Abstract

Sjögrens disease is a complex autoimmune disease with twelve established susceptibility loci. This genome-wide association study (GWAS) identifies ten novel genome-wide significant (GWS) regions in Sjögrens cases of European ancestry: CD247, NAB1, PTTG1-MIR146A, PRDM1-ATG5, TNFAIP3, XKR6, MAPT-CRHR1, RPTOR-CHMP6-BAIAP6, TYK2, SYNGR1. Polygenic risk scores yield predictability (AUROC = 0.71) and relative risk of 12.08. Interrogation of bioinformatics databases refine the associations, define local regulatory networks of GWS SNPs from the 95% credible set, and expand the implicated gene list to >40. Many GWS SNPs are eQTLs for genes within topologically associated domains in immune cells and/or eQTLs in the main target tissue, salivary glands.

11. Engaging High School Girls in Interdisciplinary STEAM

Abstract

Research into Australian students' science, technology, engineering, and maths (STEM) engagement has highlighted that there are comparably fewer women enrolling in STEM programs and working in STEM industries. In Australia, males make up 84% of the total population with STEM qualifications, for example, a report in 2015 found only 13% of all engineers in Australia were women. Science, technology, engineering, art, and mathematics (STEAM), an approach to STEM education that encourages interdisciplinarity, creativity, innovation, and entrepreneurship, is a strategy that has the potential to increase girls' engagement with STEM. This research investigated the impact of the STEAMpunk Girls Program, funded by the Australian Government, on high school girls' learning and their teachers' teaching experiences. The program uses project-learning and design thinking strategies to enable the girls to gain confidence in themselves as change-makers with the capacity to generate solutions to real-world problems. Using a mixed method approach, the findings indicated that teachers and students were positive about the STEAMpunk Girls' experience, with significant increases in confidence and motivation in the girls at the end of the program.

12. Affective Profiles of Year 9/10 Australian and South East Asian Students in Science and Science Education

Abstract

While there are research studies into students' affective perspectives revolving around science and science education over the last five decades, there are few studies on students' attitudes and beliefs in these areas in developing countries in South East Asia, particularly those that do not participate in international assessment programs. This quantitative study investigated the affective profiles of Year 9/10 students in science/science learning from seven South East Asian countries and Australia to understand the similarities and differences in students' attitudes and the implications in advancing the STEM education agenda in these countries. Comparative studies perform several important functions that are closely interlinked and this study heightens our awareness of the impact of systems and cultures on students' emotional views in the areas of science/science education, allowing country coordinators and policy makers to compare and review teaching practices to enhance student engagement and performance. The implications of the study are discussed.

13. Towards Automated Fatigue Assessment using Wearable Sensing and Mixed-Effects Models

Abstract

Fatigue is a broad, multifactorial concept that includes the subjective perception of reduced physical and mental energy levels. It is also one of the key factors that strongly affect patients' health-related quality of life. To date, most fatigue assessment methods were based on self-reporting, which may suffer from many factors such as recall bias. To address this issue, in this work, we recorded multi-modal physiological data (including ECG, accelerometer, skin temperature and respiratory rate, as well as demographic information such as age, BMI) in free-living environments and developed automated fatigue assessment models. Specifically, we extracted features from each modality and employed the random forest-based mixed-effects models, which can take advantage of the demographic information for improved performance. We conducted experiments on our collected dataset, and very promising preliminary results were achieved. Abstract

Fatigue is one of the key factors in the loss of work efficiency and health-related quality of life, and most fatigue assessment methods were based on self-reporting, which may suffer from many factors such as recall bias. To address this issue, we developed an automated system using wearable sensing and machine learning techniques for objective fatigue assessment. ECG/Actigraphy data were collected from subjects in free-living environments. Preprocessing and feature engineering methods were applied, before interpretable solution and deep learning solution were introduced. Specifically, for interpretable solution, we proposed a feature selection approach which can select less correlated and high informative features for better understanding system's decision-making process. For deep learning solution, we used state-of-the-art self-attention model, based on which we further proposed a consistency self-attention (CSA) mechanism for fatigue assessment. Abstract

There has been a significant increase in the number of diagnostic and prognostic models published in the last decade. Testing such models in an independent, external validation cohort gives some assurance the model will transfer to a naturalistic, healthcare setting. Of 2,147 published models in the PubMed database, we found just 120 included some kind of separate external validation cohort. Of these studies not all were sufficiently well documented to allow a judgement about whether that model was likely to transfer to other centres, with other patients, treated by other clinicians, using data scored or analysed by other laboratories. We offer a solution to better characterizing the validation cohort and identify the key steps on the translational pathway for diagnostic and prognostic models.

Abstract

The Receiver Operating Characteristic (ROC) curve and the Area Under the Curve (AUC) of the ROC curve are widely used to compare the performance of diagnostic and prognostic assays. The ROC curve has the advantage that it is independent of disease prevalence. However, in this note we remind readers that the performance of an assay upon translation to the clinic is critically dependent upon that very same prevalence. Without an understanding of prevalence in the test population, even robust bioassays with excellent ROC characteristics may perform poorly in the clinic. Instead, simple plots of candidate assay performance as a function of prevalence rate give a more realistic understanding of the likely real-world performance and a greater understanding of the likely impact of variation in that prevalence on translational performance in the clinic.

25. Promoting Physics Literacy through Enquiry-Based Learning Online

Abstract

In Australia, as in a number of other countries, studies have consistently shown a low enrolment trend towards Physics by students in post-secondary years, due partly to the subject being perceived as conceptually difficult and abstract to grasp. In order to promote Physics literacy, continued opportunities such as online courses for students to engage in Physics education are necessary. For courses that are aimed at reaching out to students with little Physics background, the pedagogy needs to be considered carefully, especially when it is taught entirely in an online learning environment. This research investigated a fully online, inquiry-based course design aimed at motivating students to learn Physics and its impact on students' learning experiences at an Australian university. The research compared the learning experiences of students whose career trajectories are science-related and those who are not in order to assess its effectiveness in promoting Physics literacy. An online survey containing Likert-scale items as well as open questions elicited students' perceptions of the impact of the online course on their learning. The volunteer research participants were 59 undergraduates, where about two thirds of the participants were science students and one-third non-science students. The results showed that students were positive about the pedagogical structure and content in the online Physics course. Except for one item, there were no other statistically significant differences between science and non-science students' responses in the study, suggesting that the pedagogical design catered to the needs of both groups of students, an important element in promoting Physics literacy across a broad range of students.

32. The Wanago Program: An Innovative School-University STEM Partnership Program

Abstract

University-led science, technology, engineering and mathematics (STEM) outreach plays an important role in addressing declining rates of student participation in tertiary STEM programs and declining interest in pursuing careers in STEM fields (Sadler, Eilam, Bigger & Barry, 2018). One type of outreach program that is becoming more popular is the school-university partnership. This is often a formal arrangement between a university and a single school or a group of schools. Partnerships to support innovation in school science and mathematics are a key strategy of successful STEM countries (Marginson, Tytler, Freeman & Roberts, 2013). This article reports on a school-university partnership program, known as the Wanago program, located at the University of Technology Sydney (UTS) that allows secondary school students to pursue STEM subjects on the university's campus. The impact of the program in STEM studies for secondary school students, personnel and resources that supported student learning, and how participation in the program supported leaning outcomes for the students are explored.

41. Technology-Enhanced Science Partnership Initiative: Impact on Secondary Science Teachers

Abstract

The issue of student disengagement in school science continues to pose a threat to lifting the participation rates of students undertaking STEM courses and careers in Australia and other countries globally. In Australia, several science initiatives to reverse the problem have been funded over the last two decades. Many of these initiatives involve partnerships with scientists, science educators and with industries, as is the case in this paper. The research in this paper investigated a recent partnership initiative between secondary science teachers, scientists and an educational technology company to produce science e-modules on adaptive learning platforms, enabling students to engage in personalised, inquiry-based learning and the investigation of real-world problems. One of the objectives of the partnership project was to build theoretical and pedagogical skills in teachers to deliver science by exposing them to new ways of engaging students with new digital tools, for example analytics. Using a mixed methods approach, the research investigated science teachers' pedagogical involvement in the partnership project and their perceptions of the project's impact on their teaching and students' learning. The findings indicate that the teachers believed that new technology could enhance their teaching and students' learning and that while their students were motivated by the online modules, there was still a need for scaffolding for many of the students. The effectiveness of this would depend on the teachers' ability to internalise the new technological and content knowledge resulting from the partnership and realign them with their existing pedagogical framework. The research is significant in identifying elements for successful partnership projects as well as challenges that need to be considered. It is significant in facilitating continuous discourse about new evidence-based pedagogical approaches to science education in engaging students to learn STEM subjects in a twenty-first century digitally connected future that is focused on learning at a personal level.

Abstract

Objective: This study aimed to identify peripheral and salivary gland (SG) biomarkers of response/resistance to B cell depletion based on the novel concise Composite of Relevant Endpoints for Sjögren Syndrome (cCRESS) and candidate Sjögren Tool for Assessing Response (STAR) composite endpoints. Methods: Longitudinal analysis of peripheral blood and SG biopsies was performed pre‐ and post‐treatment from the Trial of Anti–B Cell Therapy in Patients With Primary Sjögren Syndrome (TRACTISS) combining flow cytometry immunophenotyping, serum cytokines, and SG bulk RNA sequencing. Results: Rituximab treatment prevented the worsening of SG inflammation observed in the placebo arm, by inhibiting the accumulation of class‐switched memory B cells within the SG. Furthermore, rituximab significantly down‐regulated genes involved in immune‐cell recruitment, lymphoid organization alongside antigen presentation, and T cell co‐stimulatory pathways. In the peripheral compartment, rituximab down‐regulated immunoglobulins and auto‐antibodies together with pro‐inflammatory cytokines and chemokines. Interestingly, patients classified as responders according to STAR displayed significantly higher baseline levels of C‐X‐C motif chemokine ligand‐13 (CXCL13), interleukin (IL)‐22, IL‐17A, IL‐17F, and tumor necrosis factor‐α (TNF‐α), whereas a longitudinal analysis of serum T cell–related cytokines showed a selective reduction in both STAR and cCRESS responder patients. Conversely, cCRESS response was better associated with biomarkers of SG immunopathology, with cCRESS‐responders showing a significant decrease in SG B cell infiltration and reduced expression of transcriptional gene modules related to T cell costimulation, complement activation, and Fcγ‐receptor engagement. Finally, cCRESS and STAR response were associated with a significant improvement in SG exocrine function linked to transcriptional evidence of SG epithelial and metabolic restoration. Conclusion: Rituximab modulates both peripheral and SG inflammation, preventing the deterioration of exocrine function with functional and metabolic restoration of the glandular epithelium. Response assessed by newly developed cCRESS and STAR criteria was associated with differential modulation of peripheral and SG biomarkers, emerging as novel tools for patient stratification.
