Abstract

The advancement of Large Language Models (LLM) has also resulted in an equivalent proliferation in its applications. Software design, being one, has gained tremendous benefits in using LLMs as an interface component that extends fixed user stories. However, inclusion of LLM-based AI agents in software design often poses unexpected challenges, especially in the estimation of development efforts. Through the example of UI-based user stories, we provide a comparison against traditional methods and propose a new way to enhance specifications of natural language-based questions that allows for the estimation of development effort by taking into account data sources, interfaces and algorithms.

Abstract

Human pluripotent stem cell-derived cardiomyocytes (hPSC-CMs) constitute an appealing tool for drug discovery, disease modeling, and cardiotoxicity screening. However, their physiological immaturity, resembling CMs in the late fetal stage, limits their utility. Herein, we have developed a novel, scalable cell culture medium designed to enhance the maturation of hPSC-CMs. This medium facilitates a metabolic shift towards fatty acid utilization and augments mitochondrial function by targeting Acetyl-CoA carboxylase 2 (ACC2) with a specific small molecule inhibitor. Our findings demonstrate that this maturation protocol significantly advances the metabolic, structural, molecular and functional maturity of hPSC-CMs at various stages of differentiation. Furthermore, it enables the creation of cardiac microtissues with superior structural integrity and contractile properties. Notably, hPSC-CMs cultured in this optimized maturation medium display increased accuracy in modeling a hypertrophic cardiac phenotype following acute endothelin-1 induction and show a strong correlation between in vitro and in vivo target engagement in drug screening efforts. Abstract

Civil engineers are at times required to stabilize sulfate-bearing clay soils with calcium-based stabilizers. Deleterious heaving in these stabilized soils may result over time. This paper addresses critical questions regarding the consequences of treating sulfate laden soils with calcium-based stabilizers. The authors describe the nature (chemistry and structure) of the minerals (ettringite/thaumasite) blamed for deleterious reactions and explain why these structures may lead to damage. The writers also describe the mechanisms of the mineral growth, and the extent of mineral growth based on the amount of sulfate minerals present in the soil. The writers explain why the rate of ettringite growth in treated soils should not be expected to follow a controlled rate of ettringite development such as that which normally occurs in portland cement concrete. The writers compare the rate and degree of ettringite development in soils to the classical model of nucleation and growth typical of most crystal structures. Finally, the writers evaluate the role of soil mineralogy in controlling soil behavior at varying sulfate contents and verify the existence of a threshold level of soluble sulfates in soils that can trigger substantial ettringite growth. Abstract

There are numerous electronic loads connected to ac grid which requires dc supply for which rectifiers are used. Now this depreciates the power factor at the ac side. In order to improve power factor boost type or buck-boost type PFC (Power Factor Correction) converters are used. This paper proposes a new buck-boost type PFC based converter with one cycle current controller (OCCC). OCCC with voltage loop is a novel controller for controlling the line current and make it follow the input voltage improving the power factor of the system. The main feature is the topology of buck-boost type PFC which is similar to non-inverting buck-boost converter with modified bridge rectifier and the control strategy ensures low THD (Total Harmonic Distortion) of input line current.

Abstract

Conventional DC-DC boost converters are unable to provide high step-up voltage gains due to the effect of power switches, rectifier diodes and the equivalent series resistance of inductors and capacitors. In this paper, the closed loop design and analysis of voltage lift technique based non-isolated dc-dc boost converter is carried out to achieve high step-up voltage gain. In the presented converter two inductors, three capacitors and three power diodes are used. The structure of the presented converter is to be considered a very simple, since it has only two power stages. The voltage and current equations of each element with converter voltage gain under continuous conduction mode (CCM) are extracted. Moreover, the steady state analysis of voltage gains and boundary conditions are discussed in this paper. Finally, the performance verification is carried out with a prototype circuit implemented in the laboratory.

Abstract

Although transdermal route of drug delivery displays numerous benefits over conventional strategies, this pathway is yet to presents its complete potential. The chief ground for this statement is ascribed to the presence of extremely packed outermost layer of skin called stratum corneum. This compactly packed barrier is selectively permeable and furnishes smaller and lipophilic molecules to diffuse into the deeper skin layers. Due to this only a few transdermal product is commercially available. So in order to guarantee the effective transport of drug molecules, it is necessary to break the stratum corneum layer. There are two acceptable methods to break the stratum corneum architecture- active strategy which comprises the application of external energy such as in electroporation, sonophoresis, iontophoresis etc. and passive strategy which involve the application of permeation enhancers, nanoparticles etc. This article comprehensively detail the active and passive methods employed in transdermal drug delivery systems to disrupt the stratum corneum bilayers. Abstract

Skin is an attractive route for drug delivery. However poor permeation of drugs across the skin due to the presence of extremely ordered architecture of outermost layer of skin, led to several investigation to improve the permeability of drugs. Polysaccharides remain widely studied biomaterial for the sustained delivery of drug molecules across the skin. The advance of hyaluronic acid (HA) chemistry with multiple benefits has improved the attention of research groups for its application in the skin transportation of drug molecules. Beginning from the advantages of transdermal route, the present review details the application of HA in transdermal drug delivery. In the last few decades, substantial investigation in the domain has improved the requirement for an outline of all the developments, which is depicted in the review. The review also presented different types of HA based transdermal devices such as transferosomes, nanoemulsions, microneedle etc and their potential to improve the transdermal drug delivery. Furthermore the application of HA through chemical modification as a potential transdermal device is also presented.
