We investigated the spatio-temporal variations in three key biomolecular compounds (carbohydrates, proteins, and lipids) in particulate organic matter (POM) in the Yellow Sea (YS), South Sea of Korea (SS), East China Sea (ECS), and East Sea (Sea of Japan; ES) in order to estimate the regional annual calorie production rates based on the seasonal data obtained in each region. Carbohydrates were found to be most dominant, followed by lipids across all seas. The euphotic-integral calorie contents of POM during the study period were determined as 53.5 ± 12.6 Kcal m−2 in the YS; 54.2 ± 7.5 Kcal m−2 in the SS; 35.7 ± 9.2 Kcal m−2 in the ECS; and 58.7 ± 6.2 Kcal m−2 in the ES. Utilizing seasonal carbon conversion factors and primary production rates, we estimated the annual calorie productions, which were highest in the ES (1705 Kcal m−2 y−1) and lowest in the ECS (403 Kcal m−2 y−1). This can be attributed to the significantly higher energy efficiency of phytoplankton and faster turnover rate of calorie content. However, caution must be taken when comparing these estimates regionally, considering potential variations in developmental phases among the four regions during the sampling period in 2018. The calorie production rates in this study provide valuable insights into the physiological condition of phytoplankton within specific regions.

Dramatic environmental changes have been recently reported in the Yellow Sea (YS), the South Sea of Korea (SS), and the East/Japan Sea (EJS), but little information on the regional primary productions is currently available. Using the 13C-15N tracer method, we measured primary productions in the YS, the SS, and the EJS for the first time in 2018 to understand the current status of marine ecosystems in the three distinct seas. The mean daily primary productions during the observation period ranged from 25.8 to 607.5 mg C m−2 d−1 in the YS, 68.5 to 487.3 mg C m−2 d−1 in the SS, and 106.4 to 490.5 mg C m−2 d−1 in the EJS, respectively. In comparison with previous studies, significantly lower (t-test, p &lt, 0.05) spring and summer productions and consequently lower annual primary productions were observed in this study. Based on PCA analysis, we found that small-sized (pico- and nano-) phytoplankton had strongly negative effects on the primary productions. Their ecological roles should be further investigated in the YS, the SS, and the EJS under warming ocean conditions within small phytoplankton-dominated ecosystems.

The macromolecular concentrations and compositions of phytoplankton are crucial for the growth or nutritional structure of higher trophic levels through the food web in the ecosystem. To understand variations in macromolecular contents of phytoplankton, we investigated the macromolecular components of phytoplankton and analyzed their spatial pattern on the Chukchi Shelf and the Canada Basin. The carbohydrate (CHO) concentrations on the Chukchi Shelf and the Canada Basin were 50.4–480.8 μg L−1 and 35.2–90.1 μg L−1, whereas the lipids (LIP) concentrations were 23.7–330.5 μg L−1 and 11.7–65.6 μg L−1, respectively. The protein (PRT) concentrations were 25.3–258.5 μg L−1 on the Chukchi Shelf and 2.4–35.1 μg L−1 in the Canada Basin. CHO were the predominant macromolecules, accounting for 42.6% on the Chukchi Shelf and 60.5% in the Canada Basin. LIP and PRT contributed to 29.7% and 27.7% of total macromolecular composition on the Chukchi Shelf and 30.8% and 8.7% in the Canada Basin, respectively. Low PRT concentration and composition in the Canada Basin might be a result from the severe nutrient-deficient conditions during phytoplankton growth. The calculated food material concentrations were 307.8 and 98.9 μg L−1, and the average calorie contents of phytoplankton were 1.9 and 0.6 kcal m−3 for the Chukchi Shelf and the Canada Basin, respectively, which indicates the phytoplankton on the Chukchi Shelf could provide the large quantity of food material and high calories to the higher trophic levels. Overall, our results highlight that the biochemical compositions of phytoplankton are considerably different in the regions of the Arctic Ocean. More studies on the changes in the biochemical compositions of phytoplankton are still required under future environmental changes.

The biochemical composition of particulate organic matter (POM) mainly originates from phytoplankton. Transparent exopolymer particles (TEPs) depend on environmental conditions and play a role in the food web and biogeochemical cycle in marine ecosystems. However, little information on their characteristics in the Southern Ocean is available, particularly in winter. To investigate the seasonal characteristics of POM and TEPSs, seawater samples were collected once every two weeks from November 2017 to October 2018 at Jang Bogo Station (JBS) located on the coast of Terra Nova Bay in the Ross Sea. The total chlorophyll-a (Chl-a) concentrations increased from spring (0.08 ± 0.06 μg L−1) to summer (0.97 ± 0.95 μg L−1) with a highest Chl-a value of 2.15 μg L−1. After sea ice formation, Chl-a rapidly decreased in autumn (0.12 ± 0.10 μg L−1) and winter (0.01 ± 0.01 μg L−1). The low phytoplankton Chl-a measured in this study was related to a short ice-free period in summer. Strong seasonal variations were detected in the concentrations of proteins and lipids (one-way ANOVA test, p &lt, 0.05), whereas no significant difference in carbohydrate concentrations was observed among different seasons (one-way ANOVA test, p &gt, 0.05). The phytoplankton community was mostly composed of diatoms (88.8% ± 11.6%) with a large accumulation of lipids. During the summer, the POM primarily consisted of proteins. The composition being high in lipids and proteins and the high caloric content in summer indicated that the phytoplankton would make a good food source. In winter, the concentrations of proteins decreased sharply. In contrast, relatively stable concentrations of carbohydrates and lipids have been utilized for respiration and long-term energy storage in the survival of phytoplankton. The TEPS values were significantly correlated with variations in the biomass and species of the phytoplankton. Our study site was characterized by dominant diatoms and low Chl-a concentrations, which could have resulted in relatively low TEP concentrations compared to other areas. The average contributions of TEP-C to the total POC were relatively high in autumn (26.9% ± 6.1%), followed by those in summer (21.9% ± 7.1%), winter (13.0% ± 4.2%), and spring (9.8% ± 3.1%).

Investigations on marine N2 fixation have gained momentum since 1960s with eventual establishments of relevant methodologies to identify species involved and quantify the rates. The evolution of various methodologies to understand N2 fixation and to estimate its rates were underpinned by the constant efforts of pioneers in the ocean biogeochemical research field. Those efforts succeeded in introducing various methodologies that include experimental (15N2 bubble method and acetylene reduction method), geochemical (N* and P* method), mathematical modelling, and remote sensing techniques. However, the construction of an accurate N budget is still under progress due to inseparable issues associated with each method and difficulties in conducting the experiments onboard on a larger scale. Nevertheless, the contributions by each of the methodologies are significant and helped in forming basic ideas about N2 fixation activities on a global scale. It is not only important to recognize the contributions made by the formation of various methodologies by marine research pioneers, but also vital to summarize what we have achieved in the marine N2 fixation research area so far. Hence, this review is an attempt to brief on the various milestones achieved in research on the N2 fixation mechanism, species involved, evolution of methodologies to estimate N2 fixation rates, species identification, budgets, and future concerns.

Organic carbon fixed by photosynthesis of phytoplankton during the polar growing period could be important for their survival and consumers during the long polar night. Differences in biochemical traits of phytoplankton between ice-free and polar night periods were investigated in biweekly water samples obtained at the Korean “Jang Bogo Station” located in Terra Nova Bay, Antarctica. The average concentration of total Chl-a from phytoplankton dominated by micro-sized species from the entire sampling period was 0.32 μg L–1 (SD = ± 0.88 μg L–1), with the highest concentration of 4.29 μg L–1 in February and the lowest concentration of 0.01 μg L–1 during the ice-covered polar night (April–October) in 2015. The highest protein concentration coincided with the peak Chl-a concentration in February and decreased rapidly relative to the carbohydrate and lipid concentrations in the early part of polar night. Among the different biochemical components, carbohydrates were the predominant constituent, accounting for 69% (SD = ± 14%) of the total particulate organic matter (POM) during the entire study period. The carbohydrate contributions to the total POM markedly increased from 39 ± 8% during the ice-free period to 73 ± 9% during the polar night period. In comparison, while we found a significant negative correlation (r2 = 0.92, p < 0.01) between protein contributions and carbohydrate contributions, lipid contributions did not show any particular trend with relatively small temporal variations during the entire observation period. The substantial decrease in the average weight ratio of proteins to carbohydrates from the ice-free period (mean ± SD = 1.0 ± 0.3) to the ice-covered period (mean ± SD = 0.1 ± 0.1) indicates a preferential loss of nitrogen-based proteins compared to carbohydrates during the polar night period. Overall, the average food material (FM) concentration and calorific contents of FM in this study were within the range reported previously from the Southern Ocean. The results from this study may serve as important background data for long-term monitoring of the regional and interannual variations in the physiological state and biochemical compositions of phytoplankton resulting from future climate change in Antarctica.

The cellular macromolecular contents and energy value of phytoplankton as primary food source determine the growth of higher trophic levels, affecting the balance and sustainability of oceanic food webs. Especially, proteins are more directly linked with basic functions of phytoplankton biosynthesis and cell division and transferred through the food chains. In recent years, the East/Japan Sea (EJS) has been changed dramatically in environmental conditions, such as physical and chemical characteristics, as well as biological properties. Therefore, developing an algorithm to estimate the protein concentration of phytoplankton and monitor their spatiotemporal variations on a broad scale would be invaluable. To derive the protein concentration of phytoplankton in EJS, the new regional algorithm was developed by using multiple linear regression analyses based on field-measured data which were obtained from 2012 to 2018 in the southwestern EJS. The major factors for the protein concentration were identified as chlorophyll-a (Chl-a) and sea surface nitrate (SSN) in the southwestern EJS. The coefficient of determination (r2) between field-measured and algorithm-derived protein concentrations was 0.55, which is rather low but reliable. The satellite-derived estimation generally follows the 1:1 line with the field-measured data, with Pearson’s correlation coefficient, which was 0.40 (p-value &lt, 0.01, n = 135). No remarkable trend in the long-term annual protein concentration of phytoplankton was found in the study area during our observation period. However, some seasonal difference was observed in winter protein concentration between the 2003–2005 and 2017–2019 periods. The algorithm is developed for the regional East/Japan Sea (EJS) and could contribute to long-term monitoring for climate-associated ecosystem changes. For a better understanding of spatiotemporal variation in the protein concentration of phytoplankton in the EJS, this algorithm should be further improved with continuous field surveys.

Biochemical composition of phytoplankton is a key indicator of the physiological and nutritional status of phytoplankton. A balanced biochemical pattern represents a healthy and productive metabolism in the autotrophic levels which can facilitate proper functioning of higher level organisms. The estimation of biochemical compositions was initiated in the early 1970’s. However, there has been a significant set of modifications in the extraction method and improvements in the sampling and analysis techniques since then. Similarly, the extent of biochemical measurements from various aquatic ecosystems around the globe has also increased. Recently, biochemical patterns are being used as a tool to track the changes in the physiological status of phytoplankton as a response to climate change. Such investigations are also forming part of research works on marine food webs and the nutritional status of ecosystems. This article is a brief review of research works carried out so far in an attempt to understand the biochemical compositions of phytoplankton in the global oceans and the implications with regard to changing environmental conditions.

The biochemical composition of particulate organic matter (POM) is very important to understand in relation to the trophic conditions of marine ecosystems since it forms the primary trophic base. The present study investigated the biochemical compositions (i.e., carbohydrates, proteins, and lipids) of POM monthly from January to December 2015 in Geoje-Hansan Bay to determine if the macromolecular composition of POM is coupled between the water columns and sediment. A spatial difference in the macromolecular compositions was observed in the water columns between the inner and outer bays, which may be caused by the different physiological conditions of phytoplankton growth that are due to the water circulation pattern in the bay. In contrast, no distinctive spatial difference in the macromolecular compositions was found in the sedimentary organic matter. Overall, while carbohydrates were the dominant (45.7%) macromolecules of the POM in the water columns, proteins were dominant (47.9%) in the sedimentary organic matter during our observation period. Decoupling of the macromolecular compositions between the water columns and underneath the sediment in Geoje-Hansan Bay appears to be a result of the various effects of selective filter feeding by oysters and protein-dominant benthic microalgae and fouling organisms.

The macromolecular composition of phytoplankton communities and the proximate composition of zooplankton communities were measured monthly in the southwestern East/Japan Sea from April to November 2014 in order to identify seasonal changes in, and relationships among, the biochemical compositions in both phytoplankton and zooplankton. The carbohydrate content of phytoplankton was highest in June, whereas the protein content was highest in August and lipids were highest in April. Overall, carbohydrates were dominant (53.2 ± 12.5%) in the macromolecular composition of phytoplankton during the study period. This composition is believed to result from the dominance of diatoms and/or nutrient-depleted conditions. In comparison, the protein level of zooplankton was highest in November, whereas lipids were slightly higher in May than other months. Overall, proteins were the dominant organic compounds (47.9±8.6% DW) in zooplankton communities, whereas lipids were minor components (5.5±0.6% DW). The high protein content of zooplankton might be related to the abundance of copepods, whereas the low lipid content might be due to a relatively high primary production that could provide a sufficient food supply for zooplankton so that they do not require high lipid storage. The most problematic issue in the ocean color application is the presence of heavy clouds, especially in polar regions. For that reason, the demand for the ocean color application in polar regions is increased. As a way to overcome such issues, we conducted the reconstruction of the chlorophyll-a concentration (CHL) data using the machine learning-based models to raise the usability of CHL data. This analysis was first conducted on a regional scale and focused on the biologically-valued Cape Hallett, Ross Sea, Antarctica. Environmental factors and geographical information associated with phytoplankton dynamics were considered as predictors for the CHL reconstruction, which were obtained from cloud-free microwave and reanalysis data. As the machine learning models used in the present study, the ensemble-based models such as Random forest (RF) and Extremely randomized tree (ET) were selected with 10-fold cross-validation. As a result, both CHL reconstructions from the two models showed significant agreement with the standard satellite-derived CHL data. In addition, the reconstructed CHLs were close to the actual CHL value even where it was not observed by the satellites. However, there is a slight difference between the CHL reconstruction results from the RF and the ET, which is likely caused by the difference in the contribution of each predictor. In addition, we examined the variable importance for the CHL reconstruction quantitatively. As such, the sea surface and atmospheric temperature, and the photosynthetically available radiation have high contributions to the model developments. Mostly, geographic information appears to have a lower contribution relative to environmental predictors. Lastly, we estimated the partial dependences for the predictors for further study on the variable contribution and investigated the contributions to the CHL reconstruction with changes in the predictors.

Proximate analysis for proportions of lipids, proteins, ash, and water contents of fish tissue provides a powerful insight to obtain physiological information for the fish population being studied. In this study, the proximate compositions for the jack mackerel ( Trachurus japonicus ) caught in Geumo Island, Korea known as a nursery ground were examined monthly in 2013 and 2014. No significant seasonal variation was found in protein and ash contents whereas a strong seasonal variation was found in water and lipid contents. The lipid contents showed relatively higher values ranging from 3.94% to 5.75% in summer and fall which corresponds to their active feeding periods. Little information on phytoplankton is currently available for Garolim and Asan bays, which are important spawning and nursery grounds for various commercial fish. The objectives of this study were to investigate the spatial and seasonal variations in the biochemical components (carbohydrates, proteins, and lipids) and primary production by phytoplankton and to determine the major environmental factors controlling the variations in Garolim and Asan bays. All major inorganic nutrients were seasonally available to phytoplankton although PO4 concentrations were relatively lower than those of other nutrients. The chlorophyll-a (chl-a) concentration measured in this study was within the range reported previously and the phytoplankton community was mostly dominated by micro- and nano-phytoplankton (approximately 80 %). Among the different biochemical compositions, carbohydrates were the most dominant throughout our observation period. As a nitrogen source for phytoplankton growth, ammonium was more important compared to nitrate in Garolim and Asan bays because of a significantly high uptake rate of the ammonium observed in this study. The overall mean areal daily carbon uptake rate (238.8 ± 393.3 mg C m−2 d−1) measured in this study is within the range in various bays and coastal waters in Korea. The concentrations of SiO2, NO2+NO3 and micro-sized chl-a were found to be major factors for the variation in the biochemical compositions of phytoplankton. By comparison, the nano-sized chl-a concentration, NH4 concentration and temperature were observed to be major factors in the variation in the carbon uptake rate of phytoplankton in Garolim and Asan bays during our observation period. Further studies on macromolecular compositions and primary production from different cell-sized phytoplankton are warranted for a better understanding their potential effects on marine ecosystems.

Here, we investigated the elemental (C/N ratio) and isotopic signatures (δ13C) and major biomolecules (carbohydrates, proteins, and lipids) and their relative abundance (i.e., the biochemical composition) in particulate organic matter (POM) to assess their origin and fate in the Laptev and western East Siberian seas during late summer/fall of 2015. In addition, we compared our results with the summer data of 2013 collected from Laptev and northwestern East Siberian seas. In accordance with the observed hydrological structure (i.e., a northward, warmer, diluted freshwater plume than previously observed in 2013), the more depleted δ13C (−28.2 ± 0.9‰) and higher C/N ratio (10.8 ± 2.0) than those of 2013 signalled that fluvially released terrestrial organic carbon (TerrOC) was the main source of the POM, unlike in 2013, when phytoplankton was the dominant source (δ13C = −24.9 ± 1.0‰, C/N ratio = 7.6 ± 2.4; Ahn et al., 2019). During the offshore transport of heterogeneous TerrOC, carbohydrates seem to be the primary contributor to the bulk POM as a result of selective degradation and hydrodynamic sorting. Despite the TerrOC-dominated system in 2015, some marine influence was also found. The estimated phytoplankton biomass was low and comparable among the study sites. In addition, the presence of resting spores and high ammonium concentrations within the water column may suggest senescent and, to some extent, degrading conditions of the resident phytoplankton. Biological nitrogen ( N 2 ) fixation, a fundamental step in aquatic and terrestrial N cycles, is an important process through which “new” reactive nitrogen (N) is produced. The role of N 2 fixation in fueling the occurrence of harmful algal blooms (HABs) is one of the major concerns in the present scenario. Since N 2 fixation occurs under highly specific environmental conditions, determining the suitability of various ecosystems for potential N 2 fixation is important. The introduction of 15N2 bubble experiments in 1995 has led to significant N 2 fixation rate estimates from various parts of the world oceans; however, Korean coastal waters, which are prone to HABs, remained unstudied. The present study aimed to construct a research note on N 2 fixation in the Korean coastal waters on the basis of a preliminary data set of N 2 fixation rates (range; 0.31–2.35 nmol N L−1 h−1) obtained from the coastal northwestern East/Japan Sea. The results indicate that N 2 fixation is not currently a significant N source in the northwestern East/Japan Sea. However, the possibility of N 2 fixers being a future significant N source and a fuel for HABs in the East/Japan Sea under favorable environmental conditions cannot be ruled out.
