2. The phytochemistry of Eastern European forest species

Abstract

This study was a part of the ForestSpecs FP7 EU project. The aim of this project was to utilize the readily available, large volume and low price wood residues from the wood processing industry. The first part of this work involved a comparative study of three extraction techniques, shaking, soxhlet and microwave assisted extraction. The microwave extraction was found to be the most suitable method, as it allowed the results to be achieved with reduced level of solvents, lower energy consumption and faster processing time. The phytochemical investigation of six forestry species: Pinus sylvestris, Larix gmelinii, L. sibirica, L. sukaczewii, L. decidua, and Populus tremula was performed. This included many steps such as grinding, extraction of plant materials and separation, isolation and purification of a variety of compounds with potential use in pharmaceutical, cosmetic and also pest management applications. A total of ninety-three compounds were identified, with thirty-two being isolated from more than one source. Of these, four have not been described previously and are shown below. Compound 13 was isolated from L. gmelinii and compounds 32, 33 and 37 were isolated from L. sukaczewii. Components from the species investigated have been identified using 1D, 2D NMR and IR spectroscopy, mass spectrometry and optical rotation measurements. The extracts and purified compounds were submitted to the consortium screening programme. The thirteen bark extracts of the species investigated were tested for cytotoxic and apoptotic activity using the LDH and MTT assays, as well as Western blot analysis of protein p53. In both, A-375 human melanoma cells and CV1-P fibroblast cells, the following bark extracts showed selective cytotoxic effects against A-375 cancerous cells: L. gmelinii DCM (LGD) and P. sylvestris DCM (PSD), EtOAc (PSE) and MeOH (PSM) extracts. Further investigation demonstrated that L. gmelinii DCM and EtOAc and P. sylvestris DCM extracts induced the translocation of the p53 protein into the nucleus. This approach proved the significance of the use of natural products as chemopreventive agents and cancer therapeutics.

6. Effect of water chemistry on the planktonic communities and relationships among food web components across a freshwater ecotone

Abstract

Most ecological research on the food web has been focused more on the pelagic zone than on the transitional zone - ecotones between lentic and lotic habitats. The specific goals of this study were to determine whether the contact zone of waters differs in hydrochemical and biological terms from the waters of the canal and the open water zone, and to evaluate the influence of particular macro-habitats on the interactions between components of the planktonic food web. The distribution of samples in ordination space led us to conclude that the studied habitats are distributed along the rising gradient of total organic carbon and nutrients. Assemblages of all investigated groups showed a strong compositional gradient correlated with conductivity and total phosphorus, while a second strong gradient in species composition was explained by nitrate nitrogen and/or phosphate concentrations. The analysis of trophic relationships in the system bacteriaciliates- crustaceans reveals a clear differentiation and strength of mutual relations between the analyzed zones. The highest number of significant correlations was determined in the contact zone. It can also be a place of very efficient matter and energy flow in freshwater ecosystems.

Abstract

Marine organisms and their metabolites represent a unique source of potential pharmaceutical substances. In this study, we examined marine-derived substances for their bioactive properties in a cell-based Chikungunya virus (CHIKV) replicon model and for in vitro anti-inflammatory activity. In the screening of a marine sample library, crude extracts from the Indian soft coral, Sinularia kavarattiensis, showed promising activity against the CHIKV replicon. Bioassay-guided chemical fractionation of S. kavarattiensis resulted in the isolation of six known norcembranoids (1–6) and one new compound, named kavaranolide (7). The structures were elucidated on the basis of NMR and MS spectroscopic data. Compounds 1–3 and 5–7 were evaluated for their replicon-inhibiting potential in the CHIKV model by using a luminescence-based detection technique and live cell imaging. Compounds 1 and 2 showed moderate inhibition of the CHIKV replicon, but imaging studies also revealed cytotoxic properties. Moreover, the effects of the isolated compounds on primary microglial cells, an experimental model for neuroinflammation, were evaluated. Compound 2 was shown to modulate the immune response in microglial cells and to possess potential anti-inflammatory properties by dose-dependently reducing the release of pro- and anti-inflammatory cytokines.

24. Diel Horizontal Distribution of Psammonic Ciliates and Rotifers in Shallow Eutrophic Lake.

Abstract

The aim of this study was to examine the diel dynamics of the qualitative and quantitative structures of psammonic ciliates and rotifers in a shallow eutrophic lake. Furthermore, the effects of specific physical and chemical factors of water on the occurrence of microorganisms were analyzed. There were clear differences in the qualitative and quantitative structure of organisms between the examined micro-habitats in a diurnal cycle. The highest density of ciliates, irrespective of the time of day, was recorded in the higroarenal zone, while the highest density of rotifers was found in both the higro- and hydroarenal zones. The lowest diversity of ciliates was recorded in the hydroarenal zone. RDA analysis showed a clear contrast between the eupsammonic, higropsammonic, and hydropsammonic zones, regardless of the time of day. A direct impact on the occurrence of protozoa and small Metazoa was played by the content of total phosphorus (Ptot) and TOC in the water. In addition, the ciliates were significantly affected by P-PO4 and N-NO3, and also the pH affected the group of rotifers. [ABSTRACT FROM AUTHOR]
