Glucocorticoid receptor (GR) has been implicated in prostate carcinoma growth and progression. Glucocorticoid receptor beta (GRβ) acts as an inhibitor of GR; however, its function is not well understood. Serum- and glucocorticoid-regulated kinase 1 (SGK1) is a GR-responsive gene that phosphorylates N-myc downstream-regulated gene 1 (NDRG1) and is involved in cancer growth and invasion. However, the expression of GR, GRβ, SGK1, and NDRG1 in prostate cancer and their relationship with clinicopathological and functional significance remain unknown. The association between the status of GR, GRβ, SGK1, and NDRG1 immunoreactivity and clinicopathological variables was analyzed in patients with prostate carcinoma to explore their clinical significance. In prostate carcinoma cases, the relative abundance of GR and NDRG1 immunoreactivity was inversely and significantly associated with the primary tumor stage (pT), while GR immunoreactivity was inversely and significantly associated with the Ki-67 score. The relative expression status of NDRG1 was significantly associated with that of GR. However, no significant correlation was observed between any of the clinicopathological parameters and GRβ and SGK1 expression. Our findings indicate that GR and NDRG1 expression status is correlated with clinicopathological features in patients with prostate cancer.

Droplet-based microfluidic technology is a powerful tool for single-cell cultivation and rapid isolation of bacteria, yeasts and algae. However, it has been of limited use for studies of filamentous fungi due to the fast growth of their branched hyphae. The long regeneration time for fungal protoplasts and low-throughput screening methods are inherent problems for current genetic transformation techniques. Therefore, we have developed a novel droplet-based method for the filamentous fungus Trichoderma reesei expressing green fluorescent protein (GFP) as a marker. This approach presented several outstanding advantages over the traditional transformation method, including a 7-fold reduction in time for T. reesei protoplast regeneration, an 8-fold increase in regeneration frequency, and a screening speed of up to 8,000 droplets min

The National Electrostatics Corporation (NEC) 250 kV single-stage AMS (YS-AMS) was installed in March 2013 at the Laboratory for Accelerator Mass Spectrometry (LAMS), Atmosphere and Ocean Research Institute, The University of Tokyo. This is the first single stage AMS system installed in Japan. The system is equipped with a 40 solid sample ion source (MC-SNICS-II), sequential injection system at low energy mass spectrometry side, open air 250 kV high energy deck including helium gas stripper which acts as a molecular dissociation, analyzing magnet, electrostatic analyzer, sequential post-accelerator deflector, and final detector. The performance tests with 11 reference materials distributed via the International Atomic Energy Agency (IAEA), National Institute of Standards and Technology (NIST), and National Institute of Advanced Industrial Science and Technology, Japan (AIST) were in very good agreement with consensus values. Thus our routine 14C measurements have been started since August 2013 and maintaining high performance. We have measured about 8000 unknown samples during this period. Typical 12C− currents are 27 μA at low energy Faraday cup, the transmission in the accelerator is about 42%, and precision of 14C/12C and 13C/12C is better than 0.2%. Geological, archaeological, oceanographic as well as a large number of biological samples have been measured efficiently using the current system.

Sex-specific differences in the incidence of urinary bladder carcinomas are well known, and the possible involvement of sex steroids has been proposed. We previously reported the association of the loss of androgen receptors and androgen-producing enzymes with tumor progression of urinary bladder cancer patients. Clinically, the selective estrogen receptor modulators (SERMs) were reported to suppress the progression of these tumors but the status of estrogen receptors (ERs) has not been well studied in patients with bladder urinary cancer. Moreover, not only ERs but also estrogen-related enzymes, such as aromatase, steroid sulfatase (STS), and estrogen sulfotransferase (EST), have been reported in the biological/clinical behavior of various hormone-dependent carcinomas but not studied in urinary bladder carcinoma. Therefore, in this study, we immunolocalized ERs as well as estrogen metabolizing enzymes in urinary bladder carcinoma and performed immunoblotting and cell proliferation assays using the bladder urothelial carcinoma cell line, T24. The results revealed that the loss of STS and aromatase was significantly correlated with advanced stages of the carcinoma. In vitro studies also revealed that T24 cell proliferation rates were significantly ameliorated after treatment with estradiol or diarylpropionitrile (DPN). EST and aromatase were also significantly correlated with the nuclear grade of the carcinoma. The results of our present study, for the first time, demonstrated that biologically active estrogens that bind to ERs could suppress tumor progression and the inactive ones could promote its progression and the potential clinical utility of SERM treatment in selective patients with urinary bladder carcinoma.

The Cerrado is the richest savanna and is undergoing one of the planet’s most rapid land transformations for pasture and agriculture; around 45% of the biome has been deforested. Agriculture is of strategic importance to Brazil, but it also modifies ecosystems and jeopardizes habitats and biodiversity. Well-managed agricultural lands can have a favorable impact on environmental conservation. In this paper, we reviewed our current knowledge about water ecology and fire management to show that an ecosystem services perspective can bring about a conciliation of agriculture production with conservation by supporting effective land use decision-making and the optimization of public policy. The landscape/watershed scale seems to be the most relevant for decision-making on how to achieve production and conservation results. This scale appears to be an appropriate level for engaging with stakeholders. Fire frequency and timing (season) combination are determinant of individuals’ survivorship. The combination determines vegetation recovery, and it is important to maintain high biodiversity, especially for the herbaceous layer, but it is a limitation to woody vegetation recovery. A pragmatic and conciliatory land use agenda must be based on scientific knowledge and support innovative decision-making solutions for policy-makers and stakeholders, particularly farmers and donors.

Background: Fire is an important ecological factor in the Cerrado (Brazilian savanna). However, comparative studies on the effect of high temperatures experienced during fires on seed germination of native and invasive grass species are few. Aims: To assess germination responses to simulated fire temperatures by seeds of invasive and native Cerrado grasses. Methods: Heat-shock treatments (50 °C, 70 °C, 90 °C, 110 °C, 130 °C or 150 °C) were applied to seeds of 10 species of native and invasive grasses. For each temperature, the seeds were heated in a dry-air flow for 2 or 5 min. This combination of temperatures and exposure times simulated the soil conditions during typical Cerrado fires. Results: Temperature treatment was significantly related to germination, and the effect varied according to species. Heat shock did not increase germination in either the native or the invasive species. Exposure time was important for only two species, and four species showed a significant increase in mean germination time. Conclusions: Species showed different tolerances to high temperatures. It was not possible to differentiate the native and invasive grasses only by their tolerance to high temperatures, suggesting that fire alone may not be an efficient management tool to control the invasive species studied here.

Introduction Although we recently reported a detailed description of wear pattern in Caucasians, few studies have investigated severity of wear, and pattern of wear on the patella and femur in Japanese. The purpose of this study was to determine if there is a common joint specific wear pattern in the patellofemoral joint of Japanese cadavers. We further compared the wear pattern and severity between Japanese and US cadavers. Methods We examined 46 patellae and matching femurs from 23 Japanese cadavers. The wear location was transcribed to gridded templates, that were compared for frequency, gender, and sidedness of wear. Results Over 60% of the patella showed wear in the odd facet, the middle part of the medial facet, and the medial aspect of the middle part of the lateral facet. The distal femur showed common areas of wear in the posterior aspect of the medial femoral condyle and anterior aspect of the medial femoral condyle. Patella wear may differ with gender as male’s wear was more compact. Although the patella and femur wear area was similar between Japanese and US cadavers, femoral wear may be more severe in Japanese cadavers. Discussion Patella and femur showed distinct regions of articular cartilage wear. The wear patterns and locations suggest that wear occurs mainly when the knees are moderately and deeply flexed and that these wear patterns may result from common traditional Japanese style of knee positions or activities.

Clinical PET studies using 18F-THK5351 have demonstrated significant tracer retention in sites susceptible to tau burden in Alzheimer disease (AD). However, the in vivo PET signal to reflect tau aggregates remains controversial. Methods: We examined the spatial pattern of tracer binding, amyloid-β, tau, and gliosis in an autopsy-confirmed AD patient who underwent 18F-THK5351 and 11C-Pittsburgh compound B PET before death. Results: Regional in vivo 18F-THK5351 retention was significantly correlated with the density of tau aggregates in the neocortex and monoamine oxidase-B in the whole brain, but not correlated with that of insoluble amyloid-β. Furthermore, significant association was observed between the density of tau aggregates, monoamine oxidase-B, and glial fibrillary acidic protein, suggesting that neocortical tau would strongly influence the formation of reactive astrocytes. Conclusion:18F-THK5351 PET may have limited utility as a biomarker of tau pathology in AD; however, it could be used to monitor the neuroinflammatory processes in the living brain.

Background Fungi can cause a variety of infectious diseases, including invasive mycosis and non-invasive mycosis, as well as allergic diseases. The different forms of mycosis usually have been described as mutually exclusive, independent entities, with few descriptions of overlapping cases. Here, we describe the first reported case of a patient with the complication of pulmonary eosinophilia in the course of invasive mucormycosis. Case presentation A 74-year-old Japanese man with asthma-COPD overlap underwent emergency surgery for a ruptured abdominal aortic aneurysm. The surgery was successful, but fever and worsening dyspnea appeared and continued from postoperative day (POD) 10. A complete blood count showed leukocytosis with neutrophilia and eosinophilia, and the chest X-ray showed consolidation of the left upper lung at POD 15. We suspected nosocomial pneumonia together with an exacerbation of the asthma-COPD overlap, and both antibiotics and bronchodilator therapy were initiated. However, the symptoms, eosinophilia and imaging findings deteriorated. We then performed a bronchoscopy, and bronchoalveolar lavage (BAL) fluid analysis revealed an increased percentage of eosinophils (82% of whole cells) as well as filamentous fungi. We first suspected that this was a case of allergic bronchopulmonary mycosis (ABPM) caused by Aspergillus infection and began corticosteroid therapy with an intravenous administration of voriconazole at POD 27. However, the fungal culture examination of the BAL fluid revealed mucormycetes, which were later identified as Cunninghamella bertholletiae by PCR and DNA sequencing. We then switched the antifungal agent to liposomal amphotericin B for the treatment of the pulmonary mucormycosis at POD 29. Despite replacing voriconazole with liposomal amphotericin B, the patient developed septic shock and died at POD 39. The autopsy revealed that filamentous fungi had invaded the lung, heart, thyroid glands, kidneys, and spleen, suggesting that disseminated mucormycosis had occurred. Conclusions We describe the first reported case of pulmonary mucormycosis with pulmonary eosinophilia caused by Cunninghamella bertholletiae, which resulted in disseminated mucormycosis. Although it is a rather rare case, two important conclusions can be drawn: i) mycosis can simultaneously cause both invasive infection and a host allergic reaction, and ii) Cunninghamella bertholletiae rarely infects immunocompetent patients.

l-3,4-dihydroxyphenylalanine (l-DOPA) is a candidate neurotransmitter. l-DOPA is released by nicotine through nicotinic receptors. Recently, G-protein coupled receptor GPR143, was identified as a receptor for l-DOPA. In this study, genetic association studies between GPR143 genetic polymorphisms and smoking behaviors revealed that the single-nucleotide polymorphism rs6640499, in the GPR143 gene, was associated with traits of smoking behaviors in Japanese individuals. In Gpr143 gene-deficient mice, nicotine-induced hypolocomotion and rewarding effect were attenuated compared to those in wild-type mice. Our findings suggest the involvement of GPR143 in the smoking behaviors.

Several Cerrado tree species have traits and structures that protect from fires. The effectiveness of a trait depends on the fire regime, especially the frequency. We used Vochysia elliptica, a common Cerrado tree, as a model to test whether different fire frequencies alter crown architecture and flower, fruit and seed production. We analysed the effect of fire on the production of inflorescences, fruits and seeds, as well as seed germination and tree architecture of 20 trees in each of three plots of a long-term ecological experiment managed with different fire regimes: burned every 2 years (B), burned every 4 years (Q) in mid-dry season and an area protected from fire (C). We found a large negative effect of fire frequency on crown architecture and on flower and fruit production. Trees in C and Q had significantly more main branches and a larger crown area than trees in B. At its peak, a tree in C was expected to produce 2.4 times more inflorescences than Q, and 15.5 times more than B, with similar magnitudes for fruits. Sixty per cent of trees in B and 10% in Q produced no fruits. The differences in architecture might explain the reduction in sexual reproduction due to a smaller physical space to produce flowers at the branch apices. Resource limitation due to plant investment to replace burned vegetative parts may also decrease sexual reproduction. Our results indicate potentially severe consequences of high fire frequencies for population dynamics and species persistence in Cerrado communities.

Adrenocortical carcinoma (ACC) is a rare, highly malignant neoplasm harboring marked histologic heterogeneity. The Ki-67 labeling index (LI) is one of the most effective diagnostic and prognostic markers in ACC. However, its assessment has by no means been standardized. Therefore, in this study, we analyzed the Ki-67 LI in 18 ACC cases both by seven pathologists using microscopes (MA; manual analysis) and with digital image analysis (DIA) and also compared the Ki-67 LI obtained by selecting "hot spots" and formulating the "average" reading of the whole tumor specimen. In addition, we performed statistical analysis of the association between Ki-67 LI and the clinical and pathologic features of individual cases. The DIA was significantly correlated with MA in hot spots but not in the average fields. The Ki-67 LI in hot spots was significantly and consistently higher than that in average areas by both MA and DIA, indicating intratumoral heterogeneity. The Ki-67 LI was significantly correlated with the Weiss criteria (eosinophilic cytoplasm, nuclear atypia, atypical mitoses, and sinusoidal invasion) by any mode of evaluation. The clinical outcome was significantly better in the patients with a Ki-6710% than in those with a Ki-6710% by MA in hot spots. The Ki-67 LI in hot spots measured by MA best reflected the clinical and pathologic features of ACC. Employment of DIA to obtain the Ki-67 LI in ACC requires further improvement, including correction of its overestimation of the value by counting non-tumorous cells and nuclear segmentation in areas of high cell density.

Melinis minutiflora é uma gramí­nea invasora em extensas áreas de Cerrado. Neste estudo de caso, foi utilizado corte anual a fim de verificar seu efeito no recrutamento de novos indiví­duos de Melinis minutiflora. Em setembro de 2007, após a dispersão das sementes, o corte foi aplicado em uma área de 50 x 50 m e, nos quatro anos seguintes, o corte foi aplicado após a dispersão das sementes de M. minutifolia. A densidade de sementes no solo foi estimada antes de cada corte. Em 2007, a densidade de sementes de Melinis minutifolia variou de 8.025 a 22.350 sementes m-2, sendo reduzida para 88 a 313 sementes m-2 em 2011. Na primeira estação de crescimento, o recrutamento foi de oito plântulas de M. minutifolia por metro quadrado, sem recrutamento nos anos seguintes. As reduções significativas no banco de sementes e no recrutamento sugerem que o corte antes da dispersão das sementes é útil para controlar o recrutamento de M. minutiflora.

Osteoarthritis (OA) is the most common degenerative disease affecting the hip (hip OA). It is characterized by symptoms such as stiffness and pain, and results in restricted range of motion, cartilage destruction, decreased joint space width, and reduction in activities and quality of life. However, there is no adequate conservative treatment for hip OA. One of the key factors for managing hip OA is unloading or distracting the joint with traction force. Joint distraction with a surgical procedure has been used to treat OA, mostly in the knee and the ankle joints, and the effects of joint distraction on joint structure and clinical symptoms have been reported. Here, we briefly review OA and then discuss the potential benefit of distraction of the hip joint in management of OA, focusing on conservative methods.

von Hippel-Lindau (VHL) disease, caused by germline mutations in the VHL gene, is a hereditary autosomal-dominant disorder which predisposes the individual to various malignant and benign tumors. VHL acts as a tumor suppressor, mainly through the negative regulation of hypoxia-inducible factors. Molecular-targeted drugs against vascular endothelial growth factor-signaling pathways, a target of hypoxia-inducible factors, have recently been introduced into clinical practice for the treatment of patients with sporadic renal cell carcinoma and pancreatic neuroendocrine tumors. However, whether such treatments are effective in patients with VHL disease remains to be elucidated. We herein report a Japanese patient with VHL disease who was successfully treated with sunitinib for approximately 5 years.

Gastric cancer is the third leading cause of cancer mortality in Japan and worldwide. Although previous studies identify various genetic variations associated with gastric cancer, host genetic factors are largely unidentified. To identify novel gastric cancer loci in the Japanese population, herein, we carried out a large-scale genome-wide association study using 6171 cases and 27 178 controls followed by three replication analyses. Analysis using a total of 11 507 cases and 38 904 controls identified two novel loci on 12q24.11-12 (rs6490061, P = 3.20 × 10-8 with an odds ratio [OR] of 0.905) and 20q11.21 (rs2376549, P = 8.11 × 10-10 with an OR of 1.109). rs6490061 is located at intron 19 of the CUX2 gene, and its expression was suppressed by Helicobacter pylori infection. rs2376549 is included within the gene cluster of DEFB families that encode antibacterial peptides. We also found a significant association of rs7849280 in the ABO gene locus on 9q34.2 (P = 2.64 × 10-13 with an OR of 1.148). CUX2 and ABO expression in gastric mucosal tissues was significantly associated with rs6490061 and rs7849280 (P = 0.0153 and 8.00 × 10-11 ), respectively. Our findings show the crucial roles of genetic variations in the pathogenesis of gastric cancer.

Recent positron emission tomography (PET) studies have demonstrated the accumulation of tau PET tracer in the affected region of progressive supranuclear palsy (PSP) cases. To confirm the binding target of radiotracer in PSP, we performed an imaging-pathology correlation study in two autopsy-confirmed PSP patients who underwent [18F]THK5351 PET before death. One patient with PSP Richardson syndrome showed elevated tracer retention in the globus pallidus and midbrain. In a patient with PSP-progressive nonfluent aphasia, [18F]THK5351 retention also was observed in the cortical areas, particularly the temporal cortex. Neuropathological examination confirmed PSP in both patients. Regional [18F]THK5351 standardized uptake value ratio (SUVR) in antemortem PET was significantly correlated with monoamine oxidase-B (MAO-B) level, reactive astrocytes density, and tau pathology at postmortem examination. In in vitro autoradiography, specific THK5351 binding was detected in the area of antemortem [18F]THK5351 retention, and binding was blocked completely by a reversible selective MAO-B inhibitor, lazabemide, in brain samples from these patients. In conclusion, [18F]THK5351 PET signals reflect MAO-B expressing reactive astrocytes, which may be associated with tau accumulation in PSP.

Esophageal squamous cell carcinoma (ESCC) is highly malignant. Recently, the expression of myosin 5a, a member of the myosin superfamily, was reported to be associated with increased invasiveness and metastasis in many tumor types. Moreover, myosin 5a is upregulated by Snail and activated by Akt2, both of which are epithelial-mesenchymal transition (EMT) markers. In this study, we confirmed the expression of myosin 5a in ESCC surgical specimens and cell lines, revealing its correlation with tumor invasion, migration, patient prognosis, and expression of EMT-related proteins. The expression of myosin 5a, vimentin, and E-cadherin was immunohistochemically evaluated in 118 patients with ESCC who underwent esophagectomy without chemotherapy or irradiation therapy prior to surgery. We also investigated ESCC cell migration under myosin 5a silencing by siRNA induction. The high expression of myosin 5a was correlated with tumor depth, lymph node metastasis, pathological stage, high vimentin expression, and low E-cadherin expression. Patients with high expression of myosin 5a, including those with pT1 cancer, exhibited significantly worse survival. Moreover, the expression level of vimentin mRNA and the number of migrated ESCC cells decreased significantly following myosin 5a silencing. Our findings demonstrate that high expression of myosin 5a may be an independent prognostic factor in patients with ESCC, even in early invasive carcinoma, and indicate myosin 5a has a role in both cell migration and EMT.

The hip joint is a complex anatomic structure composed of osseous, ligaments, and muscular structure. It is a unique joint because it provides both stability and multiaxial mobility, and is involved in transferring the weight of the body. It is a common site of osteoarthritis (OA). OA is believed to be the most prevalent chronic joint disease of the hip joint and its incidence is rising as the population ages. A thorough anatomical understanding of the hip joint may us help to improve the management of hip conditions, including OA. This article presents a review of anatomy of the hip joint, designed to help a better understanding of the principles of management. We also discuss the clinical importance of anatomy of this joint from the perspective of conservative treatment.

Although the Cerrado vegetation is adapted to fire, anthropogenic fires may impair the reproduction of some species. This study aimed to test the role of fruits of Qualea multiflora and their position in the tree crown in protecting the seeds during fires. Ten trees were selected in an area protected from fire (NB) and ten in a biannually burned area (B). Fruits were counted before the 2008 fire and weekly thereafter for 5 weeks. Fruit dehiscence, damage and position in the tree crown were recorded. Seed germination was assessed for seeds from both areas. In NB, 5.7% of fruits were located higher than 2 m, while in B 49.5% were located at this height. One week after the fire, the proportion of dehiscent fruits was lower in NB (19.5%) than in B (34.5%). Five weeks after the fire, all fruits in NB had dispersed their seeds, whereas in B only 47% of the fruits had. Immediately before the fire, the germination rate of seeds collected in NB was 80% (0-1 m), 97% (1-2 m) and 92% (≥2 m). Fifteen days after the fire, the germination rate of seeds in the B area was 3% (0-1 m), 13% (1-2 m) and 78% (≥2 m). Protection of Q. multiflora seeds from high temperatures is related to the fruit position in the tree crown rather than to fruit protection. Therefore, the results suggest that fire may alter the recruitment of new individuals.

Endogenous fungal endophthalmitis (EFE) caused by disseminated fusariosis is a rare condition that generally has a poor outcome, even with intensive therapy. Here, we describe a case in which this type of EFE was diagnosed with vitreous sampling and was successfully treated with 25-gauge vitrectomy and antifungals, including liposomal amphotericin B and voriconazole. A 16-year-old male patient undergoing treatment for acute myeloid leukemia complained of eye pain and blurred vision in his right eye. Treatment was initiated for a vitreous opacity, possibly associated with herpetic retinitis, but the patient worsened and he was referred to us. Right-eye visual acuity was limited to light perception. We suspected endogenous endophthalmitis and performed 25-gauge vitrectomy with antibiotic perfusion of ceftazidime, vancomycin, and voriconazole. Vitreous culturing revealed the presence of Fusarium solani species complex, and enhanced computed tomography revealed disseminated fusariosis lesions in the lung, spleen, and the soft tissue of the left upper arm. The patient received antifungal treatment with liposomal amphotericin B and voriconazole, and these conditions were eliminated. Visual acuity recovered to 20/400 after additional vitrectomy for tractional retinal detachment and was maintained at this level during the 6-month follow-up period. The success of our treatment allowed the capture of optical coherence tomography images of the retina during fusarium-associated endogenous endophthalmitis and the follow-up period. Furthermore, this case showed that immediate vitrectomy for suspected EFE and intensive treatment can lead to a good clinical outcome.

Clinical PET studies using

G-protein–activated inwardly rectifying potassium (GIRK) channels are expressed in many tissues and activated by several Gi/o protein–coupled receptors, such as opioid and dopamine receptors, and thus are known to be involved in the modulation of opioid-induced analgesia, pain, and reward. We focused on a GIRK-channel subunit that plays a pivotal role in the brain, GIRK2, and investigated the contribution of genetic variations of the GIRK2 (KCNJ6) gene to individual differences in the sensitivity to opioid analgesia. In our initial linkage disequilibrium analysis, a total of 27 single-nucleotide polymorphisms (SNPs) were selected within and around the regions of the KCNJ6 gene. Among them, the rs2835859 SNP, for which associations with analgesia and pain have not been previously reported, was selected in the exploratory study as a potent candidate SNP associated with opioid analgesic sensitivity. The results were corroborated in further confirmatory study. Interestingly, this SNP was also found to be associated with sensitivity to both cold and mechanical pain, susceptibility to nicotine dependence, and successful smoking cessation. The results indicate that this SNP could serve as a marker that predicts sensitivity to analgesic and pain and susceptibility to nicotine dependence. There is insufficient information about germination, seed biology and seedling establishment of native grass species of the cerrado (Brazilian savannah) to recommend their use in restoration of disturbed areas. We evaluated the production, percentage of fertile seeds, germination, seedling growth and establishment in the field of eighteen native grass species and one exotic invasive species (Melinis minutiflora) and also the effects of different storage periods on germination. The percentage of fertile seeds varied from 6·0 to 94·2% among native species. Ten species produced a minimum of 800 fertile seeds per gram. Germination rate was in the range of 11·0 to 98·8%. Storage for 1 year reduced the germination rate in one species, did not affect germination of three species and increased germination (by 2–10 times) in 14 species. After 2 years of storage, six species showed reductions in germination. Three different patterns of resource allocation were observed and might provide mechanisms for increasing seedling establishment under different environmental conditions. In the field, only two species flowered in the first year after sowing and an additional eight in the second year of observation. Melinis minutiflora is an aggressive species with more than three times the fertile seed production of native species (2820 seeds per gram), high germination rate (55%) and a high number of germinable seeds (1551 per gram). Despite the wide variation in seed characteristics and the delay in flowering, it is concluded that most native grass species can be used in projects aiming at the restoration and management of degraded areas.

In the late 1990s, an increase in automobile shredder residue (ASR) weighed down the remaining capacity of waste landfill sites in Japan. Furthermore, an increase in landfill costs and a downturn in the ferrous scrap price made the existing end-of-life vehicle (ELV) recycling system malfunction, and there was concern about the inappropriate treatment of ELVs. Consequently, the ELV Recycling Act was enacted in 2005, and currently, 3–4 million ELVs are recycled each year. Under the ELV Recycling Act, vehicle manufacturers and importers have obligations regarding the destruction of fluorocarbons, the recycling of gas generators and the recycling and energy recovery of ASR, using the recycling fee paid by vehicle owners when they purchase a new vehicle. In the fiscal year 2011, the recycling and energy recovery rate of ASR and its equivalent was 93.3 %, which results in approximately 99 % of ELVs being reused, recycled and recovered. Thanks to the prepayment of the recycling fee, the illegal dumping and inappropriate storage of ELVs has decreased significantly by 96 % compared to the levels before the enforcement. Several challenges for future improvements of the recycling system and efforts for vehicles affected by the Great East Japan Earthquake are explained.

To make railway transportation more efficient, utilization of recuperation energy in d.c. traction power supply system is actively discussed in Japan. We, East Japan Railway Company, have already introduced two Lithium ion battery systems at Haijima Substation since 2013 and at Okegawa Substation since 2014. They contribute to reduce total traction energy supply about 5–8%. In a.c. traction power supply system, on the other hand, it is rarely discussed to make use of recuperation power because it is not an issue compared with d.c. traction power supply system in many cases. We installed, however, RPC, railway static power conditioner, at Ushiku sectioning post for recuperation energy utilization in 2014. In this paper, the difference between d.c. traction power supply system and a.c. traction power supply system from the point of view to utilize recuperation energy and its future prospect in a.c. traction power supply system are discussed.

Aim: The endogenous opioid system has been reportedly implicated in tobacco/nicotine dependence. Materials & methods: We examined the genetic effects of eight SNPs in opioid receptor-related genes on smoking status and smoking-related traits in Japanese. Results: The genotypic and allelic variations of the rs2229205 SNP in the OPRL1 gene were significantly associated with smoking status, but no significant differences were found in the genetic variations of any of the SNPs with regard to smoking-related traits. The rs2229205 SNP did not show high linkage disequilibrium with the other SNPs in the linkage disequilibrium block that contained the SNP. Conclusion: The rs2229205 SNP in the OPRL1 gene may be a genetic factor that contributes to individual differences in the vulnerability to smoking in Japanese individuals.

Smoking behavior is a multifactorial trait influenced by genetic and environmental components. Several genome-wide association studies (GWASs) have identified candidate genes or loci for smoking behavior. However, most of these results were from European studies. Genetic differences are known to exist between ethnic groups or samples from different geographic locations. This chapter focuses on the genetic aspects of Japanese smoking behavior. Focusing on the CYP2A6 polymorphism, many studies have reported that a mutant allele with an impaired or null enzyme activity was a negative risk factor for nicotine addiction; however, some studies have indicated negative associations. Regarding neuronal nicotinic acetylcholine receptors, we showed that the CHRNA5 rs16969968 polymorphism was associated with smoking cessation and a high Tobacco Dependence Screener score. We have also confirmed that the TaqIA polymorphism near the DRD2 gene was related to current smoking, which was consistent with the results of previous studies examining Japanese populations. The further accumulation of investigations, including GWASs, and functional studies is warranted to establish a wider view of the genetic factors affecting smoking behavior.

Little is known about the iliocapsularis muscle, which covers the anteromedial capsule of the hip joint. Although this muscle has rarely mentioned in anatomy texts since its first appearance in a French Anatomy text in 1843, it is considered an important muscular landmark in terms of surgery. More recently, several studies have focused on and described the morphology and possible role of this muscle. Knowledge about the iliocapsularis muscle is essential to understand its anatomy, functions, and pathology, all of which will help to improve the management of hip conditions. Therefore, this article reviews the existing knowledge about the iliocapsularis muscle, including a presentation of the muscle dissected from a Japanese cadaver. We will further discuss the clinical importance of the anatomy of this muscle from the perspective of conservative treatment.

The dopaminergic brain pathway is involved in many addictive behaviours, hence represents a good candidate in the study of smoking behaviour and nicotine addiction. Dopamine beta hydroxylase (DBH) is an enzyme that catalyses the conversion of dopamine into noradrenaline. This study, the first of its kind, was done to investigate the role of DBH rs5320 polymorphism in smoking behaviour of elderly Japanese. This was done by collecting blood samples from 2521 subjects with various smoking habits to genotype the DBH rs5320 polymorphism. Participants also had to fill out a questionnaire containing questions regarding their lifestyles. Some of the questions were from the Fagerström Test for Nicotine Dependence (FTND) and the Tobacco Dependence Screener (TDS). It was found that male ever-smokers with AA genotype smoked less cigarettes per day than those with GG and AG genotypes. FTND scores were also lowest in male ever-smokers with AA genotype and in female ever-smokers with AG genotype. There was no correlation detected between the TDS scores and any of the genotypes. This study shows that DBH rs5320 polymorphism influences nicotine dependence.

Towards total synthesis of a series of kinamycin and related antibiotics via common synthetic intermediates, total synthesis of prekinamycin was achieved via Suzuki coupling of naphthaleneboronic acid and bromobenzene derivative, intramolecular FriedelCrafts reaction of 2-(naphthalen-2-yl)benzoic acid, and diazotization in ten steps from 3,5-dimethylphenol. Synthetic studies towards kinamycin antibiotics was also examined, and the tetracyclic quinone core for kinamycins was synthesized. Palladium-catalyzed site-selective hydroxylation of a benzoic acid derivative with the AB-D ring part was successfully applied to the selective D-ring functionalizations.

We previously isolated 40 predominant strains of bacteria, the characteristics of which resembled Wolinella (presently Campylobacter ), among 422 isolates from seven human periodontal pockets; however, at that time, exact identification was not possible by conventional microbiological and biochemical tests because the criteria were ambiguous. Therefore, in the present study we aimed to clarify the classifications of the 17 representative strains by polymerase chain reaction (PCR) using a species-specific primer, 16S rDNA sequence similarity, sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), and Western immunoblotting. We also aimed to describe each strain's biochemical characteristics in detail. All of the 17 isolates studied were asaccharolytic, positive in nitrate reduction and arylsulfatase activity, negative in catalase and urease, and produced succinate as an end product of formate and fumarate for growth. Of the 17, 13 strains were identified as C. rectus , and 4 were oxidase negative strains ( C. gracilis ). Three of the C. gracilis strains were cocobacilli, which were morphologically different from the previously established strains. Evidence suggests that Campylobacter isolates from periodontal pockets in our previous work may be primarily C. rectus and secondarily C. gracilis .

The clinical significance of glutathione-S-transferase GSTM1, GSTT1 and p53 codon 72 polymorphisms in cervical carcinogenesis was investigated. Germline polymorphisms of GSTM1, GSTT1 and p53 codon 72 together with human papillomavirus (HPV) types were examined in a total of 457 blood and cervical smear samples from normal healthy women and the patients with premalignant and malignant cervical lesions. The 167 patients with low-grade squamous intraepithelial lesion (LSIL), 49 with high-grade SIL (HSIL) and 83 with squamous cell carcinoma (SCC) had significantly higher frequency of high-risk HPV than 1 58 controls. The 49 patients with HSIL and 83 with SCC had statistically higher frequency of null GSTT1 genotype than 158 controls. There was an increased odds ratio for null GSTT1 genotype in HSIL and SCC cases compared with controls among 191 patients with high-risk HPV. The 67 cases with HPV types 16 and/or 18 had higher frequency of the GSTT1 null genotype than 186 with other types of HPV. There was no statistical difference in the polymorphic frequency of GSTM1 and p53 codon 72 genotypes between SILs and controls with or without high-risk HPV. These results suggest that GSTT1 null genotype may increase the risk of cervical cancer particularly in the cases with high-risk HPV types in a Japanese population.

Fire is common in savannas but its effects on soil are poorly understood. We analyzed long-term effects of fire on surface soil of an open Brazilian savanna (campo sujo) in plots submitted to different fire regimes during 18 years. The five fire regimes were: unburned, quadrennial fires in middle dry season, and biennial fires in early, middle or late dry season. Soil was collected during the wet and the middle dry season of 2008, and analyzed for pH, organic matter, total N, potential acidity, exchangeable cations and available P, S, Mn, Cu, Zn and Fe. We applied multivariate analysis to search for patterns related to fire regimes, and to local climate, fuel, and fire behavior. Spearman test was used to establish correlations between soil variables and the multivariate analysis gradient structure. Seasonal differences were tested using t-test. We found evidence of long-term fire effects: the unburned plot was segregated mainly by lower soil pH; the quadrennial plot was also segregated by lower soil pH and higher amount of exchangeable cations; the time of burning during the dry season in biennial plots did not significantly affect soil availability of nutrients. Differences in elements amounts due to the season of soil sampling (wet or dry) were higher than due to the effect of fires. Higher availability of nutrients in the soil during the wet season was probably related to higher nutrient inputs via rainfall and higher microbial activity.

New membrane-associated placental tissue proteins (MP2 A, B, C, D, and E) were investigated by avidin-biotin immunoperoxidase technique in the human and cynomolgus monkey placentae, decidua and umbilical cords. In human early placentae, MP2 A, B, C, and E were localized mainly in the membrane of villous syncytiotrophoblasts and cytotrophoblasts. Histiocytes in the villous stroma were positive for MP2 A, B, D, and E. In human term placentae, obvious positive staining for MP2 A, B, C, and E was observed in the membrane of villous syncytiotrophoblasts, in the amniotic epithelium, and in the umbilical cord sheath. Histiocytes in the villous stroma were positive for MP2 A, B, C, E, and especially for MP2 D. Importantly, MP2 A, C, and E were positive in polymorphonuclear neutrophils, since most of these common antigens are also carcinoma-associated, suggesting clinical usage of MP2 proteins as a new tumor marker. In the cynomolgus monkey placentae, similar immuno-staining results were obtained. The monkey can thus serve as a experimental model for the investigation of the placental proteins.

Using a monoclonal immunoradiometrical assay, we measured the concentrations of the specific epitope M3 of tissue polypeptide antigen (TPA), namely, tissue polypeptide specific antigen (TPS), in serum and amniotic fluid obtained from normal, healthy, nonpregnant and pregnant Japanese women (NHNJW and NHPJW). The cut-off value of the serum TPS level was set at 130 Ul, based on the mean+2 standard deviations in the NHNJW. The serial measurement of serum TPS levels demonstrated the significant emporal elevation of TPS level near the time of ovula-tion (±3 days) in the ovulatory women. In the NHPJW, the mean serum TPS level and the positivity rate (>130 U/l) increased with the advance of gestation, reaching 183.7 U/l and 66.7%, respectively, in the third trimester. Maternal serum levels were much elevated just before (mean: 982.3 U/l) and after (mean: 824.6 U/l) delivery, and were reduced to about one-fourth by 3 days. In addition, markedly high TPS concentrations were found in the retroplacental blood (one case: 9,076 U/l) and the amniotic fluid (mean: 11,650 U/l). The serum TPS level correlated well (r=0.871) with the serum TPA level in the TPS range of 2-2,000 U/l, while a poor correlation (r=0.273) was found in the TPS range of 2–500 U/l. The present study thus obtained fundamental data on TPS in Japanese women. The fluctuation and change of serum TPS levels during the menstrual cycle and in pregnancy should be taken into consideration when we apply TPS as a tumor marker for women.
