Abstract

Tis paper deals with the question of how areas with diferent syntactic variability can be identifed. It uses data from the Syntactic Atlas of German-speaking Switzerland (SADS) which uses multiple informants in each survey location. As a starting point the well-known doubling construction with the verb aafange ‘begin’ is used to illustrate how the diferent regions differ with respect to inter-personal variation and how the diferent variants can be mapped in terms of predominance, i.e. to what extent they co-occur or compete with the other variants. As a quantitative measure, the intensity value of the dominant variant (i.e. the agreement rate between those informants providing the dominant variant as their variant) is used as the basis to create a so-called “variation index”. Tis technique is applied to a larger set of SADS data, and the results are mapped onto the survey points indicating the syntactic variability for each location. To assess the validity of the method, several subgroups are created which turn out to correlate with the whole data set at a signifcant level. By performing a hot spot analysis, regional clusters of high/low syntactic variability can be identifed.

Abstract

The Concise Guide to PHARMACOLOGY 2015/16 provides concise overviews of the key properties of over 1750 human drug targets with their pharmacology, plus links to an open access knowledgebase of drug targets and their ligands (www.guidetopharmacology.org), which provides more detailed views of target and ligand properties. The full contents can be found at http://onlinelibrary.wiley.com/doi/10.1111/bph.13355/full. G protein-coupled receptors are one of the eight major pharmacological targets into which the Guide is divided, with the others being: G protein-coupled receptors, ligand-gated ion channels, voltage-gated ion channels, other ion channels, nuclear hormone receptors, catalytic receptors and transporters. These are presented with nomenclature guidance and summary information on the best available pharmacological tools, alongside key references and suggestions for further reading. The Concise Guide is published in landscape format in order to facilitate comparison of related targets. It is a condensed version of material contemporary to late 2015, which is presented in greater detail and constantly updated on the website www.guidetopharmacology.org, superseding data presented in the previous Guides to Receptors & Channels and the Concise Guide to PHARMACOLOGY 2013/14. It is produced in conjunction with NC-IUPHAR and provides the official IUPHAR classification and nomenclature for human drug targets, where appropriate. It consolidates information previously curated and displayed separately in IUPHAR-DB and GRAC and provides a permanent, citable, point-in-time record that will survive database updates.

Abstract

The Concise Guide to PHARMACOLOGY 2015/16 provides concise overviews of the key properties of over 1750 human drug targets with their pharmacology, plus links to an open access knowledgebase of drug targets and their ligands (www.guidetopharmacology.org), which provides more detailed views of target and ligand properties. The full contents can be found at http://onlinelibrary.wiley.com/doi/10.1111/bph.13352/full. Nuclear hormone receptors are one of the eight major pharmacological targets into which the Guide is divided, with the others being: G protein-coupled receptors, ligand-gated ion channels, voltage-gated ion channels, other ion channels, catalytic receptors, enzymes and transporters. These are presented with nomenclature guidance and summary information on the best available pharmacological tools, alongside key references and suggestions for further reading. The Concise Guide is published in landscape format in order to facilitate comparison of related targets. It is a condensed version of material contemporary to late 2015, which is presented in greater detail and constantly updated on the website www.guidetopharmacology.org, superseding data presented in the previous Guides to Receptors & Channels and the Concise Guide to PHARMACOLOGY 2013/14. It is produced in conjunction with NC-IUPHAR and provides the official IUPHAR classification and nomenclature for human drug targets, where appropriate. It consolidates information previously curated and displayed separately in IUPHAR-DB and GRAC and provides a permanent, citable, point-in-time record that will survive database updates.

Abstract

The Concise Guide to PHARMACOLOGY 2015/16 provides concise overviews of the key properties of over 1750 human drug targets with their pharmacology, plus links to an open access knowledgebase of drug targets and their ligands (www.guidetopharmacology.org), which provides more detailed views of target and ligand properties. The full contents can be found at http://onlinelibrary.wiley.com/doi/10.1111/bph.13353/full. G protein-coupled receptors are one of the eight major pharmacological targets into which the Guide is divided, with the others being: G protein-coupled receptors, ligand-gated ion channels, voltage-gated ion channels, other ion channels, nuclear hormone receptors, enzymes and transporters. These are presented with nomenclature guidance and summary information on the best available pharmacological tools, alongside key references and suggestions for further reading. The Concise Guide is published in landscape format in order to facilitate comparison of related targets. It is a condensed version of material contemporary to late 2015, which is presented in greater detail and constantly updated on the website www.guidetopharmacology.org, superseding data presented in the previous Guides to Receptors & Channels and the Concise Guide to PHARMACOLOGY 2013/14. It is produced in conjunction with NC-IUPHAR and provides the official IUPHAR classification and nomenclature for human drug targets, where appropriate. It consolidates information previously curated and displayed separately in IUPHAR-DB and GRAC and provides a permanent, citable, point-in-time record that will survive database updates.

Abstract

The Concise Guide to PHARMACOLOGY 2015/16 provides concise overviews of the key properties of over 1750 human drug targets with their pharmacology, plus links to an open access knowledgebase of drug targets and their ligands (www.guidetopharmacology.org), which provides more detailed views of target and ligand properties. The full contents can be found at http://onlinelibrary.wiley.com/doi/10.1111/bph.13350/full. Voltage-gated ion channels are one of the eight major pharmacological targets into which the Guide is divided, with the others being: G protein-coupled receptors, ligand-gated ion channels, other ion channels, nuclear hormone receptors, catalytic receptors, enzymes and transporters. These are presented with nomenclature guidance and summary information on the best available pharmacological tools, alongside key references and suggestions for further reading. The Concise Guide is published in landscape format in order to facilitate comparison of related targets. It is a condensed version of material contemporary to late 2015, which is presented in greater detail and constantly updated on the website www.guidetopharmacology.org, superseding data presented in the previous Guides to Receptors & Channels and the Concise Guide to PHARMACOLOGY 2013/14. It is produced in conjunction with NC-IUPHAR and provides the official IUPHAR classification and nomenclature for human drug targets, where appropriate. It consolidates information previously curated and displayed separately in IUPHAR-DB and GRAC and provides a permanent, citable, point-in-time record that will survive database updates.

Abstract

Recently, there have been various attempts to set up a test suite covering the syntactic phenomena of a natural language (cp. [Flickinger et al. 1989], [Nerbonne et al. 1993]). The latest effort is the TSNLP project (Test Suite for Natural Language Processing) within the Linguistic Research and Engineering (LRE) framework sponsored by the European Union (cp. [Balkan et al. 1994]). These test suites are meant for the testing of NLP software regarding their coverage of syntactic phenomena. [Volk 1995] showed that a well-organised test suite can also be used to support incremental grammar development and grammar documentation. The key issues in the organisation of a test suite are the ease of extensibility and interchangeability as well as the avoidance of redundancy. We have implemented a test suite, which is optimized for the avoidance of redundancy and we report on the trade-off for extensibility and interchangeability.

Abstract

Gabmap is a web application aimed especially to facilitate explorations in quantitative dialectology — or dialectometry — by enabling researchers in dialectology to conduct computersupported explorations and calculations even if they have relatively little computational expertise. Gabmap creates various views of dialect data, from histograms of characters used to spot coding errors, to alignments of phonetic transcriptions used in measuring pronunciation distance, to colored multidimensional scaling plots intended to illustrate quantitative results insightfully. Many analyses are accompanied by facilities allowing researchers to probe further, e.g. seeking the most important linguistic bases of an areal division, or examining the results of clustering for statistical reliability. These are also intended to inform the critical discussion of quantitative techniques, i.e. a comparision between quantitative analyses and non-quantitative (qualitative) work. For this reason Gabmap also includes support for qualitative analyses, such as facilities to map the occurrence of individual features. The software is in use, and the source code is openly available., Gabmap es una aplicación destinada especialmente a facilitar los trabajos en dialectología cuantitativa —o dialectometría— permitiendo a los dialectólogos, incluso a los que tienen pocos conocimientos de técnicas computacionales, llevar a cabo análisis y cálculos asistidos por ordenador. Gabmap ofrece visualizaciones diversas de los datos dialectales, desde histogramas de los caracteres utilizados para detectar errores de codificación, a alineaciones de las transcripciones fonéticas usadas en la medida de la distancia en la pronunciación, así como gráficos multidimensionales coloreados destinados a ilustrar cuantitativamente los resultados. Muchos análisis van acompañados de herramientas que facilitan nuevas investigaciones; por ejemplo, buscando las bases lingüísticas más importantes de una división de área, o examinando los resultados de los conglomerados a partir de su fiabilidad estadística. Estos también pretenden aportar una discusión crítica a las técnicas cuantitativas; por ejemplo; una comparación entre análisis cuantitativos y no cuantitativos (cualitativos). Por esta razón, Gabmap incluye soporte para análisis cualitativos y herramientas para cartografiar las ocurrencias de rasgos individuales. El software utilizado es de código abierto.

Abstract

Rationale: Semaphorin 3A (SEMA3A)-encoded semaphorin is a chemorepellent that disrupts neural patterning in the nervous and cardiac systems. In addition, SEMA3A has an amino acid motif that is analogous to hanatoxin, an inhibitor of voltage-gated K+ channels. SEMA3A-knockout mice exhibit an abnormal ECG pattern and are prone to ventricular arrhythmias and sudden cardiac death. Objective: Our aim was to determine whether SEMA3A is a naturally occurring protein inhibitor of K(v)4.3 (I-to) channels and its potential contribution to Brugada syndrome. Methods and Results: K(v)4.3, Na(v)1.5, Ca(v)1.2, or K(v)4.2 were coexpressed or perfused with SEMA3A in HEK293 cells, and electrophysiological properties were examined via whole-cell patch clamp technique. SEMA3A selectively altered K(v)4.3 by significantly reducing peak current density without perturbing K(v)4.3 cell surface protein expression. SEMA3A also reduced I-to current density in cardiomyocytes derived from human-induced pluripotent stem cells. Disruption of a putative toxin binding domain on K(v)4.3 was used to assess physical interactions between SEMA3A and K(v)4.3. These findings in combination with coimmunoprecipitations of SEMA3A and K(v)4.3 revealed a potential direct binding interaction between these proteins. Comprehensive mutational analysis of SEMA3A was performed on 198 unrelated SCN5A genotype-negative patients with Brugada syndrome, and 2 rare SEMA3A missense mutations were identified. The SEMA3A mutations disrupted SEMA3A's ability to inhibit K(v)4.3 channels, resulting in a significant gain of K(v)4.3 current compared with wild-type SEMA3A. Conclusions: This study is the first to demonstrate SEMA3A as a naturally occurring protein that selectively inhibits K(v)4.3 and SEMA3A as a possible Brugada syndrome susceptibility gene through a K(v)4.3 gain-of-function mechanism

Abstract

In this paper we measure the degrees of association among aggregate pronunciational, lexical and syntactic differences in 70 Dutch dialect varieties. First, we show that pronunciation is marginally more strongly associated with syntax than it is with lexis and that syntax and lexis are only weakly associated. Then, we check for the influence of geography as an underlying factor because geography is known to strongly correlate with each of the linguistic levels under investigation. We find that pronunciation and syntax are more strongly associated with geography than lexis is. Finally, we refine the results by accounting for the influence of geography as an underlying factor and show that the association between pronunciation and syntax turns out to be largely based on geography. Some influence between pronunciation and syntax remains but the association between pronunciation and lexis is stronger. There is virtually no association between syntax and lexis.
