Trabalho Final de Mestrado para obtenção do grau de Mestre em Engenharia Civil na Área de Especialização em Estruturas O presente trabalho pretende aprofundar o estudo da acção do vento, analisando a quantificação da sua acção a pontes reais. Consiste na discussão da aplicação da norma NP EN 1991-1-4 (Eurocódigo 1 – Acções em estruturas; Parte 1-4: Acções gerais; Acções do vento) e na discussão das diferenças dos resultados obtidos em relação ao Regulamento de Segurança e Acções para Estruturas de Edifícios e Pontes (RSA), nomeadamente quanto aos valores característicos da acção do vento. São explicados os principais factores que influenciam a acção do vento em pontes, referenciando diversos estudos recentes e baseando-se nos regulamentos em análise. É feito um estudo cuidadoso na quantificação da acção do vento segundo o RSA e segundo a NP EN 1991-1-4, em que se discutem e analisam as diferenças encontradas. Em seguida quantifica-se a acção do vento em exemplos reais, usando os dois regulamentos. A comparação dos resultados obtidos permite verificar que, no caso específico de Portugal, apesar do período de retorno da NP EN 1991-1-4 ser bastante inferior ao do RSA, no Anexo Nacional da NP EN 1991-1-4 os valores relativos à quantificação da acção do vento, nomeadamente no valor básico da velocidade de referência, nas categorias de rugosidade do terreno e nos valores característicos da pressão dinâmica da NP EN 1991-1-4, são da mesma ordem de grandeza ou até superiores aos apresentados no RSA. Abstract: The present work has the goal of a further study of the action of the wind, analyzing the quantification of their action to real bridges. It consists in the discussion of the application of the norm NP EN 1991-1-4 (Eurocode 1 – Actions on structures; Part 1-4: General actions; Wind actions) and the discussion in the differences of the results obtain by the RSA (Buildings and Bridges Safety and Actions Design Code), regarding to the characteristic values of the wind action. It is explained the major factors that influence the action of the wind in bridges, on which are referenced by several recent studies. There is still one explanation, concerning to the regulations in study that make to the referred factors. It is made a careful study in the quantification of the action of the wind according to the RSA and the NP EN 1991-1-4. The differences found are discussed and analyzed. Then, the wind action is quantified to real examples, using the two norms (RSA and NP EN 1991-1-4). The comparison of the results obtained allow to verify that, in the specific case of Portugal, despite the return period of the NP EN 1991-1-4 being far lower than the RSA, in the National Annex of the NP EN 1991-1-4 the characteristic values of the wind action, particularly in the basic value of the reference wind speed, terrain roughness categories and in the characteristic values of peak velocity pressure of the NP EN 1991-1-4, are the same order of magnitude or even higher than those presented in the RSA.

Dissertação de mestrado em Ecologia, apresentada ao Departamento de Ciências da Vida da Faculdade de Ciências e Tecnologia da Universidade de Coimbra Nos estuários, a pesca ilegal é uma das principais atividades que afeta uma vasta gama de espécies conduzindo ao seu declínio, provocando danos nas suas populações e levando à sobre-exploração dos juvenis das espécies-alvo e não-alvo (pesca acessória). Nos estuários do Nordeste Atlântico, os juvenis de enguia-europeia (Anguilla anguilla) são um alvo preferencial durante a fase de enguia de vidro. Em Portugal, e apesar de a sua pesca não ser permitida (com a exceção do Rio Minho), uma quantidade substancial de pesca ilegal ocorre durante a estação migratória. Este caso de estudo teve como objetivo estudar os impactos da pesca ilegal da enguia de vidro no estuário do Mondego (Portugal) em espécies alvo e não-alvo. Com este trabalho, os objetivos específicos, e tendo em conta dois cenários diferentes em termos hidrológicos (ano regular vs chuvoso), foram: a) determinar a variabilidade sazonal e inter-anual das capturas de enguias de vidro e das espécies não-alvo, e b) estimar a quantidade total de capturas considerando vários cenários hidrológicos e de pressão de pesca. A amostragem foi realizada entre novembro de 2011 e março 2013. As capturas de enguia de vidro foram semelhantes entre os dois anos de estudo. No entanto, ocorreram diferenças entre as duas campanhas de pesca, com um maior número de capturas acessórias em 2012-2013 (ano chuvoso), especialmente no outono. Os resultados também indicam uma maior diversidade de espécies no ano chuvoso, durante os meses de outono, bem como o maior número de indivíduos. Capturas mais elevadas das espécies mais abundantes foram também observados durante o ano chuvoso, quando comparado com o ano regular, o que sugere que a magnitude do impacto destas práticas ilegais pode estar relacionada com os ciclos hidrológicos. O trabalho efectuado demonstra que práticas ilegais e não-seletivas tais como esta têm um impacto significativo nas espécies capturadas, e em última análise no funcionamento do ecossistema estuarino. In estuaries, illegal fishing is one of main activities that affects a wide range of species leading to the decline of total abundances, damage in fish stocks and overexploitation of juveniles in both the target and non-target species (by-catch). In North-eastern Atlantic estuaries, the European eel (Anguilla anguilla) juveniles are particularly targeted during the glass eel stage. In Portugal, and despite that fishing eel juveniles is not allowed (with the exception of the Minho River), a substantial amount of illegal fishing takes place during the migratory season. Our case study aimed at studying the impacts of glass eel illegal fishing in the Mondego estuary (Portugal) in both target and non-target species. With this work, the specific objectives, considering two different hydrological scenarios (regular vs rainy year), were; a) to determine the seasonal and interannual variability in glass eels and non-target species catches, and b) to estimate the total amount of catches considering several hydrological and fishing pressure scenarios. Sampling was performed in the Mondego estuary between November 2011 and March 2013 during the autumn and winter. Results showed differences between the two fishing seasons: a higher number of bycatch was observed in 2012-2013 (rainy year), particularly in autumn. For the glass eel, no differences between years were observed. Results also indicated higher species diversity in the rainy year, during the autumn months, as well as the highest number of individuals. Higher catches of the most abundant species were also observed in the rainy year, when compared to the regular one, suggesting that the magnitude of impact of these illegal practices is related with the hydrological cycles. The present work shows that Illegal and non-selective fishing practices such as this one have high impacts on the captures species, and ultimately on the functioning of the estuarine ecosystem.

Mestrado em Contabilidade e Auditoria Esta dissertação descreve os mecanismos de controlo interno que são utilizados na prática pelas empresas do concelho de Aveiro. Nestes mecanismos incluem-se práticas comuns, principais procedimentos, hábitos e lacunas de controlo, bem como sensibilidade das organizações face a esta temática. Numa primeira parte começa-se por fazer um enquadramento teórico do tema, depois passa-se para o estudo das suas condicionantes. De seguida, analisase os cinco componentes base do controlo interno. Nos dois últimos pontos desta parte, aborda-se as limitações de controlo interno e a responsabilidade da sua implementação e manutenção. Numa segunda parte, efectua-se a análise prática aos obtidos, com base num inquérito dirigido às empresas da região, no sentido de se aferir a posição das mesmas relativamente ao sistema de controlo interno. Com base nos resultados obtidos apresenta-se diversas conclusões. ABSTRACT: This dissertation describes the mechanisms of internal control which are used by companies of the county of Aveiro. These mechanisms include up common practices, key procedures, habits and weaknesses of control, as well as sensitivity of the organizations facing this issue. The first part starts with a theoretical framework of the theme, and than the analyses of its constraints. The next step is the study of the five basic components of internal control. In the last two chapters of this part, the limitations of internal control and responsibility of their implementation and maintenance are presented. After the theoretical approach, and based in a questionnaire of intermal control targeted to companies of the county of Aveiro, the results are discussed, interpreted and analised, in order to present the final conclusions.

Protein–protein interactions (PPIs) are emerging drug targets, which show promise for the treatment of, e.g., cancer, but are still underexploited for treating neurodegenerative diseases. 14-3-3 binding to phosphorylated Tau is a promising PPI drug target based on its reported destabilizing effect on microtubules, leading to enhanced neurofibrillary tangle formation as a potential cause of AD-related neurodegeneration. Inhibition of 14-3-3/Tau may therefore be neuroprotective. Previously, we reported the structure-guided development of modified peptide inhibitors of 14-3-3/Tau. Here, we report further efforts to optimize the binding mode and activity of our modified Tau peptides through a combination of chemical synthesis, biochemical assays, and X-ray crystallography. Most notably, we were able to characterize two different high-affinity binding modes, both of which inhibited 14-3-3-binding to full-length PKA-phosphorylated Tau protein in vitro as measured by NMR spectroscopy. Our findings, besides producing useful tool inhibitor compounds for studying 14-3-3/Tau, have enhanced our understanding of the molecular parameters for inhibiting 14-3-3/Tau, which are important milestones toward the establishment of our 14-3-3 PPI hypothesis., Published by ACS Publ., Washington, DC

However, how such interface-binding molecules can be created in a rational, bottom-up manner is a largely unanswered question. We report here how a fragment-based approach can be used to identify chemical starting points for the development of small-molecule stabilizers that differentiate between two different PPI interfaces of the adapter protein 14-3-3. The fragments discriminately bind to the interface of 14-3-3 with the recognition motif of either the tumor suppressor protein p53 or the oncogenic transcription factor TAZ. This X-ray crystallography driven study shows that the rim of the interface of individual 14-3-3 complexes can be targeted in a differential manner with fragments that represent promising starting points for the development of specific 14-3-3 PPI stabilizers., Published by ACS, Washington, DC
