WRKY Transcription factors (TFs) play critical roles in plant defence mechanisms that are activated in response to biotic and abiotic stresses. However, information on the Glycine soja WRKYs (GsoWRKYs) is scarce. Owing to its importance in soybean breeding, here we identified putative WRKY TFs in wild soybean, and compared the results with Glycine max WRKYs (GmaWRKYs) by phylogenetic, conserved motif, and duplication analyses. Moreover, we explored the expression trends of WRKYs in G. max (oomycete, fungi, virus, bacteria, and soybean cyst nematode) and G. soja (soybean cyst nematode), and identified commonly expressed WRKYs and their co-expressed genes. We identified, 181 and 180 putative WRKYs in G. max and G. soja, respectively. Though the number of WRKYs in both studied species is almost the same, they differ in many ways, i.e., the number of WRKYs on corresponding chromosomes, conserved domain structures, WRKYGQK motif variants, and zinc-finger motifs. WRKYs in both species grouped in three major clads, i.e., I-III, where group-II had sub-clads IIa-IIe. We found that GsoWRKYs expanded mostly through segmental duplication. A large number of WRKYs were expressed in response to biotic stresses, i.e., Phakospora pachyrhizi, Phytoplasma, Heterodera glycines, Macrophomina phaseolina, and Soybean mosaic virus; 56 GmaWRKYs were commonly expressed in soybean plants infected with these diseases. In recent years, numerous single nucleotide polymorphism (SNP) panel methods to genotype non-invasive faecal samples have been developed. However, none of these existing methods fit all of the criteria necessary to make a SNP panel broadly usable for conservation projects in any country-cost effective, streamlined lab protocol and user-friendly open-source bioinformatics protocols for panel design and analysis. Here, we present such a method and display its utility by developing a multiplex PCR SNP panel for conducting individual ID of snow leopards, Panthera uncia, from faecal samples. The SNP panel we present consists of 144 SNPs and utilises next-generation sequencing technology. We validate our SNP panel with paired tissue and faecal samples from zoo individuals, showing a minimum of 96.7% accuracy in allele calls per run. We then generate SNP data from 235 field-collected faecal samples from across Pakistan to show that the panel can reliably identify individuals from low-quality faecal samples of unknown age and is robust to contamination. We also show that our SNP panel has the capability to identify first-order relatives among sampled zoo individuals and provides insights into the geographic origin of samples. This SNP panel will empower the snow leopard research community in their efforts to assess local and global snow leopard population sizes. Escalating SOx and NOx emissions from industrial plants necessitates customized scrubbing solutions to improve removal efficiency and tackle cost limitations in existing wet FGD units. This work investigates the real-time intensified removal pathways via an innovative two-stage countercurrent spray tower configuration strategically integrating NaOH (M a ) and NaOH/NaClO (M a /M b ) to remove SOx and NOx emissions simultaneously from the industrial stack through a comprehensive parametric study of absorbents concentration, reaction temperature, gas flow rate, liquid to gas ratio (F L /F G ), and absorbent showering head. Flue gas stream comprising SO 2 bearing 4500 ppm, SO 3 bearing 300 ppm, 70 ppm NO, and 50 ppm NO 2 brought into contact with two scrubbing solutions as M a , and a complex absorbent of M a /M b at varying respective ratios. Ninety-two percent SOx emissions were removed using 5% NaOH with double-stage scrubbing, while NO x removal was observed below 50%. Adding NaClO facilitates additional "free radical (ClO - )" chemical pathways for gases to react and decompose into ionic forms for easier solubilization so as to significantly enhance the removal capacities for both SOx and NOx compounds. NaClO oxidizer, along with NaOH, boosted the respective removal efficiencies of SO x to 99.6% and 92% NO x , proving complementary media integration advantages arising from staged exposure and bubbly interphase mass transfer phenomena. The customized synergistic effect of M a and M b promoted the development of an additional free radical oxidation route while sustaining the solubilization of SOx/NOx in caustic, driving toward fractional detoxification. A comparative metabolomic study of three varieties of wild Rosa ( Rosa acicularis , Rosa amblyotis , and Rosa rugosa ) from a Kamchatka expedition (2024) was conducted via extraction with supercritical carbon dioxide modified with ethanol (EtOH), and detection of bioactive compounds was realized via tandem mass spectrometry. Several experimental conditions were investigated in the pressure range 50-350 bar, with the used volume of co-solvent ethanol in the amount of 2% in the liquid phase at a temperature in the range of 31-70 °C. The most effective extraction conditions are the following: pressure 200 Bar and temperature 55 °C for Rosa acicularis ; pressure 250 Bar and temperature 60 °C for Rosa amblyotis ; pressure 200 Bar and temperature 60 °C for Rosa rugosa . Three varieties of wild Rosa contain various phenolic compounds and compounds of other chemical groups with valuable biological activity. Tandem mass spectrometry (HPLC-ESI-ion trap) was applied to detect the target analytes. A total of 283 bioactive compounds (two hundred seventeen compounds from the polyphenol group and sixty-six compounds from other chemical groups) were tentatively identified in extracts from berries of wild Rosa . For the first time, forty-eight chemical constituents from the polyphenol group (15 flavones, 14 flavonols, 4 flavan-3-ols, 3 flavanones, 1 phenylpropanoid, 2 gallotannins, 1 ellagitannin, 4 phenolic acids, 1 dihydrochalcone, and 3 coumarins) were identified in supercritical extracts of R. acicularis , R. amblyotis , and R. rugosa .

Ethylene, as an important chemical raw material, could be produced through the coal-based acetylene hydrogenation route. Nickel-based catalysts demonstrate significant activity in the semihydrogenation reaction of acetylene, but they encounter challenges related to catalyst deactivation and overhydrogenation. Herein, the effect of Sn promoter on Ni/CeO 2 catalysts has been comprehensively explored for acetylene semihydrogenation. The optimized Ni/8%Sn-CeO 2 catalytic performance was significantly improved, with 100% acetylene conversion and 82.5% ethylene selectivity at 250 °C, and the catalyst maintained high catalyst performance within a 1000 min stability test. A series of characterization tests show that CeO 2 modified by moderate Sn 4+ doping is more conducive to modulating the charge structure and geometry of the Ni active center. Additionally, the in situ Diffuse Reflectance Infrared Fourier Transform Spectroscopy and density functional theory results indicated that catalysts doped with Sn 4+ facilitated more efficient desorption of ethylene from the catalyst surface compared to Ni/CeO 2 catalysts, thus improving ethylene selectivity and yield. This study highlights an effective strategy for improving the catalytic performance of rare-earth-based catalysts through the incorporation of effective metal promoters.

Embracing a circular economy in the textile industry represents a crucial step toward sustainability, where fashion and textile sectors contribute significantly to CO 2 emissions. However, transitioning from a linear "take-make-waste" model to circularity, poses multifaceted challenges, that highlight the staggering volume of annual textile waste surpassing industry predictions, thus emphasizing the urgent need for comprehensive strategies. Despite advancements in recycling technologies, challenges persist in collecting and sorting textile waste, where fragmentation in waste management and recycling processes hinders effective management of post-consumer waste. Addressing these challenges demands elevated efforts in collection, sorting, and pre-processing, alongside regulatory interventions to drive enhanced waste collection and circular business models. Efforts are underway to promote sustainable textile recycling, with initiatives like the EU's Sustainable and Circular Textiles Strategy aiming to reduce reliance on virgin resources. However, achieving a circular textile market in the near future requires collaborative action and innovative solutions. Though challenges in scaling and technological limitations still remain, recent breakthroughs in textile-recycling technologies offer promise, signaling a shift toward scalable and sustainable alternatives to virgin fibers, where bio-based chemical processes, and thermochemical recycling processes present transformative opportunities. Solid supported catalysts have several synthetic applications. Herein, finely ground eggshells were used as a solid support for the preparation of transition metal (Ni, Zn, Cu, Sn and Co) based catalysts to synthesize 2,4-dinitrophenylhydrazone (3) and dihydropyrimidinones (7 and 8). The effect of catalyst load, time and temperature on product yield was studied. Box Behnken Model was employed, and three predictors named catalyst amount (A), reaction time (B), and reaction temperature (C) were used to find the correlation of the predictors with the yield. Second order polynomial equation was used to estimate the effects of these factors. According to the statistical model, about 12% increase in yield was observed as a result of one-unit increase in reaction time while all other terms were kept constant. The values of S (18.1616) and R 2 (71.2%) indicate that the statistical model gave an adequate fit to data. Quadratic model for the response surface was used for the analysis of variance (ANOVA) results, the larger F-values, and smaller p-values indicated that the predictors are in good agreement. With the development of new technologies in recent years, researchers have made significant progress in crop breeding. Modern breeding differs from traditional breeding because of great changes in technical means and breeding concepts. Whereas traditional breeding initially focused on high yields, modern breeding focuses on breeding orientations based on different crops' audiences or by-products. The process of modern breeding starts from the creation of material populations, which can be constructed by natural mutagenesis, chemical mutagenesis, physical mutagenesis transfer DNA (T-DNA), Tos17 (endogenous retrotransposon), etc. Then, gene function can be mined through QTL mapping, Bulked-segregant analysis (BSA), Genome-wide association studies (GWASs), RNA interference (RNAi), and gene editing. Then, at the transcriptional, post-transcriptional, and translational levels, the functions of genes are described in terms of post-translational aspects. This article mainly discusses the application of the above modern scientific and technological methods of breeding and the advantages and limitations of crop breeding and diversity. In particular, the development of gene editing technology has contributed to modern breeding research.

Bioinformatics has become an interdisciplinary subject due to its universal role in molecular biology research. The current status of Russia's bioinformatics research in Russia is not known. Here, we review the history of bioinformatics in Russia, present the current landscape, and highlight future directions and challenges. Bioinformatics research in Russia is driven by four major industries: information technology, pharmaceuticals, biotechnology, and agriculture. Over the past three decades, despite a delayed start, the field has gained momentum, especially in protein and nucleic acid research. Dedicated and shared centers for genomics, proteomics, and bioinformatics are active in different regions of Russia. Present-day bioinformatics in Russia is characterized by research issues related to genetics, metagenomics, OMICs, medical informatics, computational biology, environmental informatics, and structural bioinformatics. Notable developments are in the fields of software (tools, algorithms, and pipelines), use of high computation power (e.g. by the Siberian Supercomputer Center), and large-scale sequencing projects (the sequencing of 100 000 human genomes). Government funding is increasing, policies are being changed, and a National Genomic Information Database is being established. An increased focus on eukaryotic genome sequencing, the development of a common place for developers and researchers to share tools and data, and the use of biological modeling, machine learning, and biostatistics are key areas for future focus. Universities and research institutes have started to implement bioinformatics modules. This study presents the metabolomic profiles of the four Ribes species ( Ribes pauciflorum Turcz., Ribes triste Pall., Ribes dicuscha Fisch., and Ribes aureum Purch.). The plant material was collected during two expeditions in the Russian Far East. Tandem mass spectrometry was used to detect target analytes. A total of 205 bioactive compounds (155 compounds from polyphenol group and 50 compounds from other chemical groups) were tentatively identified from the berries and extracts of the four Ribes species. For the first time, 29 chemical constituents from the polyphenol group were tentatively identified in the genus Ribes . The newly identified polyphenols include flavones, flavonols, flavan-3-ols, lignans, coumarins, stilbenes, and others. The other newly detected compounds in Ribes species are the naphthoquinone group (1,8-dihydroxy-anthraquinone, 1,3,6,8-tetrahydroxy-9(10H)-anthracenone, 8,8'-dihydroxy-2,2'-binaphthalene-1,1',4,4'-tetrone, etc.), polyhydroxycarboxylic acids, omega-3 fatty acids (stearidonic acid, linolenic acid), and others. Our results imply that Ribes species are rich in polyphenols, especially flavanols, anthocyanins, flavones, and flavan-3-ols. These results indicate the utility of Ribes species for the health and pharmaceutical industry.

This mini review delves into the unique potential of bioenergy innovations, particularly renewable diesel, bio jet fuel, and ethanol, to reduce greenhouse gas emissions and transform various industries. The article highlights critical technological advancements, supportive policies, and cross-sector collaboration essential for a sustainable energy transition. Specific challenges such as ensuring a consistent biomass feedstock supply, decentralizing processing units, and navigating complex regulatory frameworks are examined. Innovative solutions like decentralized biomass processing and enhanced biomass logistics are discussed as pathways to overcome these barriers. Drought is a major limiting factor for the growth and development of pumpkins. Plasma membrane intrinsic proteins (PIPs) are major water channels that play a crucial role in the regulation of cellular water status and solute trafficking during drought conditions. CmoPIP1-4 is a plasma membrane-localized protein that is significantly upregulated in roots and leaves under drought-stress conditions. In this study, the overexpression of CmoPIP1-4 enhances drought resistance in yeast. In contrast, CRISPR-mediated CmoPIP1-4 knockout in pumpkin roots increased drought sensitivity. This increased drought sensitivity of CmoPIP1-4 knockout plants is associated with a decline in the levels of hydrogen sulfide (H 2 S) and abscisic acid (ABA), accompanied by an increase in water loss caused by greater levels of transpiration and stomatal conductance. In addition, the sensitivity of CmoPIP1-4 CRISPR plants is further aggravated by reduced antioxidative enzyme activity, decreased proline and sugar contents, and extensive root damage. Furthermore, expression profiles of genes such as CmoHSP70s, CmoNCED3, CmoNCED4, and others involved in metabolic activities were markedly reduced in CmoPIP1-4 CRISPR plants. Moreover, we also discovered an interaction between the drought-responsive gene CmoDCD and CmoPIP1-4, indicating their potential role in activating H 2 S-mediated signaling in pumpkin, which could confer drought tolerance. Studies have reported inconsistent results about the risk of incident chronic kidney disease (CKD) in people with metabolically healthy obesity (MHO). We designed this systematic review and meta-analysis to evaluate the risk of developing CKD in people with MHO and metabolically unhealthy normal weight (MUNW). We used a predefined search strategy to retrieve eligible studies from multiple databases up to June 20, 2022. Random-effects model meta-analyses were implied to estimate the overall hazard ratio (HR) of incident CKD in obesity phenotypes. Eight prospective cohort studies, including approximately 5 million participants with a median follow-up ranging between 3 and 14 years, were included in this meta-analysis. Compared to the metabolically healthy normal weight (MHNW), the mean differences in cardiometabolic and renal risk factors in MHO, MUNW, and metabolically unhealthy obesity (MUO) were evaluated with overall HR of 1.42, 1.49, and 1.84, respectively. Compared to MHNW, the mean estimated glomerular filtration rate (eGFR) and high-density lipoprotein (HDL) were significantly lower, and low-density lipoprotein (LDL), blood pressure, blood glucose, and triglycerides were higher in MHO and MUNW. In conclusion, MHO and MUNW are not benign conditions and pose a higher risk for incident CKD. Pectinolytic enzymes are among the important group of industrial enzymes that have wide applications in different food industries. In this study, pectinase-based silica nanocarriers were synthesized using co-precipitation and cross-linking techniques. The resulting silica nanoparticles were investigated using scanning electron microscopy (SEM), energy-dispersive electron microscopy (EDEX), and X-ray diffraction (XRD) for determination of its morphology, elemental composition, and crystalline pattern. Under the optimal immobilization conditions like 1.5 % glutaraldehyde, 3000 IU/mg pectinase concentration, 90 min immobilization time and 40 °C immobilization temperature, pectinase showed maximum immobilization yield. The immobilization of pectinase onto the silica nanocarriers led to enhanced catalytic characteristics, displaying higher enzymatic activity across various temperature and pH levels compared to soluble pectinase. Moreover, the immobilization substantially improved the temperature stability of pectinase, exhibiting 100 % of its initial activity even after 120 h of pre-incubation at 50 °C. Additionally, the silica nanocarrier pectinase retained 100 % of its original activity even after being reused 10 times in a single batch of reactions. Brassica species show varying levels of resistance to salt stress. To understand the genetics underlying these differential stress tolerance patterns in Brassicas, we exposed two widely cultivated amphidiploid Brassica species having different genomes, Brassica juncea (AABB, n = 18) and Brassica napus (AACC, n = 19), to elevated levels of NaCl concentration (300 mM, half the salinity of seawater). B. juncea produced more biomass, an increased chlorophyll content, and fewer accumulated sodium (Na + ) and chloride (Cl - ) ions in its photosynthesizing tissues. Chlorophyll fluorescence assays revealed that the reaction centers of PSII of B. juncea were more photoprotected and hence more active than those of B. napus under NaCl stress, which, in turn, resulted in a better PSII quantum efficiency, better utilization of photochemical energy with significantly reduced energy loss, and higher electron transport rates, even under stressful conditions. The expression of key genes responsible for salt tolerance ( NHX1 and AVP1 , which are nuclear-encoded) and photosynthesis ( psbA , psaA , petB , and rbcL , which are chloroplast-encoded) were monitored for their genetic differences underlying stress tolerance. Under NaCl stress, the expression of NHX1 , D1 , and Rubisco increased several folds in B. juncea plants compared to B. napus , highlighting differences in genetics between these two Brassicas. The higher photosynthetic potential under stress suggests that B. juncea is a promising candidate for genetic modifications and its cultivation on marginal lands.

The lack of information regarding biodiversity status hampers designing and implementing conservation strategies and achieving future targets. Northern Pakistan consists of a unique ecoregion mosaic which supports a myriad of environmental niches for anuran diversity in comparison to the deserts and xeric shrublands throughout the rest of the country. In order to study the niche suitability, species overlap and distribution patterns in Pakistan, we collected observational data for nine anuran species across several distinct ecoregions by surveying 87 randomly selected locations from 2016 to 2018 in Rawalpindi District and Islamabad Capital Territory. Our model showed that the precipitation of the warmest and coldest quarter, distance to rivers and vegetation were the greatest drivers of anuran distribution, expectedly indicating that the presence of humid forests and proximity to waterways greatly influences the habitable range of anurans in Pakistan. Sympatric overlap between species occurred at significantly higher density in tropical and subtropical coniferous forests than in other ecoregion types. We found species such as Minervarya spp., Hoplobatrachus tigerinus and Euphlyctis spp. preferred the lowlands in proximal, central and southern parts of the study area proximal to urban settlements, with little vegetation and higher average temperatures. Duttaphrynus bengalensis and D. stomaticus had scattered distributions throughout the study area with no clear preference for elevation. Sphaerotheca pashchima was patchily distributed in the midwestern extent of the study area as well as the foothills to the north. Microhyla nilphamariensis was widely distributed throughout the study area with a preference for both lowlands and montane terrain. Endemic frogs (Nanorana vicina and Allopaa hazarensis) were observed only in locations with higher elevations, higher density of streams and lower average temperatures as compared to the other seven species sampled. It is recommended to provide legal protection to amphibians of Pakistan, especially endemic species, through revision in the existing wildlife laws. The Two-component system (TCS) consists of Histidine kinases (HKs), Phosphotransfers (HPs), and response regulator (RR) proteins. It has an important role in signal transduction to respond to a wide variety of abiotic stresses and hence in plant development. Brassica oleracea (cabbage) is a leafy vegetable, which is used for food and medicinal purposes. Although this system was identified in several plants, it had not been identified in Brassica oleracea yet. This genome-wide study identified 80 BoTCS genes consisting of 21 HKs, 8 HPs, 39 RRs, and 12 PRRs. This classification was done based on conserved domains and motif structure. Phylogenetic relationships of BoTCS genes with Arabidopsis thaliana , Oryza sativa , Glycine max, and Cicer arietinum showed conservation in TCS genes. Gene structure analysis revealed that each subfamily had conserved introns and exons. Both tandem and segmental duplication led to the expansion of this gene family. Almost all of the HPs and RRs were expanded through segmental duplication. Chromosomal analysis showed that BoTCS genes were dispersed across all nine chromosomes. The promoter regions of these genes were found to contain a variety of cis-regulatory elements. The 3D structure prediction of proteins also confirmed the conservation of structure within subfamilies. MicroRNAs (miRNAs) involved in the regulation of BoTCSs were also predicted and their regulatory roles were also evaluated. Moreover, BoTCSs were docked with abscisic acid to evaluate their binding. RNA-seq-based expression analysis and validation by qRT-PCR showed significant variation of expression for BoPHYs, BoERS1.1, BoERS2.1, BoERS2.2, BoRR10.2, and BoRR7.1 suggesting their importance in stress response. These genes showing unique expression can be further used in manipulating the plant's genome to make the plant more resistant the environmental stresses which will ultimately help in the increase of plant's yield. Sorghum is emerging as a model crop for functional genetics and genomics of tropical grasses with abundant uses, including food, feed, and fuel, among others. It is currently the fifth most significant primary cereal crop. Crops are subjected to various biotic and abiotic stresses, which negatively impact on agricultural production. Developing high-yielding, disease-resistant, and climate-resilient cultivars can be achieved through marker-assisted breeding. This current study examines the impact of renewable energy consumption, agriculture, and globalization on carbon emissions in India over the period from 1980 to 2018. For long-run estimates, we apply Gregory-Hansen's co-integration test, bootstrap ARDL approaches, fully modified ordinary least squares, and dynamic OLS. The empirical results of long-run estimates indicate that a 1% increase in renewable energy consumption, agriculture, and economic globalization will increase carbon emissions by 0.764%, 1.675%, and 0.517%, respectively. Moreover, this study confirms the detrimental effect of these variables on carbon dioxide emissions. Economic globalization coefficients indicate that the scale effect is valid in India. The 2002 economic crisis slowed down the country's growth rate, which reduced the ecological pollution. Several policy recommendations are derived from the empirical findings., (© 2023. Over the last three decades, the world has been facing the phenomenon of the ecological deficit as the ecological footprint is continuously rising due to the persistent decline of the per-capita bio-capacity. Moreover, there is a substantial increase in globalization and electricity consumption for the same period, and transportation is contributing to economic prosperity at the cost of environmental sustainability. Understanding the determinants of ecological footprint is thus critical for suggesting appropriate policies for environmental sustainability. As a result, this study analyzes the impacts of economic globalization, transportation, coal rents, and electricity consumption in ecological footprint in the context of the USA over the period 1995 to 2018. The data have been extracted from "Global Footprint Network," "Swiss Economic Institute," and "World Development Indicators." The current study has also applied the flexible Fourier form nonlinear unit root test to examine the stationarity among variables. For the empirical estimation, a novel technique, the "quantile auto-regressive distributive lag model," is applied in the study to deal with the nonlinear associations of the variables and to evaluate the long-term stability of variables across quantiles. The study's findings indicate that coal rents, transportation, and globalization significantly and positively contribute to the deterioration of ecological footprints at different quantile ranges in the short and long run. Electricity consumption is found to have a positive and significant impact at lower quantile ranges in the long run but not have a significant impact in the short run. The study suggested that lowering the dependence of the transport sector on fossil fuels, more use of hydroelectricity, and stringent strategies to curb coal consumption would be helpful to reduce the positive influence of these variables on ecological footprints in the USA., (© 2022. Based on the Pakistan Demographic and Health Survey (PDHS) 2017-18 data, the study intends to present the relative status of a particular fuel (type) in terms of its level of use in different regions of Pakistan. At the same time, the study investigates the determinants of the household type of fuel used for cooking in Pakistan. Socioeconomic characteristics of the households and household heads are taken as determinants of the main type of fuel (clean or dirty) used for cooking by the households. Binary logistic regression is applied as an estimation technique. Over the country, wood stands first as the most widely used type of fuel followed by natural gas followed by LPG. Animal dung and charcoal are respectively ranked as the fourth and fifth most commonly used types of fuel for cooking by households in Pakistan. In comparison amongst the four provinces of Pakistan, the use of wood as well as charcoal as the fuel type for cooking (in relative terms) is the most common in Balochistan province. Likewise, Punjab province is the most frequent user of liquefied petroleum gas (LPG), crop residues, and animal dung, while Sindh province is the most frequent user of natural gas. Households possessing agricultural land and livestock and with large household sizes had more probability to use dirty sources of fuel for cooking. Households with better wealth status and residing in urban areas were more likely to use clean sources of fuel for cooking. To increase its operational stability and ongoing reusability, B. subtilis pectinase was immobilized on iron oxide nanocarrier. Through co-precipitation, magnetic iron oxide nanoparticles were synthesized. Scanning electron microscopy (SEM) and energy dispersive electron microscopy (EDEX) were used to analyze the nanoparticles. Pectinase was immobilized using glutaraldehyde as a crosslinking agent on iron oxide nanocarrier. In comparison to free pectinase, immobilized pectinase demonstrated higher enzymatic activity at a variety of temperatures and pH levels. Immobilization also boosted pectinase's catalytic stability. After 120 h of pre-incubation at 50 °C, immobilized pectinase maintained more than 90% of its initial activity due to the iron oxide nanocarrier, which improved the thermal stability of pectinase at various temperatures. Following 15 repetitions of enzymatic reactions, immobilized pectinase still exhibited 90% of its initial activity. According to the results, pectinase's catalytic capabilities were enhanced by its immobilization on iron oxide nanocarrier, making it economically suitable for industrial use.

Faba bean ( Vicia faba L.) protein is a new plant protein alternative source with high nutrient content especially protein and phenolic compounds. The present study investigated physicochemical properties, phenolic content, antioxidant potential, and short chain fatty acids (SCFAs) production during in vitro digestion and colonic fermentation of faba bean hydrolysates and oil-in-water (O/W) emulsions. Results indicate that the enzymic hydrolysates of faba proteins exhibited higher protein solubility, increased electronegativity, and decreased surface hydrophobicity than native faba protein. O/W emulsions showed improved colloidal stability for the faba protein hydrolysates after ultra-high temperature processing (UHT). Furthermore, UHT processing preserved total phenolic content, DPPH and ABTS radical scavenging abilities while decreasing total flavonoid content and ferric reducing power. Besides, the release of phenolic compounds in faba bean hydrolysates (FBH) and emulsions (FBE) improved after intestinal digestion by 0.44 mg GAE/g and 0.55 mg GAE/g, respectively. For colonic fermentation, FBH demonstrated an approximately 10 mg TE/g higher ABTS value than FBE (106.45 mg TE/g). Total SCFAs production of both FBH and FBE was only 0.03 mM. The treatment of FBH with 30 min enzymatic hydrolysis displayed relatively higher antioxidant capacities and SCFAs production, indicating its potential to bring more benefits to gut health. Overall, this study showed that enzymic hydrolysis of faba proteins not only improved the colloidal emulsion stability, but also released antioxidant capacity during in vitro digestibility and colonic fermentation. Colonic fermentation metabolites (SCFAs) were related to the degree of hydrolysis for both FBH and FBE. Additional studies are required to further elucidate and differentiate the role of phenolics during faba protein processing and digestion stages in comparison to contributions of peptides, amino acids and microelements to digestion rates, antioxidant capacities and colonial SCFA production.

Melon fruits are popular because of sweet taste and pleasant aroma. Grafting has been extensively used for melons to alleviate abiotic stresses and control soil borne diseases. However, use of grafting for vegetable fruit quality improvement is less studies. In modern age fruit quality particularly sensory quality characteristics have key importance from consumer eye lens. We performed liquid chromatography-mass spectrometry and metabonomic analysis to examine sensory fruit quality of melon grafted onto ten different pumpkin rootstocks. Bases on the result of our study, 478 metabolites were detected and 184 metabolites consisting of lipids, amino acids and organic oxygen compounds were differentially expressed in grafted melon fruits. The results from metabolomic, physiochemical and sensory analysis explain the differences in melon fruit flavor from two contrasting rootstocks. In conclusion the fruits from Tianzhen No. 1 rootstock exhibited better organoleptic characteristics and higher soluble sugars content [glucose (19.87 mg/g), fructose (19.68 mg/g) and sucrose (169.45 mg/g)] compared with other rootstocks used in this study. Moreover, the contents of bitterness causing amino acids such as L-arginine, L-asparagine, Histidinyl-histidine and Acetyl-DL-valine were found lower in Tianzhen No. 1-grafted melon fruits compared with Sizhuang No. 12-grafted melon fruits. Dwarfing is an important agricultural trait for intensive cultivation and effective orchard management in modern fruit orchards. Commercial citrus production relies on grafting with rootstocks that reduce tree vigor to control plant height. Citrus growers all over the world have been attracted to dwarfing trees because of their potential for higher planting density, increased productivity, easy harvest, pruning, and efficient spraying. Dwarfing rootstocks can be used to achieve high density. As a result, the use and development of dwarfing rootstocks are important. Breeding programs in several countries have led to the production of citrus dwarf rootstocks. For example, the dwarfing rootstocks 'Flying Dragon', 'FA 517', 'HTR-051', 'US-897', and 'Red tangerine' cultivated in various regions allow the design of dense orchards. Additionally, dwarf or short-stature trees were obtained using interstocks, citrus dwarfing viroid (CDVd) and various chemical applications. This review summarizes what is known about dwarf citrus rootstocks and the mechanisms underlying rootstock-scion interactions. Despite advances in recent decades, many questions regarding rootstock-induced scion development remain unanswered. Citrus rootstocks with dwarfing potential have been investigated regarding physiological aspects, hormonal communication, mineral uptake capacity, and horticultural performance. This study lays the foundation for future research into the genetic and molecular mechanisms underlying citrus dwarfing.

Heavy metal/metalloids (HMs) are among the primary soil pollutants that limit crop production worldwide. Plants grown in HM contaminated soils exhibit reduced growth and development, resulting in a decrease in crop production. The exposure to HMs induces plant oxidative stress due to the formation of free radicals, which alter plant morphophysiological and biochemical mechanisms at cellular and tissue levels. When exposed to HM toxicity, plants evolve sophisticated physiological and cellular defense strategies, such as sequestration and transportation of metals, to ensure their survival. Plants also have developed efficient strategies by activating signaling pathways, which induce the expression of HM transporters. Plants either avoid the uptake of HMs from the soil or activate the detoxifying mechanism to tolerate HM stress, which involves the production of antioxidants (enzymatic and non-enzymatic) for the scavenging of reactive oxygen species. The metal-binding proteins including phytochelatins and metallothioneins also participate in metal detoxification. Furthermore, phytohormones and their signaling pathways also help to regulate cellular activities to counteract HM stress. The excessive levels of HMs in the soil can contribute to plant morpho-physiological, biochemical, and molecular alterations, which have a detrimental effect on the quality and productivity of crops. To maintain the commercial value of fruits and vegetables, various measures should be considered to remove HMs from the metal-polluted soils. Bioremediation is a promising approach that involves the use of tolerant microorganisms and plants to manage HMs pollution. Despite of plethora of research on hepatoprotective potential of medicinal plants, there is still need to discover potential plants with hepatoprotective activity. Iris florentina L. is a medicinal plant with traditional claims but ignored investigation regarding its hepatoprotective effects. The current study is aimed to investigate the hepatoprotective potential of I. florentina L. methanolic extract on paracetamol (PCM)-induced liver injury. The phytochemical and HPLC screening was done which showed the presence of potential constituents including flavonoids and phenols. For investigating the hepatoprotective effect of I. florentina L. methanolic extract, rats were given five different treatments for seven consecutive days. The normal control (group 1) was administered with normal saline, group 2 (Diseased) received paracetamol and group 3 (Standard) was given silymarin as reference drug. In group 4 and 5 (Treated), I. florentina L. methanolic extract (250 and 500mg/kg) were administered. Different serum biomarkers and histopathological studies were performed to assess the recovery caused by PCM in comparison to diseased group. The treatment of I. florentina methanolic extract significantly improve the serum biomarkers and restored the hepatic injury towards normal, indicating the hepatoprotective potential. Thus, we can conclude that I. florentina have significantly reversed the damage caused by paracetamol in hepatotoxic rat model due to their potential phytochemical constituents.

This research article aims to evaluate the characteristics of ecotourism destination loyalty in light of destination attachment, destination equity framed by perceived value, and tourist experience. Thus, the attributes of ecotourism destination branding in formulating tourist loyalty are examined. The study is of significant importance for developing economies having natural tourist destinations. A total of 358 questionnaires were filled through wjx, and a SmartPLS-based structural equation modeling tool was used to analyze the data obtained from eco-tourists. The software is essential for complex structural models, including multiple indicators, and relationships. The empirical results exhibit that perceived value and tourist experience significantly contribute to destination loyalty and equity, eventually influencing tourist destination loyalty. Moreover, destination memory moderates the relationship between destination attachment, destination equity, and destination loyalty. Further, destination attachment and destination equity mediate the relationship between the perceived value, experience, and destination loyalty. Additionally, the study extends the tourist consumption theory to the ecotourism literature. Besides the theoretical contribution, the study makes a practical contribution to practitioners. For instance, perceived value is a prime contributor to tourist destination loyalty. In perceived value, the most important factor is good value for money. This study was designed to evaluate the emulsifying and rheological properties of acorn protein isolate (API) in different pH mediums (pH 3, 7 and 9) and in the presence of ionic salts (1 M NaCl and 1 M CaCl2). API shows higher solubility in distilled water at pH 7, while at the same pH, a decrease in solubility was observed for API in the presence of CaCl2 (61.30%). A lower emulsifying activity index (EAI), lower stability index (ESI), larger droplet sizes and slight flocculation were observed for API in the presence of salts at different pHs. Importantly, CaCl2 treated samples showed relevantly higher EAI (252.67 m2/g) and ESI (152.67 min) values at all pH as compared to NaCl (221.76 m2/g), (111.82 min), respectively. A significant increase in interfacial protein concentration (4.61 mg/m2) was observed for emulsion at pH 9 with CaCl2, while the major fractions of API were observed in an interfacial layer after SDS-PAGE analysis. All of the emulsion shows shear thinning behavior (τc > 0 and n < 1), while the highest viscosity was observed for emulsion prepared with CaCl2 at pH 3 (11.03 ± 1.62). In conclusion, API, in the presence of ionic salts at acidic, neutral and basic pH, can produce natural emulsions, which could be substitutes for synthetic surfactants for such formulations.

The study's prime objective is to investigate the user discontinuance intention in the shed of the negative disconfirmation of user expectation. The study has derived the theoretical structure from the expectancy disconfirmation theory (EDT) enacted through the stimuli organism response (SOR) framework to study the actual cause and effect relationship of human behavioral response. To investigate the user discontinuance intention behavioral response, a total of 434 correct and complete answers were shortlisted for analysis. To examine the data set, the study has used the modern partial least square method technique or simply SmartPLS service package to run the structural equation modeling (SEM). Moreover, the study has implied the 80/20 rule run the mediating analysis of the SOR framework. The statistical results show that all three stimuli make significant positive disconfirmation of the user beliefs in terms of dissatisfaction and the anxiety that ultimately leads to the discontinuance intention in virtual network users. Further, these results are validated through the six mediating relationships, which partially mediate the relationship between the stimuli and response. Dahi is a very common and traditional fermented dairy product in Pakistan and its neighboring countries, it represents a rich source for the isolation of many new strains of lactic acid bacteria (LAB). The major objective of this study was to evaluate the probiotic potential of novel exopolysaccharide (EPS)-producing strains of S. thermophilus isolated from Dahi, sold in the local markets of Rawalpindi and Islamabad, Pakistan. In this study, 32 isolates of S. thermophilus were initially isolated from Dahi and out of these, 10 identified strains were further screened for their EPS-producing ability. Maximum EPS production was estimated for RIY strain (133.0 ± 0.06), followed by RIH4 strain (103.83 ± 0.76) and RIRT2 strain (95.77 ± 0.22), respectively. Thereafter, in vitro studies revealed that these newly identified EPS-producing strains of S. thermophilus fulfilled the basic requirements for probiotic functions; including resistance to harsh conditions of GIT, good cell surface hydrophobicity, auto-aggregation, and co-aggregation, especially against L. monocytogenes . Finally, the safety assessment displayed that these strains were also sensitive to clinical antibiotics, including vancomycin. Tomato is an important vegetable that is highly sensitive to drought (DR) stress which impairs the development of tomato seedlings. Recently, melatonin (ME) has emerged as a nontoxic, regulatory biomolecule that regulates plant growth and enhances the DR tolerance mechanism in plants. The present study was conducted to examine the defensive role of ME in photosynthesis, root architecture, and the antioxidant enzymes' activities of tomato seedlings subjected to DR stress. Our results indicated that DR stress strongly suppressed growth and biomass production, inhibited photosynthesis, negatively affected root morphology, and reduced photosynthetic pigments in tomato seedlings. Per contra, soluble sugars, proline, and ROS (reactive oxygen species) were suggested to be improved in seedlings under DR stress. Conversely, ME (100 µM) pretreatment improved the detrimental-effect of DR by restoring chlorophyll content, root architecture, gas exchange parameters and plant growth attributes compared with DR-group only. Moreover, ME supplementation also mitigated the antioxidant enzymes [APX (ascorbate peroxidase), CAT (catalase), DHAR (dehydroascorbate reductase), GST (glutathione S-transferase), GR (glutathione reductase), MDHAR (monodehydroascorbate reductase), POD (peroxidase), and SOD (superoxide dismutase)], non-enzymatic antioxidant [AsA (ascorbate), DHA (dehydroascorbic acid), GSH (glutathione), and GSSG, (oxidized glutathione)] activities, reduced oxidative damage [EL (electrolyte leakage), H 2 O 2 (hydrogen peroxide), MDA (malondialdehyde), and O 2 •- (superoxide ion)] and osmoregulation (soluble sugars and proline) of tomato seedlings, by regulating gene expression for SOD , CAT , APX , GR , POD , GST , DHAR , and MDHAR . These findings determine that ME pretreatment could efficiently improve the seedlings growth, root characteristics, leaf photosynthesis and antioxidant machinery under DR stress and thereby increasing the seedlings' adaptability to DR stress.

The major purpose of this study is to assess racial disparity and energy poverty index by measuring energy poverty index by using data envelopment analysis and regression equation from South Asia (2001-2018). An energy poverty index is quantifying the size and scope of energy poverty, and DEA is used to investigate the relevance of socioeconomic position to multidimensional energy poverty. In multidimensional energy poverty, location, house ownership position, number of dependents, and the age of the main caregiver have an important positive impact. Our research has shown that Bhutan is the most susceptible nation with an energy poverty index of (0.02), Maldives (0.03), and Bangladesh (0.11), while India (0.86) and Pakistan (0.49) are the least likely to be energy poor as regards energy poverty. Of the total energy production, 78% is based on traditional fuels, followed by 12% based on petroleum products. The Gini index indicates a positive association with the energy poverty index at a 5% significance level. This signifies that these socioeconomic indicators positively contribute to the energy poverty index level. This study developed more synchronized policies to eradicate energy poverty and can provide a way forward for policymakers to develop strategies to implement them suitably in the regional power sector., (© 2021. SARS-CoV-2, an RNA virus, has been prone to high mutations since its first emergence in Wuhan, China, and throughout its spread. Its genome has been sequenced continuously by many countries, including Pakistan, but the results vary. Understanding its genomic patterns and connecting them with phenotypic features will help in devising therapeutic strategies. Thus, in this study, we explored the mutation landscape of 250 Pakistani isolates of SARS-CoV-2 genomes to check the genome diversity and examine the impact of these mutations on protein stability and viral pathogenesis in comparison with a reference sequence (Wuhan NC 045512.2). Our results revealed that structural proteins mainly exhibit more mutations than others in the Pakistani isolates; in particular, the nucleocapsid protein is highly mutated. In comparison, the spike protein is the most mutated protein globally. Furthermore, nsp12 was found to be the most mutated NSP in the Pakistani isolates and worldwide. Regarding accessory proteins, ORF3A is the most mutated in the Pakistani isolates, whereas ORF8 is highly mutated in world isolates. These mutations decrease the structural stability of their proteins and alter different biological pathways. Molecular docking, the dissociation constant (K D ), and MM/GBSA analysis showed that mutations in the S protein alter its binding with ACE2. The spike protein mutations D614G-S943T-V622F (-75.17 kcal/mol), D614G-Q677H (-75.78 kcal/mol), and N74K-D614G (-73.84 kcal/mol) exhibit stronger binding energy than the wild type (-66.34 kcal/mol), thus increasing infectivity. Furthermore, the simulation results strongly corroborated the predicted protein servers. Our analysis findings also showed that E, M, ORF6, ORF7A, ORF7B, and ORF10 are the most stable coding genes; they may be suitable targets for vaccine and drug development., (Copyright © 2021. Globally, the interaction and vulnerability of tourism and climate change have recently been in focus. This study examines how carbon dioxide emissions respond to changes in the tourism development. Panel data from 2000 to 2017 for 70 countries are analyzed using spatial econometric method to investigate the spatial spillover effect of tourism development on environmental pollution. The direct, indirect, and overall impact of tourism on environmental pollution are estimated after the selection of the most appropriate GNS method. The findings reveal that tourism has a positive direct effect and a negative indirect effect; both are significant at the 1 % level. The negative indirect effect of tourism is greater than its direct positive effect, implying an overall significantly negative impact. Further, the outcome of financial development and carbon emissions have an inverted U-shaped and U-shaped relationship in direct and indirect impacts. Population density, trade openness and economic growth significantly influence on environmental pollution through spatial spill over. In addition, education expenditure and infrastructure play a significant moderating role in the relationship among tourism development and environmental pollution. The results have important policy implications as they establish an inverted-U-shaped relationship among tourism and environmental pollution and indicate that while a country's emissions initially rise with the tourism industry's growth, they begin declining after a limit., (Copyright © 2021 Elsevier Ltd. Arabidopsis contains 317 genes for defensin-like (DEFL) peptides. DEFLs have been grouped into different families based mainly on cysteine motifs. The DEFL0770 group contains seven genes, of which four are strongly expressed in roots. We found that the expression of these genes is downregulated in syncytia induced by the beet cyst nematode Heterodera schachtii as revealed by RNAseq analysis. We have studied one gene of this group, At3g59930 , in detail. A promoter::GUS line revealed that the gene is only expressed in roots but not in other plant organs. Infection of the GUS line with larvae of H. schachtii showed a strong downregulation of GUS expression in infection sites as early as 1 dpi, confirming the RNAseq data. The At3g59930 peptide had only weak antimicrobial activity against Botrytis cinerea . Overexpression lines had no enhanced resistance against this fungus but were more resistant to H. schachtii infection. Our data indicate that At3g59930 is involved in resistance to nematodes which is probably not due to direct nematicidal activity.

The snow leopard ( Panthera uncia ) inhabits one of the most challenging environments on Earth, referred to as the 'third pole'. Only a fraction of its vast range has been explored thus far, owing to myriad of barriers inflicted by the remote terrain and socio-ecological realities of the landscapes. Understanding distribution patterns of species is essential to devise practical management measures. This study aimed to understand the distribution pattern and factors influencing occupancy of snow leopard in the Pamir Mountain range through sign-based occupancy modelling. Our study confirmed that the Pamir range is a snow leopard stronghold, with occupancy estimated at 0.57 ± 0.02. The topographic features positively influenced the detection probability (p = 0.37 ± 0.005) of snow leopards. Occupancy was influenced by mean annual precipitation (β = -6.12 ± 1.8), density of roads (β = -1.61 ± 0.6) and water sources (β = 0.74 ± 0.4). Our findings underpin that sign-based distribution surveys provide vigorous scientific knowledge about elusive species and merit replication being used for other species.
