Background and Purpose Chronic ethanol abuse and haemorrhagic shock are major causes of global mortality and, separately, induce profound hepato- and immune-toxic effects via activation of NF-κ B. Here, we assessed the effects of chronic ethanol intake upon the pathophysiological derangements after haemorrhagic shock with subsequent resuscitation ( H/ R), with particular attention to the contribution of NF-κ B. Experimental Approach Transgenic NF-κBEGFP mice, expressing the enhanced green fluorescent protein ( EGFP) under the transcriptional control of NF-κ B cis-elements were fed a Lieber- De Carli diet containing ethanol (EtOH-diet) or an isocaloric control diet for 4 weeks and were then pairwise subjected to H/ R. Liver tissues and peripheral blood were sampled at 2 or 24 h after H/R. Cytokines in blood and tissue and leukocyte activation (as CD11b expression) were measured, along with EGFP as a marker of NF-κB activation. Key Results The EtOH-diet increased mortality at 24 h after H/ R and elevated liver injury, associated with an up-regulation of NF-κ B-dependent genes and IL-6 release; it also increased production of NF-κ B-driven intercellular adhesion molecule 1 ( ICAM-1) and EGFP in liver tissue. At 2h after the H/ R procedure in ethanol-fed mice we observed the highest proportion of NF-κ B activated non-parenchymal cells and an NF-κ B-dependent increase in polymorphonuclear leukocyte CD11b expression. Conclusions and Implications The EtOH-diet exacerbated liver injury after H/ R, accompanying an overwhelming hepatic and systemic immune response. Our findings contribute to evidence implicating NF-κ B as a key player in the orchestration of the immune response in haemorrhagic shock patients with a history of chronic ethanol abuse. To mitigate climate change, the share of renewable energies in power production needs to be increased. Renewables introduce new challenges to power grids regarding the dynamic stability due to decentralization, reduced inertia, and volatility in production. Since dynamic stability simulations are intractable and exceedingly expensive for large grids, graph neural networks (GNNs) are a promising method to reduce the computational effort of analyzing the dynamic stability of power grids. As a testbed for GNN models, we generate new, large datasets of dynamic stability of synthetic power grids and provide them as an open-source resource to the research community. We find that GNNs are surprisingly effective at predicting the highly non-linear targets from topological information only. For the first time, performance that is suitable for practical use cases is achieved. Furthermore, we demonstrate the ability of these models to accurately identify particular vulnerable nodes in power grids, so-called troublemakers. Last, we find that GNNs trained on small grids generate accurate predictions on a large synthetic model of the Texan power grid, which illustrates the potential for real-world applications., (© 2023 Author(s). The intentional deformation of human skulls in the living being was one of the most curious rituals performed in historical and ancient times. It is thought that these practices cause chronic elevated intracranial pressure and subsequent symptoms of cognitive impairment. In this report, we examine such an artificially deformed skull dating from the sixteenth century that in addition shows a fronto-ethmoidal encephalocele. However, although the mild encephalocele was already manifest at birth and deformation practices were performed over years, the encephalocele did not progress into a more severe status. We conclude that the intentional deformation of skulls does not lead to chronic elevated intracranial pressure and mental retardation.

In this prospective study we evaluated the somatostatin receptor scintigraphy in the initial staging of 19 patients suffering from Hodgkin's (HD) and non-Hodgkin's lymphomas (NHL) and in the restaging of 16 patients. Scintigraphy was compared to the results of the methods applied for adequate staging of lymphoma patients in the large multicentre trials. Planar imaging and SPECT was performed after intravenous injection of 110 or 220 MBq of 111In-pentetreotide. The patient-based analysis yielded an overall sensitivity of 88%, contrasting lesion-based sensitivities of 57%, 35%, and 43% in HD, low-grade NHL and high-grade NHL, respectively. The best results were obtained in the head-and-neck region and the worst in the abdomen (sensitivities of 61% and 24%, respectively). Bone marrow infiltration was visible in 1/12 cases only. There was no significant difference between the outcomes of patients in the initial staging and restaging and no influence of the amount of injected radiopharmaceutical on the results. In terms of the Ann-Arbor classification, 10/35 patients were concordant whereas 22 were understaged and 3 overstaged scintigraphically. In conclusion, somatostatin receptor scintigraphy is not useful in the initial staging or restaging of malignant lymphomas, especially NHL, due to low lesion detection rates most probably because of low receptor densities. In addition, intraindividual heterogeneity of somatostatin receptor expression has to be considered.

It was the aim of the present study to examine whether 111In-pentetreotide, a somatostatin analogue with predominantly renal excretion, is a suitable receptor agonist for scintigraphic imaging of endocrine gastro-entero-pancreatic (GEP) tumors, and to evaluate the contribution of the usual imaging times 4 and 24 h p.i. In 36 patients, planar scintigrams obtained 4 h, and 24 h after i.v. injection of 111 or 222 MBq 111In-pentetreotide were compared to the results of other imaging procedures and of surgery. Single photon emission computed tomography (SPECT) was also performed 24 h p.i. Positive scintigraphies were obtained in 32 out of 36 patients (18/19 patients with carcinoid syndrome, 8/9 with non hormone-producing endocrine GEP tumors, 2/4 with gastrinomas, 1/1 with glucagonoma, 1/1 with a VIPoma, 2/2 with paragangliomas). In 9 patients tumor manifestations previously not detected by conventional imaging procedures were disclosed by 111In-pentetreotide scintigraphy. 24-h images yielded significantly more true positive findings than 4-h images. In 4 patients liver metastases missed on planar scans were detected by SPECT. A discrepancy between patient-based and organ-based analysis of the results was encountered thus indicating a possible intraindividual heterogeneity in somatostatin receptor expression. In conclusion, 111In-pentetreotide is a suitable somatostatin analogue for scintigraphic in vivo demonstration of somatostatin receptors and for imaging of most tumor manifestations in patients with endocrine GEP tumors. Further studies will have to evaluate whether or not a positive receptor scintigraphy predicts response to treatment with long-acting somatostatin analogues.
