Treatment of inflammatory bowel disease (IBD), a common chronic intestinal disease, by exopolysaccharides (EPSs) produced by lactic acid bacteria has raised increasing concerns. Here, the EPS produced by Lactiplantibacillus plantarum NMGL2 was evaluated for its ameliorating effect on dextran sodium sulfate (DSS)-induced IBD in mice. Administration of the EPS was shown to decrease the body weight loss and the values of disease activity index (DAI) and alleviate the colon damage as evidenced by an improvement in colonic length shortening, a reduction in colonic coefficient, and a reduction in colonic mucosal architecture and inflammatory infiltration. Cytokine assay of the blood and colon tissue samples showed that the EPS could decrease the levels of pro-inflammatory TNF-α and IL-1β, and increase anti-inflammatory IL-10. Oxidative stress assay of the colon tissue showed that the nitric oxide (NO) and malondialdehyde (MDA) levels decreased significantly (p < 0.05), while superoxide dismutase (SOD) and glutathione (GSH) levels increased significantly (p < 0.05) after the EPS intervention. These results were further confirmed by the significantly (p < 0.05) down-regulated levels of NF-κB p65, p-IKKβ, and p-IκBα, and significantly (p < 0.05) enhanced expression of ZO-1 and occludin, as evaluated by Western-blot analysis of these proteins expressed in colonic tissue. The EPS produced by L. plantarum NMGL2 alleviated IBD by suppressing the NF-κB signaling pathway, suggesting its potential as a functional food agent in the prevention of IBD. [ABSTRACT FROM AUTHOR]

Bacterial biopolymers are changing the concepts in the food industry as customer demands the addition of no artificial additives and want products "Clean labelled." Exopolysaccharides (EPS) produced by some bacterial strains are the best replacements for these artificial additives. The monomeric composition, quality, quantity, structure, molecular weight, and charge on these bioparticles decide the qualities, that is, rheology, texture, shelf life, and physiochemical properties of the product. Exopolysaccharides are the best biothickeners and are being widely used in the food industry. They are widely used as coatings and edible films in food substances, especially fruits and vegetables. These consist of substances that are Generally Recognized as Safe. These coatings and edible films promise improved shelf life and better nutritional properties and act as a protective barrier for food items. Bacterial EPS are widely used in edible films, yogurt-like and plant-based beverages, the meat industry, and fermented low-fat meat, sourdoughs, and low-fat cheese. It interacts with other components and improves the texture, stabilization, viscosity, and mouthfeel of end products. The structural and functional properties of these substances still need to be explored. The main objective of this review is to focus on the importance of EPS produced by various strains, its structure and uses as edible coatings and films, functionality, and applications in various food products. Overall, the future of bacterial EPS in the food industry appears promising, with avenues for innovation and application across multiple facets of food production, including texture modification, nutritional enhancement, and sustainability. In the past, natural resources were used as best therapeutic sources for different diseases. Among several herbal resources, certain plant families are known for their analgesic activities, such as Dianthus crinitus , which showed a good curative potential. The mentioned plant has been used traditionally for the healing of several illnesses such as pain, inflammation, injuries, etc. The current study, therefore, evaluated the analgesic and anti-antioxidant activities of ethanolic extracts of leaves, flowers, and seeds of Dianthus crinitus in mice. For analgesic activity, acetic acid and a hot plate thermal stimulation method were used to induce writhes in mice, followed by a tail emersion process. Results showed that the extract demonstrated analgesic potential in mice, signifying both peripheral and central modes of action. Furthermore, extract also significantly inhibited the rat paw edema triggered by carrageenan seaweed and dextran as revealed by neutrophil migration toward the peritoneal cavities of animals and enhanced vascular permeability. The data suggested that the extract exhibited a dose-dependent antioxidant action. These preliminary observations provided support for further exploration and justification for the potential development of a new class of analgesic and anti-inflammatory medicines from the constituents of Dianthus crinitus extract.
