Örümceklerin ağ yapma özelliği çok eski yıllardan beri insanoğlunu etkilemiştir. Ancak bütün örümcekler ağ örmezler. Örümcekler ağ örme yeteneklerini örü aygıtlarına sahip olmalarına borçludur. Örü aygıtı ipek salgısını üreten ağ bezleri ve bu bezlere bağlı örü memelerinden oluşmaktadır. Örü memeleri, ipeğin çıkartılması ve iplik haline getirilerek işlenmesi için kullanılmaktadır. Örümceklerin ağları çok dikkat çekse de örü aygıtları hakkında çok fazla çalışma bulunmamaktadır. Bu çalışmada, oldukça dikkat çekici ağ yapısına sahip olan Agelena orientalis (C.L. Koch, 1837) huni ağ örümceğinin ağ yapısı ve ağ örme aygıtının morfolojisi taramalı elektron mikroskobu (SEM) ile çalışılmıştır. Çalışmamızda, A. orientalis’in ağ yapısı, örü memeleri ve spigotların yapısı gösterilmiş ve elde edilen veriler tartışılmıştır.

GİRİŞ ve AMAÇ: Akrep zehirleri çeşitli kimyasal maddelerin oluşturduğu oldukça kompleks heterojen karışımlardır. Bu çalışmada, bir akrep türü Mesobuthus gibbosus (Brullé, 1832) (Scorpiones: Buthidae)’un ham zehrinin kanser hücre hatları üzerine sitotoksik ve antiproliferatif etkileri araştırılmıştır. YÖNTEM ve GEREÇLER: M. gibbosus’dan elde edilen ham zehrin sitotoksik etkileri iki insan kanser hücre hattı (MCF-7 meme kanseri ve A549 akciğer karsinomu) ve normal fare fibroblast (L929) hücreleri üzerindeki sitotoksik ve antiproliferatif etkileri çalışılmış ve elde edilen veriler literatür ışığında karşılaştırılmıştır. BULGULAR: Çalışmada, akrep ham zehrinin MCF-7 kanser ve L929 fibroblast hücreleri üzerine düşük dozlarda bile sitotoksik etkisi ve hücre proliferasyonunu inhibe ettiği gözlenmiştir. Aynı yöntemlerle A549 akciğer karsinomu hücreleri üzerine zehrin sitotoksik ve antiproliferatif etkisi nispeten zayıf olarak gözlemlenmiştir. TARTIŞMA ve SONUÇ: M. gibbosus akrep ham zehrinin kanser hücreleri üzerine etkileri gözlenmiştir ancak bu kanser hücrelerinin proliferasyonunu inhibe eden zehir bileşen veya bileşenlerinin belirlenmesi için ileri çalışmalara ihtiyaç vardır.

Akrepler, Arachnida sınıfında yer alan zehirli arthropodlardan olup, bilinen en eski karasal eklembacaklılardır ve yaşayan fosiller olarak da tanımlanmaktadırlar. Akrepler kendilerine özgü bir takım özelliklere, özel yapılara sahip olmaları ve zehirlenme vakalarına sebep olmaları nedeniyle çeşitli araştırmalara konu olmuştur. Ancak, akreplere özgü bir duyu organı olan ve tarak organı olarak da bilinen pektin organın yapısı hakkında çok fazla bilgi bulunmamaktadır. Bu çalışmada, Protoiurus kraepelini (von Ubisch, 1922) akrebinin tarak organ yapısı ışık mikroskobu ve taramalı elektron mikroskop (SEM) kullanarak çalışılmış, detaylı morfolojik ve histolojik özellikleri belirlenmiştir. Örnekler Eğirdir (Isparta, Türkiye)’de yapılan arazi çalışmalarında toplanamıştır. P. kraepelini’nin tarak organı diğer akreplerde olduğu gibi bir çift olarak mesosomal ikinci segmentin ventrolateralinde yerleşmiş olup genel mimariye uygun olarak marjinal lamella, median lamella ve pektinal dişler olmak üzere üç kısımdan oluşmaktadır. Aynı zamanda bu yapılar üzerinde yer alan duyusal kılları, pektinal dişler üzerindeki peg sensillanın morfolojileri incelenmiştir. Bu çalışmada, ilk kez P. kraepelini’nin tarak organının histolojisi ortaya konulmuş ve yapı - fonksiyon arasındaki bağlantı uyarınca olası fonksiyonları tartışılmıştır. Scorpions are the oldest known terrestrial arthropods of the Arachnida class and are also known as living fossils. Scorpions have been subjected to various researches because they have special properties and unique structure, and they cause cases of poisoning. However, there is not much information about the structure of the pectine organ, a specific sense organ of the scorpion, which is also called sensory comb. In this study, the comb organ structure of Protoiurus kraepelini (von Ubisch, 1922) was studied by using light microscopy and scanning electron microscope (SEM) and detailed morphological and histological features were determined. Some of the comb organs of the P. kraepelini scorpions collected in the field studies from Eğirdir (Isparta, Turkey) were prepared for SEM by using routine methods. A portion of the comb organs were embedded in parafine and sections were taken and stained sections were stained with hematoxylin-eosin and histological features were examined in light microscope. The comb organ of P. kraepelini is located in the ventrolateral part of the mesosomal second segment as in other scorpions and consists of three parts as marginal lamella, median lamella and pectinal teeth in accordance with the general architecture. At the same time, sensory hairs on these structures and peg sensilla on pectinal teeth were examined. In this study, the histology of the comb organ of P. kraepelini was first time demonstrated and its possible functions were discussed in the relationship between structure and function.

Pyza taurica Gruber, 1979 (Opilionida, Nemastomatidae) otbiçen türü Anadolu’ya özgüdür. Bu çalışmada, P. taurica'nın dış morfolojisi ayrıntılı olarak taramalı elektron mikroskopu (SEM) kullanılarak araştırılmıştır. Özellikle taksonomik ve sistematik önem taşıyan karakterde, bacaklar, pedipalp, chelicerae ve P. taurica erkek örneklerinin dorsal üzerindeki kesici yapılar ve duyu organları incelenmekte ve tasvir edilmektedir. Bu gibi çalışmalar, özellikle otbiçenlerin fonksiyonel anatomisini ve morfolojisini anlamak için gereklidir.

The harvestmen species Pyza tauricaGruber, 1979 (Opilionida, Nemastomatidae) is endemic to Anatolia. In this study,external morphology of P. taurica was investigated in detail by using scanning electronmicroscope (SEM). Cuticular structures and sense organs on the segments oflegs, pedipalps, chelicerae, and the dorsal integument of male specimens of P.taurica are investigated and depicted, in particular, characters oftaxonomic and systematic importance. Such studies are needed especially forunderstanding the functional anatomy and morphology of opilionids., Pyza taurica Gruber,1979 (Opilionida, Nemastomatidae) otbiçen türü Anadolu’ya özgüdür. Bu çalışmada, P. taurica'nındış morfolojisi ayrıntılı olarak taramalı elektron mikroskopu (SEM)kullanılarak araştırılmıştır. WOS:000592998800004 Pseuodoscorpions (Pseudoscorpiones) have been little studied, although they are the third largest group in the Arachnida in number of species. Pseudoscorpions have features resembling those of true scorpions. Due to their small size (the largest pseudoscorpion being no larger than 1 cm) systematic studies have proven difficult. A high-performance microscope is necessary for detailed studies. In the present study. the pseudoscorpion Atemnuspolitus (Simon, 1878) collected in Osmaniye province. Turkey. was used to describe systematic and taxonomic characters, including chelicerae, pedipalps, walking legs. and cuticular structures on appendages using stereo light microscopy and scanning electron microscopy (SEM). The study revealed characteristic features of this species. This is the first SEM study of A. politus and provides an important contribution to understanding the morphology of pseudoscorpions and demonstrates the taxonomic value of SEM micrographs. Probable functions of some of the structures observed are discussed. Antimicrobial activity of the methanolic extracts of leaves and flowers of D. lamarckii Ivan, (Scophulariaceae), an endemic plant species of Turkey, was tested on ten bacterial and four yeast strains. Effective antibacterial activity was observed in four bacterial strains. Minimum inhibitory concentration (MIC) was calculated by use of liquid culture tests and in all the four effective bacterial strains, the MIC was found to beor = 199.5 mg/ml. The minimum bactericidal concentration (MBC) of B. subtilis, S. aureus, and L. monocytogenes was calculated to beor = 199.5 mg/ml, and MBC value for Shigella was calculated asor = 399 mg/ml.

Antibiyotiklere dirençli mikroorganizmaların sayısı her geçen gün artığından dolayı, tarımda ve tıpta bu dirençli mikroorganizmalarla mücadele etmek için yeni ve etkili antimikrobiyal ajanlara ihtiyaç duyulmaktadır. Antibiyotikler için alternatif bir kaynak olarak düşünülen Agelena labyrinthica (Clerck, 1757) (Araneae: Agelenidae)’nın hemolenfi 10 bakteri suşuna karşı test edilmiş ve 10 suştan 5 tanesi hemolenfe duyarlı olarak bulunmuştur. Duyarlı bakteri suşları: Bacillus subtilis, Pseudomonas aeruginosa, Shigella sp., Escherichia coli, Staphylococcus aureus’dur. Sonuçlar, örümcek hemolenfinin antibakteriyal etkiye sahip olduğunu göstermiştir. Bu çalışma A. labyrinthica hemolenfinin antibakteriyal etkisi üzerine yapılan ilk çalışmadır. Since the number of microorganisms which are resistant to antibiotics have been increasing steadily, the need for treating these pathogens requires novel and efficacious antimicrobial agents, both in medicine and in agriculture. In our study, the hemolymph of Agelena labyrinthica (Clerck, 1757) (Araneae: Agelenidae) which is considered as an alternative resource for antibiotics, was tested against ten bacterial strains, and it was found that five out of ten strains were sensitive to hemolymph. The sensitive bacterial strains were Bacillus subtilis, Pseudomonas aeruginosa, Shigella sp., Escherichia coli, Staphylococcus aureus. Our results showed that spider hemolymph had antibacterial effects. This is the first study about antibacterial effects of A. labyrinthica‘s hemolymph.

The antimicrobial activities of ethyl acetate and methanol extracts of Satureja hortensis L. were studied. These extracts were tested in vitro against 10 bacterial species, 4 yeasts species and strains by the disc diffusion method. The results indicated that the methanol extract of S. hortensis is more effective against tested microorganisms than ethyl acetate. While ethyl acetate extract showed inhibition effect against Pseudomonas aeruginosa (ATCC 27853) and Bacillus subtilis (RSHI), methanol extract affected Bacillus subtilis (RSHI), Escherichia coli (ATCC 25922), Shigella (RSHI), Staphylococcus aureus (ATCC 29213) and Pseudomonas aeruginosa (ATCC 27853). The cells of microorganisms treated with plant extracts and normal microorganism cells were observed by scanning electron microscope. It was apparent that cells after treated with S. hortensis are damaged.

The objective of the present study is to describe the functional morphology of venom apparatus of Euscorpius mingrelicus (Kessler, 1874) by using light microscope and scanning electron microscope (SEM). The venom apparatus, situates in the last segment of metasoma (telson), is composed of a pair of venom glands and sting. Telson is covered by cuticular exoskeleton as well as all body, and there are cuticular setae and pits on it as serve sensory organ. The general organization of the venom apparatus of E. mingrelicus is similar to the other scorpions' venom apparatus. The venom glands fill in the telson and are a pair and are equal size. Each venom gland is surrounded by gross striated muscular bundles. The venom is produced in the venom glands, carried by venom ducts passing through the sting and exiting from the venom pores during the muscular contractions. The venom pores situate both side of the tip of sting and are used for injecting venom. The venom produces left venom gland is carried by left venom duct and exited from left venom pore as well as right. In the transverse section of the sting, both venom ducts can be easily seen by light microscope.

Örümcek zehirleri; hem avları olan böceklerde hem de memelilerde hayatsal fizyolojik fonksiyonları etkileyen biyo-aktif maddeler olup esas bileşenleri; protein, polipeptid, poliamin nörotoksinler, enzimler, nükleik asitler, serbest amino asitler, monoaminler ve inorganik tuzlardır. Örümcek zehirinde bulunan proteinlerin tanımlanması, zehirin tanımlanmasında önemli bir adım teşkil etmektedir. Bu çalışmada; ülkemizde oldukça yaygın bir örümcek türü olan Agelena labyrinthica'nm uyanlmamış ve uyarılmış durumdaki zehir bezlerinden elde edilen protein profilleri saf zehir salgısı ile karşılaştırılmıştır. Uyanlmamış zehir bezi ekstraktı ve elektriksel uyarımla elde edilmiş saf zehirin protein profilleri, SDS-PAGE analitik yöntemi kullanılarak karşılaştırıldığında, zehir proteininin yapısal proteinlerden kesin olarak ayrıldığı ve saf zehirde molekül ağırlıkları bakımından yedi farklı polipeptidin bulunduğu belirlenmiştir. Spider venoms are biologically active substances which affect a variety of vital physiological functions in both insects and mammals. The major constituents of spider venoms are protein, polypeptide and polyamine neurotoxins, enzymes, nucleic acids, free amino acids, monoamines and inorganic salts. The identification of the proteins in spider venoms is an essential step for identification of venom. In this study, we compared with protein profiles of Agelena labyrinthica venom obtained by electrostimulation of the prosoma and extracted directly from gland. There were seven components identified in whole venom, when whole venom and venom gland extracts composition of A labyrinthica were compared by SDS-PAGE.

Bu çalışmada, ağ örücü örümcek olan Agelena orientalis C.L. Koch, 1837, Uloborus walckenaerius Latreille 1806, Argiope lobato (Pallas,1772), Argiope bruennichi'n (Scopoli, 1772) ağ yapısı ve ağ örme aygıtının yapısal organizasyonu, taramalı elektron mikroskobu (SEM) kullanılarak gözlemlenmiştir. Örümceklerin örü aygıtı, ipek salgısını üreten ağ bezleri, bu salgıyı örü memeleri taşıyan kanallar, örü memeleri ve örü alanında bulunan spigotlardan oluşmaktadır. Çalışmamızda, dört farklı örümcek türünün ağ yapıları, örü memeleri özellikle posterior örü memeleri ve spigotların düzenlenmesi gösterilmiş ve elde edilen veriler literatür ışığında tartışılmıştır. Spiders are creatures that are best adapted to terrestrial life after insects in invertebrates and are represented by approximately 48,000 species in the world. The webs of spiders are literally a work of art. But all spiders do not weave a web. The main reason for spider web weaving is hunting. Each of the spiders has different silk glands. Spiders therefore provide the formation of different webs and silk fibers. Some spider species live in the nature dependent on the own web, while others continue to live without being dependent on the own web. The silk glands in the spiders are divided into six groups as ampullate, tubuliform, flagelliform, aggregate, aciniform and pyriform glands and different types of silk are secreted from each gland. Although basic taxonomic features generally remain unchanged, some spider-silk devices may undergo adaptive variations. In this study, the web structure of the weaving web spider, Agelena orientalis C.L. Koch, 1837, Uloborus walckenaerius Latreille 1806, Argiope lobato (Pallas,1772), Argiope bruennichi (Scopoli, 1772) and the structural organization of the web weaving apparatus was observed by using scanning electron microscopy (SEM). The web weaving apparatus of the spiders consists of the silk glands which produce silk, the ducts that carry this silk secretion to spinnerets, spinnerets and spigots on the weaving surfaces. In our study, the webs structures of four different species of spiders, especially the arrangement of the posterior spinnerets and spigots are shown and discussed in the light of the literature. YÖK Tez ID: 515785 Çalışmada, beyin tümörünün farklı tiplerindeki GSTP1 ve GSTM1 izoenzimlerinin ekspresyonları üzerinde inceleme yapılıp günümüzdeki literatürlerle kıyaslaması yapıldı. 2016-2017 yılları arasındaki beyin tümörlü 55 hastada immunohistokimyasal metod kullanılarak, GSTP1 ve GSTM1 izozimlerin ekspresyonları çalışıldı. Demografik parametreler de dikkate alınarak izoenzimlerin ekspresyon sonuçlarıyla karşılaştırıldı. Beyin tümörlü hastaların yaş ortalaması 46.72 olup 29 hasta bayanlardan, 26 hasta erkeklerden oluşturuldu.55 beyin tümörlü hastada 57 beyin tümör çeşidine ulaşıldı. Bunlar arasında 12 olguda meningioma,12 olguda metastaz,9 hastada glioblastioma, 5 örnekte hipofiz adenomu tanısı bulundu. Hipofiz bezi adenom dokularda GSTP1 ve GSTM1 ekspresyon miktarının yüksek olduğu gözlendi. Buna karşılık glioblastoma dokularında GSTP1 ekspresyon miktarı az olduğu belirlendi.Hipofiz bezindeki tümörlü dokuların, diğer tümörlü dokulara oranla ilaç metabolizma kapasitesinin (direncinin) yüksek olduğu bulundu.. Bu bulguların ilaç tedavisini güçleştidiği düşünüldü The aim of the present study is to show the expression of GSTP1 and GSTM1 in different types of brain tumors and to compare the results with current literature. The expression of GSTP1 and GSTM1 was analyzed using immunostaining in 55 patient with intracranial tumors between 2016 and 2017. The demograhic features of the patients were documented and the results of expressions were compared. The mean age of the patients was 46.72 years and 29 patients were female, 26 were male. Fiftyseven specimens were obtanied from 55 patients. Among them, meningioma was diagnosed in 12, metastases in 12, glioblastoma in 9 and pituitary adenoma in 5 samples. The highest GSTP1 and GSTM1 expressions were observed in pituitary adenomas, while the least GSTP1 expression was detected in glioblastoma. Various dilutions of crude venom obtained from scorpion by means of electrostimulation technique were tested on two type cancer cells (MCF-7 breast cancer and A549 lung carcinoma cell line) and L929 fibroblast cells, and then anti-proliferative effects on these cell lines were examined. At the end of the present study, it was determined that mcf-7 and fibroblast cells were more effective at low dilutions of venom doses than A549. It was observed that the amount of dilution decreased the venom effect on the cells. Although M. gibbossus scorpion venom has been tried on cancer cell lines by using different methods, the antiproliferative effect of this study was first time tried to be demonstrated by using real-time cell analysis system.

Dilüsyon miktarı artıkça zehir etkisinin azaldığı ve h1299 hücreleri üzerinde apoptoz ve nekroz oranlarının düştüğü, hücre proliferasyonuna etki yapmadığı gözlenmiştir. mcf-7 hücrelerine bakıldığında apoptoz ve nekroz oranların birbirine yakın olduğu, h1299 hücrelerinde apoptoz oranının nekroz oranından iki kat fazla olduğu tespit edilmiştir. E. mingrelicus akrep zehri her ne kadar mcf-7 hücreleri üzerine kısmen etkili bulunsa da kesin bir şey söyleyebilmek için daha detaylı ileri çalışmalara ihtiyaç vardır. Bazı özellikleri bakımından gerçek akreplere benzeyen yalancı akrepler oldukça ilginç canlılar olup, boyu en fazla 1cm kadardır. Yalancı akrepler küçük yapılı olduklarından sistematik olarak çalışılmaları zordur. Bu canlıların çalışılması için yüksek performanslı mikroskoplara ihtiyaç duyulmaktadır. Bu çalışmada, Atemnus politus (Simon, 1878)' un tanımlanmasında kullanılabilecek önemli sistematik ve taksonomik karakterler stereo ışık mikroskobu ve taramalı elektron mikroskop (SEM) kullanılarak çalışılmıştır. Özellikle A. politus' un keliser, pedipalp, yürüme bacakları ve bunların üzerindeki kutikular yapılar SEM kullanılarak ilk defa detaylı bir şekilde incelenmiştir. Yalancı akreplerin morfolojisine önemli katkılar sağlayacağı düşünülen bu çalışmada, bazı incelenen yapıların muhtemel fonksiyonları da tartışılmıştır. Pseudoscorpions (Pseudoscorpiones) are the third largest group in the Arachnida class in terms of number of species. Pseudoscorpions that look like real scorpions in terms of some features are quite interesting creatures, and the biggest body is not bigger than 1cm. Due to the small bodies of pseudosorpions, there are difficulties in systematic studies. To study these creatures is needed for high performance microscope. In this study, the some important systematic and taxonomic characters that could be used to describe Atemnus politus (Simon, 1878) were studied using stereo light microscopy and scanning electron microscopy (SEM). In particular, the chelicerea, pedipalp, walking legs of A. politus and the cututicular structures on their appendages are investigated the first time by using SEM in the present study. This study, which is thought to provide important contributions to the morphology of pseudoscorpions, has discussed the probable functions of the some structures studied.

Yapılan çalışmalar sonucunda bu 2 familyaya ait 3 cins ve 5 tür tespit edilmiştir. İncelenen türler; Gylippus (Paragylippus) monoceros Werner, Gylippus (Gylippus) syriacus (Simon), Gylippus (Hemigylippus) bayrami sp. n., Eusimonia nigrescens Kraepelin ve Karschia (Karschia) mastigofera (Birula)'dır. Tanımlanan türlerin sistematik, morfolojik, etolojik ve ekolojik özellikleri verilmiş ve bu taksonlarla ilgili yorumlar yapılmıştır. Türlerin keliser, pedipalp, yürüme bacakları gibi diagnostik yapılarının taksonomik özellikleri SEM (Taramalı Elektron Mikroskobu) kulanılarak ortaya konmuş ve türlerin redeskripsiyonları yapılmıştır. Ayrıca Gylippus (Hemigylippus) bayrami sp. n. türü bilim dünyası için yeni bir türdür. Ayrıca Gylippus (Hemigylippus) Birula alt cinsi ülkemiz için yeni kayıttır. In this study, collected from the various regions of Turkey between the 2010 and 2013 years and with the museum specimens, a total of 111 specimens were examined belong to Gylippidae Roewer and Karschiidae Kraepelin families. As a result of, 3 genera and 5 species have been identified belong to these 2 families. Examined species are Gylippus (Paragylippus) monoceros Werner, Gylippus (Gylippus) syriacus (Simon), Gylippus (Hemigylippus) bayrami sp. n. , Eusimonia nigrescens Kraepelin and Karschia (Karschia) mastigofera (Birula). Detailed systematical, morphological, ethological and ecological characteristics of diagnosed species and remarks about these taxa were given. The taxonomic details of the diagnostic structures (Chelicerae, pedipalps, walking legs etc.) of the species were presented using SEM (Scanning Electron Microscope) and species were redescribed. Gylippus (Hemigylippus) bayrami sp. n. is a new species for the world of science. Also, the subgenus Gylippus (Hemigylippus) Birula is new record for our country.
