Abstract P2X receptors are a family of seven trimeric non-selective cation channels that are activated by extracellular ATP to play roles in the cardiovascular, neuronal, and immune systems. Although it is known that the P2X1 receptor subtype has increased sensitivity to ATP and fast desensitization kinetics, an underlying molecular explanation for these subtype-selective features is lacking. Here we report high-resolution cryo-EM structures of the human P2X1 receptor in the apo closed, ATP-bound desensitized, and the high-affinity antagonist NF449-bound inhibited states. The apo closed and ATP-bound desensitized state structures of human P2X1 define subtype-specific properties such as distinct pore architecture and ATP-interacting residues. The NF449-bound inhibited state structure of human P2X1 reveals that NF449 has a unique dual-ligand supramolecular binding mode at the interface of neighboring protomers, inhibiting channel activation by overlapping with the canonical P2X receptor ATP-binding site. Altogether, these data define the molecular pharmacology of the human P2X1 receptor laying the foundation for structure-based drug design.

Urban environments worldwide face toxic heavy metal pollution originating from industrial discharge, municipal waste disposal, vehicular emissions, and atmospheric deposition. Kazakhstan, experiencing accelerated economic growth and extensive mining activities, contends with widespread heavy metal contamination in its soil-plant-air-water ecosystems. This study explores the potential of hyperaccumulating plants for phytoremediation in urban soils of Kazakhstan contaminated with Pb, Cd, and Co. Twelve plant species, including Korean Mint (Lamiaceae), Ornamental Cabbage (Brassica oleracea), Ageratum (Ageratum houstonianum), Coneflower (Echinacea purpurea), Amaranth (Amaranthus Perfect and Amaranthus Emerald), Fescue (Festuca glauca), Burning Bush (Kochia scoparia), Marigold (Tagetes patula nana), White Cabbage (Brassica-Cavolo cappuccino BIANKO), Tepary Bean (Phaseolus acutifolius), and Rapeseed (Brassica napus), were evaluated for growth and biomass production in urban soils spiked with two maximum permissible addition (MPA) treatments of Pb, Co, and Cd. The selected plants demonstrated varied responses to heavy metal stress, with Marigold (8.4 g shoot biomass/plant), Korean mint (10.5 g shoot biomass/plant), Rapeseed (19.9 g/shoot biomass), and Tepary bean (25.9 g shoot biomass/plant) exhibiting resilience or tolerance to Pb, Co, and Cd stresses. The results highlight the significant potential of these plants for efficient phytoremediation, showcasing their unique abilities to absorb and accumulate specific metals. Marigold, particularly, displayed noteworthy Pb accumulation (40.3 mg/kg biomass), resulting in reduced residual Pb concentrations in the soil (74.7 mg/kg). Conversely, White cabbage and Amaranth showed limited efficiency in Cd extraction, while Rapeseed and Tepary bean emerged as promising candidates for Cd phytoremediation. This study emphasizes the critical role of tailored plant species selection in designing effective phytoremediation strategies for specific metal-contaminated urban sites. A comprehensive understanding of the dynamics of metal accumulation and residual concentrations is crucial for the development of sustainable and efficient environmental remediation approaches. Further research is warranted to explore the long-term effects of different plant species on soil metal concentrations, refining and optimizing phytoremediation methods for urban soils grappling with toxic heavy metal contamination

Abstract Global climate‐driven ocean warming has decreased dissolved oxygen (DO) levels (ocean deoxygenation) leading to expansions of hypoxic zones, which will affect the movements, behaviour, physiology and distributions of marine animals. However, the precise responses of animals to low DO remains poorly understood because movements and activity levels are seldom recorded alongside instantaneous DO in situ. We describe a new animal‐attached (dissolved oxygen measuring, DOME) archival tag with an optical oxygen sensor for recording DO, in addition to sensors for temperature and depth, a triaxial accelerometer for fine‐scale movements and activity, and a GPS for tag recovery. All sensors were integrated on a single electronic board. Calibration tests demonstrated small mean difference between DOME tag and factory‐calibrated DO sensors (mean relative error of 5%). No temporal drift occurred over a test period three times longer than the maximum deployment time. Deployments on four blue sharks (Prionace glauca) in the central North Atlantic Ocean showed regular vertical oscillations from the surface to a maximum of 404 m. Profiles from diving sharks recorded DO concentrations ranging from 217 to 272 μmol L−1, temperatures between 13°C and 23°C, and identified an oxygen maximum at ~45 m depth, all of which were consistent with ship‐based measurements. Interestingly, the percentage of time sharks spent burst swimming was greater in the top 85 m compared to deeper depths, potentially because of higher prey availability in the surface layer. The DOME tag described blue shark fine‐scale movements and activity levels in relation to accurately measured in situ DO and temperature, with the potential to offer new insights of animal performance in low oxygen environments. Development of a tag with physico‐chemical and movement sensors on a single electronic board is a first step towards satellite relay of these data over broader spatiotemporal scales (months over thousands of kilometres) to determine direct and indirect responses of marine animals to heatwave and deoxygenation events.

Abstract P2X receptors are trimeric ATP-gated ion channels that activate diverse signaling cascades. Due to its role in apoptotic pathways, selective activation of P2X7 is a potential experimental tool and therapeutic approach in cancer biology. However, mechanisms of high-affinity P2X7 activation have not been defined. We report high-resolution cryo-EM structures of wild-type rat P2X7 bound to the high-affinity agonist BzATP as well as significantly improved apo receptor structures in the presence and absence of sodium. Apo structures define molecular details of pore architecture and reveal how a partially hydrated Na+ ion interacts with the conductance pathway in the closed state. Structural, electrophysiological, and direct binding data of BzATP reveal that three residues just outside the orthosteric ATP-binding site are responsible for its high-affinity agonism. This work provides insights into high-affinity agonism for any P2X receptor and lays the groundwork for development of subtype-specific agonists applicable to cancer therapeutics.

Summary: Zika virus (ZIKV) is a neurotropic flavivirus that can persist in several tissues. The late consequences of ZIKV persistence and whether new rounds of active replication can occur, remain unaddressed. Here, we investigated whether neonatally ZIKV-infected mice are susceptible to viral reactivation in adulthood. We found that when ZIKV-infected mice are treated with immunosuppressant drugs, they present increased susceptibility to chemically induced seizures. Levels of subgenomic flavivirus RNAs (sfRNAs) were increased, relative to the amounts of genomic RNAs, in the brains of mice following immunosuppression and were associated with changes in cytokine expression. We investigated the impact of immunosuppression on the testicles and found that ZIKV genomic RNA levels are increased in mice following immunosuppression, which also caused significant testicular damage. These findings suggest that ZIKV can establish new rounds of active replication long after acute stages of disease, so exposed patients should be monitored to ensure complete viral eradication.

Field level data from a voluntary water use reporting program in the Delta region of Mississippi (MS), USA, provides empirical evidence that crop growers increase the amount of groundwater pumped for irrigation during periods of high temperature (degree days above 32 C). Regression analysis reveals that growers apply excess irrigation to cope with high temperature conditions while growing season evapotranspiration and precipitation are not significant factors in their decision of how much irrigation to apply. The existing literature indicates that excess water does not alleviate the harmful effect of extreme heat, making the excess irrigation wasteful and costly. Additional degree days above 32 C are associated with a 4.6 mm increase in irrigation depth (46 m3ha−1). This result may be partially explained by the fact that most farmers use visual cues (such as leaf folding which correlate with air temperature) to initiate irrigation events regardless of soil moisture contents. To quantify the impact of this mismatch, we estimate that shifting the underlying sensitivity in the farmers’ minds by 1 C would reduce groundwater pumpage by 800862 mega liters across the Delta region of Mississippi, USA. The monetary cost of mismanaging heat stress with excessive irrigation is estimated at a minimum of over US$33 million per year. We further identify two additional possible distortions affecting the demand for irrigation groundwater: (i) overestimated benefits from irrigation; and (ii) underestimated groundwater pumping cost. The policy implication is that more efforts and incentives should be placed for programs that affect the producers’ water management mindset, such as ''Master Irrigator'' programs that will reduce or eliminate the identified groundwater distortions and optimize benefits from irrigation.

This article deals with a comprehensive analysis and performance evaluation of a fully integrated two-phase cooling system for power converters. A suitable test bed has been properly manufactured to perform the experimental campaign for the evaluation of the benefits and to deploy a dedicated management procedure of the two-phase cooling. The system is tested under real operating conditions, in which the system is employed for cooling a 1200-V 100-A insulated-gate bipolar transistor (IGBT) power module of an inverter controlling a permanent magnet synchronous machine. The experimental results show the inverter temperature behavior in start-up for different flow rates of coolant and different temperatures of the fluid on the secondary system. In the steady-state operation, using lower coolant flow rates in the primary circuit achieved equal or even better cooling for the IGBT module compared with higher flow rates. With respect to traditional cooling approaches, the proposed arrangement allows a greater extraction of the heat at a very low flow rate of the cooling fluid, even with standard industrial grade heat sinks, which motivates the use of this cooling technology for the next generation of power electronics converters.

Abstract Butyrate‐producing bacteria are found in many outdoor ecosystems and host organisms, including humans, and are vital to ecosystem functionality and human health. These bacteria ferment organic matter, producing the short‐chain fatty acid butyrate. However, the macroecological influences on their biogeographical distribution remain poorly resolved. Here we aimed to characterise their global distribution together with key explanatory climatic, geographical and physicochemical variables. We developed new normalised butyrate production capacity (BPC) indices derived from global metagenomic (n = 13,078) and Australia‐wide soil 16S rRNA (n = 1331) data, using Geographic Information System (GIS) and modelling techniques to detail their ecological and biogeographical associations. The highest median BPC scores were found in anoxic and fermentative environments, including the human (BPC = 2.99) and non‐human animal gut (BPC = 2.91), and in some plant–soil systems (BPC = 2.33). Within plant–soil systems, roots (BPC = 2.50) and rhizospheres (BPC = 2.34) had the highest median BPC scores. Among soil samples, geographical and climatic variables had the strongest overall effects on BPC scores (variable importance score range = 0.30–0.03), with human population density also making a notable contribution (variable importance score = 0.20). Higher BPC scores were in soils from seasonally productive sandy rangelands, temperate rural residential areas and sites with moderate‐to‐high soil iron concentrations. Abundances of butyrate‐producing bacteria in outdoor soils followed complex ecological patterns influenced by geography, climate, soil chemistry and hydrological fluctuations. These new macroecological insights further our understanding of the ecological patterns of outdoor butyrate‐producing bacteria, with implications for emerging microbially focused ecological and human health policies.

SN 2023ixf was discovered in M101 within a day of the explosion and rapidly classified as a Type II supernova with flash features. Here we present ultraviolet (UV) spectra obtained with the Hubble Space Telescope 14, 19, 24, and 66 days after the explosion. Interaction between the supernova ejecta and circumstellar material (CSM) is seen in the UV throughout our observations in the flux of the first three epochs and asymmetric Mg ii emission on day 66. We compare our observations to CMFGEN supernova models that include CSM interaction ( $\dot{M}\lt {10}^{-3}$ M _⊙ yr ^−1 ) and find that the power from CSM interaction is decreasing with time, from L _sh ≈ 5 × 10 ^42 erg s ^−1 to L _sh ≈ 1 × 10 ^40 erg s ^−1 between days 14 and 66. We examine the contribution of individual atomic species to the spectra on days 14 and 19, showing that the majority of the features are dominated by iron, nickel, magnesium, and chromium absorption in the ejecta. The UV spectral energy distribution of SN 2023ixf sits between that of supernovae, which show no definitive signs of CSM interaction, and those with persistent signatures assuming the same progenitor radius and metallicity. Finally, we show that the evolution and asymmetric shape of the Mg ii λ λ 2796, 2802 emission are not unique to SN 2023ixf. These observations add to the early measurements of dense, confined CSM interaction, tracing the mass-loss history of SN 2023ixf to ∼33 yr prior to the explosion and the density profile to a radius of ∼5.7 × 10 ^15 cm. They show the relatively short evolution from a quiescent red supergiant wind to high mass loss.

Abstract Background There is an expanding unregulated market for a psychotropic compound called ∆8-Tetrahydrocannabinol (delta-8-THC) that is being derived from hemp, but a summary of adverse events related to delta-8-THC has not been publicly reported. Methods This case series assessed adverse events reported by delta-8-THC users on the Reddit forum r/Delta8 and compared these to delta-8-THC AEs in the US Food and Drug Administration Adverse Event Reporting System (FAERS). Delta-8-THC and cannabis AEs reported in FAERS were also compared. The r/Delta8 forum was selected because it includes a large sample of 98,700 registered individuals who publicly discuss their experiences using delta-8-THC. All r/Delta8 posts were obtained from August 20, 2020, through September 25, 2022. A random sample of r/Delta8 posts was drawn (n = 10,000) and filtered for posts in which delta-8-THC users reported an adverse event (n = 335). FAERS reports that listed delta-8-THC (N = 326) or cannabis (N = 7076) as a suspect product active ingredient were obtained. Adverse events claimed to result from delta-8-THC use were coded using Medical Dictionary for Regulatory Activities to system organ class and preferred term categories. Results The absolute number of delta-8-THC adverse event reports (N = 2184, 95% CI = 1949–2426) and serious adverse event reports (N = 437; 95% CI = 339–541) on r/Delta 8 were higher than the adverse event reports (N = 326) and serious adverse event reports (N = 289) to FAERS. Psychiatric disorders were the most frequently cited system organ class in r/Delta8 adverse event reports, mentioned in 41.2% (95% CI = 35.8%-46.3%) of reports, followed by respiratory, thoracic and mediastinal disorders (29.3%, 95% CI = 25.1%-34.0%) and nervous system disorders (23.3%, 95% CI = 18.5%-27.5%). Anxiety (16.4%, 95% CI = 12.8–20.6), Cough (15.5%, 95% CI = 11.9–20.0) and Paranoia (9.3%, 95% CI = 6.3–12.5) were the most frequently cited preferred terms in adverse event reports. The overall prevalence of AEs reported for cannabis and delta-8-THC on FAERS were also similar when analyzed by system organ class (Pearson’s r = 0.88). Conclusions The findings of this case series suggest that most of the adverse events reported by delta-8-THC users are like those reported during acute cannabis intoxication. This finding suggests that health care professionals follow similar treatment and management protocols, and that jurisdictions should clarify whether delta-8-THC can be sold as a hemp product.

Plastic pollution is a pressing global issue, necessitating a focus on consumer behavior to curb this problem at its source. To effectively promote sustainable practices, communication strategies that employ future visions have gained attention. This study examines the effects of a narrative video intervention depicting an optimistic future vision concerning single-use plastic bag consumption in South Africa, compared to a representation of the prevailing status quo. Using a preregistered within-subject design, we assess the psychological and emotional responses to two scenarios of which one is illustrating adaptive behaviors toward reduced plastic bag usage, and the other showcasing current consumption patterns. Parametric analyses revealed a shift in emotional states, characterized by a greater experience of positive emotions and a reduced experience of negative emotions following the exposure to the optimistic future scenario video, as compared to the status quo video. Moreover, engagement with the optimistic future scenario was associated with higher levels of perceived behavioral control and behavioral intentions. No significant changes were found regarding sense of responsibility. These findings point to the potential of optimistic future visions to influence individuals at psychological and emotional levels. This renders optimistic future vision communication as an effective tool for sustainable behavior change, particularly in relation to the sustainable use of plastic shopping bags.

Abstract Increased soybean (Glycine max L. Merril) seed costs have motivated interest in reduced seeding rates to improve profitability while maintaining or increasing yield. However, little is known about the effect of early-season plant-to-plant spatial uniformity on the yield of modern soybean varieties planted at reduced seeding rates. The objectives of this study were to (i) investigate traditional and devise new metrics for characterizing early-season plant-to-plant spatial uniformity, (ii) identify the best metrics correlating plant-to-plant spatial uniformity and soybean yield, and (iii) evaluate those metrics at different seeding rate (and achieved plant density) levels and yield environments. Soybean trials planted in 2019 and 2020 compared seeding rates of 160, 215, 270, and 321 thousand seeds ha−1 planted with two different planters, Max Emerge and Exact Emerge, in rainfed and irrigated conditions in the United States (US). In addition, trials comparing seeding rates of 100, 230, 360, and 550 thousand seeds ha−1 were conducted in Argentina (Arg) in 2019 and 2020. Achieved plant density, grain yield, and early-season plant-to-plant spacing (and calculated metrics) were measured in all trials. All site-years were separated into low- (2.7 Mg ha−1), medium- (3 Mg ha−1), and high- (4.3 Mg ha−1) yielding environments, and the tested seeding rates were separated into low ( 300 seeds m−2) levels. Out of the 13 metrics of spatial uniformity, standard deviation (sd) of spacing and of achieved versus targeted evenness index (herein termed as ATEI, observed to theoretical ratio of plant spacing) showed the greatest correlation with soybean yield in US trials (R2 = 0.26 and 0.32, respectively). However, only the ATEI sd, with increases denoting less uniform spacing, exhibited a consistent relationship with yield in both US and Arg trials. The effect of spatial uniformity (ATEI sd) on soybean yield differed by yield environment. Increases in ATEI sd (values > 1) negatively impacted soybean yields in both low- and medium-yield environments, and in achieved plant densities below 200 thousand plants ha−1. High-yielding environments were unaffected by variations in spatial uniformity and plant density levels. Our study provides new insights into the effect of early-season plant-to-plant spatial uniformity on soybean yields, as influenced by yield environments and reduced plant densities.

The 2015 Paris Agreement requires all nations to combat climate change and to adapt to its effects. Countries promise to reduce their greenhouse gas (GHG) emissions through their Nationally Determined Contributions. Pledges to reduce emissions, however, have implications for economic growth. We estimate the link between economic growth and CO2 pollution levels and find that this relationship is highly non-linear. A country's GHG emissions rise rapidly as its economic activity rises, relative to global activity, meaning that fast-growing countries contribute most heavily to current GHG emissions. Then, using real per-capita GDP as our metric, we estimate how much the carbon price should be in order to remove the economic growth benefit from excess GHG emissions. We find that the implied prices are far higher than the prices on any existing market for emissions as well as estimates of the social cost of carbon. Our findings also have important implications for the global dialogue regarding responsibility for climate mitigation as well as for the choice of policies to support mitigation efforts.

We present photometric and spectroscopic observations of SN 2023fyq, a Type Ibn supernova (SN) in the nearby galaxy NGC 4388 ( D ≃ 18 Mpc). In addition, we trace the 3 yr long precursor emission at the position of SN 2023fyq using data from DLT40, ATLAS, Zwicky Transient Facility, ASAS-SN, Swift, and amateur astronomer Koichi Itagaki. The double-peaked postexplosion light curve reaches a luminosity of ∼10 ^43 erg s ^−1 . The strong intermediate-width He lines observed in the nebular spectrum imply the interaction is still active at late phases. We found that the precursor activity in SN 2023fyq is best explained by the mass transfer in a binary system involving a low-mass He star and a compact companion. An equatorial disk is likely formed in this process (∼0.6 M _⊙ ), and the interaction of SN ejecta with this disk powers the second peak of the SN. The early SN light curve reveals the presence of dense extended material (∼0.3 M _⊙ ) at ∼3000 R _⊙ ejected weeks before the SN explosion, likely due to final-stage core silicon burning or runaway mass transfer resulting from binary orbital shrinking, leading to rapid-rising precursor emission within ∼30 days prior to explosion. The final explosion could be triggered either by the core collapse of the He star or by the merger of the He star with a compact object. SN 2023fyq, along with SN 2018gjx and SN 2015G, forms a unique class of Type Ibn SNe, which originate in binary systems and are likely to exhibit detectable long-lasting pre-explosion outbursts with magnitudes ranging from −10 to −13.

Understanding that design involves trade-offs, thinking at multiple levels of abstraction, and considering the cohesion and coupling between sub-components of a larger whole is an important part of software (and other) engineering. It can be challenging to convey such abstract design concepts to novice engineers, especially for materials that are themselves abstract (e.g. software). Such challenges are compounded when teaching at the secondary school stage where students have limited experience of large-scale design problems that motivate the need for abstraction at all. In this paper, we describe a method for introducing these concepts to secondary school students using LEGO® and Raspberry Pi computers, asking them to build musical instruments as an entertaining way of motivating engagement with learning about design through play. The method has been successfully piloted in a series of three classroom sessions and key observations and experiences of using the method are presented.

Single-use plastic bags contribute significantly to the plastic pollution in South Africa posing a serious environmental challenge. Hence, there is a need for comprehensive strategies to reduce plastic bag consumption. We report the results of an online workshop aimed to create a positive, shared future vision. The workshop was attended by 12 invited participants from diverse backgrounds and disciplines and utilized a collaborative approach to propose a framework for reducing plastic bag usage during grocery shopping in South Africa. The findings suggest that a combination of economic, structural and behavioral interventions is necessary to promote lasting behavior change, including financial incentives, a broad introduction of more environmentally friendly alternatives, and promoting bag reuse. Additionally, the vision underscored the importance of using social channels such as education and social norms. The invited stakeholders did not endorse conventional approaches such as increasing an already existing levy or implementing a complete ban on plastic bags. They also questioned the introduction of fully recycled plastic bags. Instead, the proposed vision encourages an integrated approach to reducing plastic bag usage involving individual behavior change and community engagement. The development of a creative product such as a video to showcase a future vision offers a powerful tool for communication with further stakeholders and political authorities, to engage with the community and broader population and for inspiring collective action. Overall, the findings have implications for policymakers, business leaders, environmental educators, and individuals committed to reducing plastic pollution and promoting sustainable practices in South Africa.

The rate and extent of anthropogenic alteration of the global nitrogen cycle over the past four decades has been extensive, resulting in cascading negative impacts on riverine and coastal water quality. In this paper, we investigate the individual effects of a set of management, technology, and policy mechanisms that alter total reactive nitrogen (TN) flux through rivers, using a modified, spatially detailed SPARROW TN model, between 1980 and 2019 in the Northeast (NE) and Midwest (MW) of the United States. Using the recalibrated model, we simulate and validate a historical baseline, to which we compare a set of climate and non-climate single factor experiments (SFEs) in which individual factors are held at 1980s levels while all other factors change dynamically. We evaluate SFE performance in terms of differences in TN flux and willingness to pay. The largest effect on TN flux are related to reduction in cropland area and atmospheric nitrogen deposition. Multi-factor experiments (MFEs) suggest that increasingly efficient corn cultivars had a larger influence than increasing fertilizer application rate, while population growth has a larger influence than wastewater treatment. Extreme climate SFEs suggest that persistent wet conditions increase TN flux throughout the study region. Meanwhile, persistent hot years result in reduced TN flux. The persistent dry climate SFE leads to increased TN flux in the NE and reduced TN flux in the MW. We find that the potential for TN removal through aquatic decay is greatest in MW, due to the role of long travel time of rivers draining into the Lower Mississippi River. This paper sheds light on how a geographically and climatologically diverse region would respond to a representative selection of management options.

The continuous generation and accumulation of solid waste is a critical problem at the world level and require urgent action. Therefore, the present paper contains an alternative of using two solid plastic wastes, determining the use of polymeric synthesis made from recycled polypropylene (PP) and expanded polystyrene (EPS). First, the research developed a new material using these two recycled raw materials and evaluated its physical-mechanical properties. The next step was the comparison of the properties with different potentially replaceable materials. Finally, polyvinyl chloride (PVC) was the material identified as possible to be replaced by the mixture (EPS+PP) because of the similar values of its properties, with specific use for coating electrical installations and internal communications. The study uses the Life Cycle Assessment methodology (LCA) to evaluate the material's environmental performance. This paper reports the inventory data of the new material process validated in a plastic plant in Bogotá (Colombia). Consequently, by using the mass and energy balances, the authors evaluate the environmental indicators for the process proposed and then compare the results with the ones of PVC in the Ecoinvent databases. The study used a "cradle to door" approach and SIMAPRO software in this step. The LCA results show that the new material generates less environmental impact than conventional (PVC). Furthermore, the present research results allow new possibilities for incorporating circular economy models in the construction sector. As a result, the new material produces a lower environmental impact than PVC.

We analyze pre-explosion near- and mid-infrared (IR) imaging of the site of SN 2023ixf in the nearby spiral galaxy M101 and characterize the candidate progenitor star. The star displays compelling evidence of variability with a possible period of ≈1000 days and an amplitude of Δ m ≈ 0.6 mag in extensive monitoring with the Spitzer Space Telescope since 2004, likely indicative of radial pulsations. Variability consistent with this period is also seen in the near-IR J and K _s bands between 2010 and 2023, up to just 10 days before the explosion. Beyond the periodic variability, we do not find evidence for any IR-bright pre-supernova outbursts in this time period. The IR brightness ( ${M}_{{K}_{s}}=-10.7$ mag) and color ( J − K _s = 1.6 mag) of the star suggest a luminous and dusty red supergiant. Modeling of the phase-averaged spectral energy distribution (SED) yields constraints on the stellar temperature ( ${T}_{\mathrm{eff}}={3500}_{-1400}^{+800}$ K) and luminosity ( $\mathrm{log}L/{L}_{\odot }=5.1\pm 0.2$ ). This places the candidate among the most luminous Type II supernova progenitors with direct imaging constraints, with the caveat that many of these rely only on optical measurements. Comparison with stellar evolution models gives an initial mass of M _init = 17 ± 4 M _⊙ . We estimate the pre-supernova mass-loss rate of the star between 3 and 19 yr before explosion from the SED modeling at $\dot{M}\approx 3\times {10}^{-5}$ to 3 × 10 ^−4 M _⊙ yr ^−1 for an assumed wind velocity of v _w = 10 km s ^−1 , perhaps pointing to enhanced mass loss in a pulsation-driven wind.

We present very early photometric and spectroscopic observations of the Type Ia supernova (SN Ia) 2023bee, starting about 8 hr after the explosion, which reveal a strong excess in the optical and nearest UV ( U and UVW1 ) bands during the first several days of explosion. This data set allows us to probe the nature of the binary companion of the exploding white dwarf and the conditions leading to its ignition. We find a good match to the Kasen model in which a main-sequence companion star stings the ejecta with a shock as they buzz past. Models of double detonations, shells of radioactive nickel near the surface, interaction with circumstellar material, and pulsational delayed detonations do not provide good matches to our light curves. We also observe signatures of unburned material, in the form of carbon absorption, in our earliest spectra. Our radio nondetections place a limit on the mass-loss rate from the putative companion that rules out a red giant but allows a main-sequence star. We discuss our results in the context of other similar SNe Ia in the literature.

We present five far- and near-ultraviolet spectra of the Type II plateau supernova, SN 2022acko, obtained 5, 6, 7, 19, and 21 days after explosion, all observed with the Hubble Space Telescope/Space Telescope Imaging Spectrograph. The first three epochs are earlier than any Type II plateau supernova has been observed in the far-ultraviolet revealing unprecedented characteristics. These three spectra are dominated by strong lines, primarily from metals, which contrasts with the featureless early optical spectra. The flux decreases over the initial time series as the ejecta cool and line blanketing takes effect. We model this unique data set with the non–local thermodynamic equilibrium radiation transport code CMFGEN , finding a good match to the explosion of a low-mass red supergiant with energy E _kin = 6 × 10 ^50 erg. With these models we identify, for the first time, the ions that dominate the early ultraviolet spectra. We present optical photometry and spectroscopy, showing that SN 2022acko has a peak absolute magnitude of V = − 15.4 mag and plateau length of ∼115 days. The spectra closely resemble those of SN 2005cs and SN 2012A. Using the combined optical and ultraviolet spectra, we report the fraction of flux as a function of bluest wavelength on days 5, 7, and 19. We create a spectral time-series of Type II supernovae in the ultraviolet, demonstrating the rapid decline of flux over the first few weeks of evolution. Future observations of Type II supernovae are required to map out the landscape of exploding red supergiants, with and without circumstellar material, which is best revealed in high-quality ultraviolet spectra.

We present the optical spectroscopic evolution of SN 2023ixf seen in subnight cadence spectra from 1.18 to 15 days after explosion. We identify high-ionization emission features, signatures of interaction with material surrounding the progenitor star, that fade over the first 7 days, with rapid evolution between spectra observed within the same night. We compare the emission lines present and their relative strength to those of other supernovae with early interaction, finding a close match to SN 2020pni and SN 2017ahn in the first spectrum and SN 2014G at later epochs. To physically interpret our observations, we compare them to CMFGEN models with confined, dense circumstellar material around a red supergiant (RSG) progenitor from the literature. We find that very few models reproduce the blended N iii ( λλ 4634.0,4640.6)/C iii ( λλ 4647.5,4650.0) emission lines observed in the first few spectra and their rapid disappearance thereafter, making this a unique diagnostic. From the best models, we find a mass-loss rate of 10 ^−3 –10 ^−2 M _⊙ yr ^−1 , which far exceeds the mass-loss rate for any steady wind, especially for an RSG in the initial mass range of the detected progenitor. These mass-loss rates are, however, similar to rates inferred for other supernovae with early circumstellar interaction. Using the phase when the narrow emission features disappear, we calculate an outer dense radius of circumstellar material R _CSM,out ≈ 5 × 10 ^14 cm, and a mean circumstellar material density of ρ = 5.6 × 10 ^−14 g cm ^−3 . This is consistent with the lower limit on the outer radius of the circumstellar material we calculate from the peak H α emission flux, R _CSM,out ≳ 9 × 10 ^13 cm.

We present the densely sampled early light curve of the Type II supernova (SN) 2023ixf, first observed within hours of explosion in the nearby Pinwheel Galaxy (Messier 101; 6.7 Mpc). Comparing these data to recently updated models of shock-cooling emission, we find that the progenitor likely had a radius of 410 ± 10 R _⊙ . Our estimate is model dependent but consistent with a red supergiant. These models provide a good fit to the data starting about 1 day after the explosion, despite the fact that the classification spectrum shows signatures of circumstellar material around SN 2023ixf during that time. Photometry during the first day after the explosion, provided almost entirely by amateur astronomers, does not agree with the shock-cooling models or a simple power-law rise fit to data after 1 day. We consider the possible causes of this discrepancy, including precursor activity from the progenitor star, circumstellar interaction, and emission from the shock before or after it breaks out of the stellar surface. The very low luminosity (−11 mag > M > −14 mag) and short duration of the initial excess lead us to prefer a scenario related to prolonged emission from the SN shock traveling through the progenitor system.

B cells, follicular helper T (Tfh) cells and follicular regulatory T (Tfr) cells are part of a circuit that may play a role in the development or progression of rheumatoid arthritis (RA). With the aim of providing further insight into this topic, here we evaluated the frequency of different subsets of Tfh and Tfr in untreated and long-term treated RA patients from a cohort of Argentina, and their potential association with particular human leukocyte antigen (HLA) class-II variants and disease activity. We observed that the frequency of total Tfh cells as well as of particular Tfh subsets and Tfr cells were increased in seropositive untreated RA patients. Interestingly, when analyzing paired samples, the frequency of Tfh cells was reduced in synovial fluid compared to peripheral blood, while Tfr cells levels were similar in both biological fluids. After treatment, a decrease in the CCR7loPD1hi Tfh subset and an increase in the frequency of Tfr cells was observed in blood. In comparison to healthy donors, seropositive patients with moderate and high disease activity exhibited higher frequency of Tfh cells while seropositive patients with low disease activity presented higher Tfr cell frequency. Finally, we observed that HLA-DRB1*09 presence correlated with higher frequency of Tfh and Tfr cells, while HLA-DRB1*04 was associated with increased Tfr cell frequency. Together, our results increase our knowledge about the dynamics of Tfh and Tfr cell subsets in RA, showing that this is altered after treatment.

Abstract Investigation of the microbial ecology of terrestrial, aquatic and atmospheric ecosystems requires specific sampling and analytical technologies, owing to vastly different biomass densities typically encountered. In particular, the ultra-low biomass nature of air presents an inherent analytical challenge that is confounded by temporal fluctuations in community structure. Our ultra-low biomass pipeline advances the field of bioaerosol research by significantly reducing sampling times from days/weeks/months to minutes/hours, while maintaining the ability to perform species-level identification through direct metagenomic sequencing. The study further addresses all experimental factors contributing to analysis outcome, such as amassment, storage and extraction, as well as factors that impact on nucleic acid analysis. Quantity and quality of nucleic acid extracts from each optimisation step are evaluated using fluorometry, qPCR and sequencing. Both metagenomics and marker gene amplification-based (16S and ITS) sequencing are assessed with regard to their taxonomic resolution and inter-comparability. The pipeline is robust across a wide range of climatic settings, ranging from arctic to desert to tropical environments. Ultimately, the pipeline can be adapted to environmental settings, such as dust and surfaces, which also require ultra-low biomass analytics.

Extracellular ATP is a critical signaling molecule that is found in a wide range of concentrations across cellular environments. The family of nonselective cation channels that sense extracellular ATP, termed P2X receptors (P2XRs), is composed of seven subtypes (P2X1-P2X7) that assemble as functional homotrimeric and heterotrimeric ion channels. Each P2XR is activated by a distinct concentration of extracellular ATP, spanning from high nanomolar to low millimolar. P2XRs are implicated in a variety of physiological and pathophysiological processes in the cardiovascular, immune, and central nervous systems, corresponding to the spatiotemporal expression, regulation, and activation of each subtype. The therapeutic potential of P2XRs is an emerging area of research in which structural biology has seemingly exceeded medicinal chemistry, as there are several published P2XR structures but currently no FDA-approved drugs targeting these ion channels. Cryogenic electron microscopy is ideally suited to facilitate structure-based drug design for P2XRs by revealing and characterizing novel ligand-binding sites. This review covers structural elements in P2XRs including the extracellular orthosteric ATP-binding site, extracellular allosteric modulator sites, channel pore, and cytoplasmic substructures, with an emphasis on potential therapeutic ligand development.

Abstract Background The western flower thrips, Frankliniella occidentalis (Pergande), is a globally invasive pest and plant virus vector on a wide array of food, fiber, and ornamental crops. The underlying genetic mechanisms of the processes governing thrips pest and vector biology, feeding behaviors, ecology, and insecticide resistance are largely unknown. To address this gap, we present the F. occidentalis draft genome assembly and official gene set. Results We report on the first genome sequence for any member of the insect order Thysanoptera. Benchmarking Universal Single-Copy Ortholog (BUSCO) assessments of the genome assembly (size = 415.8 Mb, scaffold N50 = 948.9 kb) revealed a relatively complete and well-annotated assembly in comparison to other insect genomes. The genome is unusually GC-rich (50%) compared to other insect genomes to date. The official gene set (OGS v1.0) contains 16,859 genes, of which ~ 10% were manually verified and corrected by our consortium. We focused on manual annotation, phylogenetic, and expression evidence analyses for gene sets centered on primary themes in the life histories and activities of plant-colonizing insects. Highlights include the following: (1) divergent clades and large expansions in genes associated with environmental sensing (chemosensory receptors) and detoxification (CYP4, CYP6, and CCE enzymes) of substances encountered in agricultural environments; (2) a comprehensive set of salivary gland genes supported by enriched expression; (3) apparent absence of members of the IMD innate immune defense pathway; and (4) developmental- and sex-specific expression analyses of genes associated with progression from larvae to adulthood through neometaboly, a distinct form of maturation differing from either incomplete or complete metamorphosis in the Insecta. Conclusions Analysis of the F. occidentalis genome offers insights into the polyphagous behavior of this insect pest that finds, colonizes, and survives on a widely diverse array of plants. The genomic resources presented here enable a more complete analysis of insect evolution and biology, providing a missing taxon for contemporary insect genomics-based analyses. Our study also offers a genomic benchmark for molecular and evolutionary investigations of other Thysanoptera species.

The ever-changing nature of the power industry will require the implementation of hybrid energy systems. Integration of tightly coupled components in hybrids often involves the diversion of exhaust gas flow. An innovative smart material actuation technology is proposed to replace traditional electro-mechanical actuated valve mechanisms with lighter and less expensive actuators. A shape memory alloy (SMA) spring-actuated valve was designed for high-temperature service to demonstrate the promise of smart materials in control valve applications. With SMA springs only generating a maximum force of 3.2 N, an innovative valve design was necessary. To demonstrate the concept, a 3-inch Nominal Pipe Size valve was designed, and 3D printed using the stereolithography technique. Increasing the electrical current to actuate the SMA springs reduced actuation time. The maximum current of 10 A produced the lowest actuation time of 2.85 s, with an observed maximum stroke rate of more than 100 stroke completion %/s (considering actuation open/close as 100% stroke) at the midrange. The final assembly of the valve was estimated to provide a cost reduction of more than 30% and a weight reduction of more than 80% compared to the other available automatic valves in the present market.

In the present work we study how the number of simulated customers (occupancy) affects social distance in an ideal supermarket, considering realistic typical dimensions and processing times (product selection and checkout). From the simulated trajectories we measure social distance events of less than 2 m, and their duration. Among other observables, we define a physical distance coefficient that informs how many events (of a given duration) each agent experiences.

We perform a comprehensive search for optical precursor emission at the position of SN 2023ixf using data from the DLT40, ZTF, and ATLAS surveys. By comparing the current data set with precursor outburst hydrodynamical model light curves, we find that the probability of a significant outburst within 5 yr of explosion is low, and the circumstellar material (CSM) ejected during any possible precursor outburst is likely smaller than ∼0.015 M _⊙ . By comparing to a set of toy models, we find that, if there was a precursor outburst, the duration must have been shorter than ∼100 days for a typical brightness of M _r ≃ −9 mag or shorter than 200 days for M _r ≃ −8 mag; brighter, longer outbursts would have been discovered. Precursor activity like that observed in the normal Type II SN 2020tlf ( M _r ≃ −11.5) can be excluded in SN 2023ixf. If the dense CSM inferred by early flash spectroscopy and other studies is related to one or more precursor outbursts, then our observations indicate that any such outburst would have to be faint and only last for days to months, or it occurred more than 5 yr prior to the explosion. Alternatively, any dense, confined CSM may not be due to eruptive mass loss from a single red supergiant progenitor. Taken together, the results of SN 2023ixf and SN 2020tlf indicate that there may be more than one physical mechanism behind the dense CSM inferred around some normal Type II supernovae.

The challenge of a depleting Mississippi River Valley Alluvial Aquifer (MRVAA) requires reducing groundwater withdrawal for irrigation, increasing aquifer recharge, and protecting water quality for sustainable water use. To meet the challenge, the National Center for Alluvial Aquifer Research (NCAAR) is oriented towards producing scientific work aimed at improving irrigation methods and scheduling, employing alternative water sources, and improving crop management and field practices to increase water use efficiency across the region. Big data is key for NCAAR success. Its scientists use big data for research in the form of various soil, weather, geospatial, and water monitoring and management devices to collect agronomic or hydrogeologic data. They also produce, process, and analyze big data which are converted to scientific publications and farm management recommendations via technology transfer. Similarly, decision tools that would help producers leverage the wealth of data they generate from their operations will also be developed and made available to them. This article outlines some of the many ways big data is intertwined with NCAAR’s mission.

Coexistence of ecologically similar species occupying the same geographic location (sympatry) poses questions regarding how their populations persist without leading to competitive exclusion. There is increasing evidence to show that micro-variations in habitat use may promote coexistence through minimizing direct competition for space and resources. We used two sympatric marine predators that show high fidelity to a small, remote coral atoll as a model to investigate how temporally dynamic partitioning of space use may promote coexistence. Using novel methods (difference network analysis and dynamic space occupancy analysis), we revealed that even though blacktip reef sharks Carcharhinus melanopterus and sicklefin lemon sharks Negaprion acutidens both show focused use of the same atoll habitats, the spatio-temporal dynamics of their use was partitioned such that they only shared the same microhabitats 26% of the time. Moreover, the degree of overlap was strongly influenced by the tidal cycle, peaking at ∼35% at higher tides as both species appear to target similar intertidal micro-habitats despite the increase in available space. Our work provides a rare example of how two marine predators with similar ecological roles and habitat preferences may coexist in the same place through dynamic segregation of habitat use in space and time, potentially reflecting adaptive behavioral traits for minimizing interactions. The strong influence of small tidal variation on species habitat use and partitioning also raises concerns over how atoll ecosystem dynamics may be influenced by sea level rises that could alter tidal dynamics.

Lead trihalide perovskites are notable for their excellent optoelectronic properties and uncommon phase behavior. Here, Kirschner et al. show that cesium lead bromide nanocrystals experience a reversible orthorhombic-to-cubic phase transition at moderate excitation fluences and become amorphous at higher fluences.

Humidity has been shown to influence many aspects of plant physiology. Here Mwimba et al. show that oscillating humidity entrains the circadian clock under constant light conditions and enhances clock amplitude in simulated natural environments, while also improving immunity and overall growth.

Mechanical twins are common microstructures in deformed calcite. Calcite twins have been used for a long time as indicators of stress/strain orientations and magnitudes. Developments during the last decade point toward significant improvements of existing techniques as well as new applications of calcite twin analysis in tectonic studies. This review summarises the recent progress in the understanding of twin formation, including nucleation and growth of twins, and discusses the concept of CRSS and its dependence on several factors such as strain, temperature and grain size. Classical and recent calcite twin measurement techniques are also presented and their pros and cons are discussed. The newly proposed inversion techniques allowing for the use of calcite twins as indicators of orientations and/or magnitudes of stress and strain are summarized. Benefits for tectonic studies are illustrated through the presentation of several applications, from the scale of the individual tectonic structure to the continental scale. The classical use of calcite twin morphology (e.g., thickness) as a straightforward geothermometer is critically discussed in the light of recent observations that thick twins do not always reflect deformation temperature above 170–200 °C. This review also presents how the age of twinning events in natural rocks can be constrained while individual twins cannot be dated yet. Finally, the review addresses the recent technical and conceptual progress in calcite twinning paleopiezometry, together with the promising combination of this paleopiezometer with mechanical analysis of fractures or stylolite roughness.

The present work correlates the quasi-static, tensile mechanical properties of additively manufactured Ti-6Al-4V extra low interstitial (ELI, Grade 23) alloy to the phase constituents, microstructure, and fracture surface characteristics that changed with post-heat treatment of stress relief (670 °C for 5 h) and hot isostatic pressing (HIP with 100 MPa at 920 °C for 2 h under an Ar atmosphere). Ti-6Al-4V ELI alloy tensile specimens in both the horizontal (i.e., X and Y) and vertical (Z) directions were produced by the laser powder bed fusion (LPBF) technique. Higher yield strength (1141 MPa), higher tensile strength (1190 MPa), but lower elongation at fracture (6.9%), along with mechanical anisotropy were observed for as-stress-relieved (ASR) samples. However, after HIP, consistent and isotropic mechanical behaviors were observed with a slight reduction in yield strength (928 MPa) and tensile strength (1003 MPa), but with a significant improvement in elongation at fracture (16.1%). Phase constituents of acicular α′ phase in ASR and lamellar α + β phases in HIP samples were observed and quantified to corroborate the reduction in strength and increase in ductility. The anisotropic variation in elongation at fracture observed for the ASR samples, particularly built in the build (Z) direction, was related to the presence of “keyhole” porosity.

Abstract Single-molecule RNA fluorescence in situ hybridization (smFISH) provides unparalleled resolution in the measurement of the abundance and localization of nascent and mature RNA transcripts in fixed, single cells. We developed a computational pipeline (BayFish) to infer the kinetic parameters of gene expression from smFISH data at multiple time points after gene induction. Given an underlying model of gene expression, BayFish uses a Monte Carlo method to estimate the Bayesian posterior probability of the model parameters and quantify the parameter uncertainty given the observed smFISH data. We tested BayFish on synthetic data and smFISH measurements of the neuronal activity-inducible gene Npas4 in primary neurons.

Though coronary arteries are crucial for heart function, the mechanisms guiding their formation are largely unknown. Here, Wang et al. identify a unique, endocardially-derived angiogenic precursor cell population for coronary artery formation in mice and show that a DLL4/NOTCH1/VEGFA/VEGFR2 signaling axis is key for coronary artery development.
