Objective Microvesicles (MV) released from blood cells play an important role in the progress of diseases. The purpose of this study is to detect circulating MV level of patients suffering from severe fever with thrombocytopenia syndrome (SFTS). Methods The plasma samples of SFTS patients and healthy controls were collected. And MV from samples were isolated and MV levels were detected quantitatively. Results The results showed that the level of the circulating MV in SFTS patients was obviously higher than that of the healthy control, with statistically significant difference. Further analysis revealed that MV level was relevant to the severity of SFTS patients, namely the higher the concentration of MV, the more severity of the disease. Linear correlation analysis showed that the circulating MV level in SFTS was correlated positively with leukocyte count (r = 0.243, P < 0.05), but no correlation with the platelet count (r = 0.193, P >0.05). Conclusions We demonstrated that there existed high level of circulating MV in SFTS patients, and the level of circulating MV had a close relationship with the severity of the disease and leukocyte count. Therefore, monitoring the level of circulating MV may provide a new indicator for predicting the prognosis of SFTS patients.

It was reported that urinary modified nucleosides with abnormally high amounts were found in many cancer patients. This study was designed to investigate the usefulness of urinary nucleosides in the diagnosis of gastric carcinoma.The concentrations of 15 kinds of urinary nucleosides from 50 healthy persons and 48 patients with gastric carcinoma were determined by high-performance liquid chromatography (HPLC). Of 48 patients with gastric carcinoma, 25 underwent serum CEA examination.The average levels of 14 kinds of urinary nucleosides, m5U excepted, from patients with gastric carcinoma were higher than those from health persons (P0.05). The level of nucleoside I was positively correlated with the tumor size (P0.05). The level of nucleoside X was positively correlated with lymph node metastasis (P0.05). Using the concentrations of 15 nucleosides as the data vectors, principal component analysis was applied to classify gastric cancer patients and normal adults, 63%(30/48) of cancer patients were correctly classified, in which the identification rate was higher than that of CEA method (12%).Urinary modified nucleoside increased in the patients with gastric carcinoma, and it may be helpful in the diagnosis of gastric carcinoma.

A rapid/high replicative strain of human immunodeficiency virus type 1 (HIV-1) (BC9101) was isolated directly in the Jurkat-tat III cell line from a Chinese patient with AIDS. The thin-section electron microscopy was performed and revealed high efficiency of replication of BC9101 with some unusual biological properties. Many vacuoles, most of them filled with HIV particles, were found close to the nucleus. Double-cored virions and double budding were frequently observed in the vacuoles and at the vacuolar membrane. Virus particles matured by budding both into intracytoplasmic vacuoles and through the plasma membrane. Inclusion bodies of varying sizes, some consisting of thousands of HIV particles, were found in the cytoplasm. All the illustrated features describing formation of inclusion bodies were compatible with the observation that HIV particles were assembled at and budded from the cytoplasmic vacuole membrane. They were then released from the membrane into the vacuoles, and subsequently, the maturation occurred. Some of the vacuoles accumulated to such a high number of mature virus particles that inclusion bodies were formed. During the disintegration of the cells, the inclusion bodies surrounded by the vacuolar membrane were released from the cells. The nucleotide sequence of the vpu gene of BC9101 was investigated and indicated that the unusual biological properties may due to the lack of a start codon for translation of the vpu protein.
