Abstract

Preclinical testing of novel therapeutics for chronic hepatitis B (CHB) requires suitable animal models. Equids host homologs of hepatitis C virus (HCV). Because coinfections of hepatitis B virus (HBV) and HCV occur in humans, we screened 2,917 specimens from equids from five continents for HBV. We discovered a distinct HBV species (Equid HBV, EqHBV) in 3.2% of donkeys and zebras by PCR and antibodies against EqHBV in 5.4% of donkeys and zebras. Molecular, histopathological, and biochemical analyses revealed that infection patterns of EqHBV resembled those of HBV in humans, including hepatotropism, moderate liver damage, evolutionary stasis, and potential horizontal virus transmission. Naturally infected donkeys showed chronic infections resembling CHB with high viral loads of up to 2.6 × 10(9) mean copies per milliliter serum for >6 mo and weak antibody responses. Antibodies against Equid HCV were codetected in 26.5% of donkeys seropositive for EqHBV, corroborating susceptibility to both hepatitis viruses. Deltavirus pseudotypes carrying EqHBV surface proteins were unable to infect human cells via the HBV receptor NTCP (Na(+)/taurocholate cotransporting polypeptide), suggesting alternative viral entry mechanisms. Both HBV and EqHBV deltavirus pseudotypes infected primary horse hepatocytes in vitro, supporting a broad host range for EqHBV among equids and suggesting that horses might be suitable for EqHBV and HBV infections in vivo. Evolutionary analyses suggested that EqHBV originated in Africa several thousand years ago, commensurate with the domestication of donkeys. In sum, EqHBV naturally infects diverse equids and mimics HBV infection patterns. Equids provide a unique opportunity for preclinical testing of novel therapeutics for CHB and to investigate HBV/HCV interplay upon coinfection.

Abstract

Introdução: Drimys brasiliensis Miers, conhecida popularmente como cataia ou casca-d'anta, é uma árvore nativa da Mata Atlântica de grande importância fitoterápica. As folhas e cascas da espécie são utilizadas na medicina popular com ação estimulante, antiespasmódica, antidiarreica, antifebril, antibacteriana, no tratamento de asma, bronquite e em certas afecções do trato digestivo. Objetivo: Avaliar a influência da época de coleta no teor e composição química do óleo essencial de folhas e cascas de D. brasiliensis. Métodos: O material vegetal foi coletado em área nativa, a partir de 15 indivíduos em duas épocas distintas, verão e inverno de 2016. Folhas e cascas foram submetidas à hidrodestilação em aparelho tipo Clevenger durante 4 horas e o teor de óleo essencial foi determinado pesando o óleo extraído e levando em consideração o peso de massa seca, sendo expresso em peso de óleo por peso de material vegetal em Base Livre de Umidade (% p/p BLU). A identificação e quantificação dos constituintes químicos foram realizadas por cromatografia gasosa acoplada a espectrometria de massas. Os dados referentes ao teor do óleo essencial foram submetidos ao teste t de Student. Resultados: Não houve influência da época de coleta no teor do óleo essencial de folhas e cascas de D. brasiliensis, porém o teor do óleo de folhas (1,09 a 1,39 %) foi significativamente superior ao de cascas (0,14 a 0,15 %), sendo a composição química dos óleos muito semelhantes. Foram identificados 62 compostos químicos nos óleos essenciais de D. brasiliensis, com predominância de monoterpenos. O constituinte majoritário observado foi o ?-pineno tanto em folhas (17,73 a 18,44 %) quanto em cascas (60,12 a 60,39 %). Conclusão: O teor dos óleos essenciais de folhas é altamente superior ao teor dos óleos de cascas, sendo a composição química dos óleos muito semelhantes, sugerindo-se a utilização de folhas em substituição às cascas. Abstract

Background: Globally 6,300 people die per day as a result of occupational accidents or work- related diseases and a portion of them are those working in health care sectors . Healthcare sector employ over 59 million workers who work in hazardous and high risk work environments which expose them to get work-related injury (WRI). One way of WRI prevention is by doing educational intervention which need to be tailored to their needs to ensure its effectiveness . Materials and Methods: Articles were chosen by using a systematic search via three electronic database, namely PubMed, Scopus and ScienceDirect coupled with a combination of keywords namely "work-related injury" OR "occupational hazard" OR "healthcare workers" OR "hospital" AND "educational intervention" OR "injury prevention" OR "workplace education". Other inclusion criteria included fully accessible, original studies in English language, published in a peer-reviewed journals from 1 January 2010 to 31 March 2018. Result and Discussion: A number of 10 studies were included in this review. The results were categorised into type of intervention, subject population, health behaviour model used (if available) and the results of the selected studies. All 10 selected studies showed significant improvement post intervention in the research subject using education as part of their intervention strategy. Two of those studies mentioned and used specific behavioural or training model namely Health Belief Model (HBM) and Kirkpatrick's Model. Among all the reviewed articles, three of them used components of participatory training in addition to educational intervention. Conclusion: This review provides strong evidence that WRIs can be prevented with an educational intervention programme or module. Despite differences in methodological approaches, the studies consistently showed educational programs are effective in reducing the risk of WRIs in healthcare settings. [ABSTRACT FROM AUTHOR]
