The double sucrose-gap method was applied to single muscle fibers of Xenopus. From the "artificial node" of the fiber, action potentials were recorded under current-clamping condition together with twitches of the node. The action potentials were stored on magnetic tape. The node was then made inexcitable by tetrodotoxin or by a sodium-free solution, and the wave form of the action potential stored on magnetic tape was imposed on the node under voltage-clamp condition (simulated AP). The twitch height caused by the simulated AP's was always smaller than the twitch height produced by the real action potentials, the ratio being about 0.3 at room temperature. The results strongly suggest that the transverse tubular system is excitable and is necessary for the full activation of twitch, and that the action potential of the tubules contributes to about 70 % of the total mechanical output of the normal isotonic twitch at 20°C. Similar results were obtained in the case of tetanic contraction. At a temperature near 10°C, twitches produced by the simulated AP were not very different (85 % of control amplitude) from the twitches caused by real action potentials. This indicates that the excitability of the tubules becomes less necessary for the full activation of twitch as the temperature becomes lower.

Passive electrical properties (internal conductance, membrane conductance, low frequency capacity, and high frequency capacity obtained from the foot of the action potential) of normal and glycerol-treated muscle of Xenopus were determined with the intracellular microelectrode technique. The results show that the electrical properties of Xenopus muscle are essentially the same as those of frog muscle. Characteristics of the action potential of Xenopus muscle were also similar to those of frog muscle. Twitch tension of glycerol-treated muscle fibers of Xenopus recovered partially when left in normal Ringer for a long time (more than 6 h). Along with the twitch recovery, the membrane capacity increased. Single isolated muscle fibers of Xenopus were subjected to the double sucrose-gap technique. Action potentials under the sucrose gap were not very different from those obtained with the intracellular electrode, except for the sucrose-gap hyperpolarization and a slight tendency toward prolongation of the shape of action potential. Twitch contraction of the artificial node was recorded as a change of force from one end of the fiber under the sucrose gap. From the time-course of the recorded force and the sinusoidal stress-strain relationship at varying frequencies of the resting muscle fiber, the time-course of isotonic shortening of the node was recovered by using Fourier analysis. It was revealed that the recorded twitch force can approximately be regarded as isotonic shortening of the node.

Voltage clamp analyses, combined with pharmacological tools demonstrate the independence of reactive Na and K channels in electrically excitable membrane of eel electroplaques. Spike electrogenesis is due to Na activation and is eliminated by tetrodotoxin or mussel poison, or by substituting choline, K, Cs, or Rb for Na in the medium. The K channels remain reactive, but K activation is always absent, the electroplaques responding only with K inactivation. This is indicated by an increased resistance when the membrane is depolarized by more than about 30 mv. The resting resistance (1 to 5 ohm cm2) is dependent upon the ionic conditions, but when K inactivation occurs the resistance becomes about 10 ohm cm2 in all conditions. K inactivation does not change the EMF significantly. The transition from low to high resistance may give rise to a negative-slope voltage current characteristic, and to regenerative inactivation responses under current clamp. The further demonstration that pharmacological K inactivation (by Cs or Rb) leaves Na activation and spike electrogenesis unaffected emphasizes the independence of the reactive processes and suggests different chemical compositions for the membrane structures through which they operate.

Voltage clamp measurements on squid giant axons show that externally applied puffer fish poison, tetrodotoxin, eliminates only the initial inward current component of spike electrogenesis and does not affect the subsequent outward current. The selective effect on Na activation, which is reversible, confirms the view that the movements of Na and K during spike electrogenesis occur at structurally different sites on the membrane. Spike electrogenesis is also blocked when tetrodotoxin is injected into the axon, but the interior of the membrane appears to be somewhat less sensitive to the poison. Differences in reactivity of various electrogenic membrane components to tetrodotoxin are discussed as signifying differences in chemical structures.

Delayed rectification was elicited in frog's skeletal muscles bathed in choline-Ringer's solution, in normal Ringer's solution with tetrodotoxin, in 40 mM Na2SO4 solution with tetrodotoxin, and even in 40 mM K2SO4 solution when the membrane had been previously hyperpolarized. However, after a sustained depolarization current-voltage relations in 40 mM K2SO4 and in 40 mM Na2SO4 solutions revealed a rectifier property in the anomalous direction. This indicates that the increase in potassium conductance which is brought about upon depolarization is a transient phenomenon and is inactivated by a maintained depolarization, and that this potassium inactivation process converts the delayed rectification into the anomalous rectification. In normal Ringer's solution with tetrodotoxin and in the 40 mM Na2SO4 solution with tetrodotoxin the apparent resistance was increased when the membrane was hyperpolarized beyond about -150 mv. This is thought to be due to a decrease of K conductance caused by a strong hyperpolarizing current. In the 40 mM Na2SO4 solution with tetrodotoxin a de- or hyperpolarizing current pulse induced a prolonged depolarizing response. During the early phase of this response the effective resistance was lower, and during the following phase greater than that in the resting fiber. An interpretation in terms of the ionic hypothesis was made of the nature of this response.
