12. Assessment of Weed Dominance Dynamics Across Varied Weeding Interval in Onion Cultivation on Ultisol Soils

Weed management is a crucial aspect of agricultural productivity, particularly in onion cultivation, where competition with weeds can significantly impact crop yields. This study investigates the dynamics of weed dominance in onion cultivation on Ultisol soil, focusing on various weeding intervals. A field experiment assessed weed species composition and density across different weed-free periods: 0-20, 20-40, and 40-60 days after planting. Results indicated that weed diversity fluctuated, revealing 29 weed species consisting of broadleaf, grass, and sedge types. Overall, broadleaf weeds were the dominant species in the onion fields, constituting up to 77.78% of the weed population by the end of the study. This dominance was influenced by the weeding intervals, with a higher presence of aggressive species observed in each treatment. Frequent weeding intervals resulted in high density of broadleaf weeds. These findings have direct implications for onion farmers, highlighting the need for optimized weeding strategies to enhance onion production and providing insights into sustainable agricultural practices on Ultisol soils. By understanding the dynamics of weed dominance, farmers can better manage their crops, leading to increased yields and improved sustainability. This research contributes to a deeper understanding of weed-plant interactions and effective weed management in onion cultivation.

Abstract This paper explores the potential implementation of Nature-Based Solutions (NBSs) in agriculture, specifically focusing on soil and water management in Southern Italy, particularly in the Apulia and Basilicata regions. Through a tailored questionnaire, it investigates farmers’ perceptions of the utility of NBSs, addressing key issues in the region and evaluating their role in addressing soil and water management challenges. Findings reveal primary challenges such as drought, floods, and water pollution, with soil erosion being a major concern. Several NBSs, including wetlands and bioswales, demonstrate consistent utility and performance, while disparities exist for agroforestry and strip cropping. The study underscores a significant gap in the economic valuation of NBSs, emphasizing the need for comprehensive assessments that incorporate livability improvements, water quality enhancement, and socio-cultural benefits. Additionally, an analysis of NBS implementation across Italian agriculture reveals limited case studies, suggesting the need for strategic expansions to meet Sustainable Development Goals. This research offers critical insights into the effectiveness and challenges of NBSs in agricultural soil and water management, advocating for enhanced stakeholder engagement and the development of multidimensional evaluation frameworks to support sustainable practices.

This study aimed to determine the effect of different starting feeds on the growth and survival rate of kelabau fish larvae (Ostechilus melanopleurus, Bleeker 1852). The research was conducted in a Complete Randomized Design (CRD), 3 treatments and 3 replicates. The tested animals used were 7-Dpc (Day post hatch) 90 kelabau fish (Ostechilus melanopleurus) larvae with an initial average length of 0.50 ± 0.06 cm and an initial weight of 0.03 ± 0.02 g. Treatment of one (P1) Artemia nauplii, (P2) Tubifex sp., and (P3) Daphnia. Aquarium research containers size 30×30×20 cm3 9 pieces with a volume of 5 L of water without water flow in the first week onwards from the second week to the 4th week are increased to 10 L with a water recirculation system with a water flow of about 0.5 L per minute. Measurement of the length and weight of the larvae is carried out once a week for 4 weeks. The results showed that the highest length and weight growth was obtained in the P2 treatment of 3.92 ± 0.13c cm and 1.25 ± 0.03c g, and the lowest growth was in the P3 treatment of 1.73 ± 0.05a cm and 0.093 ± 0.010a g. Duano language in Sabak and Tungkal is included into the extinct language category. One way to avoid language extinction is through language revitalization or language documentation. The purpose of this research is to ascertain and explain the proto language and the kinship of Duano tribe in Sabak and Tungkal through dialectology. Then, language documentation is carried out to look at all related aspects from cognate to morpheme positioning. The writers used Proto Malayic from Adelaar (1992) as IPD instrument, The data sources are Duano language from Sabak and Tungkal regencies spoken by 10 respondents. This is a qualitative study. The data were collected through interview, note-taking, recording, and documentation. Then, they were analyzed comparatively. This research found 920 zero data, 510 lexical differences data, 1210 cognate, and 570 phonological differences data. This showed that almost 50% of the data has zero difference, which means they have the same vocabulary. Then, phonological difference is 7.01% indicating that the two languages are different Speech. Meanwhile, lexical difference is 35.29% implying that the two are different subdialects, The relationship between the two regions is very close., the number of vocabularies that belong to vocabulary shift was higher than the vocabulary maintenance. This means that language extinction is very high, especially in the Tungkal area.

Abstract Two-dimensional transition metal dichalcogenides, particularly MoS2, are interesting materials for many applications in aerospace research, radiation therapy and bioscience more in general. Since in many of these applications MoS2-based nanomaterials can be placed in an aqueous environment while exposed to ionizing radiation, both experimental and theoretical studies of their behaviour under these conditions is particularly interesting. Here, we study the effects of tiny imparted doses of 511 keV photons to MoS2 nanoflakes in water solution. To the best of our knowledge, this is the first study in which ionizing radiation on 2D-MoS2 occurs in water. Interestingly, we find that, in addition to the direct interaction between high-energy photons and nanoflakes, reactive chemical species, generated by γ-photons induced radiolysis of water, come into play a relevant role. A radiation transport Monte Carlo simulation allowed determining the elements driving the morphological and spectroscopical changes of 2D-MoS2, experimentally monitored by SEM microscopy, DLS, Raman and UV–vis spectroscopy, AFM, and X-ray photoelectron techniques. Our study demonstrates that radiolysis products affect the Molybdenum oxidation state, which is massively changed from the stable + 4 and + 6 states into the rarer and more unstable + 5. These findings will be relevant for radiation-based therapies and diagnostics in patients that are assuming drugs or contrast agents containing 2D-MoS2 and for aerospace biomedical applications of 2DMs investigating their actions into living organisms on space station or satellites.

This service program aims to educate the community about the ecoprint method, an environmentally friendly printing technique that uses natural materials, such as local plant leaves, to create value-added products. The service method includes counseling, skill development, as well as lectures, practice, mentoring, monitoring, and evaluation. The data analysis used is descriptive analysis. The results of this service activity are increased community knowledge and skills in optimally utilizing natural potential and supporting sustainable economic development in Nyatnyono Village. Evaluation results showed high satisfaction with the relevance of the material, clarity of presentation, team responsiveness, and resource availability, although areas for improvement included language clarity, presentation structure, and ensuring adequate time for skill mastery. Overall, the community service activity had a significant positive impact on enhancing technical skills and creating new economic opportunities for the community.

Regression analysis is one of the statistical methods used to determine the causal relationship between one or more explanatory variables to the affected variable. The problem that often occurs in regression analysis is that there are multicollonity and outliers. To deal with such problems can be solved using ridge regression analysis and robust regression. Ridge regression can solve the problem of multicollinearas by assigning a constant k to the matrix Z′Z. But in this method the resulting bias value is still high, so to overcome this problem, the jackknife ridge regression method is used. Meanwhile, to overcome outliers in the data using robust regression methods which have several estimation methods, two of which are the Generalized-M (GM) estimator and the Least Trimmed Square (LTS) estimator. The aim of the study is to solve the problem of multicollinearity and outliers simultaneously using robust jackknife ridge regression method with GM estimators and LTS estimators. The results showed that the robust ridge jackknife regression method with LTS estimator can control multicollinearity and outliers simultaneously better based on MSE, AIC and BIC values compared to the robust ridge jackknife regression method with GM estimators. This is indicated by the value MSE = -6.60371, AIC = 75.823 and BIC = 81.642 on LTS estimators that are of lower value than GM estimators.

19. HiViPore: a highly viable in-flow compression for a one-step cell mechanoporation in microfluidics to induce a free delivery of nano- macro-cargoes

Abstract Background Among mechanoporation techniques for intracellular delivery, microfluidic approaches succeed in high delivery efficiency and throughput. However, especially the entry of large cargoes (e.g. DNA origami, mRNAs, organic/inorganic nanoparticles) is currently impaired since it requires large cell membrane pores with the need to apply multi-step processes and high forces, dramatically reducing cell viability. Results Here, HiViPore presents as a microfluidic viscoelastic contactless compression for one-step cell mechanoporation to produce large pores while preserving high cell viability. Inducing an increase of curvature at the equatorial region of cells, formation of a pore with a size of ~ 1 μm is obtained. The poration is coupled to an increase of membrane tension, measured as a raised fluorescence lifetime of 12% of a planarizable push-pull fluorescent probe (Flipper-TR) labelling the cell plasma membrane. Importantly, the local disruptions of cell membrane are transient and non-invasive, with a complete recovery of cell integrity and functions in ~ 10 min. As result, HiViPore guarantees cell viability as high as ~ 90%. In such conditions, an endocytic-free diffusion of large nanoparticles is obtained with typical size up to 500 nm and with a delivery efficiency up to 12 times higher than not-treated cells. Conclusions The proposed one-step contactless mechanoporation results in an efficient and safe approach for advancing intracellular delivery strategies. In detail, HiViPore solves the issues of low cell viability when multiple steps of poration are required to obtain large pores across the cell plasma membrane. Moreover, the compression uses a versatile, low-cost, biocompatible viscoelastic fluid, thus also optimizing the operational costs. With HiViPore, we aim to propose an easy-to-use microfluidic device to a wide range of users, involved in biomedical research, imaging techniques and nanotechnology for intracellular delivery applications in cell engineering.

Recently, we designed a chimeric proteomimetic of SOCS3, mimicking the interfacing regions of a ternary complex composed of SOCS3, JAK2 (Janus kinase 2) and gp130 (glycoprotein 130) proteins. The derived chimeric peptide, KIRCONG chim, demonstrated limited mimetic function owing to its poor water solubility.Methods: We report investigations concerning a PEGylated variant of KIRCONG mimetic, named KIRCONG chim, bearing a PEG (Polyethylene glycol) moiety as a linker of noncontiguous SOCS3 regions. Its ability to bind to the catalytic domain of JAK2 was evaluated through MST (MicroScale Thermophoresis), as well as its stability in biological serum assays. The structural features of the cyclic compounds were investigated by CD (circular dichroism), nuclear magnetic resonance (NMR), and molecular dynamic (MD) studies. Abstract AQP4 is expressed in the endfeet membranes of subpial and perivascular astrocytes and in the ependymal cells that line the ventricular system. The sporadic appearance of obstructive congenital hydrocephalus (OCHC) has been observed in the offspring of AQP4−/− mice (KO) due to stenosis of Silvio’s aqueduct. Here, we explore whether the lack of AQP4 expression leads to abnormal development of ependymal cells in the aqueduct of mice. We compared periaqueductal samples from wild-type and KO mice. The microarray-based transcriptome analysis reflected a large number of genes with differential expression (809). Gene sets (GS) associated with ependymal development, ciliary function and the immune system were specially modified qPCR confirmed reduced expression in the KO mice genes: (i) coding for transcription factors for ependymal differentiation (Rfx4 and FoxJ1), (ii) involved in the constitution of the central apparatus of the axoneme (Spag16 and Hydin), (iii) associated with ciliary assembly (Cfap43, Cfap69 and Ccdc170), and (iv) involved in intercellular junction complexes of the ependyma (Cdhr4). By contrast, genes such as Spp1, Gpnmb, Itgax, and Cd68, associated with a Cd11c-positive microglial population, were overexpressed in the KO mice. Electron microscopy and Immunofluorescence of vimentin and γ-tubulin revealed a disorganized ependyma in the KO mice, with changes in the intercellular complex union, unevenly orientated cilia, and variations in the planar cell polarity of the apical membrane. These structural alterations translate into reduced cilia beat frequency, which might alter cerebrospinal fluid movement. The presence of CD11c + microglia cells in the periaqueductal zone of mice during the first postnatal week is a novel finding. In AQP4−/− mice, these cells remain present around the aqueduct for an extended period, showing peak expression at P11. We propose that these cells play an important role in the normal development of the ependyma and that their overexpression in KO mice is crucial to reduce ependyma abnormalities that could otherwise contribute to the development of obstructive hydrocephalus.

Informasi lainnya seperti perubahan format terbitan Buletin Psikologi turut dilaporkan. Hasil analisis deskriptif menunjukkan bahwa terdapat 320 artikel yang telah dipublikasikan dalam Buletin Psikologi, 54 diantaranya (16.83%) terbit dalam Bahasa Inggris. Rerata jumlah halaman terbitan adalah 12 halaman per artikel dan rata-rata jumlah sitasi sebesar 28,453. Temuan studi ini menjadi informasi penting berupa journal metrics Buletin Psikologi. Dengan demikian, studi ini merupakan fondasi yang dapat dilanjutkan pada masa mendatang seiring bertambahnya data terbitan pada volume dan edisi selanjutnya.

This community service project aims to raise awareness and demonstrate the process of converting corn plant waste into compost. The project involved socializing, training, and practical demonstrations on composting corn crop waste. These activities have been conducted through counseling sessions, focus group discussions, and hands-on training from June to August 2022. Following the activities, an evaluation was conducted through discussions with the participants. The project's initial result was that ten stakeholders participated in the socialization of using corn crop waste for composting. Additionally, 15 Batang Piarau Jorong Sago farmer group members in Kenagarian Manggopoh have received training and successfully produced compost from corn stalk waste generated from animal feed crops.

Abstract Patients on hemodialysis (HD) have a high risk of death from COVID-19. We evaluated the humoral and cell-mediated immune response to BNT162b2 (Pfizer-BioNTech) vaccine in HD patients, comparing HD with Poly-methyl-methacrylate (PMMA) and HD with Polysulphone (PS). Samples were collected before vaccination (T0) and 14-days after the 2ndvaccine (T2) in a TG (TG, n = 16-Foggia) and in a VG (CG, n = 36-Novara). Anti-SARS-CoV-2-Ig were titrated in the cohort 2-weeks after the 2nddose of vaccine. Abstract Natural polymers have found increased use in a wider range of applications due to their less harmful effects. Notably, bacterial cellulose has gained significant consideration due to its exceptional physical and chemical properties and its substantial biocompatibility, which makes it an attractive candidate for several biomedical applications. This study attempts to thoroughly unravel the microstructure of bacterial cellulose precursors, known as bioflocculants, which to date have been poorly characterised, by employing both electron and optical microscopy techniques. Here, starting from bioflocculants from Symbiotic Culture of Bacteria and Yeast (SCOBY), we proved that their microstructural features, such as porosity percentage, cellulose assembly degree, fibres’ density and fraction, change in a spatio-temporal manner during their rising toward the liquid–air interface. Furthermore, our research identified a correlation between electron and optical microscopy parameters, enabling the assessment of bioflocculants' microstructure without necessitating offline sample preparation procedures. The ultimate goal was to determine their potential suitability as a novel cellulose-based building block material with tuneable structural properties. Our investigations substantiate the capability of SCOBY bioflocculants, characterized by distinct microstructures, to successfully assemble within a microfluidic device, thereby generating a cellulose sheet endowed with specific and purposefully designed structural features.

The study investigates the effect of Hydroxyapatite (HA) coating thickness on the inflammation and osseointegration properties of Ti–29Nb–13Ta-4.6Zr (TNTZ) implants for orthopedic implant applications. TNTZ screws were used as based samples, and the HA coating was deposited using an Electrophoretic Deposition (EPD) method to achieve various coating thickness. The thin (50–70 μm), medium (70–90 μm), and thick (90–110 μm) coating thicknesses were obtained by applying different voltage and time exposure during the EPD process, while the uncoated screw was used as the control sample. The screws were implanted into the right tibia of male Rattus norvegicus Wistar rats, and the animals were kept in cages and provide with ad libitum food for two weeks. After termination of the test animals, blood samples were collected to measure levels of Tumor Necrosis Factor-α (TNF-α) using an ELISA method, and a removal torque test was performed on the implanted screws. Additionally, tibial bone tissue around the implant area was collected for histopathology analysis. The result of the study indicate that HA coating thickness has an effect on TNF-α concentration, removal torque value, and new bone growth. The optimal thickness of the HA layer was found to be in the medium range (70–90 μm), resulting in low inflammation levels, relatively high osseointegration rates, and optimal new bone tissue growth. These finding are expected to be applicable in future orthopedic implant applications.

Dalam pembelajaran matematika, penalaran berperan sebagai dasar yang memungkinkan siswa untuk mendalamai serta memahami konsep-konsep umum dalam pembelajaran matematika serta membantu siswa mencapai menarik suatu kesimpulan dari permasalahan matematis. Dengan menerapkan model pembelajaran berbasis masalah dalam kegiatan pembelajaran yang dilaksanakan, dapat membantu siswa dalam menyelesaikan/mencari solusi dari setiap permasalahan matematis yang berkaitan dengan permasalahan yang sedang terjadi. Dalam penelitian ini, jenis penelitian yang digunakan yakni kualitatif dengan pendekatan fenomenologi. Sehingga, penelitian ini bermaksud untuk menganalisis serta mendeskripsikan kemampuan penalaran matematis siswa dengan menerapkan model pembelajaran berbasis masalah. Penelitian dilaksanakan di SMP Negeri 4 Gunungsitoli Utara. Hasil perolehan nilai siswa setelah diberikan tes kemampuan penalaran matematis dengan perolehan skor yang bervariasi yakni skor tertinggi adalah 87, skor terendah 31 serta rata-rata 63,35 dengan kriteria tergolong sedang. Berdasarkan hasil analisis, ditemukan beberapa faktor yang menyebabkan siswa tidak memenuhi kriteria kemampuan penalaran matematis, antara lain kurangnya pemahaman konsep, ketidakkonsistenan dalam mengidentifikasi masalah, kurangnya ketelitian, serta kesulitan dalam menemukan rumus untuk menyelesaikan soal.

Abstract The biological effects of ionizing radiation are exploited in the clinical practice of radiotherapy to destroy tumour cells while sparing the surrounding normal tissue. While most of the radiotherapy research focused on DNA damage and repair, recently a great attention is going to cells' interactions with the mechanical microenvironment of both malignant and healthy tissues after exposure. In fact, the stiffness of the extracellular matrix can modify cells' motility and spreading through the modulation of transmembrane proteins and surface receptors' expression, such as CD-44. CD-44 receptor has held much interest also in targeted-therapy due to its affinity with hyaluronic acid, which can be used to functionalize biodegradable nanoparticles loaded with chemotherapy drugs for targeted therapy. We evaluated changes in CD-44 expression in two mammary carcinoma cell lines (MCF10A and MDA-MB-231) after exposure to X-ray (2 or 10 Gy). To explore the role of the mechanical microenvironment, we mimicked tissues' stiffness with polyacrylamide's substrates producing two different elastic modulus values (0.5 and 15 kPa). We measured a dose dependent increase in CD-44 relative expression in tumour cells cultured in a stiffer microenvironment. These findings highlight a crucial connection between the mechanical properties of the cell's surroundings and the post-radiotherapy expression of surface receptors.

Background: Diabetic Mastopathy (DMP) is an uncommon benign fibro-inflammatory condition that occurs in women with long-standing diabetes mellitus (DM), particularly type 1. It often mimics breast cancer (BC) in clinical and imaging presentations, leading to diagnostic challenges. Methods: A retrospective monocentric study was conducted, analyzing clinical, radiologic, and pathological data from 28 women diagnosed with DMP over 10 years at the European Institute of Oncology. Data on DM type, age at DMP diagnosis, associated autoimmune conditions, imaging features, and surgical outcomes were collected and compared with the existing literature. Results: The majority (82%) of the patients had type 1 DM, with most diagnosed with DMP before age 40. Common complications included retinopathy (46%) and neuropathy (35%). Imaging often suggested malignancy, necessitating core needle biopsies for diagnosis. Surgical intervention occurred in 55% of cases, with a recurrence rate of 32%. One case of BC was observed. Conclusions: DMP remains challenging due to its resemblance to BC. Conservative management is typical, but the recurrence rate post-surgery highlights the importance of ongoing monitoring. Although DMP does not significantly increase BC risk, caution is advised, especially for immunocompromised patients. Further studies are needed to comprehensively understand DMP’s relationship with BC.

34. Effect of light intensity on the photo-oxidation stability of red and yellow palm olein mixture

Palm oil is an edible oil derived from the mesocarp of oil palm fruit (Elaeis guineensis), which has a high content of carotenoids and tocopherol components. This research aimed to study the effects of light intensity on the photo-oxidation stability of a red and yellow palm olein mixture. The red and yellow palm oleins were mixed into 100, 200, and 350 ppm carotene content. The photo-oxidation stability of the palm olein mixture was investigated under fluorescent light intensities of 5,000 and 10,000 lux at 31 ±2 °C for 7 days. Changes in the content of chlorophyll, carotene, tocopherols, and peroxide value (PV) were evaluated daily. The results showed that an increase in carotene and tocopherol contents effectively improved the photo-oxidative stability of the palm olein mixture. Degradation of chlorophyll, tocopherols, and increased PV were proportional to light intensity during photo-oxidation. There were no significant changes in carotene content at 5,000 lux light intensity exposure. The degradation rates of chlorophyll and tocopherols can be described as first-order reaction kinetics. In contrast, the increase rate of PV can be described as a zero-order kinetics model with k-values of 6.6 x 10-2, 4.9 x 10-2, 3.7 x 10-2 mequiv.kg-1.h-1, and 8.3 x 10-2, 6.8 x 10-2, and 5.6 x 10-2 mequiv.kg-1.h-1 in palm olein mixture, which contains 100, 200, and 350 ppm carotene at 5,000 and 10,000 lux light intensity exposure, respectively. These results suggested that carotene protected tocopherol in palm olein and that tocopherol and carotene synergistically acted as singlet oxygen quenchers during photo-oxidation.

The stiffness of the extracellular matrix plays a crucial role in cell motility and spreading, influencing cell morphology through cytoskeleton organization and transmembrane proteins’ expression. In this context, mechanical characterization of both cells and the extracellular matrix gains prominence for enhanced diagnostics and clinical decision-making. Here, we investigate the combined effect of mechanotransduction and ionizing radiations on altering cells’ mechanical properties, analysing mammary cell lines (MCF10A and MDA-MB-231) after X-ray radiotherapy (2 and 10 Gy). We found that ionizing radiations sensitively affect adenocarcinoma cells cultured on substrates mimicking cancerous tissue stiffness (15 kPa), inducing an increased structuration of paxillin-rich focal adhesions and cytoskeleton: this process translates in the augmentation of tension at the actin filaments level, causing cellular stiffness and consequently affecting cytoplasmatic/nuclear morphologies. Deeper exploration of the intricate interplay between mechanical factors and radiation should provide novel strategies to orient clinical outcomes.

The COVID-19 pandemic outbreak has caused disruptions in various aspects of life and the economy, including the movement of stock indexes. The objective of this study is to examine the influence of macroeconomic factors on the volatility of the Jakarta Composite Index both before and after the COVID-19 pandemic. Monthly secondary data from June 2007 to December 2019 were analyzed to identify pre-COVID trends, while data from June 2007 to December 2021 were used to identify post-COVID-19 patterns. The analysis incorporated the Granger causality test, Cointegration test, Vector Error Correction Model, and Augmented Dickey-Fuller. The results of the causality analysis revealed variations in the impact of interest rates, global oil prices, inflation, and exchange rates on the Jakarta Composite Index before and following the COVID-19 pandemic. These findings hold significance for investors as they enable the identification of potential risks associated with investments during global crises.

Mammalian sperm motility is getting more relevant due to rising infertility rates worldwide, generating the need to improve conventional analysis and diagnostic approaches. Nowadays, computer assisted sperm analysis (CASA) technologies represent a popular alternative to manual examination which is generally performed by observing sperm motility in very confined geometries. However, under physiological conditions, sperm describe three-dimensional motility patterns which are not well reconstructed by the limited depth of standard acquisition chambers. Therefore, affordable and more versatile alternatives are needed. Here, a motility analysis in unconfined conditions is proposed. In details, the analysis is characterized by a significant longer duration -with respect to conventional systems- with the aim to observe eventually altered motility patterns. Brightfield acquisition in rectangular glass capillaries captured frozen–thawed bovine spermatozoa which were analyzed by means of a self-written tracking routine and classified in sub-populations, based on their curvilinear velocity. To test the versatility of our approach, cypermethrin -a commonly used pesticides- known to be responsible for changes in sperm motility was employed, assessing its effect at three different time-steps. Experimental results showed that such drug induces an increase in sperm velocity and progressiveness as well as circular pattern formation, likely independent of wall interactions. Moreover, this resulted in a redistribution of sperm with the rapid class declining in number with time, but still showing an overall velocity increase. The flexibility of the approach permits parameter modifications with the experimental needs, allowing us to conduct a comprehensive examination of sperm motility. This adaptability facilitated data acquisition which can be computed at different frame rates, extended time periods, and within deeper observation chambers. The suggested approach for sperm analysis exhibits potential as a valuable augmentation to current diagnostic instruments.

Tissue-engineered skin substitutes are promising tools to cover large and deep skin defects. However, the lack of a synergic and fast regeneration of the vascular network, nerves, and skin appendages limits complete skin healing and impairs functional recovery. It has been highlighted that an ideal skin substitute should mimic the structure of the native tissue to enhance clinical effectiveness. Here, we produced a pre-vascularized dermis (PVD) comprised of fibroblasts embedded in their own extracellular matrix (ECM) and a capillary-like network. Upon implantation in a mouse full-thickness skin defect model, we observed a very early innervation of the graft in 2 weeks. In addition, mouse capillaries and complete epithelialization were detectable as early as 1 week after implantation and, skin appendages developed in 2 weeks. These anatomical features underlie the interaction with the skin nerves, thus providing a further cue for reinnervation guidance. Further, the graft displays mechanical properties, collagen density, and assembly features very similar to the host tissue. Taken together our data show that the pre-existing ECM components of the PVD, physiologically organized and assembled similarly to the native tissue, support a rapid regeneration of dermal tissue. Therefore, our results suggest a promising potential for PVD in skin regeneration.

As HPC systems grow in complexity, efficient and manageable operation is increasingly critical. Many centers are thus starting to explore the use of Operational Data Analytics (ODA) techniques, which extract knowledge from massive amounts of monitoring data and use it for control and visualization purposes. As ODA is a multi-faceted problem, much effort has gone into researching its separate aspects: however, accounts of production ODA experiences are still hard to come across. In this work we aim to bridge the gap between ODA research and production use by presenting our experiences with ODA in production, involving in particular the control of cooling infrastructures and visualization of job data on two HPC systems. We cover the entire development process, from design to deployment, highlighting our insights in an effort to drive the community forward. Candida auris is an emerging yeast considered a serious threat to global health. We report the first case of C. auris candidemia in Southern Italy, characterized using whole genome sequencing (WGS), and compared with a second strain isolated from a patient who presented as C. auris-colonized following screening. The C. auris strain was isolated from clinical samples, identified via MALDI-TOF, and subjected to WGS. Antifungal susceptibility testing was performed using commercial broth microdilution plates, and resistance protein sequences were evaluated with TBLASTN-2.15.0. Following the initial C. auris isolation from patient A, active surveillance and environmental investigations were implemented for all ICU patients. Of the 26 ICU surfaces sampled, 46.1% tested positive for C. auris via real-time PCR. Screening identified a second patient (patient B) as C. auris-colonized. The phylogenetic characterization of strains from patients A and B, based on the D1/D2 region of the 28s rDNA and the internal transcribed spacer (ITS) region, showed high similarity with strains from Lebanon. SNP analysis revealed high clonality, assigning both strains to clade I, indicating a significant similarity with Lebanese strains. This case confirms the alarming spread of C. auris infections and highlights the need for stringent infection control measures to manage outbreaks.

New strategies for enhancing drug delivery to the blood–brain barrier (BBB) represent a major challenge in treating cerebral diseases. Nanoemulsion-based nanocarriers represent an ideal candidate to improve drug delivery thanks to their versatility in functionalization and cargo protection. In this work, a paclitaxel-loaded nano-emulsion has been firstly functionalized and stabilized with two layers constituted of chitosan and hyaluronic acid, and, secondly, the latter has been conjugated to the CRT peptide. CRT is a bioactive peptide that selectively recognizes bEnd.3 cells, a model of the BBB, thanks to its interactions with transferrin (Tf) and its receptor (TfR). Cytotoxic results showed a 41.5% higher uptake of CRT functionalized nano-emulsion than the negative control, demonstrating the ability of this novel tool to be accumulated in brain endothelium tissue. Based upon these results, our approach can be fully generalizable to the design of multifunctional nanocarriers for delivery of therapeutic agents to the central nervous systems.

Currently, the biomimetic approach of drawing inspiration from nature has frequently been employed in designing drug nanocarriers (NCs) of actively target various diseases, ranging from cancer to neuronal and inflammation pathologies. The cell-membrane coating can confer upon the inner nanomaterials a biological identity and the functions exhibited by the cells from which the membrane is derived. Monocyte- and macrophage-membrane-coated nanomaterials have emerged as an ideal delivery system to target inflamed vasculature. Herein, we developed two biomimetic NCs using a human-derived leukaemia monocytic cell line (THP-1), either undifferentiated or differentiated by phorbol 12-myristate 13-acetate (PMA) into adherent macrophage-like cells as membrane sources for NC coating. We employed a secondary oil-in-water nano-emulsion (SNE) as the inner core, which served as an optimal NC for high payloads of lipophilic compounds. Two different biomimetic systems were produced, combining the biomimetic features of biological membranes with the physicochemical and nano-sized characteristics of SNEs. These systems were named Monocyte NEsoSome (M-NEsoSome) and Macrophage NEsoSome (M0-NEsoSome). Their uptake ability was investigated in tumour necrosis factor alfa (TNFα)-treated human umbilical vein endothelial cells (HUVECs), selected as a model of inflamed endothelial cells. The M0 membrane coating demonstrated accelerated internalisation compared with the monocyte coating and notably surpassed the uptake rate of bare NCs. In conclusion, M0-NEsoSome NCs could be a therapeutic system for targeting inflamed endothelial cells and potentially delivering anti-inflammatory drugs in vascular inflammation.

The rationale for conducting this research stems from the observation that the government's efforts in administering the Pariaman traditional market have not been fully optimised, leading to inefficient development strategies aimed at enhancing the regional revenue of Kota Pariaman. The objective of this study is to examine the local government's approach to administering traditional markets in Pariaman City, Indonesia. The present study employs qualitative research methodology, specifically utilising descriptive methodologies. The application of SWOT analysis was employed throughout the many stages of data processing, data analysis, data reduction, and drawing conclusions. The results of this study indicate that (1) infrastructure facilities such as playgrounds can be an attraction for, (2) good organizational governance, (3) Being able to create tourism-based innovations, (4) Providing freedom to rent shops for yellow card owners so that they get full benefits from the sales proceeds, (5) conduct socialization to all yellow card owners, (6) carry out repairs to leaky buildings, (7) conduct training to improve employee performance, (8) make regulations regarding shop rental fees, (9) improve supervision so that visitors are more comfortable, (10) Improve facilities and infrastructure in traditional markets in dealing with home industry (11) Involving security officers to carry out their functions in controlling morning market traders and street vendors around Pariaman tradisional market, (12) taking action against elements who set prices for renting kiosks outside the arena (13) increased supervision. it can be interpreted that the Pariaman government needs to take better steps and strategies.

This community service program aims to provide knowledge and skills to the community on how to manage and process household waste, especially plastic, into valuable and marketable products. The methods for implementing community service activities included lectures, practical exercises, and guidance. The evaluation used in the community service activities involved the utilization of questionnaires and their subsequent qualitative descriptive analysis. The outcome of the community service activities was an increase in knowledge about waste bank management and skills in utilizing plastic waste to create woven plastic bags for water bottle holders. In conclusion, community service has proven to be a highly effective endeavor, providing valuable knowledge and skills to the partners while promoting environmental awareness and sustainability through plastic crochet.

The biblical story has significance for the Christian faith and Christian ethics and serves as a strong basis for a complete worldview to face the challenge of the postmodern world. According to this viewpoint, the biblical story offers solutions to the most important problems in life, such as: Who are we? We are where? What is the issue? What is the remedy? The biblical story offers a strong basis for an all-encompassing worldview that responds to these concerns and aids in resolving any potential separation in our lives were postmodern gives questions in higher education.

Background Transthyretin cardiac amyloidosis (ATTR‐CA) is a progressive and ultimately fatal cardiomyopathy. Biomarkers reflecting multiorgan dysfunction are of increasing importance in patients with heart failure; however, their significance in ATTR‐CA remains largely unknown. The aims of this study were to characterize the multifaceted nature of ATTR‐CA using blood biomarkers and assess the association between blood biomarkers and prognosis. Methods and Results This is a retrospective cohort study of 2566 consecutive patients diagnosed with ATTR‐CA between 2007 and 2023. Anemia (39%), high urea (52%), hyperbilirubinemia (18%), increased alkaline phosphatase (16%), increased CRP (C‐reactive protein; 27%), and increased troponin (98.2%) were common findings in the overall population, whereas hyponatremia (6%) and hypoalbuminemia (2%) were less common. These abnormalities were most common in patients with p.(V142I) hereditary ATTR‐CA, and became more prevalent as the severity of cardiac disease increased. Multivariable Cox regression analysis demonstrated that anemia (hazard ratio [HR], 1.19 [95% CI, 1.04–1.37]; P=0.01), high urea (HR, 1.23 [95% CI, 1.04–1.45]; P=0.01), hyperbilirubinemia (HR, 1.32 [95% CI, 1.13–1.57; P=0.001), increased alkaline phosphatase (HR, 1.20 [95% CI, 1.01–1.42; P=0.04), hyponatremia (HR, 1.65 [95% CI, 1.28–2.11]; P56 ng/L (HR, 1.72 [95% CI, 1.46–2.03]; P

Tapai is a traditional Indonesian food usually made from cassava and glutinous rice. The study aims to determine natural dyes that produce the highest quality probiotic glutinous rice tapai. The study was conducted experimentally using a complete randomized design consisting of four treatments, and each treatment was repeated four times. The treatment in this study was the soaking of glutinous rice into natural dye extracts, namely red dragon fruit extract, purple sweet potato extract, and suji leaf extract, with different concentrations, namely 10%, 15%, 20%, and 25%. The data obtained were statistically analyzed using variance analysis and continued with Duncan's new multiple-range test (DNMRT) at a level of 5%. The results showed that natural dye extracts with different concentrations have a noticeable influence on total lactic acid, alcohol content, total lactic acid bacteria, antioxidant activity, and sensory characteristics of color, aroma, taste, and hardness. The soaking treatment in red dragon fruit extract and purple sweet potato extract with a concentration of 20% resulted in glutinous rice tapai, preferred by panelists. Furthermore, the characteristic pH value of 3.35-3.41, total lactic acid 0.61-0.70%, alcohol content 0.33-0.42%, total lactic acid bacteria 9.11-9.40 CFU/mL, and antioxidant activity 167.35-102.51 ppm.

This study analyzes the species composition, length–weight relationship (LWR), condition factor, and morphometric and meristic characteristics of eleven freshwater fish species in Kota Panjang Reservoir, Indonesia. Fresh specimens were collected from September 2023 to February 2024. Results show that among the sampled species, two belong to Order Anabantiformes, three to Order Siluriformes, and six to Order Cypriniformes. The Order Siluriformes has the lowest species composition, with Hemibagrus wyckii at 3.09%, while the Order Cypriniformes has the highest, with Barbonymus schwanefeldii at 15.25%. The LWR coefficient b ranges from 1.693 to 3.151. Of the species studied, only Hemisilurus heterorhynchus (b = 2.174), Clarias teijsmanni (b = 2.046), Anabas testudineus (b = 2.375), Labiobarbus festivus (b = 2.305), and Rasbora argyrotaenia (b = 1.693) fall outside the expected range of 2.5

50. Analysis of Fatty Acids and Amino Acids of Three Local Freshwater Bagridae Fish Species in the Kampar Kanan River, Indonesia, for Food Security

Fish have become an irreplaceable dietary source of animal protein, especially among households with low socioeconomic status in rural and urban areas of Indonesia. This study is aimed at analysing the proximate composition, minerals, fatty acids, and amino acids of three local Bagridae fish species in the Kampar Kanan river, Indonesia. The standard AOAC method was employed to examine the proximate composition of the carcass, and the analysis of amino acids and fatty acids was conducted through HPLC and GC-MS, respectively. The mineral content was determined using AAS. The nutrient composition results of Hemibagrus nemurus, Hemibagrus wyckii, and Mystus nigriceps revealed that the protein content was 24.26%, 22.57%, and 21.39% (% dry weight), whereas the total lipid content was 6.64%, 7.47%, and 7.75%, respectively. Regarding mineral contents, the calcium levels ranged from 1.49 to 1.66 mg/g, iron levels from 28.35 to 40.36 μg/g, and zinc levels from 24.03 to 54.46 μg/g. Among the fatty acids, palmitic acid was the most abundant in all three species, accounting for 25.59–30.70% of the total fatty acids. Additionally, significant amounts of C18:1 (oleic acid), C18:0 (stearic acid), and C20:4 (arachidonic acid) were also detected as primary fatty acids. The calculated atherogenic index values in the three species of Bagridae fish ranged from 0.73 to 0.99, while the thrombogenic index values varied between 0.54 and 0.75. The predominant amino acids found in the three species of Bagridae fish were glutamic acid with their concentrations ranging from 9.10 to 24.34%. These results indicate that consuming the meat of these three freshwater Bagridae fish species caught in the wild does not pose any health risks to consumers. They can be considered a safe and suitable food source with good nutritional quality.
