Typ I IFN gehört zur ersten Abwehrlinie des Körpers nach Infektionen. Neben der direkten antiviralen Wirkung auf infizierte Zellen, wirkt es auf Zellen des angeborenen und adaptiven Immunsystems. Die Wirkung von Typ I IFN auf CD8+ T-Zellen, die als Teil der adaptiven Immunantwort infizierte Zellen eliminieren, ist widersprüchlich. Zum einen kann Typ I IFN die Immunantwort unterstützen, zum anderen übt IFN negative Effekte auf CD8+ T Zellen aus. Wie diese vielfältigen Wirkungen zustande kommen ist bisher unklar. Sowohl die Bindung von Typ I IFN an den entsprechenden Rezeptor als auch eine Stimulation des T-Zell-Rezeptors induzieren Signalkaskaden. Es sollte daher untersucht werden, welchen Einfluss die Signalkaskaden aufeinander ausüben, ob es einen „Crosstalk“ zwischen den Signalkaskaden gibt und welche Konsequenzen dies für die Aktivierung, Differenzierung und Effektorfunktion von T-Zellen hat. Um dies näher zu untersuchen, wurden OT-I T-Zellen TZR-spezifisch stimuliert und der Einfluss einer IFN-ß-Stimulation untersucht. Es konnte gezeigt werden, dass Typ I IFN abhängig von Zeitpunkt und Dauer der Stimulation Proliferation und Effektorfunktion von Antigen-stimulierten T-Zellen inhibiert, während Aktivierung und Differenzierung der T-Zellen nicht beeinflusst werden. Typ I IFN inhibiert die Induktion von IL-2, was ursächlich für die verminderte Proliferation ist. Zusätzlich konnte gezeigt werden, dass Typ I IFN Moleküle der T-Zell-Rezeptor-Signalkaskade beeinflusst. Der Kalzium-Signalweg, der Ras/MAPK-Signalweg sowie die Aktivierung von NF-kB werden durch Typ I IFN inhibiert, während die MAP-Kinasen JNK und p38 nicht beeinflusst werden. Geninduktionsanalysen zeigen, dass Typ I IFN zu einer verminderten Induktion einer Vielzahl T-Zell-Rezeptor-induzierter Gene führt, jedoch auch IFN-induzierte Gene durch eine T-Zell-Rezeptor-Stimulation vermindert induziert werden. Insgesamt zeigen die Daten, dass ein „Crosstalk“ zwischen T-Zell-Rezeptor- und Typ I IFN-induzierten Signalwegen existiert und in Folge dessen T-Zell-Funktionen beeinflusst werden., Type I IFN provides the first line of defense after infections. Apart from its direct antiviral effects on infected cells, Type I IFN can also modulate cells of the innate and adaptive immunity. CD8+ T cells, a part of the adaptive immunity, recognize and eliminate infected cells. Previously described effects of Type I IFN on CD8+ T cells are contradictory. CD8+ T cell-mediated responses can be supported or are negatively affected. The reason for these multifaceted outcomes has not been elucidated in detail. Binding of Type I IFN to its corresponding receptor as well as the stimulation of the T cell receptor induces subsequent downstream signaling cascades. This study aimed at identifying, how these signaling cascades interact with each other and what the consequences of these interactions are for T cell activation, differentiation and effector-function. For this purpose, the effect of IFN-ß stimulation on antigen-specific stimulated OT-I T cells was investigated. Type I IFN leads to a time-dependent inhibition of T cell-proliferation and effector-function, whereas activation and differentiation of T cells remain unaffected. Type I IFN reduces the induction of Interleukin-2, leading to the observed reduced proliferation. Furthermore, Type I IFN can influence certain TCR-induced signaling cascades. The TCR-induced Calcium pathway is inhibited, the activation of MAP-Kinase Erk1/2 as well as NF-kB is reduced, while the activation of the MAP-Kinases JNK and p38 is not altered. Microarray-analysis revealed a Type I IFN-mediated decrease in the induction of a multitude of TCR-stimulated genes, whereas a TCR-stimulation led to a diminished IFN-stimulated gene induction. These results indicate the existence of a “cross-talk” between TCR- and IFN-induced signaling cascades influencing T cell function.
