The hippocampus is a limbic structure involved in anxiety-like behaviors. We aimed to evaluate the role of the dorsal (DH) and ventral (VH) hippocampus in anxiety-like behaviors in the elevated plus maze (EPM). We inhibited these brain regions using cobalt chloride (CoCl2: 1.0 nmol) microinjections. We also investigated the involvement of corticotropin-releasing factor (CRF) action and protein kinase A (PKA) pathway using intra-DH and intra-VH microinjections of the CRF1 receptor antagonist CP376395 (0, 3.0, or 6.0 nmol) and the PKA inhibitor H-89 (0, 2.5, or 5.0 nmol). The results indicated that intra-VH CoCl2 microinjection increased the percentage of time spent and entries in the open arms. The mice also exhibited fewer stretch attend postures in the protected area and increased percentage of open arm entries. Further, intra-VH injection of 3.0 nmol CP376395 increased time spent in the open arms. Intra-DH injection of 6.0 nmol CP376395 increased the frequency of unprotected head dipping, whereas intra-VH injection of 6 nmol CP376395 increased the frequency of protected head dipping. Intra-VH, but not intra-DH, microinjection of 2.5 nmol H-89 increased the percentages of open arm entries and time spent in the open arms. Microinjection of 2.5 and 5.0 nmol H-89 reduced the frequency of protected head dipping behavior. This study demonstrated that VH modulates anxiety-like behaviors in EPM. Moreover, CRF and the cAMP/PKA pathway seem to modulate these effects.

Essas respostas são moduladas por diferentes neurotransmissores e moduladores neurais em estruturas do sistema límbico, como o hipocampo. Dentre os neurotransmissores envolvidos, tem sido atribuído ao fator liberador de corticotrofina (CRF) um importante papel na modulação de comportamentos defensivos relacionados à ansiedade, uma vez que foi identificada a presença de neurônios CRFérgicos e receptores para CRF em sítios extrahipolâmicos. A ligação com esses receptores acoplados à proteína Gs deflagra uma cascata de reações intracelulares que envolvem a ativação da via do AMPc/PKA. Este trabalho teve como objetivo avaliar o papel do neurotransmissor CRF e da PKA no hipocampo de camundongos nas respostas comportamentais relacionadas à ansiedade quando expostos ao labirinto em cruz elevado (LCE), através dos efeitos da microinjeção de H-89, inibidor de PKA, e de CP 376395 376395, antagonista de CRF1 e ao teste de exposição ao rato (TER), através da expressão nuclear de proteína Fos e efeitos da microinjeção de CP 376395. A partir dos resultados obtidos com a exposição ao LCE, foi possível verificar que o tratamento com CP 376395 intra-hipocampo ventral (HV) elevou o tempo de permanência em braços abertos do LCE na dose 3,0nmol, porém, a dose de 6,0nmol aumentou o número de mergulhos desprotegidos quando injetada no hipocampo dorsal (HD) e o número de mergulhos protegidos quando injetada em HV. Nenhuma das doses promoveu alterações nos comportamentos de esticadas e chegadas ao final do braço aberto. A microinjeção do composto H-89 a 2,5nmol no HV, mas não no HD aumentou o tempo de permanência e número de entradas nos braços abertos, mas a dose de 5,0nmol elevou o número de entradas em braços fechados. Ambas as doses reduziram a expressão do comportamento de mergulho protegido para a mesma sub-região. Quando expostos ao TER, houve apenas diminuição do número de células Fos-positivas no HD de camundongos expostos repetidamente ao rato e relação ao grupo exposto a pelúcia. Em contrapartida, no HV, houve aumento da expressão de Fos nos grupos expostos ao rato de maneira única e repetida, em relação aos grupos controle e exposição ao brinquedo. A avaliação dos comportamentos desses animais revelou que a exposição repetida ao rato diminuiu o tempo de permanência na superfície do aparato, bem como o tempo de contato com a tela e escalada. A exposição única ao rato também diminuiu o tempo de escalada na tela e aumentou o número de esticadas. A microinjeção de CP 376395 nas doses 3,0 e 6,0nmol em HV aumentou o tempo de permanência na superfície do TER, mas só a dose de 6,0nmol foi capaz de exercer esse efeito em HD. Quanto aos comportamentos complementares, apenas a dose de 3,0nmol, injetada em HV, diminuiu o número de esticadas. Assim, esse estudo demonstrou participação do CRF principalmente no HV comparado ao HD na mediação de comportamentos relacionados à ansiedade e, além disso, que a via do AMPc/PKA no HV também parece mediar tais efeitos. Anxiety disorders involve several behavioral and autonomic reactions in face of aversive stimuli that may threaten life. These responses are modulated by different neurotransmitters and neural modulators in limbic system structures, such as hippocampus. Among the neurotransmitters involved, it has been attributed to corticotrophin releasing fator (CRF) an important role in regulating anxiety-related defensive behaviors, since the presence of CRFergic neurons and CRF receptors in extrahipolamic sites has been identified. When CRF binds to these Gs-protein coupled receptors it triggers a cascade of intracellular reactions involving the activation of the cAMP/Protein Kinase A (PKA) pathway. This study aimed to evaluate the role of the CRF neurotransmitter and PKA into the hippocampus of mice in anxiety-related behavioral responses when exposed to the Elevated Plus Maze (EPM) through the effects of CP 376395 376395, CRF1 antagonist, and H-89 microinjection, a PKA inhibitor, and rat exposure test (RET), through nuclear expression of Fos protein and effects CP 376395 microinjection. From EPM exposure results, it was possible to verify that the intra-ventral hippocampus (VH) treatment with CP 376395 increased the time in the open arms of the EPM at 3.0nmol dose, however, the 6.0nmol dose increased the number of unprotected head dipping when injected into the dorsal hippocampus (DH) and the number of protected head dipping when injected into VH. None of the doses promoted changes in the stretched-attend posture (SAP) behavior and arrival at the end of the open arms. Microinjection of H-89 at 2.5nmol dose in VH, but not in DH increased the time and number of entries in the open arms, but the 5.0nmol dose increased the number of entries in closed arms. Both doses reduced the expression of protected head dipping behavior on the same subregion. When exposed to RET, there was a decrease only in the number of Fos-positive cells in DH from mice repeatedly exposed to the rat in relation to the toy exposed group. On the other hand, in VH, there was an increase in Fos expression in the groups exposed to the rat in a single or repeated way, in relation to the control and toy exposed groups. The behavioral analysis of these animals revealed that the repeated exposure to the rat decreased the time spent on the surface of the apparatus, as well as the time of contact with the screen and climbing. The single exposure to rat also decreased the time climbing on the screen and increased the number of SAP. Microinjection of CP 376395 at 3.0 and 6.0nmol doses in VH increased the residence time on the surface of the RET, but only the dose of 6.0nmol was able to exert this effect in DH. In relation to the complementary behaviors, only the 3.0nmol dose, injected in VH, decreased the number of SAP. Thus, this study has demonstrated a CRF participation mainly into the VH, in the mediation of anxiety-like behaviors and, moreover, that cAMP/PKA pathway is also involved in the mediation of these effects.
