Abstract

Advantages and disadvantages of alphabetic and logographic writing systems are discussed, with particular emphasis on Blissymbolics as an example of a logographic system. It is concluded that logographic systems might be easier to learn than alphabetic systems and, in particular, that Blissymbols could be a useful communication device for people with various kinds of handicaps. This suggestion is substantiated by empirical research.

Abstract

An alternative pathway, which involves several serine proteases for the production of angiotensin II (ANG II), exists in human detrusor smooth muscle. This serine protease in human detrusor smooth muscle changes ANG I to ANG II. Serial protease inhibitors such as soybean trypsin inhibitor and chymostatin completely block ANG II formation, which remains uninhibited by angiotension-converting enzyme.

Abstract

General cognitive function is substantially heritable across the human life course from adolescence to old age. We investigated the genetic contribution to variation in this important, health-and well-being-related trait in middle-aged and older adults. We conducted a meta-analysis of genome-wide association studies of 31 cohorts (N = 53 949) in which the participants had undertaken multiple, diverse cognitive tests. A general cognitive function phenotype was tested for, and created in each cohort by principal component analysis. We report 13 genome-wide significant single-nucleotide polymorphism (SNP) associations in three genomic regions, 6q16.1, 14q12 and 19q13.32 (best SNP and closest gene, respectively: rs10457441, P = 3.93 x 10(-9), MIR2113; rs17522122, P = 2.55 x 10(-8), AKAP6; rs10119, P = 5.67 x 10(-9), APOE/TOMM40). We report one gene-based significant association with the HMGN1 gene located on chromosome 21 (P = 1x10(-6)). These genes have previously been associated with neuropsychiatric phenotypes. Meta-analysis results are consistent with a polygenic model of inheritance. To estimate SNP-based heritability, the genome-wide complex trait analysis procedure was applied to two large cohorts, the Atherosclerosis Risk in Communities Study (N = 6617) and the Health and Retirement Study (N = 5976). The proportion of phenotypic variation accounted for by all genotyped common SNPs was 29% (s.e. = 5%) and 28% (s.e. = 7%), respectively. Using polygenic prediction analysis, similar to 1.2% of the variance in general cognitive function was predicted in the Generation Scotland cohort (N = 5487; P = 1.5 x 10(-17)). Abstract

The Enhancing NeuroImaging Genetics through Meta-Analysis (ENIGMA) Consortium is a collaborative network of researchers working together on a range of large-scale studies that integrate data from 70 institutions worldwide. Organized into Working Groups that tackle questions in neuroscience, genetics, and medicine, ENIGMA studies have analyzed neuroimaging data from over 12,826 subjects. In addition, data from 12,171 individuals were provided by the CHARGE consortium for replication of findings, in a total of 24,997 subjects. By meta-analyzing results from many sites, ENIGMA has detected factors that affect the brain that no individual site could detect on its own, and that require larger numbers of subjects than any individual neuroimaging study has currently collected. ENIGMA's first project was a genome-wide association study identifying common variants in the genome associated with hippocampal volume or intracranial volume. Continuing work is exploring genetic associations with subcortical volumes (ENIGMA2) and white matter microstructure (ENIGMA-DTI). Working groups also focus on understanding how schizophrenia, bipolar illness, major depression and attention deficit/hyperactivity disorder (ADHD) affect the brain. We review the current progress of the ENIGMA Consortium, along with challenges and unexpected discoveries made on the way.

37. Leisure-time activity in old age as predictors of impending dementia: A 15-year prospective study.

Abstract

This study examined the relationship between leisure activities and risk of dementia in a sample of healthy older individuals, dementia free at the beginning of the project. Data were drawn from a population-based longitudinal study (the Betula project) and the participants were followed up for 15 years. At baseline, participants were asked about their frequency of participation in 15 selected leisure activities. When age, gender, education, APOE and other potential confounders were controlled for, results revealed quite moderate effects on dementia after analysis of the activities separately. However, by weighting each activity into a mental, social and physical dimension (based on valuation by the participants), and then summarizing into a score for each dimension, we further investigated if level of engagement could predict impending dementia. Preliminary results indicate that the dimensions may have influence on the risk of dementia for certain age groups. The study also showed that the strongest predictor of dementia is being a carrier of the APOE ɛ4 allele. The outcomes are discussed in terms of important methodological difference between studies concerning the effects of leisure activities in preventing dementia diseases.

Abstract

Two representative samples of adult Norwegians (n=2000) were asked a set of general and specific questions regarding their beliefs and opinions about human memory. The results indicate that on many questions, such as time of the earliest memories, inhibiting effects of collaboration, and memory for dramatic versus ordinary events, the views of the general public concurred with current research findings, and people in general had realistic views about their own memory performance. On other questions, such as the reliability of olfactory as compared with visual and auditory memory, the memory of small children in comparison with that of adults, the likelihood of repression of adult traumatic memories, and on more general questions such as the possibility of training memory and the capacity limitations of long-term memory, a large proportion of the participants expressed views that are less supported by scientific evidence. Implications of these findings are briefly discussed. © 2006 Psychology Press Ltd.

Abstract

Malfunctioning of cyclin-dependent kinase 5 (CDK5) through aberrant proteolytic cleavage of its neuronal activators p35 and p39 is involved in neurodegeneration in Alzheimer's disease (AD) and other neurodegenerative brain diseases. By extensive genetic analysis of the genes encoding CDK5 (CDK5), p35 (CDK5RI) and p39 (CDK5R2), we excluded causal mutations in 70 familial early-onset AD patients. We performed an association study with five informative SNPs in CDK5 in two independent samples of early-onset AD patients and matched control individuals from The Netherlands and northern Sweden. Association was observed with g.149800G > C in intron 5 of CDK5, and a two times increased risk was observed in both patient samples for carriers of the C-allele. Our data are indicative for a role of the CDK5 molecular complex in the genetic etiology of early-onset AD, and suggest that a yet unknown functional variant in CDK5 or in a nearby gene might lead to increased susceptibility for early-onset AD.

48. Cerebral atrophy as predictor of cognitive function in old, community-dwelling individuals

Abstract

Objectives: The impact of cortical and subcortical atrophy on cognitive function was examined in a sample of older community-dwelling men and women. Material and methods: Magnetic resonance imaging was performed on a sample of 129 individuals [age: 68.4 ± 3.6 years (mean ± SD), range 64–74 years, 64 women and 65 men, Mini-Mental State Examination scores above 23] to assess cortical and subcortical atrophy. Participants also performed a number of cognitive tasks, and the measures of atrophy were used to predict performance in these tasks. Results: In men, frontal cortical atrophy predicted worse performance in word fluency and the Stroop test, and occipital cortical atrophy was associated with poor performance in motor speed. In women, poor performance in motor speed was associated with subcortical atrophy at the level of the caudate nucleus. Conclusion: Atrophy in certain areas was associated with poor performance in specific cognitive tasks, although the amount of explained variance was rather limited in this quite homogenous sample.
