Background: Diabetes mellitus, a metabolic disorder caused by a defect in the secretion or function of insulin. Arginine is an amino acid that is lower in these patients. Octopine is an arginine derivative. Therefore, the present study evaluated the effect of opine on glucose and stress oxidative indices in streptozotocin-induced diabetic rats. Methods: In this study, 16 male mice were divided into two groups including diabetic and non-diabetic controls and 16 male mice in two experimental groups. The first experimental group received opine treatment at 50 mg and second experimental group received opine treatment at 100 mg of opine for 3 weeks. Diabetes was induced in diabetic rats through intraperitoneal injection of 60 mg/kg of Streptozotocin. Blood samples were enrolled from mice in fasting status and the level of malondialdehyde (MDA) was measured by thiobarbituric acid method, glucose levels using Pars-Azmoon Company Kit, superoxide dismutase (SOD) and glutathione peroxidase (GPX) activity using Randox (England) kits. Results: The opine consumption had no significant effect on serum glucose and MDA, while 50 mg opine treatment significantly increased the activity of SOD and GPX enzymes in comparison with diabetic rats. Conclusion: The consumption of octopine had no improving effect on blood glucose levels but it improved antioxidant status in diabetic rats.

Background: Helicobacter pylori (H. pylori) plays the primary role in increasing oxidative stress and causing stomach inflammation, peptic ulcers, and gastric malignancy in the infected patients. L-arginine (Arg) has antibacterial and anti-inflammatory effects. Objectives: The current study aimed at investigating the beneficial effects of L-arginine on inflammation and oxidative stress in patients infected with H. pylori with dyspeptic symptoms. Methods: The current randomized, double-blind controlled, clinical trial was conducted on 34 patients with H. pylori infection referred to the center of digestive disorders affiliated to Isfahan University of Medical Sciences, Isfahan, Iran, in order to undergo endoscopy from December 2016 to September 2017. Patients were classified into two groups (control and treatment); the control group only received triple-drug therapy (including Amoxicillin, Clarithromycin, and Omeprazole), and the treatment group received standard triple-drug therapy and L-Arg capsules for three weeks. Gastric biopsies and serum samples were taken from all patients before and after the study. H. pylori infection was examined by a rapid urease test and antioxidant indices including superoxide dismutase (SOD), glutathione peroxidase (GPX), and total antioxidant capacity (TAC) were evaluated in gastric biopsies. In addition, serum samples were used to measure the inflammation factors including interleukin (IL)-8 and tumor necrosis factor (TNF)-α. Results: Level of SOD activity increased significantly in the treatment group compared with that of the control group (4.91 ± 95.21 vs. 4.0 ± 44.11 IU/mg) (P = 0.001). In the treatment group, compared with the control group, the level of TAC increased significantly (0.35 ± 0.60 vs. 0.30 ± 0.9 mM/L) (P = 0.006) and the level of GPX activity increased significantly in the treatment group compared with the control group (10.68 ± 2.39 vs. 5.16 ± 2.12 IU/mg) (P = 0.000). Regarding the inflammation factor, IL-8 decreased significantly in the treatment group compared with the control group (8.00 ± 1.94 vs. 10.28 ± 2.10 pg/mL) (P = 0.002); also TNF-α decreased significantly in the treatment group compared with the control group (9.71 ± 2.69 vs. 12.24 ± 3.29 pg/mL) (P = 0.036), while there was no significant difference in high sensitivity C-reactive protein (hs-CRP) decrease between the treatment and the control groups (2.34 ± 1.28 vs. 3.04 ± 1.58 mg/L) (P = 0.16). Conclusions: Consumption of L-arginine increased antioxidant indices and decreased inflammation in patients infected with H. pylori.

Actin is one of the most widely studied structural and multifunctional housekeeping proteins in eukaryotic cells with important roles in many cell functions. Antibodies against β-actin and other housekeeping gene-encoded proteins are used as internal loading controls in Western blot analyses. The aim of this study was to produce a monoclonal antibody (mAb) against a synthetic peptide derived from N-terminal region of β-actin and to study its reactivity with different organisms. A synthetic peptide, derived from β-actin, was designed and used to produce a mAb by hybridoma technology. The produced antibody (clone 4E5- A10) was purified by an affinity chromatography column followed by characterization of purified mAb using SDS-PAGE, ELISA and Western blot. Our results showed that 4E5-A10 was an IgM and had desired purity and excellent reactivity with the immunizing peptide with an affinity constant of 2.7x10 8 M -1 >. It could detect a band of about 45 kDa, corresponding to β-actin, in Western blot. Furthermore, it could react in a more sensitive manner and with a wider range of organisms than a known commercial anti β-actin antibody. Our data suggest that 4E5-A10 can act as a sensitive probe for detection of β-actin as an internal loading control, for a wide range of organisms, in Western blot analyses.

Objective . To investigate the prevalence, characteristics (malocclusion, location, type) and gender distribution of supernumerary teeth (ST) in an Iranian orthodontic population. Material and methods . A retrospective study was carried out using radiographs (periapicals, panoramic) and study models of 1751 subjects (870 girls, 881 boys; age range 9–27 years). The Pearson chi-square test was used to determine differences in the distribution of ST when stratified by gender and malocclusion type. Results . Fourteen ST were found in 13 patients (six males, seven females), giving a prevalence rate of 0.74%. The prevalence rate of ST was higher in Class III malocclusions (5.2%) and was a rare finding among Class II malocclusions (0.1%). The occurrence rate of ST was higher in maxilla (78.5%) than in mandible (21.5%). The most common type of ST was the conical form (43%). We did not observe any late-developing paramolar or distomolar ST. Conclusion . The prevalence rate of ST in our study was 0.74% and...

To investigate the prevalence, characteristics (ie, malocclusion, location, type), and sex distribution of hypodontia in an Iranian orthodontic population. A retrospective study was conducted using periapical and panoramic radiographs and study models of 1,751 subjects attending university orthodontic clinics (870 females, 881 males, age 9-27 years). The Chi-square test was used to analyze differences in the distribution of hypodontia, after stratification by sex and malocclusion type. A total of 197 congenitally missing teeth were observed in 160 patients (9.1%; 74 boys and 86 girls); there were no statistically significant differences between sexes (Chi-square = 0.832, P = 0.36). Hypodontia was more common in patients with Class III malocclusion (45.2%), and was more prevalent in the maxilla (71%) than in the mandible (29%). Maxillary lateral incisors (35.6%) and maxillary second premolars (13.0%) were the most commonly missing teeth, followed by mandibular lateral incisors (9.6%) and mandibular second premolars (8.2%). The prevalence of missing teeth was higher in the anterior segment (incisors and canines) than in the posterior segment (premolars and molars). The prevalence of oligodontia was 0.34%. The prevalence and characteristics of the most frequently missing teeth accorded with the findings of most studies conducted in other countries.

Objectives: This in-vitro study aimed at evaluating the effect of three debonding sequences on the shear bond strength (SBS) of new stainless steel (SS) brackets. Materials and Methods: Stainless steel twin brackets (0.022-inch, American Orthodontics, Sheboygan, WI, USA) were bonded with light cure adhesive (Transbond XT, 3M Unitek, St. Paul, MN, USA) to 80 newly extracted human premolars after acid etching with 37% phosphoric acid (30 s). Brackets were debonded with a universal testing machine, and new brackets were bonded to teeth using the same adhesive and same manner. This process was repeated twice, and brackets were debonded within 24 h after bonding. The longitudinal changes of average SBS were assessed with the repeated measures ANOVA. Post-hoc tests using the Bonferroni correction were also used to compare the average SBS at three debonding sequences. Result: The mean SBS decreased significantly after each debonding sequence (P < 0.01). The corresponding mean values (standard deviation, 95% CI) after the first, second, and third debonding sequences were 22.88 MPa (4.08, 21.97-22.79), 19.36 MPa (4.54, 18.62-20.64), and 16.67 MPa (4.27, 15.72-17.62), respectively. There was no significant difference among the adhesive remnant index (ARI) scores of three debonding sequences (χ2 = 5.067, df = 6, P = 0.53). Conclusion: Average SBS after three debonding sequences was significantly decreased, but was above the recommended 5.9-7.8 MPa. In-vivo studies are required to validate the finding of this study.

To compare the in-vitro Weekly Cumulative Fluoride Release (WCFR) of three Glass Ionomer Cements (GICs) used for orthodontic banding.The GICs tested were Granitec (Confi-Dental, Louisville, CO, USA), Bandtite (American Orthodontics, Sheboygan, WI, USA) and Ariadent (Apadana Tak Co, Tehran, Iran). Fifteen discs of each GIC were constructed (6mm diameter and 1.5mm depth). Specimens were immersed in 5 ml of deionised water and the WCFR was measured at weekly intervals, on days 1, 8, 15, 22 and 29 after immersion in deonised water, using the potentiometery device and single junction saturated calomel electrode technique (Jenway, England, UK). To compare the WCFR profile of 3 GICS, data were subjected to the one-way analysis of variance (ANOVA), and were appropriate, the Scheffe or Tamhane multiple comparison tests (post-hoc). For assessing the longitudinal changes of average WCFRs in 3 GICs, the repeated measures ANOVA were used. Post hoc tests using the Bonferroni correction was also used to compare the average WCFRs at different time-points.One-way ANOVA and post-hoc multiple comparison tests revealed significant differences in WCFR among 3 GICs at five time-points (p0.05). The post-hoc multiple comparison test revealed Bandtite cement had consistently higher WCFR at all time-points, compared to Granitec and Ariadent cements (p0.05). The one-way ANOVA test revealed significant differences in WCFR at different time-points for all GIC groups (p0.05). The day 8 exhibited the highest WCFR for all GICs. The repeated measures ANOVA test revealed significant differences in WCFR at different time-points for all GIC groups (p0.0005). Reviewing average WCFR on days 22 and 29, these values for Granitec, Bandtite and Ariadent GICs, were higher (p0.05), not different (p0.05), and significantly lower (p0.05) than the day 1 values, respectively.Bandtite followed by Granitec showed higher WCFR compared to Ariadent.
