1. The Influence of Seismic Parameters on Site Response

Abstract

Geotechnical earthquake engineers have been trying to determine the most appropriate solution techniques for ground response analysis under earthquake loadings. This paper presents the importance of the adequate soil behavior model to simulate earthquake site response analysis. The influence of nonlinearity and linearity of soils on the site response is also investigated. Many methods have been proposed for the analysis of one-dimensional ground response such as nonlinear methods and equivalent linear with reduced modulus. Using 13 well known earthquake records and geotechnical information of sand in the Qeshm Island (Iran) and classification of earthquakes based on seismic parameters, the influence of each parameter on the Response spectra and Fourier Spectrum of surface of ground becomes evident. Analysis was done using two softwares; EERA (equivalent linear analysis) and NERA (nonlinear analysis) and compared. Finally, output of the software is compared with the reference result on site.

5. A step toward cleaner energy production: A water saving-based optimization approach for economic dispatch in modern power systems

Abstract

This paper proposes a novel approach to discover an optimal solution of multi-area dynamic economic dispatch integrated with wind power and pumped hydroelectric storage. Minimizing of water consumption associated with power production in thermal power plants is yet to be addressed in economic dispatch problem. Thus, it is crucial to modernize the conventional power industry to save the water utilization in the process of generating electric energy. As a result, making a feasible power generation schedule by finding a trade-off between total generation cost and water consumption is the main focus of this paper. Toward this end, this study introduces an effective optimization approach for decreasing the contaminants ‎produced by the combustion-based power plants via reflecting the impacts of wind power and storage systems in the proposed problem. An efficient optimization algorithm considering the merits of whale optimization algorithm and wavelet mutation is also proposed to handle the complications of the trade-off between generation cost and water consumption. The proposed approach is applied to 40-unit 4-area IEEE system by 1) scrutinizing the performance of the hybrid framework from different standpoints, 2) achieving an improvement about 5.4% in the operation cost, and 3) promoting the decarbonized economy by implementing ‎renewable energy.

Abstract

Interleukin 6 (IL-6) is a multifunctional cytokine with both pro- and anti-inflammatory properties with a heritability estimate of up to 61%. The circulating levels of IL-6 in blood have been associated with an increased risk of complex disease pathogenesis. We conducted a two-staged, discovery and replication meta genome-wide association study (GWAS) of circulating serum IL-6 levels comprising up to 67428 (n(discovery)=52654 and n(replication)=14774) individuals of European ancestry. The inverse variance fixed effects based discovery meta-analysis, followed by replication led to the identification of two independent loci, IL1F10/IL1RN rs6734238 on chromosome (Chr) 2q14, (P-combined=1.8x10(-11)), HLA-DRB1/DRB5 rs660895 on Chr6p21 (P-combined=1.5x10(-10)) in the combined meta-analyses of all samples. We also replicated the IL6R rs4537545 locus on Chr1q21 (P-combined=1.2x10(-122)). Our study identifies novel loci for circulating IL-6 levels uncovering new immunological and inflammatory pathways that may influence IL-6 pathobiology.

Abstract

Reproductive longevity is essential for fertility and influences healthy ageing in women1,2, but insights into its underlying biological mechanisms and treatments to preserve it are limited. Here we identify 290 genetic determinants of ovarian ageing, assessed using normal variation in age at natural menopause (ANM) in about 200,000 women of European ancestry. These common alleles were associated with clinical extremes of ANM; women in the top 1% of genetic susceptibility have an equivalent risk of premature ovarian insufficiency to those carrying monogenic FMR1 premutations3. The identified loci implicate a broad range of DNA damage response (DDR) processes and include loss-of-function variants in key DDR-associated genes. Integration with experimental models demonstrates that these DDR processes act across the life-course to shape the ovarian reserve and its rate of depletion. Furthermore, we demonstrate that experimental manipulation of DDR pathways highlighted by human genetics increases fertility and extends reproductive life in mice. Causal inference analyses using the identified genetic variants indicate that extending reproductive life in women improves bone health and reduces risk of type 2 diabetes, but increases the risk of hormone-sensitive cancers. These findings provide insight into the mechanisms that govern ovarian ageing, when they act, and how they might be targeted by therapeutic approaches to extend fertility and prevent disease.Copyright © 2021, The Author(s), under exclusive licence to Springer Nature Limited.

Abstract

Reproductive longevity is essential for fertility and influences healthy ageing in women1,2, but insights into its underlying biological mechanisms and treatments to preserve it are limited. Here we identify 290 genetic determinants of ovarian ageing, assessed using normal variation in age at natural menopause (ANM) in about 200,000 women of European ancestry. These common alleles were associated with clinical extremes of ANM; women in the top 1% of genetic susceptibility have an equivalent risk of premature ovarian insufficiency to those carrying monogenic FMR1 premutations3. The identified loci implicate a broad range of DNA damage response (DDR) processes and include loss-of-function variants in key DDR-associated genes. Integration with experimental models demonstrates that these DDR processes act across the life-course to shape the ovarian reserve and its rate of depletion. Furthermore, we demonstrate that experimental manipulation of DDR pathways highlighted by human genetics increases fertility and extends reproductive life in mice. Causal inference analyses using the identified genetic variants indicate that extending reproductive life in women improves bone health and reduces risk of type 2 diabetes, but increases the risk of hormone-sensitive cancers. These findings provide insight into the mechanisms that govern ovarian ageing, when they act, and how they might be targeted by therapeutic approaches to extend fertility and prevent disease.

However, this study shows that the most economic schedule of power generation cannot satisfy echo-system expectation; therefore, making a compromise between fuel cost and environmental issues, a hot-button subject in industrialized nations, seems to be crucial. To reach the goals a bi-objective multi-area dynamic economic dispatch approach, which can handle intertemporal and multi-area constraints concurrently, is proposed to assist power system operators more and more. Finally, a hybrid algorithm, namely gray wolf optimizer-particle swarm optimization is introduced to solve the proposed problem and also a set of benchmark problems. By implementing the proposed approach on two small (10-unit, three areas) and large (40-unit, four areas) scale test systems, about 3.1% and 3.3% improvement in generation cost is obtained, respectively compare to the best reported results in the literature.

Abstract

This paper introduces a novel, practical model for Multi-Area Dynamic Economic Dispatch (MADED) problem whose primary targets are to minimize both operational cost and water consumption. To make the proposed study more practical, several additional requirements that are inevitable in forthcoming power systems are taken into account including wind power penetration, pumped storage systems, and different interconnected areas. Such a complicated problem needs to be solved by a potent multi-objective optimization tool. In this regard, one of the relatively new optimization approaches, Whale Optimization Algorithm (WOA), is implemented to solve the proposed problem. The obtained optimal solutions on a 40-unit power network illustrate the effectiveness of the proposed approach and reveal a substantial improvement in reducing the operational cost, i.e., more than $30 million is reduced annually.

14. Strategies for Empowering Collective Design

Abstract

Collective activities such as crowdfunding, competitions, jams and hackathons are gaining more attention in innovation management. However, within the design domain such activities are often considered as a way for mass generation of ideas, rather than processes for developing proper design solutions. Information acquisition, gaining sufficient knowledge and generating feasible ideas are among the main challenges of collective design. In this study we applied three strategies of rapid design, informed participation, and knowledge sharing to overcome these challenges. During a one- week design hackathon, we conducted a collective design project in which three teams of participants conducted autoethnography research, shared their findings by an idea crowdsourcing platform, and tested their 3D printed prototypes with potential users. Although the tests showed efficiency of rapid design and informed participation strategy, we did not find knowledge sharing effective, mainly due to the absence of collaboration among teams.

Abstract

Density functional theory (DFT) based calculations have been performed to examine the relaxations of tautomers of 4–hydroxy–6–methylpyridin–2(1H)–one (MPO), as a representative of pyridinone derivatives, at the fullerene (C60) surfaces. Optimized molecular properties including energies, dipole moments and atomic scale quadrupole coupling constants (CQ) have been evaluated to investigate the structural and electronic properties of the models. The structural configurations of tautomers show different relaxations at the C60 surface yielding different magnitudes of total and binding energies. Moreover, deformation of each tautomer due to relaxation at the C60 surface with respect to the initial singular structure has been examined. Complimentary parameters of energy gaps and dipole moments exhibit the effects of relaxations at the C60 surface for the MPO counterparts. Atomic scale CQ properties also indicate that the electronic properties of atoms show significant changes for tautomers and hybrid systems. As a final note, the tautomeric structures in singular and hybrid forms exhibit different electronic properties because of effects of interactions with C60, especially for the interaction regions.

Abstract

The present study aimed to prepare Fe3O4 nanocarriers (NCs) by a thermal treatment method. After the Fe3O4 (Fe) NCs was prepared, zinc oxide and silica nanoparticles were added to it as Photosensitizer. The structure, morphology, and magnetic properties of Fe3O4@ZnO (Fe@Zn) and Fe3O4@SiO2 (Fe@Si) NCs were determined by XRD, FT-IR, FESEM, and VSM. Then, the loading and drug release of Fe, Fe@Zn, and Fe@Si NCs were investigated. The curcumin (CUR) release of Fe@Zn+CUR and Fe@Si+CUR increased from 30% and 26% at pH 7.4 to 53% and 57% at pH 5.5, respectively. The cytotoxicity of Fe@Zn and Fe@Si NCs were determined by MTT assay, hemolysis test, acute toxicity, and lethal dose test. The results showed that Fe@Zn and Fe@Si were appropriate for Photodynamic Therapy (PDT) and in the next step, the effect of Fe@Zn, Fe@Si, Fe@Zn+CUR, and Fe@Si+CUR NCs on MCF-7 cells under visible light were studied. Finally, the ranking of the destruction of cancerous cells of MCF-7 using NCs under visible light was: Fe@Zn+CUR>Fe@Zn>Fe@Si+CUR>Fe@Si. [ABSTRACT FROM AUTHOR]

23. Research on simulation and modeling of simple and cost-effective BLDC motor drives.

Abstract

Brushless DC (BLDC) motors have been increasingly considered in variable speed drives in wide array of applications due to their significant features such as high efficiency, power density, and torque density. Cost minimization of variable speed drives is accomplished by topological and control approaches. Minimum number of switches and current sensors, and eliminating the position sensors are desired in the topological approach. Moreover, the control algorithms must be simple, effective, and adjustable with drive topology to produce the desired speed-torque characteristics. In this paper, a review of four different drives of BLDC motor is presented, with emphasis on control strategies, drive topologies, and analysis of simulation results. In addition, speed control of these drives using Particle Swarm Optimization based on tuned Proportional Integral controller is regulated because classic control theory is complex and requires the precise recognition of system to develop a desired controller. Abstract

Interleukin 6 receptor (IL-6R), mediating IL-6’s biological functions, plays an important role in different diseases such as diabetes, obesity and cardio-vascular diseases. In this study, we investigated the effects of two single nucleotide polymorphisms (SNPs), within the IL-6Rloci, previously associated with C-reactive protein (CRP) and coronary heart diseases risk, and with controversial effects on lipids traits: SNP rs4845625 and SNP rs4537545. The results showed that both investigated SNPs were antagonistically related with CRP levels; the minor rs4845625*T allele was associated with increased CRP levels (P-value=0.011), while the minor rs4537545*T allele was associated with decreased CRP levels (P-value=0.009). Interestingly, the minor rs4845625*T allele was significantly associated with higher low-density lipoprotein cholesterol (LDL-C) and ApoB levels (P=0.007 and P=0.009 respectively). Haplotype analysis showed that the TC haplotype, having the minor rs4845625*T allele, was related simultaneously with increased levels of CRP, LDL-C and ApoB levels, thus could be considered as a risk factor. Our investigation detects for the first time an independent effect of rs4845625 on LDL-C and ApoB traits, explaining an important range of those traits variability (3.49 and 5.57% respectively). Our findings might be of high clinical significance in pharmacogenomics studies of tocilizumab for which IL-6Ris target.

In this study, Shuffled Frog Leaping Algorithm (SFLA) for optimizing generators fuel cost, active power transmission losses and voltage deviation of system buses simultaneously and with FACTS devices in power system have been used. In this suggested approach, optimal power flow has been formulized as a multi-purpose optimal problem. Applied FACTS devices include Thyristor Controlled Series Capacitor (TCSC) and Unified Power Flow Controller (UPFC). Proposed method have been examined on 5 buses and 30 buses IEEE standard test systems and obtained results of suggested algorithm have been compared with obtained results of Improvement Particle Swarm Optimization method. Abstract

Aims: Internal evaluation is a kind of self-evaluation that is applied by educational units in order to determine the educational and research situation of the department. The purpose of this study was the application of internal evaluation model in the education technology department of Tarbiat-Moallem University considering the aims and expectations, in order to offer needed suggestions for its quality improvement. Methods: This descriptive applied research was carried out in 2008 & 2009. All faculty members, students, librarians and the head of the department were studied due to the limitation of statistical society. Available sampling method was used for evaluating the alumni and their direct employers. To conduct the internal evaluation, at first, the goals of the department were determined in three categories: input, process and output. Bearing national and international experiences in mind, 7 factors, 40 criteria and 124 indicators along with judgment criteria was determined. Data collection tools including questionnaires and checklists were prepared and distributed among the research samples. Data analysis was carried out by descriptive statistical methods using SPSS 16 software. Results: The current status of the education technology department of Tarbiat-Moallem University in terms of input, process and output was quite desirable leading to a satisfactory level in general. Conclusion: The education technology department of Tarbiat-Moallem University is in a quite desirable level in terms of input, process and output factors. Abstract With cAMP signaling having a profound inhibitory effect on DNA damage-induced apoptosis in B-cell precursor acute lymphoblastic leukemia (BCP-ALL) cells, understanding how this signaling pathway affects the survival capacity of the cell has important implications for cancer therapy. We have recently shown that p53 is critical for the inhibitory effect of cAMP on genotoxic agents-mediated apoptosis in BCP-ALLs. Here, we show that elevation of cAMP levels in cells exposed to DNA damage enhances the nuclear translocation and DNA binding of NF-κB by accelerating the phosphorylation of IKKβ and thereby phosphorylation and degradation of IκBα. Furthermore, we show that the ability of cAMP to potentiate the ionizing radiation-induced activation of NF-κB requires the activity of MEK. Importantly, pharmacological or genetic ablation of NF-κB reversed the inhibitory effect of cAMP on DNA damage-induced apoptosis, demonstrating that, in addition to p53, cAMP relies on the activity of NF-κB to provide cells with a survival advantage in the face of DNA damage. Collectively, our results uncover a novel and important interaction between the cAMP and NF-κB pathways that may have implications for the targeted treatment of lymphoid malignancies, such as BCP-ALL, in which aberrant NF-κB activity functions as a driving force for treatment resistance.

Abstract

International audience; C-reactive protein (CRP) is a sensitive biomarker of chronic low-grade inflammation and is associated with multiple complex diseases. The genetic determinants of chronic inflammation remain largely unknown, and the causal role of CRP in several clinical outcomes is debated. We performed two genome-wide association studies (GWASs), on HapMap and 1000 Genomes imputed data, of circulating amounts of CRP by using data from 88 studies comprising 204,402 European individuals. Additionally, we performed in silico functional analyses and Mendelian randomization analyses with several clinical outcomes. The GWAS meta-analyses of CRP revealed 58 distinct genetic loci (p < 5 × 10-8). After adjustment for body mass index in the regression analysis, the associations at all except three loci remained. The lead variants at the distinct loci explained up to 7.0% of the variance in circulating amounts of CRP. We identified 66 gene sets that were organized in two substantially correlated clusters, one mainly composed of immune pathways and the other characterized by metabolic pathways in the liver. Mendelian randomization analyses revealed a causal protective effect of CRP on schizophrenia and a risk-increasing effect on bipolar disorder. Our findings provide further insights into the biology of inflammation and could lead to interventions for treating inflammation and its clinical consequences.Copyright © 2018 American Society of Human Genetics. Abstract

Background: Patients with schizophrenia spectrum disorders (SSD) have a shortened life expectancy related to cardiovascular diseases. We investigated the association of cognitive, positive, and negative symptoms with cardiometabolic dysregulations in SSD patients., Methods: Overall, 1,119 patients from the Genetic Risk and Outcome in Psychosis (GROUP) study were included. Cognitive function, positive and negative symptoms were assessed at baseline, 3-year, and 6-year. Cardiometabolic biomarkers were measured at 3-year follow-up. We used linear and multinomial logistic regression models to test the association between cardiometabolic biomarkers and clinical trajectories and performed mediation analyzes, while adjusting for clinical and demographic confounders., Results: Cognitive performance was inversely associated with increased body mass index (mean difference [β], β high  = -1.24, 95% CI = -2.28 to 0.20, P  = 0.02) and systolic blood pressure (β mild  = 2.74, 95% CI = 0.11 to 5.37, P  = 0.04). The severity of positive symptoms was associated with increased glycated hemoglobin (HbA1c) levels (β low  = -2.01, 95% CI = -3.21 to -0.82, P  = 0.001). Increased diastolic blood pressure (OR high-decreased  = 1.04, 95% CI = 1.01 to 1.08, P  = 0.02; OR high-increased  = 1.04, 95% CI = 1.00 to 1.08, P  = 0.048) and decreased high-density lipoprotein (OR high-increased  = 6.25, 95% CI = 1.81 to 21.59, P  = 0.004) were associated with more severe negative symptoms. Increased HbA1c (OR moderate  = 1.05, 95% CI = 1.01 to 1.10, P  = 0.024; OR high  = 1.08, 95% CI = 1.02 to 1.14, P  = 0.006) was associated with more severe positive symptoms. These associations were not mediated by antipsychotics., Conclusions: We showed an association between cardiometabolic dysregulations and clinical and cognitive symptoms in SSD patients. The observed associations underscore the need for early identification of patients at risk of cardiometabolic outcomes.

Abstract

Background: This study was designed to identify common genetic susceptibility and shared genetic variants associated with acute radiation-induced toxicity across 4 cancer types (prostate, head and neck, breast, and lung)., Methods: A genome-wide association study meta-analysis was performed using 19 cohorts totaling 12 042 patients. Acute standardized total average toxicity (STATacute) was modelled using a generalized linear regression model for additive effect of genetic variants, adjusted for demographic and clinical covariates (rSTATacute). Linkage disequilibrium score regression estimated shared single-nucleotide variation (SNV-formerly SNP)-based heritability of rSTATacute in all patients and for each cancer type., Results: Shared SNV-based heritability of STATacute among all cancer types was estimated at 10% (SE = 0.02) and was higher for prostate (17%, SE = 0.07), head and neck (27%, SE = 0.09), and breast (16%, SE = 0.09) cancers. We identified 130 suggestive associated SNVs with rSTATacute (5.0 × 10‒8 < P < 1.0 × 10‒5) across 25 genomic regions. rs142667902 showed the strongest association (effect allele A; effect size ‒0.17; P = 1.7 × 10‒7), which is located near DPPA4, encoding a protein involved in pluripotency in stem cells, which are essential for repair of radiation-induced tissue injury. Gene-set enrichment analysis identified 'RNA splicing via endonucleolytic cleavage and ligation' (P = 5.1 × 10‒6, P = .079 corrected) as the top gene set associated with rSTATacute among all patients. In silico gene expression analysis showed that the genes associated with rSTATacute were statistically significantly up-regulated in skin (not sun exposed P = .004 corrected; sun exposed P = .026 corrected)., Conclusions: There is shared SNV-based heritability for acute radiation-induced toxicity across and within individual cancer sites. Future meta-genome-wide association studies among large radiation therapy patient cohorts are worthwhile to identify the common causal variants for acute radiotoxicity across cancer types., (© The Author(s) 2023. Abstract

Schizophrenia spectrum disorders (SSD) are complex mental disorders, and while treatment with antipsychotics is important, many patients do not respond or develop serious side effects. Genetic variation has been shown to play a considerable role in determining an individual's response to antipsychotic medication. However, previous pharmacogenetic (PGx) studies have been limited by small sample sizes, lack of consensus regarding relevant genetic variants, and cross-sectional designs. The current study aimed to investigate the association between PGx variants and long-term clinical outcomes in 691 patients of European ancestry with SSD. Using evidence from the literature on candidate genes involved in antipsychotic pharmacodynamics, we created a polygenic risk score (PRS) to investigate its association with clinical outcomes. We also created PRS using core variants of psychotropic drug metabolism enzymes CYP2D6 and CYP2C19. Furthermore, the CYP2D6 and CYP2C19 functional activity scores were calculated to determine the relationship between metabolism and clinical outcomes. We found no association for PGx PRSs and clinical outcomes; however, an association was found with CYP2D6 activity scores by the traditional method. Higher CYP2D6 metabolism was associated with high positive and high cognitive impairment groups relative to low symptom severity groups. These findings highlight the need to test PGx efficacy with different symptom domains. More evidence is needed before pharmacogenetic variation can contribute to personalized treatment plans.

41. The Progress and Pitfalls of Pharmacogenetics-Based Precision Medicine in Schizophrenia Spectrum Disorders: A Systematic Review and Meta-Analysis.

Abstract

The inadequate efficacy and adverse effects of antipsychotics severely affect the recovery of patients with schizophrenia spectrum disorders (SSD). We report the evidence for associations between pharmacogenetic (PGx) variants and antipsychotics outcomes, including antipsychotic response, antipsychotic-induced weight/BMI gain, metabolic syndrome, antipsychotic-related prolactin levels, antipsychotic-induced tardive dyskinesia (TD), clozapine-induced agranulocytosis (CLA), and drug concentration level (pharmacokinetics) in SSD patients. Through an in-depth systematic search in 2010-2022, we identified 501 records. We included 29 meta-analyses constituting pooled data from 298 original studies over 69 PGx variants across 39 genes, 4 metabolizing phenotypes of CYP2D9 , and 3 of CYP2C19 . We observed weak unadjusted nominal significant ( p < 0.05) additive effects of PGx variants of DRD1 , DRD2 , DRD3 , HTR1A , HTR2A , HTR3A , and COMT (10 variants) on antipsychotic response; DRD2 , HTR2C , BDNF , ADRA2A , ADRB3 , GNB3 , INSIG2 , LEP , MC4R , and SNAP25 (14 variants) on weight gain; HTR2C (one variant) on metabolic syndrome; DRD2 (one variant) on prolactin levels; COMT and BDNF (two variants) on TD; HLA-DRB1 (one variant) on CLA; CYP2D6 (four phenotypes) and CYP2C19 (two phenotypes) on antipsychotics plasma levels. In the future, well-designed longitudinal naturalistic multi-center PGx studies are needed to validate the effectiveness of PGx variants in antipsychotic outcomes before establishing any reproducible PGx passport in clinical practice.

42. The genetic contribution of the X chromosome in age-related hearing loss.

Abstract

Age-related (AR) hearing loss (HL) is the most common sensory impairment with heritability of 55%. The aim of this study was to identify genetic variants on chromosome X associated with ARHL through the analysis of data obtained from the UK Biobank. We performed association analysis between self-reported measures of HL and genotyped and imputed variants on chromosome X from ∼460,000 white Europeans. We identified three loci associated with ARHL with a genome-wide significance level (p < 5 × 10 -8 ), ZNF185 (rs186256023, p = 4.9 × 10 -10 ) and MAP7D2 (rs4370706, p = 2.3 × 10 -8 ) in combined analysis of males and females, and LOC101928437 (rs138497700, p = 8.9 × 10 -9 ) in the sex-stratified analysis of males. In-silico mRNA expression analysis showed MAP7D2 and ZNF185 are expressed in mice and adult human inner ear tissues, particularly in the inner hair cells. We estimated that only a small amount of variation of ARHL, 0.4%, is explained by variants on the X chromosome. Abstract

Background and Purpose: We aimed to the genetic components and susceptibility variants associated with acute radiation-induced toxicities (RITs) in patients with head and neck cancer (HNC)., Materials and Methods: We performed the largest meta-GWAS of seven European cohorts (n = 4,042). Patients were scored weekly during radiotherapy for acute RITs including dysphagia, mucositis, and xerostomia. We analyzed the effect of variants on the average burden (measured as area under curve, AUC) per each RIT, and standardized total average acute toxicity (STAT acute ) score using a multivariate linear regression. We tested suggestive variants (p < 1.0x10 -5 ) in discovery set (three cohorts; n = 2,640) in a replication set (four cohorts; n = 1,402). We meta-analysed all cohorts to calculate RITs specific SNP-based heritability, and effect of polygenic risk scores (PRSs), and genetic correlations among RITS., Results: From 393 suggestive SNPs identified in discovery set; 37 were nominally significant (p replication  < 0.05) in replication set, but none reached genome-wide significance (p combined  < 5 × 10 -8 ). In-silico functional analyses identified "3'-5'-exoribonuclease activity" (FDR = 1.6e-10) for dysphagia, "inositol phosphate-mediated signalling" for mucositis (FDR = 2.20e-09), and "drug catabolic process" for STAT acute (FDR = 3.57e-12) as the most enriched pathways by the RIT specific suggestive genes. The SNP-based heritability (±standard error) was 29 ± 0.08 % for dysphagia, 9 ± 0.12 % (mucositis) and 27 ± 0.09 % (STAT acute ). Positive genetic correlation was rg = 0.65 (p = 0.048) between dysphagia and STAT acute . PRSs explained limited variation of dysphagia (3 %), mucositis (2.5 %), and STAT acute (0.4 %)., Conclusion: In HNC patients, acute RITs are modestly heritable, sharing 10 % genetic susceptibility, when PRS explains < 3 % of their variance. 44. Nigella sativa and its main constituent, thymoquinone protect against glycerol-induced acute kidney injury in rats.

Abstract

Objective: Rhabdomyolysis is a life-threatening disease caused by releasing myoglobin from injured myocytes, which results in acute kidney injury. In this study, the effect of aqueous-alcoholic extract of Nigella sativa (NS) and thymoquinone (TQ) on rhabdomyolysis-induced kidney damage in rats was investigated., Materials and Methods: There were five groups of rats (n=8): Control, rhabdomyolysis, rhabdomyolysis treated with NS aqueous-alcoholic extract (200 and 400 mg/kg) and TQ (15 mg/kg). Treatments were given for 7 days (two days before and four days after glycerol injection). Glycerol was injected intramuscularly on the third day of the experiment for induction of rhabdomyolysis. Renal function parameters on the first, fourth, and seventh days of the experiment and renal oxidative stress and histological changes at the end of this study were assessed., Results: Glycerol injection caused a significant increase in serum level of urea, creatinine, creatine phosphokinase, urine output and tissue MDA compared to the control animals (p<0.05-0.001). Administration of NS extract and TQ significantly decreased serum urea and creatinine on days 4 and 7, creatine phosphokinase on day 4, and urine output on day 7 compared to the rhabdomyolysis group (p<0.05-0.001). Compared to the rhabdomyolysis group, treatment with NS extract and TQ improved kidney histological abnormalities (p<0.01-0.001). The catalase enzyme activity in the group treated with NS 400 mg/kg and thiol content in the NS 400 mg/kg and TQ groups were significantly higher than those of the rhabdomyolysis group (p<0.05-0.01)., Conclusion: NS extract and to some extent TQ protect the kidney from rhabdomyolysis-induced injury., Competing Interests: The authors have declared that there is no conflict of interest.

Abstract

Purpose: A two-stage genome-wide association study was carried out in head and neck cancer (HNC) patients aiming to identify genetic variants associated with either specific radiotherapy-induced (RT) toxicity endpoints or a general proneness to develop toxicity after RT., Materials and Methods: The analysis included 1780 HNC patients treated with primary RT for laryngeal or oro/hypopharyngeal cancers. In a non-hypothesis-driven explorative discovery study, associations were tested in 1183 patients treated within The Danish Head and Neck Cancer Group. Significant associations were later tested in an independent Dutch cohort of 597 HNC patients and if replicated, summary data obtained from discovery and replication studies were meta-analysed. Further validation of significantly replicated findings was pursued in an Asian cohort of 235 HNC patients with nasopharynx as the primary tumour site., Results: We found and replicated a significant association between a locus on chromosome 5 and mucositis with a pooled OR for rs1131769*C in meta-analysis = 1.95 (95% CI 1.48-2.41; p pooled  = 4.34 × 10 -16 )., Conclusion: This first exploratory GWAS in European cohorts of HNC patients identified and replicated a risk locus for mucositis. Background: Most head and neck cancer (HNC) patients receive radiotherapy (RT) and develop toxicities. This genome-wide association study (GWAS) was designed to identify single nucleotide polymorphisms (SNPs) associated with common acute radiation-induced toxicities (RITs) in an HNC cohort., Methods: A two-stage GWAS was performed in 1279 HNC patients treated with RT and prospectively scored for mucositis, xerostomia, sticky saliva, and dysphagia. The area under the curve (AUC) was used to estimate the average load of toxicity during RT. At the discovery study, multivariate linear regression was used in 957 patients, and the top-ranking SNPs were tested in 322 independent replication cohort. Next, the discovery and the replication studies were meta-analyzed., Results: A region on 5q21.3 containing 16 SNPs showed genome-wide (GW) significance association at P-value < 5.0 × 10 -8 with patient-rated acute xerostomia in the discovery study. The top signal was rs35542 with an adjusted effect size of 0.17*A (95% CI 0.12 to 0.23; P-value <  = 3.78 × 10 -9 ). The genome wide significant SNPs were located within three genes (EFNA5, FBXL17, and FER). In-silico functional analysis showed these genes may be involved in DNA damage response and co-expressed in minor salivary glands. We found 428 suggestive SNPs (P-value < 1.0 × 10 -5 ) for other toxicities, taken to the replication study. Eleven of them showed a nominal association (P-value < 0.05)., Conclusions: This GWAS suggested novel SNPs for patient-rated acute xerostomia in HNC patients. If validated, these SNPs and their related functional pathways could lead to a predictive assay to identify sensitive patients to radiation, which may eventually allow a more individualized RT treatment., (© 2021. Abstract

Reproductive longevity is essential for fertility and influences healthy ageing in women 1,2 , but insights into its underlying biological mechanisms and treatments to preserve it are limited. Here we identify 290 genetic determinants of ovarian ageing, assessed using normal variation in age at natural menopause (ANM) in about 200,000 women of European ancestry. These common alleles were associated with clinical extremes of ANM; women in the top 1% of genetic susceptibility have an equivalent risk of premature ovarian insufficiency to those carrying monogenic FMR1 premutations 3 . The identified loci implicate a broad range of DNA damage response (DDR) processes and include loss-of-function variants in key DDR-associated genes. Integration with experimental models demonstrates that these DDR processes act across the life-course to shape the ovarian reserve and its rate of depletion. Furthermore, we demonstrate that experimental manipulation of DDR pathways highlighted by human genetics increases fertility and extends reproductive life in mice. Causal inference analyses using the identified genetic variants indicate that extending reproductive life in women improves bone health and reduces risk of type 2 diabetes, but increases the risk of hormone-sensitive cancers. These findings provide insight into the mechanisms that govern ovarian ageing, when they act, and how they might be targeted by therapeutic approaches to extend fertility and prevent disease., (© 2021. Abstract

Objective: Phenylketonuria (PKU) is one of the most common inherited metabolic diseases, which is classified into classic and non-classic types. It is estimated that 2% of children with PKU develop a severe and progressive neurological disease, called non-classic (malignant) PKU. This study aimed to demonstrate the clinical features, laboratory findings, and diagnostic/therapeutic characteristics of non-classic PKU patients referred to a tertiary referral center for children in Tehran, Iran., Materials & Methods: In this study, background information, such as gender and age, clinical manifestations, laboratory findings, and response rate to conventional treatment, was investigated in patients with non-classic PKU, who were referred to Mofid Children's Hospital in Tehran, Iran, through neonatal screening., Results: Twenty patients with a diagnosis of non-classic PKU were included in this study. The mean age of the patients was 6.00±2.81 years (range: 2-12 years), and 45.0% were male. In patients with a late diagnosis, the most common presentations were motor developmental delay (15.0%), skin and cutaneous manifestations (15.0%), seizure (5.0%), and restlessness (5.0%). The overall response rate to treatment was 85.0%. Factors that predict good response to treatment included female gender, higher neopterin level, and lower age at diagnosis and management., Conclusion: In conclusion, about half of patients with non-classic PKU remain asymptomatic, which is due to early diagnosis via neonatal screening. 49. Cervical Cancer Prediction by Merging Features of Different Colposcopic Images and Using Ensemble Classifier.

Abstract

Background: Cervical cancer is a significant cause of cancer mortality in women, particularly in low-income countries. In regular cervical screening methods, such as colposcopy, an image is taken from the cervix of a patient. The particular image can be used by computer-aided diagnosis (CAD) systems that are trained using artificial intelligence algorithms to predict the possibility of cervical cancer. Artificial intelligence models had been highlighted in a number of cervical cancer studies. However, there are a limited number of studies that investigate the simultaneous use of three colposcopic screening modalities including Greenlight, Hinselmann, and Schiller., Methods: We propose a cervical cancer predictor model which incorporates the result of different classification algorithms and ensemble classifiers. Our approach merges features of different colposcopic images of a patient. The feature vector of each image includes semantic medical features, subjective judgments, and a consensus. The class label of each sample is calculated using an aggregation function on expert judgments and consensuses., Results: We investigated different aggregation strategies to find the best formula for aggregation function and then we evaluated our method using the quality assessment of digital colposcopies dataset, and our approach performance with 96% of sensitivity and 94% of specificity values yields a significant improvement in the field., Conclusion: Our model can be used as a supportive clinical decision-making strategy by giving more reliable information to the clinical decision makers. Abstract

Interleukin 6 (IL-6) is a multifunctional cytokine with both pro- and anti-inflammatory properties with a heritability estimate of up to 61%. The circulating levels of IL-6 in blood have been associated with an increased risk of complex disease pathogenesis. We conducted a two-staged, discovery and replication meta genome-wide association study (GWAS) of circulating serum IL-6 levels comprising up to 67 428 (ndiscovery = 52 654 and nreplication = 14 774) individuals of European ancestry. The inverse variance fixed effects based discovery meta-analysis, followed by replication led to the identification of two independent loci, IL1F10/IL1RN rs6734238 on chromosome (Chr) 2q14, (Pcombined = 1.8 × 10-11), HLA-DRB1/DRB5 rs660895 on Chr6p21 (Pcombined = 1.5 × 10-10) in the combined meta-analyses of all samples. We also replicated the IL6R rs4537545 locus on Chr1q21 (Pcombined = 1.2 × 10-122). Our study identifies novel loci for circulating IL-6 levels uncovering new immunological and inflammatory pathways that may influence IL-6 pathobiology., (© The Author(s) 2021.
