2. Three‐domain microbial communities in the gut of Pachnoda marginata larvae: A comparative study revealing opposing trends in gut compartments.

Abstract

This study aimed to examine the bacterial, methanogenic archaeal, and eukaryotic community structure in both the midgut and hindgut of Pachnoda marginata larvae using an amplicon sequencing approach. The goal was to investigate how various diets and the soil affect the composition of these three‐domain microbial communities within the gut of insect larvae. The results indicated a notable variation in the microbial community composition among the gut compartments. The majority of the bacterial community in the hindgut was composed of Ruminococcaceae and Christensenellaceae. Nocardiaceae, Microbacteriaceae, and Lachnospiraceae were detected in midgut samples from larvae feeding on the leaf diet, whereas Sphingomonadaceae, Rhodobacteraceae, and Promicromonasporaceae dominated the bacterial community of midgut of larvae feeding on the straw diet. The diet was a significant factor that influenced the methanogenic archaeal and eukaryotic community patterns. The methanogenic communities in the two gut compartments significantly differed from each other, with the midgut communities being more similar to those in the soil. A higher diversity of methanogens was observed in the midgut samples of both diets compared to the hindgut. Overall, the microbiota of the hindgut was more host‐specific, while the assembly of the midgut was more influenced by the environmental microorganisms. [ABSTRACT FROM AUTHOR]

Abstract

Summary: Anaerobic digestion (AD) is one of the oldest biotechnological processes and originally referred to biomass degradation under anoxic conditions in both natural and engineered systems. It has been used for decades to treat various waste streams and to produce methane-rich biogas as an important energy carrier, and it has become a major player in electrical power production. AD is a popular, mature technology, and our knowledge about the influencing process parameters as well as about the diverse microbial communities involved in the process has increased dramatically over the last few decades. To avoid competition with food and feed production, the AD feedstock spectrum has constantly been extended to waste products either rich in recalcitrant lignocellulose or containing inhibitory substances such as ammonia, which requires application of various pre-treatments or specific management of the microbial resources. Extending the definition of AD, it can also convert gases rich in hydrogen and carbon dioxide into methane that can substitute natural gas, which opens new opportunities by a direct link to traditional petrochemistry. Furthermore, AD can be coupled with emerging biotechnological applications, such as microbial electrochemical technologies or the production of medium-chain fatty acids by anaerobic fermentation. Ultimately, because of the wide range of applications, AD is still a very vital field in science. This Special Issue highlights some key topics of this research field.

Abstract

Tarım atıkları, yüksek enerji içerikleri ve üretim miktarları ile düşük maliyetleri nedeniyle anaerobik çürütücüler için en önemli biyokütle olarak değerlendirilmesine rağmen karmaşık lignoselülozik yapıları bu maddelerin hidroliz aşamalarını sınırlandırmaktadır. Hidroliz verimini artırmak için etkin lignoselüloz degradasyon stratejileri geliştirilmektedir. Bu çalışmanın amacı, Pachnoda marginata larvalarının arka bağırsağından lignoselüloz parçalayabilen anaerobik mikrobiyal topluluğun zenginleştirilmesidir. Bu kapsamda, Pachnoda marginata larvaları 3 hafta süre ile lignoselülozik substrat ile beslenmiş, sonrasında disekte edilmiştir. Arka bağırsak kültür şişelerine transfer edilmiştir. Bakteriyel topluluk çeşitliliği 16S rRNA amplikon dizileme yöntemi ile Illumina MiSeq platformunda analiz edilmiş, metanojenik arkeal topluluk ise T-RFLP yöntemi ile incelenmiştir. Kültür şişelerinde biyogaz üretimi zaman ile artış göstermiş, tüm şişelerde metan üretimi gözlenmiştir. Zenginleştirme prosedürü sonucunda, kültürün bakteriyel topluluk profili değişiklik göstermiş, üç transfer sonrasında alınan örneklerde Porphyromonadaceae (phylum: Bacteroidetes) bolluğunun artarak baskıladığı görülmüştür. Metanojenik topluluk ise Methanobrevibacter ile baskılanmıştır. Çalışmada elde edilen sonuçlar, Pachnoda marginata larvasının sindirim sisteminden üretilen zenginleştirilmiş kültürün, lignoselülozca zengin kompleks biyokütleyi etkin bir şekide parçalayabildiğini göstermiştir.
