Abstract

A novel porcine intestinal in vitro organ culture (IVOC) model is developed by using cell crown (CC) technology (CCIVOC) (Scaffdex). Although Lactobacillus plantarum JC1 has not reduced the association of Salmonella Typhimurium strain SL1344 to the jejunal IVOC tissues, the detection of increased acidic mucin secretion, host cytoskeletal rearrangements, and proteins involved in the porcine immune response have shown that the strain of Lactobacillus plantarum has contributed to protecting the pig from infections by Salmonella Typhimurium or other pathogens.

21. Spectroscopic characterisation of the reaction centre of photosystem II from higher plants

Abstract

Two different photosystem II particles isolated from pea, a core complex and the D1/D2/Cyt b-559 reaction centre (RC) complex, have been characterised by absorption, linear dichroism (LD) and circular dichroism (CD) spectroscopy. Only one carotenoid contributes to the LD of the reaction centre, in agreement with the biochemical analysis. This carotenoid is oriented parallel to the long axis of the reaction centre. The chlorophyll Q Ycontribution to the LD is oriented perpendicular to the long axis of the reaction centre. The LD of the reaction centre carotenoid is reversed in the core complex. In addition, the contribution of a second carotenoid species can be observed. In the core, the two pools of carotenoid have a rather different orientation with respect to the membrane plane: one parallel, the other perpendicular. In addition, in the core the sign of the LD in the chlorophyll Q Yregion is reversed and red-shifted as compared to the reaction centre. These observations suggest that the reaction centre is oriented with its long axis perpendicular to the long axis of the PS II core and to the membrane plane. The circular dichroism CD of the reaction centre has intense peaks of opposite sign at 444 nm (negative) and 435 nm (positive), which we attribute to exciton coupling between Chl amolecules in the reaction centre. The RC has no CD in the range 460–650 nm; thus there is no exciton coupling between the carotenoid and the other pigments. The lack of CD in this region is consistent with the biochemical analysis of only one carotenoid per reaction centre. The larger core complex exhibits much weaker CD (per Chl). The CD in the Q Yabsorption band indicates exciton coupling between chlorophyll molecules. The sign of the pair of peaks in this region is reversed in the core with respect to the reaction centre.

Abstract

Abstract: Electron impact excitation of vibrational levels in the ground electronic state and seven excited electronic states in O2 have been simulated for an International Brightness Coefficient-Category 2+ (IBC II+) night-time aurora, in order to predict O2 excited state number densities and volume emission rates (VERs). These number densities and VERs are determined as a function of altitude (in the range 80–350km) in the present study. Recent electron impact excitation cross-sections for O2 were combined with appropriate altitude dependent IBC II+ auroral secondary electron distributions and the vibrational populations of the eight O2 electronic states were determined under conditions of statistical equilibrium. Pre-dissociation, atmospheric chemistry involving atomic and molecular oxygen, radiative decay and quenching of excited states were included in this study. This model predicts relatively high number densities for the metastable electronic states and could represent a significant source of stored energy in O2* for subsequent thermospheric chemical reactions. Particular attention is directed towards the emission intensities of the infrared (IR) atmospheric (1.27μm), Atmospheric (0.76μm) and the atomic oxygen 1 S→1 D transition (5577Å) lines and the role of electron-driven processes in their origin. Aircraft, rocket and satellite observations have shown both the IR atmospheric and Atmospheric lines are dramatically enhanced under auroral conditions and, where possible, we compare our results to these measurements. Our calculated 5577Å intensity is found to be in good agreement with values independently measured for a medium strength IBC II+ aurora. [Copyright &y& Elsevier]
