Among grapevine yellows, Bois noir (BN), associated with ‘Candidatus Phytoplasma solani’, represents the biggest threat in the main wine-growing areas worldwide, causing significant losses in berry quality and yields. BN epidemiology involves multiple plant hosts and several insect vectors, making considerably complex the development of effective management strategies. Since application of insecticides on the grapevine canopy is not effective to manage vectors, BN management includes an integrated approach based on treatments to the canopy to make the plant more resistant to the pathogen and/or inhibit the vector feeding, and actions on reservoir plants to reduce possibilities that the vector reaches the grapevine and transmit the phytoplasma. Innovative sustainable strategies developed in the last twenty years to improve the BN management are reviewed and discussed.

The HERD experiment is a future experiment for the direct detection of high-energy cosmic rays and is to be installed on the Chinese space station in 2027. The main objectives of HERD are the first direct measurement of the knee of the cosmic ray spectrum, the extension of electron+positron flux measurement up to tens of TeV, gamma ray astronomy, and the search for indirect signals of dark matter. The main component of the HERD detector is an innovative calorimeter composed of about 7500 LYSO scintillating crystals assembled in a spherical shape. Two independent readout systems of the LYSO scintillation light will be installed on each crystal: the wavelength-shifting fibers system developed by IHEP and the double photodiode readout system developed by INFN and CIEMAT. In order to measure protons in the cosmic ray knee region, we must be able to measure energy release of about 250 TeV in a single crystal. In addition, in order to calibrate the system, we need to measure typical releases of minimum ionizing particles that are about 30 MeV. Thus, the readout systems should have a dynamic range of about 107. In this article, we analyze the development and the performance of the double photodiode readout system. In particular, we show the performance of a prototype readout by the double photodiode system for electromagnetic showers as measured during a beam test carried out at the CERN SPS in October 2021 with high-energy electron beams.

Storage pests and the food spoilage they cause are problems of great concern. Using essential oil obtained from different plants as an insecticide against these storage pests can be considered an environmentally friendly pest management option. Lantana camara Linn. (family Verbenaceae) is a flowering species, and is also a noxious weed that can proliferate well in nearly all geographical habitats. A biopesticide derived from the essential oil extracted from this plant can offer an effective solution for controlling storage pests. The goal of this study is to extract and analyse the chemical composition of essential oil obtained from L. camara leaves, and assess its effectiveness as a bioactive substance against three storage pests: Tribolium castaneum, Lasioderma serricorne, and Callosobruchus chinensis. The yield of essential oil extracted from L. camara leaves was about 0.24 ± 0.014%. By employing the GC-MS technique, the major phytochemicals contained in L. camara leaf essential oil were identified as caryophyllene (69.96%), isoledene (12%), and ɑ-copaene (4.11%). The essential oil exhibited excellent fumigant toxicity (LC50 of 16.70 mg/L air for T. castaneum, 4.141 mg/L air for L. serricorne and 6.245 mg/L air for C. chinensis at 24 h), contact toxicity (LC50 of 8.93 mg/cm2 for T. castaneum, 4.82 mg/cm2 for L. serricorne and 6.24 mg/cm2 for C. chinensis after 24 h) along with effective repellent activity towards the test insects. In addition, the oil showed no significant phytotoxicity on the germination of paddy seeds. This presents the potential to utilize a weed in developing a biopesticide for effectively managing stored product insects because of its strong bioactivity.

Pathogens and pests constantly challenge food security and safety worldwide. The use of plant protection products to manage them raises concerns related to human health, the environment, and economic costs. Basic substances are active, non-toxic compounds that are not predominantly used as plant protection products but hold potential in crop protection. Basic substances’ attention is rising due to their safety and cost-effectiveness. However, data on their protection levels in crop protection strategies are lacking. In this review, we critically analyzed the literature concerning the field application of known and potential basic substances for managing diseases and pests, investigating their efficacy and potential integration into plant protection programs. Case studies related to grapevine, potato, and fruit protection from pre- and post-harvest diseases and pests were considered. In specific cases, basic substances and chitosan in particular, could complement or even substitute plant protection products, either chemicals or biologicals, but their efficacy varied greatly according to various factors, including the origin of the substance, the crop, the pathogen or pest, and the timing and method of application. Therefore, a careful evaluation of the field application is needed to promote the successful use of basic substances in sustainable pest management strategies in specific contexts.

The mosaic leafhopper, Orientus ishidae (Matsumura), is an Asian species widespread in Europe that can cause leaf damage in wild trees and transmit disease phytoplasmas to grapevines. Following an O. ishidae outbreak reported in 2019 in an apple orchard in northern Italy, the biology and damage caused by this species to apples were investigated during 2020 and 2021. Our studies included observations on the O. ishidae life cycle, leaf symptoms associated to its trophic activity, and its capability to acquire “Candidatus Phytoplasma mali,” a causal agent of Apple Proliferation (AP). The results indicate that O. ishidae can complete the life cycle on apple trees. Nymphs emerged between May and June, and adults were present from early July to late October, with the peak of flight between July and early August. Semi-field observations allowed for an accurate description of leaf symptoms that appeared as a distinct yellowing after a one-day exposure. In field experiments, 23% of the leaves were found damaged. In addition, 16–18% of the collected leafhoppers were found carrying AP phytoplasma. We conclude that O. ishidae has the potential to be a new apple tree pest. However, further studies are required to better understand the economic impact of the infestations.

Scaphoideus titanus is the main vector of phytoplasmas associated with Flavescence dorée (FD), one of the most serious threats to viticulture in many European countries. To minimize the spread of this disease, mandatory control measures against S. titanus were decided in Europe. In the 1990s, the repeated application of insecticides (mainly organophosphates) proved to be an effective measure to control the vector and the related disease in north-eastern Italy. These insecticides and most of the neonicotinoids were recently banned from European viticulture. Serious FD issues detected in the recent years in northern Italy could be related to the use of less effective insecticides. Trials aimed at evaluating the efficacy of the most used conventional and organic insecticides in the control of S. titanus have been performed in semi-field and field conditions to test this hypothesis. In efficacy trials, carried out in four vineyards, etofenprox and deltamethrin proved to be the best conventional insecticides, while pyrethrins were the most impactful among organic insecticides. Insecticide residual activity was evaluated in semi-field and field conditions. Acrinathrin showed the most significant residual effects in both conditions. In semi-field trials, most of the pyrethroids were associated with good results in terms of residual activity. However, these effects declined in field conditions, probably due to high temperatures. Organic insecticides showed poor results in terms of residual efficacy. Implications of these results in the context of Integrated Pest Management in conventional and organic viticulture are discussed.

A space-based detector dedicated to measurements of γ-rays and charged particles has to achieve a balance between different instrumental requirements. A good angular resolution is necessary for the γ-rays, whereas an excellent geometric factor is needed for the charged particles. The tracking reference technique of γ-ray physics is based on a pair-conversion telescope made of passive material (e.g., tungsten) coupled with sensitive layers (e.g., silicon microstrip). However, this kind of detector has a limited acceptance because of the large lever arm between the active layers, needed to improve the track reconstruction capability. Moreover, the passive material can induce fragmentation of nuclei, thus worsening charge reconstruction performances. The Tracker-In-Calorimeter (TIC) project aims to solve all these drawbacks. In the TIC proposal, the silicon sensors are moved inside a highly-segmented isotropic calorimeter with a couple of external scintillators dedicated to charge reconstruction. In principle, this configuration has a good geometrical factor, and the angle of the γ-rays can be precisely reconstructed from the lateral profile of the electromagnetic shower sampled, at different depths in the calorimeter, by silicon strips. The effectiveness of this approach has been studied with Monte Carlo simulations and validated with beam test data of a small prototype.

Calorimetric space experiments were employed for the direct measurements of cosmic-ray spectra above the TeV region. According to several theoretical models and recent measurements, relevant features in both electron and nucleus fluxes are expected. Unfortunately, sizable disagreements among the current results of different space calorimeters exist. In order to improve the accuracy of future experiments, it is fundamental to understand the reasons of these discrepancies, especially since they are not compatible with the quoted experimental errors. A few articles of different collaborations suggest that a systematic error of a few percentage points related to the energy-scale calibration could explain these differences. In this work, we analyze the impact of the nonproportionality of the light yield of scintillating crystals on the energy scale of typical calorimeters. Space calorimeters are usually calibrated by employing minimal ionizing particles (MIPs), e.g., nonshowering proton or helium nuclei, which feature different ionization density distributions with respect to particles included in showers. By using the experimental data obtained by the CaloCube collaboration and a minimalist model of the light yield as a function of the ionization density, several scintillating crystals (BGO, CsI(Tl), LYSO, YAP, YAG and BaF2) are characterized. Then, the response of a few crystals is implemented inside the Monte Carlo simulation of a space calorimeter to check the energy deposited by electromagnetic and hadronic showers. The results of this work show that the energy scale obtained by MIP calibration could be affected by sizable systematic errors if the nonproportionality of scintillation light is not properly taken into account.

HERD is a future experiment for the direct detection of high energy cosmic rays. The instrument is based on a calorimeter optimized not only for a good energy resolution but also for a large acceptance. Each crystal composing the calorimeter is equipped with two read-out systems: one based on wavelength-shifting fibers and the other based on two photodiodes with different active areas assembled in a monolithic package. In this paper, we describe the photodiode read-out system, focusing on experimental requirements, design and estimated performances. Finally, we show how these features lead to the flight model project of the photodiode read-out system.

The encapsulation of pesticides within nanoparticles is a promising approach of advanced technology in sustainable agriculture. Lambda-cyhalothrin (LC) was encapsulated by the ionotropic gelation technique into chitosan (CS)/tripolyphosphate (TPP) and CS/alginate (ALG) matrixes. CS-LC nanoparticles were characterized, and their efficacy was then evaluated against the key pest of soft fruits in Europe and the United States, Drosophila suzukii. The encapsulation efficiency (74%), nanoparticle yield (80%), polydispersity index (0.341), zeta potential (−23.1 mV) and particle size (278 nm) were determined at the optimum conditions. FTIR confirmed the cross-linkage between CS and TPP/ALG in the nanoparticles. The optimum formula recommended by the fractional factorial design was associated with the formulation variables of CS of high or low molecular weight, cross-linking agent (TPP), LC concentration (1.5% w/v) and stirring rate (1500 rpm), showing the highest desirability value (0.5511). CS-LC nanoparticles of the lowest particle size (278 nm) exhibited the highest percent mortality of D. suzukii males (86%) and females (84%), exceeding that caused by the commercial product (Karate-zeon® 10% CS) at 2 HAT. This is the first work to use the ionic gelation technique to make LC nanoparticles, to the best of our knowledge. The encapsulation of chemical pesticides within biodegradable polymeric nanoparticles could be helpful for establishing a sustainable IPM strategy with benefits for human and environmental health and the lifetime of pesticides.

A new generation magnetic spectrometer in space will open the opportunity to investigate the frontiers in direct high-energy cosmic ray measurements and to precisely measure the amount of the rare antimatter component in cosmic rays beyond the reach of current missions. We propose the concept for an Antimatter Large Acceptance Detector In Orbit (ALADInO), designed to take over the legacy of direct measurements of cosmic rays in space performed by PAMELA and AMS-02. ALADInO features technological solutions conceived to overcome the current limitations of magnetic spectrometers in space with a layout that provides an acceptance larger than 10 m2 sr. A superconducting magnet coupled to precision tracking and time-of-flight systems can provide the required matter–antimatter separation capabilities and rigidity measurement resolution with a Maximum Detectable Rigidity better than 20 TV. The inner 3D-imaging deep calorimeter, designed to maximize the isotropic acceptance of particles, allows for the measurement of cosmic rays up to PeV energies with accurate energy resolution to precisely measure features in the cosmic ray spectra. The operations of ALADInO in the Sun–Earth L2 Lagrangian point for at least 5 years would enable unique revolutionary observations with groundbreaking discovery potentials in the field of astroparticle physics by precision measurements of electrons, positrons, and antiprotons up to 10 TeV and of nuclear cosmic rays up to PeV energies, and by the possible unambiguous detection and measurement of low-energy antideuteron and antihelium components in cosmic rays.

The control of Bactrocera oleae is fundamental to decreasing the significant production loss in olive cultivation. However, traditional containment based on the use of synthetic insecticides has been encountering serious limitations due to their negative effect on human health and the environment. Within the scope of integrated olive fly management, the use of products with repellency and oviposition deterrent activity might represent a more eco-friendly solution. In this study, we tested the oviposition deterrent activity of some commercial formulations already used in olive tree crops as fungicides (copper oxychloride, dodine, mancozeb, pyraclostrobin and difeconazole) and plant bio-stimulants (tannins, clay, flavonoids and a zinc-copper-citric acid biocomplex). The trials were conducted testing the oviposition behavior of mated olive fly females in both choice and no-choice assays. Our results showed that most of the substances have affected the ovipositional activity of the olive fly, except for difeconazole. Moreover, some products (copper oxychloride, flavonoids and tannins) have proven to differently influence the flies’ oviposition comparing the two tests. The repellent effect of these commercial products should be further studied to prove whether the repellency was due either to the active ingredient or to the co-formulants, and to assess their effect in the open field.

Grapevine Bois noir (BN) is associated with infection by “Candidatus Phytoplasma solani” (CaPsol). In this study, an array of CaPsol strains was identified from 142 symptomatic grapevines in vineyards of northern, central, and southern Italy and North Macedonia. Molecular typing of the CaPsol strains was carried out by analysis of genes encoding 16S rRNA and translation elongation factor EF-Tu, as well as eight other previously uncharacterized genomic fragments. Strains of tuf-type a and b were found to be differentially distributed in the examined geographic regions in correlation with the prevalence of nettle and bindweed. Two sequence variants were identified in each of the four genomic segments harboring hlyC, cbiQ-glyA, trxA-truB-rsuA, and rplS-tyrS-csdB, respectively. Fifteen CaPsol lineages were identified based on distinct combinations of sequence variations within these genetic loci. Each CaPsol lineage exhibited a unique collective restriction fragment length polymorphism (RFLP) pattern and differed from each other in geographic distribution, probably in relation to the diverse ecological complexity of vineyards and their surroundings. This RFLP-based typing method could be a useful tool for investigating the ecology of CaPsol and the epidemiology of its associated diseases. Phylogenetic analyses highlighted that the sequence variants of the gene hlyC, which encodes a hemolysin III-like protein, separated into two clusters consistent with the separation of two distinct lineages on the basis of tufB gene sequences. Alignments of deduced full protein sequences of elongation factor-Tu (tufB gene) and hemolysin III-like protein (hlyC gene) revealed the presence of critical amino acid substitutions distinguishing CaPsol strains of tuf-type a and b. Findings from the present study provide new insights into the genetic diversity and ecology of CaPsol populations in vineyards.

Hyalesthes obsoletus is the vector of “Candidatus Phytoplasma (Ca. P.) solani,” the causal agent of grapevine yellows Bois noir (BN). The relationships among the planthopper, its main herbaceous hosts as phytoplasma reservoirs (Convolvolus arvensis and Urtica dioica) and BN spreading were studied in northern Italy. In two areas the relationship between host plants and the phenology and survival of planthopper adults was investigated in potted plants and in field conditions. Moreover, H. obsoletus ecology, newly symptomatic grapevine occurrence and “Ca. P. solani” tuf-types’ presence were studied in two vineyards (2014–2019). An earlier occurrence of H. obsoletus adults on C. arvensis than U. dioica and better adult survival of the originating host were observed. When U. dioica was prevalent, the vector occurred almost exclusively along the ditch outside the vineyard. Hyalesthes obsoletus amount varied widely from year to year and nymphal mortality due to late frosts was supposed. In one vineyard, the amount of newly symptomatic grapevines was significantly correlated with vector abundance in the previous year. The “Ca. P. solani” tuf-type was influenced by vector population levels on the two hosts. Since the abundance of H. obsoletus populations on the two hosts influences BN epidemiology and dynamics and the “Ca. P. solani” tuf-type, this must be considered in BN control strategies.

The meadow spittlebug, Philaenus spumarius (L.) (Hemiptera Aphrophoridae), the main vector of Xylella fastidiosa Wells et al. in Europe, has few known natural enemies. The endoparasitoid Verrallia aucta (Fallén) (Diptera, Pipunculidae) was first noticed a long time ago but very little is known about its biology and prevalence. In this study, the presence and prevalence of V. aucta were investigated in different regions of northern Italy, both in plain–foothill and montane zones. Parasitic larvae were identified by the dissection of spittlebug adults, P. spumarius and Neophilaenus campestris (Fallén), and by a new species-specific molecular tool targeting the ITS2 and COI genomic regions, developed in this work. A small-scale rearing was set up to gain information on the life cycle of V. aucta on its main host P. spumarius. During the four-year investigation (2016–2019) the pipunculid parasitoid displayed low prevalence, reaching a maximum parasitization rate of 17.5% (calculated over the adult spittlebug season) in vineyards of the Piemonte region. Over the whole period, no significant difference in the prevalence was found between male and female spittlebugs. Collected data and rearing observations suggest that V. aucta is monovoltine and synchronous with P. spumarius, laying eggs in newly emerged adults, developing as an endoparasitoid through two larval stages during the whole summer, and overwintering as a pupa in the soil.

Precise measurements of the energy spectra and of the composition of cosmic rays in the PeV region could improve our knowledge regarding their origin, acceleration mechanism, propagation, and composition. At the present time, spectral measurements in this region are mainly derived from data collected by ground-based detectors, because of the very low particle rates at these energies. Unfortunately, these results are affected by the high uncertainties typical of indirect measurements, which depend on the complicated modeling of the interaction of the primary particle with the atmosphere. A space experiment dedicated to measurements in this energy region has to achieve a balance between the requirements of lightness and compactness, with that of a large acceptance to cope with the low particle rates. CaloCube is a four-year-old R&D project, approved and financed by the Istituto Nazionale di Fisica Nucleare (INFN) in 2014, aiming to optimize the design of a space-borne calorimeter. The large acceptance needed is obtained by maximizing the number of entrance windows, while thanks to its homogeneity and high segmentation this new detector achieves an excellent energy resolution and an enhanced separation power between hadrons and electrons. In order to optimize detector performances with respect to the total mass of the apparatus, comparative studies on different scintillating materials, different sizes of crystals, and different spacings among them have been performed making use of MonteCarlo simulations. In parallel to simulations studies, several prototypes instrumented with CsI(Tl) (Caesium Iodide, Tallium doped) cubic crystals have been constructed and tested with particle beams. Moreover, the last development of CaloCube, the Tracker-In-Calorimeter (TIC) project, financed by the INFN in 2018, is focused on the feasibility of including several silicon layers at different depths in the calorimeter in order to reconstruct the particle direction. In fact, an important requirement for γ -ray astronomy is to have a good angular resolution in order to allow precise identification of astrophysical sources in space. In respect to the traditional approach of using a tracker with passive material in front of the calorimeter, the TIC solution can save a significant amount of mass budget in a space satellite experiment, which can then be exploited to improve the acceptance and the resolution of the calorimeter. In this paper, the status of the project and perspectives for future developments are presented.

Muon absorption radiography is an imaging technique based on the measurement of the absorption of cosmic ray muons. This technique has recently been used successfully to investigate the presence of unknown cavities in the Bourbon Gallery in Naples and in the Chephren Pyramid at Cairo. The MIMA detector (Muon Imaging for Mining and Archaeology) is a prototype muon tracker for muon radiography for application in the fields of archaelogy and mining. It is made of three pairs of X-Y planes each consisting of 21 scintillator bars with a silicon photomultiplier readout. The detector is compact, robust, easily transportable, and has a low power consumption: all of which makes the detector ideal for measurements in confined and isolated environments. With this detector, a measurement from inside the Temperino mine in the San Silvestro archaeo-mining park in Tuscany was performed. The park includes about 25 km of mining tunnels arranged on several levels that have been exploited from the Etruscan time. The measured muon absorption was compared to the simulated one, obtained from the information provided by 3D laser scanner measurements and cartographic maps of the mountain above the mine, in order to obtain information about the average density of the rock. This allowed one to confirm the presence of a partially accessible exploitation opening and provided some hints regarding the presence of a high-density body within the rock.

Drosophila suzukii causes considerable economic damage to small and thin-skinned fruits including cherry, blueberry, raspberry, grape and strawberry. Since it attacks fruits at the ripening stage, the use of chemical pesticides is limited due to the high risk of residues on fruit. Biological control is thus expected to play an essential role in managing this pest. The Gram-negative bacterium, Photorhabdus luminescens and its symbiotic Heterorhabditis spp. nematode have been shown to be highly pathogenic to insects, with a potential for replacing pesticides to suppress several pests. Insecticidal activity of P. luminescens at different bacterial cell concentrations and its cell-free supernatant were assessed against third-instar larvae and pupae of D. suzukii under laboratory conditions. P. luminescens suspensions had a significant oral and contact toxicity on D. suzukii larvae and pupae, with mortalities up to of 70⁻100% 10 days after treatment. Cell-free supernatant in the diet also doubled mortality rates of feeding larvae. Our results suggest that P. luminescens may be a promising candidate for biological control of D. suzukii, and its use in integrated pest management (IPM) programs is discussed.

The leafhopper Scaphoideus titanus is the vector of ‘Candidatus Phytoplasma vitis’, the causal agent of Flavescence dorée (FD) a key disease for European viticulture. In organic vineyards, the control of S. titanus relies mostly on the use of pyrethrins that have suboptimal efficacy. During 2016, three field trials were conducted to evaluate the efficacy of kaolin, orange oil, insecticidal soap and spinosad against S. titanus nymphs, in comparison with pyrethrins. The activity of kaolin was evaluated also in the laboratory. In all field trials, kaolin had an efficacy against nymphs comparable to pyrethrins, while the other products were not effective. Laboratory results confirmed that kaolin increased nymph mortality. In organic vineyards, kaolin and pyrethrins are valuable tools in the management of FD. Nevertheless, their efficacy is lower compared to that of the synthetic insecticides used in conventional viticulture. Therefore, further research should be conducted in order to identify alternatives to synthetic insecticides for S. titanus control in the context of organic viticulture.

The course of ‘flavescence dorée’ (FD) and ‘bois noir’ (BN) diseases can result in either recovery or death of affected grapevines. When farmers observe symptomatic grapevines, they must choose whether to replace or maintain the plants. To establish whether there is an advantage in replacing symptomatic grapevines, data were collected on the costs of replacing them (removing the diseased plants and planting new grapevines, with resultant yield loss during the rearing period) and growing them on (yield losses in symptomatic grapevines over the following years). To calculate the cost of maintaining FD-infected plants, the possibility was also considered that symptomatic grapevines may be sources of phytoplasmas for the vector Scaphoideus titanus Ball. The symptomatic course of BN was observed in ‘Chardonnay’, and of FD in ‘Chardonnay’, ‘Merlot’ and ‘Perera’ grape cultivars. The costs of replacement decreased with the increase in the productive lifetime of the vineyards. The cost of maintenance was greatly influenced by the course of the diseases, and in the case of FD, also by the risk of new infections due to the fact that S. titanus acquires phytoplasmas from infected grapevines. The replacement of plants affected by BN is not profitable when recovery is the most frequent course of the disease, particularly when it is considered that replantings can, in turn, become infected. The replacement of plants affected by FD is not profitable for cultivars with a recovery near to 100% (‘Merlot’), whereas it is necessary for cultivars where the course of the disease is frequently lethal (‘Perera’). For cultivars with intermediate sensitivity, the decision varies in relation to agronomic/economic factors and to the risk of new infections (‘Chardonnay’). For FD, both replacement and maintenance strategies need to be associated with S. titanus control inside and outside the vineyards. In the case of maintenance the infected plants can be sources of phytoplasmas, and in the case of replacement, the vector can also inoculate the new grapevines.
