13. Downregulation of type I interferon signalling pathway by urate in primary human PBMCs.

Abstract

Type I interferons (IFN1s) mediate innate responses to microbial stimuli and regulate interleukin (IL)‐1 and IL‐1 receptor antagonist (Ra) production in human cells. This study explores interferon‐stimulated gene (ISG) alterations in the transcriptome of patients with gout and stimulated human primary cells in vitro in relation to serum urate concentrations. Peripheral blood mononuclear cells (PBMCs) and monocytes of patients with gout were primed in vitro with soluble urate, followed by lipopolysaccharide (LPS) stimulation. Separately, PBMCs were stimulated with various toll‐like receptor (TLR) ligands. RNA sequencing and IL‐1Ra cytokine measurement were performed. STAT1 phosphorylation was assessed in urate‐treated monocytes. Cytokine responses to IFN‐β were evaluated in PBMCs cultured with or without urate and restimulated with LPS and monosodium urate (MSU) crystals. Transcriptomics revealed suppressed IFN‐related signalling pathways in urate‐exposed PBMCs or monocytes which was supported by diminishment of phosphorylated STAT1. The stimulation of PBMCs with IFN‐β did not modify the urate‐induced inflammation. Interestingly, in vivo, serum urate concentrations were inversely correlated to in vitro ISG expression upon stimulations with TLR ligands. These findings support a deficient IFN1 signalling in the presence of elevated serum urate concentrations, which could translate to increased susceptibility to infections. [ABSTRACT FROM AUTHOR]

15. Advancements in Ti3C2TxMXene Stability: Synergistic Antioxidant Strategies and Their Impact on Long-Lasting Flexible Sensors

Abstract

Two-dimensional (2D) transition metal carbides (Ti3C2TxMXene) have demonstrated substantial application potential across various fields, owing to their excellent metallic conductivity and solution processability. However, the rapid oxidation of Ti3C2Txin aqueous environments, leading to a loss of stability within mere days, poses a significant obstacle for its practical applications. Herein, we introduce an antioxidant strategy that combines free radical scavenging with surface passivation, culminating in the design and synthesis of imidazolium-based ionic liquids (ILs) incorporating siloxane groups. By deploying a straightforward hydrolysis-addition reaction, we successfully fabricated IL-modified Ti3C2Txmaterials (Ti3C2Tx-IL). The Ti3C2Tx-IL not only displayed exceptional conductivity exceeding 3.85 × 104S/m and hydrophilic contact angles below 45° but also showcased its superior chemical stability and antioxidation mechanisms through various analyses, including visual color change experiments, spectroscopic and energy spectrum characterization, free radical scavenging tests, and density-functional-theory-based molecular simulations. Furthermore, when utilized as a conductive filler in the fabrication of a poly(vinyl alcohol)/nanocellulose fiber (PVA/CNF) composite hydrogel (PCMIL), the resultant sensors exhibited remarkable mechanical performance with up to 535% strain, 1.59 MPa strength, 4.35 MJ/m3toughness, and a conductivity of 3.40 mS/cm, as well as a high sensitivity gauge factor of 3.3. Importantly, even after 45 days of storage, the PCMIL retained most of its functionalities, demonstrating superior performance in human-machine interaction applications compared to hydrogels made from unmodified Ti3C2Tx. This research establishes a robust antioxidant protection strategy for Ti3C2Tx, offering substantial technical reinforcement for its prospective applications in the realm of flexible electronics and sensing technologies.

Abstract

Gout, an inflammatory disease orchestrated by interleukin-1β activation and release, is more prevalent in men. The clinical profiles of patients with gout report differences by sex. This study aims to investigate sex-specific cytokine profiles in circulation and in stimulated peripheral blood mononuclear cells (PBMCs) of patients with gout and controls. Participants included in the gout group met the criteria of the American College of Rheumatology/European League Against Rheumatism (ACR/EULAR). The control group included individuals with varying levels of serum urate and absence of gout. PBMCs were stimulated in vitro for 24 h with various TLR ligands. Cytokines were determined in culture supernatants and plasma. Plasma IL-1Ra and high-sensitivity C-reactive protein (hsCRP) were higher in men with gout compared to men without gout whereas no significant differences in circulating cytokines were observed in women. PBMCs of patients with gout showed higher cytokine production of IL-1β, IL-1Ra, and TNF-α following 24 h stimulation, predominantly observed in women. We identified sex-specific cytokine production in gout in response to in vitro stimulation. While men with gout had higher levels of circulating cytokines, stimulated PBMCs of women with gout show an enhanced capacity for cytokine production. Abstract

Prostate malignancy represents the second leading causeof cancer-specificdeath among the male population worldwide. Herein, enhanced intracellularmagnetic fluid hyperthermia is applied in vitro totreat prostate cancer (PCa) cells with minimum invasiveness and toxicityand highly specific targeting. We designed and optimized novel shape-anisotropicmagnetic core-shell-shell nanoparticles (i.e., trimagneticnanoparticles - TMNPs) with significant magnetothermal conversionfollowing an exchange coupling effect to an external alternating magneticfield (AMF). The functional properties of the best candidate in termsof heating efficiency (i.e., Fe3O4@Mn0.5Zn0.5Fe2O4@CoFe2O4) were exploited following surface decoration with PCa cellmembranes (CM) and/or LN1 cell-penetrating peptide (CPP). We demonstratedthat the combination of biomimetic dual CM-CPP targeting and AMF responsivenesssignificantly induces caspase 9-mediated apoptosis of PCa cells. Furthermore,a downregulation of the cell cycle progression markers and a decreaseof the migration rate in surviving cells were observed in responseto the TMNP-assisted magnetic hyperthermia, suggesting a reductionin cancer cell aggressiveness.
