9. Comparison of Two DoseResponse Techniques to Study the Pancreatic Secretory Response to Intraduodenal Tryptophan in the Absence and Presence of the M1Receptor Antagonist Telenzepine

Abstract

To answer the questions if the type of continuous dose-response technique influences the pancreatic secretory response to intraduodenal tryptophan and if the M1-receptor antagonist telenzepine influences the intestinal absorption of tryptophan, we determined, in 12 conscious dogs with chronic gastric and duodenal fistulas, pancreatic bicarbonate and protein secretion and tryptophan plasma concentrations following intraduodenal tryptophan perfusion using two dose-response techniques. With an ascending continuous dose-response technique (aDRT), tryptophan was perfused in loads ranging from 0.12 to 10.0 mmol h−1starting with the lowest load and tripling it every 45 min. With the descending continuous dose-response technique (dDRT), the order of tryptophan loads was reversed, with the highest load being given first. All studies were done on a fixed background of intravenous secretin (20.5 pmol kg−1h−1) and repeated in the presence of the anticholinergic M1-receptor antagonist telenzepine (243 nmol kg−1h−1). The bicarbonate and protein response as well as the tryptophan plasma concentrations to the same loads of tryptophan did not differ significantly between the two techniques. Using both techniques, telenzepine significantly (p< 0.05) inhibited the overall pancreatic protein response by 65 (dDRT) to 81 (aDRT). The overall bicarbonate response was only numerically, and not statistically significantly, inhibited by telenzepine. Tryptophan plasma concentrations after duodenal perfusion with tryptophan were neither influenced by the order of tryptophan loads nor altered by telenzepine. These findings indicate that in the intact animal (a) either the aDRT or the dDRT may be used to study the pancreatic secretory response to enteral infusion of amino acids, (b) at least part of the pancreatic secretory response to tryptophan is mediated through cholinergic M1receptors, and (c) the duodenal absorption of tryptophan is not affected by telenzepine and therefore most likely not mediated by neuronal M1receptors.

Abstract

Background: Epigenetic changes in DNA methylation have recently been demonstrated to be involved in effector T-cell polarization, resulting in differential secretion of T(H)1 and T(H)2 cytokines. However, the contribution to the development of a chronic inflammatory phenotype remains still unclear., Objective: We sought to investigate changes in DNA methylation in marker genes of T-cell subsets during allergen sensitization/challenge and their influence on the development of an allergic airway inflammatory response., Methods: The relationship between changes in DNA methylation and phenotype development were examined in a well-established model of experimental asthma. DNA methylation was investigated at genomic loci associated with T(H)1 (IFNG promoter) or T(H)2 (conserved noncoding sequence 1 [CNS1]) cytokine production by using bisulfite pyrosequencing., Results: Analysis of CD4(+) T cells revealed a significant increase in DNA methylation at the IFNG promoter after allergen sensitization/challenge, which correlated with decreased IFN-γ cytokine expression, whereas only minor changes were observed at the CNS1 locus. Furthermore, the increase in DNA methylation at the IFNG promoter could be reversed with a DNA methyltransferase (DNMT) inhibitor in vitro and in vivo with beneficial effects on sensitization status and allergic phenotype. The specific importance of the DNA methylation status in CD4(+) T cells could be confirmed by using adoptive transfer experiments., Conclusion: We here report the novel finding that epigenetic regulation in T cells contributes to the development of experimental asthma and can be targeted pharmacologically., (Copyright © 2012 American Academy of Allergy, Asthma & Immunology. 13. The risk of involuntary firearms discharge.

Abstract

Objectives: To investigate whether unintended muscle activity induced by motor overflow can result in the unintentional discharge of a firearm., Background: In the past few years an increasing number of people have been injured, sometimes fatally, as a result of police weapons being discharged involuntarily., Methods: Two experiments were performed using a pistol fitted with sensors to register the pressure exerted on the hand stock and the trigger. In Study 1, 34 police officers reacted to a set case using the prepared pistol. In Study 2, 25 participants performed 13 tasks requiring the use of different limbs., Results: In Study 1, about 20% of participating officers made contact with the trigger, thus violating police regulations to keep the trigger finger outside the trigger guard at all times until a conscious decision to fire has been made. Study 2 showed that motor activity in different limbs leads to a significant increase in grip force exerted on a firearm (significant results for 12 of 13 trials, Bonferroni-corrected p < .004). Furthermore, pressure on the trigger resulting from involuntary muscle contractions was, in certain cases, sufficient to unintentionally discharge a weapon., Conclusion: Unintentional discharges attributable to involuntary muscle activity can be a real danger for police officers when carrying out their duties., Application: Results may help improve police training with the aim of reducing the danger of involuntary discharges in real-life situations.
