Abstract

All parts of Neem (Azadirachta indica A. Juss) show a broad spectrum efficacy against insect pests including insecticidal, anti-feedant or insect repellent activities. Several studies have shown that plant cell cultures can produce azadirachtins. We induced more than 40 novel Neem cell lines in modified Murashige and Skoog (MS) media containing different concentrations of auxins and cytokinins. To enhance the Azadirachtin production from 1 mg/l, it was necessary to optimize the media composition separately for biomass and secondary metabolite production. In light of this complex challenge we used our novel fully automated high-throughput microbioreactor system that allows us a fast and controlled batch and fedbatch screening in 48-well microtiter plates. There is increasing evidence that plants like Azadirachta indica contain endophytes which are able to colonize internal plant tissue without causing visible disease symptoms. The estimated high species diversity of endophytes suggests a rich and almost untapped source of new secondary metabolites. We isolated more than 340 endophytes from various plant tissues and tested if they were able to produce Azadirachtin-A. Here, we present data on isolation of endophytes and induction of callus as well as first results of our microbioreactor system.

Abstract

The detection of groups of molecules that co-localize with histopathological patterns or sub-structures is an important step to combine the rich high-dimensional content of mass spectrometry imaging (MSI) with classic histopathological staining. Here we present the evolution of GRINE to COBI-GRINE, an interactive web tool that maps MSI data onto a graph structure to detect communities of laterally similar distributed molecules and co-visualizes the communities with Hematoxylin and Eosin (HE) stained images. Thereby the tool enables biologists and pathologists to examine the MSI image graph in a target-oriented manner and links molecular co-localization to pathology. Another feature is the manual optimization of cluster results with the assist of graph statistics in order to improve the community results. As the graphs can become very complex, those statistics provide good heuristics to support and accelerate the detection of sub-clusters and misclusterings. This kind of edited cluster optimization allows the integration of expert background knowledge into the clustering result and a more precise analysis of links between molecular co-localization and pathology.

Abstract

Hydrangea serrata, also knowen as the Japanese tea hortensia, is known for its sweet taste and health properties of bevarages produced from this plant. The H. serrata 3,4-dihydroisocoumarins, hydrangenol and phyllodulcin harbour a variety of biological activities and pharmacological properties. Therefore, a detailed understanding of dihydroisocoumarin biosynthesis in H. serrata is of major interest. Their biosynthesis is assumed to be enhanced by elicitors and mediated by polyketide synthases like in cases of phenylpropanoid derived phytoalexins. A de-novo transcriptome assembly of leaves and roots from the aluminium chloride treatment group versus the control group alongside with annotation was generated. Secondary plant metabolites were analysed by LCMS. It revealed that a terpene synthase and a triterpenoid synthase gene as well as lignin biosynthesis encoding genes were upregulated in roots. Many genes for transporters, glycosyl, and other transferases as well as glycosylases were found to be differentially expressed in both organs. As no differentially expressed polyketide synthase gene homolog was found, the relative leaf and root 3,4-dihydroisocoumarin content was analysed by LC-MS measurement. Although Hydrangea species are known for their aluminium detoxification using phenylpropanoid-derived compounds, the levels of 3,4-dihydroisocoumarins were not enhanced. In this metabolite analysis, an organ-specific accumulation profile of hydrangenol, phyllodulcin, hydrangeic acid and their mono- and diglycosides was figured out. [ABSTRACT FROM AUTHOR]

Abstract

Sigma factors are transcriptional regulators that are part of complex regulatory networks for major cellular processes, as well as for growth phase-dependent regulation and stress response. Actinoplanes sp. SE50/110 is the natural producer of acarbose, an α-glucosidase inhibitor that is used in diabetes type 2 treatment. Acarbose biosynthesis is dependent on growth, making sigma factor engineering a promising tool for metabolic engineering. ACSP50_0507 is a homolog of the developmental and osmotic-stress-regulating Streptomyces coelicolor σHSc. Therefore, the protein encoded by ACSP50_0507 was named σHAs. Here, an Actinoplanes sp. SE50/110 expression strain for the alternative sigma factor gene ACSP50_0507 (sigHAs) achieved a two-fold increased acarbose yield with acarbose production extending into the stationary growth phase. Transcriptome sequencing revealed upregulation of acarbose biosynthesis genes during growth and at the late stationary growth phase. Genes that are transcriptionally activated by σHAs frequently code for secreted or membrane-associated proteins. This is also mirrored by the severely affected cell morphology, with hyperbranching, deformed and compartmentalized hyphae. The dehydrated cell morphology and upregulation of further genes point to a putative involvement in osmotic stress response, similar to its S. coelicolor homolog. The DNA-binding motif of σHAs was determined based on transcriptome sequencing data and shows high motif similarity to that of its homolog. The motif was confirmed by in vitro binding of recombinantly expressed σHAs to the upstream sequence of a strongly upregulated gene.
