The pathogenesis of choroid plexus papillomas, intraventricular papillary neoplasms most often occurring sporadically in children and young adults, remains poorly understood. To identify pathways operative in the development of choroid plexus papillomas, gene expression profiles obtained from laser-microdissected human choroid plexus papilloma cells (n = 7) were compared with that of normal choroid plexus epithelial cells laser microdissected from autopsy tissue (n = 8). On DNA microarray data analysis, 53 probe sets were differentially expressed in choroid plexus papilloma tumor cells (>7-fold). Up-regulation of TWIST1, WIF1, TRPM3, BCLAF1, and AJAP1, as well as down-regulation of IL6ST was confirmed using quantitative reverse transcription-PCR. Knockdown of Twist1 gene expression in the rat choroid plexus epithelial cell line Z310 significantly reduced proliferation as assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and cell invasion in a Matrigel assay, whereas cell migration was not affected. Screening for expressional changes of cancer-related genes upon Twist1 knockdown revealed up-regulation of Cdkn1a, Cflar, and Serpinb2 and down-regulation of Figf. To conclude, using gene expression profiling, several genes differentially expressed in human choroid plexus papillomas could be identified. Among those, TWIST1 is highly expressed in choroid plexus papillomas and promotes proliferation and invasion. [Cancer Res 2009;69(6):2219–23]

International audience; INTRODUCTION: Organic compounds in asteroids and comets contain information about the early history of the Solar System. They could also have delivered organic material to early Earth. The Hayabusa2 spacecraft visited the carbonaceous asteroid Ryugu and collected samples of its surface materials, which were brought to Earth in December 2020. RATIONALE: We investigated the macromolecular organic matter in the Ryugu samples, measuring its elemental, isotopic, and functional group compositions along with its small-scale structures and morphologies. Analytical methods used included spectro-microscopies, electron microscopy, and isotopic microscopy. We examined intact Ryugu grains and insoluble carbonaceous residues isolated by acid treatment of the Ryugu samples. RESULTS: Organic matter is abundant in the Ryugu grains, distributed as submicrometersized organic grains and as organic matter dispersed in matrix. Context. After landing on C-type asteroid Ryugu, MASCOT imaged brightly colored, submillimeter-sized inclusions in a small rock. Hayabusa2 successfully returned a sample of small particles from the surface of Ryugu, but none of these appear to harbor such inclusions. The samples are considered representative of Ryugu. Aims. To understand the apparent discrepancy between MASCOT observations and Ryugu samples, we assess whether the MASCOT landing site, and the rock by implication, is perhaps atypical for Ryugu. Methods. We analyzed observations of the MASCOT landing area acquired by three instruments on board Hayabusa2: a camera (ONC), a near-infrared spectrometer (NIRS3), and a thermal infrared imager. We compared the landing area properties thus retrieved with those of the average Ryugu surface. Results. We selected several areas and landforms in the landing area for analysis: a small crater, a collection of smooth rocks, and the landing site itself. The crater is relatively blue and the rocks are relatively red. The spectral and thermophysical properties of the landing site are very close to those of the average Ryugu surface. The spectral properties of the MASCOT rock are probably close to average, but its thermal inertia may be somewhat higher. Conclusions. The MASCOT rock can also be considered representative of Ryugu. Some of the submillimeter-sized particles in the returned samples stand out because of their atypical spectral properties. Such particles may be present as inclusions in the MASCOT rock.

Perseverance’s Mastcam-Z instrument provides high-resolution stereo and multispectral images with a unique combination of spatial resolution, spatial coverage, and wavelength coverage along the rover’s traverse in Jezero crater, Mars. Images reveal rocks consistent with an igneous (including volcanic and/or volcaniclastic) and/or impactite origin and limited aqueous alteration, including polygonally fractured rocks with weathered coatings; massive boulder-forming bedrock consisting of mafic silicates, ferric oxides, and/or iron-bearing alteration minerals; and coarsely layered outcrops dominated by olivine. Pyroxene dominates the iron-bearing mineralogy in the fine-grained regolith, while olivine dominates the coarse-grained regolith. Solar and atmospheric imaging observations show significant intra- and intersol variations in dust optical depth and water ice clouds, as well as unique examples of boundary layer vortex action from both natural (dust devil) and Ingenuity helicopter–induced dust lifting. High-resolution stereo imaging also provides geologic context for rover operations, other instrument observations, and sample selection, characterization, and confirmation.

This paper describes the development of a SBHC with an innovative model of care that grew out of a partnership between a public-school district and a university nursing programme in the midwestern region of the United States.Persistent barriers to health and health care experienced by youth are well documented. School-based health centres (SBHCs) can improve educational and health outcomes, positively impacting health equity. Academic systems are positioned to address health care needs of the school-aged population, yet educators face challenges of accessing quality learning placements for students and faculty practice sites.A community-based collaborative methodology guided the planning phases that were driven by priority needs identified by families and stakeholders. With the mission of "partnering with students, families, and communities in the promotion of health and wellness through engagement in practice, education, and research," an ongoing dialogue over a two-year period led to articulating a vision, designing a plan and implementing a nurse-managed SBHC. The Standards for Reporting Qualitative Research (SRQR) checklist was considered in the preparation of this paper.In three years, this SBHC has addressed and identified priority needs and served individual youth and families. The SBHC provides opportunities for the faculty to fulfil a practice requirement for certification and accreditation. Nursing students engage with youth and families in health education and health promotion while strengthening their technical and relational skills. Family nurse practitioner students gain valuable clinical experience. Faculty with expertise in family nursing guide family assessments, support family resiliency and direct therapeutic conversations with family units.SBHCs serve youth, families, and community. This academic-practice partnership has the added benefit of providing faculty practice opportunities and nursing student experiential learning.SHBCs provide practice opportunities that address needs in individuals, families, and communities. Partnerships should be considered at academic nursing programmes to support their needs and fulfil commitments to address health equity gaps.

Polygonal patterned ground is ubiquitous in the martian mid-latitudes and in the polar regions of Earth. The latitude dependence of martian patterned ground and its morphological similarity to terrestrial patterned ground suggests that thermal contraction cracking may have been the leading formation mechanism for those polygons. Due to a lack of ground truthing on martian patterned ground, the role of liquid water in its formation and weather freeze-thaw processes lead to their origin is still debated. This study uses a quantitative approach, based on geomorphometrical and soil characteristics of patterned ground in continental Antarctica and glacial deposits with low inclination of Terra Nova Bay as terrestrial analogues, to understand polygon formation in martian hyper-arid conditions. We investigated polygons in ice-free parts of the mountain range of Helliwell Hills (~71°43S / 161°2E) in continental Antarctica and the Northern Foothills in the coastal Terra Nova Bay area (74°45S / 164°E).Field observations were made during the austral summer on the GANOVEX XI and GANOVEX XIII expeditions in Dec-Jan 2015/2016 and Oct-Nov 2018, respectively. The polygonal troughs have been mapped and digitized in ArcGIS based on high resolution satellite images. For Helliwell Hills we used World View 2 images with a pixel size of 50 cm. For Terra Nova Bay, Quickbird satellite imagery has been used with a pixel size of 60 cm. Based on these datasets, parameters such as area, perimeter, length, and width have been measured, and size, circularity, orientation, and aspect ratio of each polygon were derived from these measurements. Additionally, we used a DTM derived from World View 2 stereo imagery (ground sampling distance: 8 m) to calculate the average slope, aspect, and solar irradiation of each polygon. The quantitative analysis shows that the geomorphometric characteristics of polygons in the Helliwell Hills differ significantly from those in Terra Nova Bay. Polygons in the Helliwell Hills are significantly smaller than in Terra Nova Bay and are organized as orthogonal, random-orthogonal and hexagonal polygon networks, while all polygons in Terra Nova Bay form hexagonal polygon-net geometries. The correlation of polygon-net geometries and the slope gradient shows that hexagonal polygon-net geometries dominate in flat terrains, while orthogonal geometries have developed on steeper slopes or in the immediate proximity of sharp terrain margins such as topographic slopes. The polygons in Helliwell Hills do not display significant local relief, but overall, the polygon centres are slightly higher than the bounding cracks (i.e. high-centered polygons). In Terra Nova Bay the appearance of high centred polygons and a deeper trough is even more developed and well distinguishable on satellite images.During the fieldwork in Helliwell Hills, excavations were made in the center of polygons and across the bounding cracks. Typically, the uppermost ∼40 cm of regolith are dry and unconsolidated. Below that, there is commonly a sharp transition to ice-cemented material or very clear ice with no bubbles. The grain size analysis indicated no significant trend of sorting. We will present the results of our analysis and compare them with selected polygon sites on Mars.

An expedition EMM-Etna to simulate the Lunar and Martian volcanic and soil environment will be carried out at Mount.Etna’s Cratere del Laghetto in Sicily, near Catania Italy by the EuroMoonMars TUDublin and LEAPS ExoMars groups. This scouting campaign intends to train in using instruments to be used on MoonMars landers and rovers, with a perspective of ARCHES DLR telerobotics campaign to be conducted in June 2022, and in preparation for ExoMars rover instruments (PANCAM, CLUPI and spectrometers) science and operations. Figure 1: Lunar Lander and REMMI Rover for Sample Analysis The aim of this EMM-Etna expedition is to investigate and analyse the terrain with the use of different scientific instruments. The topography of the landscape will be photographed using a 360° panoramic camera and drone; it will be processed, and a 3D model developed. The terrain will also be investigated using the REMMI Rover, the abilities of the rover to operate and transport equipment will be monitored. This will further develop the knowledge available of the terrain and help future expeditions to identify different landmarks. The use of a Radio Jove Antenna will permit the team to monitor transmissions from both the Sun and Jupiter. This will allow different cosmic events or changes in the celestial objects to be studied and explored. On site a selection of different samples will also be collected and examined using the REMMI Rover. An Ocean Optics UV-Vis-NIR spectrometer will a be operated in order to evaluate the existence of biological compounds and substances within these samples and in the area itself. It is key to understand the molecular makeup of one’s surroundings when in an unknown environment. By analysing samples collected, spectroscopy can be used to identify and determine a diagnostic for each substance. This process will be monitored by a Logitech camera to ensure it is carried put correctly. A selection of photographs will be captured of each sample using a portable optical microscope. This will allow an in-depth analysis of the microscopic structure of each collected sample. The use of all of the instruments mentioned above is key in the investigation and research into the Moon and Martian-like volcanic environment that is Mount Etna. We would also like to thank Prof I. Alteration processes on asteroid and comet surfaces, such as thermal fracturing, (micrometeorite) impacts or volatile outgassing, are complex mechanisms that form diverse surface morphologies and roughness on various scales. These mechanisms and their interaction may differ on the surfaces of different bodies. Asteroid Ryugu and comet 67P/Churyumov–Gerasimenko, both, have been visited by landers that imaged the surfaces in high spatial resolution. We investigate the surface morphology and roughness of Ryugu and 67P/Churyumov–Gerasimenko based on high-resolution in situ images of 0.2 and 0.8 mm pixel resolution over an approximately 25 and 80 cm wide scene, respectively. To maintain comparability and reproducibility, we introduce a method to extract surface roughness descriptors (fractal dimension, Hurst exponent, joint roughness coefficient, root-mean-square slope, hemispherical crater density, small-scale roughness parameter, and Hapke mean slope angle) from in situ planetary images illuminated by LEDs. We validate our method and choose adequate parameters for an analysis of the roughness of the surfaces. We also derive the roughness descriptors from 3D shape models of Ryugu and orbiter camera images and show that the higher spatially resolved images result in a higher roughness. We find that 67P/Churyumov–Gerasimenko is up to 6 per cent rougher than Ryugu depending on the descriptor used and attribute this difference to the different intrinsic properties of the materials imaged and the erosive processes altering them. On 67P/Churyumov–Gerasimenko sublimation appears to be the main cause for roughness, while on Ryugu micrometeoroid bombardment as well as thermal fatigue and solar weathering may play a significant role in shaping the surface.

Stack, K. et al., The Mars 2020 Perseverance rover landing site is located within Jezero crater, a ∼50km diameter impact crater interpreted to be a Noachian-aged lake basin inside the western edge of the Isidis impact structure. Jezero hosts remnants of a fluvial delta, inlet and outlet valleys, and infill deposits containing diverse carbonate, mafic, and hydrated minerals. Prior to the launch of the Mars 2020 mission, members of the Science Team collaborated to produce a photogeologic map of the Perseverance landing site in Jezero crater. Mapping was performed at a 1:5000 digital map scale using a 25 cm/pixel High Resolution Imaging Science Experiment (HiRISE) orthoimage mosaic base map and a 1 m/pixel HiRISE stereo digital terrain model. Mapped bedrock and surficial units were distinguished by differences in relative brightness, tone, topography, surface texture, and apparent roughness. Mapped bedrock units are generally consistent with those identified in previously published mapping efforts, but this study’s map includes the distribution of surficial deposits and sub-units of the Jezero delta at a higher level of detail than previous studies. This study considers four possible unit correlations to explain the relative age relationships of major units within the map area. Unit correlations include previously published interpretations as well as those that consider more complex interfingering relationships and alternative relative age relationships. The photogeologic map presented here is the foundation for scientific hypothesis development and strategic planning for Perseverance’s exploration of Jezero crater., With funding from the Spanish government through the "María de Maeztu Unit of Excellence" accreditation (MDM-2017-0737)

The ESA-Russia Rosalind Franklin (ExoMars 2022) rover, with its unique 2m drill, will address key questions in the search for life on Mars. PanCam will establish the surface context, with other instruments, providing geology, atmospheric science and 3D vision. PanCam uses a pair of Wide Angle Cameras (WACs), each with an 11 position filter wheel, and a High Resolution Camera (HRC). The cameras and electronics are in an optical bench that provides the interface to the rover and planetary protection. PanCam includes a calibration target mounted on the rover deck for radiometric calibration, fiducial markers for geometric calibration and a rover inspection mirror.

The ExoMars mission will deploy a stationary surface platform and a rover in Oxia Planum (OP), a region at the transition between the heavily cratered highlands of Mars and the ancient and filled impact basin, Chryse Planitia. While the fundamental geologic characteristics of the area have been investigated during the landing site selection process, detailed geologic or morpho-stratigraphic mapping is still missing. To fill this knowledge gap, two complementary mapping approaches were initiated by the ExoMars RSOWG: (1) Local HiRISE-scale mapping of the landing ellipse(s) area (reported elsewhere: Sefton-Nash et al., LPSC 2020). (2) Regional mapping at ~CTX-scale [this study] will provide a more synoptic view of the wider landing site within OP, enabling the contextualization of the units within the stratigraphy of western Arabia Terra and Chryse Planitia, and a comparison to other sites with similar key geologic and physiographic characteristics. It is also expected that this map will serve as a geologic reference throughout the mission and subsequent data analysis. The study area is located between 16.5°N and 19.5°N, and 334°E to 338°E. The data sets used for mapping include HRSC, THEMIS IR (day and night), CTX, and CaSSIS. Mapping scale in a GIS environment is 1:100,000, which will result in a final printable map at a scale of 1:1M. Mapping started in mid-October 2019. Overall, the identified map units are very similar to those described by Quantin et al. (Astrobiology, submitted): The spatially most widespread units are the phyllosilicate-bearing unit that is the prime ExoMars target (with distinctly enhanced THEMIS nighttime temperatures when compared to its surroundings), a dark resistant unit of possibly volcanic or sedimentary origin, and a mantling unit that was likely emplaced by eolian processes. Multiple channels of various morphology and degradation state as well as sedimentary fan-shaped deposits (with low nighttime temperatures) imply a diverse and possibly long-lived history of surface runoff, perhaps accompanied or replaced by groundwater processes such as sapping. Inverted landforms (channels, impact craters) are the result of intense erosion. Additional mapped features include tectonic structures such as wrinkle ridges and lobate scarps (delineating a basin-like depression in the central mapping area), remnant erosional buttes in the northwestern portion of the mapping area (i.e. towards Chryse Planitia), craters and their ejecta blankets, and fields of eolian bedforms and secondary craters. At the time of writing, the mapping is incomplete and only initial and limited conclusions can be drawn. Overall, the mapping confirms previous geologic analyses. However, some features (e.g., contractional structures, channels, possible sapping landforms) need further attention as the may provide important constraints on the tectonic and aqueous evolution of the ExoMars landing area. A comparison to a distant, but geologically very similar site in Xanthe Terra, southeast of the Hypanis fan-shaped deposits, may enable testing of hypotheses raised by the geologic mapping of OP.

Although the present environmental conditions on Mars prohibit the generation of significant volumes of liquid water, observations of several very young landforms, such as gullies and recurrent slope lineae, have been interpreted as signals for aqueous processes. To explore the range of conditions under which such features can be formed on Earth, a field site in northern Victoria Land, East Antarctica, was geomorphologically investigated. Despite the small size of the icefree area, the site displays gullies, water tracks and other traces of liquid water. The gullies show clear evidence of sediment transport by debris flows, and are typical of paraglacial processes on steep slopes in a recently deglaciated area. Water tracks appear in different forms, and seem to recur seasonally in the austral summer. Melting of snow and surface glacier ice is the major water source for both debris flows and water tracks. The observations presented here highlight the potential for hyperarid polar deserts to generate morphogenetically significant amounts of meltwater. The gullies are morphologically analogous to Martian gullies, and water tracks on steep slopes appear very similar to recurrent slope lineae. The observations suggest that even small icefree sites in continental Antarctica may enable observations which can serve as a basis for working hypotheses in Mars analogue studies, and future field work should consider more areas in Antarctica in addition to the McMurdo Dry Valleys to search for Mars analogue landforms.

After 3.5 years of cruise, and about 3 months in the vicinity of its target, the MASCOT lander was deployed successfully on October 3, 2018 by the Hayabusa2 spacecraft onto the C-type near-Earth asteroid (162173) Ryugu. After a free-fall of 5 ​min 51 ​s from an altitude of 41 ​m MASCOT experienced its first contact with the asteroid hitting a big boulder. The lander bounced for ~11 ​min 3 ​s before it came to rest. MASCOT was able to perform science measurements with its payload suite at 3 different locations on the surface of Ryugu. It investigated the fine-scale structure, multispectral reflectance, thermal characteristics and magnetic properties. The surface consists of very rugged terrain littered with large surface boulders. The in-situ measurements confirmed the absence of fine particles and dust as already implied by the remote sensing instruments aboard the Hayabusa2 spacecraft. After about 17 ​h of operations, the MASCOT mission terminated with the last communication contact as its primary batteries depleted. This paper summarizes the MASCOT mission covering its four years of in-flight operations, its preparation for the descent, landing and in-situ investigation on the asteroid Ryugu until the end of its operation.

20 years after the successful ground deployment test of a (20 m)2 solar sail at DLR Cologne, and in the light of the upcoming U.S. NEAscout mission, we provide an overview of the progress made since in our mission and hardware design studies as well as the hardware built in the course of our solar sail technology development. We outline the most likely and most efficient routes to develop solar sails for useful missions in science and applications, based on our developed ‘now-term’ and near-term hardware as well as the many practical and managerial lessons learned from the DLR-ESTEC Gossamer Roadmap. Mission types directly applicable to planetary defense include single and Multiple NEA Rendezvous ((M)NR) for precursor, monitoring and follow-up scenarios as well as sail-propelled head-on retrograde kinetic impactors (RKI) for mitigation. Other mission types such as the Displaced L1 (DL1) space weather advance warning and monitoring or Solar Polar Orbiter (SPO) types demonstrate the capability of near-term solar sails to achieve asteroid rendezvous in any kind of orbit, from Earth-coorbital to extremely inclined and even retrograde orbits. Some of these mission types such as SPO, (M)NR and RKI include separable payloads. For one-way access to the asteroid surface, nanolanders like MASCOT are an ideal match for solar sails in micro-spacecraft format, i.e. in launch configurations compatible with ESPA and ASAP secondary payload platforms. Larger landers similar to the JAXA-DLR study of a Jupiter Trojan asteroid lander for the OKEANOS mission can shuttle from the sail to the asteroids visited and enable multiple NEA sample-return missions. The high impact velocities and re-try capability achieved by the RKI mission type on a final orbit identical to the target asteroid's but retrograde to its motion enables small spacecraft size impactors to carry sufficient kinetic energy for deflection.

After its release and a descent and bouncing phase, the Hayabusa2 lander MASCOT came to a final rest and MASCOT’s camera MASCam acquired a set of images of the surface of Ryugu. With MASCam’s instantaneous field of view of about 1 mrad, the images provide pixel scales from 0.2 to 0.5 mm pixel−1in the foreground and up to 1 cm pixel−1for surface parts in the background. Using a stereo-photogrammetric analysis of the MASCam images taken from slightly different positions due to commanded and unintentional movements of the MASCOT lander, we were able to determine the orientation for the different measurement positions. Furthermore, we derived a 3D surface model of MASCOT’s vicinity. Although the conditions for 3D stereo processing were poor due to very small stereo angles, the derived 3D model has about 0.5 cm accuracy in the foreground at 20 cm distance and about 1.5 cm at a distance of 40–50 cm.

Any effort which intends to physically interact with specific asteroids requires understanding at least of the composition and multi-scale structure of the surface layers, sometimes also of the interior. Therefore, it is necessary first to characterize each target object sufficiently by a precursor mission to design the mission which then interacts with the object. In small solar system body (SSSB) science missions, this trend towards landing and sample-return missions is most apparent. It also has led to much interest in MASCOT-like landing modules and instrument carriers. They integrate at the instrument level to their mothership and by their size are compatible even with small interplanetary missions.\ud \ud The DLR-ESTEC Gossamer Roadmap NEA Science Working Groups‘ studies identified Multiple NEA Rendezvous (MNR) as one of the space science missions only feasible with solar sail propulsion. Parallel studies of Solar Polar Orbiter (SPO) and Displaced L1 (DL1) space weather early warning missions studies outlined very lightweight sailcraft and the use of separable payload modules for operations close to Earth as well as the ability to access any inclination and a wide range of heliocentric distances.\ud \ud These and many other studies outline the unique capability of solar sails to provide access to all SSSB, at least within the orbit of Jupiter. Since the original MNR study, significant progress has been made to explore the performance envelope of near-term solar sails for multiple NEA rendezvous.\ud \ud However, although it is comparatively easy for solar sails to reach and rendezvous with objects in any inclination and in the complete range of semi-major axis and eccentricity relevant to NEOs and PHOs, it remains notoriously difficult for sailcraft to interact physically with a SSSB target object as e.g. the Hayabusa missions do.\ud \ud The German Aerospace Center, DLR, recently brought the Gossamer solar sail deployment technology to qualification status in the Gossamer-1 project. Development of closely related technologies is continued for very large deployable membrane-based photovoltaic arrays in the GoSolAr project.\ud \ud We expand the philosophy of the Gossamer solar sail concept of efficient multiple sub-spacecraft integration to also include landers for one-way in-situ investigations and sample-return missions. These are equally useful for planetary defence scenarios, SSSB science and NEO utilization. We outline the technological concept used to complete such missions and the synergetic integration and operation of sail and lander.\ud \ud We similarly extend the philosophy of MASCOT and use its characteristic features as well as the concept of Constraints-Driven Engineering for a wider range of operations.

Images from the Optical Navigation Camera system (ONC) onboard the Hayabusa2 spacecraft show the MASCOT lander during its descent to the surface of asteroid (162173) Ryugu. We used results from a previous stereo-photogrammetric analysis that provided precise ONC image orientation data (camera position and pointing), ONC orthoimages, and an ONC-based 3D surface model to combine them with the visibilities of MASCOT itself and its shadow on-ground within the ONC images. We integrated additional information from instruments onboard MASCOT (MASMag, MARA, MASCam) and derived MASCOT’s release position and modeled its free-fall descent path and its velocity over 350 s from its release at ∼41 m altitude above ground until its first contact with the surface of Ryugu. After first contact, MASCOT bounced over the surface of Ryugu for 663 s and came to rest at its first settlement point after four intermediate surface contacts. We again used ONC images that show MASCOT and partly its shadow and reconstructed the bouncing path and the respective velocities of MASCOT. The achieved accuracy for the entire descent and bouncing path is ∼0.1 m (1σ).

We analyze images of a rock on Ryugu acquired in situ by MASCam, the camera on the mascot lander, with the aim of identifying possible carbonaceous chondrite (CC) analogs. The rock’s reflectance ( r F = 0.034 ± 0.003 at phase angle 4.5 ∘ ± 0.1 ∘ ) is consistent with Ryugu’s average reflectance, suggesting that the rock is typical for this asteroid. A spectrophotometric analysis of the rock’s inclusions provides clues to its CC group membership. Inclusions are generally brighter than the matrix. The dominant variation in their color is a change in the visible spectral slope, with many inclusions being either red or blue. Spectral variation in the red channel hints at the presence of the 0.7 μm absorption band linked to hydrated phyllosilicates. The inclusions are unusually large for a CC; we find that their size distribution may best match that of the Renazzo (CR2) and Leoville (CV3) meteorites. The Ryugu rock does not easily fit into any of the CC groups, consistent with the idea that typical Ryugu-type meteorites are too fragile to survive atmospheric entry.

MASCOT (‘Mobile Asteroid Surface Scout’) is a 10 kg mobile surface science package on board JAXA’s Hayabusa2 sample return mission, currently on its way to the near-Earth asteroid (162173) Ryugu. The mission was launched in December 2014 from Tanegashima Space Center, Japan. The Hayabusa2 spacecraft will reach the target asteroid in summer 2018. Hayabusa2 will return its samples to Earth in December 2020. After arrival at the target asteroid ‘Ryugu’ a detailed mapping phase will be performed and the landing site of MASCOT will be selected. The deployment of MASCOT to the asteroids surface is planned for the beginning of October 2018. MASCOT has been developed by the German Aerospace Center (DLR) in cooperation with the Centre National d’Etudes Spatiales (CNES). The main objective of MASCOT is to perform in-situ investigations of the asteroid surface and to support the sampling site selection for the mother spacecraft. Mascot is equipped with four scientific instruments a wide angle camera, a hyperspectral infrared microscope, a radiometer and a magnetometer. The camera (MasCam) provides ground truth for the orbital measurements of the Hayabusa2 orbiter instruments and the in-situ MASCOT sensor suite as well as geological context of the samples. The radiometer (MARA) determines the surface brightness temperature, the thermal inertia of the surface material and the spectral slope in infrared. The radiometer field of view is correlated with the wide angle camera field of view. The magnetometer (MasMAG) observes the magnetic field profile during descent and bouncing and determines any global and local magnetization of the asteroid.

Sphingosine‐1‐phosphate receptor 1 (S1P1) modulators sequester circulating lymphocytes within lymph nodes, thereby preventing potentially pathogenic autoimmune cells from exiting into the blood stream and reaching inflamed tissues. S1P1 receptor modulation may thus offer potential to treat various autoimmune diseases. The first nonselective S1P1‐5 receptor modulator FTY720/fingolimod/Gilenya® has successfully demonstrated clinical efficacy in relapsing forms of multiple sclerosis. However, cardiovascular, hepatic, and respiratory side‐effects were reported and there is a need for novel S1P1 receptor modulators with better safety profiles. Here, we describe the discovery of cenerimod, a novel, potent and selective S1P1 receptor modulator with unique S1P1 receptor signaling properties and absence of broncho‐ and vasoconstrictor effects ex vivo and in vivo. Cenerimod dose‐dependently lowered circulating lymphocyte counts in rats and mice after oral administration and effectively attenuated disease parameters in a mouse experimental autoimmune encephalitis (EAE) model. Cenerimod has potential as novel therapy with improved safety profile for autoimmune diseases with high unmet medical need.

ISEM (Infrared Spectrometer for ExoMars) is a pencil-beam infrared spectrometer that will measure reflected solar radiation in the near infrared range for context assessment of the surface mineralogy in the vicinity of the ExoMars rover. The instrument will be accommodated on the mast of the rover and will be operated together with the panoramic camera (PanCam), high-resolution camera (HRC). ISEM will study the mineralogical and petrographic composition of the martian surface in the vicinity of the rover, and in combination with the other remote sensing instruments, it will aid in the selection of potential targets for close-up investigations and drilling sites. Of particular scientific interest are water-bearing minerals, such as phyllosilicates, sulfates, carbonates, and minerals indicative of astrobiological potential, such as borates, nitrates, and ammonium-bearing minerals. The instrument has an ∼1° field of view and covers the spectral range between 1.15 and 3.30 μm with a spectral resolution varying from 3.3 nm at 1.15 μm to 28 nm at 3.30 μm. The ISEM optical head is mounted on the mast, and its electronics box is located inside the rover's body. The spectrometer uses an acousto-optic tunable filter and a Peltier-cooled InAs detector. The mass of ISEM is 1.74 kg, including the electronics and harness. The science objectives of the experiment, the instrument design, and operational scenarios are described. Objective Novel biomarkers for monitoring progression in neurodegenerative conditions are needed. Measurement of microstructural changes in white matter (WM) using diffusion tensor imaging (DTI) may be a useful outcome measure. Here we report trajectories of WM change using serial DTI in a cohort with behavioral variant frontotemporal dementia (bvFTD). Methods Twenty-three patients with bvFTD (12 having genetic mutations), and 18 age-matched control participants were assessed using DTI and neuropsychological batteries at baseline and ∼1.3 years later. Baseline and follow-up DTI scans were registered using a groupwise approach. Annualized rates of change for DTI metrics, neuropsychological measures, and whole brain volume were calculated. DTI metric performances were compared, and sample sizes for potential clinical trials were calculated. OX40, a member of the TNF receptor superfamily, is expressed on activated T cells and implicated in stimulation of T cells and T-dependent humoral responses. We generated OX40 −/− mice and found that the formation of extrafollicular plasma cells, germinal centers, and antibody responses was independent of OX40. After infection with LCMV and influenza virus, OX40 −/− mice retain primary and memory cytotoxic T cell responses with normal expansion and decline of specific CTL. In contrast, CD4 + T cell proliferation and the number of IFN-γ-producing CD4 + T cells were reduced in OX40 −/− mice. Moreover, the number of CD4 + T cells infiltrating the lungs of influenza virus–infected OX40 −/− mice was reduced. These results define a unique role of OX40 in the generation of optimal CD4 + T cell responses in vivo.

Intranasal (i.n.) immunization aims to induce local as well as systemic immune responses. In the present study, we assessed a vaccine platform based on virus-like particles (VLP) derived from the RNA phage Qbeta for i.n. immunization. We found that both i.n. and subcutaneous (s.c.) administration of Qbeta-VLP elicited strong and comparable specific IgG responses in serum and lung. Surprisingly, both routes also induced high levels of specific IgA in serum. In contrast, only i.n. administration of Qbeta-VLP resulted in local IgA production in the lung. Efficient induction of B cell responses by i.n. administration of VLP was further supported by the presence of large numbers of germinal centers (GC) as well as memory B cells in the spleen and plasma cells in the bone marrow. Results obtained for the VLP itself could be extended to an antigen covalently attached to it. Specifically, i.n. immunization of mice with VLP displaying the influenza virus derived ectodomain of the M2 protein resulted in strong M2-specific antibody responses as well as anti-viral protection. In contrast, i.n. immunization with VLP displaying p33 peptide, the major CTL epitope of lymphocytic choriomeningitis virus, induced relatively inefficient cytotoxic T cell responses, resulting in low numbers of specific T cells and poor effector cell differentiation. Taken together, these results suggest that effective antibody-based vaccines are achievable by i.n. administration of Qbeta-VLP displaying specific antigens.

Suppression by natural CD4+CD25+regulatory T cells (Tregs) is one mechanism by which tolerance is maintained. However, the way in which Tregs mediate suppression is not well understood. Here, we show that secreted phospholipase A2 (sPLA2)-IID is selectively produced by Tregs. sPLA2-IID is a potent mediator of Treg function, because it strongly suppressed proliferation of CD4+and CD8+T cells in vitro and in vivo in a manner independent of its catalytic activity. Furthermore, sPLA2-IID promoted the differentiation of Tregs, presumably via attenuating signaling through the PI3K/Akt/mammalian target of rapamycin pathway. Importantly, administration of a sPLA2-IID-Fc fusion protein inhibited disease development in murine models of colitis and multiple sclerosis, suggesting that sPLA2-IID's immunosuppressive function might be exploited therapeutically.

Background: 22q11.2 deletion syndrome (22q11DS) is caused by a microdeletion on chromosome 22q11.2 and associated with an increased risk to develop psychosis. The gene coding for catechol-O-methyl-transferase (COMT) is located at the deleted region, resulting in disrupted dopaminergic neurotransmission in 22q11DS, which may contribute to the increased vulnerability for psychosis. A dysfunctional motivational reward system is considered one of the salient features in psychosis and thought to be related to abnormal dopaminergic neurotransmission. The functional anatomy of the brain reward circuitry has not yet been investigated in 22q11DS. Methods: This study aims to investigate neural activity during anticipation of reward and loss in adult patients with 22q11DS. We measured blood-oxygen-level dependent (BOLD) activity in 16 patients with 22q11DS and 12 healthy controls during a monetary incentive delay task using a 3T Philips Intera MRI system. Data were analysed using SPM8. Results: During anticipation of reward, the 22q11DS group alone displayed significant activation in bilateral middle frontal and temporal brain regions. Compared to healthy controls, significantly less activation in bilateral cingulate gyrus extending to premotor, primary motor and somatosensory areas was found. During anticipation of loss, the 22q11DS group displayed activity in the left middle frontal gyrus and anterior cingulate cortex, and relative to controls, they showed reduced brain activation in bilateral (pre)cuneus and left posterior cingulate. Within the 22q11DS group, COMT Val hemizygotes displayed more activation compared to Met hemizygotes in right posterior cingulate and bilateral parietal regions during anticipation of reward. During anticipation of loss, COMT Met hemizygotes compared to Val hemizygotes showed more activation in bilateral insula, striatum and left anterior cingulate. Conclusions: This is the first study to investigate reward processing in 22q11DS. Our preliminary results suggest that people with 22q11DS engage a fronto-temporal neural network. Compared to healthy controls, people with 22q11DS primarily displayed reduced activity in medial frontal regions during reward anticipation. COMT hemizygosity affects responsivity of the reward system in this condition. Alterations in reward processing partly underlain by the dopamine system may play a role in susceptibility for psychosis in 22q11DS

JANUS (Jovis Amorum Ac Natorum Undique Scrutator) is a high-resolution visible camera designed for the ESA space mission JUICE (Jupiter Icy moons Explorer). The main scientific goal of JANUS is to observe the surface of the Jupiter satellites Ganymede and Europa in order to characterize their physical and geological properties. During the design phases, we have proposed two possible optical configurations: a Three Mirror Anastigmat (TMA) and a Ritchey-Chrétien (RC) both matching the performance requirements. Here we describe the two optical solutions and compare their performance both in terms of achieved optical quality, sensitivity to misalignment and stray light performances.

A vaccine protecting against all influenza strains is a long-sought goal, particularly for emerging pandemics. As previously shown, vaccines based on the highly conserved extracellular domain of M2 (M2e) may protect against all influenza A strains. Here, we demonstrate that M2e-specific monoclonal antibodies (mAbs) protect mice from a lethal influenza infection. To be protective, antibodies had to be able to bind to Fc receptors and fix complement. Furthermore, mAbs of IgG2c isotype were protective in mice, while antibodies of identical specificity, but of the IgG1 isotype, failed to prevent disease. These findings readily translated into vaccine design. A vaccine targeting M2 in the absence of a toll-like receptor (TLR) 7 ligand primarily induced IgG1, whilst the same vaccine linked to a TLR7 ligand yielded high levels of IgG2c antibodies. Although both vaccines protected mice from a lethal challenge, mice treated with the vaccine containing a TLR7 ligand showed significantly lower morbidity. In accordance with these findings, vaccination of TLR7(-/-) mice with a vaccine containing a TLR7 ligand did not result in protection from a lethal challenge. Hence, the innate immune system is required to direct isotype switching toward the more protective IgG2a/c antibodies.

Early identification of subjects with an increased risk of psychosis is necessary to develop interventions to delay or prevent disease onset. We recently reported that decreased semantic verbal fluency performance in ultra high risk (UHR) subjects predicts the development of psychosis (Becker et al., 2010). The present study investigated whether semantic and verbal fluency scores correlate with grey matter density in UHR subjects. Thirty-seven UHR subjects underwent structural MRI scanning and verbal fluency assessment after which they were followed up for 2 years. Using voxel-based morphometry, we investigated whether grey matter density correlated with verbal fluency scores in 10 UHR subjects who developed psychosis during follow-up and 27 UHR subjects who did not develop psychosis. In UHR subjects developing psychosis, lower semantic fluency scores correlated significantly with reduced grey matter density in the right superior and middle temporal gyrus, the right insula, and the left anterior cingulate cortex. This study shows that a correlation between semantic fluency performance and grey matter density in task-related areas can differentiate between UHR subjects who subsequently will develop psychosis and those who will not. Combining these two measures could improve psychosis prediction in UHR subjects.

Objective: The relationship between cannabis use and psychosis has been studied intensively. Few data, however, are available on the relationship between cannabis use, ultra-high risk for developing psychosis and neurocognition. The aim of the present cross-sectional study was therefore to investigate the relationship between cannabis use, ultra-high-risk (UHR) symptoms and cognitive functioning in UHR patients and healthy controls.Methods: A total of 63 ultra-high-risk patients (34 cannabis users) and 58 control subjects (28 cannabis users) were assessed with clinical measures and a neuropsychological test battery. Patients were eligible for the study if they were between the ages of 12 and 35 years and if they fell into one or more of the following inclusion groups: familial risk and reduced functioning, attenuated psychotic symptoms, brief limited intermittent psychotic symptoms and basic symptoms. Control subjects were eligible for the study if they were between the ages 12 and 35, had no present or past psychiatric illness, no family history of psychiatric illness, no drug use in the non-cannabis-using group, and use of at least four joints per week in the cannabis-using control group.Results: In the UHR and the control group, cannabis users experienced more basic symptoms and UHR symptoms than the non-cannabis users. Moreover, cannabis users in the control group performed at the level of the UHR subjects on a test of verbal memory and verbal fluency. Frequency of cannabis use correlated with severity of several UHR symptoms.Conclusions: Cannabis-using UHR patients have more basic symptoms than non-using patients. In addition, healthy cannabis users have more subclinical UHR and basic symptoms and more neuropsychological dysfunctions than non-cannabis users. More frequent cannabis use was related to increased severity of certain UHR symptoms.

Background: Autistic Spectrum Disorder (ASD), including Asperger syndrome and autism, is a highly genetic neurodevelopmental disorder. There is a consensus that ASD has a biological basis, and it has been proposed that it is a “connectivity” disorder. Diffusion Tensor Magnetic Resonance Imaging (DT-MRI) allows measurement of the microstructural integrity of white matter (a proxy measure of “connectivity”). However, nobody has investigated the microstructural integrity of whole brain white matter in people with Asperger syndrome. Methods: We measured the fractional anisotropy (FA), mean diffusivity (MD) and radial diffusivity (RD) of white matter, using DT-MRI, in 13 adults with Asperger syndrome and 13 controls. The groups did not differ significantly in overall intelligence and age. FA, MD and RD were assessed using whole brain voxel-based techniques. Results: Adults with Asperger syndrome had a significantly lower FA than controls in 13 clusters. These were largely bilateral and included white matter in the internal capsule, frontal, temporal, parietal and occipital lobes, cingulum and corpus callosum. Conclusions: Adults with Asperger syndrome have widespread significant differences from controls in white matter microstructural integrity.

Allergen-specific desensitization is the only disease-modifying therapy currently available for the treatment of allergies. These therapies require application of allergen over several years and some may induce life-threatening anaphylactic reactions. An ideal vaccine for desensitization should be highly immunogenic and should alleviate allergic symptoms upon few injections while being nonreactogenic. We describe such a vaccine for the treatment of cat allergy, consisting of the major cat allergen Fel d1 coupled to bacteriophage Qbeta-derived virus-like particles (Qbeta-Fel d1). Qbeta-Fel d1 was highly immunogenic, and a single vaccination was sufficient to induce protection against type I allergic reactions. Allergen-specific immunoglobulin G antibodies were shown to be the critical effector molecules and alleviated symptoms by two distinct mechanisms. Although allergen-induced systemic basophil degranulation was inhibited in an FcgammaRIIb-dependent manner, inhibition of local mast cell degranulation in tissues occurred independently of FcgammaRIIb. In addition, treatment with Qbeta-Fel d1 abolished IgE memory responses upon antigen recall. Despite high immunogenicity, the vaccine was essentially nonreactogenic and vaccination induced neither local nor systemic anaphylactic reactions in sensitized mice. Moreover, Qbeta-Fel d1 did not induce degranulation of basophils derived from human volunteers with cat allergies. These data suggest that vaccination with Qbeta-Fel d1 may be a safe and effective treatment for cat allergy.

IL-17-producing CD4 + T cells have been recognized as key players in organ-related autoimmune disease; however, the parameters that govern their development are yet to be elucidated fully. By using both in vivo and in vitro systems, we have investigated the role of antigen dose, pathogen-associated molecular patterns, and CD40–CD40 ligand (CD40L) cross-talk in Th17 differentiation. We found that the strength of antigenic stimulation critically influenced the extent of Th17 differentiation, because high, but not low or intermediate, antigen concentrations led to IL-17 production. Strong antigenic stimulation of T cells up-regulated CD40L expression, which in concert with certain microbial stimuli (i.e., cytosine phosphate guanine, curdlan, and zymosan) synergistically increased dendritic cell (DC) IL-6 production and Th17 polarization. CD40-deficient DCs exhibited reduced cytokine release and failed to drive Th17 development in vitro. These results were confirmed in vivo where the absence of CD40–CD40L cross-talk was found to prevent the expansion of IL-17-producing cells and accordingly the development of experimental autoimmune encephalitis. Our data demonstrate that CD40–CD40L cross-talk is important for Th17 development by translating strong T cell receptor and microbial stimuli into IL-6 production.

The JUICE (JUpiter ICy moons Explorer) satellite of the European Space Agency (ESA) is dedicated to the detailed study of Jupiter and its moons. Among the whole instrument suite, JANUS (Jovis, Amorum ac Natorum Undique Scrutator) is the camera system of JUICE designed for imaging at visible wavelengths. It will conduct an in-depth study of Ganymede, Callisto and Europa, and explore most of the Jovian system and Jupiter itself, performing, in the case of Ganymede, a global mapping of the satellite with a resolution of 400 m/px. The optical design chosen to meet the scientific goals of JANUS is a three mirror anastigmatic system in an off-axis configuration. To ensure that the achieved contrast is high enough to observe the features on the surface of the satellites, we also performed a preliminary stray light analysis of the telescope. We provide here a short description of the optical design and we present the procedure adopted to evaluate the stray-light expected during the mapping phase of the surface of Ganymede. We also use the results obtained from the first run of simulations to optimize the baffle design.
