23. Plasma diamine oxidase activity decline with diarrhea severity in calves indicating systemic dysfunction related to intestinal mucosal damage

type:Article, The aim of the present study was to investigate whether abnormalities in plasma diamine oxidase (DAO) activity reflect the degree of intestinal mucosal disorder in calves with diarrhea. A total of 50 Holstein calves were enrolled. Thirty-six of the 50 calves presented diarrhea and were sub-classified by severity based on fecal status (0: firm, 1: pasty, 2: loose, and 3: watery) and blood pH (acidemia: blood pH <7.25) as follows: Seventeen calves exhibiting watery diarrhea and/or fall into acidemia were sub-categorized into the severe group. The other nineteen calves exhibiting pasty or loose diarrhea and not fall into acidemia were sub-categorized into the moderate group. The remaining 14 calves without diarrhea were assigned to the control group. The plasma DAO activity was significantly lower (p < .01) in the calves with severe or moderate diarrhea than in the control group. In addition, the plasma DAO activity was significantly lower (p < .05) in the severe group than in the moderate group. The relationship between plasma DAO activity and fecal score (r = −0.55, p < .01) in calves with diarrhea were found to have significantly and negatively correlated by Spearman's rank test in this study. Our results suggested that plasma DAO activity reflect the degree of intestinal mucosal disorder due to diarrhea.

Abstract

第1報にて，試作した乳質測定装置を用いて静置した状態で生乳の乳成分を高い精度で測定できることがわかった。本報では実際の搾乳時に本装置を用いて，乳質（乳成分 : 乳脂肪，乳タンパク質，乳糖）の測定を行った。その結果，乳脂肪の測定精度は決定係数r2＝0.94，標準誤差SEP＝0.55％，乳タンパク質の測定精度はr2＝0.80，SEP＝0.13％，乳糖の測定精度はr2＝0.83，SEP＝0.09％と，いずれも良好であった。さらに，試作した装置により搾乳時の乳成分をリアルタイムに測定することができた。また，乳質測定装置で測定した乳成分と搾乳時の乳量から搾乳1回分の乳成分を算出したところ，実測値と良く一致した。, We previously showed in part 1 that a near-infrared spectroscopic sensing system constructed on an experimental basis can be used to assess milk quality in the laboratory. In this study, we developed calibration models for predicting three major milk constituents (fat, protein and lactose) during milking by the sensing system and validated the precision and accuracy of the models. Coefficient of determination (r2) and standard error of prediction (SEP) of the validation set for fat were 0.94 and 0.55%, respectively. The values of r2 and SEP for protein were 0.80 and 0.13%, respectively, and the values of r2 and SEP for lactose were 0.83 and 0.09%, respectively. These results indicate that the sensing system can be used to assess milk quality in real-time during milking. From the data of milk constituents measured by the sensing system and milk flow rate during milking, we calculated milk constituents of whole one milking. The calculated values of milk constituents well agreed with the reference values.

Abstract

搾乳時における乳質の連続測定技術の開発を目標に，近赤外分光法を用いた乳質測定装置を試作した。乳成分を測定する検量線を作成し，装置の測定精度を検証した。その結果，乳成分を精度良く測定できた。また実用を想定し，生乳の温度変動が測定精度に与える影響を検証した。その結果，検量線作成に用いた試料の温度範囲内の生乳であれば，乳成分を高い精度で測定できた。よって様々な温度の試料を採取し検量線作成に用いる事で，検量線の温度補償が可能になる事がわかった。, We constructed an online near-infrared spectroscopic sensing system on an experimental basis. Calibration models for determining three major milk constituents (fat, protein and lactose) were developed, and the precision and accuracy of the models were validated. The sensing system can be used to assess milk quality with sufficient precision and accuracy. Moreover we evaluated the influence of milk temperature on the performance of calibration models in practical use. When the temperature of the validation subset was in the temperature range of the calibration subset, the performance of calibration models was good. Therefore, temperature compensation of a calibration model was possible using calibration subset that covers a variation in temperature of the sample to be measured.

Abstract

The present study examined the effects of substituting kraft pulp (KP) with corn silage (CS) on dry matter intake (DMI), ruminal mat and rumen fermentation characteristics, and rumination. Four non‐lactating, rumen‐cannulated Holstein cows were fed a CS diet comprising 36% grass silage (GS) and 64% CS or a KP diet comprising 36% GS, 57% KP, and 7% soybean meal. DMI was significantly lower in cows fed the KP diet than in those fed the CS diet (p < 0.05), whereas rumination time did not significantly differ between the treatments. Dry matter content in the rumen immediately before and 3 h after feeding was significantly higher in cows fed the KP diet than in those fed the CS diet (p < 0.05). The consistency and thickness of the ruminal mat did not significantly differ between the treatments. The ruminal mean retention time of feed particles tended to be longer in cows fed the KP diet than in those fed the CS diet (p < 0.10). The ruminal digestion rate of KP was comparable to that of GS and CS. Because ruminal mat was formed and rumination was stimulated, KP was considered to have the equivalent physical effectiveness as CS. [ABSTRACT FROM AUTHOR]

Abstract

Agriculture in Hokkaido dawned in 1868; the time when the Meiji Government was established. Warriors who were deprived of their positions by the new government after the Meiji Revolution, and farmers who could afford only small area in the main islands of Japan, immigrated to Hokkaido. They cleared the land and planted crops facing sever winters of Hokkaido. Presently, Hokkaido leads agriculture in Japan, proudly tops the production of rice, wheat, potato, beans, sugar beet, vegetables, and especially dairy products. The people of Hokkaido, however, shall never forget that it is the hard work of their predecessors for more than hundred years that has earned the present position for Hokkaido. At the same time, misunderstanding between immigrants and native Ainu people, and environmental degradation caused by reclamation of Hokkaido, should be matters of concern. This book describes past, present and future of Hokkaido's agriculture. The articles for this have been contributed by the professors outstanding in their respective fields in Hokkaido University. All chapters have been written in a simple language and illustrated with relevant photographs. Though this book has been produced as a textbook for the foreign students at Hokkaido University, it can serve a wide range of readers. It will be a great pleasure for us if this book is read by all concerned including visitors to this lovely land of Hokkaido.

44. Plasma and urine levels of electrolytes, urea and steroid hormones involved in osmoregulation of cetaceans

Abstract

Cetaceans are well adapted to their hyperosmotic environment by properly developed osmoregulatory ability. A question here is how they regulate water and mineral balances in marine habitats. In the present study, we determined blood and urine levels of various chemicals involved in osmoregulation, compared them with those in artiodactyls, and characterized the values in the whales. Blood and urine samples obtained from baleen whales of common minke (Balaenoptera acutorostrata), sei (B. borealis), and Bryde's whales (B. brydei), and toothed whales of sperm whales (Physeter macrocephalus) were analyzed for osmolality, major electrolytes, urea, steroid hormones and glucose. The urine osmolality and Na+ concentrations in the cetaceans were much higher than those in the cattle. Furthermore, the cetaceans had 5 to 11-fold urea in plasma than the cattle, and 2 to 4-fold urea in urine. There were no significant difference in the plasma concentrations of corticosteroids between the cetaceans and the cattle. The present results indicate that the osmoregulatory parameters seem to be not affected by the reproductive stage and sex steroid hormones. The concentrations of urea in plasma and urine of the baleen whales were higher than those of the sperm whales, indicating a possibility that their osmoregulatory mechanisms may be correlated to their feeding habits. The present results suggest that cetaceans have unique osmoregulatory mechanisms by which they excrete strongly hypertonic urine to maintain fluid homeostasis in marine habitats.
