(1) Background: Producing active antimicrobial peptides with disulfide bonds in bacterial strains is challenging. The cytoplasm of Escherichia coli has a reducing environment, which is not favorable to the formation of disulfide bonds. Additionally, E. coli may express proteins as insoluble aggregates known as inclusion bodies and have proteolytic systems that can degrade recombinant peptides. Using E. coli strains like SHuffle and tagging the peptides with fusion proteins is a common strategy to overcome these difficulties. Still, the larger size of carrier proteins can affect the final yield of recombinant peptides. Therefore, a small fusion protein that can be purified using affinity chromatography may be an ideal strategy for producing antimicrobial peptides in E. coli. (2) Methods: In this study, we investigated the use of the small metal-binding protein SmbP as a fusion partner for expressing and purifying the antimicrobial peptide scygonadin in E. coli. Two constructs were designed: a monomer and a tandem repeat; both were tagged with SmbP at the N-terminus. The constructs were expressed in E. coli SHuffle T7 and purified using immobilized metal-affinity chromatography. Finally, their antimicrobial activity was determined against Staphylococcus aureus. (3) Results: SmbP is a remarkable fusion partner for purifying both scygonadin constructs, yielding around 20 mg for the monomer and 30 mg for the tandem repeat per 1 mL of IMAC column, reaching 95% purity. Both protein constructs demonstrated antimicrobial activity against S. aureus at MICs of 4 μM and 40 μM, respectively. (4) Conclusions: This study demonstrates the potential of SmbP for producing active peptides for therapeutic applications. The two scygonadin constructs in this work showed promising antimicrobial activity against S. aureus, suggesting they could be potential candidates for developing new antimicrobial drugs.

We have recently shown that SmbP, the small metal-binding protein of Nitrosomonas europaea, can be employed as a fusion protein to express and purify recombinant proteins and peptides in Escherichia coli. SmbP increases solubility, allows simple, one-step purification through affinity chromatography, and provides superior final yields due to its low molecular weight. In this work, we report for the first time the use of SmbP to produce a recombinant peptide with anticancer activity: the antitumor-analgesic peptide (BmK-AGAP), a neurotoxin isolated from the venom of the Chinese scorpion Buthus martensii Karsch. This peptide was expressed in Escherichia coli SHuffle for correct, cytoplasmic, disulfide bond formation and tagged with SmbP at the N-terminus to improve its solubility and allow purification using immobilized metal affinity chromatography. SmbP_BmK-AGAP was found in the soluble fraction of the cell lysate. After purification and removal of SmbP by digestion with enterokinase, 1.8 mg of pure and highly active rBmK-AGAP was obtained per liter of cell culture. rBmK-AGAP exhibited antiproliferative activity on the MCF-7 cancer cell line, with a half-maximal inhibitory concentration value of 7.24 μM. Based on these results, we considered SmbP to be a suitable carrier protein for the production of recombinant, biologically active BmK-AGAP. We propose that SmbP should be an attractive fusion protein for the expression and purification of additional recombinant proteins or peptides that display anticancer activities.

Dengue virus NS5 protein plays multiple functions in the cytoplasm of infected cells, enabling viral RNA replication and counteracting host antiviral responses. Here, we demonstrate a novel function of NS5 in the nucleus where it interferes with cellular splicing. Using global proteomic analysis of infected cells together with functional studies, we found that NS5 binds spliceosome complexes and modulates endogenous splicing as well as minigene-derived alternative splicing patterns. In particular, we show that NS5 alone, or in the context of viral infection, interacts with core components of the U5 snRNP particle, CD2BP2 and DDX23, alters the inclusion/exclusion ratio of alternative splicing events, and changes mRNA isoform abundance of known antiviral factors. Interestingly, a genome wide transcriptome analysis, using recently developed bioinformatics tools, revealed an increase of intron retention upon dengue virus infection, and viral replication was improved by silencing specific U5 components. Different mechanistic studies indicate that binding of NS5 to the spliceosome reduces the efficiency of pre-mRNA processing, independently of NS5 enzymatic activities. We propose that NS5 binding to U5 snRNP proteins hijacks the splicing machinery resulting in a less restrictive environment for viral replication.

Abstract We study the dynamically induced flexural topography in subduction numerical mechanical models. We focus on the topographic changes at the overriding plate (OP) surface induced by variations in OP kinematics, particularly when the subducting plate (SP) has a stationary motion after having reached the rigid base of the upper mantle. Our models consist of two viscoelastic plates with free surfaces and an isoviscous mantle. Friction is imposed along the planar subduction interface. We first characterize the main topographic features at a constant OP velocity, using spatial definitions based on geometrical estimations of the volcanic arc position. The models exhibit the formation of a bulge in the forearc area followed landwards by a depression and a smaller second bulge, both bracketing the arc region. The steady‐state distance to the trench of these features increases with OP velocity. Their amplitude is affected by the far‐field OP tectonic regime that depends on kinematics, and plates and subduction interface strength. We next test the effect of sudden changes in OP velocity. An OP acceleration yields a transient topographic tilt, during which the outer forearc quickly subsides whereas the arc region uplifts, and that is followed by reverse slower motions. An OP slowdown induces opposite motions. The rates of elevation change during the tilt are approximately proportional to velocity variations and mainly sensitive to the SP strength. The rates are higher than 0.1 mm/yr for velocity changes higher than 1 cm/yr. We suggest that topographic accommodations of OP velocity changes should be considered when quantifying nonisostatic topography.

(1) Background: The cathelicidin peptide LL-37 is a prominent molecule with many biological activities, including antimicrobial. Due to its importance, here, we describe the production of LL-37 tagged with SmbP, a relatively new carrier protein that improves the production of recombinant proteins and peptides in Escherichia coli. We present an alternative method for the rapid expression, purification, and antimicrobial evaluation of LL-37, that involves only one purification step. (2) Methods: A DNA construct of SmbP_LL-37 was transformed into E. coli BL21(DE3); after overnight expression, the protein was purified directly from the cell lysate using immobilized metal-affinity chromatography. SmbP_LL-37 was treated with Enterokinase to obtain the free LL-37 peptide. The antimicrobial activity of both SmbP_LL-37 and free LL-37 was determined using the colony forming unit assay method. (3) Results: SmbP_LL-37 was observed in the soluble fraction of the cell lysate; after purification with IMAC, protein gel electrophoresis, and analysis by ImageJ, it showed 90% purity. A total of 3.6 mg of SmbP_LL-37 was produced from one liter of cell culture. SmbP_LL-37 and free LL-37 both showed inhibition activity against Staphylococcus aureus and Escherichia coli. (4) Conclusions: The SmbP fusion protein is a valuable tool for producing biologically-active LL-37 peptide. The production method described here should be of interest for the expression and purification of additional cationic peptides, since it cuts the purification time considerably prior to determination of antimicrobial activity.

A new species of Leptodactylus frog (Anura: Leptodactylidae) from the South American Gran Chaco, morphologically similar and previously confused with the widespread Leptodactylus mystacinus, is described through the use of multiple sources of evidence (molecular, external morphology, coloration, osteology, bioacoustics, and behavior). The phylogenetic analysis with partial sequences of mitochondrial rDNA genes (12S and 16S) recovered the new species within the L. fuscus group, being highly divergent (>3% genetic distance in 16S). The new species was recovered as sister taxa of L. mystacinus, from which it is distinguished by tympanum coloration, cephalic index, dorsum and legs coloration, and some osteological differences in nasals and prevomers. This new frog is characterized by a moderate body size (SVL 46.80–66.21 mm), distinctive color pattern (reddish dorsal surfaces of body with noticeable black stripes in the dorsolateral folds), a circular and dark tympanum with dark tympanic annuli, and behavior of males that call on top of fallen logs and tree branches close to the ground.

Fluid and melt transport in the solid mantle can be modeled as a two-phase flow in which the liquid flow is resisted by the compaction of the viscously deforming solid mantle. Given the wide impact of liquid transport on the geodynamical and geochemical evolution of the Earth, the so-called "compaction equations" are increasingly being incorporated into geodynamical modeling studies. When implementing these equations, it is common to use a regularization technique to handle the porosity singularity in the dry mantle. Moreover, it is also common to enforce a positive porosity (liquid fraction) to avoid unphysical negative values of porosity. However, the effects of this "capped" porosity on the liquid flow and mass conservation have not been quantitatively evaluated. Here, we investigate these effects using a series of 1- and 2-dimensional numerical models implemented using the commercial finite-element package COMSOL Multiphysics®. The results of benchmarking experiments against a semi-analytical solution for 1- and 2-D solitary waves illustrate the successful implementation of the compaction equations. We show that the solutions are accurate when the element size is smaller than half of the compaction length. Furthermore, in time-evolving experiments where the solid is stationary (immobile), we show that the mass balance errors are similarly low for both the capped and uncapped (i.e., allowing negative porosity) experiments. When Couette flow, convective flow, or subduction corner flow of the solid mantle is assumed, the capped porosity leads to overestimations of the mass of liquid in the model domain and the mass flux of liquid across the model boundaries, resulting in intrinsic errors in mass conservation even if a high mesh resolution is used. Despite the errors in mass balance, however, the distributions of the positive porosity and peaks (largest positive liquid fractions) in both the uncapped and capped experiments are similar. Hence, the capping of porosity in the compaction equations can be reasonably used to assess the main pathways and first-order distribution of fluids and melts in the mantle. [ABSTRACT FROM AUTHOR]
