Like many coastal lagoons in several countries, the "Navío Quebrado" lagoon (La Guajira, Colombia) is a very delicate and precious environment; indeed, it is a nationally recognized Flora and Fauna Sanctuary. Several factors, including climate change, are threatening its existence because of changes in the governing hydro-morphological and biological processes. Certainly, the first step to addressing this problem is to understand its hydrological behavior and to be able to replicate, via simulation, its recent history before inferring likely futures. These potential futures will be marked by changes in the water input by its tributary, the Camarones River, and by modified water exchange with the sea, according to a foreseen sea level rise pattern, as well as by a different evaporation rate from the free surface, according to temperature changes. In order to achieve the required ability to simulate future scenarios, data on the actual behavior have to be gathered, i.e., a monitoring system has to be set up, which to date is non-existent. Conceptually, designing a suitable monitoring system is not a complex issue and seems easy to implement. However, the environmental, socio-cultural, and socio-economic context makes every little step a hard climb. An extremely simple—almost "primitive"—monitoring system has been set up in this case, which is based on very basic measurements of river flow velocity and water levels (river, lagoon, and sea) and the direct participation of local stakeholders, the most important of which is the National Park unit of the Sanctuary. All this may clash with the latest groovy advances of science, such as in situ automatized sensors, remote sensing, machine learning, and digital twins, and several improvements are certainly possible and desirable. However, it has a strong positive point: it provides surprisingly reasonable data and operates at almost zero additional cost. Several technical difficulties made this exercise interesting and worthy of being shared. Its novelty lies in showing how old, simple methods may offer a working solution to new challenges. SUMMARY: Polyamines are involved in several plant physiological processes. In Arabidopsis thaliana, five FAD‐dependent polyamine oxidases (AtPAO1 to AtPAO5) contribute to polyamine homeostasis. AtPAO5 catalyzes the back‐conversion of thermospermine (T‐Spm) to spermidine and plays a role in plant development, xylem differentiation, and abiotic stress tolerance. In the present study, to verify whether T‐Spm metabolism can be exploited as a new route to improve stress tolerance in crops and to investigate the underlying mechanisms, tomato (Solanum lycopersicum) AtPAO5 homologs were identified (SlPAO2, SlPAO3, and SlPAO4) and CRISPR/Cas9‐mediated loss‐of‐function slpao3 mutants were obtained. Morphological, molecular, and physiological analyses showed that slpao3 mutants display increased T‐Spm levels and exhibit changes in growth parameters, number and size of xylem elements, and expression levels of auxin‐ and gibberellin‐related genes compared to wild‐type plants. The slpao3 mutants are also characterized by improved tolerance to drought stress, which can be attributed to a diminished xylem hydraulic conductivity that limits water loss, as well as to a reduced vulnerability to embolism. Altogether, this study evidences conservation, though with some significant variations, of the T‐Spm‐mediated regulatory mechanisms controlling plant growth and differentiation across different plant species and highlights the T‐Spm role in improving stress tolerance while not constraining growth. Summary: Woody plants display some photosynthetic activity in stems, but the biological role of stem photosynthesis and the specific contributions of bark and wood to carbon uptake and oxygen evolution remain poorly understood.We aimed to elucidate the functional characteristics of chloroplasts in stems of different ages in Fraxinus ornus. Our investigation employed diverse experimental approaches, including microsensor technology to assess oxygen production rates in whole stem, bark, and wood separately. Additionally, we utilized fluorescence lifetime imaging microscopy (FLIM) to characterize the relative abundance of photosystems I and II (PSI : PSII chlorophyll ratio) in bark and wood.Our findings revealed light‐induced increases in O2 production in whole stem, bark, and wood. We present the radial profile of O2 production in F. ornus stems, demonstrating the capability of stem chloroplasts to perform light‐dependent electron transport. Younger stems exhibited higher light‐induced O2 production and dark respiration rates than older ones.While bark emerged as the primary contributor to net O2 production under light conditions, our data underscored that wood chloroplasts are also photosynthetically active. The FLIM analysis unveiled a lower PSI abundance in wood than in bark, suggesting stem chloroplasts are not only active but also acclimate to the spectral composition of light reaching inner compartments. Understanding which species are able to recover from drought, under what conditions, and the mechanistic processes involved, will facilitate predictions of plant mortality in response to global change. In response to drought, some species die because of embolism-induced hydraulic failure, whilst others are able to avoid mortality and recover, following rehydration. Several tree species have evolved strategies to avoid embolism, whereas others tolerate high embolism rates but can recover their hydraulic functioning upon drought relief. Here, we focus on structures and processes that might allow some plants to recover from drought stress via embolism reversal. We provide insights into how embolism repair may have evolved, anatomical and physiological features that facilitate this process, and describe possible trade-offs and related costs. Recent controversies on methods used for estimating embolism formation/repair are also discussed, providing some methodological suggestions. Although controversial, embolism repair processes are apparently based on the activity of phloem and ray/axial parenchyma. The mechanism is energetically demanding, and the costs to plants include metabolism and transport of soluble sugars, water and inorganic ions. We propose that embolism repair should be considered as a possible component of a ‘hydraulic efficiency-safety’ spectrum. We also advance a framework for vegetation models, describing how vulnerability curves may change in hydrodynamic model formulations for plants that recover from embolism.

Plant hydraulic traits related to leaf drought tolerance, like the water potential at turgor loss point (TLP) and the water potential inducing 50% loss of hydraulic conductance (P 50), are extremely useful to predict the potential impacts of drought on plants. While novel techniques have allowed the inclusion of TLP in studies targeting a large group of species, fast and reliable protocols to measure leaf P 50 are still lacking. Recently, the optical method coupled with the gas injection (GI) technique has been proposed as a possibility to speed up the P 50 estimation. Here, we present a comparison of leaf optical vulnerability curves (OVcs) measured in three woody species, namely Acer campestre (Ac), Ostrya carpinifolia (Oc) and Populus nigra (Pn), based on bench dehydration (BD) or GI of detached branches. For Pn, we also compared optical data with direct micro-computed tomography (micro-CT) imaging in both intact saplings and cut shoots subjected to BD. Based on the BD procedure, Ac, Oc and Pn had P 50 values of −2.87, −2.47 and −2.11 MPa, respectively, while the GI procedure overestimated the leaf vulnerability (−2.68, −2.04 and −1.54 MPa for Ac, Oc and Pn, respectively). The overestimation was higher for Oc and Pn than for Ac, likely reflecting the species-specific vessel lengths. According to micro-CT observations performed on Pn, the leaf midrib showed none or very few embolized conduits at −1.2 MPa, consistent with the OVcs obtained with the BD procedure but at odds with that derived on the basis of GI. Overall, our data suggest that coupling the optical method with GI might not be a reliable technique to quantify leaf hydraulic vulnerability since it could be affected by the 'open-vessel' artifact. Accurate detection of xylem embolism in the leaf vein network should be based on BD, preferably of intact up-rooted plants. [ABSTRACT FROM AUTHOR]

Grapevines store non-structural carbohydrates (NSC) during late summer to sustain plant development at the onset of the following spring’s growth. Starch is the main stored carbohydrate, found in the wood-ray parenchyma of roots and canes. Although the relationship between hydraulic and plant photosynthetic performance is well-recognized, little research has been done on the long-term effects of drought in grapevines adopting different strategies to cope with water stress (i.e. isohydric and anisohydric). We performed our study by exposing two different grape cultivars (Syrah and Cabernet Sauvignon) to a short but severe drought stress, at two stages of the growing season (July and September). No marked differences in the physiological and hydraulic responses of the two varieties were found, probably due to our experimental conditions. However, anatomical and biochemical characterization of overwintering canes pointed out several interesting outcomes. We found a significant and parallel increase of starch and medullar ray number in both cultivars exposed to early water stress. We hypothesize that stressed vines limited their carbon allocation to growth, while shifting it to starch accumulation, with a most evident effect in the period of intense photosynthetic activity. We also speculate that a different aptitude to osmotic adjustment may underlay variation in starch increase and the specific involvement of bark NSC in the two cultivars.

Plant water content is a simple and promising parameter for monitoring drought‐driven plant mortality risk. However, critical water content thresholds leading to cell damage and plant failure are still unknown. Moreover, it is unclear whether whole‐plant or a specific organ water content is the most reliable indicator of mortality risk. We assessed differences in dehydration thresholds in leaf, stem and root samples, hampering the organ‐specific rehydration capacity and increasing the mortality risk. We also tested eventual differences between a fast experimental dehydration of uprooted plants, compared to long‐term water stress induced by withholding irrigation in potted plants. We investigated three species with different growth forms and leaf habits i.e., Helianthus annuus (herbaceous), Populus nigra (deciduous tree) and Quercus ilex (evergreen tree). Results obtained by the two dehydration treatments largely overlapped, thus validating bench dehydration as a fast but reliable method to assess species‐specific critical water content thresholds. Regardless of the organ considered, a relative water content value of 60% induced significant cell membrane damage and loss of rehydration capacity, thus leading to irreversible plant failure and death. Summary statements: Across three species with different growth forms and leaf habits, a relative water content (RWC) of 60% increased the risk of mortality. The loss of rehydration capacity and cell membrane damage were correlated to RWC, that turned out as a reliable indicator of incipient plant death under drought. [ABSTRACT FROM AUTHOR]
