Abstract

We analysed the clustering of a photometric sample of galaxy clusters selected from the Third Data Release of the Kilo-Degree Survey, focusing on the redshift-space two-point correlation function (2PCF). We compared our measurements to theoretical predictions of the standard $\Lambda$ cold dark matter ($\Lambda$CDM) cosmological model. We measured the 2PCF of the sample in the cluster centric radial range $r\in[5,80]$ $h^{-1}$Mpc, considering 4934 galaxy clusters with richness $\lambda^*\geq15$ in the redshift range $z\in[0.1,0.6]$. A Markov chain Monte Carlo analysis has been performed to constrain the cosmological parameters $\Omega_{\rm m}$, $\sigma_8$, and $S_8 \equiv \sigma_8(\Omega_{\rm m}/0.3)^{0.5}$, assuming Gaussian priors on the mass-richness relation given by the posteriors obtained from a joint analysis of cluster counts and weak lensing. In addition, we constrained the normalisation of the mass-richness relation, $\alpha$, with fixed cosmological parameters. We obtained $\Omega_{\rm m}=0.28^{+0.05}_{-0.04}$, $\sigma_8=0.82^{+0.14}_{-0.12}$, and $S_8=0.80^{+0.08}_{-0.08}$. The constraint on $S_8$ is consistent within 1$\sigma$ with the results from WMAP and Planck. Furthermore, by fixing the cosmological parameters to those provided by Planck, we obtained $\alpha=0.12^{+0.06}_{-0.06}$, which is fully consistent with the result obtained from the joint analysis of cluster counts and weak lensing performed for this sample., Comment: 8 pages, 5 figures, 1 table. Abstract

We present a cosmological analysis of abundances and stacked weak-lensing profiles of galaxy clusters, exploiting the AMICO KiDS-DR3 catalogue. The sample consists of 3652 galaxy clusters with intrinsic richness $\lambda^*\geq20$, over an effective area of 377 deg$^2$, in the redshift range $z\in[0.1,\,0.6]$. We quantified the purity and completeness of the sample through simulations. The statistical analysis has been performed by simultaneously modelling the comoving number density of galaxy clusters and the scaling relation between the intrinsic richnesses and the cluster masses, assessed through a stacked weak-lensing profile modelling. The fluctuations of the matter background density, caused by super-survey modes, have been taken into account in the likelihood. Assuming a flat $\Lambda$CDM model, we constrained $\Omega_{\rm m}$, $\sigma_8$, $S_8 \equiv \sigma_8(\Omega_{\rm m}/0.3)^{0.5}$, and the parameters of the mass-richness scaling relation. We obtained $\Omega_{\rm m}=0.24^{+0.03}_{-0.04}$, $\sigma_8=0.86^{+0.07}_{-0.07}$, $S_8=0.78^{+0.04}_{-0.04}$. The constraint on $S_8$ is consistent within 1$\sigma$ with the results from WMAP and Planck. Abstract

Bioimage classification plays a crucial role in many biological problems. In this work, we present a new General Purpose (GenP) ensemble that boosts performance by combining local features, dense sampling features, and deep learning approaches. First, we introduce three new methods for data augmentation based on PCA/DCT; second, we show that different data augmentation approaches can boost the performance of an ensemble of CNNs; and, finally, we propose a set of handcrafted/learned descriptors that are highly generalizable. Each handcrafted descriptor is used to train a different Support Vector Machine (SVM), and the different SVMs are combined with the ensemble of CNNs. Our method is evaluated on a diverse set of bioimage classification problems. Results demonstrate that the proposed GenP bioimage ensemble obtains state-of-the-art performance without any ad-hoc dataset tuning of parameters (thus avoiding the risk of overfitting/overtraining)., Comment: 27 pages, 1 figure, 5 tables, manuscript

Abstract

Background Although children with special health care needs (CSHCN) represent a minority of the population, they go through more hospitalizations, more admissions to the Emergency Department (ED), and receive a major number of medical prescriptions, in comparison to general pediatric population. Objectives of the study were to determine the reasons for admission to the ED in Italian CSHCN, and to describe the association between patient’s demographic data, clinical history, and health services requirements. Methods Ad hoc web site was created to collect retrospective data of 3479 visits of CSHCN to the ED in 58 Italian Hospitals. Results Seventy-two percent of patients admitted to ED were affected by a previously defined medical condition. Most of the ED admissions were children with syndromic conditions (54%). 44.2% of the ED admissions were registered during the night-time and/or at the weekends. The hospitalization rate was of 45.6% among patients admitted to the ED. The most common reason for admission to the ED was the presence of respiratory symptoms (26.6%), followed by gastrointestinal problems (21.3%) and neurological disorders (18.2%). 51.4% of the access were classified as ‘urgent’, with a red/yellow triage code. Considering the type of ED, 61.9% of the visits were conducted at the Pediatric EDs (PedEDs), 33.5% at the Functional EDs (FunEDs) and 4.6% at the Dedicated EDs (DedEDs). Patients with more complex clinical presentation were more likely to be evaluated at the PedEDs. CSHCN underwent to a higher number of medical procedures at the PedEDs, more in comparison to other EDs. Children with medical devices were directed to a PedED quite exclusively when in need for medical attention. Subjects under multiple anti-epileptic drug therapy attended to PedEDs or FunEDs generally. Patients affected by metabolic diseases were more likely to look for medical attention at FunEDs. Syndromic patients mostly required medical attention at the DedEDs. Conclusions Access of CSHCN to an ED is not infrequent. For this reason, it is fundamental for pediatricians working in any kind of ED to increase their general knowledge about CHSCN and to gain expertise in the management of such patients and their related medical complexity.

Abstract

Background Zinc finger and BTB domain-containing protein 12 (ZBTB12) is a predicted transcription factor with potential role in hematopoietic development. Recent evidence linked low methylation level of ZBTB12 exon1 to myocardial infarction (MI) risk. However, the role of ZBTB12 in the pathogenesis of MI and cardiovascular disease in general is not yet clarified. We investigated the relation between ZBTB12 methylation and several blood parameters related to cardio-cerebrovascular risk in an Italian family-based cohort. Results ZBTB12 methylation was analyzed on white blood cells from the Moli-family cohort using the Sequenom EpiTYPER MassARRAY (Agena). A total of 13 CpG Sequenom units were analyzed in the small CpG island located in the only translated ZBTB12 exon. Principal component analysis (PCA) was performed to identify groups of CpG units with similar methylation estimates. Linear mixed effect regressions showed a positive association between methylation of ZBTB12 Factor 2 (including CpG units 8, 9–10, 16, 21) and TNF-ɑ stimulated procoagulant activity, a measure of procoagulant and inflammatory potential of blood cells. In addition, we also found a negative association between methylation of ZBTB12 Factor 1 (mainly characterized by CpG units 1, 3–4, 5, 11, and 26) and white blood cell and granulocyte counts. An in silico prediction analysis identified granulopoiesis- and hematopoiesis-specific transcription factors to potentially bind DNA sequences encompassing CpG1, CpG3–4, and CpG11. Conclusions ZBTB12 hypomethylation is linked to shorter TNF-ɑ stimulated whole blood coagulation time and increased WBC and granulocyte counts, further elucidating the possible link between ZBTB12 methylation and cardiovascular disease risk. Abstract

BackgroundPlatelet-endothelial aggregation receptor 1 (PEAR-1) is a transmembrane receptor involved in platelet activation and megakaryopoiesis whose expression is driven by DNA methylation.PEAR1variants were associated with differential platelet response to activation and cardiovascular outcomes. We aimed at investigating the link betweenPEAR1methylation and platelet and leukocyte function markers in a family-based population.ResultsWe measuredPEAR1methylation in 605 Moli-family participants with available blood counts, plasma P-selectin and C-reactive protein, whole blood platelet P-selectin, and platelet-leukocyte mixed conjugate measurements. We performed principal component analysis (PCA) to identify groups of highly correlated CpG sites. We used linear mixed regression models (using age, gender, BMI, smoking, alcohol drinking, being a proband for family recruitment, being a member of myocardial infarction (MI) family as fixed effects, and family as a random effect) to evaluate associations betweenPEAR1methylation and phenotypes.PEAR1methylation Factor2, characterized by the previously identified megakaryocyte-specific CpG sites, was inversely associated with platelet-monocyte conjugates, P-selectin, and WBC counts, while positively associated with the platelet distribution width (PDW) and with leukocyte CD11b and L-selectin. Moreover,PEAR1Factor2 methylation was negatively associated with INFLAscore, a low-grade inflammation score. The latter was partially mediated by thePEAR1methylation effect on platelet variables.PEAR1methylation association with WBC measurements and INFLAscore was confirmed in the independent cohort FLEMENGHO.ConclusionsWe report a significant link between epigenetic signatures in a platelet functional gene and inflammation-dependent platelet function variability measured in two independent cohorts.
