Initiation of colorectal carcinogenesis may be induced by chromosomal instability caused by oxidative stress or indirectly by bacterial infections. Moreover, proliferating tumor cells are characterized by reprogrammed glucose metabolism, which is associated with upregulation of PDK1 and LDHA enzymes. In the present study, some 4,5,6,7-tetrahydrobenzo[b]thiophene derivatives in addition to Fe3O4 and Fe3O4/SiO2 nanoparticles (NPs) supported with a new Schiff base were synthesized for biological evaluation as PDK1 and LDHA inhibitors as well as antibacterial, antioxidant, and cytotoxic agents on LoVo and HCT-116 cells of colorectal cancer (CRC). The results showed that compound 1b is the most active as PDK1 and LDHA inhibitor with IC50 values (μg/mL) of 57.10 and 64.10 compared to 25.75 and 15.60, which were produced by the standard inhibitors sodium dichloroacetate and sodium oxamate, respectively. NPs12a,b and compound 1b exhibited the strongest antioxidant properties with IC50 values (μg/mL) of 80.0, 95.0, and 110.0 μg/mL, respectively, compared to 54.0 μg/mL, which was produced by butylated hydroxy toluene. Moreover, NPs12a and carbamate derivative 3b exhibited significant cytotoxic activities with IC50 values (μg/mL) of 57.15 and 81.50 (LoVo cells) and 60.35 and 71.00 (HCT-116 cells). Thus, NPs12a and compound 3b would be considered as promising candidates suitable for further optimization to develop new chemopreventive and chemotherapeutic agents against these types of CRC cell lines. Besides, molecular docking in the colchicine binding site of the tubulin (TUB) domain revealed a good binding affinity of 3b to the protein; in addition, the absorption, distribution, metabolism, and excretion (ADME) analyses showed its desirable drug-likeness and oral bioavailability characteristics.

In this study, the chemical compositions of the ethanolic and aqueous extracts of the leaves of Origanum syriacum and Salvia lanigera were identified based on GC-MS spectrometric analyses. The in vitro anti-inflammatory potential of the different extracts was evaluated by determining the membrane stabilization of human red blood cells and the percent inhibition of the COX1/2, 5LOX, and sPLA2-V enzymes. Both ethanolic extracts showed maximum membrane stabilization (≤ 91%, at 100 μg/mL) compared to the aqueous extracts (≤ 45%) and the reference drug diclofenac sodium (90.75%). The membrane-stabilizing effects of the ethanolic extracts could be directly correlated to their anti-inflammatory activity. While both ethanolic fractions strongly inhibited the 5LOX and COX-1 enzymes at 100 μg/mL, only the O. syriacum ethanolic extract selectively inhibited sPLA2-V (99.35%, at 50 μg/mL). The differences in the pharmacological efficiencies of the different extracts could be attributed to the variation in their chemical compositions particularly the content of oxygenated monoterpenoids. Additionally, none of the ethanolic extracts demonstrated cytotoxicity to human colorectal cancer cell lines (HCT-116 and Lovo), even at the highest concentration tested (200 μg/mL). The safe profiles of these extracts towards the tested cell lines may be due to the absence of the toxic phthalic acid ester substances. Collectively, these findings clearly suggest that the studied ethanolic extracts of O. syriacum and S. lanigera can be considered interesting candidates for the treatment of human inflammatory diseases related to oxidative stress and microbial infections.

In the present work, QSAR (quantitative structure−activity relationship) analysis has been executed for RG7834 analogs. RG7834 is a first-in- class selective and orally available dihydroquinolizinone (DHQ)-based small molecule Hepatitis B virus expression inhibitor. OECD’s guidelines have been followed for developing multiple QSAR models for Hepatitis B virus expression inhibitory activity of 73 RG7834 analogs. The present multiple QSAR models are not only easily interpretable but possess high external predictive ability, as well. These are effective in the recognition of many privileged and underprivileged molecular descriptors, which could be very valuable for the use of these models by the experts and nonexperts of QSAR in future optimizations. The models satisfy threshold values for many fitting, internal and external validation parameters, such as R2 = 0.83, Q2 = 0.80, CCCext = 0.88, etc., thereby demonstrating good external predictive ability of the models. The multiple QSAR and pharmacophoric models successfully identified a good number of important positively and negatively related structural features of RG7834 analogs that govern their Hepatitis B virus expression inhibitory activity. The results could be very beneficial to synthetic/medicinal chemists for future alterations of RG7834 analogs as better drug candidates.

In the present work, OECD guidelines have been followed for developing QSAR (Quantitative Structure-Activity Relationship) models for anti-HAT (Human African trypanosomiasis) activity of two hundred and seventy pyrazole sulphonamides. The newly developed easily interpretable multiple QSAR models have been successful in identification of many privileged as well as under-privileged molecular descriptors, which could be highly useful for future use of these models by expert and non-experts of QSAR. The multiple QSAR models satisfy threshold values for many statistical parameters such as R2 = 0.80–0.83, Q2 = 0.79–0.81, CCCext = 0.82–0.84, etc. thereby assuring good external predictive ability of the models. The multiple QSAR and pharmacophoric models successfully identified that N,4-disubstituted-benzenesulfonamide moiety, frequency of occurrence of H-bond acceptor from Oxygen atom within nine bonds and some other pharmacophoric features that govern the anti-HAT activity of pyrazole sulphonamides. The results could be very useful to synthetic/medicinal chemists for future modifications of pyrazole sulphonamides as drug candidates.

The pathogenesis of colorectal cancer is a multifactorial process. Dysbiosis and the overexpression of COX-2 and LDHA are important effectors in the initiation and development of the disease through chromosomal instability, PGE2 biosynthesis, and induction of the Warburg effect, respectively. Herein, we report the in vitro testing of some new quinoxalinone and quinazolinone Schiff’s bases as: antibacterial, COX-2 and LDHA inhibitors, and anticolorectal agents on HCT-116 and LoVo cells. Moreover, molecular docking and SAR analyses were performed to identify the structural features contributing to the biological activities. Among the synthesized molecules, the most active cytotoxic agent, (6d) was also a COX-2 inhibitor. In silico ADMET studies predicted that (6d) would have high Caco-2 permeability, and %HIA (99.58%), with low BBB permeability, zero hepatotoxicity, and zero risk of sudden cardiac arrest, or mutagenicity. Further, (6d) is not a potential P-gp substrate, instead, it is a possible P-gpI and II inhibitor, therefore, it can prevent or reverse the multidrug resistance of the anticancer drugs. Collectively, (6d) can be considered as a promising lead suitable for further optimization to develop anti-CRC agents or glycoproteins inhibitors.

Due to the genetic similarity between SARS-CoV-2 and SARS-CoV, the present work endeavored to derive a balanced Quantitative Structure−Activity Relationship (QSAR) model, molecular docking, and molecular dynamics (MD) simulation studies to identify novel molecules having inhibitory potential against the main protease (Mpro) of SARS-CoV-2. The QSAR analysis developed on multivariate GA–MLR (Genetic Algorithm–Multilinear Regression) model with acceptable statistical performance (R2 = 0.898, Q2loo = 0.859, etc.). QSAR analysis attributed the good correlation with different types of atoms like non-ring Carbons and Nitrogens, amide Nitrogen, sp2-hybridized Carbons, etc. Thus, the QSAR model has a good balance of qualitative and quantitative requirements (balanced QSAR model) and satisfies the Organisation for Economic Co-operation and Development (OECD) guidelines. After that, a QSAR-based virtual screening of 26,467 food compounds and 360 heterocyclic variants of molecule 1 (benzotriazole–indole hybrid molecule) helped to identify promising hits. Furthermore, the molecular docking and molecular dynamics (MD) simulations of Mpro with molecule 1 recognized the structural motifs with significant stability. Molecular docking and QSAR provided consensus and complementary results. The validated analyses are capable of optimizing a drug/lead candidate for better inhibitory activity against the main protease of SARS-CoV-2.

Multiple discrete quantitative structure–activity relationships (QSARs) models were constructed for the anticancer activity of α, β-unsaturated carbonyl-based compounds, oxime and oxime ether analogues with a variety of substituents like Br, OH, –OMe, etc. at different positions. A big pool of descriptors was considered for QSAR model building. Genetic algorithm (GA), available in QSARINS-Chem, was executed to choose optimum number and set of descriptors to create the multi-linear regression equations for a dataset of sixty-nine compounds. The newly developed five parametric models were subjected to exhaustive internal and external validation along with Y-scrambling using QSARINS-Chem, according to the OECD principles for QSAR model validation. The models were built using easily interpretable descriptors and accepted after confirming statistically robustness with high external predictive ability. The five parametric models were found to have R2 = 0.80 to 0.86, R2ex = 0.75 to 0.84, and CCCex = 0.85 to 0.90. The models indicate that frequency of nitrogen and oxygen atoms separated by five bonds from each other and internal electronic environment of the molecule have correlation with the anticancer activity.

Synthesis of some new pyrazole, pyridazine, pyridine, chalcone, hydrazone derivatives containing thiophene ring has been carried out by simple, efficient and good yielding routes starting from the versatile and readily accessible ethyl 5-acetyl-4-phenyl- 2-(phenylamino)thiophene-3-carboxylate (1). The structure of the products has been elucidated from their spectral and elemental analyses. X-ray crystallography of compound 1 confirmed its structure.

In the present work, sixty substituted 2-Phenylimidazopyridines previously reported with potent anti-human African trypanosomiasis (HAT) activity were selected to build genetic algorithm (GA) based QSAR models to determine the structural features that have significant correlation with the activity. Multiple QSAR models were built using easily interpretable descriptors that are directly associated with the presence or the absence of a structural scaffold, or a specific atom. All the QSAR models have been thoroughly validated according to the OECD principles. All the QSAR models are statistically very robust (R2 = 0.80–0.87) with high external predictive ability (CCCex = 0.81–0.92). The QSAR analysis reveals that the HAT activity has good correlation with the presence of five membered rings in the molecule.

Fine particulate matter (PM 2.5) is the principal instigators of adverse health events, yet gaps still remain in understanding the mechanism mediating its toxic response. Similar to nanoparticles, PM 2.5, with large surface area to volume ratio, can absorb multipollutants in air, displaying toxicity profiles that are very different from those of coarse particles of the same composition. One particularly relevant interaction is that of PM 2.5 and the anthropogenic metals. In this study, we used carbon black nanoparticle (CBs) and metal ions as model materials to investigate the synergistic pulmonary toxicity and its mechanism. We demonstrated that excessive metal contaminants adsorbed on CBs contributed to the observed toxic effects both in vitro and in C57BL6 mice intratracheal instillation model. Significantly, we found that autophagy and lysosomal dysfunction accounted for the synergistic pulmonary toxic effect of co-exposure to CBs and metals. Our findings provide a new insight into understanding the toxicological and healthy effects of fine particles, which have potential to aid in mitigating their adverse health effect.

Some new 3H-quinazolin-4-one derivatives were synthesised and screened for anticancer, antiphospholipases, antiproteases, and antimetabolic syndrome activities. Compound 15d was more potent in reducing the cell viabilities of HT-29 and SW620 cells lines to 38%, 36.7%, compared to 5-FU which demonstrated cell viabilities of 65.9 and 42.7% respectively. The IC50 values of 15d were ∼20 µg/ml. Assessment of apoptotic activity revealed that 15d decreased the cell viability by down regulating Bcl2 and BclxL. Moreover, compounds, 8j, 8d/15a/15e, 5b, and 8f displayed lowered IC50 values than oleanolic acid against proinflammatory isoforms of hGV, hG-X, NmPLA2, and AmPLA2. In addition, 8d, 8h, 8j, 15a, 15b, 15e, and 15f showed better anti-α-amylase than quercetin, whereas 8g, 8h, and 8i showed higher anti-α-glucosidase activity than allopurinol. Thus, these compounds can be considered as potential antidiabetic agents. Finally, none of the compounds showed higher antiproteases or xanthine oxidase activities than the used reference drugs.

ABBV-075 (Mivebresib) analogues were reported to have BET family bromodomain binding affinity Ki in nano-molar (nM) range (0.8 to 3000 nM). But future optimizations are required to achieve a drug-like molecule with retention of high binding affinity and optimum ADMET (Absorption, Distribution, Metabolism, Excretion and Toxicity) profile. This could be achieved by identifying the pharmacophoric features (salient and concealed) using modern techniques like CADD (Computer-Aided Drug Designing). Therefore, in the present work, QSAR (Quantitative structure-activity relationship) analysis, a thriving CADD branch, has been executed to achieve the determined objectives. The developed QSAR model is statistically acceptable with robust fitting, high internal and external predictive ability. The developed model fulfils the threshold values for a good number of statistical parameters like R2 = 0.80, R2CV = 0.77, etc. The analysis reveals that non-ring Carbon/Nitrogen atoms, frequency of occurrence of specific combinations of Carbon/Nitrogen atoms with acceptor/donor atoms are important pharmacophoric features for BET binding affinity. Thus, the developed QSAR has a balance of quantitative and qualitative approaches. The results could be useful for future optimizations of Mivebresib analogues .

Autophagy is a basic cellular process that decomposes damaged organelles and aberrant proteins. Dysregulation of autophagy is implicated in pathogenesis of neurodegenerative disorders, including Parkinson’s disease (PD). Pharmacological compounds that stimulate autophagy can provide neuroprotection in models of PD. Nanoparticles have emerged as regulators of autophagy and have been tested in adjuvant therapy for diseases. In this present study, we explore the effects of quantum dots (QDs) that can induce autophagy in a cellular model of Parkinson’s disease. CdTe/CdS/ZnS QDs protect differentiated rat pheochromocytoma PC12 cells from MPP+-induced cell damage, including reduced viability, apoptosis and accumulation of α-Synuclein, a characteristic protein of PD. The protective function of QDs is autophagy-dependent. In addition, we investigate the interaction between quantum dots and autophagic pathways and identify beclin1 as an essential factor for QDs-induced autophagy. Our results reveal new promise of QDs in the theranostic of neurodegenerative diseases.

On-site detection of biomarkers in biofluids found at the crime scene is critically important forensic analysis. However, it remains difficult due to the lack of portable, fast and cheap on-site analytical devices. Traditional methods including polymerase chain reaction (PCR) and electrophoresis requires complicated instrumentation and critical environment. Optical analytical methods can be affected by intrinsic adsorption of complicated sample such as serum. In response, we developed a smartphone-interfaced electrochemical chip device for on-site gender verification. The detection is based on the known difference of biomarkers (creatine kinase (CK) and alanine transaminase (ALT)) between male and female groups. Enzyme cascade reaction converted the enzyme level in biofluids to the consumption of NADH, which can be electrochemically detected by our designed electrochemical chip. Our device retained the capability of gender verification when used in serum and serum stains. The detection can be completed in 20 min. Gender verification of real samples (39 serum samples) demonstrated excellent sensitivity and specificity of this smartphone based device.

Many types of biocompatible nanomaterials have proven of low cytotoxicity and hold great promise for various applications in nanomedicine. Whereas they generally do not cause apparent organ toxicity or tissue damage in adult animals, it is yet to determine their biological consequences in more general contexts. In this study, we investigate how silica nanoparticles (NPs) affect cellular activities and functions under several physiological or pathological conditions. Although silica NPs are generally regarded as "inert" nanocarriers and widely employed in biomedical studies, we find that they actively affect Wnt signaling in various types of cell lines, diminishing its anti-adipogenic effect in preadipocytes and pro-invasive effect in breast cancer cells, and more significantly, impair Wnt-regulated embryonic development in Zebrafish. We further demonstrate that intracellular silica NPs block Wnt signal transduction in a way resembling signaling molecules. Specifically, silica NPs target the Dvl protein, a key component of Wnt signaling cascade, for lysosomal degradation. As Wnt signaling play significant roles in embryonic development and adipogenesis, the observed physiological effects beyond toxicity imply potential risk of obesity, or developmental defects in somitogenesis and osteogenesis upon exposure to silica NPs. In addition, given the clinical implications of Wnt signaling in tumorigenesis and cancer metastasis, our work also establishes for the first time a molecular link between nanomaterials and the Wnt signaling pathway, which opens new door for novel applications of unmodified silica NPs in targeted therapy for cancers and other critical illness.

In the present work, 2D- and 3D-quantitative structure-activity relationship (QSAR) analysis has been employed for a diverse set of eighty-nine quinoxalinones to identify the pharmacophoric features with significant correlation with the aldose reductase inhibitory activity. Using genetic algorithm (GA) as a variable selection method, multivariate linear regression (MLR) models were derived using a pool of molecular descriptors. All the six-descriptor based GA-MLR QSAR models are statistically robust with coefficient of determination (R2 )>0.80 and cross-validated R2 >0.77. The derived GA-MLR models were thoroughly validated using internal and external and Y-scrambling techniques. The CoMFA like model, which is based on a combination of steric and electrostatic effects and graphically inferred using contour plots, is highly robust with R2 >0.93 and cross-validated R2 >0.73. The established QSAR and CoMFA like models are proficient in identify key pharmacophoric features that govern the aldose reductase inhibitory activity of quinoxalinones.

Multiple separate quantitative structure–activity relationships (QSARs) models were built for the antiproliferative activity of substituted Phenyl 4-(2-Oxoimidazolidin-1-yl)-benzenesulfonates (PIB-SOs). A variety of descriptors were considered for PIB-SOs through QSAR model building. Genetic algorithm (GA), available in QSARINS, was employed to select optimum number and set of descriptors to build the multi-linear regression equations for a dataset of PIB-SOs. The best three parametric models were subjected to thorough internal and external validation along with Y -randomization using QSARINS, according to the OECD principles for QSAR model validation. The models were found to be statistically robust with high external predictivity. The best three parametric model, based on steric, 3D- and finger print descriptors, was found to have R 2 = 0.91, R 2 ex = 0.89, and CCC ex = 0.94. The CoMFA model, which is based on a combination of steric and electrostatic effects and graphically inferred using contour plots, gave F = 229.34, R 2 CV = 0.71 and R 2 = 0.94. Steric repulsion, frequency of occurrence of carbon and nitrogen at topological distance of seven, and internal electronic environment of the molecule were found to have correlation with the anti-tumor activity of PIB-SOs.

Some novel hydrazone derivatives 6a-o were synthesized from the key intermediate 4-Chloro-N-(2-hydrazinocarbonyl-phenyl)-benzamide 5 and characterized using IR, 1H-NMR, 13C-NMR, mass spectroscopy and elemental analysis. The inhibitory potential against two secretory phospholipase A2 (sPLA2), three protease enzymes and eleven bacterial strains were evaluated. The results revealed that all compounds showed preferential inhibition towards hGIIA isoform of sPLA2 rather than DrG-IB with compounds 6l and 6e being the most active. The tested compounds exhibited excellent antiprotease activity against proteinase K and protease from Bacillus sp. with compound 6l being the most active against both enzymes. Furthermore, the maximum zones of inhibition against bacterial growth were exhibited by compounds; 6a, 6m, and 6o against P. aeruginosa; 6a, 6b, 6d, 6f, 6l, 6m, 6n, and 6o against Serratia; 6k against S. mutans; and compounds 6a, 6d, 6e, 6m, and 6n against E. feacalis. The docking simulations of hydrazones 6a-o with GIIA sPLA2, proteinase K and hydrazones 6a-e with glutamine-fructose-6-phosphate transaminase were performed to obtain information regarding the mechanism of action.

Several new pyrazole, pyridine, [1,2,4]triazolo[1,5-α]pyrimidine, benzimidazo[1,2-a]pyrimidine and 1,2,4-triazolo[3,4-c][1,2,4]triazine derivatives incorporating a thiophene moiety were synthesized from (E)-ethyl 5-(3-(dimethylamino)acryloyl)-4-phenyl-2-(phenylamino)thiophene-3-carboxylate (1). The structures of the newly synthesized compounds were confirmed by IR, ¹H-, (13)C-NMR, mass spectral data and elemental analysis. The antibacterial and antifungal activities of all the synthesized compounds were evaluated. The results indicated that compounds 9, 12, and 19 were found to be more potent than the standard drug Amphotericin B against Aspergillus fumigates. Additionally, compound 12 exhibited higher activity than the standard drug Amphotericin B against Syncephalastrum racemosum.

The thiosemicarbazide derivatives 3a and 3b were cyclized in the presence of concentrated sulfuric acid to give the 5-cyanomethyl-1,3,4-thiadiazole derivatives 4a and 4b , respectively. The latter products were used for many heterocyclic transformations to form coumarin, 1,3,4-thiadiazolo[4,5-a]pyridine, and 5-thiophenylthiophene. In addition, compound 3b underwent cyclization in NaOH (2 N) solution to give the 1,2,4-triazole derivative 16 . The reactivity of the latter product towards some chemical reagents was studied. The antimicrobial activities of the newly synthesized products were measured and showed high activities.

The reaction of pregnenolone with cyanoacetylhydrazine and ammonium acetate at 120°C gave the Knoevenagel condensation product 3. The latter reacted with different reagents to give thiophene, thieno[2,3-d]pyrimidine, 1,2,4-triazole and pyran derivatives. The anti-inflammatory and anti-ulcer evaluations of the newly synthesized products were evaluated and the results showed that compounds 4, 8c, 10, 11, 13c, 15a, 15c, 17a, 17b, 17e, 18a and 18f possessed higher activity compared to the rest of the compounds. In addition to this, the toxicity of these active compounds was studied against shrimp larvae where compounds 15a, 15c and 18a showed non-toxicity against the tested organisms.

The reaction of pregnenolone with either 2-aminoprop-1-ene-1,1,3-tricarbonitrile or 3-oxo-3-phenylpropanenitrile gave the Knoevenagel condensation products 3 and 6, respectively. Separation of the E and Z isomeric compounds of 3 and 6 together with their structure elucidation were carried out. Some chemical transformations of the latter products were carried out and the cytotoxicity of the newly obtained products was evaluated against some cancer cell lines and a human normal cell line. The results indicated that compounds 15, 17a, 18 and 20e among the tested compounds showed the highest cytotoxicity against the cancer cell lines.

Biocompatible nanoparticles hold a great promise for biomedical applications, whereas their biosafety has raised extensive concerns. Nanodiamonds (NDs) are generally regarded as “inert” nanocarriers and widely employed in biomedical studies; however, it is yet to explore their biological effects in more general contexts. In this study, the authors observe that intracellular NDs block signal transduction of the Wnt signaling pathway, an effect that is not caused by general cytotoxicity. The authors find that NDs attenuate activities of Wnt signaling in several types of cell lines and in Zebrafish, and interfere with Wnt signaling-controlled biological processes, including cancer cell migration, adipocyte differentiation, and embryonic development. Significantly, the authors show that intracellular NDs trigger degradation of the disheveled protein, a key component of Wnt signaling cascade, while they do not affect protein stability of other Wnt signal transducers or other signaling molecules. This work thus illustrates a novel crosstalk between nanoparticles and the Wnt signaling pathway, and expands the understanding of biological effects induced by nanoparticles. In addition, given the clinical implications of Wnt signaling in tumorigenesis and cancer metastasis, this study also provides the rationale for potential applications of NDs in cancer therapies.

Tetrasubstituted 2-acetylthiophene derivative 5 was synthesized and then condensed with various nitrogen nucleophiles such as 5-amino-1,2,4-triazole, 2-aminobenzimidazole, aniline or p-chloroaniline to afford the corresponding iminothiophene derivatives 6-8a,b. Condensation of thiophene 5 with malononitrile as carbon nucleophile afforded compound 9, which underwent nucleophilic addition with DMF-DMA to afford compound 10. The newly synthesized products were characterized by elemental analysis, IR, MS, ¹H-(13)C-NMR and CHN analysis and then evaluated for their antimicrobial activity. Results of the in vitro antibacterial activity showed that thiophene derivative 7 was found to be more potent than the standard drug gentamicin against Pseudomonas aeruginosa. Some of these compounds showed potential antimicrobial activities. Molecular docking and Osiris/Molinspiration analyses show the crucial role and impact of substituents on bioactivity and indicate the unfavorable structural parameters in actual drug design: more substitution with electronic donor group doesn't guarantee more effective bioactivity. This study should greatly help in an intelligent and a controlled pharmacomodulation of antibiotics.
