Intraoperative (n = 290) and autoptic (n = 246) materials obtained from tuberculosis hospitals in 2005-2007 were examined to study the pathomorphism of pulmonary tuberculosis. The study revealed an increase in the number of patients with acute pulmonary tuberculosis. Fibrocavernous tuberculosis halved as compared to the 1980s. Incapsulated pulmonary tuberculosis is prevalent in the phthisiosurgical material (more than 70% of cases). The leading cause of pulmonary tuberculosis was progression of a specific inflammatory process while cardiopulmonary failure and visceral amyloidosis became rare causes of death. The higher incidence of poorly progressive pulmonary tuberculosis in males (they accounted for 85% of the number of death cases) was associated with concomitant and aggravating diseases, such as alcoholism, drug addiction, and HIV infection (68.5% of cases).

We studied the possibility of evaluation of the intensity of pathological oxidative processes in rat liver using an integral bioluminescent test with controlled perfusion of the isolated organ. The test revealed a significant correlation between the level of TBA-reactive products and bioluminescence intensity.

ATP activity in mouse bone marrow cells was studied in in vitro experiments. ATP in physiological and supraphysiological concentrations stimulated blebbing of the plasma membrane. Subacute administration of doxorubicin (inductor of oxidative stress) decreased cell sensitivity to ATP, which corresponded to changes in membrane lipid peroxidation.

Studies by the method of splenic exocolonies with serotonin receptor blockers methysergide and cyproheptadine showed that S2 receptors are involved in the stimulatory effect of serotonin on hemopoietic bone marrow stem cells.

We studied in vitro cytostatic effect of doxorubicin on bone marrow cells preincubated with NAD and reduced glutathione. The presence of NAD and glutathione in the medium potentiated membrane toxicity of this antibiotic, but prevented its toxic effects on intracellular pool of pyridine nucleotides during oxidative stress.

We studied the effects of cis-dichlorodiammine platinum (II) on different pathways of cell death in cultured Ehrlich ascites carcinoma in vitro and on subsequent growth of transplanted tumor in vivo. One-hour incubation with the cytostatic modulated apoptosis in cell culture. However, exposure of cell culture to a minimal effective concentration of cis-platinum(II)diammine dichloride promoted the growth of transplanted tumor.

The metabolic state of rat liver was studied in vivo 1, 6, and 18 h after single hyperthermia (42 degrees C). It was shown that intracellular compensatory reactions involved in restoration of the energy state and realization of endogenous cytoprotective mechanisms play a role during recovery.

The metabolic changes in rat liver were studied during a reduction period after the overheating (25 min, 42 degrees C) using the isolated organ perfusion. First six hours after the treatment were characterised by a decrease of the respiration rate, the activation of glycolysis and an exhaustion of hepatic carbohydrate resources. The development of adaptive reaction resulted in the recovery of respiration rate to 18 h. This may provide subsequent restoration of hepatic metabolism and normalisation of the organ functioning.

The work shows the results of experimental study of the characteristics of serotonin regulation of cell proliferation and programmed cell death in bone-marrow cells cultivated in diffusion chambers. It was found that the change in cell colony and cluster formation and death induced by this biogenic enzyme is disturbed by acrylamide, an inducer of oxidizing stress. Nicotinamide levels out the effect of acrylamide, which is evidence of the existence of NAD-dependent mechanisms in the serotonin-mediated regulation of hemopoiesis.

Haemopoietic stem cells content and proliferative activity were studied in the bone marrow of female F1 (CBA x C57Bl6) mice after single (50 mg/kg) and chronic (0.5 mg/kg daily for 7 days) serotonin (S) injections. It is shown that 9-day and 12-day COEs contents in the bone marrow of experimental mice has been increasing for 24 h after single S injection. After chronic S injections twofold increase of 12-day COEs is observed without any increasing of 9-day COEs. Total myelokaryocyte number is increased too. The study of proliferative status by in vitro incubation of bone marrow cells with ARA-C has shown that the numbers of 9-day and 12-day COEs in S-phase have increased both after single and chronic S injections. Possible mechanisms of stimulating effect of S on bone marrow stem cells are discussed.

Hypoxemia (45-minute) influence in vivo on erythropoietic activity of the kidney, liver, spleen, and sternum was studied by normoxemic perfusion of the isolated organs. The erythropoietic activity proved to increase after 6-hour perfusion of the liver; this confirmed the participation of this organ in the extrarenal secretion of the erythropoietic factor.

The balance of erythropoietin production by the dog kidney and liver was studied during controlled normoxic perfusion. The hormone production was stimulated by acute posthemorrhagic anemia (bloodletting of 25% total blood volume) combined with subcutaneous injection of cobaltous chloride (250 microM/kg body weight). The increase in erythropoietin level was revealed in posthypoxic animal perfusate after 6 hours of perfusion. The amount of hepatic erythropoietin was shown to be 2.5 times higher than that excreted by kidneys.

The influence of exogenous serotonin on cell division of L929 and L-41 cell strains has been investigated under various conditions of cell growth in culture (the incubation either in 10% serum medium without changing the medium, or in the medium with 0.5% serum). The data obtained show that serotonin in physiological concentration (10(-7) M) stimulates proliferation of resting cells. In proliferating cells, compared to resting ones, the sensitivity to exogenous amine appeared statistically non-significant. Exogenous serotonin is suggested to be a proliferating stimulus for resting cells.

The dynamics of the amino acid composition of the medium under conditions of adequate perfusion of the isolated organs of a dog (sternum, kidney and liver) was studied. It was found that after a 6-hour perfusion of the complex of organs the amount in the perfusion medium of such amino acids as histidine, lysine, alanine, considerably increased, whereas the amount of arginine, serine, aspartic acid, threonine with glutamine, isoleucine, proline, leucine and valine decreased as compared with their initial concentration. The dynamics of the amino acid medium composition during a 4-hour perfusion was studied in experiments with the isolated sternum. The concentration of alanine, lysine and histidine increased in the medium. At the same time there was seen a decrease in the concentration of serine, aspartic acid, isoleucine, tyrosine and phenyl-alanine.

The dynamics of the medium amino acid composition was studied during a 6-hour perfusion of dog kidney and liver by a mixture of autogenic plasma and medium 199 (a ratio of 2 : 3). In comparison to the initial level the amount of histidine during 6-hour cultivation of the kidney was found to increase 2.2 times, that of glutamic acid--1.7 times, and of alanine and lysine--1.6 times. At the same time the amount of arginine, serine and asparagic acid became 3.3 times and of glutamine and threonine--2.5 times lower. With the liver perfusion the level of glutaminic acid became 2.9 times, of alanine--2.3 times, of cystine--2 times and of glycine--1.5 times higher. With the liver perfusion the level of arginine decreased so rapidly that none was found in the medium after a 2-hour perfusion. The described method of amino acid analysis during the cultivation of the organs could serve as means for elaborating and correcting the culture media.
