completed questionnaires regarding the child, including the Selective Mutism Questionnaire (SMQ). Twenty-four subjects were identified and evaluated. Their mean age ± SD of onset of SM symptoms, beginning of treatment, and age at follow-up were 3.4 ± 1.4, 6.4 ± 3.1, and 9.3 ± 3.4 years, respectively. There was robust improvement from beginning of treatment to follow-up evaluation in SM, social anxiety disorder, and specific phobia symptoms. The recovery rate from SM was 84.2 %., Conclusion: SM-focused MCBT is feasible in children and possibly effective in inducing long-term reduction of SM and comorbid anxiety symptoms., What Is Known: • There are limited empirical data on selective mutism (SM) treatment outcome and specifically on cognitive-behavioral therapy, with the majority of studies being uncontrolled case reports of 1 to 2 cases each. • There is also limited data on the long-term outcome of children with SM following treatment. What is New: • Modular cognitive behavioral treatment is a feasible and possibly effective treatment for SM. Intervention at a younger age is more effective comparing to an older age. • Treatment for SM also decreases the rate of psychiatric comorbidities, including separation anxiety disorder and specific phobia.

Background: Depending on the definition used, malnutrition is prevalent among 20-50% of hospitalized patients. Routine nutritional screening is necessary to identify patients with or at increased risk for malnutrition. The Nutrition Risk Screening (NRS 2002) has been recommended as an efficient tool to identify the risk of malnutrition in adult inpatients., Objectives: To utilize the NRS 2002 to estimate the prevalence of malnutrition among newly hospitalized adult patients, and to identify risk factors for malnutrition., Methods: During a 5 week period, all adult patients newly admitted to all inpatient departments (except Maternity and Emergency) at Wolfson Medical Center, Holon, were screened using the NRS 2002. An answer of "yes" recorded for any of the Step 1 questions triggered the Step 2 screen on which an age-adjusted total score > or = 3 indicated high malnutrition risk., Results: Data were obtained from 504 newly hospitalized adult patients, of whom 159 (31.5%) were identified as being at high risk for malnutrition. Malnutrition was more prevalent in internal medicine than surgical departments: 38.6% vs. 19.1% (P < 0.001). Body mass index was within the normal range among subjects at high risk for malnutrition: 23.9 +/- 5.6 kg/m2 but significantly lower than in subjects at low malnutrition risk: 27.9 +/- 5.3 kg/m2 (P < 0.001). Malnutrition risk did not differ by gender or smoking status, but subjects at high malnutrition risk were significantly older (73.3 +/- 16.2 vs. 63.4 +/- 18.4 years, P < 0.001). Total protein, albumin, total cholesterol, low density lipoprotein-cholesterol, hemoglobin and % lymphocytes were all significantly lower, whereas urea, creatinine and % neutrophils were significantly higher in patients at high malnutrition risk., Conclusions: Use of the NRS 2002 identified a large proportion of newly hospitalized adults as being at high risk for malnutrition. These findings indicate the need to intervene on a system-wide level during hospitalization.

The aims of this study were to (1) delineate the sociodemographic profile of Israeli primary caregivers of stroke survivors; (2) examine the changes in caregivers' burden, physical and emotional health, social support network, and quality of life (QOL) during the first 6 months after the stroke occurrence; and (3) identify the predictors of caregivers' QOL for that time period. Primary caregivers (N = 140) were assessed at three intervals during this period: within 2 weeks after the stroke occurrence, in the geriatric rehabilitation ward, and at 3 and 6 months poststroke in the community. Results of the study revealed that during the 6-month period, caregivers' physical health remained stable. After 3 months, depression levels decreased, as did the need for instrumental support in and outside the home. Nevertheless, satisfaction with informal support and family relationships decreased, as did QOL. Burden decreased consistently at 3- and 6-month intervals. After 6 months, perception of health and QOL improved, whereas the other variables remained stable. These results indicate that overall, this population of caregivers adapted to their role over the 6-month period. At each of the three intervals, QOL was explained by the same four variables, although with differential weights: number of diseases, confidence in the support system, burden, and sharing a household with the survivor. The results of this study contribute to nurses' ability to identify caregivers at risk and develop appropriately timed interventions for empowering caregivers in their role fulfillment.
