Scalable continuous flow synthesis of dendritic fibrous nanospheres of silica (DFNS).

The influence of dispersity in the molecular weight distributions in the core forming block for block copolymer (BCP) self-assembly is analyzed via an automated flow synthesis approach.

We integrate continuous flow organocatalysed ring-opening polymerisation with a core-grafting strategy to rapidly and readily prepare multifunctional, processable polyesters. Their potential use in materials applications is subsequently highlighted.

CsPbBr3-(SH)polyHIPE composite was produced by adhering CsPbBr3 nanocrystals to a (SH)-polyHIPE substrate. This well-designed composite has an extremely high sensitivity to copper ions in organic solvents.

Tendons are integral to our daily lives by allowing movement and locomotion but are frequently injured, leading to patient discomfort and impaired mobility. Current clinical procedures are unable to fully restore the native structure of the tendon, resulting in loss of full functionality, and the weakened tissue following repair often re-ruptures. Tendon tissue engineering, involving the combination of cells with biomaterial scaffolds to form new tendon tissue, holds promise to improve patient outcomes. A key requirement for efficacy in promoting tendon tissue formation is the optimal differentiation of the starting cell populations, most commonly adult tissue-derived mesenchymal stem/stromal cells (MSCs), into tenocytes, the predominant cellular component of tendon tissue. Currently, a lack of consensus on the protocols for effective tenogenic differentiation is hampering progress in tendon tissue engineering. In this review, we discuss the current state of knowledge regarding human stem cell differentiation towards tenocytes and tendon tissue formation. Tendon development and healing mechanisms are described, followed by a comprehensive overview of the current protocols for tenogenic differentiation, including the effects of biochemical and biophysical cues, and their combination, on tenogenesis. Lastly, a synthesis of the key features of these protocols is used to design future approaches. The holistic evaluation of current knowledge should facilitate and expedite the development of efficacious stem cell tenogenic differentiation protocols with future impact in tendon tissue engineering. STATEMENT OF SIGNIFICANCE: The lack of a widely-adopted tenogenic differentiation protocol has been a major hurdle in the tendon tissue engineering field. Building on current knowledge on tendon development and tendon healing, this review surveys peer-reviewed protocols to present a holistic evaluation and propose a pathway to facilitate and expedite the development of a consensus protocol for stem cell tenogenic differentiation and tendon tissue engineering.

Highly porous emulsion templated polymers (PolyHIPEs) provide a number of potential advantages in the fabrication of scaffolds for tissue engineering and regenerative medicine. Porosity enables cell ingrowth and nutrient diffusion within, as well as waste removal from, the scaffold. The properties offered by emulsion templating alone include the provision of high interconnected porosity, and, in combination with additive manufacturing, the opportunity to introduce controlled multiscale porosity to complex or custom structures. However, the majority of monomer systems reported for PolyHIPE preparation are unsuitable for clinical applications as they are nondegradable. Thiol-ene chemistry is a promising route to produce biodegradable photocurable PolyHIPEs for the fabrication of scaffolds using conventional or additive manufacturing methods; however, relatively little research has been reported on this approach. This study reports the groundwork to fabricate thiol- and polycaprolactone (PCL)-based PolyHIPE materials via a photoinitiated thiolene click reaction. Two different formulations, either three-arm PCL methacrylate (3PCLMA) or four-arm PCL methacrylate (4PCLMA) moieties, were used in the PolyHIPE formulation. Biocompatibility of the PolyHIPEs was investigated using human dermal fibroblasts (HDFs) and human osteosarcoma cell line (MG-63) by DNA quantification assay, and developed PolyHIPEs were shown to be capable of supporting cell attachment and viability.

Diffusion-ordered NMR spectroscopy (DOSY) allows for accurate molecular weight calibration and determination that can be corrected for solvent influences. Polystyrene and poly(ethylene glycol) standards have been used to calibrate DOSY diffusion data for a variety of solvents, showing a high correlation of data when the bulk viscosity of the solvent is accounted for following the Stokes-Einstein equation. In this way, a type of universal calibration is introduced that allows for determinations of average molecular weight that are at least as accurate as those of traditional size-exclusion chromatography (SEC), if not better. Further, we demonstrate that DOSY calibrations can be used between laboratories, hence removing the need for individual calibration of setups as currently done.

The flocculation efficiency of polyelectrolytes in a high-ionic-strength environment is often affected and reduced due to shielding of the active ionizable functional groups, as well as changes in the surface chemistry of the solid slurry. To address this problem, a series of well-defined novel ABA triblock copolymers were employed for the flocculation of high-ionic-strength kaolin slurries at three different Ca2+ concentrations (0.05, 0.10, and 0.50 M). The primary focus was on the advancement in the polymer architecture, where the anionic functionalities were localized at the terminal ends. Typical commercial flocculants tend to have anionic functionalities randomly distributed throughout the polymer chain and hence a higher propensity toward condensed conformation and formation of insoluble species. In comparison to a control random copolymer, the ABA triblock copolymers were able to flocculate kaolin slurries to give faster settlement rates, particularly at the high Ca2+ concentrations of 0.10 and 0.50 M. In addition, these polymers had significantly better clarification ability at higher Ca2+ concentrations compared to the control random copolymer. The ABA triblock copolymer architecture may therefore have potential as a flocculant in high-ionic-strength applications.

This study investigates the utility of a tailored poly(ethylene glycol) diacrylate-crosslinked porous polymeric tissue engineering scaffold, with mechanical properties specifically optimised to be comparable to that of mammalian brain tissue for 3D human neural cell culture. Results obtained here demonstrate the attachment, proliferation and terminal differentiation of both human induced pluripotent stem cell- and embryonic stem cell-derived neural precursor cells (hPSC-NPCs) throughout the interconnected porous network within laminin-coated scaffolds. Phenotypic data and functional analyses are presented demonstrating that this material supports terminal in vitro neural differentiation of hPSC-NPCs to a mixed population of viable neuronal and glial cells for periods of up to 49 days. This is evidenced by the upregulation of TUBB3, MAP2, SYP and GFAP gene expression, as well as the presence of the proteins βIII-TUBULIN, NEUN, MAP2 and GFAP. Functional maturity of neural cells following 49 days 3D differentiation culture was tested via measurement of intracellular calcium. These analyses revealed spontaneously active, synchronous and rhythmic calcium flux, as well as response to the neurotransmitter glutamate. This tailored construct has potential application as an improved in vitro human neurogenesis model with utility in platform drug discovery programs. STATEMENT OF SIGNIFICANCE: The interconnected porosity of polyHIPE scaffolds exhibits the ability to support three-dimensional neural cell network formation due to limited resistance to cellular migration and re-organisation. The previously developed scaffold material displays mechanical properties similar to that of the mammalian brain. This research also employs the utility of pluripotent stem cell-derived neural cells which are of greater clinical relevance than primary neural cell lines. This scaffold material has future potential in better mimicking three-dimensional neural networks found in the human brain and may result in improved in vitro models for disease modelling and drug screening applications.

Commercial nerve guidance conduits (NGCs) for repair of peripheral nerve discontinuities are of little use in gaps larger than 30 mm, and for smaller gaps they often fail to compete with the autografts that they are designed to replace. While recent research to develop new technologies for use in NGCs has produced many advanced designs with seemingly positive functional outcomes in animal models, these advances have not been translated into viable clinical products. While there have been many detailed reviews of the technologies available for creating NGCs, none of these have focussed on the requirements of the commercialisation process which are vital to ensure the translation of a technology from bench to clinic. Consideration of the factors essential for commercial viability, including regulatory clearance, reimbursement processes, manufacturability and scale up, and quality management early in the design process is vital in giving new technologies the best chance at achieving real-world impact. Here we have attempted to summarise the major components to consider during the development of emerging NGC technologies as a guide for those looking to develop new technology in this domain. We also examine a selection of the latest academic developments from the viewpoint of clinical translation, and discuss areas where we believe further work would be most likely to bring new NGC technologies to the clinic. Statement of significance NGCs for peripheral nerve repairs represent an adaptable foundation with potential to incorporate modifications to improve nerve regeneration outcomes. In this review we outline the regulatory processes that functionally distinct NGCs may need to address and explore new modifications and the complications that may need to be addressed during the translation process from bench to clinic.

High internal phase emulsions (HIPEs), with densely packed droplets of internal phase and monomers dispersed in the continuous phase, are now an established medium for porous polymer preparation (polyHIPEs). The ability to influence the pore size and interconnectivity, together with the process scalability and a wide spectrum of possible chemistries are important advantages of polyHIPEs. In this review, the focus on the biomedical applications of polyHIPEs is emphasised, in particular the applications of polyHIPEs as scaffolds/supports for biological cell growth, proliferation and tissue (re)generation. An overview of the polyHIPE preparation methodology is given and possibilities of morphology tuning are outlined. In the continuation, polyHIPEs with different chemistries and their interaction with biological systems are described. A further focus is given to combined techniques and advanced applications.

Platelets are a reservoir of growth factors, cytokines and chemokines involved in spontaneous wound repair. In this study, a platelet-rich and fibrin-rich hydrogel was generated from expired platelet components that would have otherwise been transfused. The material contained physiological concentrations of transforming growth factor β1 (TGF-β1, platelet-derived growth factor AB (PDGF-AB), PDGF-BB, insulin-like growth factor-1 (IGF-1), fibroblast growth factor 2 (FGF-2), and epidermal growth factor (EGF). The effect of the hydrogel on wound repair was investigated in SKH-1 mice. Full thickness dorsal wounds were created on the mice and treated with the hydrogel at various concentrations. Immunohistochemical staining with CD31 (endothelial cell marker) revealed that wounds treated with the hydrogel showed significantly enhanced vascularisation in the wound bed. Moreover, high levels of interleukin-6 (IL-6) and KC (IL-8 functional homologue) in treated wounds were sustained over a longer period of time, compared to untreated wounds. We postulate that sustained IL-6 is a driver, at least partly, of enhanced vascularisation in full thickness wounds treated with the hydrogel. Future work is needed to explore whether this hydrogel can be utilised as a treatment option when vascularisation is a critical limitation. STATEMENT OF SIGNIFICANCE: The economic cost of wound repair is estimated in billions of dollars each year. In many cases time required to vascularise wounds is a major contributor to slow wound repair. In this study, we developed a blood-derived platelet- and fibrin-rich hydrogel. It contains a number of growth factors actively involved in spontaneous wound healing. This hydrogel significantly improved dermal repair and vascularisation in a full-thickness wound mouse model. This study provides an action mechanism for modulation of localised inflammation.

Thermoresponsive gold nanoparticles were prepared by conjugation of short pentapeptide sequences, containing the elastin-like peptide (ELP) repeat unit Val-Pro-Gly-Val-Gly (VPGVG), to a well-defined active ester prepolymer, poly(pentafluorophenyl acrylate) (pPFPA), itself prepared by RAFT polymerisation. Polymers with number-average chain lengths of 25, 50, 75 and 100 were prepared. The cloud points of their aqueous solutions was found to decrease with chain length and peptide sequence, and varied from 76 to 42 oC. Circular dichroism indicated that the cloud point behaviour was accompanied by a reversible change in peptide conformation from a random coil to a -turn. The resulting thermoresponsive elastin peptide side-chain polymers (ESPs) were then conjugated to gold nanoparticles (GNPs) by ligand exchange. The resulting ESPGNPs similarly displayed thermo-reversible aggregation with transition temperature (Tt) values of 65 and 35 oC for ESPs of chain length 75 and 100, respectively.

Direct glycan-glycan interactions are increasingly implicated in survival and pathogenicity of bacteria. Here, we show that they can be exploited by protozoan parasites in their insect hosts. Force spectroscopy revealed that Leishmania promastigotes display a high-affinity biomolecular interaction between their lipophosphoglycan glycocalyx and mimics of N-acetyl-D-galactosamine, commonly expressed on the midguts of a wide range of sand fly vector species. This enabled gut-adhesive nectomonad promastigotes of Leishmania mexicana to efficiently bind to membrane-bound mucin-like, O-linked glycoproteins of the sand fly Lutzomyia longipalpis, an event crucial for parasite survival, and accounts for a permissive mode of binding. Thus, direct interaction between parasite and sand fly midgut glycans are key to permitting vector competence for all forms of leishmaniasis worldwide. In addition, these studies demonstrate the feasibility of interfering with these interactions as transmission-blocking vaccines.

Methods with which to tailor the morphology of polystyrene-based emulsion-templated (PolyHIPE) materials are presented. Increasing the temperature of the aqueous phase used to prepare the parent emulsion leads to an increase in average void and interconnect size in the resulting porous material. Additionally, the presence in the aqueous phase of small quantities of organic additives that are capable of partitioning between the two emulsion phases also affects the morphology of the porous material obtained. The additives examined were tetrahydrofuran (THF), methanol and poly(ethylene glycol) (PEG), all of which were found to increase the average void and interconnect diameters. It is suggested that THF and, to a lesser extent, PEG enhance Ostwald ripening, resulting in emulsion coarsening over time. Evidence for this was gleaned from NMR experiments to determine the rates of water diffusion in each emulsion. However, methanol was shown not to affect the rate of water diffusion. An alternative mechanism by which methanol could affect the emulsion stability is by depleting surfactant from the interface. However, higher levels of surfactant in emulsions containing methanol did not have a significant effect on morphology. To explain this, we suggest that methanol may result in depletion of surfactant from the emulsion interface, however additional surfactant serves not only to replace this depleted surfactant but also to increase the number of w/o micelles in the continuous phase. These facilitate transport of water between droplets, thus negating the effect of replacing the surfactant lost from the interface.

Efforts in the synthesis of macroporous polymers have mostly been directed towards the formation of stable high internal phase emulsions (HIPEs) from commercially available monomers, limiting their scope of application. Therefore, the development of simple synthetic approaches to access tailor-made macromonomers that can be used as precursors for the formation of HIPEs, allowing the design of new generations of polyHIPE materials with bespoke chemical and physical properties, is desirable in the search for new applications. In this work, cobalt(II) mediated catalytic chain transfer polymerisation (CCTP) is used to polymerise ethylene glycol dimethacrylate (EGDMA), producing multi vinyl-terminated branched EGDMA polymers with tuneable branching density and degree of unsaturation. These materials are subsequently implemented as macromonomer crosslinking agents for the formulation of HIPEs. The use of acrylate comonomers as propagation promoters is found to be essential and 2-ethylhexyl acrylate (EHA), isobornyl acrylate (IBOA) and 2-methoxyethyl acrylate (MEA) are investigated as comonomers in the formulations to both facilitate the photochemical curing of the HIPEs and to impart material properties to the products. The CCTP derived branched macromonomers are fully charaterised by GPC, 1H-NMR and MALDI–ToF spectroscopy. Scanning electron microscopy (SEM) is used to explore the morphology of the produced materials. Surface wettability experiments are conducted to evaluate the hydrophilicity of the polyHIPE surface. Compression tests are used to investigate the influence of the branching density of the CCTP macromonomers as well as the nature of comonomers on the mechanical properties of the materials.

Engineered dermal templates have revolutionised the repair and reconstruction of skin defects. Their interaction with the wound microenvironment and linked molecular mediators of wound repair is still not clear. This study investigated the wound bed and acellular &ldquo, off the shelf&rdquo, dermal template interaction in a mouse model. Full-thickness wounds in nude mice were grafted with allogenic skin, and either collagen-based or fully synthetic dermal templates. Changes in the wound bed showed significantly higher vascularisation and fibroblast infiltration in synthetic grafts when compared to collagen-based grafts (P &le, 0.05). Greater tissue growth was associated with higher prostaglandin-endoperoxide synthase 2 (Ptgs2) RNA and cyclooxygenase-2 (COX-2) protein levels in fully synthetic grafts. Collagen-based grafts had higher levels of collagen III and matrix metallopeptidase 2. To compare the capacity to form a double layer skin substitute, both templates were seeded with human fibroblasts and keratinocytes (so-called human skin equivalent or HSE). Mice were grafted with HSEs to test permanent wound closure with no further treatment required. We found the synthetic dermal template to have a significantly greater capacity to support human epidermal cells. In conclusion, the synthetic template showed advantages over the collagen-based template in a short-term mouse model of wound repair.

A synthetic cell mimic in the form of giant glycosylated polymersomes (GGPs) comprised of a novel amphiphilic diblock copolymer is reported. A synthetic approach involving a poly(dimethylsiloxane) (PDMS) macro-chain transfer agent (macroCTA) and postpolymerization modification was used to marry the hydrophobic and highly flexible properties of PDMS with the biological activity of glycopolymers. 2-Bromoethyl acrylate (BEA) was first polymerized using a PDMS macroCTA ( Mn,th ≈ 4900 g·mol-1, Đ = 1.1) to prepare well-defined PDMS- b-pBEA diblock copolymers ( Đ = 1.1) that were then substituted with 1-thio-β-d-glucose or 1-thio-β-d-galactose under facile conditions to yield PDMS- b-glycopolymers. Compositions possessing ≈25% of the glycopolymer block (by mass) were able to adopt a vesicular morphology in aqueous solution (≈210 nm in diameter), as indicated by TEM and light scattering techniques. The resulting carbohydrate-decorated polymersomes exhibited selective binding with the lectin concanavalin A (Con A), as demonstrated by turbidimetric experiments. Self-assembly of the same diblock copolymer compositions using an electroformation method yielded GGPs (ranging from 2-20 μm in diameter). Interaction of these cell-sized polymersomes with fimH positive E. coli was then studied via confocal microscopy. The glucose-decorated GGPs were found to cluster upon addition of the bacteria, while galactose-decorated GGPs could successfully interact with (and possibly immobilize) the bacteria without the onset of clustering. This demonstrates an opportunity to modulate the response of these synthetic cell mimics (protocells) toward biological entities through exploitation of selective ligand-receptor interactions, which may be readily tuned through a considered choice of carbohydrate functionality.

Ultra-high molecular weight (UHMW, Mn > 1000 kDa) polymeric drift control adjuvants (DCAs) for agricultural spraying are prone to mechanical degradation and rapidly lose performance. To overcome this, we have designed linear coordination polymers (LCPs) composed of 400 kDa telechelic bis-terpyridine end-functionalised polyacrylamide units, which ‘self-heal’ upon shearing through reformation of coordination bonds. After addition of Fe(II) to dilute aqueous solutions of the terpyridine telechelics, UHMW LCPs were obtained as demonstrated by UV-vis spectroscopy, MALS GPC and intrinsic viscosity measurements. Importantly, these UHMW LCPs were shown to function as effective DCAs, reducing the formation of fine ‘driftable’ droplets during spray testing at concentrations as low as 100 ppm. Following mechanically-induced coordination bond-scission, the UHMW LCPs were found to recover up to 90% of their performance compared to un-sheared samples, at a rate dependent on the transition metal ion used to form the complex.

Novel approaches for culturing primary human cells in vitro are increasingly needed to study cell and tissue physiology and to grow replacement tissue for regenerative medicine. Conventional 2D monolayer cultures of endometrial epithelial and stromal cells fail to replicate the complex 3D architecture of tissue. A fully synthetic scaffold that mimics the microenvironment of the human endometrium can ultimately provide a robust platform for investigating tissue physiology and, hence, take significant steps toward tackling female infertility and IVF failure. In this work, emulsion-templated porous polymers (known as polyHIPEs) were investigated as scaffolds for the culture of primary human endometrial epithelial and stromal cells (HEECs and HESCs). Infiltration of HEECs and HESCs into cell-seeded polyHIPE scaffolds was assessed by histological studies, and phenotype was confirmed by immunostaining. Confocal microscopy revealed that the morphology of HEECs and HESCs is representative of that found in vivo. RNA sequencing was used to investigate transcriptome differences between cells grown on polyHIPE scaffolds and in monolayer cultures. The differentiation status of HEECs and HESCs grown in polyHIPE scaffolds and in monolayer cultures was further evaluated by monitoring the expression of endometrial marker genes. Our observations suggest that a 3D cell culture model that could approximate native human endometrial architecture and function can be developed using tailored polyHIPE scaffolds.\ud \ud

Thermally-responsive, short elastin-like peptides (ELPs) of sequence VPGVG (V, P and G represent valine, proline and glycine respectively), bearing different N-terminal functional groups (amino-, N-acetyl and thiol) and a non-ionisable C-terminal group, were prepared by solid phase synthesis. The conformation and aggregation properties of the ELPs were studied in different pH aqueous buffer solutions using UV-vis spectroscopy and circular dichroism (CD). The thiol-capped ELPs were used to prepare functionalized gold nanoparticles (GNPs), which were found to undergo thermally-triggered reversible aggregation at 40 °C. The peptide conformation and nanoparticle aggregation behaviour of the ELP-GNPs in aqueous solution were investigated by transmission electron microscopy (TEM), circular dichroism (CD) and UV-vis spectroscopy. It was found that the ELP-GNP conjugates were capable of reversible, thermally triggered aggregation at near-physiological temperatures (transition temperature of 40 °C at pH = 7.4), opening up applications in photothermal cancer therapy and diagnosis.

In the last decade, interest in the field of three-dimensional (3D) bioprinting has increased enormously. 3D bioprinting combines the fields of developmental biology, stem cells, and computer and materials science to create complex bio-hybrid structures for various applications. It is able to precisely place different cell types, biomaterials and biomolecules together in a predefined position to generate printed composite architectures. In the field of tissue engineering, 3D bioprinting has allowed the study of tissues and organs on a new level. In clinical applications, new models have been generated to study disease pathogenesis. One of the most important components of 3D bio-printing is the bio-ink, which is a mixture of cells, biomaterials and bioactive molecules that creates the printed article. This review describes all the currently used bio-printing inks, including polymeric hydrogels, polymer bead microcarriers, cell aggregates and extracellular matrix proteins. Amongst the polymeric components in bio-inks are: natural polymers including gelatin, hyaluronic acid, silk proteins and elastin; and synthetic polymers including amphiphilic block copolymers, PEG, poly(PNIPAAM) and polyphosphazenes. Furthermore, photocrosslinkable and thermoresponsive materials are described. To provide readers with an understanding of the context, the review also contains an overview of current bio-printing techniques and finishes with a summary of bio-printing applications.

Accurate and reliable analysis of polymers by GPC is vital in the field of controlled radical polymerisation. Often, water-soluble polymers are analysed by aqueous gel permeation chromatography (GPC) in a solvent containing dilute sodium azide as an anti-microbial agent. Previous reports have shown that sodium azide at high concentration is able to remove terminal CTA groups from polymer chains, producing thiol-terminated polymers. This study demonstrates that GPC sample preparation of RAFT polymers in aqueous solvents containing dilute (200 ppm) sodium azide can cause significant changes in the measured molecular weight distribution. These changes occur within hours of dissolving the polymer sample and are shown to be due to cleavage of the CTA in the polymer chain together with disulfide coupling of the resulting polymeric thiols. The extent to which this occurs is strongly dependent on the CTA attached to the polymer; an almost 10-fold difference in the rate of CTA removal is observed between different RAFT agents. The by-product of the reaction between sodium azide and RAFT polymers is also investigated and shown to be an unstable thiatriazole-functionalised Z group. The thiatriazole then degrades further to form a nitrile-functionalised Z group, N2 and elemental sulfur.

A new facile and efficient route for the chemical functionalisation of thiol-acrylate polyHIPE materials with responsive macromolecules using the highly emissive dithiomaleimide (DTM) linker is demonstrated. Functionalisation is found to be reversible upon addition of a thiol-containing compound, glutathione, resulting in switchable surface properties including fluorescence and wettability, hence broadening the scope of applications.

The work here describes the synthesis of tailor-made, porous, polymeric materials with elastic moduli in the range associated with mammalian brain tissue (0.1–24 kPa). Three new emulsion-templated porous polymer materials (polyHIPEs) were synthesised by thiol–ene photopolymerisation from hexanediol diacrylate (HDDA) and polyethylene glycol diacrylate (PEGDA) crosslinkers and compared with a previously reported material prepared from trimethylolpropane triacrylate (TMPTA). The materials were found to have an average pore diameter of 30–63 μm and a porosity of 77% and above. PEGDA crosslinked materials at 80 and 85% porosity, when swollen in PBS at 37 °C, were found to have an elastic modulus of 18 and 9.0 kPa respectively. PEGDA crosslinked materials were also found to have a swelling ratio of 700% in PBS at 37 °C. PEGDA crosslinked materials had improved visible light transmission properties when compared to TMPTA crosslinked materials under a bright field microscope. All materials were shown via hematoxylin and eosin staining to support the infiltration and attachment of induced pluripotent stem cell (iPSC)-derived human neural progenitor cells (hNPCs). HNPCs on all materials were demonstrated in short term 3D cultures to maintain a phenotype consistent with early neural lineage specification via immunohistochemical staining for the intermediate filament protein vimentin.

The movement of a pesticide or herbicide to an off-target site during agricultural spraying can cause injury to wildlife, plants and contamination of surface water. This phenomenon is known as spray drift and can be controlled by spraying during favorable environmental conditions, and by using low drift nozzles and drift control adjuvants (DCAs). Polymeric DCAs are the most common type of DCA and function by increasing the droplet size produced during spraying. There are, however, two main drawbacks of polymeric DCAs; they are prone to mechanical degradation during spraying which reduces their performance and they can produce oversized drops which reduces the efficacy of the spray. In this trend article, existing DCA technology is reviewed including the mechanism through which they function. This then provides a platform for the discussion of novel polymeric architectures which have currently not been applied in DCA formulations.

Skin-like energy devices can be conformally attached to the human body, which are highly desirable to power soft wearable electronics in the future. Here, a skin-like stretchable fuel cell based on ultrathin gold nanowires (AuNWs) and polymerized high internal phase emulsions (polyHIPEs) scaffolds is demonstrated. The polyHIPEs can offer a high porosity of 80% yet with an overall thickness comparable to human skin. Upon impregnation with electronic inks containing ultrathin (2 nm in diameter) and ultrahigh aspect-ratio (>10 000) gold nanowires, skin-like strain-insensitive stretchable electrodes are successfully fabricated. With such designed strain-insensitive electrodes, a stretchable fuel cell is fabricated by using AuNWs@polyHIPEs, platinum (Pt)-modified AuNWs@polyHIPEs, and ethanol as the anode, cathode, and fuel, respectively. The resulting epidermal fuel cell can be patterned and transferred onto skin as "tattoos" yet can offer a high power density of 280 µW cm-2 and a high durability (>90% performance retention under stretching, compression, and twisting). The results presented here demonstrate that this skin-thin, porous, yet stretchable electrode is essentially multifunctional, simultaneously serving as a current collector, an electrocatalyst, and a fuel host, indicating potential applications to power future soft wearable 2.0 electronics for remote healthcare and soft robotics.

Catechol-Fe(III) complexes contain some of the strongest known metal-chelate coordination bonds. Despite this, they have until now not been utilized in (polymeric linker) linear coordination polymer (LCP) synthesis. With the view of generating catechol end-functional polymers, a new, symmetrical bis-catechol functionalized trithiocarbonate reversible addition fragmentation chain transfer (RAFT) agent is synthesized (CatDMAT). Acrylamide (AM) and dimethylacrylamide (DMA) polymerizations are conducted with CatDMAT using direct photoactivation RAFT polymerization to yield bis-catechol end-functionalized homo- and block-copolymers of molecular weight 10-15 kDa. Catechol-Fe(III) LCPs are successfully formed from the telechelic catechol polymers by bis-complexation to Fe(III). The tetrahedral bis-complex is detected by UV-vis spectroscopy (λmax = 570 nm), while increases in relative viscosity and Mn,GPC over their respective uncomplexed polymers confirm the occurrence of supramolecular polymerization. The catechol-LCPs are shown to undergo oxidation and crosslinking in aqueous solution after 24 h.

A novel strategy for the surface functionalization of emulsion-templated highly porous (polyHIPE) materials as well as its application to in vitro 3D cell culture is presented. A heterobifunctional linker that consists of an amine-reactive N-hydroxysuccinimide ester and a photoactivatable nitrophenyl azide, N-sulfosuccinimidyl-6-(4'-azido-2'-nitrophenylamino)hexanoate (sulfo-SANPAH), is utilized to functionalize polyHIPE surfaces. The ability to conjugate a range of compounds (6-aminofluorescein, heptafluorobutylamine, poly(ethylene glycol) bis-amine, and fibronectin) to the polyHIPE surface is demonstrated using fluorescence imaging, FTIR spectroscopy, and X-ray photoelectron spectroscopy. Compared to other existing surface functionalization methods for polyHIPE materials, this approach is facile, efficient, versatile, and benign. It can also be used to attach biomolecules to polyHIPE surfaces including cell adhesion-promoting extracellular matrix proteins. Cell culture experiments demonstrated that the fibronectin-conjugated polyHIPE scaffolds improve the adhesion and function of primary human endometrial stromal cells. It is believed that this approach can be employed to produce the next generation of polyHIPE scaffolds with tailored surface functionality, enhancing their application in 3D cell culture and tissue engineering whilst broadening the scope of applications to a wider range of cell types.

This study investigates the utility of a novel poly(ethylene glycol) diacrylate-crosslinked porous polymeric tissue engineering scaffold, with mechanical properties specifically optimised to be comparable to that of mammalian brain tissue for 3D human neural cell culture. Results obtained here demonstrate the attachment, proliferation and terminal differentiation of both human induced pluripotent stem cell- and embryonic stem cell-derived neural precursor cells (hPSC-NPCs) throughout the interconnected porous network within laminin-coated scaffolds. Phenotypic data and functional analyses are presented demonstrating that this material supports terminal in vitro neural differentiation of hPSC-NPCs to a mixed population of viable neuronal and glial cells for periods of up to 49 days. This is evidenced by the upregulation of TUBB3, MAP2, SYP and GFAP gene expression, as well as the presence of the proteins βIII-TUBULIN, NEUN, MAP2 and GFAP. Functional maturity of neural cells following 49 days 3D differentiation culture was tested via measurement of intracellular calcium. These analyses revealed spontaneously active, synchronous and rhythmic calcium flux, as well as response to the neurotransmitter glutamate. This novel construct has potential application as an improved in vitro human neurogenesis model with utility in platform drug discovery programs.

Film-forming polymer latex particles of diameter300 nm can be prepared in the complete absence of surfactants, stabilised in part by silica nanoparticles through a Pickering type emulsion polymerisation. Control of the silica wettability through modulation of reaction pH or by reaction of the nanoparticles with a hydrophobic silane results in silica-covered latex particles.The oil-in-water polymerisation process used methyl methacrylate (MMA) and n-butyl acrylate (BA) as co-monomers, potassium persulphate (KPS) as an initiator and a commercially available colloidal nano-silica (Ludox®-TM40). It was found that pH control before polymerisation using methacrylic acid (MAA) facilitated the formation of armoured latexes, and mechanistic features of this process are discussed. An alternative, more robust protocol was developed whereby addition of vinyltriethoxysilane (VTES) to control wettability resulted in latexes completely armoured in colloidal nano-silica. The latexes were characterised using SEM, cryo-TEM and AFM imaging techniques. The mechanism behind the adsorption was investigated through surface pressure and contact angle measurements to understand the factors that influence this irreversible adsorption.Results indicate that nanoparticle attachment (but intriguingly not latex size) is dependent on particle wettability, providing new insight into the formation of nanoparticle-armoured latexes, along with opportunities for further development of diversely functionalized inorganic/organic polymer composite particles.

Flocculation is a common technique that is widely used in many industrial applications to promote solid–liquid separation processes. The addition of a polymeric flocculant allows for the destabilization of suspended colloidal particles, and thus significantly increases their sedimentation rate. Polymeric flocculants are generally divided into four categories, which include non-ionic, cationic, anionic, and amphoteric polymers. This minireview article summarises important information on the recent design and synthesis of polymeric materials from these four categories. In addition, their properties and flocculation efficiency are also presented and discussed.\ud \ud

Poly(ionic liquid)s (P(IL)s) of different degrees of polymerization (10, 50, and 100) were prepared via RAFT polymerization using an alkyne-terminated xanthate as transfer agent, with a monomer conversion of up to ∼80% and a ĐM of 1.5 for P(IL)100. Subsequently, P(IL) chains were coupled to 15N-labeled azido-functionalized hydroxyethyl cellulose (HEC), forming graft copolymers of HEC with different chain length and graft densities, which were characterized using (13C and 15N) CP-MAS NMR and FT-IR spectroscopies. The antibacterial activities of HEC-g-P(IL)s were tested against Escherichia coli and Staphylococcus aureus and were comparable to ampicillin, a well-known antibiotic, demonstrating efficient activity of the graft copolymers against bacteria. Moreover, HEC-g-P(IL)s were slightly more effective against E. coli than S. aureus. A decrease in graft density of P(IL)10 on the HEC backbone decreased the activity of the graft copolymers against both bacteria. These findings suggest that HEC-g-P(IL) could find applications as an antiseptic compound, for example, in paint formulation.\ud \ud

Emulsion-templated highly porous polymers (polyHIPEs), containing distinct regions differing in composition, morphology, and/or properties, are prepared by the simultaneous polymerization of two high internal phase emulsions (HIPEs) contained within the same mould. The HIPEs are placed together in the mould and subjected to thiol-acrylate photopolymerization. The resulting polyHIPE material is found to contain two distinct semicircular regions, reflecting the composition of each HIPE. The original interface between the two emulsions becomes a copolymerized band between 100 and 300 μm wide, which is found to be mechanically robust. The separate polyHIPE layers are distinguished from one another by their differing average void diameter, chemical composition, and extent of contraction upon drying.

The emulsion templating process offers a route to highly porous polymers with well-defined morphologies. This study describes the preparation of such porous polymers (polyHIPEs) via the photopolymerization of a multi-functional thiol and polycaprolactone macromonomer. The resulting materials have nominal porosities of 90% and 95%, and are seen to have an interconnected pore morphology, with an average pore diameter of approximately 60 μm. Initial biocompatibility assessments with fibroblast cells (L929) have shown that the polymers are capable of supporting cell growth over 7 days and degradation products are non-toxic to cells up to a concentration of 0.1 mg ml−1.\ud \ud

The synthesis of ultra-high molecular weight (UHMW) polymers using reversible deactivation radical polymerisation techniques remains a challenge and has been the centre of attention only in a limited number of studies. Although UHMW polymers were synthesised in these research studies, the complexity in architecture has mainly been limited to linear homopolymers and AB diblock copolymers. We hereby report a new pathway to synthesise UHMW ABA triblock copolymers using a combination of reversible addition–fragmentation chain transfer (RAFT) polymerisation, end-group modification by aminolysis and chain coupling. A simple aqueous RAFT-mediated gel polymerisation technique was initially employed to synthesise high molecular weight AB diblock copolymers with low dispersities (Đ < 1.50). The use of the said gel polymerisation method in combination with a redox initiation system allowed for the rapid chain propagation of water-soluble monomers at a low reaction temperature of 20 °C. These polymers were subsequently treated by aminolysis to convert the chain end into a thiol functionality, which spontaneously coupled under oxidative conditions to form a disulfide bridge between the AB diblock copolymers to produce the final UHMW ABA triblock copolymers (Mn,SEC > 1000k; Đ < 1.70).

The chemical and mechanical properties of macroporous polymer substrates play a crucial role in the determination of their end-application. The preparation of highly (macro)porous monolithic polymers (polyHIPEs) by emulsion templating and thiol-ene/yne photopolymerisation, using multifunctional acrylate, allyl ether and alkyne-based monomers with trimethylolpropane tris (3-mercaptopropionate) (TMPTMP), is described in this work. Issues associated with monomer solubility and/or stability of the produced high internal phase emulsions (HIPEs) are tackled. Scanning electron microscopy (SEM) is used to study the morphology and porosity (average void diameters) of the obtained materials. Due to the nature of the photoinitiated thiol-ene reactions, materials obtained from acrylate monomers display residual thiols that are quantified by a colourimetric (Ellman's) assay. Raman spectroscopy is also shown to be a complementary technique to evaluate the residual thiol content. The influence of the monomer functionality on the mechanical properties of the material is explored using compression tests. Significant differences in the surface functionality and mechanical behavior between materials prepared with comonomers able to homopolymerise (acrylates) and those unable to homopolymerise (allyl ethers; alkynes) are demonstrated.\ud \ud

Emulsion-templated (polyHIPE) scaffolds for bone tissue engineering were produced by photopolymerisation of a mixture of trimethylolpropane tris(3-mercaptopropionate) and dipentaerythritol penta-/hexaacrylate in the presence of hydroxyapatite (HA) or strontium-modified hydroxyapatite (SrHA) nanoparticles. Porous and permeable polyHIPE materials were produced regardless of the type or incorporation level of the bioceramic, although higher loadings resulted in a larger average pore diameter. Inclusion of HA and SrHA into the scaffolds was confirmed by EDX-SEM, FTIR and XPS and quantified by thermogravimetry. Addition of HA to polyHIPE scaffolds significantly enhanced compressive strength (148–216 kPa) without affecting compressive modulus (2.34–2.58 MPa). The resulting materials were evaluated in vitro as scaffolds for the 3D culture of MG63 osteoblastic cells vs. a commercial 3D cell culture scaffold (Alvetex®). Cells were able to migrate throughout all scaffolds, achieving a high density by the end of the culture period (21 days). The presence of HA and in particular SrHA gave greatly enhanced cell proliferation, as determined by staining of histological sections and total protein assay (Bradford). Furthermore, Von Kossa and Alizarin Red staining demonstrated significant mineralisation from inclusion of bioceramics, even at the earliest time point (day 7). Production of alkaline phosphatase (ALP), an early osteogenic marker, was used to investigate the influence of HA and SrHA on cell function. ALP levels were significantly reduced on HA- and SrHA-modified scaffolds by day 7, which agrees with the observed early onset of mineralisation in the presence of the bioceramics. The presented data support our conclusions that HA and SrHA enhance osteoblastic cell proliferation on polyHIPE scaffolds and promote early mineralisation.
