Breast tumor is a kind of woman′s disease with high incidence rate, it is also a kind of disease that can be diagnosed early and treated early. Thus the mortality rate could be reduced. The method of combining grey symbiotic matrix with BP neural network is proposed to improve the recognition rate of breast tumors. Firstly, the infrared breast image was pretreated to highlight the texture of lesions and blood vessels, and the texture features of such gray-scale curve images of the breast were extracted by using the gray co-occurrence matrix. Then, the sample data were trained through BP neural network. The trained BP neural network could effectively identify the lesion area. The experimental results show that the method presented in this paper has good recognition effect on the breast tumor lesion area.

In an effort to suppress cavitation in low-specific speed centrifugal pump, a method of changing the axial angle of slotted blade was proposed, and three oblique slotted blade schemes were designed. By using numerical simulation methods to compare the experimental results of centrifugal pumps, analyze the effect of blade oblique slotting on the suppression of cavitation in centrifugal pump. The simulation results show that the oblique slotted blade with an axial angle of 5° has the most significant cavitation suppression result on low-specific speed centrifugal pump, and the fracture head is increased by 0.281 m. The oblique slotted blade suppress the development of cavitation in the flow field of the centrifugal pump impeller, and the maximum reduction in cavitation volume is 67.25% during the severe cavitation stage; The oblique slotted blade optimize the flow field structure of the impeller, reduces the low-pressure area at the impeller inlet, and reduces the turbulent kinetic energy in the impeller channel. Esophageal cancer (EC) is the most aggressive malignancy in the gastrointestinal tract. Long noncoding RNA cyclin‐dependent kinase inhibitor 2 B antisense RNA 1 (CDKN2B‐AS1) is implicated in EC development. However, the specific mechanisms involved remain poorly defined. Therefore, this research aimed to explore the mechanism of action of CDKN2B‐AS1 in EC. Quantitative real‐time polymerase chain reaction was conducted to measure CDKN2B‐AS1 expression in EC cells and western blotting was utilized to evaluate transcription factor AP‐2 alpha (TFAP2A) and fascin actin‐bundling protein 1 (FSCN1) expression. After gain‐of‐function and loss‐of‐function assays, cell proliferation, migration, invasion, apoptosis, and apoptosis‐related protein expression were assessed using cell counting kit‐8, scratch tests, Transwell assays, flow cytometry, and western blotting, respectively. The binding relationship between CDKN2B‐AS1 and TFAP2A was assessed by RNA immunoprecipitation and RNA pull‐down assays. The binding relationship between TFAP2A and FSCN1 was evaluated using dual‐luciferase reporter and chromatin immunoprecipitation assays. Tumor xenografts from nude mice were used for in vivo verification. CDKN2B‐AS1, TFAP2A, and FSCN1 were upregulated in EC cells. Mechanistically, CDKN2B‐AS1 transcriptionally activated FSCN1 by recruiting TFAP2A to the FSCN1 promoter. Silencing CDKN2B‐AS1 or TFAP2A suppressed EC cell proliferative, migrating, and invasive properties and augmented apoptosis. TFAP2A was bound to CDKN2B‐AS1 and the FSCN1 promoter. Overexpression of TFAP2A or FSCN1 abolished the effects of CDKN2B‐AS1‐silencing on EC cell function. CDKN2B‐AS1 silencing curtailed tumorigenesis in nude mice, which was nullified by the upregulation of TFAP2A or FSCN1. Our findings demonstrated the antioncogenic effects of silencing CDKN2B‐AS1 in EC through inactivation of the TFAP2A/FSCN1 axis. [ABSTRACT FROM AUTHOR]

Objective This study aimed to investigate the effect of transfecting SOX2-shRNA vector lentivirus to SACC cell lines on the biological behavior of salivary adenoid cystic carcinoma (SACC)-LM and SACC-83. Methods Three types of SOX2-shRNA lentiviral vectors (817, 818, and 819) were constructed and transfected successfully. The shRNA with the best inhibitory effect was screened out and transfected into SACC cells. Real-time fluorescence quantitative polymerase chain reaction (RT-qPCR) and Western blot were used to detect the expressions of Survivin, E-cadherin, and N-cadherin of SACC-LM and SACC-83. CCK-8 and flow cytometry were used to detect SACC-LM and SACC-83 proliferation and apoptosis. Cell scratch test and Transwell method were used to detect the migration and invasion capabilities of SACC-LM and SACC-83. Results SOX2-shRNA-819 had the best interference effect among the three lentiviruses. After transfecting SOX2-shRNA-819 into SACC-LM and SACC-83, the expressions of SOX2, Survivin, and N-cadherin were significantly reduced, and that of E-cadherin was significantly increased (P<0.05). The cell proliferation ability decreased, and the number of apoptotic cells increased (P<0.05). The cell migration and invasion ability decreased (P<0.05). Conclusion shRNA interference technology reduced SOX2 expression while downregulating Survivin expression. These two expressions may be related. Low SOX2 expression inhibits the proliferation, migration, and invasion of SACC cells and promotes the apoptosis of SACC cells. SOX2 may be involved in the epithelial-mesenchymal transition process. Homogeneous copper incorporated nickel sulfide (NS) nanosheets were realized via an in situ ion exchange method and one-pot hydrothermal process. Nickel foam (NF) and copper foam (CF) were used as metal sources for the co-formation of Cu1.8S–Ni3S2 in the presence of thiourea. As a result of the Cu incorporation, the NF-supported Ni3S2 nanosheets exhibited improved electronic conductivity with higher electrochemical surface area (ECSA). When evaluated as electrode materials for supercapacitors, the as-obtained active materials demonstrated high specific capacitance (1686 F g−1 at 1 A g−1) with excellent cycling performance (95.39% capacitive retention after 10 000 cycles), which may be attributed to the unique structural features as well as the synergistic effect of localized copper species with Ni3S2 nanosheets. An image restoration method based on noise gray scale difference estimation is proposed. In order to ensure the quality of restored punctate image, the image is convoluted respectively by various directional uniform arithmetic operators, and the results from convolution are subtracted respectively from the original images. The estimation image of noise gray scale difference is constructed by extracting the pixels with minima of gray scale differences. Then, the intensity and the position of the spot in the image can be got, and the spots are smoothed by neighboring pixels. Test results show that noise gray scale difference estimation can express exactly intensity and position of the spots in the punctate image with small operation amount needed. The proposed method has been used experimentally to restore three dimension biological microscopic image, and its results show that the lighteness of spots is controlled efficiently and small spots are wiped off in restored image. Objective: To study the expression of tyrosine kinase receptor 2 (Tie2) in oral squamous cell carcinoma (OSCC) and its effect on cell proliferation and migration and the epithelial-mesenchymal transition (EMT) process., Methods: Immunohistochemistry (IHC) tests were conducted to examine the expression of Tie2 in OSCC tissues and normal oral mucosa tissues. Western blot was performed to examine the expression of Tie2 in dysplastic oral keratinocyte (DOK) cell line and OSCC cell lines, and the cell line with high Tie2 expression was selected as the experimental cell line. The Tie 2-silenced lentiviral vector was successfully transfected onto the experimental cell line for subsequent experiments. Cell proliferation and cloning abilities were examined with CCK-8 and clone formation assays. Cell migration ability was examined with scratch and Transwell assays. The remodeling ability of cytoskeletal F-actin and the expressions of E-cadherin and N-cadherin were examined with confocal laser scanning microscope. Western blot was performed to examine the expression of EMT-related signature proteins, including E-cadherin, N-cadherin, and vimentin, and the expression of the protein kinase B (AKT) and extracellular signal-regulated kinase (ERK)., Results: IHC results showed that the Tie2-positive rate of the OSCC group (74.5%) was significantly higher than that of the control group (19.4%) ( P <0.0001). The expression of Tie2 was higher in HSC-4 and SCC-9 cell lines compared to that in DOK cells. The lentiviral shRNA-162 group showed the best silencing effect, which was used as the experimental group and applied in subsequent experiments. Compared with those of the control group, the proliferation, cloning and migration capacities of the cells of the experimental group were significantly reduced. Furthermore, the green fluorescence intensity of the cytoskeleton F-actin was reduced, the number of filamentous pseudopods at the leading edge of the cells decreased and their length was shortened, and the expression of E-cadherin was significantly increased, while the expression of N-cadherin and vimentin was significantly reduced in the experimental group in comparison with those of the control group. The expression of p-AKT and p-ERK proteins decreased, while AKT and ERK protein expression increased., Conclusion: Tie2 was highly expressed in most OSCC cells. Silencing Tie2 can inhibit the proliferation, cloning, and migration ability of OSCC cells, inhibit F-actin remodeling, and alter the expression of its EMT-related signature proteins by regulating AKT and ERK signaling pathway, which suggests that Tie2 may be involved in the growth, metastasis and EMT process of OSCC., (Copyright© by Editorial Board of Journal of Sichuan University (Medical Sciences).)

Three-dimensional (3D) nanostructures are commonly endowed with numerous active sites, large specific surface area, and good mechanical strength, which make them as an efficient candidate for energy storage and conversion. Herein, by considering the advantages of 3D nanostructures, we successfully fabricated carbon-coated nickel sulfide on a nickel foam (C@NS@NF) with a unique 3D treelike superstructure via a two-step hydrothermal process. By virtue of its hierarchical superstructures, 3D treelike architecture, and carbon shell encapsulation, the as-fabricated carbon-coated Ni 3 S 2 can be directly served as binder-free bifunctional electrodes for supercapacitor and hydrogen evolution reaction, where high specific areal capacitance (6.086 F cm -2 at 10 mA cm -2 ) for supercapacitors and low overpotential (92 mV at 10 mA cm -2 ) for the electrocatalyst have been demonstrated. These inspiring results of this material make it as a potential candidate for energy storage and conversion.
