Background: Immune checkpoint inhibitors have revolutionized cancer therapy, but many patients fail to respond or develop resistance, often due to reduced T cell activity. Costimulation via 4-1BB has emerged as a promising approach to enhance the effector function of antigen-primed T cells. Bispecific T cell-engaging (TCE) antibodies are an effective way to provide tumor-specific T cell receptor-mediated signaling to tumor-infiltrating lymphocytes. mRNA-based delivery of bispecific antibodies, offer a novel approach to enhance tumor-specific immune responses while minimizing adverse effects. Methods: Two bispecific antibodies were generated: the EGFR x CD3 TCE antibody (LiTE) and the PD-L1 x 4-1BB costimulatory antibody (LiTCo), which was further fused to a high FcRn albumin variant (Albu-LiTCo). The mRNA encoding these bispecific antibodies contains an N1-methylpseudouridine modified nucleoside and regulatory sequences to ensure proper expression and stability. A series of in vitro assays and cell-based analyses were performed to characterize both antibodies. The in vivo efficacy of the mRNA-encoded bispecific antibodies was evaluated in xenograft tumor models expressing EGFR. Results: We investigated the combined effect of two mRNA-encoded Fc-free bispecific antibodies with complementary mechanisms of action: an EGFR-targeting TCE and a half-life extended PD-L1 x 4-1BB costimulatory antibody. The mRNAs encoding both bispecific LiTE
RNA and Albu-LiTCoRNA , showed similar binding specificity and in vitro function to their protein analogues. Pharmacokinetic studies demonstrated sustained expression of both bispecific antibodies following intravenous administration of the mRNAs formulated using a polymer/lipid-based nanoparticle (LNP) but different pharmacokinetic profiles, shorter for the TCE and longer for the PD-L1 x 4-1BB. When administered as a mRNA-LNP combination (ComboRNA ), the growth of EGFR-positive tumors in immunocompetent mice was significantly inhibited, resulting in tumor regression in 20% of cases with no associated toxicity. Histological analysis confirmed increased T cell infiltration in the tumors treated with LITERNA and ComboRNA . Repeated administration resulted in sustained production of bispecific antibodies with different exposure cycles and potent antitumor activity with a favorable safety profile. Conclusions: These results highlight the potential of combining two mRNA-encoded bispecific antibodies with different mechanisms of action and programmable half-life for cancer immunotherapy. WASTEWATER treatment , *FACTORIES , *AQUEOUS solutions , *PILOT plants , *FLOCCULATION - Abstract
Electrocoagulation (EC) and coagulation – flocculation (CF), have been employed for the treatment of different organic contaminants present in aqueous solutions owing to their efficiency. This review article reports the results obtained by different authors in the last 5 years, where EC, CF, and their coupling with other processes, are employed at the laboratory, pilot plant, and industrial scales. The evaluation of operating conditions, efficiencies, applications, limitations, and opportunities that arise in scaling both processes are presented. In this way, the present paper is novel because it presents evidence on the advantages and disadvantages of EC and CF technologies implemented at pilot/full scale to treat real wastewater, which has not been collected and synthesised in previous studies. The average elimination percentages (%COD) were evaluated at a pilot/industrial plant scale, where EC (%COD = 70%) presents higher efficiencies compared to CF (%COD = 60%). In CF, the %COD increases when using longer times, contrary to EC (%COD = 75% at t = 10 h in CF and %COD = 80% at t = 0.33 h in EC). Achieving neutral conditions in treated wastewater promotes high efficiencies and low energy consumption, causing costs to be reduced. The origin and composition of the wastewater also influence the pH and efficiencies. EC is the most studied method at laboratory scale, however, its development at an industrial level is minimal, unlike CF, where more studies are reported on a pilot and industrial scale. In both processes, the use of oxidising agents and ultraviolet energy favours the formation of metal radicals and the transformation of contaminants, increasing removal efficiencies. - Abstract

- Abstract
Administration of neutralizing antibodies (nAbs) has proved to be effective by providing immediate protection against SARS-CoV-2. However, dual strategies combining virus neutralization and immune response stimulation to enhance specific cytotoxic T cell responses, such as dendritic cell (DC) cross-priming, represent a promising field but have not yet been explored. Here, a broadly nAb, TNT, are first generated by grafting an anti-RBD biparatopic tandem nanobody onto a trimerbody scaffold. Cryo-EM data show that the TNT structure allows simultaneous binding to all six RBD epitopes, demonstrating a high-avidity neutralizing interaction. Then, by C-terminal fusion of an anti-DNGR-1 scFv to TNT, the bispecific trimerbody TNTDNGR-1 is generated to target neutralized virions to type 1 conventional DCs (cDC1s) and promote T cell cross-priming. Therapeutic administration of TNTDNGR-1, but not TNT, protects K18-hACE2 mice from a lethal SARS-CoV-2 infection, boosting virus-specific humoral responses and CD8+ T cell responses. These results further strengthen the central role of interactions with immune cells in the virus-neutralizing antibody activity and demonstrate the therapeutic potential of the Fc-free strategy that can be used advantageously to provide both immediate and long-term protection against SARS-CoV-2 and other viral infections.

- Abstract
Immune checkpoint blockade (ICB) with antibodies has shown durable clinical responses in a wide range of cancer types, but the overall response rate is still limited. Other effective therapeutic modalities to increase the ICB response rates are urgently needed. New bispecific antibody (bsAb) formats combining the ICB effect and a direct action on cancer cells could improve the efficacy of current immunotherapies. Here, we report the development of a PD-L1/EGFR symmetric bsAb by fusing a dual-targeting tandem trimmer body with the human IgG1 hinge and Fc regions. The bsAb was characterized in vitro and the antitumor efficacy was evaluated in humanized mice bearing xenografts of aggressive triple-negative breast cancer and lung cancer. The IgG-like hexavalent bsAb, designated IgTT-1E, was able to simultaneously bind both EGFR and PD-L1 antigens, inhibit EGF-mediated proliferation, effectively block PD-1/PD-L1 interaction, and induce strong antigen-specific antibody-dependent cellular cytotoxicity activity in vitro. Potent therapeutic efficacies of IgTT-1E in two different humanized mouse models were observed, where tumor growth control was associated with a significantly increased proportion of CD8+ T cells. These results support the development of IgTT-1E for the treatment of EGFR+ cancers.

Immune checkpoint blockade has changed the treatment paradigm for advanced solid tumors, but the overall response rates are still limited. The combination of checkpoint blockade with anti-4-1BB antibodies to stimulate tumor-infiltrating T cells has shown anti-tumor activity in human trials. However, the further clinical development of these antibodies has been hampered by significant off-tumor toxicities. Here, we generated an anti-4-1BB/EGFR/PD-L1 trispecific antibody consisting of a triple-targeting tandem trimerbody (TT) fused to an engineered silent Fc region. This antibody (IgTT-4E1-S) was designed to combine the blockade of the PD-L1/PD-1 axis with conditional 4-1BB costimulation specifically confined to the tumor microenvironment (TME). The antibody demonstrated simultaneous binding to purified EGFR, PD-L1, and 4-1BB in solution, effective blockade of the PD-L1/PD1 interaction, and potent 4-1BB-mediated costimulation, but only in the presence of EGFR-expressing cells. These results demonstrate the feasibility of IgTT-4E1-S specifically blocking the PD-L1/PD-1 axis and inducing EGFR-conditional 4-1BB agonist activity. [ABSTRACT FROM AUTHOR]

- Abstract
Retargeting of T lymphocytes toward cancer cells by bispecific antibodies has demonstrated its therapeutic potential, with one such antibody approved for the treatment of acute lymphoblastic leukemia (blinatumomab) and several other in clinical trials. However, improvement of their efficacy and selectivity for solid tumors is still required. Here, we describe a novel tandem T-cell recruiting trispecific antibody for the treatment of colorectal cancer (CRC). This construct, termed trispecific T-cell engager (TriTE), consists of a CD3-specific single-chain Fv (scFv) flanked by anti-epidermal growth factor receptor (EGFR) and anti-epithelial cell adhesion molecule (EpCAM) single-domain VHH antibodies. The TriTE was well expressed in mammalian and yeast cells, bound the cognate antigens of the three parental antibodies, and enabled the specific cytolysis of EGFR- and/or EpCAM-expressing cancer cells, without inducing T cell activation and cytoxicity against double-negative (EGFR−EpCAM−) cancer cells. Bivalent bispecific targeting of double-positive HCT116 cells by TriTE improved in vitro potency up to 100-fold compared to single-positive cells and significantly prolonged survival in vivo. In addition, it was less efficient at killing single-positive target cells than the corresponding bispecific controls, leading to potentially enhanced tumor specificity. Moreover, dual targeting of two tumor-associated antigens may contribute toward preventing the tumor escape by antigen loss caused by selective pressures from conventional single-targeting T-cell engagers, and may help to overcome antigenic heterogeneity.

We have previously shown that self-initiation improves accuracy in visual Short Term Memory (STM) (Loyola-Navarro et al., 2021). Nevertheless, we don't know if these results are generalizable to Working Memory (WM) tasks, since our previous design has little ecological validity, and thus our results are not easily generalizable. Besides, it is not clear whether this effect is multimodal, or if it is specific to the visual domain. To prove if self-initiation improves STM and WM, we will recruit undergrade students and ask them to engage in two Digit Span experiments with varying initiation styles (i.e. encoding). We designed two initiation styles: Automatic (Auto) and Self-initiated presentation. Auto consisted of the automatic presentation of the digits, 550 ms after a cue. Self-initiated consisted of the presentation of the digits 550 ms after the subject pressed the spacebar. To asses the multimodality of the effect, stimulus will be presented in the visual and auditory modality. Experiment 1 will assess the efects of initiation styles and sensory modality using a Forward Digit Span task, while in Experiment 2 we will use a Backward Digit Span task. The experiments will involve a 2x2 factorial repeated measures design. We will assess the effects of self-initiation and sensory modalities on Accuracy (proportion of correct answers) and Response times using two Generalized Linear Mixed Models per experiment. Pilot data retrieved from 19 undergraduate students shows no effect of initiation style nor modality on Forward Digit Span Accuracy, while an effect of initiation style on Backward Digit Span Acurracy in the visual modality only.

ELECTRIC actuators ,MECHATRONICS ,AUTOMATIC control systems ,INTERACTIVE learning ,LEARNING ability - Abstract
Kinematics is a fundamental topic in engineering, robotics, mechatronics, and control systems and significantly resolves some of these fields' most pressing issues. It is essential to assess the balance between a topic's theoretical framework and its empirical validation to succeed in engineering. Educational tools have gained significant attention for their ability to enhance the learning experience by providing the hands-on experiences necessary to assess theoretical frameworks and empirical validations. This paper presents a system incorporating state-of-the-art features, including a fuzzy controller enabling precise control of a linear actuator and a USB camera, to provide an interactive experience. The USB camera captures the position of the actuator, providing real-time visual feedback and allowing the students to validate their theoretical understanding through practical experiments. Precision, accuracy, resolution, and the implementation of the fuzzy controller are measured to evaluate the whole system's performance. The design, implementation, and control of our educational electrical linear actuator for teaching kinematics concepts contribute to a practical educational tool and advance interactive learning approaches in the field. Cardiology and Cardiovascular Medicine - Abstract
The 2-Minute Step Test (2MST) has been presented as an alternative to the 6-Minute Walk Test (6MWT) based on the association between the two tests in older adults; however, some authors propose that it should not be a substitute but rather a complement to the latter in the fitness evaluation. Specifically, in coronary disease, despite the potential and clinical utility of 2MST, the relationship of both tests in this population is unknown. This study aimed to determine the relationship between 6MWT and 2MST and to explore the relationship of biodemographic factors for both tests in subjects with treated coronary artery disease. For this, the 6MWT and the 2MST were applied to patients with coronary artery disease treated in 6 hospitals in Chile between May 2019 and February 2020. Additionally, lower limb strength was assessed by a chair-stand test, grip strength was assessed by a dynamometer, and physical measurements were applied. In total, 163 participants underwent both tests (average age = 58.7 ± 9.8 years; 73.6% men; 64.4% revascularized by angioplasty; 28.2% revascularized by surgery, and 7.4% treated by drugs or thrombolysis). Heart rate was higher at the end of the 6MWT, while the perception of effort was greater at the end of the 2MST. There was a weak positive correlation between the 6MWT and the 2MST in subjects with treated coronary disease (r = 0.28, p = 0.0003). While age (r = –0.27), weight (r = 0.25), height (r = 0.49), and strength of both lower limbs (r = 0.41) and grip strength (r = 0.53) correlated weakly or moderately to the covered distance in 6MWT, the number of steps by the 2MST correlated only weakly to height (r = 0.23), lower limb strength (r = 0.34), and grip strength (r = 0.34). Age, weight, height, lower limb strength, and grip strength would explain better the meters walked in the 6MWT than the steps achieved in the 2MST. With these findings, we can conclude that, in patients with treated coronary artery disease, it does not seem advisable to replace 6MWT with 2MST when it is possible to do so. Additionally, the 2MST may provide additional information in the fitness evaluation. However, the usefulness of 2MST in this population needs to be further studied.

Agricultural greenhouses must accurately predict environmental factors to ensure optimal crop growth and energy management efficiency. However, the existing predictors have limitations when dealing with dynamic, non-linear, and massive temporal data. This study proposes four supervised learning techniques focused on linear regression (LR) and Support Vector Regression (SVR) to predict the internal temperature of a greenhouse. A meteorological station is installed in the greenhouse to collect internal data (temperature, humidity, and dew point) and external data (temperature, humidity, and solar radiation). The data comprises a one year, and is divided into seasons for better analysis and modeling of the internal temperature. The study involves sixteen experiments corresponding to the four models and the four seasons and evaluating the models' performance using R 2 , RMSE, MAE, and MAPE metrics, considering an acceptability interval of ±2 ° C. The results show that LR models had difficulty maintaining the acceptability interval, while the SVR models adapted to temperature outliers, presenting the highest forecast accuracy among the proposed algorithms. The work consists in the analysis of the consumption of traditional television and YouTube digital platform. The main objective is to confirm if the substitute for television is YouTube. For this reason, two studies have been carried out. On the one hand, one about the different content on television and on YouTube, and, on the other hand, another comparing the different audiences in both audiovisual productions. It is shown that the rise of digital platforms harms television audiences. However, a third study has been carried out to prove if YT and TV can become allies. In this case, the Top Gamers Academy television program has been the topic of the study obtaining as a result: a failed alliance.

This project consists of the creation and development of the Kawati brand, which is dedicated to the sale of personalized items by hand, focusing for the moment on bags and shoes, but with a view to expanding the product offering to many others, all of them handmade and personalized. To do this, a market study has been carried out on the sector in which it is intended to influence, the competition and the audience you want to attract, in order to make the right decisions regarding brand identity, branding and all corporate visual identity. Each of the details that make up a brand have been carefully chosen, effectively and consistently so that they convey the same values, thus reinforcing the brand identity from all possible points to improve its inclusion in the market. Once the brand is created and developed, decisions have been made across the entire digital framework, The profiles have been created in the different social networks in which the Kawati public moves and, in addition, content has been generated for them.

In this work, we show that TGF‐β produced by colorectal cancer cells activates pericytes that in turn promote their tumorigenic properties through the release of soluble factors, including IGFBP‐3, with a prominent role in cancer cell migration and invasion. Moreover, co‐implantation of colorectal cancer cells and pericytes in immunodeficient mice increased tumor growth., The crosstalk between cancer cells and the tumor microenvironment has been implicated in cancer progression and metastasis. Fibroblasts and immune cells are widely known to be attracted to and modified by cancer cells. However, the role of pericytes in the tumor microenvironment beyond endothelium stabilization is poorly understood. Here, we report that pericytes promoted colorectal cancer (CRC) cell proliferation, migration, invasion, stemness, and chemoresistance in vitro, as well as tumor growth in a xenograft CRC model. We demonstrate that coculture with human CRC cells induced broad transcriptomic changes in pericytes, mostly associated with TGF‐β receptor activation. The prognostic value of a TGF‐β response signature in pericytes was analyzed in CRC patient data sets. This signature was found to be a good predictor of CRC relapse. Moreover, in response to stimulation by CRC cells, pericytes expressed high levels of TGF‐β1, initiating an autocrine activation loop. Investigation of secreted mediators and underlying molecular mechanisms revealed that IGFBP‐3 is a key paracrine factor from activated pericytes affecting CRC cell migration and invasion. In summary, we demonstrate that the interplay between pericytes and CRC cells triggers a vicious cycle that stimulates pericyte cytokine secretion, in turn increasing CRC cell tumorigenic properties. Overall, we provide another example of how cancer cells can manipulate the tumor microenvironment.

- Abstract
What does it say about the industry?, donde se muestra las relaciones entre sectores y colores.
