Abstract The circadian clock of cyanobacteria, which predicts daily environmental changes, typically includes a standard oscillator consisting of proteins KaiA, KaiB, and KaiC. However, several cyanobacteria have diverse Kai protein homologs of unclear function. In particular, Synechocystis sp. PCC 6803 harbours, in addition to a canonical kaiABC gene cluster (named kaiAB1C1), two further kaiB and kaiC homologs (kaiB2, kaiB3, kaiC2, kaiC3). Here, we identify a chimeric KaiA homolog, named KaiA3, encoded by a gene located upstream of kaiB3. At the N-terminus, KaiA3 is similar to response-regulator receiver domains, whereas its C-terminal domain resembles that of KaiA. Homology analysis shows that a KaiA3-KaiB3-KaiC3 system exists in several cyanobacteria and other bacteria. Using the Synechocystis sp. PCC 6803 homologs, we observe circadian oscillations in KaiC3 phosphorylation in vitro in the presence of KaiA3 and KaiB3. Mutations of kaiA3 affect KaiC3 phosphorylation, leading to growth defects under both mixotrophic and chemoheterotrophic conditions. KaiC1 and KaiC3 exhibit phase-locked free-running phosphorylation rhythms. Deletion of either system (∆kaiAB1C1 or ∆kaiA3B3C3) alters the period of the cellular backscattering rhythm. Furthermore, both oscillators are required to maintain high-amplitude, self-sustained backscatter oscillations with a period of approximately 24 h, indicating their interconnected nature.

Abstract Cyanobacteria are a diverse group of prokaryotic organisms that have been the subject of intense basic research, resulting in a wealth of knowledge about fundamental cellular processes such as photosynthesis. However, the translation of that research towards industry-relevant applications is still limited. To understand the reasons for this contradictory situation, we conducted a quantitative survey among researchers in the cyanobacterial community, a set of individual interviews with established researchers, and a literature analysis. Our results show that the community seems to be committed to embracing cyanobacterial diversity and promoting collaboration. Additionally, participants expressed a strong desire to develop standardized protocols for research and establish larger consortia to accelerate progress. The results of the survey highlight the need for a more integrated approach to cyanobacterial research that encompasses both basic and applied aspects. Based on the survey and interview results as well as our literature analysis, we highlight areas for potential improvement, strategies to enhance cyanobacterial research, and open questions that demand further exploration. Addressing these challenges should accelerate the development of industrial applications based on cyanobacterial research.

Insights in age- and sex-specific coronary atherosclerotic plaque characteristics may contribute to a better understanding of coronary artery disease and, ultimately, to its prevention and treatment. In 307 women and 406 men aged 20 to 90 years undergoing intravascular ultrasound imaging, sex-based differences in coronary atherosclerotic plaque characteristics were mainly present in younger patients, while these differences were less pronounced at advanced age.

Abstract Background People with serious mental illness (SMI) and people with intellectual disabilities/developmental disabilities (ID/DD) are at higher risk for COVID-19 and more severe outcomes. We compare a tailored versus general best practice COVID-19 prevention program in group homes (GHs) for people with SMI or ID/DD in Massachusetts (MA). Methods A hybrid effectiveness-implementation cluster randomized control trial compared a four-component implementation strategy (Tailored Best Practices: TBP) to dissemination of standard prevention guidelines (General Best-Practices: GBP) in GHs across six MA behavioral health agencies. GBP consisted of standard best practices for preventing COVID-19. TBP included GBP plus four components including: (1) trusted-messenger peer testimonials on benefits of vaccination; (2) motivational interviewing; (3) interactive education on preventive practices; and (4) fidelity feedback dashboards for GHs. Primary implementation outcomes were full COVID-19 vaccination rates (baseline: 1/1/2021–3/31/2021) and fidelity scores (baseline: 5/1/21–7/30/21), at 3-month intervals to 15-month follow-up until October 2022. The primary effectiveness outcome was COVID-19 infection (baseline: 1/1/2021–3/31/2021), measured every 3 months to 15-month follow-up. Cumulative incidence of vaccinations were estimated using Kaplan-Meier curves. Cox frailty models evaluate differences in vaccination uptake and secondary outcomes. Linear mixed models (LMMs) and Poisson generalized linear mixed models (GLMMs) were used to evaluate differences in fidelity scores and incidence of COVID-19 infections. Results GHs (n=415) were randomized to TBP (n=208) and GBP (n=207) including 3,836 residents (1,041 ID/DD; 2,795 SMI) and 5,538 staff. No differences were found in fidelity scores or COVID-19 incidence rates between TBP and GBP, however TBP had greater acceptability, appropriateness, and feasibility. No overall differences in vaccination rates were found between TBP and GBP. However, among unvaccinated group home residents with mental disabilities, non-White residents achieved full vaccination status at double the rate for TBP (28.6%) compared to GBP (14.4%) at 15 months. Additionally, the impact of TBP on vaccine uptake was over two-times greater for non-White residents compared to non-Hispanic White residents (ratio of HR for TBP between non-White and non-Hispanic White: 2.28, p = 0.03). Conclusion Tailored COVID-19 prevention strategies are beneficial as a feasible and acceptable implementation strategy with the potential to reduce disparities in vaccine acceptance among the subgroup of non-White individuals with mental disabilities. ABSTRACT Aurora kinases are crucial regulators of mitotic cell cycle progression in eukaryotes. The protozoan malaria parasite Plasmodium falciparum replicates via schizogony, a specialized mode of cell division characterized by consecutive asynchronous rounds of nuclear division by closed mitosis followed by a single cytokinesis event producing dozens of daughter cells. P. falciparum encodes three Aurora-related kinases (PfARKs) that have been reported essential for parasite proliferation, but their roles in regulating schizogony have not yet been explored in great detail. Here, we engineered transgenic parasite lines expressing GFP-tagged PfARK1–3 to provide a systematic analysis of their expression timing and subcellular localization throughout schizogony as well as in the non-dividing gametocyte stages, which are essential for malaria transmission. We demonstrate that all three PfARKs display distinct and highly specific and exclusive spatiotemporal associations with the mitotic machinery. In gametocytes, PfARK3 is undetectable, and PfARK1 and PfARK2 show male-specific expression in late-stage gametocytes, consistent with their requirement for endomitosis during male gametogenesis in the mosquito vector. Our combined data suggest that PfARK1 and PfARK2 have non-overlapping roles in centriolar plaque maturation, assembly of the mitotic spindle, kinetochore–spindle attachment and chromosome segregation, while PfARK3 seems to be exquisitely involved in daughter cell cytoskeleton assembly and cytokinesis. These important new insights provide a reliable foundation for future research aiming at the functional investigation of these divergent and possibly drug-targetable Aurora-related kinases in mitotic cell division of P. falciparum and related apicomplexan parasites.IMPORTANCEMalaria parasites replicate via non-conventional modes of mitotic cell division, such as schizogony, employed by the disease-causing stages in the human blood or endomitosis during male gametogenesis in the mosquito vector. Understanding the molecular mechanisms regulating cell division in these divergent unicellular eukaryotes is not only of scientific interest but also relevant to identify potential new antimalarial drug targets. Here, we carefully examined the subcellular localization of all three Plasmodium falciparum Aurora-related kinases (ARKs), distantly related homologs of Aurora kinases that coordinate mitosis in model eukaryotes. Detailed fluorescence microscopy-based analyses revealed distinct, specific, and exclusive spatial associations for each parasite ARK with different components of the mitotic machinery and at different phases of the cell cycle during schizogony and gametocytogenesis. This comprehensive set of results closes important gaps in our fragmentary knowledge on this important group of kinases and offers a valuable source of information for future functional studies.

With innovative portable force plate systems being widely implemented for lower-body neuromuscular performance assessment in an applied sports setting and the existing gap in the scientific literature regarding player performance during in-game competitive scenarios, the purpose of the present study was to compare changes in countermovement vertical jump (CVJ) performance pre-post a simulated 3×3 basketball tournament. Seven current or former members of a 3×3 national basketball team volunteered to participate in the present investigation. Upon completing standardized warm-up procedures, athletes stepped on a uni-axial force plate system sampling at 1,000 Hz and performed three maximal-effort CVJs with no arm swing. Then, the athletes proceeded to play a simulated 3×3 basketball tournament composed of two consecutive games, separated by a 15-min rest interval. Immediately following the completion of the second game, the identical CVJ testing procedures were repeated. Paired sample t-tests were used to examine pre-post-tournament differences in nineteen CVJ performance metrics (p < 0.05). The results reveal that force-time metrics during both eccentric and concentric phases of the CVJ remain relatively unchanged pre-post simulated 3×3 basketball tournament. However, multiple force-time metrics within the eccentric phase of the CVJ changed by 12.1%–19.1% (e.g., eccentric peak power and peak velocity, eccentric duration), suggesting that the eccentric phase of CVJ might be responsive to performance stimulus to a greater extent than the concentric phase. Overall, these findings further support the importance of comprehensive CVJ analysis when intending to measure changes in neuromuscular performance.

Immune checkpoint inhibitors (ICI) have transformed the management of cancer, particularly for older adults, who constitute a majority of the global cancer patient population. This study aimed to assess the inclusion, characteristics, and reporting of older adults enrolled in Food and Drug Administration (FDA) registration clinical trials of ICI between 2018 and 2022. Clinical trials of ICI leading to an FDA approval in solid tumor oncology between 2018 and 2022 were included. Primary study reports and all available secondary publications were assessed. The availability and completeness of older subgroup data for protocol-defined clinical efficacy endpoints, health-related quality of life (HRQOL) and toxicity outcomes, and baseline characteristics were assessed according to predefined criteria which categorized reporting completeness hierarchically in relation to the availability of published data, including effect size, sample size, and measures of precision. 53 registration trials were included, involving a total of 37,094 participants. Most trials (64.2%) were of ICI combination therapy. 42.3% of patients were aged≥65 years; 11.1% were aged≥75. No trials specified an upper age limit for eligibility. 98.1% of trials excluded patients with European Cooperative Oncology Group performance status>1. 87.2% of primary efficacy endpoints and 17.9% of secondary efficacy endpoints were reported completely for older adults. Five studies (9.4%) reported baseline characteristics, three (6.1%) reported HRQOL assessments, and four (7.5%) reported toxicity outcomes completely among older subgroups. No trials conducted baseline geriatric assessments or reported geriatric-specific symptoms or quality of life scales. This analysis highlights significant deficits in the enrollment and reporting of older subgroups in pivotal trials of ICI therapy. The findings highlight an urgent need for improved reporting and inclusion standards in clinical trials of ICI to better inform treatment decisions for older adults.

IntroductionAdvances in motion capture technology include markerless systems to facilitate valid data collection. Recently, the technological reliability of this technology has been reported for human movement assessments. To further understand sources of potential error, biological reliability must also be determined. The aim of this study was to determine the day-to-day reliability for a three-dimensional markerless motion capture (MMC) system to quantify 4 movement analysis composite scores, and 81 kinematic variables.MethodsTwenty-two healthy men (n = 11; X¯±SD; age = 23.0 ± 2.6 years, height = 180.4.8 cm, weight = 80.4 ± 7.3 kg) and women (n = 11; age = 20.8 ± 1.1 years, height = 172.2 ± 7.4 cm, weight = 68.0 ± 7.3 kg) participated in this study. All subjects performed 4 standardized test batteries consisting of 14 different movements on four separate days. A three-dimensional MMC system (DARI Motion, Lenexa, KS) using 8 cameras surrounding the testing area was used to quantify movement characteristics. 1 × 4 RMANOVAs were used to determine significant differences across days for the composite movement analysis scores, and RM-MANOVAs were used to determine test day differences for the kinematic data (p 50% total variability).DiscussionCombined, these findings effectively add to the body of literature suggesting sufficient reliability for MMC solutions in capturing kinematic features of human movement.

Abstract The ever‐increasing importance of critical metals (CMs) in modern society underscores their resource security and circularity. Waste‐printed circuit boards (WPCBs) are particularly attractive reservoirs of CMs due to their gamut CM embedding and ubiquitous presence. However, the recovery of most CMs is out of reach from current metal‐centric recycling industries, resulting in a flood loss of refined CMs. Here, 41 types of such spent CMs are identified. To deliver a higher level of CM sustainability, this work provides an insightful overview of paradigm‐shifting pathways for CM recovery from WPCBs that have been developed in recent years. As a crucial starting entropy‐decreasing step, various strategies of metal enrichment are compared, and the deployment of artificial intelligence (AI) and hyperspectral sensing is highlighted. Then, tailored metal recycling schemes are presented for the platinum group, rare earth, and refractory metals, with emphasis on greener metallurgical methods contributing to transforming CMs into marketable products. In addition, due to the vital nexus of CMs between the environment and energy sectors, the upcycling of CMs into electro‐/photo‐chemical catalysts for green fuel synthesis is proposed to extend the recycling chain. Finally, the challenges and outlook on this all‐round upgrading of WPCB recycling are outlined.

Streptococcus pneumoniae is a major pathogen responsible for severe complications in patients with prior influenza A virus (IAV) infection. We have previously demonstrated that S. pneumoniae exhibits increased intracellular survival within IAV-infected cells. Fluoroquinolones (FQs) are widely used to treat pneumococcal infections. However, our prior work has shown that S. pneumoniae can develop intracellular FQ persistence, a phenomenon triggered by oxidative stress within host cells. This persistence allows the bacteria to withstand high FQ concentrations. In this study, we show that IAV infection enhances pneumococcal FQ persistence during intracellular survival within pneumocytes, macrophages, and neutrophils. This enhancement is partly due to increased oxidative stress induced by the viral infection. We find that this phenotype is particularly pronounced in autophagy-proficient host cells, potentially resulting from IAV-induced blockage of autophagosome-lysosome fusion. Moreover, we identified several S. pneumoniae genes involved in oxidative stress response that contribute to FQ persistence, including sodA (superoxide dismutase), clpL (chaperone), nrdH (glutaredoxin), and psaB (Mn+2 transporter component). Our findings reveal a novel mechanism of antibiotic persistence promoted by viral infection within host cells. This underscores the importance of considering this phenomenon when using FQs to treat pneumococcal infections, especially in patients with concurrent influenza A infection.

IntroductionWith recent increases in the popularity of studying the physical construct of horizontal deceleration performance in team-sport athletes, the aim of the present study was to assess the inter-rater and intra-rater reliability of processing and quantifying horizontal deceleration ability using radar technology.MethodsData from 92 NCAA Division 1 athletes from two different athletic teams (American football and Lacrosse) were used for the present investigation. All athletes performed two trials of the modified acceleration to deceleration assessment (ADA), which consisted of a maximal 10 m sprint acceleration, followed by a rapid deceleration. Four individual raters manually processed raw, radar-derived instantaneous velocity data for the ADA, and an automated script was used to calculate metrics of interest.ResultsPrimary study findings suggest moderate to excellent levels of agreement (ICC = 0.56–0.91) for maximal horizontal deceleration metrics between the four individual raters. The intra-rater analyses revealed poor to excellent consistency (ICC = 0.31–0.94) between ADA trials, with CV%'s ranging from 3.1% to 13.2%, depending on the respective metric and rater.DiscussionOur data suggests that if a foundational understanding and agreement of manual data processing procedures for radar-derived data is given between raters, metrics may be interpreted with moderate to excellent levels of confidence. However, when possible, and when using the Stalker ATS radar technology, authors recommend that practitioners use one trained individual to manually process raw data. Ideally, this process should become fully automated, based on selected filters or algorithms, rather than the subjectivity of the rater.

En América Latina las revueltas urbanas son cada vez más frecuentes y tienen importantes consecuencias. Sin embargo, la mayoría de las investigaciones existentes se basan en estudios de caso, lo cual impide obtener generalizaciones sobre sus causas, dinámicas y consecuencias. En este artículo discutimos la literatura sobre revueltas urbanas en América Latina y proponemos un marco conceptual para estudiarlas. Definimos las revueltas urbanas con base en cuatro atributos, las distinguimos de otras formas de política contenciosa y presentamos proposiciones sobre su desarrollo y dinámicas. Hipotetizamos que las revueltas urbanas son más probables cuando los sectores urbanos y populares no han sido completamente incluidos política, económica y culturalmente; cuando existen lazos débiles e inorgánicos entre élites políticas y ciudadanía; y cuando los sectores populares están débilmente articulados en asociaciones conectadas horizontalmente. Sin embargo, el momento y dinámica de las revueltas dependen de los factores precipitantes, las respuestas gubernamentales y las condiciones para la difusión de las protestas.

Given its fast-growing popularity and unique on-court competitive demands, 3 × 3 basketball has captured a considerable amount of attention over recent years. However, unlike research focused on studying 5 × 5 basketball players, there is a lack of scientific literature focused on examining countermovement vertical jump (CMJ) and sprint performance characteristics of 3 × 3 athletes. Thus, the purpose of the present study was to compare force-time metrics during both eccentric and concentric phases of the CMJ and acceleration and deceleration capabilities between 3 × 3 and 5 × 5 top-tier professional male basketball athletes. IntroductionWhile using force-plate derived measures of vertical jump performance, reflective of stretch-shortening-cycle (SSC) efficiency is common practice in sport science, there is limited evidence as to which tests and measures may be most sensitive toward neuromuscular fatigue. The aim of this study was to explore the SSC fatigue response to a one-week high-intensity fatiguing phase of training in National Collegiate Athletic Association (NCAA) Division-I basketball players.MethodsThe study timeline consisted of three weeks of baseline measures, one week of high-intensity training, and two weeks of follow-up testing. Countermovement jumps (CMJ) and 10-5 hop tests were performed at baseline, as well as at two time-points during, and three time-points following the fatiguing training period, allowing for performance-comparisons with baseline.ResultsCompared to the weekly training sum at baseline, during the high intensity training phase, athletes were exposed to very large increases in selected external load metrics (ES = 1.44–3.16), suggesting that athletes experienced fatigue acutely, as well as potential longer lasting reductions in performance. Vertical jump data suggested that in the CMJ, traditional metrics such as jump height, as well as metrics reflecting kinetic outputs and movement strategies, were sensitive to the stark increase in high-intensity training exposure. The 10-5 hop test suggested a fatigue-induced loss of tolerance to ground impact reflected by performance reductions in metrics related to jump height and reactive strength qualities.DiscussionThese findings emphasize that when monitoring neuromuscular fatigue, variables and assessments may not be looked at individually, but rather as part of a more global monitoring approach.

In a sport-specific setting, the external load represents the cumulative amount of stress placed on the athlete during a specific activity, while the internal load describes the athlete’s individual physiological response to that particular activity. The purpose of this study was to determine the relationship between external load measured with a tri-axial accelerometer and salivary testosterone (T), cortisol (C), and testosterone-to-cortisol ratio (T/C) as measures of internal load during a 5-on-5 basketball game. Six professional male basketball players volunteered to participate in the investigation (age=28.3±1.9 years; height=193.3±10.6 cm; body mass=90.7±10.7 kg). Upon completing a standardized warm-up protocol, athletes played a simulated basketball game composed of four 10-minute quarters with the Elam Ending. Each athlete wore a tri-axial inertial measurement unit (StriveTech, Bothell, WA, USA) embedded within tightly fitted compression shorts throughout the competitive period. Cyanobacterial blooms are a persistent concern to water management and treatment, with blooms potentially causing the release of toxins and degrading water quality. However, previous models have not considered the zero inflation of cyanobacteria count data. Typically, a relatively large proportion of measured count data are zeros or non-detects of cyanobacteria, representing either no cyanobacteria was present or the cell number was too low to be detected. Commonly used Poisson and negative binomial models for count data underestimate the probability of zero data, making these models less reliable. This study proposes a Bayesian approach to fit the cyanobacteria abundance data with mixture models that handle zero-inflated data. Predictor variables considered included weather and water quality measures that can easily be obtained day-to-day. The optimal model (zero-inflated negative binomial) was used to predict cyanobacteria alert levels on a separate test set. The ability to predict narrow alert levels was limited, however, 76% accuracy was achieved in predicting cyanobacteria counts above or below 1,000 cells/mL. Parameter estimates were highly variable and demonstrated that complex and uncertain factors influence cyanobacteria count predictions. The modelling approach can be applied to a wide range of environmental problems where zero-inflated data is common. Quantifying viral growth rates is key to understanding evolutionary dynamics and the potential for mutants to escape antiviral drugs. Defining evolutionary escape paths and their impact on viral fitness allows for the development of drugs that are resistant to escape. In the case of HIV, combination antiretroviral therapy can successfully prevent or treat infection, but it relies on strict adherence to prevent escape. Here, we present a method termed QuickFit that enables the quantification of viral fitness by employing large numbers of parallel viral cultures to measure growth rates accurately. QuickFit consistently recapitulated HIV growth measurements obtained by traditional approaches, but with significantly higher throughput and lower rates of error. This method represents a promising tool for rapid and consistent evaluation of viral fitness.

Abstract This study evaluates the quality of published research using artificial intelligence (AI) for ovarian cancer diagnosis or prognosis using histopathology data. A systematic search of PubMed, Scopus, Web of Science, Cochrane CENTRAL, and WHO-ICTRP was conducted up to May 19, 2023. Inclusion criteria required that AI was used for prognostic or diagnostic inferences in human ovarian cancer histopathology images. Risk of bias was assessed using PROBAST. Information about each model was tabulated and summary statistics were reported. The study was registered on PROSPERO (CRD42022334730) and PRISMA 2020 reporting guidelines were followed. Searches identified 1573 records, of which 45 were eligible for inclusion. These studies contained 80 models of interest, including 37 diagnostic models, 22 prognostic models, and 21 other diagnostically relevant models. Common tasks included treatment response prediction (11/80), malignancy status classification (10/80), stain quantification (9/80), and histological subtyping (7/80). Models were developed using 1–1375 histopathology slides from 1–776 ovarian cancer patients. A high or unclear risk of bias was found in all studies, most frequently due to limited analysis and incomplete reporting regarding participant recruitment. Limited research has been conducted on the application of AI to histopathology images for diagnostic or prognostic purposes in ovarian cancer, and none of the models have been demonstrated to be ready for real-world implementation. Key aspects to accelerate clinical translation include transparent and comprehensive reporting of data provenance and modelling approaches, and improved quantitative evaluation using cross-validation and external validations. Given the multidirectional nature of the sport, handball athletes must frequently perform high-intensity decelerations to avoid defenders, generate space, or perform directional changes. The aim of the present study was twofold: (i) to investigate different kinematic measures of horizontal deceleration performance by comparing the acceleration-deceleration assessment (ADA) with the 5-0-5 test and (ii) to investigate relationships between force-time characteristics derived from the countermovement vertical jump (CVJ) and measures of horizontal deceleration performance. Eleven female handball players competing in the first-tier professional league in Europe performed three CVJs while standing on a uni-axial force plate system sampling at 1000 Hz, followed by two ADAs (i.e., maximal-effort acceleration over a 10 m distance, followed by rapid deceleration) and 5-0-5 test trials. Tripod-mounted radar sampling at 47 Hz, placed 5 m behind the start line, was used to record horizontal velocity data. Each test was separated by a 5–7 min rest interval to minimize the influence of fatigue. No statistically significant differences were found in horizontal deceleration performance parameters between ADA and the 5-0-5 test. However, athletes with a higher CVJ height and reactive strength index-modified showed better performance in terms of horizontal deceleration measures such as maximal approach velocity and average and maximal deceleration. Overall, these results may be of interest to practitioners working with multidirectional sport athletes such as handball players as they provide critical insight for the selection of assessments and training strategies targeted toward optimizing on-court athlete performance.

Left ventricular outflow tract obstruction (LVOTO) is a notorious complication of transcatheter mitral valve replacement (TMVR). Computed tomography–derived simulations can predict neo-LVOTO post-TMVR, whereas alcohol septal ablation can mitigate neo-LVOTO risk. We report a case of sequential alcohol septal ablation of 2 adjacent septal branches to resolve unexpected neo-LVOTO post-TMVR.

CyanoCyc is a web portal that integrates an exceptionally rich database collection of information about cyanobacterial genomes with an extensive suite of bioinformatics tools. It was developed to address the needs of the cyanobacterial research and biotechnology communities. The 277 annotated cyanobacterial genomes currently in CyanoCyc are supplemented with computational inferences including predicted metabolic pathways, operons, protein complexes, and orthologs; and with data imported from external databases, such as protein features and Gene Ontology (GO) terms imported from UniProt. Five of the genome databases have undergone manual curation with input from more than a dozen cyanobacteria experts to correct errors and integrate information from more than 1,765 published articles. CyanoCyc has bioinformatics tools that encompass genome, metabolic pathway and regulatory informatics; omics data analysis; and comparative analyses, including visualizations of multiple genomes aligned at orthologous genes, and comparisons of metabolic networks for multiple organisms. CyanoCyc is a high-quality, reliable knowledgebase that accelerates scientists’ work by enabling users to quickly find accurate information using its powerful set of search tools, to understand gene function through expert mini-reviews with citations, to acquire information quickly using its interactive visualization tools, and to inform better decision-making for fundamental and applied research.

This review aimed to specify different concepts that are essential to the development of medical devices (MDs) with artificial intelligence (AI) (AI-based MDs) and shed light on how algorithm performance, interpretability, and explainability are key assets. First, a literature review was performed to determine the key criteria needed for a health technology assessment of AI-based MDs in the existing guidelines. Then, we analyzed the existing assessment methodologies of the different criteria selected after the literature review. The scoping review revealed that health technology assessment agencies have highlighted different criteria, with 3 important ones to reinforce confidence in AI-based MDs: performance, interpretability, and explainability. We give recommendations on how and when to evaluate performance on the basis of the model structure and available data. In addition, should interpretability and explainability be difficult to define mathematically, we describe existing ways to support their evaluation. We also provide a decision support flowchart to identify the anticipated regulatory requirements for the development and assessment of AI-based MDs. The importance of explainability and interpretability techniques in health technology assessment agencies is increasing to hold stakeholders more accountable for the decisions made by AI-based MDs. The identification of 3 main assessment criteria for AI-based MDs according to health technology assessment guidelines led us to propose a set of tools and methods to help understand how and why machine learning algorithms work as well as their predictions.

New targeted treatments are urgently needed to improve triple-negative breast cancer (TNBC) patient survival. Previously, we identified the cell surface protein A Disintegrin And Metalloprotease 8 (ADAM8) as a driver of TNBC tumor growth and spread via its metalloproteinase and disintegrin (MP and DI) domains. In proof-of-concept studies, we demonstrated that a monoclonal antibody (mAb) that simultaneously inhibits both domains represents a promising therapeutic approach. Here, we screened a hybridoma library using a multistep selection strategy, including flow cytometry for Ab binding to native conformation protein and in vitro cell-based functional assays to isolate a novel panel of highly specific human ADAM8 dual MP and DI inhibitory mAbs, called ADPs. The screening of four top candidates for in vivo anti-cancer activity in an orthotopic MDA-MB-231 TNBC model of ADAM8-driven primary growth identified two lead mAbs, ADP2 and ADP13. Flow cytometry, hydrogen/deuterium exchange–mass spectrometry (HDX-MS) and alanine (ALA) scanning mutagenesis revealed that dual MP and DI inhibition was mediated via binding to the DI. Further testing in mice showed ADP2 and ADP13 reduce aggressive TNBC characteristics, including locoregional regrowth and metastasis, and improve survival, demonstrating strong therapeutic potential. The continued development of these mAbs into an ADAM8-targeted therapy could revolutionize TNBC treatment.

Generations of scientists have pursued the goal of defining beauty. While early scientists initially focused on objective criteria of beauty ('feature-based aesthetics'), philosophers and artists alike have since proposed that beauty arises from the interaction between the object and the individual who perceives it. The aesthetic theory of fluency formalizes this idea of interaction by proposing that beauty is determined by the efficiency of information processing in the perceiver's brain ('processing-based aesthetics'), and that efficient processing induces a positive aesthetic experience. The theory is supported by numerous psychological results, however, to date there is no quantitative predictive model to test it on a large scale. In this work, we propose to leverage the capacity of deep convolutional neural networks (DCNN) to model the processing of information in the brain by studying the link between beauty and neuronal sparsity, a measure of information processing efficiency. Whether analyzing pictures of faces, figurative or abstract art paintings, neuronal sparsity explains up to 28% of variance in beauty scores, and up to 47% when combined with a feature-based metric. However, we also found that sparsity is either positively or negatively correlated with beauty across the multiple layers of the DCNN. Our quantitative model stresses the importance of considering how information is processed, in addition to the content of that information, when predicting beauty, but also suggests an unexpectedly complex relationship between fluency and beauty.

With force plates being widely implemented for neuromuscular performance assessment in sport-specific settings and various force-time metrics being able to differentiate athletes based on their performance capabilities, the purpose of the present study was to examine the differences in countermovement vertical jump (CVJ) characteristics between starting and non-starting professional male basketball players (e.g., ABA League). IntroductionA study was conducted to investigate if an individual’s trust in law enforcement affects their perception of the emotional facial expressions displayed by police officers.MethodsThe study invited 77 participants to rate the valence of 360 face images. Images featured individuals without headgear (condition 1), or with a baseball cap (condition 2) or police hat (condition 3) digitally added to the original photograph. The images were balanced across sex, race/ethnicity (Asian, African American, Latine, and Caucasian), and facial expression (Happy, Neutral, and Angry). After rating the facial expressions, respondents completed a survey about their attitudes toward the police.ResultsThe results showed that, on average, valence ratings for “Angry” faces were similar across all experimental conditions. However, a closer examination revealed that faces with police hats were perceived as angrier compared to the control conditions (those with no hat and those with a baseball cap) by individuals who held negative views of the police. Conversely, participants with positive attitudes toward the police perceived faces with police hats as less angry compared to the control condition. This correlation was highly significant for angry faces (p < 0.01), and stronger in response to male faces compared to female faces but was not significant for neutral or happy faces.DiscussionThe study emphasizes the substantial role of attitudes in shaping social perception, particularly within the context of law enforcement.

Admixture, the exchange of genetic information between distinct source populations, is thought to be a major source of adaptive genetic variation. Unlike mutation events, which periodically generate single alleles, admixture can introduce many selected alleles simultaneously. As such, the effects of linkage between selected alleles may be especially pronounced in admixed populations. However, existing tools for identifying selected mutations within admixed populations only account for selection at a single site, overlooking phenomena such as linkage among proximal selected alleles. Here, we develop and extensively validate a method for identifying and quantifying the individual effects of multiple linked selected sites on a chromosome in admixed populations. Our approach numerically calculates the expected local ancestry landscape in an admixed population for a given multi-locus selection model, and then maximizes the likelihood of the model. After applying this method to admixed populations of Drosophila melanogaster and Passer italiae, we found that the impacts between linked sites may be an important contributor to natural selection in admixed populations. Furthermore, for the situations we considered, the selection coefficients and number of selected sites are overestimated in analyses that do not consider the effects of linkage among selected sites. Our results imply that linkage among selected sites may be an important evolutionary force in admixed populations. This tool provides a powerful generalized method to investigate these crucial phenomena in diverse populations.

The purpose of the present study was to examine the acute impact of resistance exercise on basketball shooting mechanics and accuracy. Ten resistance-trained recreationally active men with previous basketball playing experience (x̄ ± SD; height = 182.6 ± 9.7 cm; body mass = 79.2 ± 13.9 kg; age = 25.6 ± 5.5 years) performed control, upper-body, and lower-body training sessions in randomized order followed by 5 sets of stationary free-throw (4.57 m), two-point (5.18 m) and three-point (6.75 m) basketball shooting drills in 30 min time increments. This research explores early Event-Related Potentials (ERPs) sensitivity to facial stimuli, investigating various facial features aimed to unveil underlying neural mechanisms. Two experiments, each involving 15 undergraduate students, utilized a multidimensional stimulus set incorporating race, gender, age, emotional expression, face masks, and stimulus orientation. Findings highlight significant modulations in N170 and P200 amplitudes and latencies for specific attributes, replicating prior research and revealing novel insights. Notably, age-related facial feature variations, facial inversion, and the presence of face masks significantly impact neural responses. Several speculative explanations are proposed to elucidate these results: First, the findings lend support to the idea that the increased N170 amplitude observed with facial inversion is closely tied to the activation of object-sensitive neurons. This is further bolstered by a similar amplitude increase noted when masks (effective objects) are added to faces. Second, the absence of an additional amplitude increase, when inverting face images with face masks suggests that neural populations may have reached a saturation point, limiting further enhancement. Third, the study reveals that the latency deficit in N170 induced by facial inversion is even more pronounced in the subsequent ERP component, the P200, indicating that face inversion may impact multiple stages of face processing. Lastly, the significant increase in P200 amplitude, typically associated with face typicality, for masked faces in this study aligns with previous research that demonstrated elevated P200 amplitudes for scrambled faces. This suggests that obscured faces may be processed as typical, potentially representing a default state in face processing.

Background: Thrombolysis in Myocardial Infarction (TIMI) 0 flow often characterizes ST-segment Elevation Myocardial Infarction (STEMI) patients, but may also feature in non-ST-segment Elevation Acute Coronary Syndrome (NSTE-ACS). Since recanalization usually occurs later in NSTE-ACS patients, the aim of this study was to assess whether patients presenting with NSTE-ACS and TIMI 0 flow have worse clinical outcomes as compared to patients presenting with STEMI and TIMI 0 flow. Methods: A single-center retrospective cohort study was conducted with patients treated for NSTE-ACS and STEMI with TIMI 0 flow at diagnostic angiogram between January 2015 and December 2019. The two patient groups were 1:1 matched using a propensity score logistic regression model. The primary outcome was Major Adverse Cardiac Events (MACE), a composite of all-cause mortality, any myocardial infarction, coronary artery bypass graft, urgent target vessel revascularization or stroke during long term follow-up. Results: The total population consisted of 1255 ACS patients, of which 249 NSTE-ACS and 1006 STEMI patients. After propensity score matching, 234 NSTE-ACS patients were matched with 234 STEMI patients. In this matched population, the mean age was 62.6 (±12.4) years and 75.2 % of the patients was male. The median follow-up time was 3.2 years. MACE rates during follow-up were similar between the two matched groups (HR = 0.84 [95 % CI 0.60 – 1.12] with p = 0.33) with cumulative event-free survival of 63.3 % in the NSTE-ACS group vs 59.3 % in the STEMI group at 6 year follow-up. Conclusion: In this retrospective study, a culprit lesion with TIMI 0 flow has similar clinical outcome in NSTE-ACS and STEMI patients. Further research is warranted to determine optimal the timing of PCI in NSTE-ACS patients with TIMI 0 flow.

Basketball is a sport that relies heavily on an athlete’s ability to rapidly decelerate in order to change direction, avoid a defender, or create space. Recent literature has proposed novel ways of measuring maximal horizontal deceleration using radar technology. The aim of this study was to investigate the relationships between different countermovement jump (CMJ) force-time characteristics and metrics related to maximal horizontal deceleration for a sample of professional male basketball players. To gain further insight into performance qualities that influence horizontal deceleration performance, athletes were separated into high- and low-performance groups for all horizontal deceleration metrics, using a median split analysis, and differences in CMJ force-time metrics were investigated between groups. The results revealed no significant correlations between any CMJ force-time metrics and horizontal deceleration performance. However, athletes’ height and body mass were correlated with different deceleration performance measures, such as average deceleration, horizontal deceleration impulse, and time to stop. Higher performing athletes with regards to average horizontal deceleration and horizontal braking impulse relative to body mass generated greater concentric power (effect size (ES) = 1.04, ES = 0.86) and concentric velocities (ES = 1.17, ES = 0.97), as well as greater jump heights (ES = 1.19, ES = 0.99). Reactive Strength Index modified values were also greater in the higher performing group for horizontal braking impulse relative to body mass (ES = 1.06). On the other hand, higher-performing athletes with regard to horizontal braking impulse generated greater eccentric deceleration force (ES = 0.81) and eccentric power values (ES = 0.88) in the CMJ. Findings may be of interest to practitioners physically preparing basketball players for the sport-specific deceleration actions they may encounter.

Streptococcus pneumoniae is a gram-positive, aerotolerant bacterium that naturally colonizes the human nasopharynx, but also causes invasive infections and is a major cause of morbidity and mortality worldwide. This pathogen produces high levels of H2O2 to eliminate other microorganisms that belong to the microbiota of the respiratory tract. However, it also induces an oxidative stress response to survive under this stressful condition. Furthermore, this self-defense mechanism is advantageous in tolerating oxidative stress imposed by the host’s immune response. This review provides a comprehensive overview of the strategies employed by the pneumococcus to survive oxidative stress. These strategies encompass the utilization of H2O2 scavengers and thioredoxins, the adaptive response to antimicrobial host oxidants, the regulation of manganese and iron homeostasis, and the intricate regulatory networks that control the stress response. Here, we have also summarized less explored aspects such as the involvement of reparation systems and polyamine metabolism. A particular emphasis is put on the role of the oxidative stress response during the transient intracellular life of Streptococcus pneumoniae, including coinfection with influenza A and the induction of antibiotic persistence in host cells.

Background Cardiac death or myocardial infarction still occurs in patients undergoing contemporary percutaneous coronary intervention (PCI). We aimed to identify adverse clinical and vessel characteristics related to hard outcomes after PCI and to investigate their individual and combined prognostic implications. BackgroundA steep rise in the use of transcatheter aortic valve implantation (TAVI) for the management of symptomatic severe aortic stenosis occurred. Minimalist TAVI procedures and streamlined patient pathways within experienced Heart Valve Centres are designed to overcome the challenges of ever-increasing procedural volume.AimsThe 2022 European TAVI Pathway Survey aims to describe contemporary TAVI practice across Europe.Materials and methodsBetween October and December 2022, TAVI operators from 32 European countries were invited to complete an online questionnaire regarding their current practice.ResultsResponses were available from 147 TAVI centres in 26 countries. In 2021, the participating centres performed a total number of 27,223 TAVI procedures, with a mean of 185 TAVI cases per centre (median 138; IQR 77–194). Treatment strategies are usually (87%) discussed at a dedicated Heart Team meeting. Transfemoral TAVI is performed with local anaesthesia only (33%), with associated conscious sedation (60%), or under general anaesthesia (7%). Primary vascular access is percutaneous transfemoral (99%) with secondary radial access (52%). After uncomplicated TAVI, patients are transferred to a high-, medium-, or low-care unit in 28%, 52%, and 20% of cases, respectively. Time to discharge is day 1 (12%), day 2 (31%), day 3 (29%), or day 4 or more (28%).ConclusionReported adoption of minimalist TAVI techniques is common among European TAVI centres, but rates of next-day discharge remain low. This survey highlights the significant progress made in refining TAVI treatment and pathways in recent years and identifies possible areas for further improvement.

Background: Recently, non-hyperemic pressure ratios (NHPRs) have been validated as a reliable alternative to fractional flow reserve (FFR). However, a discordance between FFR and NHPRs is observed in 20–25% of cases. The aim of this study is to evaluate predictors of discordance between FFR and diastolic Pressure ratio (dPR). Methods: PREDICT is a retrospective, single center, investigator-initiated study including 813 patients (1092vessels) who underwent FFR assessment of intermediate coronary lesions (angiographic 30%-80% stenosis). dPR was calculated using individual pressure waveforms and dedicated software. Clinical, angiographic and hemodynamic variables were compared between patients with concordant and discordant FFR and dPR values. Results: Median age was 65 (IQR:59–73) years and 70% were male. Hemodynamically significant lesions, as defined by FFR ≤ 0.80, and dPR ≤ 0.89, were identified in 29.6% and 30.3% of cases, respectively. Overall, FFR and dPR values were discordant in 22.1% patients (17.4% of the vessels). The countermovement vertical jump (CVJ) is one of the most commonly implemented non-invasive and time-efficient testing modalities for lower-body neuromuscular performance assessment. With more practitioners having access to portable force plates, the purpose of this study was to examine position-specific differences in CVJ force-time metrics within a cohort of elite professional male basketball athletes. Twenty-eight athletes competing in top-tier European basketball leagues volunteered to participate in the present study. Following familiarization with testing procedures and a standardized warm-up protocol, each athlete performed three maximal-effort CVJ on a uni-axial force plate system with hands on the hips during the entire movement. To minimize the possible influence of fatigue, each jump trial was separated by an approximately 15-s rest interval. The mean value across three jumps was used for performance analysis purposes. The findings of the present study reveal notable position-specific differences during the eccentric phase of the CVJ, with centers having greater braking impulse, mean force, and mean power when compared to guards. However, when normalized by body mass, the observed differences during the eccentric phase of the CVJ were nonexistent. On the other hand, no significant differences in absolute mean and peak force and power were detected during the concentric phase of the CVJ. Yet, when normalized by the player’s body mass, centers demonstrated inferior performance than guards for the same force-time metrics. Overall, these findings may help practitioners obtain a better insight into position-specific differences with regards to CVJ force-time characteristics as well as aid with individually tailored training regimen design.

Background Little is known about the impact of transcatheter mitral valve edge‐to‐edge repair on changes in left ventricular ejection fraction (LVEF) and the effect of an acute reduction in LVEF on prognosis. We aimed to assess changes in LVEF after transcatheter mitral valve edge‐to‐edge repair for both primary and secondary mitral regurgitation (PMR and SMR, respectively), identify rates and predictors of LVEF reduction, and estimate its impact on prognosis. Methods and Results In this international multicenter registry, patients with both PMR and SMR undergoing transcatheter mitral valve edge‐to‐edge repair were included. We assessed rates of acute LVEF reduction (LVEFR), defined as an acute relative decrease of >15% in LVEF, its impact on all‐cause mortality, major adverse cardiac event (composite end point of all‐cause death, mitral valve surgery, and residual mitral regurgitation grade ≥2), and LVEF at 12 months, as well as predictors for LVEFR. Knowing cardiac physiology is essential for health care professionals working in the cardiovascular field. Pressure–volume loops (PVLs) offer a unique understanding of the myocardial working and have become pivotal in complex pathophysiological scenarios, such as profound cardiogenic shock or when mechanical circulatory supports are implemented. This review provides a comprehensive summary of the left and right ventricle physiology, based on the PVL interpretation.

The strength of the affective priming effect is influenced by various factors, including the duration of the prime. Surprisingly, short-duration primes that are around the threshold for conscious awareness typically result in stronger effects compared to long-duration primes. The misattribution effect theory suggest that subliminal primes do not provide sufficient cognitive processing time for the affective feeling to be attributed to the prime. Instead, the neutral target being evaluated is credited for the affective experience. In everyday social interactions, we shift our gaze from one face to another, typically contemplating each face for only a few seconds. It is reasonable to assume that no affective priming takes place during such interactions. To investigate whether this is indeed the case, participants were asked to rate the valence of faces displayed one by one. Each face image simultaneously served as both a target (primed by the previous trial) and a prime (for the next trial). Depending on the participant’s response time, images were typically displayed for about 1–2 s. As predicted by the misattribution effect theory, neutral targets were not affected by positive affective priming. However, non-neutral targets showed a robust priming effect, with emotional faces being perceived as even more negative or positive when the previously seen face was emotionally congruent. These results suggest that a “correct attribution effect” modulates how we perceive faces, continuously impacting our social interactions. Given the importance of faces in social communication, these findings have wide-ranging implications.

Introduction Sedentary work is associated with poor health outcomes. Many healthcare occupations, including office-based care coordination, are largely sedentary. Many nurses do not achieve the recommended levels of daily physical activity, however, the physical activity levels among nurses working in care coordination are not known. Objective To assess the physical activity levels, self-reported health and well-being, overall quality of life, and work-related satisfaction of office-based care coordinators, and compare these among nursing and non-nursing staff. Methods This study collected objective physical activity data using accelerometry along with self-reported information on work-related quality of life and satisfaction from 42 healthcare staff working in a hospital-affiliated office-based care coordination program. Results were compared among nursing and non-nursing staff. Results Nurses had lower moderate-to-vigorous physical activity levels compared to non-nursing staff (25 min/day vs. 45, p = .007). There were no differences in daily sedentary time, light activity, or steps between nursing and non-nursing staff. Nurses reported high quality of life scores compared to non-nursing staff (4.4 vs. 4.1, p = .02), but similar levels of work-related quality of life, happiness, self-rated health, and well-being. Conclusion Nurses working in an office-based care coordination program had lower levels of physical activity but reported a higher overall quality of life than non-nurse work colleagues working in a similar environment. Given known health risks associated with sedentary occupational work and the growing number of care coordination programs, health policies and initiatives aimed at increasing the physical activity levels of care coordination workers is of prime importance.

While various quantifiable physical attributes have been found to contribute to athletes’ performance, there is a lack of scientific literature focused on examining how they relate to success during competition performance. The aim of this study was to investigate different countermovement jump (CMJ)-derived force–time characteristics and their utility in distinguishing high from low performers within a measure of on-court contribution (i.e., minutes per game played). Twenty-nine collegiate athletes (n = 15 males and n = 14 females) volunteered to participate in this investigation and performed CMJs on dual force plates sampling at 1000 Hz, weekly over the course of their basketball season. The athletes’ average of their three best test-days across the season was used for further analysis. To identify their on-court contribution, athletes were divided into groups with high and low minutes per game, based on a median-split analysis. The findings suggest that at the overall group level (i.e., both genders), the modified reactive strength index (mRSI) and braking rate of force development (RFD) revealed the greatest between-group magnitudes of difference, with athletes playing more minutes per game showing greater performance. At the team-specific level, the braking RFD, average braking velocity, and mRSI were shown to be the greatest differentiators between groups for the men’s team. The women’s high-minutes group displayed greater magnitudes of mRSI and jump height. By identifying the neuromuscular qualities seen in top performers within their respective populations, the attributed physical performance underpinning these qualities may be identified, providing practitioners with insights into physical performance qualities and training methodologies that have the potential to influence basketball performance.

Although different strategies have been implemented to manage recovery-fatigue status in athletes, there is still a lack of consensus on which recovery protocols have the greatest impact and effectiveness when implemented with basketball players, including both physiological and psychological recovery methods. Thus, the purpose of this systematic review is to: (a) determine which recovery methods attain the greatest benefit in restoring the process of attenuating fatigue and (b) provide sports practitioners with guidelines on how some of the most effective recovery strategies can be used to optimize athletes’ recovery and ultimately enhance their performance. Using the PRISMA guidelines, a total of 3931 research reports were obtained through four database searches (i.e., PubMed, Scopus, Cochrane, and Web of Science), from which only 25 met the inclusion and exclusion criteria. The recovery protocols analyzed in this systematic review were: sleep, nutrition, hydration, ergogenic aids, cold-water immersion, compression garments, massage, acupuncture, tapering, mindfulness, and red-light irradiation. The results revealed that all recovery strategies are capable of attenuating fatigue and enhancing recovery in basketball players to a certain degree. However, an individualized approach should be promoted, where a combination of proactive recovery modalities appears to result in the most rapid rates of recovery and athletes’ ability to maintain high-level performance. Recovery should be programmed as an integral component of training regimens. Also, cooperation and communication between coaches, players, and the rest of the team staff members are essential in minimizing the risk of non-functional overreaching or injury and optimizing basketball players’ on-court performance.

OncoboxPD (Oncobox pathway databank) available at https://open.oncobox.com is the collection of 51 672 uniformly processed human molecular pathways. Superposition of all pathways formed interactome graph of protein–protein interactions and metabolic reactions containing 361 654 interactions and 64 095 molecular participants. Pathways are uniformly classified by biological processes, and each pathway node is algorithmically functionally annotated by specific activator/repressor role. This enables online calculation of statistically supported pathway activation levels (PALs) with the built-in bioinformatic tool using custom RNA/protein expression profiles. Each pathway can be visualized as static or dynamic graph, where vertices are molecules participating in a pathway and edges are interactions or reactions between them. Differentially expressed nodes in a pathway can be visualized in two-color mode with user-defined color scale. For every comparison, OncoboxPD also generates a graph summarizing top up- and downregulated pathways.

Background: Aortic valve calcification correlates with the severity of aortic valve stenosis and a high calcium score is associated with conduction disturbances and paravalvular leakage after transcatheter aortic valve replacement. The 3mensio Structural Heart is a semiautomated software package to facilitate aortic root analysis by multislice computed tomography.The aim of the contemporary study is to validate a semiautomated calcium quantification scoring tool with a conventional manual calcium quantification tool. Methods: Fifty randomly selected patients who underwent multislice computed tomography for preprocedural planning were retrospectively selected to compare the semiautomated aortic valve Agatston calcium score by 3mensio with the manually obtained score using IntelliSpace Portal as standard reference. Results: Patients had a mean age of 76.7 ± 7.4 years and 60% were male. The median Agatston score was 3390 [interquartile range 1877-4509] with 3mensio and 3434 [interquartile range 1839-4620] with IntelliSpace.The mean difference was -0.18 [95% confidence interval (CI) −53.8 to 53.4]. The intraclass correlation coefficient between the Agatston scores using IntelliSpace and 3mensio showed an excellent correlation of 0.995 [95% CI 0.992-0.997], p ≤ 0.001. Youth with Down syndrome (DS) have a higher prevalence of overweight and obesity compared to the general youth population. Due to physiological and cognitive differences observed in youth with DS, weight management recommendations developed for the general population, may not be suitable for youth with DS. However, there are no recent recommendations for weight management in youth with DS. A workgroup of clinicians and researchers with extensive experience working with youth with DS came together to give clinicians and families guidance for weight management in youth with DS. Recommendations were developed by the workgroup via a methodical, deliberative process. After the initial development of the recommendations, they were shared with an expert review panel and caregivers who rated the strength of the recommendation and strength of the evidence. The workgroup moved forward the recommendations which the review panels rated as strong. Eight recommendations were developed which focused on screening for overweight and obesity, screening for health conditions that may impact dietary intake and physical activity, screening for feeding difficulties, setting appropriate recommendations for dietary intake and physical activity, and well as prevention and treatment of overweight and obesity using evidence-based strategies. These recommendations can be implemented by clinicians working with youth with Down syndrome as well as the family, school, and other relevant entities.
