Communal grazing is the predominant farming system in Ethiopia and the livelihood of millions of people depend on it. Gastrointestinal nematodes represent a serious threat to the sustainability of these systems, and there is further concern due to the worldwide spread of anthelmintic resistance (AR). This study aimed to assess nematode control practices and anthelmintic efficacy in three districts of central Ethiopia, where sheep, goats and cattle are reared together by resource-poor farmers. A total of 153 farmers were interviewed and 36 faecal egg count reduction tests (FECRTs) were conducted for each ruminant species grazing in the communal pastures, comprising a control group and three treatment groups (albendazole ABZ, tetramisole TET and ivermectin IVM) for each district. The questionnaire survey indicated that nematode control relies on poor practices and is substantially based on pharmacological treatments. About 91.8% of animals are treated at least twice a year often in absence of clinical signs and by untrained farmers, with no monitoring nor drug rotation implemented. In the FECRT-based survey, susceptibility to TET was confirmed in all trials, while treatment failure was a consistent finding for ABZ. IVM showed inefficacy in cattle and, to a lesser extent, in goats, while it was always effective in sheep. Research on AR in African countries is focused on small ruminants but in this study cattle had the worst results in terms of the number and severity of ineffective treatments, suggesting their inclusion in future studies on AR in this type of settings., Competing Interests: Declarations. Soybean cyst nematode (SCN) is a major pest of soybean crops, causing significant yield losses and economic impact. Current management strategies primarily rely on resistant varieties, cover crops, and seed treatments. In addition, there is a growing interest in developing sustainable, ecologically based approaches to integrate SCN risk reduction into soybean production systems. This study aimed to evaluate the efficacy of various compost and manure amendments in suppressing SCN populations and promoting soybean productivity. An in vitro egg hatching assay was conducted to screen the inhibitory effects of different compost and manure extracts on SCN egg hatching. Results indicated that poultry manure, Layer Ash Blend, and swine manure extracts significantly inhibited SCN hatching compared with other treatments across multiple time points. Greenhouse trials further validated the effectiveness of Layer Manure, poultry manure, High Carbon Dairy Doo, and Seed Starter 101 in suppressing SCN cysts, eggs, and juveniles. A field microplot trial confirmed the potential of Layer Ash Blend and poultry manure in SCN management, with significant reductions in SCN populations and increased soybean yields. The study also investigated the impact of these amendments on promoting the population of bacterivorous and frugivorous nematodes, contributing to a biological diverse soil ecosystem. Overall, the results indicate that amending SCN-infested soil with specific compost or manure formulations can effectively suppress nematode populations while improving soybean productivity. There is a pressing need to develop alternative management strategies for the soybean cyst nematode ( Heterodera glycines ), the most costly pathogen to soybeans. Plant elicitor peptides (PEPs), which are produced by plants in response to stress and stimulate broad-spectrum disease resistance, were previously shown to reduce soybean cyst nematode infection on soybeans when applied as a seed treatment. Here, we introduce an alternative method to deliver PEPs to soybean using a common plant growth-promoting rhizobacterium, Bacillus subtilis , as a bacterial expression system. Similar to the empty vector control, B. subtilis engineered to express a PEP from soybean (GmPEP3) was able to colonize soybean roots and persisted on roots more than a month after treatment. Compared with water or the empty vector control, plants that received a seed treatment with B. subtilis expressing GmPEP3 (B.+GmPEP3) were significantly taller early in vegetative growth (V1 stage) and had lower chlorophyll content in the reproductive stage (R3/R4); these results suggest that GmPEP3 may hasten growth and subsequent senescence. When plants were inoculated with soybean cyst nematode at the V1 stage, those pretreated with B.+GmPEP3 supported significantly fewer nematode eggs at the reproductive stage (R3/R4) than plants treated with water or the empty vector. The effects of B.+GmPEP3 on nematode infection and plant growth appeared to be due primarily to the peptide itself because no significant differences were observed between plants treated with water or with B. subtilis expressing the empty vector. The use of medicinal plants as a means of combating parasites is becoming increasingly popular due to general resistance to synthetic anthelmintics. Goats typically respond less well to anthelminthic treatments, which may increase their resistance to nematodes. For this purpose, the anthelmintic effect of Pelargonium quercetorum Agnew (P. quercetorum) extract was tested in an in vivo study against gastrointestinal parasites of goats. A total of 40 goats naturally infected with mixed gastrointestinal nematode species were divided into four groups: the first group was treated with albendazole (7.5 mg/kg), the second group received a single dose of closantel (7.5 mg/kg), the third group received plant extract (7.5 mg/kg), and the fourth group served as an untreated control. Fecal egg counts (FEC) were carried out on day 14, as well as day 0, which was prior to the first treatment dose. According to the results, maximum reduction in FEC percentage was observed in P. quercetorum treated group (63.41%). Furthermore, nematode parasites responded poorly to synthetic drugs, although there was a 4.72% and 45.54% decrease in FEC in the albendazole and closantel-treated groups, respectively. Although no significant difference was found between the treatment groups, the P. quercetorum treated group showed a significant reduction in fecal egg count after treatment. Based on findings, a P. quercetorum based anthelmintic could be a sustainable alternative to combat parasite infestation. Background: Human parasitic infections caused by Adenophorean nematodes encompass a range of diseases, including dioctophymiasis, trichuriasis, capillariasis, trichinellosis, and myositis. These infection can result in adverse impacts on human health and cause societal and economic concerns in tropical and subtropical regions., Methods: This review conducted searches in PubMed, Embase and Google Scholar for relevant studies that published in established databases up to April 26, 2024. Studies that focused on the common morphology, life cycle, disease distribution, clinical manifestations, and prevention and control strategies for Adenophorean parasitic diseases in humans were included., Results: Adenophorean nematodes exhibit shared morphological characteristics with a four-layered cuticle; uninucleate epidermal cells; pseudocoelom with six or more coelomocytes; generally three caudal glands; five esophageal glands; two testes in males with median-ventral supplementary glands in a single row; tail in males rarely possessing caudal alae; amphids always postlabial; presence of cephalic sensory organs; absence of phasmids; and a secretory-excretory system consisting of a single ventral gland cell, usually with a non-cuticularized terminal duct. Humans play two important roles in the life cycle of the nematode class, Adenophorea: 1) as a definitive host infected by ingesting undercooked paratenic hosts, embryonated eggs, infective larvae in fish tissue and meat contaminated with encysted or non-encysted larvae, and 2) as an accidental host infected by ingesting parasitic eggs in undercooked meat. Many organs are targeted by the Adenophorean nematode in humans such as the intestines, lungs, liver, kidneys, lymphatic circulation and blood vessels, resulting in gastrointestinal problems, excessive immunological responses, cell disruption, and even death. Most of these infections have significant incidence rates in the developing countries of Africa, Asia and Latin America; however, some parasitic diseases have restricted dissemination in outbreaks. To prevent these diseases, interventions together with education, sanitation, hygiene and animal control measures have been introduced in order to reduce and control parasite populations., Conclusions: The common morphology, life cycle, global epidemiology and pathology of human Adenophorean nematode-borne parasitic diseases were highlighted, as well as their prevention and control. Extensive farming systems form an integral part of sheep production systems across Europe. However, with innate production handicaps, declining sheep numbers and narrow economic margins, production is becoming increasingly challenging threatening the future sustainability of the industry. Gastrointestinal nematodes (GINs) are a significant cause of production losses to the global sheep industry, with well-established resistance to the major anthelmintic groups. Traditionally, extensive farming systems are not thought to have a significant parasite challenge compared with intensive farms, but there is a need to identify the scale and importance of GINs on extensive farms to inform the need for sustainable control strategies. In this study, a questionnaire of extensive farmers (n=34) was conducted and parasitological data were collected from nine study farms to investigate the perceived versus actual GIN and anthelmintic resistance challenge faced by extensive farms. The results showed a production-limiting challenge on most farms, with a higher GIN challenge observed on improved pastures. Furthermore, over half of the extensive farmers perceived anthelmintic resistance to be a greater problem for intensive farmers, with only 20% of respondents reporting known anthelmintic resistance. However, all study farms had evidence of resistance to at least one group of anthelmintics. Gastrointestinal nematode (GIN) infections impact small ruminant health, welfare, and production across farming systems. Rising anthelmintic resistance and regulation of synthetic drug use in organic farming is driving research and development of sustainable alternatives for GIN control. One alternative is the feeding of plants that contain secondary metabolites (PSMs) e.g., proanthocyanidins (PA, syn. condensed tannins) that have shown anthelmintic potential. However, PSMs can potentially impair performance, arising from reduced palatability and thus intake, digestibility or even toxicity effects. In this study, we tested the trade-off between the antiparasitic and anti-nutritional effects of heather consumption by lambs. The impact of additional feeding of a nematophagous fungus (Duddingtonia flagrans) on larval development was also explored. Lambs infected with Teladorsagia circumcincta or uninfected controls, were offered ad libitum heather, or a control chopped hay for 22 days during the infection patent period. Eight days into the patent period, parasitised lambs were supplemented (or remained unsupplemented) with D. flagrans for a 5-day period. Performance and infection metrics were recorded, and polyphenol levels in the heather and control hay were measured to investigate their association with activity. The lambs consumed heather at approximately 20% of their dry matter intake, which was sufficient to exhibit significant anthelmintic effects via a reduction in total egg output (P = 0.007), compared to hay-fed lambs; the magnitude of the reduction over time in heather fed lambs was almost 10-fold compared to control lambs. Negative effects on production were shown, as heather-fed lambs weighed 6% less than hay-fed lambs (P < 0.001), even though dry matter intake (DMI) of heather increased over time. D. flagrans supplementation lowered larval recovery in the faeces of infected lambs by 31.8% (P = 0.003), although no interactions between feeding heather and D. flagrans were observed (P = 0.337). There was no significant correlation between PA, or other polyphenol subgroups in the diet and egg output, which suggests that any association between heather feeding and anthelmintic effect is not simply and directly attributable to the measured polyphenols. The level of heather intake in this study showed no antagonistic effects on D. flagrans, demonstrating the methods can be used in combination, but provide no additive effect on overall anthelmintic efficacies. Gastrointestinal parasitic nematode (GIN) infections are the cause of severe losses to farmers in countries where small ruminants such as sheep and goat are the mainstay of livestock holdings. There is a need to develop effective and easy-to-administer anti-parasite vaccines in areas where anthelmintic resistance is rapidly rising due to the inefficient use of drugs currently available. In this review, we describe the most prevalent and economically significant group of GIN infections that infect small ruminants and the immune responses that occur in the host during infection with an emphasis on mucosal immunity. Furthermore, we outline the different prevention strategies that exist with a focus on whole and purified native parasite antigens as vaccine candidates and their possible oral-nasal administration as a part of an integrated parasite control toolbox in areas where drug resistance is on the rise.

Duddingtonia flagrans is a nematode trapping fungus used for the control of gastrointestinal nematodes in livestock. The quantity of chlamydospores of D. flagrans required for the reduction of third-stage larvae (L3) of sheep gastrointestinal nematodes (GIN) is largely unknown, and a matter of discussion. The aim of this experiment was to determine in vitro the nematophagous activity of four different concentrations of D. flagrans (1000, 3000, 6250, or 11000 chlamydospores/ml) in the presence of varying numbers of GIN third-stage larvae (L3) (500, 1000, 1500). Additionally, the study sought to evaluate the efficacy of this fungus on Haemonchus contortus, Teladorsagia circumcincta, Trichostrongylus colubriformis and Chabertia ovina. The results showed that as fungal concentrations increased, so did the larval reduction of third-stage infective larvae in each test. L3s number was not a determining factor in the efficacy against GIN. The comparison between various concentrations of chlamydospores revealed significant differences, particularly between 1000 and 11000 chlamydospores (P≤0.05). Regarding the larval reduction of the GIN species considered, D. flagrans demonstrated the same effectiveness across all species tested. The results of the current study confirm the efficacy and underscore the importance of D. flagrans as an alternative for controlling of GIN.

Disease cross-transmission between wild and domestic ungulates can negatively impact livelihoods and wildlife conservation. In Pin valley, migratory sheep and goats share pastures seasonally with the resident Asiatic ibex ( Capra sibirica ), leading to potential disease cross-transmission. Focussing on gastro-intestinal nematodes (GINs) as determinants of health in ungulates, we hypothesized that infection on pastures would increase over summer from contamination by migrating livestock. Consequently, interventions in livestock that are well-timed should reduce infection pressure for ibex. Using a parasite life-cycle model, that predicts infective larval availability, we investigated GIN transmission dynamics and evaluated potential interventions. Migratory livestock were predicted to contribute most infective larvae onto shared pastures due to higher density and parasite levels, driving infections in both livestock and ibex. The model predicted a c. 30-day antiparasitic intervention towards the end of the livestock's time in Pin would be most effective at reducing GINs in both hosts. Albeit with the caveats of not being able to provide evidence of interspecific parasite transmission due to the inability to identify parasite species, this case demonstrates the usefulness of our predictive model for investigating parasite transmission in landscapes where domestic and wild ungulates share pastures. Helminth infections are ubiquitous in grazing ruminants and cause significant costs due to production losses. Moreover, anthelmintic resistance (AR) in parasites is now widespread throughout Europe and poses a major threat to the sustainability of modern ruminant livestock farming. Epidemiological data on the prevalence and distribution of gastrointestinal nematodes, cestodes and liver- and rumen-flukes in Italian small ruminants are outdated and fragmentary. However, anthelmintics are commonly used to control these infections and often without prior diagnosis. Compared to other European countries, few reports of AR in small ruminants against levamisole, ivermectin and benzimidazoles have been published in Italy, but recent studies suggest that this phenomenon is spreading. Increased and integrated research efforts in developing innovative approaches to control helminth infections and AR are needed and must be tailored to the peculiarities of each context in order to be effectively implemented. This manuscript provides an overview on helminth prevalence and distribution, sustainable treatment strategies and integrated control approaches in small ruminants in Italy. The implementation of targeted treatments and targeted selective treatments is discussed based on different parameters, such as fecal egg count, degree of anaemia (FAMACHA © method), milk production and body condition score. In addition, several Italian studies have also investigated the implementation of alternative strategies such as the use of different natural bioactive compounds or genetic selection for resistance and resilience to helminth infections. These concrete solutions for helminth management in small ruminant farms in the country are reported and discussed, representing a valid example for other Mediterranean countries.

Targeted Selective Treatment (TST) is a gastrointestinal nematode (GIN) control strategy where anthelmintic treatment decisions are made at an individual animal level. TST has been proven to reduce anthelmintic use and subsequently slow down anthelmintic resistance development, however questions remain regarding optimal TST methods and their applicability across farms. In this study, the influence of Mineral and Vitamin (MV) supplementation on optimal energy utilisation (EU) TST thresholds was assessed on three Welsh farms. In total, 360 lambs were split into two groups, MV supplemented and control, and were treated with an anthelmintic against GIN at the midway point of the experiment. Lambs that improved their EU efficiency post treatment were deemed to have benefited from anthelmintic treatment. Optimal EU TST thresholds was determined for each treatment group per farm using Youden's J statistic where the treatment threshold retrospectively exhibiting the greatest combined sensitivity and specificity in correctly identifying lambs benefiting from treatment was deemed to be optimal. Results demonstrated that the optimal EU TST threshold was higher in MV supplemented groups at 0.72, 0.71 and 0.56 versus 0.58, 0.67, 0.51 for control groups on each respective farm. Identification of lambs for TST was more effective when using an optimised EU TST threshold, compared to when using the standard EU TST threshold of 0.66. The study highlights that applying standard EU TST thresholds may not be appropriate on all commercial farms with factors including MV status as noted in this study likely to influence optimal EU TST thresholds. Refugia-based strategies associated with a combination of anthelmintic drugs belonging to different drug classes are becoming more common management practices to mitigate anthelmintic resistance (AR) in gastrointestinal nematodes (GIN) in small ruminants. Though refugia-based strategies have been largely demonstrated in small ruminants, cattle veterinarians and producers are considering such management strategies in grazing cattle production systems. Implementing refugia-based strategies lowers the amount of anthelmintics used in the herd and therefore slows the progression of AR by allowing a proportion of worms to escape drug selection pressure. The objective of this study was to observe the effect of a refugia-based strategy on body weight (BW), average daily gain (ADG) and fecal egg counts (FEC) of trichostongyle-type nematodes in naturally infected beef calves over a 131-day grazing season when compared with a whole herd treatment strategy, using the same combination of drugs. Stocker calves (n = 160) were ranked by body weight within sex then allocated to 16 paddocks, which were randomly assigned to one of two treatment groups. All calves in Group 1 (n = 80) were administered treatment, while in Group 2 (n = 80) the steer with the highest FEC in eggs per gram (EPG) within the paddock was left untreated. Treated calves received an extended release injectable 5 % eprinomectin (LongRange®, Boehringer Ingelheim Animal Health USA Inc.; 1 mL/50 kg of BW) and a 22.5 % oxfendazole oral suspension (Synanthic®, Boehringer Ingelheim Animal Health USA Inc.; 1 mL/50 kg of BW). Fecal egg counts and BW were recorded on days (D) -35, 0, 21, 131, and 148 to calculate the average fecal egg count reduction (FECR) and ADG for both groups. Linear mixed models, with paddock as the experimental unit, were used for analyses. The EPG differed on D21 (p < 0.01) and D131 (p = 0.057) with Group 2 having a higher average FEC (15.2 EPG D21; 57 EPG D131) compared with Group 1 (0.4 EPG D21; 37.25 EPG D131). However, there was no significant difference in average BW or ADG between treatment groups throughout the study. To counter the global spread of anthelmintic resistance (AR), considerable efforts have been invested in the development and dissemination of sustainable alternatives to control gastrointestinal nematode (GIN) infections in small ruminants. The degree to which these have been accepted and integrated by farmers, particularly in organic systems, where the drive to reduce chemical inputs is arguably even more pressing, has been little studied. To identify whether more comprehensive actions are needed to support the uptake of alternative GIN controls amongst organic farmers, this study conducted a survey in five European countries on organic dairy goat and meat sheep farmers to gain insight into current GIN control strategies and farmer attitudes towards AR and alternative measures in these countries. The structured survey was disseminated in the five European countries Switzerland, France, Netherlands, Lithuania and United Kingdom, receiving a total of 425 responses, 106 from organic dairy goat farmers and 319 from organic meat sheep farmers. Regression analyses were carried out to identify factors impacting anthelmintic drenching on meat sheep production systems, whereas all data were analysed descriptively. Four key findings emerged: i) The frequency of anthelmintic treatments averaged less than two per animal a year in all production systems; ii) Overall, organic farmers seemed well informed on the availability of alternative GIN control methods, but fewer stated to put them into practice; iii) Targeted selective treatment (TST) strategies of anthelmintics appears to be not commonly incorporated by organic farmers; iv) Despite operating under national and EU organic regulations, each of the organic dairy goat (Switzerland, France and Netherlands) and meat sheep (Switzerland, Lithuania and United Kingdom) production systems developed distinct approaches for GIN control. Since 2003, the Sustainable Control Of Parasites in Sheep (SCOPS) group have provided the UK sheep farming industry with guidance on ways to mitigate the development and dissemination of anthelmintic resistance (AR). However our empirical understanding of sheep farmers' influences towards such 'best practice' parasite control approaches is limited, and therefore requires further assessment and evaluation to identify the potential factors influencing their implementation. In 2015, a telephone questionnaire was conducted in order to elicit Scottish sheep farmers' attitudes and behaviours regarding the SCOPS recommended practices, as well as gauging farmers' general attitudes to gastrointestinal nematodes (GIN; term roundworm used in questionnaire) control. A quantitative structural equation modelling (SEM) approach was employed to determine the influences of socio-psychological factors and the uptake of individual anthelmintic resistance mitigating practices including: the implementation of a quarantine strategy for parasite control and the use of parasite diagnostic testing for monitoring faecal egg counts (FEC) and detecting AR. The proposed models established a good fit with the observed data and explained 61%, 54% and 27% of the variance in the adoption of AR testing, FEC monitoring, and quarantine behaviours respectively. The results presented highlight a number of consistent and distinct factors significantly influencing the implementation of selected SCOPS recommended practices. The negative influences of topography and farmer experience was frequently demonstrated in relation to multiple GIN control practices, as well as the positive influences of social norms, worm control knowledge, AR risk perception and positive attitudes to the services provided by the veterinary profession. Factors that were shown to have the greatest relative effects on individual parasite control practices included: the perceived expectation of others (i.e. Social norms) for implementing a quarantine strategy, farmer's suspicions to the presence of AR on the holding for instigating AR testing and the confirmation of AR for adopting FEC monitoring. Determining the influences of behaviour-specific factors on farmers' decision making processes will help to identify and address positive and negative influences concerning implementation of AR mitigating practices, as well as contribute to the development of more evidence based intervention strategies in the future., (Copyright © 2022 Elsevier B.V. Intestinal nematode infections caused by soil-transmitted helminths (STH), such as the roundworm Ascaris lumbricoides, the whipworm Trichuris trichiura, and the hookworms Ancylostoma duodenale, and Necator americanus, infect more than 1 billion people throughout the world. School-aged children tend to harbor the greatest numbers of intestinal worms, and as a result, experience more adverse health consequences, such as poor growth, anemia, and cognitive decline. Clinicians should maintain a high degree of suspicion in endemic areas when patients present with surgical abdomens, particularly children. The nematophagous fungus Duddingtonia flagrans is used in integrated management of gastrointestinal nematodes in ruminants. The chlamydospores of the fungus, orally administered, pass through the segments of the ruminant digestive tract and, in the feces, capture the nematodes preventing their migration to grasslands. The drastic conditions of the gastrointestinal segments can negatively affect the fungus' biocontrol activity. The aim of this study was to assess the effect of in vitro conditions of the sheep's main gastrointestinal segments on the concentration, viability and nematode predatory ability of D. flagrans chlamydospores. The segments evaluated separately in vitro were the oral cavity, rumen, abomasum, and small intestine. The results showed that chlamydospores concentration was not affected by exposure to the different segments. The viability of the chlamydospores after exposure to the oral cavity (2.53 × 10 6  CFU/mL) and small intestine (1.24 × 10 5  CFU/mL) was significantly lower than its control treatment, with values of 6.67 × 10 6  CFU/mL and 2.31 × 10 5  CFU/mL respectively. Nematode predatory ability after rumen exposure was reduced by 7% compared to the control treatment, by 25% after abomasum exposure and by 17% after small intestine. This study revealed the individual in vitro effect of each segment of ovine gastrointestinal tract on the integrity of this strain of the fungus D. flagrans affecting its viability and nematode predatory ability under the evaluated conditions. This review article provides an historical perspective on some of the major research advances of relevance to ruminant livestock gastrointestinal nematode control over the last 50 years. Over this period, gastrointestinal nematode control has been dominated by the use of broad-spectrum anthelmintic drugs. Whilst this has provided unprecedented levels of successful control for many years, this approach has been gradually breaking down for more than two decades and is increasingly unsustainable which is due, at least in part, to the emergence of anthelmintic drug resistance and a number of other factors discussed in this article. We first cover the remarkable success story of the discovery and development of broad-spectrum anthelmintic drugs, the changing face of anthelmintic drug discovery research and the emergence of anthelmintic resistance. This is followed by a review of some of the major advances in the increasingly important area of non-pharmaceutical gastrointestinal nematode control including immunology and vaccine development, epidemiological modelling and some of the alternative control strategies such as breeding for host resistance, refugia-based methods and biological control. The last 50 years have witnessed remarkable innovation and success in research aiming to improve ruminant livestock gastrointestinal nematode control, particularly given the relatively small size of the research community and limited funding. In spite of this, the growing global demand for livestock products, together with the need to maximise production efficiencies, reduce environmental impacts and safeguard animal welfare - as well as specific challenges such as anthelmintic drug resistance and climate change- mean that gastrointestinal nematode researchers will need to be as innovative in the next 50 years as in the last., (Copyright © 2021 Australian Society for Parasitology. Background: Farm management practices have a major impact on nematode population dynamics. The presented study aimed to understand current nematode management practices on UK sheep farms; with a particular focus on Nematodirus battus because of the changing epidemiology, and emerging anthelmintic resistance observed in this species., Methods: A 42 question online survey covering grazing management, farm demographics and parasite control strategies was developed and distributed to the farming community in 2016. Analysis of the 187 completed questionnaires explored regional variations in practices., Results: Uptake of recommendations was variable, particularly quarantine practices and monitoring tools. Results also highlighted variation in the epidemiology of N. battus; respondents in the north (Scotland, north-west and north-east England) typically reported N. battus in spring with a perception of more severe clinical symptoms than those from the south (Midlands, Wales, south-east and south-west England; p = 0.03). Farms in the south observed greater changes in the timing of disease (p = 0.006) with N. battus being reported throughout the year on some holdings and more frequent use of faecal egg count monitoring (p = 0.006)., Conclusions: Control of N. battus infection is challenging and 'one-size-fits-all' advice is not applicable; however, the information gathered will enable the development of effective, adaptable control strategies., (© 2021 British Veterinary Association.)

Recent reports indicate that gastrointestinal nematodes (GIN) are contributing to significant losses in deer productivity and that anthelmintic resistance has become an issue of concern for deer farmers in New Zealand. The aim of this study was to evaluate cross-grazing of deer with sheep or cattle as an aid for control of gastrointestinal and pulmonary nematode parasites of farmed red deer (Cervus elaphus) in New Zealand. This was a field study replicated over two years (2012 and 2013) for 16 weeks each year at two locations (Massey University, Palmerston North and Invermay AgResearch Centre, Mosgiel). Each farm replicate included four groups (19-20 deer) at each location: red deer cross-grazing with cattle (Deer/Cattle); red deer cross-grazing with sheep (Deer/Sheep); red deer grazing on their own (DeerOwn); and red deer grazing on their own and treated with anthelmintics every two weeks to suppress worm burdens, as a positive control (DeerSup). The key outcome was the number of anthelmintic treatments (AT) given to deer. The decision to treat individual resident deer in Deer/Cattle, Deer/Sheep and DeerOwn groups was based on "trigger" criteria including faecal egg count (FEC)≥250 eggs/g or Dictyocaulus faecal larval count (FLC)≥100 larvae/g or when growth rate was less than 80 % of the mean of the DeerSup group in the previous two weeks. In addition, to quantify the species of parasites cycling in each group, sets of three "tracer" deer were introduced to graze with each group at the mid-point and again at the end of each 16 week period in both years at both locations. Least squares means (LSM) of the number of AT given per animal for Deer/Sheep (3.4) and DeerOwn (3.3) groups were significantly higher than for the Deer/Cattle (2.7) group (p < 0.001). In tracer animals, the LSM of abomasal Trichostrongylus spp. were significantly fewer in the DeerOwn (17), Deer/Cattle (37) and DeerSup (54) groups than in the Deer/Sheep (952; p < 0.001) group. The LSM of the nematodes in the subfamily Ostertagiinae (=Ostertagia-type) were significantly more in the DeerOwn (1950) than in Deer/Sheep (370; p = 0.003) and DeerSup (238; p < 0.001) groups, but the number in the Deer/Cattle group (689) was not different to DeerOwn (p> 0.05). The LSM of lungworm were fewer in Deer/Sheep (3), Deer/Cattle (4) and DeerSup (3; p < 0.001) groups than in DeerOwn (40) group. The Deer/Cattle and DeerSup groups had significantly higher LSM of liveweight gain over the 16 weeks (p < 0.001) than the other two groups. This study demonstrated that cross-grazing with either sheep or cattle aided control of lungworm and gastrointestinal nematodes in young deer during autumn. The use of nematophagous fungi is an alternative for the biological control of nematodes in ruminants. In this study, the compatibility of joint growth of the fungi Monacrosporium sinense and Pochonia chlamydosporia and the joint nematicidal activity of these fungal isolates on bovine infective larvae were evaluated. For that, tests of direct confrontation, the effect of volatile compounds and antibiosis were conducted. In order to carry out the tests, the fungi were inoculated in potato dextrose agar culture medium and, after the incubation period, the growth of the colonies, the formation of an inhibition halo and the effect of volatile metabolites were verified. The compatibility between fungi isolates M. sinense and P. chlamydosporia was confirmed and the nematicidal evaluation proved the best effectiveness was when both were used together, with a 98.90% reduction in the number of bovine nematode infective larvae under in vitro conditions. It was concluded that M. sinense and P. chlamydosporia presented synergistic action, suggesting that the joint application of the fungi increases the effectiveness of biological control of bovine infective larvae.

Biological control is a strategy to decrease parasitic populations, and the action takes place through natural antagonists in the environment. We studied the predatory activity of the fungus Duddingtonia flagrans in infective larvae (L3) of gastrointestinal nematodes after gastrointestinal transit. Ten heifers were divided into two groups: treated (animals received pellets containing fungus) and control (animals received pellets without fungus). Twelve hours after administration, faeces samples were collected for in vitro efficacy tests. The animals then remained for 7 h in the experimental pasture area. At the end of this period, 20 faecal pads (ten treated and ten control) were selected at random. Pasture, faecal pad and soil collections occurred with an interval of 7 days, totalling four assessments. In vitro activity demonstrated that fungi effectively preyed on L3, achieving a reduction percentage of 88%. In the faecal pad of the pasture area, there was a difference (P < 0.05) between collections 3 and 4 for both groups; in the treated group a reduction of 65% was obtained, while in the control group there was an increase of 217% in the number of L3. The recovery of L3 in the soil and in the pasture was similar in both groups. There was no influence (P = 0.87) of the passage time on the fungus predatory activity. Duddingtonia flagrans demonstrated the ability to survive gastrointestinal transit in the animals, reducing the number of L3 in the faeces, indicating that this biological control has great potential in the control of worm infections.

Gastrointestinal nematodes (GIN) negatively impact productivity and welfare in sheep globally and are estimated to cost the European sheep industry €157-477 million annually. GIN are mainly controlled by anthelmintic treatment, however, as anthelmintic resistance becomes prominent, the routine treatment of ewes against GIN has been questioned. A questionnaire survey of 383 sheep farmers in Great Britain was conducted to identify strategies currently used to control GIN infections in ewes. Ordinal and binary regression analysis were used to identify factors associated with use of practices known to influence anthelmintic resistance development, including number and timing of ewe GIN anthelmintic treatment, targeted selective treatment (TST) of ewes, drench and move of ewes and long-acting moxidectin treatment of periparturient ewes. Participating farmers treated their ewes against GIN 1.68 times per year on average, with 42.3% and 32.1% of participating farmers worming their ewes once or twice a year on average, respectively. 17.2% of participating farmers wormed their ewes more than twice a year, and 8.4% never worm their ewes. Participating farmers who devised GIN control strategies based on SCOPS guidelines treated their ewes significantly less per year (P < 0.001), whilst those determining treatment timing based on ewe DAG scores or the time of year treated their ewes significantly more frequently (P < 0.001). Farmers who devised GIN control strategies in conjunction with their vet had greater odds of using TST (P < 0.001), as well as farmers who determined flock treatment timing based on ewe condition (P = 0.027). The use of narrow spectrum flukicides was significantly associated with reduced number of annual ewe GIN anthelmintic treatments (P < 0.001), TST of ewes against GIN (P < 0.001) and the avoidance of moving ewes to clean pastures following GIN treatment (P < 0.001). The presence of sheep scab on a farm or in the area was significantly associated with increased annual GIN treatments for ewes (P = 0.002), not using TST strategies to control GIN in ewes (P < 0.001) and moving ewes to clean pasture after anthelmintic treatment, whilst using macrocyclic lactones treatments to prevent sheep scab was significantly associated with the treatment of periparturient ewes with long-acting moxidectin (P = 0.001). This research suggests that by encouraging the application of evidence based targeted or targeted selective treatment strategies, further interaction between farmers and veterinarians/SCOPS guidance, and the uptake of best practices for controlling liver fluke and sheep scab on farms, sustainable GIN control strategies can become the common practice in ewes., (Copyright © 2021 Elsevier B.V. The pine wilt disease (PWD), for which no effective treatment is available at the moment, is a constant threat to Pinus spp. plantations worldwide, being responsible for significant economic and environmental losses every year. It has been demonstrated that elicitation with chitosan increases plant tolerance to the pinewood nematode (PWN) Bursaphelenchus xylophilus, the causal agent of the PWD, but the biochemical and genetic aspects underlying this response have not been explored. To understand the influence of chitosan in Pinus pinaster tolerance against PWN, a low-molecular-weight (327 kDa) chitosan was applied to mock- and PWN-inoculated plants. Nematode population, malondialdehyde (MDA), catalase, carotenoids, anthocyanins, phenolic compounds, lignin and gene expression related to oxidative stress (thioredoxin 1, TRX) and plant defence (defensin, DEF, and a-farnesene synthase, AFS), were analysed at 1, 7, 14, 21 and 28 days post-inoculation (dpi). At 28 dpi, PWN-infected plants elicited with chitosan showed a sixfold lower nematode population when compared to non-elicited plants. Higher levels of MDA, catalase, carotenoids, anthocyanins, phenolic compounds, and lignin were detected in chitosan-elicited plants following infection. The expression levels of DEF gene were higher in elicited plants, while TRX and AFS expression was lower, possibly due to the disease containment-effect of chitosan. Combined, we conclude that chitosan induces pine defences against PWD via modulation of metabolic and transcriptomic mechanisms related with plant antioxidant system.

Gastrointestinal nematodes (GIN) are a cause of significant losses in animal production worldwide. In recent years, there have been important advances in the biological control of GIN of ruminants and horses. While these measures are still relatively under-utilised in practice, interest will undoubtedly grow due to the emergence of drug resistant parasite populations, the rise in demand for organically farmed products (which does not allow prophylactic use of drugs, including anthelmintics) and legislation, which regulates and restricts the use of anthelmintics. This review provides an overview of the most promising biocontrol agents of GIN of grazing animals including nematophagous fungi, dung beetles, earthworms, predacious nematodes and nematophagous mites. Recent advancements in these fields are evaluated, and the potential reasons for the delayed development and slow uptake of biocontrol agents are discussed. It is now widely believed that no method of GIN control is sustainable alone, and a combination of strategies (i.e. integrated pest management) is required for long term, effective parasite control. The aim of this study was to evaluate the effect of feed additives (pumpkin seed cake and cloves) on the egg excretion of gastrointestinal nematodes (GIN) in sheep. Thirty ewes naturally infected with GIN were randomly selected from a flock and assigned to the following groups of 10 animals each: clove group (received 1.8 g ground cloves/ewe/day, for 7 days), pumpkin seed cake group (200 g pumpkin seed cake/ewe/day, for 7 days) and control group. Before starting the study, on day 0, and 5 days after the 7-day supplementation, on day 12, the body condition and FAMACHA scores were assessed and individual faecal egg counts (FEC) were performed. The mean body condition and the FAMACHA scores did not change significantly between day 0 and 12 with the exception of a significantly deteriorated FAMACHA score in the clove group. The percentage reduction of FEC was 40.7% on day 12 in the clove group and 52.9% in the pumpkin seed cake group. In the control group, FEC increased by 8.7%. A coproculture of faecal samples from four of the most infected animals on day 0 revealed Trichostrongylus spp. larvae L3 in all four selected ewes, Ostertagia spp. and Cooperia spp. in three and Haemonchus contortus in one ewe. These results are promising and encourage further studies aimed to evaluate the possibility that these plant supplements could be a complementary method for parasite control, thus reducing the need for chemotherapy., (© J. Gastrointestinal parasitism is one of the factors that discourages farmers from raising small ruminants in cultivated pastures. To validate a soil treatment strategy to control the free-living stages of gastrointestinal nematodes (GIN), castor cake (CC) was used as a fertilizer on a pasture where sheep grazed on guinea grass under continuous stocking. On day zero, the pasture was divided into three paddocks, contaminated by GIN and treated, respectively, with CC divided into two applications (2CC1/2), CC in a single application (CC1) and organic compost in a single application (control). On day 21, eight GIN-free sheep were placed in each paddock. On day 58, significant differences (P<0.05) were observed: reduction of up to 66.10% in larvae.g-1 of dry mass in pastures fertilized with CC, decrease of up to 60.72% in infection rates among the animals in the groups treated with CC, higher average daily weight gain (over 185 g.day-1) and packed cell volume (over 26%) in the groups treated with CC, when compared to the control (128 g.day-1; 20.9%). In view of the results, the use of CC, mainly CC1, as a fertilizer for guinea grass pastures, under continuous stocking, proved to be promising, with 63.41% effectiveness in controlling worm infestations.

The objective was to examine the effects of supplementary quebracho on control of coccidiosis and gastrointestinal nematodes in lambs and kids. In Exp. 1, naturally infected lambs weaned (87.8 ± 0.4 days of age; day 0) in January (winter) were blocked by sex and randomly assigned (n = 10/treatment) to receive supplement with or without 100 g/lamb of quebracho for 28 days. In Exp. 2, single or twin rearing ewes were randomly assigned into two groups, and naturally infected lambs were fed control (n = 28) or quebracho (100 g/lamb of quebracho tannins in feed; n = 27) between -28 and 21 days (weaning = day 0; 70.8 ± 0.1 days of age). In Exp. 3, weaned doe kids (57.6 ± 2.0 days of age) were randomly assigned to receive alfalfa (Medicago sativa) supplement with (n = 9) or without (n = 8) 50 g/kid quebracho or sericea lespedeza (Lespedeza cuneata) with quebracho (n = 8) for 21 days. Fecal oocyst count (FOC), nematode egg counts (FEC), fecal score, dag score (soiling around rear quarters), and blood packed cell volume (PCV) were determined every 7 days. Data were analyzed as repeated measures using mixed models. In Exp. 1, FOC decreased in quebracho-fed lambs (diet × time, P < 0.001) but FEC was similar between treatments during the feeding period (P = 0.19). Packed cell volume (P = 0.19) and fecal score (P = 0.42) were similar between groups. Quebracho-fed lambs had a greater dag score initially (diet × time, P = 0.02), but were similar by day 42 (P = 0.72). In Exp. 2, FOC remained low (P = 0.02), PCV tended to decrease (P = 0.06), but FEC increased on days 14 and 21 (diet × time; P < 0.001) in quebracho compared with control-fed lambs. Quebracho-fed lambs had lower fecal score (diet × time; P = 0.005) but higher dag score (diet × time; P < 0.001). In Exp. 3, FOC of kids fed quebracho (alfalfa or sericea lespedeza supplement) was lower than control (P < 0.001). Fecal score of kids fed sericea lespedeza compared with alfalfa were lower regardless of quebracho (P = 0.01). There were no differences among treatments for dag, FEC, PCV, or body weight (P> 0.10). Quebracho was effective in reducing FOC but not clinical signs of coccidiosis in both lambs and kids, and may not be highly digestible in lambs as it caused loose stools., (Copyright © 2020 Elsevier B.V. Nematodiosis are responsible for financial losses in cattle production systems due to treatment costs, falling production rates and animal deaths. The incorporation of sodium alginate pellets containing nematophagous fungi in the bovine diet is a method for the control of nematodiosis. The aims of this study were to evaluate the viability of Arthrobotrys cladodes and Pochonia chlamydosporia contained in sodium alginate pellets after passage through the bovine gastrointestinal tract, as well as to evaluate the effects of single and combined use of these fungi against bovine parasitic nematodes. The reduction in parasitic nematode infective larvae of bovines by the combined use of Arthrobotrys cladodes and Pochonia chlamydosporia was more than the reduction in infective larvae by Arthrobotrys cladodes or Pochonia chlamydosporia alone in the collections 12, 24 and 36 h after giving the pellets to the animals. Arthrobotrys cladodes and Pochonia chlamydosporia combined promoted maximum reduction of infective larvae of 86.3%. Therefore, the combination of Arthrobotrys cladodes and Pochonia chlamydosporia may be an effective method to control gastrointestinal nematodiosis affecting grazing cattle., (Copyright © 2020 Elsevier Inc. The objective of this study was to evaluate the reduction in nematode faecal egg count (FEC) in Pelibuey lambs segregated as resistant (RES), susceptible (SUS) and intermediate (INT) to gastrointestinal nematodes. Twenty-nine weaned Pelibuey lambs, aged five months old, free of nematode infection, were used. Nine lambs were RES, six were SUS and 14 were INT lambs. The study consisted of two phases: in Phase 1 the lambs were infected experimentally with Haemonchus contortus. In Phase 2, the lambs were naturally infected by grazing. Faecal and blood samples were taken every week. The packed cell volume and total protein were quantified. The FEC value (FECmax) per lamb was recorded together with a natural reduction in FEC in the two phases. The data were analysed with a model of measures repeated over time. During Phase 1, the RES lambs showed the lowest FEC (1061 ± 1053) compared to the other groups (INT: 2385 ± 1794 eggs per gram of faeces (EPG); and SUS: 3958 ± 3037 EPG). However, in Phase 2 no significant differences (p > 0.05) were observed between the groups of lambs (RES: 275 ± 498 EPG; SUS: 504 ± 1036 EPG; and INT: 603 ± 1061 EPG). At the end of Phase 1, the FEC of RES lambs was naturally reduced by 75.5% in respect to FECmax (p < 0.05), and at the end of Phase 2 the reduction in FEC was 90% in respect to FECmax (p > 0.05); the same behaviour was observed in RES and SUS lambs. It is concluded that the artificial infection in the lambs induced a more rapid immune response in RES than SUS lambs, and all lambs developed high acquired resistance by continuous infection.

Soil-transmitted nematodiasis was once widely prevalent in Jiangsu Province, which seriously threatened human health and hindered socioeconomic development. The control efforts over decades resulted in a remarkable decline in the prevalence of soil-transmitted nematode human infections in Jiangsu Province, with a reduction from 59.32% in 1989 to 0.12% in 2019, and the human prevalence remains at < 0.5% since 2013. Since 1987, an integrated strategy has been adopted for the control of soil-transmitted nematodiasis in Jiangsu Province; however, the core interventions varies at different stages, which mainly include deworming, water and sanitation service improvement, health education, and monitoring and assessment. The criteria of effective soil-transmitted nematodiasis control had been achieved in all epidemic counties (districts) of Jiangsu Province by 2019. Further actions to strengthen health education and monitoring and implement precision control measures are required to consolidate the achievements of soil-transmitted nematodiasis control and eliminate the harm of soil-transmitted nematodiasis to humans. This review summarizes the epidemiology, control progress and evolution of control strategy of soil-transmitted nematodiasis in Jiangsu Province.

The control of gastrointestinal nematodes among ruminants maintained in zoological parks remains difficult due to infective stages develop in the soil. For the purpose to improve the possibilities of the control of gastrointestinal nematodes (genera Trichostrongylus, Nematodirus, Chabertia and Haemonchus) affecting wild captive bovidae ruminants belonging to the subfamilies Antilopinae, Caprinae, Bovinae and Reduncinae, commercial pelleted feed enriched with a blend of 104-105 spores of both filamentous fungi Mucor circinelloides + Duddingtonia flagrans per kg meal was provided for a period of 3.5 years. All animals were dewormed at the beginning of the trial and also when exceeding a cut-off point of 300 eggs per gram of feces (EPG). The anthelmintic efficacy ranged between 96 and 100%. The need for repeating the administration of parasiticide treatment disappeared at the 24th month of study in the Antilopinae individuals, and at the 8th month in the Caprinae, Bovinae and Reduncinae. No side-effects were observed on the skin or in the digestive, respiratory or reproductive system. It was concluded that this strategy provides a sustainable tool for preventing the contamination of paddocks where captive ruminants are maintained, decreasing the risk of infection by gastrointestinal nematodes and consequently the need of frequent deworming.

Cattle production in extensive systems favours the occurrence of gastrointestinal nematodes, and the use of nematophagous fungi complements the control strategies for these nematodes. The aim of this study was to evaluate the effectiveness of the fungi Arthrobotrys cladodes and Pochonia chlamydosporia in the biological control of gastrointestinal parasitic nematodes in grazing cattle. Twenty-four calves were randomly divided into four groups and allocated to independent paddocks from February 2018 to January 2019. In the first group, the animals received pellets containing P. chlamydosporia. In the second group, the animals received pellets containing A. cladodes. In the third group, the animals received pellets containing a combination of the fungi A. cladodes and P. chlamydosporia. In the control group, the animals received pellets without fungus. The combined use of A. cladodes and P. chlamydosporia showed greater efficacy in the biological control of bovine gastrointestinal parasitic nematodes than the same fungi used separately. The parasite load was lower and weight gain was greater (P ⩽ 0.05) in the groups of cattle treated with nematophagous fungi. Therefore, the use of A. cladodes and P. chlamydosporia is promising in the biological control of nematodiosis in cattle.

In contrast to many other European countries, no data were available on the presence of anthelmintic resistance in gastrointestinal nematodes in sheep in Belgium. A faecal egg count reduction test was performed in 26 sheep flocks in Flanders, Northern Belgium. Results indicated widespread resistance against benzimidazoles (albendazole, fenbendazole and mebendazole), with treatment failure on all 8 farms investigated. Haemonchus contortus and Teladorsagia circumcincta were the predominant species after treatment failure. Amino acid substitutions associated with benzimidazole resistance were detected at the codon positions 167 (8%) and 200 (92%) of the isotype-1 beta tubulin gene in H. contortus, codon positions 198 (47%) and 200 (43%) in T. circumcincta and position 200 (100%) in T. colubriformis. Resistance against macrocyclic lactones (ivermectin, doramectin and moxidectin) was recorded on 7 out of 20 flocks, mainly in H. contortus and T. circumcincta. Treatment failure was also observed for closantel (in combination with mebendazole) and for monepantel, on one farm each. Trichostrongylus spp. were implicated with resistance against monepantel. A questionnaire survey on farm management and worm control measures indicated that worm control was often not sustainable. Ewes and lambs were treated frequently (on average 2.6 and 3.2 times per year), mostly without weighing. Only few sheep farmers (9%) regularly used faecal egg counts to monitor worm infections. Strongylid gastrointestinal nematodes are an important cause of disease and economic loss in small ruminants. These parasites are important in most of the United States, with the bloodsucking parasite Haemonchus contortus being the predominant species of concern. Sheep and goats are infected while grazing, and the biology of infective larvae on pastures is important in the design of parasite management programs. Parasite species infecting cattle throughout northern North American are generally the same as those found throughout North America. Throughout Canada, cattle are primarily infected with Ostertagia ostertagi and Cooperia oncophora, whose larvae survive cold winters within soil of pastures. Overwintering larvae of these species maintain a temporary population of refugia available in spring to grazing cattle. Cattle from northern United States are also infected with Cooperia punctata and Haemonchus placei, whose larvae cannot survive cold winters within pastures. Control of gastrointestinal nematodes (GIN) can have both economic and health benefits for cattle operations in the southern United States. In the past several decades, GIN control has relied almost exclusively on the use of anthelmintics. With the increase in anthelmintic resistance new strategies must be developed. Anthelmintic use should be minimized by integrating grazing management and a good herd health program into GIN control programs. Duddingtonia flagrans is a nematode-trapping fungus that has shown promising results as a tool to combat parasitic nematode infections in livestock. The fungus interrupts the parasitic lifecycle by trapping and killing larval stages on pasture to prevent re-infection of animals. One barrier to the fungus' commercial use is scaling up production of the fungus, and specifically of chlamydospores, which survive the digestive tract to grow in fecal pats on pasture, thus have potential as a feed through anthelmintic. The purpose of this study was to evaluate the effect of dehydration on sporulation of the fungus. Disks of Duddingtonia flagrans type strain (ATCC® 13423™) were grown on 17% cornmeal agar for 26 days at 30 °C, then split into three groups; dried quickly at 38 °C and 37% humidity over 48 h ("incubated"), dried more slowly at 24 °C and 55% humidity over 10 days ("air-dried"), or kept at 30 °C and sealed with parafilm to prevent loss of moisture as a control ("wet"). Half of each dried culture was resuspended in water, then heated to liquify and homogenized through vortexing. Spores were then counted in a Neubauer hematocytometer. Both the "air-dried" and "incubated" drying techniques yielded significantly more spores than the "wet" control (Welch's two sample t test p values of .0359 and .0411, respectively). The difference in average chlamydospores per milliliter was insignificant between the two drying techniques, although a visual representation of the data shows less spore count variability in the "air-dried" technique.

Control of sheep gastrointestinal nematode (GIN) infections mostly relies on the use of anthelmintics. Refugia-based control strategies as targeted selective treatments (TST) can delay anthelmintic resistance development, but the optimal decision criteria for selecting individuals to be treated in subclinical infection scenarios remain unclear. The present study aimed to evaluate the suitability of body condition score (BCS) and faecal egg counts (FEC) as treatment indicators and to determine their optimized threshold values for treatment in TST by determining the relationships of BCS, FEC and anthelmintic treatment with several productive parameters in pre-mating and pre-partum periods at an individual level. Deworming in pre-mating period increased BCS gain, but its magnitude was directly associated with strongyle FEC before treatment. Deworming also increased fertility in ewes with BCS < 2.75 and the proportion of lambing ewes that got pregnant during the first ovulation cycle. Nevertheless, treatment did not yield productive benefits in ewes with higher BCS values. On the other hand, deworming in the pre-partum lowered lamb peri-partum mortality and increased lamb weight at birth and the growth of lambs during lactation, especially in lambs born from ewes with strongyle FEC > 400 epg before treatment. These results showed that FEC and especially BCS can be potential decision criteria for the implementation of TST in these types of scenarios. The TST scheme derived from the present results on the control of GIN infections should include anthelmintic treatment of ewes with BCS lower than ≈ 3 approximately five weeks before mating and lambing. In the pre-lambing period, treatment may be unnecessary if flock mean strongyle FEC is lower than ≈ 200 epg, as the proportion of ewes with individual FEC > 400 epg would be very low. The results suggest that implementation of this TST scheme would provide benefits, such as the improvement of productivity, a rational management of parasites in refugia, and preservation of future efficacy of anthelmintics, in comparison to traditional deworming schemes., (Copyright © 2019 Elsevier B.V. The present study compared the feeding behaviour of goats in the early morning (EM = 7:00-8:30 a.m.) and late morning (LM = 9:30-11:00 a.m.) in response to their natural gastrointestinal nematode (GIN) infection status. Twelve female adult goats (37 ± 7.7 kg live weight (LW); 5 ± 1 years) with browsing experience in the tropical deciduous forest were divided into two groups (n = 6): INF group, with natural GIN infection, and non-infected (NI) group, dewormed with moxidectin (0.4 mg/kg LW subcutaneous). Feeding behaviour (dry matter intake (DMI)) of two resource types (grasses vs. shrubs + herbs) was estimated by direct observation for 4 weeks on two grazing moments (EM vs. LM). Environmental temperature and relative humidity at pasture level were measured twice weekly. The GIN egg counts and goats' LW were measured on days 0, 14 and 28. Temperature (mean ± standard deviation) at EM (26.2 ± 1.5 °C) was lower than at LM (38.7 ± 1 °C; P < 0.01). Humidity was higher on the EM (85.1 ± 2.6%) compared to LM (60.4 ± 5.6%; P < 0.01). Irrespective of the infection status, goats consumed similar amounts of grass and shrubs + herbs during EM (P > 0.05). On the other hand, the experimental groups consumed more grass than shrubs + herbs during LM (P < 0.05). The latter suggested prophylactic behaviours strategies to (a) avoid GIN infective larvae, (b) balance the protein:energy ratio of the diet and (c) avoid saturation of detoxification pathways for the secondary compounds consumed from shrubs + herbs. The constant consumption of shrubs + herbs during the study may have reduced the GIN egg count of the INF group.

Nematophagous fungi are used in the biological control of the parasitic helminths of animals and plants. The association of ovicidal and predator nematophagous fungi may present a complementary and increased action on the biological control of helminths. Joint growth compatibility and predation tests were carried out on infective larvae of nematode parasites of bovines with the nematophagous fungus ovicide Pochonia chlamydosporia and the nematophagous fungus predator Arthrobotrys cladodes. The tests of antagonism in direct confrontation, antibiosis and the effect of volatile metabolites between the isolates of P. chlamydosporia and A. cladodes indicated the viability of joint growth of these fungi. The association of the fungi P. chlamydosporia and A. cladodes presented a higher predatory capacity of infective larvae of the parasitic nematodes of bovines when compared to the predation of the fungi used alone. Therefore, under laboratory conditions, the fungi studied presented growth compatibility and the association of these increased the nematicidal activity against parasitic helminths of cattle.

The efficacy and safety of a new topical formulation containing selamectin plus sarolaner (Revolution® Plus / Stronghold® Plus, Zoetis) was evaluated for the prevention of heartworm (Dirofilaria immitis) disease and the treatment of roundworm infection in cats enrolled as veterinary patients in two field studies conducted in Japan. In the heartworm field study, 91 cats negative for D. immitis antigen and anti-D. immitis antibody were enrolled and received 9 monthly topical treatments with selamectin plus sarolaner during the period of April to December 2015. Efficacy was assessed by testing post-treatment blood samples collected 8, 12, and 15 months after initiation of treatment for the presence of D. immitis antigen and anti-D. immitis antibody. Eighty-seven cats completed the entire study and were included in the determination of efficacy. No D. immitis antigen or anti-D. immitis antibody were detected in any of the post-treatment samples. In the roundworm field study, completed in the period from April to November 2015, 64 cats with ≥100 roundworm eggs per gram (EPG) of feces were enrolled and allocated randomly in a 1:1 ratio, to receive either selamectin plus sarolaner or emodepside plus praziquantel (Profender®, Bayer). Treatments were administered topically on Days 0 and 30, and efficacy was assessed by fecal EPG counts conducted on Days 14, 30, and 60. All cats completed the entire study. At enrollment, all cats were infected with Toxocara cati. Compared to pre-treatment, geometric mean T. cati EPG counts on Days 14, 30, and 60 were reduced by >99.9% in both treatment groups. There were no treatment-related adverse events in either study. Monthly topical administration of Revolution® Plus / Stronghold® Plus providing a minimum of 6 mg/kg selamectin and 1 mg/kg sarolaner was safe and effective in the prevention of heartworm disease and the treatment of roundworm infection in cats enrolled as veterinary patients in Japan., (Copyright © 2019 Zoetis Services LLC. The current status of soil-transmitted nematodiasis in China is analyzed. The achievements in prevention and control of soil-transmitted nematodiasis are illustrated. Moreover, the challenges in condition of the decline of soil-transmitted nematodiasis in China are demonstrated. Finally, the working principals for prevention and control of soil-transmitted nematodiasis in the new period are proposed, combining with the health poverty alleviation project, focusing on the goals of control plan for important parasitic diseases, establishing monitoring system for soil-transmitted nematodiasis, and implementing the principles of precise prevention and control.

The present study was conducted to further characterize the anthelmintic activity of the 0.5% w/v topical formulation of eprinomectin (EPRINEX ® Pour-on, Merial) when administered at 1 mg/kg body weight to sheep in preventing the establishment of induced gastrointestinal and pulmonary nematode infections. Thirty-six female Merino sheep (∼4 months of age, weighing 27.0-36.0 kg) were blocked by pre-treatment body weight to form blocks of four animals. Within blocks, the animals were randomly allocated to either remain untreated (control) or be treated once with EPRINEX ® either on Day 0, Day 7 or Day 14. Starting on Day 15, the sheep were given trickle infections with infective larvae of seven species of gastrointestinal nematodes and Dictyocaulus filaria lungworms daily for seven consecutive days. Five weeks after completion of the daily challenge (Day 56), the animals were necropsied for parasite recovery and count. Treatment with EPRINEX ® prevented the establishment (>90%, p ≤ 0.027) of D. filaria, Teladorsagia circumcincta (pinnata/trifurcata), Cooperia curticei, Nematodirus battus, Trichostrongylus colubriformis and Oesophagostomum venulosum for at least 21 days, and of Haemonchus contortus and Trichostrongylus axei for at least 14 days. Sheep in the groups treated with EPRINEX ® at Days 7 and 14 had significantly (p ≤ 0.018) higher Day -1 to Day 56 wt gains than the untreated controls. Background: Fluralaner provides efficacy against feline ectoparasites following topical administration. Moxidectin is routinely used to treat gastrointestinal nematode infections and prevent heartworm disease caused by Dirofilaria immitis. Praziquantel is routinely used to treat feline tapeworm infections. The safety of a fluralaner plus moxidectin combination topical solution (Bravecto™ Plus, MSD Animal Health) was assessed when administered concurrently with a commercially available praziquantel topical solution (Droncit™ Spot-on, Bayer Animal Health GmbH). The highest dose rates in clinical use were tested., Results: Concurrent topical administration of a fluralaner plus moxidectin and a praziquantel product did not result in adverse findings. One out of ten cats receiving praziquantel only (control group), and two out of ten cats receiving fluralaner plus moxidectin and praziquantel (treatment group) had dandruff-like flakes in their coat at the application site. Two out of the ten control cats and three cats out of the ten treatment group cats had very small amounts of unidentified material (minute crusts or crumbs) at the application site which was only visible during close inspection., Conclusions: The concurrent treatment of cats with fluralaner plus moxidectin and praziquantel at the maximum dose in clinical use was well tolerated.

Inoculation with antagonistic soil microorganisms has shown potential to suppress replant disease of apple in orchard soils. Pseudomonas spp. may have the potential to reduce Pratylenchus penetrans populations on apple. Pseudomonas spp. were isolated from the rhizosphere of sweet cherry and screened for antagonistic characteristics. Two highly antagonistic Pseudomonas isolates, P10-32 and P10-42, were evaluated for growth promotion of apple seedlings, suppression of P. penetrans populations, and root colonization in soil from three orchards. During the isolate screening, Pseudomonas fluorescens P10-32 reduced in vitro growth of fungal pathogens, had protease activity, had capacity to produce pyrrolnitrin, suppressed P. penetrans populations, and increased plant biomass. Pseudomonas fluorescens P10-42 reduced in vitro growth of fungal pathogens, had protease activity, suppressed P. penetrans populations, and increased plant biomass. In potted orchard soil, inoculating apple with P. fluorescens P10-32 suppressed P. penetrans populations in one of the three soils examined. Inoculation with P. fluorescens P10-42 improved plant growth in two of the soils and suppressed P. penetrans abundance in one soil. In one of the soils, P. fluorescens P10-42 was detected on the roots 56 days postinoculation. Overall, we conclude that Pseudomonas spp. play a role in suppressing P. penetrans on apple in orchard soil.

Biological control through the use of nematophagous fungi is a sustainable alternative for combatting helminthes in domestic animals and allows a reduction in the use of anthelmintics. The objective of this research was to evaluate the efficacy of the Arthrobotrys cladodes var macroides fungus in a pelleted formulation, based on sodium alginate and administered twice a week orally, as an alternative for the biological control of nematodes in field-grown young cattle. The experiment was conducted in a farm located in the municipality of Viçosa, MG, where 12 cattle, seven to nine months old, were allocated in two groups (treated group and control group) and distributed in pickets of Brachiaria decumbens, naturally infested with nematode larvae. The animals in the treated group received 1g of sodium alginate matrix pellets for every 10 kg of animal live weight, containing the nematophagous fungus Arthrobotrys cladodes var macroides and administered twice a week in conjunction with commercial feed. In the control group, each animal received 1 g of pellets for every 10 kg of animal live weight, without fungal mycelium added to the feed. Samples of feces and pastures were collected fortnightly for 12 months. The results showed that the most prevalent nematode genera in the coprocultures were Haemonchus sp., Cooperia sp. and Oesophagostomum sp., reflecting the results found in forage. The pasture that contained the animals that received feed with the fungus presented a reduction of 59% and 52% of larvae recovered at distances of 20 cm and 40 cm from the fecal pats, respectively. The mean number of eggs per gram of feces each month and animal body weight did not differ (p > 0.05) between the treated and control groups. Stool and soil samples from both groups were colonized by A. cladodes fungus and other fungi. Administration of Arthrobotrys cladodes var macroides mycelium by means of a sodium alginate matrix twice weekly reduced larval infestation of the surrounding pasture, indicating that this fungus may be a promising biological control of infecting forms of nematodes present in the environment., (Copyright © 2018 Elsevier Inc. This study was conducted to evaluate effectiveness of an integrated management intervention in the control of endo- and ectoparasites of pigs kept by smallholder farmers in Mbeya Rural and Mbozi districts of southern highlands of Tanzania. A repeated cross-sectional group randomization design was employed, with 10 villages in the intervention group and six villages in the control group. The intervention consisted of improving pig confinement, feeds and feeding practices, and strategic anthelmintic treatment of pigs with oxfendazole, to primarily control Taenia solium (porcine) cysticercosis and nematodes; and with ivermectin to control ectoparasites and nematodes. Blood and faecal samples, ectoparasite specimens and skin scrapings were collected at baseline and two follow-up rounds, at an interval of seven months. A total of 482, 460 and 421 pigs were sampled in 221, 196 and 139 households at baseline, first and second follow-up visits respectively. Sero-prevalence of PC, prevalence and burden of gastrointestinal (GI) nematodes and prevalence of ectoparasites were determined using Ag-ELISA, McMaster faecal egg counting technique and body searches/skin scrapings, respectively. Mann Whitney test was used to measure intervention effect by comparing changes from baseline values of prevalence and faecal egg counts between the two groups. No significant difference (p > 0.05) was observed between the two groups, in changes from baseline of sero-prevalence of PC. The intervention significantly reduced prevalence of Trichuris suis (p = 0.044) and mean faecal egg counts of Ascaris suum (p = 0.02) from baseline to first follow-up. The intervention showed a substantial effect on the prevalence of ectoparasites as it significantly reduced overall prevalence of ectoparasites (p = 0.026), lice (p = 0.045), ticks (p = 0.049) and mites (p = 0.013) from baseline to first follow-up, and prevalence of ectoparasites, lice and mites from baseline to second follow-up (p = 0.052, p = 0.03, p = 0.017 respectively). This study has demonstrated a moderate effect of the intervention on controlling GI nematodes and ectoparasites, but no effect on sero-prevalence of PC. The reported ineffectiveness of the intervention against PC is probably an underestimation because serology is not able to provide quantitative data. It may also be a result of reduced compliance as this was a field study. Background: Foodborne nematodiasis are caused by the ingestion of food contaminated by helminths. In Europe, these diseases are present in all countries., Objectives: To review the available data on epidemiology and management of foodborne nematodiasis in the European Union, detect any trends and determine the possible causes of the observed changes., Methods: A review of available literature published between 2000 and 2016 was conducted., Results: Out of 1523 cases described in the literature, 1493 cases were autochthonous and 30 cases were imported. The detected parasites were Toxocara spp (34.7%), Ascaris lumbricoides (27.1%), Trichinella spp (21.9%), Anisakis spp (15.5%) and Angiostrongylus cantonensis (0.8%)., Conclusions: Foodborne nematodiasis remains a public health challenge for the European Union. Autochthonous cases of nematodiasis present the greatest health risk within the European Union. Foodborne nematodes due to lack of hygiene in food processing are diseases that can be avoided by increasing.
