1. Focused Active Learning for Histopathological Image Classification

Abstract: Active Learning (AL) has the potential to solve a major problem of digital pathology: the efficient acquisition of labeled data for machine learning algorithms. However, existing AL methods often struggle in realistic settings with artifacts, ambiguities, and class imbalances, as commonly seen in the medical field. The lack of precise uncertainty estimations leads to the acquisition of images with a low informative value. To address these challenges, we propose Focused Active Learning (FocAL), which combines a Bayesian Neural Network with Out-of-Distribution detection to estimate different uncertainties for the acquisition function. Specifically, the weighted epistemic uncertainty accounts for the class imbalance, aleatoric uncertainty for ambiguous images, and an OoD score for artifacts. We perform extensive experiments to validate our method on MNIST and the real-world Panda dataset for the classification of prostate cancer. The results confirm that other AL methods are 'distracted' by ambiguities and artifacts which harm the performance. FocAL effectively focuses on the most informative images, avoiding ambiguities and artifacts during acquisition. For both experiments, FocAL outperforms existing AL approaches, reaching a Cohen's kappa of 0.764 with only 0.69% of the labeled Panda data.

2. The Debris of the 'Last Major Merger' is Dynamically Young

Abstract: The Milky Way's (MW) inner stellar halo contains an [Fe/H]-rich component with highly eccentric orbits, often referred to as the "last major merger." Hypotheses for the origin of this component include Gaia-Sausage/Enceladus (GSE), where the progenitor collided with the MW proto-disk 8-11 Gyr ago, and the Virgo Radial Merger (VRM), where the progenitor collided with the MW disk within the last 3 Gyr. These two scenarios make different predictions about observable structure in local phase space, because the morphology of debris depends on how long it has had to phase mix. The recently-identified phase-space folds in Gaia DR3 have positive caustic velocities, making them fundamentally different than the phase-mixed chevrons found in simulations at late times. Roughly 20\% of the stars in the prograde local stellar halo are associated with the observed caustics. Based on a simple phase-mixing model, the observed number of caustics are consistent with a merger that occurred 1--2 Gyr ago. We also compare the observed phase-space distribution to FIRE-2 Latte simulations of GSE-like mergers, using a quantitative measurement of phase mixing (2D causticality). The observed local phase-space distribution best matches the simulated data 1--2 Gyr after collision, and certainly not later than 3 Gyr. This is further evidence that the progenitor of the "last major merger" did not collide with the MW proto-disk at early times, as is thought for the GSE, but instead collided with the MW disk within the last few Gyr, consistent with the body of work surrounding the VRM.

3. Resting-State Functional Connectivity Reveals Differences in Large-Scale Network Interactions between Eminent and Non-Eminent Thinkers

Abstract: Prior work suggests that differences in brain morphology and task-evoked neural activity may underlie extraordinary creative achievement. Here, we extend these findings by focusing on resting blood oxygen level-dependent (rsBOLD) functional connectivity differences between eminent creators from diverse fields of expertise and a "smart" comparison group of age- and education-matched non-eminent thinkers. Participants underwent rsBOLD imaging, along with a high-resolution structural brain scan, and completed a series of demographic and behavioral measures. The results revealed significant differences in rsBOLD connectivity between the eminent and noneminent thinkers. Eminent thinkers showed increased connectivity between (a) the right central operculum and the bilateral parietal operculum, right supramarginal gyrus, and left precentral gyrus; (b) the right precentral gyrus and the right postcentral gyrus; (c) the left parietal operculum and the right central operculum and the right sensorimotor cortex; and (d) the posterior cingulate gyrus and the right precentral gyrus. In contrast, they showed decreased connectivity between (a) the posterior cingulate gyrus and the left supracalcarine cortex and (b) between the left occipital fusiform gyrus and the left frontal operculum. These results suggest that extraordinary creativity may be associated with measurable differences in functional connectivity, particularly in areas that support creative functions.

4. Exploring asymmetric substructures of the outer disk based on the conjugate angle of the radial action

Abstract: We use the conjugate angle of radial action ($\theta_R$), the best representation of the orbital phase, to explore the "mid-plane, north branch, south branch" and "Monoceros area" disk structures that were previously revealed in the LAMOST K giants (Xu et al. 2020). The former three substructures, identified by their 3D kinematical distributions, have been shown to be projections of the phase space spiral (resulting from nonequilibrium phase mixing). In this work, we find that all of these substructures associated with the phase spiral show high aggregation in conjugate angle phase space, indicating that the clumping in conjugate angle space is a feature of ongoing, incomplete phase mixing. We do not find the $Z-V_Z$ phase spiral located in the "Monoceros area", but we do find a very highly concentrated substructure in the quadrant of conjugate angle space with the orbital phase from the apocenter to the guiding radius. The existence of the clump in conjugate angle space provides a complementary way to connect the "Monoceros area" with the direct response to a perturbation from a significant gravitationally interactive event. 5. Primary Objective Grating Telescopy: Optical Properties and Feasibility of Applications

Abstract: We develop the theoretical foundation for primary objective grating (POG) telescopy. In recent years, a wide range of telescope designs that collect the light over a large grating and focus it with a secondary receiving optic that is placed at grazing exodus have been proposed by Thomas D. Ditto, and are sometimes referred to as Dittoscopes. Applications include discovery and characterization of exoplanets, discovery of near-Earth asteroids, and spectroscopic surveys of the sky. These telescopes would have small aerial mass, and therefore provide a path forward to launch large telescopes into space. Because this series of telescope designs departs from traditional telescope designs, it has been difficult to evaluate which applications are most advantageous for this design. Here, we define a new figure of merit, the "modified etendue," that characterizes the photon collection capability of a POG. It is demonstrated that the diffraction limit for observations is determined by the length of the grating. We evaluate the effects of atmospheric seeing for ground-based applications and the disambiguation of position vs. wavelength in the focal plane using a second dispersing element. 6. A Swing of the Pendulum: The Chemodynamics of the Local Stellar Halo Indicate Contributions from Several Radial Merger Events

Abstract: We find that the chemical abundances and dynamics of APOGEE and GALAH stars in the local stellar halo are inconsistent with a scenario in which the inner halo is primarily composed of debris from a single, massive, ancient merger event, as has been proposed to explain the Gaia-Enceladus/Gaia Sausage (GSE) structure. The data contains trends of chemical composition with energy which are opposite to expectations for a single massive, ancient merger event, and multiple chemical evolution paths with distinct dynamics are present. We use a Bayesian Gaussian mixture model regression algorithm to characterize the local stellar halo, and find that the data is best fit by a model with four components. We interpret these components as the VRM, Cronus, Nereus, and Thamnos; however, Nereus and Thamnos likely represent more than one accretion event because the chemical abundance distributions of their member stars contain many peaks. Although the Cronus and Thamnos components have different dynamics, their chemical abundances suggest they may be related. We show that the distinct low and high alpha halo populations from Nissen & Schuster (2010) are explained by VRM and Cronus stars, as well as some in-situ stars. Because the local stellar halo contains multiple substructures, different popular methods of selecting GSE stars will actually select different mixtures of these substructures, which may change the apparent chemodynamic properties of the selected stars. We also find that the Splash stars in the solar region are shifted to higher v_phi and slightly lower [Fe/H] than previously reported.

8. Bilateral Carotid Calcification Correlates with Regional Cerebral Glucose Metabolism: Insights from PET/CT Imaging of Patients with Cardiovascular Risk Factors

Abstract: Background: Cardiovascular disease is a leading cause of illness and death globally, primarily due to atherosclerosis. This disease reduces blood flow and oxygen delivery to organs, and when it affects the carotid arteries, it can lead to cognitive impairment and dementia. In a population of 104 individuals, comprising both healthy controls and individuals at elevated risk for developing cardiovascular diseases (CVD) due to identified risk factors, we used PET imaging with 18F-fluorodeoxyglucose (FDG) to assess cerebral glucose metabolism and 18F-sodium fluoride (NaF) to detect atherosclerotic calcification. Our statistical analysis revealed significant differences in metabolic activity between healthy and at-risk individuals in specific brain regions. 18F-FDG uptake in the brain varied inversely with respect to the clinical assessment of cardiovascular risk in regions such as the cuneus (β = −0.030, SE = 0.014, p = 0.035), middle occipital gyrus (β = −0.032, SE = 0.011, p = 0.005), and posterior cingulate gyrus (β = −0.032, SE = 0.015, p = 0.044). In contrast, areas including the basis pontis (β = 0.025, SE = 0.012, p = 0.038) and the pons (β = 0.034, SE = 0.013, p = 0.008) exhibited direct correlations. Notably, carotid 18F-NaF uptake had inverse associations with 18F-FDG uptake in the cerebellum (β = −0.825, SE = 0.354, p = 0.021), medulla (β = −0.888, SE = 0.405, p = 0.029), and posterior cingulate gyrus (β = −1.253, SE = 0.567, p = 0.028), while increased carotid calcification influenced metabolic activity in the fusiform gyrus (β = 1.660, SE = 0.498, p = 0.001) and globus pallidus (β = 1.505, SE = 0.571, p = 0.009). We observed that atherosclerotic plaque accumulation, especially in the carotid arteries, has potential implications for metabolic changes in brain regions governing cognition, emotion, sensory perception, and motor activities. Our findings underscore the possible early interventions that can be used to preempt or delay cognitive deterioration linked with cardiovascular ailments.

9. 60 candidate high-velocity stars originating from the Sagittarius dwarf spheroidal galaxy in Gaia EDR3

Abstract: Using proper motions from Gaia Early Data Release 3 (Gaia EDR 3) and radial velocities from several surveys, we identify 60 candidate high-velocity stars with total velocity greater than 75\% escape velocity that probably origin from Sagittarius dwarf spheroidal galaxy (Sgr) by orbital analysis. Sgr's gravity has little effect on the results and the Large Magellanic Cloud's gravity has non-negligible effect on only a few stars. The closest approach of these stars to the Sgr occurs when the Sgr passed its pericenter ($\sim$ 38.2 Myr ago), which suggest they were tidally stripped from the Sgr. The positions of these stars in the HR diagram and the chemical properties of 19 of them with available [Fe/H] are similar with the Sgr stream member stars. This is consistent with the assumption of their accretion origin. 11. Neuroimaging evaluation of the long term impact of a novel paired meditation practice on brain function

Abstract: BackgroundA growing number of advanced neuroimaging studies have compared brain structure and function in long term meditators to non-meditators. The goal is to determine if there may be long term effects on the brain from practicing meditation. In this paper, we present new data on the long term effects of a novel meditation practice in which the focus is on clitoral stimulation. The findings from such a study have implications for potential therapeutic uses with regard to various neurological or psychiatric conditions.MethodsWe evaluated the cerebral glucose metabolism in 40 subjects with an extended history (>1 year of practice, 2–3 times per week) performing the meditation practice called Orgasmic Meditation (OM) and compared their brains to a group of non-meditating healthy controls (N = 19). Both meditation and non-meditation subjects underwent brain PET after injection with 148 to 296 MBq of FDG using a standard imaging protocol. Resting FDG PET scans of the OM group were compared to the resting scans of healthy, non-meditating, controls using statistical parametric mapping.ResultsThe OM group showed significant differences in metabolic activity at rest compared to the controls. Specifically, there was significantly lower metabolism in select areas of the frontal, temporal, and parietal lobes, as well as the anterior cingulate, insula, and thalamus, in the OM group compared to the controls. In addition, there were notable distinctions between the males and females with the females demonstrating significantly lower metabolism in the thalamus and insula.ConclusionsOverall, these findings suggest that the long term meditation practitioners of OM have different patterns of resting brain metabolism. Since these areas of the brain in which OM practitioners differ from controls are involved in cognition, attention, and emotional regulation, such findings have implications for understanding how this meditation practice might affect practitioners over long periods of time.

Abstract: Mutationally activated BRAF is detected in approximately 7% of human lung adenocarcinomas, with BRAFT1799A serving as a predictive biomarker for treatment of patients with FDA-approved inhibitors of BRAFV600E oncoprotein signaling. In genetically engineered mouse (GEM) models, expression of BRAFV600E in the lung epithelium initiates growth of benign lung tumors that, without additional genetic alterations, rarely progress to malignant lung adenocarcinoma. To identify genes that cooperate with BRAFV600E for malignant progression, we used Sleeping Beauty-mediated transposon mutagenesis, which dramatically accelerated the emergence of lethal lung cancers. Among the genes identified was Rbms3, which encodes an RNA-binding protein previously implicated as a putative tumor suppressor. Silencing of RBMS3 via CRISPR/Cas9 gene editing promoted growth of BRAFV600E lung organoids and promoted development of malignant lung cancers with a distinct micropapillary architecture in BRAFV600E and EGFRL858R GEM models. BRAFV600E/RBMS3Null lung tumors displayed elevated expression of Ctnnb1, Ccnd1, Axin2, Lgr5, and c-Myc mRNAs, suggesting that RBMS3 silencing elevates signaling through the WNT/β-catenin signaling axis. Although RBMS3 silencing rendered BRAFV600E-driven lung tumors resistant to the effects of dabrafenib plus trametinib, the tumors were sensitive to inhibition of porcupine, an acyltransferase of WNT ligands necessary for their secretion. Analysis of The Cancer Genome Atlas patient samples revealed that chromosome 3p24, which encompasses RBMS3, is frequently lost in non-small cell lung cancer and correlates with poor prognosis. Collectively, these data reveal the role of RBMS3 as a lung cancer suppressor and suggest that RBMS3 silencing may contribute to malignant NSCLC progression.SignificanceLoss of RBMS3 cooperates with BRAFV600E to induce lung tumorigenesis, providing a deeper understanding of the molecular mechanisms underlying mutant BRAF-driven lung cancer and potential strategies to more effectively target this disease.

Abstract: Differential mRNA expression between ancestry groups can be explained by both genetic and environmental factors. We outline a computational workflow to determine the extent to which germline genetic variation explains cancer-specific molecular differences across ancestry groups. Using multi-omics datasets from The Cancer Genome Atlas (TCGA), we enumerate ancestry-informative markers colocalized with cancer-type-specific expression quantitative trait loci (e-QTLs) at ancestry-associated genes. This approach is generalizable to other settings with paired germline genotyping and mRNA expression data for a multi-ethnic cohort. 15. Estimate of the Mass and Radial Profile of the Orphan-Chenab Stream's Dwarf Galaxy Progenitor Using MilkyWay@home

Abstract: We fit the mass and radial profile of the Orphan-Chenab Stream's (OCS) dwarf galaxy progenitor by using turnoff stars in the Sloan Digital Sky Survey (SDSS) and the Dark Energy Camera (DEC) to constrain N-body simulations of the OCS progenitor falling into the Milky Way on the 1.5 PetaFLOPS MilkyWay@home distributed supercomputer. We infer the internal structure of the OCS's progenitor under the assumption that it was a spherically symmetric dwarf galaxy comprised of a stellar system embedded in an extended dark matter halo. We optimize the evolution time, the baryonic and dark matter scale radii, and the baryonic and dark matter masses of the progenitor using a differential evolution algorithm. The likelihood score for each set of parameters is determined by comparing the simulated tidal stream to the angular distribution of OCS stars observed in the sky. We fit the total mass of the OCS's progenitor to ($2.0\pm0.3$) $\times 10^7 M_\odot$ with a mass-to-light ratio of $\gamma=73.5\pm10.6$ and ($1.1\pm0.2$)$\times10^6M_{\odot}$ within 300 pc of its center. Within the progenitor's half-light radius, we estimate total a mass of ($4.0\pm1.0$)$\times10^5M_{\odot}$. We also fit the current sky position of the progenitor's remnant to be $(\alpha,\delta)=((166.0\pm0.9)^\circ,(-11.1\pm2.5)^\circ)$ and show that it is gravitationally unbound at the present time. The measured progenitor mass is on the low end of previous measurements, and if confirmed lowers the mass range of ultrafaint dwarf galaxies. Abstract: Abstract RNA splicing is an important biological process associated with cancer initiation and progression. However, the contribution of alternative splicing to pancreatic cancer (PDAC) development is not well understood. Here, we identify an enrichment of RNA binding proteins (RBPs) involved in splicing regulation linked to PDAC progression from a forward genetic screen using Sleeping Beauty insertional mutagenesis in a mouse model of pancreatic cancer. We demonstrate downregulation of RBFOX2, an RBP of the FOX family, promotes pancreatic cancer progression and liver metastasis. Specifically, we show RBFOX2 regulates exon splicing events in transcripts encoding proteins involved in cytoskeletal remodeling programs. These exons are differentially spliced in PDAC patients, with enhanced exon skipping in the classical subtype for several RBFOX2 targets. RBFOX2 mediated splicing of ABI1, encoding the Abelson-interactor 1 adapter protein, controls the abundance and localization of ABI1 protein isoforms in pancreatic cancer cells and promotes the relocalization of ABI1 from the cytoplasm to the periphery of migrating cells. Using splice-switching antisense oligonucleotides (AONs) we demonstrate the ABI1 ∆Ex9 isoform enhances cell migration. Together, our data identify a role for RBFOX2 in promoting PDAC progression through alternative splicing regulation.

Abstract: We have developed a method for estimating the properties of the progenitor dwarf galaxy from the tidal stream of stars that were ripped from it as it fell into the Milky Way. In particular, we show that the mass and radial profile of a progenitor dwarf galaxy evolved along the orbit of the Orphan Stream, including the stellar and dark matter components, can be reconstructed from the distribution of stars in the tidal stream it produced. We use MilkyWay@home, a PetaFLOPS-scale distributed supercomputer, to optimize our dwarf galaxy parameters until we arrive at best-fit parameters. The algorithm fits the dark matter mass, dark matter radius, stellar mass, radial profile of stars, and orbital time. The parameters are recovered even though the dark matter component extends well past the half light radius of the dwarf galaxy progenitor, proving that we are able to extract information about the dark matter halos of dwarf galaxies from the tidal debris. Our simulations assumed that the Milky Way potential, dwarf galaxy orbit, and the form of the density model for the dwarf galaxy were known exactly; more work is required to evaluate the sources of systematic error in fitting real data. This method can be used to estimate the dark matter content in dwarf galaxies without the assumption of virial equilibrium that is required to estimate the mass using line-of-sight velocities. 18. The Local Stellar Halo is Not Dominated by a Single Radial Merger Event

Abstract: We use halo dwarf stars with photometrically determined metallicities that are located within 2 kpc of the Sun to identify local halo substructure. The kinematic properties of these stars do not indicate a single, dominant radial merger event (RME). The retrograde Virgo Radial Merger (VRM) component has [Fe/H] = -1.7. A second, non-rotating RME component we name Nereus is identified with [Fe/H] = -2.1 and has similar energy as the VRM. A possible third RME we name Cronus is identified that is co-rotating with the disk, has lower energy than the VRM, and has [Fe/H] = -1.2. We identify the Nyx Stream in the data. In addition to these substructures, we observe metal-poor halo stars ([Fe/H] ~ -2.0 and $\sigma_v$ ~ 180 km s$^{-1}$) and a disk/Splash component with lower rotational velocity than the disk and lower metallicity than typically associated with the Splash. An additional excess of halo stars with low velocity and metallicity of [Fe/H] = -1.5 could be associated with the shell of a lower energy RME or indicate that lower energy halo stars have higher metallicity. Abstract: Abstract Somatic PTEN mutations are common and have driver function in some cancer types. However, in colorectal cancers (CRCs), somatic PTEN-inactivating mutations occur at a low frequency (~8–9%), and whether these mutations are actively selected and promote tumor aggressiveness has been controversial. Analysis of genomic data from ~53,000 CRCs indicates that hotspot mutation patterns in PTEN partially reflect DNA-dependent selection pressures, but also suggests a strong selection pressure based on protein function. In microsatellite stable (MSS) tumors, PTEN alterations co-occur with mutations activating BRAF or PI3K, or with TP53 deletions, but not in CRC with microsatellite instability (MSI). Unexpectedly, PTEN deletions are associated with poor survival in MSS CRC, whereas PTEN mutations are associated with improved survival in MSI CRC. These and other data suggest use of PTEN as a prognostic marker is valid in CRC, but such use must consider driver mutation landscape, tumor subtype, and category of PTEN alteration.

Abstract: Although Blue Horizontal Branch (BHB) stars are commonly used to trace halo substructure, the stars bluer than (g-r)<-0.3 are ignored due to the difficulty in determining their absolute magnitudes. The blue extention of the horizontal branch (HBX) includes BHB tail stars and Extreme Horizontal Branch (EHB) stars. We present a method for identifying HBX stars in the field, using spectra and photometry from the Sloan Digital Sky Survey Data Release 14 (SDSS DR14). We derive an estimate for the absolute magnitudes of BHB tail and EHB stars as a function of color, and use this relationship to calculate distances. We identify an overdensity of HBX stars that appears to trace the northern end of the Hercules-Aquila Cloud (HAC). We identify three stars that are likely part of a tidal stream, but this is not enough stars to explain the observed overdensity. Combining SDSS data with Gaia DR2 proper motions allows us to show that the orbits of the majority of the HBX stars in the overdensity are on high eccentricity orbits similar to those in the Virgo Radial Merger/Gaia-Enceladus/Gaia Sausage structure, and that the overdensity of high eccentricity orbits extends all the way to the Virgo Overdensity. We use stellar kinematics to separate the HBX stars into disk stars andhalo stars. The halo stars are primarily on highly eccentric (radial) orbits. Abstract: Background The relationship between sexuality, or the libido, and spirituality or religion has long been debated in psychiatry. Recent studies have explored the neurophysiology of both sexual experiences and spiritual practices such as meditation or prayer. In the present study, we report changes in cerebral glucose metabolism in a unique meditation practice augmented by clitoral stimulation called, Orgasmic Meditation, in which a spiritual state is described to be attained by both male and female participants engaged in the practice as a pair. Methods Male (N=20) and female (N=20) subjects had an intravenous catheter connected to a bag of normal saline inserted prior to the practice. During the practice, men stimulated their partner’s clitoris for exactly 15 minutes (he received no sexual stimulation). Midway through the practice, researchers injected 18F-fluorodeoxyglucose so the scan would reflect cerebral metabolism during the practice. Positron emission tomography (PET) imaging was performed approximately 30 minutes later. Results In the female participants, the meditation state showed significant decreases in the left inferior frontal, inferior parietal, insula, middle temporal, and orbitofrontal regions as well as in the right angular gyrus, anterior cingulate and parahippocampus compared to a neutral state (p Conclusions These findings represent a distinct pattern of brain activity, for both men and women, that is a hybrid between that of other meditation practices and sexual stimulation. 27. Promoting healthful and diverse eating behaviours through an extracurricular culinary skills intervention in Philadelphia

Abstract: In the current study we evaluated an afterschool nutrition education programme, called Vetri Cooking Lab (VCL), for promoting healthy and diverse eating habits among at-risk children in the Greater Philadelphia area. To understand potential programme impacts, we conducted a longitudinal analysis of survey data collected before and after participation in VCL. Main study included cooking confidence, cooking knowledge, changes in dietary consumption behaviours, and changes in vegetable preferences. Participants included students in grades 3–11 enrolled in VCL during the 2018–19 school year at VCL sites (n = 60) throughout Philadelphia, PA, and Camden, NJ. Eligible participants completed surveys both before and after participating in the programme. We found that students’ confidence and knowledge increased (P < 0.001) after the cooking intervention. Knowledge and confidence were positively associated (r = 0.55; P < 0.001). Confidence was correlated with consumption behaviour changes (r = 0.18; P = 0.022). Confidence was positively associated with consumption changes in both our adjusted (OR = 1.81; P < 0.001) and unadjusted models (aOR = 1.88; P = 0.013). Compared to Black students, White students were more likely to report consumption changes (aOR = 5.83; P = 0.013). Hispanic/Latino participants and participants who spoke Spanish had nearly three times higher odds of consumption behaviour changes (Hispanic/Latino OR = 2.55; P = 0.007; Spanish OR = 3.04; P = 0.005). Student age and gender were not associated with behaviour changes. Our research demonstrates that programmes integrating practical cooking skills education along with nutrition, food, and cooking education can improve confidence and knowledge about healthy food choices amongst children driving an overall improvement in children’s eating habits.

Abstract: Mild traumatic brain injury (mTBI) is a public health concern. The present study aimed to develop an automatic classifier to distinguish between patients with chronic mTBI (n = 83) and healthy controls (HCs) (n = 40). Resting-state functional MRI (rs-fMRI) and positron emission tomography (PET) imaging were acquired from the subjects. We proposed a novel deep-learning-based framework, including an autoencoder (AE), to extract high-level latent and rectified linear unit (ReLU) and sigmoid activation functions. Single and multimodality algorithms integrating multiple rs-fMRI metrics and PET data were developed. We hypothesized that combining different imaging modalities provides complementary information and improves classification performance. Additionally, a novel data interpretation approach was utilized to identify top-performing features learned by the AEs. Our method delivered a classification accuracy within the range of 79–91.67% for single neuroimaging modalities. However, the performance of classification improved to 95.83%, thereby employing the multimodality model. The models have identified several brain regions located in the default mode network, sensorimotor network, visual cortex, cerebellum, and limbic system as the most discriminative features. We suggest that this approach could be extended to the objective biomarkers predicting mTBI in clinical settings.

Abstract: Mild traumatic brain injury (mTBI) is a significant public health concern, specially characterized by a complex pattern of abnormal neural activity and functional connectivity. It is often associated with a broad spectrum of short-term and long-term cognitive and behavioral symptoms including memory dysfunction, headache, and balance difficulties. Furthermore, there is evidence that oxidative stress significantly contributes to these symptoms and neurophysiological changes. The purpose of this study was to assess the effect of N-acetylcysteine (NAC) on brain function and chronic symptoms in mTBI patients. Fifty patients diagnosed with chronic mTBI participated in this study. They were categorized into two groups including controls (CN, n = 25), and patients receiving treatment with N-acetyl cysteine (NAC, n = 25). NAC group received 50 mg/kg intravenous (IV) medication once a day per week. In the rest of the week, they took one 500 mg NAC tablet twice per day. Each patient underwent rs-fMRI scanning at two timepoints including the baseline and 3 months later at follow-up, while the NAC group received a combination of oral and IV NAC over that time. Three rs-fMRI metrics were measured including fractional amplitude of low frequency fluctuations (fALFF), degree centrality (DC), and functional connectivity strength (FCS). Abstract: The recent discovery of a spiral feature in $Z-V_Z$ phase plane in the solar neighborhood implies that the Galactic disk has been remarkably affected by a dwarf galaxy passing through it some hundreds of millions of years ago. Using 429,500 LAMOST K giants stars, we show that the spiral feature exits not only in the solar vicinity; it also extends to about 15 kpc from the Galactic center, and then disappears beyond this radius. Moreover, we find that when the spiral features in a plot of $V_\phi$ as a function of position in the $Z-V_Z$ plane, at various Galactocentric radii, are re-mapped to $R-Z$ plane, the spiral can explain well the observed asymmetric velocity substructures. This is evidence that the phase spiral features are the same as the bulk motions found in previous as well as this work. Test-particle simulations and N-body simulations show that an encounter with a dwarf galaxy a few hundred million years ago will induce a perturbation in the Galactic disk. In addition, we find that the last impact of Sgr dSph can also contribute to the flare. 32. Existence of the Metal-Rich Stellar Halo and High-velocity Thick Disk in the Galaxy

Abstract: Based on the second Gaia data release (DR2), combined with the LAMOST and APOGEE spectroscopic surveys, we study the kinematics and metallicity distribution of the high-velocity stars that have a relative speed of at least 220 ${\rm km\ s^{-1}}$ with respect to the local standard of rest in the Galaxy. The rotational velocity distribution of the high-velocity stars with [Fe/H] $>-1.0$ dex can be well described by a two-Gaussian model, with peaks at $V_{\phi}\sim +164.2\pm0.7$ and $V_{\phi}\sim +3.0\pm0.3$ ${\rm km\ s^{-1}}$, associated with the thick disk and halo, respectively. This implies that there should exist a high-velocity thick disk (HVTD) and a metal-rich stellar halo (MRSH) in the Galaxy. The HVTD stars have the same position as the halo in the Toomre diagram and but show the same rotational velocity and metallicity as the canonical thick disk. The MRSH stars have basically the same rotational velocity, orbital eccentricity, and position in the Lindblad and Toomre diagram as the canonical halo stars, but they are more metal-rich. Furthermore, the metallicity distribution function (MDF) of our sample stars are well fitted by a four-Gaussian model, associated with the outer-halo, inner-halo, MRSH, and HVTD, respectively. Timing arguments for these shells indicate that their progenitor dwarf galaxy passed through the Galactic center 2.7 +/- 0.2 Gyr ago. Based on the time of collision, it is also possible that the VRM is related to the phenomenon that created phase-space spirals in the vertical motion of the disk and/or the Splash, and could have caused a burst of star formation in the inner disk. We analyze phase mixing in a collection of radial merger N-body simulations, and find that shell structure similar to that observed in Milky Way data disappears by 5 Gyr after collision with the Galactic center. The method used to calculate the merger time of the VRM was able to reliably recover the correct merger times for these simulations. Previous work supports the idea that the VRM and the Gaia Sausage/Gaia-Enceladus Merger are the same. However, the Gaia Sausage is widely believed to be 8--11 Gyr old. The disparate ages could be reconciled if the larger age is associated with an infall time when the progenitor crossed the virial radius; we do not constrain the time at which the progenitor became bound to the Milky Way. Alternatively, the Gaia Sausage could be younger than previously thought.

Abstract: Summary: Background: While vaccines have established utility against COVID-19, phase 3 efficacy studies have generally not comprehensively evaluated protection provided by previous infection or hybrid immunity (previous infection plus vaccination). Individual patient data from US government-supported harmonized vaccine trials provide an unprecedented sample population to address this issue. We characterized the protective efficacy of previous SARS-CoV-2 infection and hybrid immunity against COVID-19 early in the pandemic over three-to six-month follow-up and compared with vaccine-associated protection. Methods: In this post-hoc cross-protocol analysis of the Moderna, AstraZeneca, Janssen, and Novavax COVID-19 vaccine clinical trials, we allocated participants into four groups based on previous-infection status at enrolment and treatment: no previous infection/placebo; previous infection/placebo; no previous infection/vaccine; and previous infection/vaccine. The main outcome was RT-PCR-confirmed COVID-19 >7–15 days (per original protocols) after final study injection. We calculated crude and adjusted efficacy measures. Findings: Previous infection/placebo participants had a 92% decreased risk of future COVID-19 compared to no previous infection/placebo participants (overall hazard ratio [HR] ratio: 0.08; 95% CI: 0.05–0.13). Among single-dose Janssen participants, hybrid immunity conferred greater protection than vaccine alone (HR: 0.03; 95% CI: 0.01–0.10). Too few infections were observed to draw statistical inferences comparing hybrid immunity to vaccine alone for other trials. Vaccination, previous infection, and hybrid immunity all provided near-complete protection against severe disease. Interpretation: Previous infection, any hybrid immunity, and two-dose vaccination all provided substantial protection against symptomatic and severe COVID-19 through the early Delta period. Thus, as a surrogate for natural infection, vaccination remains the safest approach to protection. Funding: National Institutes of Health.

40. Dynamically Produced Moving Groups in Interacting Simulations

Abstract: We show that Smoothed Particle Hydrodynamics (SPH) simulations of dwarf galaxies interacting with a Milky Way-like disk produce moving groups in the simulated stellar disk. We analyze three different simulations: one that includes dwarf galaxies that mimic the Large Magellanic Cloud, Small Magellanic Cloud and the Sagittarius dwarf spheroidal; another with a dwarf galaxy that orbits nearly in the plane of the Milky Way disk; and a null case that does not include a dwarf galaxy interaction. We present a new algorithm to find large moving groups in the $V_R, V_\phi$ plane in an automated fashion that allows us to compare velocity sub-structure in different simulations, at different locations, and at different times. We find that there are significantly more moving groups formed in the interacting simulations than in the isolated simulation. A number of dwarf galaxies are known to orbit the Milky Way, with at least one known to have had a close pericenter approach. Our analysis of simulations here indicates that dwarf galaxies like those orbiting our Galaxy produce large moving groups in the disk. Our analysis also suggests that some of the moving groups in the Milky Way may have formed due to dynamical interactions with perturbing dwarf satellites. 41. New nearby hypervelocity stars and their spatial distribution from Gaia DR2

Abstract: Base on about 4,500 large tangential velocity ($V_\mathrm{tan}>0.75V_\mathrm{esc}$) with high-precision proper motions and $5\sigma$ parallaxes in Gaia DR2 5D information derived from parallax and proper motion, we identify more than 600 high velocity stars with $50\%$ unbound probability. Of these, 28 nearby (less than 6 kpc) late-type Hypervelocity stars (HVSs) with over $99\%$ possibility of unbound are discovered. In order to search for the unbound stars from the full Gaia DR2 6D phase space information derived from parallax, proper motion and radial velocity, we also identify 28 stars from the total velocity ($V_\mathrm{gc}>0.75V_\mathrm{esc}$) that have probabilities greater than $50\%$ of being unbound from the Galaxy. Of these, only three have a nearly $99\%$ probabilities of being unbound. On the whole HVSs subsample, there is 12 sources reported by other surveys. We study the spatial distribution of angular positions and angular separation of HVSs. We find the unbound HVSs are spatially anisotropic that is most significant in the Galactic longitude at more than $3\sigma$ level, and lower unbound probability HVSs are systematically more isotropic. 42. The Virgo Overdensity Explained

Abstract: We suggest that the Virgo Overdensity (VOD) of stars in the stellar halo is the result of a radial dwarf galaxy merger that we call the Virgo Radial Merger. Because the dwarf galaxy passed very near to the Galactic center, the debris has a large range of energies but nearly zero $L_z$ angular momentum. The debris appears to extend from 5 to 50 kpc from the Sun in the Virgo region. We connect different parts of this merger debris to the Perpendicular and Parallel Streams (the Virgo Stellar Stream is associated with either or both of these streams), the Hercules-Aquila Cloud (HAC), and possibly the Eridanus Phoenix Overdensity (EriPhe). This radial merger can explain the majority of the observed moving groups of RR Lyrae and blue horizontal branch stars that have previously been identified in Virgo. This merger also produces debris in the Solar neighborhood similar to that identified as the Gaia-Enceladus or it Gaia-sausage merger. 43. A Cold Stellar Stream in Pegasus

Abstract: We report the serendipitous discovery of a stellar stream in the constellation Pegasus in the south Galactic hemisphere. The stellar stream was detected using the SDSS Data Release 14 by means of a matched filter in the color--magnitude diagram that is optimised for a stellar population that is 8 Gyr old with [Fe/H] = $-$0.46 dex, and located at heliocentric distance of 18 kpc. The candidate stream is faint (turnoff point at $r_0 \sim$ 19.6), sparse and barely visible in SDSS photometry. It is also detected in the (shallower) Pan-STARRs data. The residual stellar density in the $(u-g)_0$, $(g-r)_0$ color--color diagram gives the same estimate for the age and [Fe/H] of this stellar population. The stream is located at a Galactic coordinates $(l,b) = (79.4,-24.6)$ and extends over 9$^\circ$ (2.5 kpc), with a width of 112 pc. 44. An Orbit Fit to Likely Hermus Stream Stars

Abstract: We selected blue horizontal branch (BHB) stars within the expected distance range and sky position of the Hermus Stream from Data Release 10 of the Sloan Digital Sky Survey. We identify a moving group of $19$ BHB stars that are concentrated within two degrees of the Hermus Stream, between $10$ and $14$ kpc from the Sun. The concentration in velocity is inconsistent with a Gaussian distribution with 98% confidence (2.33 sigma).The stars in the moving group have line-of-sight velocities of v$_{\rm gsr} \sim 50$ km s$^{-1}$, a velocity dispersion of $\sigma_v\lesssim11$ km s$^{-1}$, a line-of-sight depth of $\sim 1$ kpc, and a metallicity of [Fe/H] $=-2.1 \pm 0.4$. The best-fit orbit has a perigalacticon of $\sim4$ kpc, apogalacticon of $\sim 17$ kpc, orbital period of $\sim247$ Myr, eccentricity $e = 0.62$, and inclination $i \sim75^\circ$ from $b=90^\circ$. The BHB stars in the stream are estimated to be $12$ Gyr old. An N-body simulation of a mass-follows-light ultrafaint dwarf galaxy with mass $10^6M_{\odot}$ and radius $40$ pc is consistent with the observed properties. The properties of the identified moving group of 19 BHB stars are close enough to those of the Hermus Stream (which is traced predominantly in turnoff stars) that we find it likely that they are associated. Abstract: Based on the second Gaia data release (Gaia DR2) and spectroscopy from the Large Sky Area Multi-Object Fiber Spectroscopic Telescope (LAMOST) Data, we identified 20,089 halo stars kinematically and chemically. The halo streams in the solar neighborhood could be detected in the space of energy and angular momentum. We reshuffle the velocities of these stars to determine the significance of substructure. Finally, we identify 4 statistically significant substructures that are labeled GL-1 through 4. Among these substructures, GL-1 is previously known stream ("N2" stream) and the rest 3 substructures are new. Abstract: PurposeAfrican American (AFR) men have the highest mortality rate from prostate cancer (PCa) compared with men of other racial/ancestral groups. Differences in the spectrum of somatic genome alterations in tumors between AFR men and other populations have not been well-characterized due to a lack of inclusion of significant numbers in genomic studies.Experimental designTo identify genomic alterations associated with race, we compared the frequencies of somatic alterations in PCa obtained from four publicly available datasets comprising 250 AFR and 611 European American (EUR) men and a targeted sequencing dataset from a commercial platform of 436 AFR and 3018 EUR men.ResultsMutations in ZFHX3 as well as focal deletions in ETV3 were more frequent in tumors from AFR men. TP53 mutations were associated with increasing Gleason score. MYC amplifications were more frequent in tumors from AFR men with metastatic PCa, whereas deletions in PTEN and rearrangements in TMPRSS2-ERG were less frequent in tumors from AFR men. KMT2D truncations and CCND1 amplifications were more frequent in primary PCa from AFR men. Genomic features that could impact clinical decision making were not significantly different between the two groups including tumor mutation burden, MSI status, and genomic alterations in select DNA repair genes, CDK12, and in AR.ConclusionsAlthough we identified some novel differences in AFR men compared with other populations, the frequencies of genomic alterations in current therapeutic targets for PCa were similar between AFR and EUR men, suggesting that existing precision medicine approaches could be equally beneficial if applied equitably.

Abstract: We evaluated ancestry effects on mutation rates, DNA methylation, and mRNA and miRNA expression among 10,678 patients across 33 cancer types from The Cancer Genome Atlas. We demonstrated that cancer subtypes and ancestry-related technical artifacts are important confounders that have been insufficiently accounted for. Once accounted for, ancestry-associated differences spanned all molecular features and hundreds of genes. Biologically significant differences were usually tissue specific but not specific to cancer. However, admixture and pathway analyses suggested some of these differences are causally related to cancer. Specific findings included increased FBXW7 mutations in patients of African origin, decreased VHL and PBRM1 mutations in renal cancer patients of African origin, and decreased immune activity in bladder cancer patients of East Asian origin.

50. The origin of high velocity stars from Gaia and LAMOST

Abstract: Based on the second Gaia data (Gaia DR2) and spectroscopy from the LAMOST Data Release 5, we defined the high-velocity (HiVel) stars sample as those stars with $v_{\mathrm{gc}} > 0.85 v_{\mathrm{esc}}$, and derived the final sample of 24 HiVel stars with stellar astrometric parameters and radial velocities. Most of the HiVel stars are metal-poor and $\alpha$-enhanced. In order to further explore the origin of these HiVel stars, we traced the backwards orbits of each HiVel star in the Galactic potential to derive probability parameters which are used to classify these HiVel stars.
