Abstract: International audience; A science is defined by a set of encyclopedic knowledge related to facts or phenomena following rules or evidenced by experimentally-driven observations. Computer Science and in particular computer networks is a relatively new scientific domain maturing over years and adopting the best practices inherited from more fundamental disciplines. The design of past, present and future networking components and architectures have been assisted, among other methods, by experimentally-driven research and in particular by the deployment of test platforms, usually named as testbeds. However, often experimentally-driven networking research used scattered methodologies, based on ad-hoc, small-sized testbeds, producing hardly repeatable results. We believe that computer networks needs to adopt a more structured methodology, supported by appropriate instruments, to produce credible experimental results supporting radical and incremental innovations. This paper reports lessons learned from the design and operation of test platforms for the scientific community dealing with digital infrastructures. We introduce the SLICES initiative as the outcome of several years of evolution of the concept of a networking test platform transformed into a scientific instrument. We address the challenges, requirements and opportunities that our community is facing to manage the full research-life cycle necessary to support a scientific methodology.

Abstract: Developments of future scenarios of Antarctic ecosystems are still in their infancy, whilst predictions of the physical environment are recognized as being of global relevance and corresponding models are under continuous development. However, in the context of environmental change simulations of the future of the Antarctic biosphere are increasingly demanded by decision makers and the public, and are of fundamental scientific interest. This paper briefly reviews existing predictive models applied to Antarctic ecosystems before providing a conceptual framework for the further development of spatially and temporally explicit ecosystem models. The concept suggests how to improve approaches to relating species’ habitat description to the physical environment, for which a case study on sea urchins is presented. In addition, the concept integrates existing and new ideas to consider dynamic components, particularly information on the natural history of key species, from physiological experiments and biomolecular analyses. Thereby, we identify and critically discuss gaps in knowledge and methodological limitations. These refer to process understanding of biological complexity, the need for high spatial resolution oceanographic data from the entire water column, and the use of data from biomolecular analyses in support of such ecological approaches. Our goal is to motivate the research community to contribute data and knowledge to a holistic, Antarctic-specific, macroecological framework. Abstract: Fragile X syndrome (FXS) is the first cause of inherited intellectual disability, due to the silencing of the X-linked Fragile X Mental Retardation 1 gene encoding the RNA-binding protein FMRP. While extensive studies have focused on the cellular and molecular basis of FXS, neither human Fragile X patients nor the mouse model of FXS—the Fmr1-null mouse—have been profiled systematically at the metabolic and neurochemical level to provide a complementary perspective on the current, yet scattered, knowledge of FXS. Using proton high-resolution magic angle spinning nuclear magnetic resonance (1H HR-MAS NMR)-based metabolic profiling, we have identified a metabolic signature and biomarkers associated with FXS in various brain regions of Fmr1-deficient mice. Our study highlights for the first time that Fmr1 gene inactivation has profound, albeit coordinated consequences in brain metabolism leading to alterations in: (1) neurotransmitter levels, (2) osmoregulation, (3) energy metabolism, and (4) oxidative stress response. To functionally connect Fmr1-deficiency to its metabolic biomarkers, we derived a functional interaction network based on the existing knowledge (literature and databases) and show that the FXS metabolic response is initiated by distinct mRNA targets and proteins interacting with FMRP, and then relayed by numerous regulatory proteins. This novel “integrated metabolome and interactome mapping” (iMIM) approach advantageously unifies novel metabolic findings with previously unrelated knowledge and highlights the contribution of novel cellular pathways to the pathophysiology of FXS. These metabolomic and integrative systems biology strategies will contribute to the development of potential drug targets and novel therapeutic interventions, which will eventually benefit FXS patients.

Abstract: International audience; Like all retroviruses, human immunodeficiency virus type 1 (HIV-1) undergoes reverse transcription during its replication cycle. The cellular cofactors potentially involved in this process still remain to be identified. We show here that A-kinase anchoring protein 149 (AKAP149) interacts with HIV-1 reverse transcriptase (RT) in both the yeast two-hybrid system and human cells. The AKAP149 binding site has been mapped to the RNase H domain of HIV-1 RT. AKAP149 silencing by RNA interference in HIV-1-infected cells inhibited viral replication at the reverse transcription step. We selected single-point mutants of RT defective for AKAP149 binding and demonstrated that mutant G462R, despite retaining significant intrinsic RT activity in vitro, failed to carry out HIV-1 reverse transcription correctly in infected cells. This suggests that the interaction between RT and AKAP149 in infected cells may play an important role in HIV-1 reverse transcription.

Abstract: International audience; A series of chiral cyclometalated iridium complexes of the type [Ir(C boolean AND N)(2)(C boolean AND C:)], {(C boolean AND N) = ppy (2); dfppy (3)} featuring a naphthalimide N-heterocyclic carbene ligand (C boolean AND C:) = (Naphthalimide-NHC) are described and fully characterized. The racemic complexes rac-2 and rac-3 were resolved via chiral column chromatography techniques into their corresponding enantiopure species Delta-2, Lambda-2, Delta-3, Lambda-3 as confirmed by their CD curves. This unique class of molecules containing organic and inorganic chromophores might be used as a platform to probe the stereochemical effect on the photophysical properties. Vibrational circular dichroism (VCD) was used as an important tool to assign successfully the stereochemistry of the enantiomers. TD-DFT calculations are also advanced to support the experimental studies and to rationalize the observed results.

Abstract: Sickle cell disease and β-thalassemia affect the production of the adult β-hemoglobin chain. The clinical severity is lessened by mutations that cause fetal γ-globin expression in adult life (i.e., the hereditary persistence of fetal hemoglobin). Mutations clustering ~200 nucleotides upstream of the HBG transcriptional start sites either reduce binding of the LRF repressor or recruit the KLF1 activator. Here, we use base editing to generate a variety of mutations in the −200 region of the HBG promoters, including potent combinations of four to eight γ-globin-inducing mutations. Editing of patient hematopoietic stem/progenitor cells is safe, leads to fetal hemoglobin reactivation and rescues the pathological phenotype. Creation of a KLF1 activator binding site is the most potent strategy – even in long-term repopulating hematopoietic stem/progenitor cells. Compared with a Cas9-nuclease approach, base editing avoids the generation of insertions, deletions and large genomic rearrangements and results in higher γ-globin levels. Our results demonstrate that base editing of HBG promoters is a safe, universal strategy for treating β-hemoglobinopathies.

Abstract: This article considers the evolution of brain architectures for predictive processing. We argue that brain mechanisms for predictive perception and action are not late evolutionary additions of advanced creatures like us. Rather, they emerged gradually from simpler predictive loops (e.g. autonomic and motor reflexes) that were a legacy from our earlier evolutionary ancestors—and were key to solving their fundamental problems of adaptive regulation. We characterize simpler-to-more-complex brains formally, in terms ofgenerative modelsthat include predictive loops of increasing hierarchical breadth and depth. These may start from a simple homeostatic motif and be elaborated during evolution in four main ways: these include themultimodal expansionof predictive control into an allostatic loop; itsduplicationto form multiple sensorimotor loops that expand an animal's behavioural repertoire; and the gradual endowment of generative models withhierarchical depth(to deal with aspects of the world that unfold at different spatial scales) andtemporal depth(to select plans in a future-oriented manner). In turn, these elaborations underwrite the solution to biological regulation problems faced by increasingly sophisticated animals. Our proposal aligns neuroscientific theorising—about predictive processing—with evolutionary and comparative data on brain architectures in different animal species.This article is part of the theme issue ‘Systems neuroscience through the lens of evolutionary theory’.

Abstract: Local interactions and communication are key features in swarm robotics, but they are most often fixed at design time, limiting flexibility and causing a stiff and inefficient response to changing environments. Motivated by the need for higher adaptation abilities, we propose that information about emergent collective structures should percolate onto the individual behavior, modifying it in a way that determines suitable responses in the face of new working conditions and organizational challenges. Indeed, complex societies are driven by an evolving set of individual and social norms subject to cultural propagation, which contribute to determining the individual behaviors. We leverage ideas from the evolution of natural language – an undoubtedly efficient cultural trait – and exploit the resulting social dynamics to select and propagate microscopic behavioral parameters that adapt continuously to macroscopic conditions, which in turn affect the agents’ communication topography, and, therefore, feed back onto the social dynamics. This concept is demonstrated on a self-organized aggregation behavior, which is a building block for most swarm robotics behaviors and a striking example of how collective dynamics are sensitive to experimental parameters. By means of experiments with simulated and real robots, we show that the cultural evolution of aggregation rules outperforms conventional approaches in terms of adaptivity to multiple experimental settings.

Abstract: Synthetic vitreous silica is currently the preferred material for the production of optical fibres because of the several excellent properties of this glass, e.g. high transmission in the visible and IR domains, high mechanical strength, chemical durability, and ease of doping with various materials. For instance, fiber lasers and amplifiers exploit the light amplification properties provided by rare-earth ions employed as dopants in the core of silica-based optical fibers. The structure and composition of the nearest neighbor shell surrounding rare-earth ions in silica-based optical fibers and amplifiers have been intensively debated in the last decade. To reduce aggregation effects between rare-earth ions, co-dopants such as phosphorus and aluminium are added as structural modifiers; phosphorus-doping, in particular, has proved to be very efficient in dissolving rare-earth ions. In this work, we provide further insights concerning the embedding of P atoms into the silica network, which may be relevant for explaining the ease of formation of a phosphorus pentoxide nearest-neighbor shell around a rare-earth dopant. In particular, by means of first-principles calculations, we discuss alternative models for an irradiation (UV, x–, γ-rays) induced paramagnetic center, i.e. the so called room-temperature phosphorus-oxygen-hole center, and its precursors. We report that the most likely precursor of a room-temperature phosphorus-oxygen-hole center comprises of a micro-cluster of a few (at least two) neighboring phosphate tetrahedra, and correspondingly that the occurrence of isolated [(O-)2P(=O)2]− units is unlikely even at low P-doping concentrations. In fact, this work predicts that the symmetric stretching of P=O bonds in isolated [(O-)2P(=O)2]− units appears as a Raman band at a frequency of ~1110 cm−1, and only by including at least another corner-sharing phosphate tetrahedron, it is shown to shift to higher frequencies (up to ~40 cm−1) due to the shortening of P=O bonds, thereby leading to an improved agreement with the observed Raman band located at ~1145 cm−1.

Abstract: International audience; Existing European observation tools and services have the potential to take advantage of cutting-edge technologies to obtain a wide range of data at a much higher spatial resolution and temporal regularity and duration. The EU-funded NAUTILOS project will develop a new generation of sensors and samplers for physical, chemical, and biological essential ocean variables in addition to micro-and nano-plastics. The project will improve our understanding of environmental variations and anthropogenic impacts connected with aquaculture, fi sheries, and marine litter. The project will integrate recently advanced marine technologies into different observing platforms and deploy them through innovative and cost-effective methods in a wide range of key environmental settings and EU policy-related applications. The project aims to complement and expand existing European observation instruments and services and further enable and democratise the monitoring of the marine environment for both traditional and non-traditional data users.; Les outils et services d'observation européens existants ont le potentiel de tirer parti des technologies de pointe pour obtenir un large éventail de données à une résolution spatiale et une régularité temporelle et une durée beaucoup plus élevées. Abstract: International audience; Oxytocin is an important regulator of the social brain. In some animal models of autism, notably in Magel2 tm1.1Mus -deficient mice, peripheral administration of oxytocin in infancy improves social behaviors until adulthood. However, neither the mechanisms responsible for social deficits nor the mechanisms by which such oxytocin administration has long-term effects are known. Here, we aimed to clarify these oxytocin-dependent mechanisms, focusing on social memory performance. Using in situ hybridization (RNAscope), we have established that Magel2 and oxytocin receptor are co-expressed in the dentate gyrus and CA2/CA3 hippocampal regions involved in the circuitry underlying social memory. Then, we have shown that Magel2 tm1.1Mus - deficient mice, evaluated in a three-chamber test, present a deficit in social memory. Next, in hippocampus, we conducted neuroanatomical and functional studies using immunostaining, oxytocin-binding experiments, ex vivo electrophysiological recordings, calcium imaging and biochemical studies. We demonstrated: an increase of the GABAergic activity of CA3-pyramidal cells associated with an increase in the quantity of oxytocin receptors and of somatostatin interneurons in both DG and CA2/CA3 regions. We also revealed a delay in the GABAergic development sequence in Magel2 tm1.1Mus -deficient pups, linked to phosphorylation modifications of KCC2. Above all, we demonstrated the positive effects of subcutaneous administration of oxytocin in the mutant neonates, restoring hippocampal alterations and social memory at adulthood. Although clinical trials are debated, this study highlights the mechanisms by which peripheral oxytocin administration in neonates impacts the brain and demonstrates the therapeutic value of oxytocin to treat infants with autism spectrum disorders.

Abstract: Current global change substantially threatens pollinators, which directly impacts the pollination services underpinning the stability, structure and functioning of ecosystems. Amongst these threats, many synergistic drivers, such as habitat destruction and fragmentation, increasing use of agrochemicals, decreasing resource diversity, as well as climate change, are known to affect wild and managed bees. Therefore, reliable indicators for pollinator sensitivity to such threats are needed. Biological traits, such as phenotype (e.g. shape, size and asymmetry) and storage reserves (e.g. fat body size), are important pollinator traits linked to reproductive success, immunity, resilience and foraging efficiency and, therefore, could serve as valuable markers of bee health and pollination service potential. This data paper contains an extensive dataset of wing morphology and fat body content for the European honeybee (Apis mellifera) and the buff-tailed bumblebee (Bombus terrestris) sampled at 128 sites across eight European countries in landscape gradients dominated by two major bee-pollinated crops (apple and oilseed rape), before and after focal crop bloom and potential pesticide exposure. The dataset also includes environmental metrics of each sampling site, namely landscape structure and pesticide use. The data offer the opportunity to test whether variation in the phenotype and fat bodies of bees is structured by environmental factors and drivers of global change. Overall, the dataset provides valuable information to identify which environmental threats predominantly contribute to the modification of these traits.

Abstract: Riverine systems act as converging pathways for discarded litter within drainage basins, becoming key elements in gauging the transfer of mismanaged waste into the ocean. However, riverine litter data are scarce and biased towards microplastics, generally lacking information about larger items. Based on the first ever database of riverine floating macrolitter across Europe, we have estimated that between 307 and 925 million litter items are released annually from Europe into the ocean. The plastic fraction represented 82% of the observed litter, mainly fragments and single-use items (that is, bottles, packaging and bags). Our modelled estimates show that a major portion of the total litter loading is routed through small-sized drainage basins (, We acknowledge the additional members (coordinators and observers) of RiLON, listed in Supplementary Data 5, for their support in the field data collection and feedback during the data quality control. Abstract: International audience; Cardiometabolic disorders are among the leading causes of mortality in the human population. Dietary polyphenols exert beneficial effects on cardiometabolic health in humans. Molecular mechanisms, however, are not completely understood. Aiming to conduct in-depth integrative bioinformatic analyses to elucidate molecular mechanisms underlying the protective effects of polyphenols on cardiometabolic health, we first conducted a systematic literature search to identify human intervention studies with polyphenols that demonstrate improvement of cardiometabolic risk factors in parallel with significant nutrigenomic effects. Applying the predefined inclusion criteria, we identified 58 differentially expressed genes at mRNA level and 5 miRNAs, analyzed in peripheral blood cells with RT-PCR methods. Subsequent integrative bioinformatic analyses demonstrated that polyphenols modulate genes that are mainly involved in the processes such as inflammation, lipid metabolism, and endothelial function. We also identified 37 transcription factors that are involved in the regulation of polyphenol modulated genes, including RELA/NFKB1, STAT1, JUN, or SIRT1. Integrative bioinformatic analysis of mRNA and miRNA-target pathways demonstrated several common enriched pathways that include MAPK signaling pathway, TNF signaling pathway, PI3K-Akt signaling pathway, focal adhesion, or PPAR signaling pathway. These bioinformatic analyses represent a valuable source of information for the identification of molecular mechanisms underlying the beneficial health effects of polyphenols and potential target genes for future nutrigenetic studies.

Abstract: International audience; The paper presents some recent trends in multi-fidelity digital modelling for marine engineering applications. Digital modelling is achieved by machine learning methods, namely multi-fidelity surrogate models, trained by computational fluid dynamics (CFD). Adaptative approaches are discussed for radial basis functions and Gaussian process models. Simulation-based design optimisation problems are presented to discuss the use and effects of different adaptivity concepts: (1) adaptive refinement of the computational-domain discretization in CFD; (2) adaptive sampling of the design/operational space; (3) adaptive selection of the fidelity used for the surrogate model training in a multi-fidelity environment; (4) adaptivity of the models to noise. Model adaptation allows for the efficient training of machine learning models, reducing the computational cost associated to building the training sets and improving the overall accuracy of the digital representation.

Abstract: In the context of ionic transport in solids, the variation of a migration barrier height under electric fields is traditionally assumed to be equal to the classical electric work of a point charge that carries the transport charge. However, how reliable is this phenomenological model and how does it fare with respect to Modern Theory of Polarization? In this work, we show that such a classical picture does not hold in general as collective dipole effects may be critical. Such effects are unraveled by an appropriate polarization decomposition and by an expression that we derive, which defines the equivalent polarization-work charge. The equivalent polarization-work charge is not equal neither to the transported charge, nor to the Born effective charge of the migrating atom alone, but it is defined by the total polarization change at the transition state. Our findings are illustrated by oxygen charged defects in MgO and in SiO2., Ionic transport in solids is important for applications including rapid access memories and ion-based batteries. Here, the authors introduce polarization work contributions from the migrating ion and its environment, demonstrating the representative profile is given by a unique charge parameter.

Abstract: International audience; An adaptive-fidelity approach to metamodeling from noisy data is presented for design-space exploration and design optimization. Computational fluid dynamics (CFD) simulations with different numerical accuracy (spatial discretization) provides metamodel training sets affected by unavoidable numerical noise. The-fidelity approximation is built by an additive correction of a low-fidelity metamodel with metamodels of differences (errors) between higher-fidelity levels whose hierarchy needs to be provided. The approach encompasses two core metamodeling techniques, namely: i) stochastic radial-basis functions (SRBF) and ii) Gaussian process (GP). The adaptivity stems from the sequential training procedure and the auto-tuning capabilities of the metamodels. The method is demonstrated for an analytical test problem and a CFD-based optimization of a NACA airfoil, where the fidelity levels are defined by an adaptive grid refinement technique of a Reynolds-averaged Navier-Stokes (RANS) solver. The paper discusses: i) the effect of using more than two fidelity levels; ii) the use of least squares regression as opposed to exact interpolation; iii) the comparison between SRBF and GP; and iv) the use of two sampling approaches for GP. Results show that in presence of noise, the use of more than two fidelity levels improves the model accuracy with a significant reduction of the number of high-fidelity evaluations. Both least squares SRBF and GP provide promising results in dealing with noisy data.

Abstract: Background: Several automatic tools have been implemented for semi-quantitative assessment of brain [ 18 ]F-FDG-PET. Objective: We aimed to head-to-head compare the diagnostic performance among three statistical parametric mapping (SPM)-based approaches, another voxel-based tool (i.e., PALZ), and a volumetric region of interest (VROI-SVM)-based approach, in distinguishing patients with prodromal Alzheimer's disease (pAD) from controls. Methods: Sixty-two pAD patients (MMSE score = 27.0±1.6) and one hundred-nine healthy subjects (CTR) (MMSE score = 29.2±1.2) were enrolled in five centers of the European Alzheimer's Disease Consortium. The three SPM-based methods, based on different rationales, included 1) a cluster identified through the correlation analysis between [ 18 ]F-FDG-PET and a verbal memory test (VROI-1), 2) a VROI derived from the comparison between pAD and CTR (VROI-2), and 3) visual analysis of individual maps obtained by the comparison between each subject and CTR (SPM-Maps). The VROI-SVM approach was based on 6 VROI plus 6 VROI asymmetry values derived from the pAD versus CTR comparison thanks to support vector machine (SVM). Results: The areas under the ROC curves between pAD and CTR were 0.84 for VROI-1, 0.83 for VROI-2, 0.79 for SPM maps, 0.87 for PALZ, and 0.95 for VROI-SVM. Pairwise comparisons of Youden index did not show statistically significant differences in diagnostic performance between VROI-1, VROI-2, SPM-Maps, and PALZ score whereas VROI-SVM performed significantly (p < 0.005) better than any of the other methods. Conclusion: The study confirms the good accuracy of [ 18 ]F-FDG-PET in discriminating healthy subjects from pAD and highlights that a non-linear, automatic VROI classifier based on SVM performs better than the voxel-based methods.

Abstract: Among the common vacancy-related point defects in silicon, the E center is one of the most prominent due to its degrading effect in silicon-based technology. Even though it has been the subject of extensive experimental and theoretical studies, a comprehensive theoretical model capable of reproducing the experimental evidence for all three dopants (P, As, and Sb) is still missing. Guided by a Jahn-Teller model, we are able to reproduce the absorption bands and the transition probability between equivalent geometries of the defect at low temperatures by including many-body-perturbation corrections based on the GW approximation on top of the density functional theory. At higher temperatures, vacancies become mobile centers, enabling the reorientation of the whole defect and contributing to the dopant diffusion. The underlying mechanisms of the vacancy-mediated dopant diffusion are revisited, characterizing the activation energies of such technologically relevant processes and obtaining quantitative results in good agreement with experiment.

Abstract: International audience; Integrated forest management (IFM) can help reconcile critical trade-offs between goals in forest management, such as nature conservation and biomass production. The challenge of IFM is dealing with these trade-offs at the level of practical forest management, such as striving for compromises between biomass extraction and habitat retention. This paper reviews some of the driving factors that influence the integration of nature conservation into forest management. The review was conducted in three steps-a literature review, an expert workshop and an expert-based cooperative analysis. Of 38 driving factors identified, three were prioritised by more of the participants than any of the others: two are socio-cultural factors, identity (how people identify with forest) as well as outreach and education, and one is economic-competitiveness in forest value chains. These driving factors correspond to what are considered in the literature as enablers for IFM. The results reveal that targeted, group-oriented, adaptive and innovative policy designs are needed to integrate nature conservation into forest management. Further, the results reveal that a "one-size-fits-all" governance approach would be ineffective, implying that policy instruments need to consider contextually specific driving factors. Understanding the main driving factors and their overall directions can help to better manage trade-offs between biodiversity conservation and biomass production in European forests.

Abstract: Flavanol intake positively influences several cardiometabolic risk factors in humans. However, the specific molecular mechanisms of action of flavanols, in terms of gene regulation, in the cell types relevant to cardiometabolic disease have never been systematically addressed. On this basis, we conducted a systematic literature review and a comprehensive bioinformatic analysis of genes whose expression is affected by flavanols in cells defining cardiometabolic health: hepatocytes, adipocytes, endothelial cells, smooth muscle cells and immune cells. A systematic literature search was performed using the following pre-defined criteria: treatment with pure compounds and metabolites (no extracts) at low concentrations that are close to their plasma concentrations. Differentially expressed genes were analyzed using bioinformatics tools to identify gene ontologies, networks, cellular pathways and interactions, as well as transcriptional and post-transcriptional regulators. The systematic literature search identified 54 differentially expressed genes at the mRNA level in in vitro models of cardiometabolic disease exposed to flavanols and their metabolites. Global bioinformatic analysis revealed that these genes are predominantly involved in inflammation, leukocyte adhesion and transendothelial migration, and lipid metabolism. We observed that, although the investigated cells responded differentially to flavanol exposure, the involvement of anti-inflammatory responses is a common mechanism of flavanol action. We also identified potential transcriptional regulators of gene expression: transcriptional factors, such as GATA2, NFKB1, FOXC1 or PPARG, and post-transcriptional regulators: miRNAs, such as mir-335-5p, let-7b-5p, mir-26b-5p or mir-16-5p. In parallel, we analyzed the nutrigenomic effects of flavanols in intestinal cells and demonstrated their predominant involvement in the metabolism of circulating lipoproteins. In conclusion, the results of this systematic analysis of the nutrigenomic effects of flavanols provide a more comprehensive picture of their molecular mechanisms of action and will support the future setup of genetic studies to pave the way for individualized dietary recommendations., COST (European Cooperation in Science and Technology) Action FA1403.

Abstract: International audience; Sovjan, a pile-dwelling settlement located in southeastern Albania with a stratigraphic sequence extending from the Early Bronze Age to the Early Iron Age attesting continuous habitation, serves as a starting point for a discussion on the applicability of Network Analysis (NA) in the southwestern Balkans. The excavations of several settlements in the Lakes Region divided between modern Albania, Greece, and North Macedonia, together with an extensive survey of the area around the former Lake Maliq where Sovjan is located, permit an analysis of connectivity conducted at different spatial (from micro- to macro-regional) and temporal levels. Important changes in connectivity patterns can be discerned on the basis of technological and stylistic changes observable in the ceramics and the different interregional influences at play in Sovjan. The Early Bronze Age, with its contact networks that extend from the Carpathian region to Greece, is separated from the Late Bronze Age by the hitherto poorly researched Middle Bronze Age. Following this interim period in which networks shrink and change, far-reaching networks arise once again demonstrating influences from the Danube region and, above all, central Macedonia.

Abstract: 1. River ecosystems worldwide are affected by altered flow regimes, and an advanced science and practice of environmental flows has developed to understand and reduce these impacts. But most environmental flows approaches ignore flow intermittency, which is a natural feature of 30% of the global river network length. Ignoring flow intermittency when setting environmental flows in naturally intermittent rivers might lead to deleterious ecological effects. \ud \ud 2. We review evidence of the ecological effects of flow intermittency and provide guidance to incorporate intermittency (non-flow events) into existing methods judged as suitable for application in temporary waterways. \ud \ud 3. To better integrate non-flow events into hydrological methods, we propose a suite of new indicators to be used in the Range of Variability Approach. These indicators reflect dry periods and the unpredictable nature of temporary waterways. We develop a predictability index for protecting those species adapted to temporary conditions. \ud \ud 4. For hydraulic habitat models, we find that mesohabitat methods are particularly effective for describing complex habitat dynamics during dry phases. We present an example of the European eel to show the relationship between discharge and non-flow days and wet area, habitat suitability, and connectivity. \ud \ud 5. We find that existing holistic approaches may be applied to temporary waterways without significant structural alteration to their stepwise frameworks, but new component methods are needed to address flow-related aspects across both flow and non-flow periods of the flow regime. \ud \ud 6. Synthesis and applications. Setting environmental flow requirements for temporary waterways requires modification and enhancement of existing approaches and methodologies, most notably the explicit consideration of non‐flow events and greater integration of specific geomorphic, hydrogeologic, and hydraulic elements. Temporary waterways are among the freshwater ecosystems most vulnerable to alterations in flow regimes, and they are also under great pressure. The methodological modifications recommended in this paper will aid water managers in protecting key components of temporary flow regimes, thereby preserving their unique ecology and associated services.

Abstract: The primary objective of this study is to leverage the integration of surface mapping data derived from optical, radar, and historic topographical studies with archaeological sampling to identify ancient settlement areas in the Northern Nile Delta, Egypt. This study employed the following methods: digitization of topographic maps, band indices techniques on optical data, the creation of a 3D model from SRTM data, and Sentinel-1 interferometric wide swath (IW) analysis. This type of study is particularly relevant to the search for evidence of otherwise hidden ancient settlements. Due to its geographical situation and the fertility of the Nile, Egypt witnessed the autochthonous development of predynastic and dynastic civilizations, as well as an extensive history of external influences due to Greek, Roman, Coptic, Islamic, and Colonial-era interventions. Excavation work at Buto (Tell el-Fara’in) in 2017–18, carried out by the Kafrelsheikh University (KFS) in cooperation with the Ministry of Antiquities, demonstrated that remote sensing data offers considerable promise as a tool for developing regional settlement studies and excavation strategies. This study integrates the mission work in Buto with the satellite imagery in and around the area of the excavation. The results of the initial Buto area research serve as a methodological model to expand the study area to the North Delta with the goal of detecting the extent of the ancient kingdoms of Buto and Sakha. The results of this research include the creation of a composite historical database using ancient references and early topographical maps (1722, 1941, 1950, and 1997), Optical Corona (1965), Landsat MSS (Multispectral Scanner System) (1973, 1978, and 1988), TM (Thematic Mapper) (2005) data, and Radar SRTM (2014) and Sentinel1 (2018 and 2019) data. The data in this study have been analyzed using the ArcMap, Envi, and SNAP software. The results from the current investigation highlight the rapid changes in the land use/land cover in the last century in which many ancient sites were lost due to agriculture and urban development. Three potential settlement areas have been identified with the Sentinel1 Radar data, and have been integrated with the early maps. These discoveries will help develop excavation strategies aimed at elucidating the ancient settlement dynamics and history of the region during the next phase of research.

Abstract: International audience; Limb-girdle muscular dystrophy type 2D (LGMD2D) is a rare autosomal-recessive disease, affecting striated muscle, due to mutation of SGCA, the gene coding for α-sarcoglycan. Nowadays, more than 50 different SGCA missense mutations have been reported. They are supposed to impact folding and trafficking of α-sarcoglycan because the defective polypeptide, although potentially functional, is recognized and disposed of by the quality control of the cell. The secondary reduction of α-sarcoglycan partners, β-, γ- and δ-sarcoglycan, disrupts a key membrane complex that, associated to dystrophin, contributes to assure sarcolemma stability during muscle contraction. The complex deficiency is responsible for muscle wasting and the development of a severe form of dystrophy. Here, we show that the application of small molecules developed to rescue ΔF508-CFTR trafficking, and known as CFTR correctors, also improved the maturation of several α-sarcoglycan mutants that were consequently rescued at the plasma membrane. Remarkably, in myotubes from a patient with LGMD2D, treatment with CFTR correctors induced the proper re-localization of the whole sarcoglycan complex, with a consequent reduction of sarcolemma fragility. Although the mechanism of action of CFTR correctors on defective α-sarcoglycan needs further investigation, this is the first report showing a quantitative and functional recovery of the sarcoglycan-complex in human pathologic samples, upon small molecule treatment. It represents the proof of principle of a pharmacological strategy that acts on the sarcoglycan maturation process and we believe it has a great potential to develop as a cure for most of the patients with LGMD2D.

Abstract: In this paper we study the effect on the electric fields on the formation of bulk Frenkal Pairs and on the migration of oxygen interstitials, I O , and oxygen vacancies, V O , within the framework of Density Functional Theory and Modern Theory of Polarization. At typical OXRRAM field conditions, We show that a significant effect of the electric field is observed only for charged defect. Analyzing the polarization work, we found anomalously high polarization work, for the case of $\mathbf{I}_{\mathcal{O}}^{-2}$, with respect to the classical picture of the electric work of an isolated point charge. This large difference has to be ascribed to collective contributions coming from the environment.

Abstract: The aim of metamodelling in CFD-based automatic shape optimization is to replace expensive CFD computations in the optimization process by evaluations of a cheap surrogate model, created from a limited training set of simulations. Multi-fidelity metamodels [1] make this process even more efficient by basing a part of the metamodel on inexpensive low-fidelity simulations and introducing a correction based on a few high-fidelity simulations. To adaptively define the training sets of the low-fidelity and correction metamodels, different sampling strategies can be used [2]. However, the CFD results on which the metamodels are based contain numerical errors. Especially if new grids are generated for each simulation, the numerical errors for two similar geometries may be different. These differences manifest themselves as numerical noise in the training sets for the metamodels. Adaptive sampling strategies which determine new training points by evaluating the uncertainty of the metamodel, may react to this noise by placing new points in noisy regions, rather than in places where the overall metamodel is unreliable. This deteriorates the metamodel quality. In this paper, we study multifidelity metamodeling approaches which eliminate a part of the numerical noise in the metamodel. For metamodels based on Dynamic Radial Basis Functions [3], this is obtained by introducing less RBF degrees of freedom than training points and computing the RBF fits using least-squares approximation. The number of DoF will be defined adaptively as the training progresses, based on a metric like leave-one-out cross-validation combined with in-the-loop optimization of the metamodel. Applications to the shape optimisation of 2D NACA four-digit airfoils and a destroyer-type vessel will be presented. The CFD simulations are based on a Reynolds-averaged Navier Stokes solver that uses adaptive grid refinement to create the meshes.

Abstract: International audience; Silica-based optical fibers, fiber-based devices and optical fiber sensors are today integrated in a variety of harsh environments associated with radiation constraints. Under irradiation, the macroscopic properties of the optical fibers are modified through three main basic mechanisms: the radiation induced attenuation, the radiation induced emission and the radiation induced refractive index change. Depending on the fiber profile of use, these phenomena differently contribute to the degradation of the fiber performances and then have to be either mitigated for radiation tolerant systems or exploited to design radiation detectors and dosimeters. Considering the strong impact of radiation on key applications such as data transfer or sensing in space, fusion and fission-related facilities or high energy physics facilities, since 1970′s numerous experimental and theoretical studies have been conducted to identify the microscopic origins of these changes. The observed degradation can be explained through the generation by ionization or displacement damages of point defects in the differently doped amorphous glass (SiO2) of the fiber's core and cladding layers. Indeed, the fiber chemical composition (dopants/concentrations) and elaboration processes play an important role. Consequently, identifying the nature, the properties and the generation and bleaching mechanisms of these point defects is mandatory in order to imagine ways to control the fiber radiation behaviors. In this review paper, the responses of the main classes of silica-based optical fibers are presented: radiation tolerant pure-silica core or fluorine doped optical fibers, germanosilicate optical fibers and radiation sensitive phosphosilicate and aluminosilicate optical fibers. Our current knowledge about the nature and optical properties of the point defects related to silica and these main dopants is presented. The efficiency of the known defects to reproduce the transient and steady state radiation induced attenuation between 300 nm and 2 µm wavelength range is discussed. The main parameters, related to the fibers themselves or extrinsic - harsh environments, profile of use - affecting the concentration, growth and decay kinetics of those defects are also reviewed. Finally, the main remaining challenges are discussed, including the increasing needs for accurate and multi-physics modeling tools.

Abstract: International audience; In vertebrates, a remarkable diversity of sex determining mechanisms is observed, ranging from environmental to genetic sex determination (ESD and GSD). In teleosts, sex determination mechanisms include both environmental and genetic regulation and are extremely diverse, changeable, and labile. In addition, many sex-determining genes have been already found in this group of vertebrates showing that SD genes can be extremely variable even between closely related organisms. Hence, teleost fish have emerged as interesting models to study the macroevolution of sex determining (SD) mechanisms within vertebrates. The cavefish Astyanax mexicanus belongs to the Characiform group. Different populations of Astyanax have been described, including pigmented river-dwelling and several depigmented blind cave populations. These populations are still inter-fertile and cave populations are known to have evolved from a surface ancestral population around 100,000 years ago. In addition these Astyanax populations can be also reared under laboratory conditions quite easily and are amendable to genetic manipulations. Altogether this makes surface and cave populations a particularly interesting evolutionary genetic model system for comparative and microevolution studies. We thus initiated studies on the microevolution of SD and SD genes in the cavefish Astyanax mexicanus, and used restriction site-associated DNA (RAD) sequencing and pool-sequencing approaches to identify sex-biased molecular markers in both cave and surface populations. Our first results led to the identification of a candidate master sex-determining gene that potentially triggers SD in both cave and surface populations. Despite this conservation of a strict GSD and the same master sex-determining gene, our results also pin point that SD in Astyanax mexicanus has already evolved between surface and cave populations and suggest a quick evolutionary transition between an old polygenic SD system in surface population towards a more recent and simple monofactorial system in cave population.

Abstract: International audience; A new technology for reducing wheat flour toxicity for celiac disease patients through the in situ formation of gluten-chitosan interlocked self-assembled supramolecular architecture was developed. To have a deeper insight into the microstructure of this new molecular organization and its impact on the dough properties, its small and large deformation rheological properties and the macromolecular features of gluten-chitosan polymers were studied. The reduction of gluten proteins followed by spontaneous oxidation in the presence of the chitosan template in the range of 7.5:1 to 1.3:1 protein to chitosan weight ratio imposed a different reorganization of wheat flour proteins in the polymeric fraction changing conformation from homogeneous spherical molecules to polymer molecules with random-coil conformation. The polymeric fraction increased with decreasing protein to chitosan weight ratio attaining a maximum value at the 1.9:1 ratio. Moreover, the formation of the novel supramolecular architecture at this ratio allowed dough to maintain its ability to form a network after water addition and kneading showing a higher elastic and viscous moduli when compared to the control flour and the other studied formulations. It also presented a significantly higher resistance to extension, didn't inhibit the fermentation process, and retained the original dough ball shape while the dough made with the untreated flour presented a considerable extension during baking. Results show that it is possible to obtain a fully-functional wheat-based product when using a 1.9:1 protein to chitosan weight ratio with a reduced toxicity for celiac patients, opening in this way a new perspective concerning the quest for alternatives of gluten-exclusion diet.

Abstract: International audience; Human LARP4A belongs to a superfamily of RNA binding proteins called La-related proteins (LARPs). Whilst being a positive regulator of protein synthesis and a promoter of mRNA stability, LARP4A also controls cell morphology and motility in human breast and prostate cancer cells. All LARPs share a characteristic RNA binding unit named the La-module, which despite a high level of primary structure conservation exhibits a great versatility in RNA target selection. Human LARP4A La-module is the most divergent compared with other LARPs and its RNA recognition properties have only recently started to be revealed. Given the key role of LARP4A protein in cancer cell biology, we have initiated a complete NMR characterisation of its La-module and here we report the assignment of 1 H, 15 N and 13 C resonances resulting from our studies.

Abstract: International audience; Objective Charcot-Marie-Tooth (CMT) disease 4B1 and 4B2 (CMT4B1/B2) are characterized by recessive inheritance, early onset, severe course, slowed nerve conduction, and myelin outfoldings. CMT4B3 shows a more heterogeneous phenotype. All are associated with myotubularin-related protein (MTMR) mutations. We conducted a multicenter, retrospective study to better characterize CMT4B. Methods We collected clinical and genetic data from CMT4B subjects in 18 centers using a predefined minimal data set including Medical Research Council (MRC) scores of nine muscle pairs and CMT Neuropathy Score. Results There were 50 patients, 21 of whom never reported before, carrying 44 mutations, of which 21 were novel and six representing novel disease associations of known rare variants. CMT4B1 patients had significantly more-severe disease than CMT4B2, with earlier onset, more-frequent motor milestones delay, wheelchair use, and respiratory involvement as well as worse MRC scores and motor CMT Examination Score components despite younger age at examination. Vocal cord involvement was common in both subtypes, whereas glaucoma occurred in CMT4B2 only. Nerve conduction velocities were similarly slowed in both subtypes. Regression analyses showed that disease severity is significantly associated with age in CMT4B1. Slopes are steeper for CMT4B1, indicating faster disease progression. Almost none of the mutations in the MTMR2 and MTMR13 genes, responsible for CMT4B1 and B2, respectively, influence the correlation between disease severity and age, in agreement with the hypothesis of a complete loss of function of MTMR2/13 proteins for such mutations. Interpretation This is the largest CMT4B series ever reported, demonstrating that CMT4B1 is significantly more severe than CMT4B2, and allowing an estimate of prognosis. ANN NEUROL 2019

Abstract: International audience; The paper presents how to efficiently and effectively solve stochastic shape optimization problems by combing Reynolds-averaged Navier-Stokes (RANS) equation solvers with design-space augmented dimensionality reduction (ADR). This study has been conducted within the NATO Science and Technology Organization, Applied Vehicle Technology, Task Group AVT-252 "Stochastic Design Optimization for Naval and Aero Military Vehicles." The application pertains to the robust and the reliability-based robust design optimization of a destroyer hull-form for resistance in calm water and waves and seakeeping performance, under stochastic environmental and operating conditions (speed, sea state, heading). The current work extends previous research by the authors, presented at earlier AIAA conferences [1-3], where only potential flow solvers were used. In the present work, the expected value of the total resistance is reduced respectively by 4.4 and 3% in calm water and waves. An 8% improvement of the seakeeping performance is also achieved. Design-space assessment by ADR is demonstrated to be a viable option in solving the curse of dimensionionality in shape optimization, especially when high-fidelity CPU-expensive solvers are used.

Abstract: Background: While documentation of clinical aspects of General Practice/Family Medicine (GP/FM) is assured by the International Classification of Primary Care (ICPC), there is no taxonomy for the professional aspects (context and management) of GP/FM. Objectives: To present the development, dissemination, applications, and resulting face validity of the Q-Codes taxonomy specifically designed to describe contextual features of GP/FM, proposed as an extension to the ICPC. Development: The Q-Codes taxonomy was developed from Lamberts’ seminal idea for indexing contextual content (1987) by a multi-disciplinary team of knowledge engineers, linguists and general practitioners, through a qualitative and iterative analysis of 1702 abstracts from six GP/FM conferences using Atlas.ti software. A total of 182 concepts, called Q-Codes, representing professional aspects of GP/FM were identified and organized in a taxonomy. Dissemination: The taxonomy is published as an online terminological resource, using semantic web techniques and web ontology language (OWL) (http://www.hetop.eu/Q). Each Q-Code is identified with a unique resource identifier (URI), and provided with preferred terms, and scope notes in ten languages (Portuguese, Spanish, English, French, Dutch, Korean, Vietnamese, Turkish, Georgian, German) and search filters for MEDLINE and web searches. Applications: This taxonomy has already been used to support queries in bibliographic databases (e.g., MEDLINE), to facilitate indexing of grey literature in GP/FM as congress abstracts, master theses, websites and as an educational tool in vocational teaching, Conclusions: The rapidly growing list of practical applications provides face-validity for the usefulness of this freely available new terminological resource.

Abstract: In this paper, the mechanical behavior of austenitic stainless steel 316L with ultrafine grains is investigated using monotonic tensile tests and small punch tests at various temperatures ranging from 20 to 500 °C. Samples were elaborated by powder metallurgy coupled with spark plasma sintering with both unimodal and bimodal grain size distributions. The latter case is obtained by mixing two different powders. The ratio between the largest and lowest grain size was set to 50 and three different volume fractions (i.e. 25, 50 and 75%) of grains with conventional size were investigated. Results show that the bimodal grain size distribution increases the fracture strain to failure in tensile conditions but no effect is observed for small punch tests at room temperature. Abstract: Annals of Clinical and Translational Neurology published by Wiley Periodicals, Inc on behalf of American Neurological Association. Mutations in the sodium-activated potassium channel gene KCNT1 have been associated with nonlesional sleep-related hypermotor epilepsy (SHE). We report the co-occurrence of mild malformation of cortical development (mMCD) and KCNT1 mutations in four patients with SHE. Focal cortical dysplasia type I was neuropathologically diagnosed after epilepsy surgery in three unrelated MRI-negative patients, periventricular nodular heterotopia was detected in one patient by MRI. Our findings suggest that KCNT1 epileptogenicity may result not only from dysregulated excitability by controlling Na+K+ transport, but also from mMCD. Therefore, pathogenic variants in KCNT1 may encompass both lesional and nonlesional epilepsies. Refereed/Peer-reviewed

Abstract: Tractography based on non-invasive diffusion imaging is central to the study of human brain connectivity. To date, the approach has not been systematically validated in ground truth studies. Based on a simulated human brain data set with ground truth tracts, we organized an open international tractography challenge, which resulted in 96 distinct submissions from 20 research groups. Here, we report the encouraging finding that most state-of-the-art algorithms produce tractograms containing 90% of the ground truth bundles (to at least some extent). However, the same tractograms contain many more invalid than valid bundles, and half of these invalid bundles occur systematically across research groups. Taken together, our results demonstrate and confirm fundamental ambiguities inherent in tract reconstruction based on orientation information alone, which need to be considered when interpreting tractography and connectivity results. Our approach provides a novel framework for estimating reliability of tractography and encourages innovation to address its current limitations., Though tractography is widely used, it has not been systematically validated. Here, authors report results from 20 groups showing that many tractography algorithms produce both valid and invalid bundles.

Abstract: International audience; The hereby proposed terminology called "Q-Codes" can be defined as an extension of the International Classification of Primary Care (ICPC-2). It deals with non-clinical concepts that are relevant in General Practice/Family Medicine (GP/FM). This terminology is a good way to put an emphasis on underestimated topics such as Teaching, Patient issues or Ethics. It aims at indexing GP/FM documents such as congress abstracts and theses to get a more comprehensive view about the GP/FM domain. The 182 identified Q-Codes have been very precisely defined by a college of experts (physicians and terminologists) from twelve countries. The result is available on the Health Terminology/Ontology Portal (http://www.hetop.org/Q) and formatted in OWL-2 for further semantic considerations and will be used to index the 2016 WONCA World congress communications.
