Brewer's spent grains (BSG) offer valuable opportunities for valorization beyond its conventional use as animal feed. Among its components, lignin—a natural polymer with inherent antioxidant properties—holds significant industrial potential. This work investigates the use of microwave-assisted extraction combined with acidic natural deep eutectic solvents (NaDESs) for efficient lignin recovery, evaluating three different NaDES formulations. The results indicate that choline chloride–lactic acid (ChCl-LA), a NaDES with superior thermal stability as confirmed via thermogravimetric analysis (TGA), is an ideal solvent for lignin extraction at 150 °C and 15 min, achieving a balance of high yield and quality. ChCl-LA also demonstrated good solubility and cell disruption capabilities, while microwaves significantly reduced processing time and severity. Under optimal conditions, i.e., 150 °C, 15 min, in the presence of ChCl-LA NaDES, the extracted lignin achieved a purity of up to 79% and demonstrated an IC50 (inhibitory concentration 50%) of approximately 0.022 mg/L, indicating a relatively strong antioxidant activity. Fourier transform infrared (FTIR) and 2D-HSQC NMR (heteronuclear single quantum coherence nuclear magnetic resonance) spectroscopy confirmed the successful isolation and preservation of its structural integrity. Natural Deep Eutectic Solvents (NADESs) are a form of green solvents used to extract phenolic compounds and are considered a more environmentally friendly alternative to traditional solvents. The study aimed to explore a more eco-friendly and effective approach to extracting anthocyanins from Prunus domestica by utilizing NADESs. Four NADESs were created by making combinations of choline chloride with citric acid, lactic acid, glycerol, and ethylene glycol. The study investigated the anthocyanins profile, antioxidant, and anti-sickling properties of fruit extracts that were obtained using NADESs. The examination of NADESs revealed that the hydrogen bond acceptors and donors had a substantial impact on the viscosity, pH, density, and chemical structure of the NADESs. The resulting extracts demonstrated the highest recovery rates, resulting in the greatest total phenolic content (35.05 ± 0.15 mg GAE/100 g) and total anthocyanin content (17.22 ± 0.296 mg CGE/100 g). This extract from the Prunus domestica fruit, which contains anthocyanin, has anti-sickling effects on sickle cell erythrocytes. The Emmel test was used to assess the in vitro anti-sickling effects of the NADES extract. The percent sickling of negative control was found to be 40.88%. And it was discovered that the NADES extract of CHCL: ETHGly was 14.71% at an effective concentration of 0.2% (v/v), and the NADES extract of CHCl: GLY was discovered to be 20.94% at an effective concentration of 0.2% (v/v). An open new perspective has been established toward synthesizing eco‐friendly CSA@g‐C3N4 employing surface engineering. The carbon nitride modified through camphorsulfonic acid was designed and developed in a category of the new generation of photocatalysts for the oxidation of benzyl alcohol and thioanisole in the existence of a natural deep eutectic solvent (NADES). In comparison with pure g‐C3N4, not only does CSA@g‐C3N4 exhibit an extraordinarily higher ability for harvesting visible light stemming from declining the recombination rate of electrons/holes dependent on PL results but it also reveals notable photocatalytic oxidation capability in the transformation of alcohols as well as thiols into relevant compounds. In addition, non‐metal compound (CSA) incorporation would result in considerably diminishing the energy band gap value from 2.8 to 2.28 eV to escalate the visible‐light absorption of g‐C3N4. While the conventional consensus implies that inherent properties of photocatalysts bring on high photoactivity, this study indicates that deploying choline chloride–urea deep eutectic solvent as an external factor plays the role of photoactivity accelerator. Furthermore, readily recycling and reusability can be achieved for the photocatalytic setup of CSA@g‐C3N4 ascribed to its heterogeneous nature with no drop in the photoactivity. The objective of this study was to investigate the impact of nutrient starvation on the growth, biochemical, metabolomic, and biological traits of Lavandula viridis L'Hér and Thymus lotocephalus G. López and R. Morales in vitro cultures. In both species, a reduction in shoot growth and in the production of chlorophyll and carotenoids was observed in cultures grown under nutrient-deficient media (especially Fe and N) compared to those grown under control conditions. The highest levels of hydrogen peroxide and lipid peroxidation, two indicators of oxidative stress, were observed in L. viridis cultures grown under N deficiency and in T. lotocephalus under P and Fe limitation. The results demonstrated that nutrient deficiency led to a 72% and 62% increase in the quantified phenolic compounds in L. viridis and T. lotocephalus, respectively. The highest concentrations of the major compound in both species—rosmarinic acid—were observed in cultures grown under Mg-deficient (60.7 ± 1.0 mg/gDW) and Fe-deficient (50.0 ± 0.4 mg/gDW) conditions in L. viridis and T. lotocephalus, respectively. Furthermore, nutrient starvation enhanced the capacity of green extracts to inhibit three enzymes (tyrosinase, elastase, and hyaluronidase) associated with anti-aging and their antioxidant properties. Mangifera pajang Kosterm., or 'Bambangan,' a fruit known for its rich phytochemical content, was investigated for the effects of ultrasound‐assisted osmotic dehydration (UAOD) on pretreated M. pajang fruit pulp (PMPF) and drying at different temperatures (40, 50, 60, 70, and 80°C). Thin‐layer drying kinetics were analyzed using six models, with the Midilli and Kucuk model being the most suitable, showing optimal effective moisture diffusivity and activation energy. PMPF dried at 50°C, using natural deep eutectic solvents (NADES) as the extraction solvent, exhibited the highest extraction yield (64.13 ± 1.08%), mangiferin content (0.5818 ± 0.00164 mg/g), total phenolic content (146.2081 ± 1.33 mg GAE/g DW), total flavonoid content (2.4737 ± 0.06 mg RE/g DW), and DPPH inhibition activity (95.87 ± 0.16%), along with the best color retention compared to untreated M. pajang fruit pulp. This highlights its potential for nutraceutical and functional food applications, with UAOD as pretreatment effectively preserving food quality and color. Practical Applications: This article provides new insights into the drying and pretreatment of Mangifera pajang fruits. Rich in phytochemicals, M. pajang offers numerous health benefits. Pretreatment methods such as ultrasound‐assisted osmotic dehydration (UAOD) and oven drying at different temperatures significantly enhance the retention of bioactive compounds like phenolics and flavonoids, as well as antioxidant activity and color stability. The study identified optimal drying temperatures, conditions, drying rates, and activation energy to maximize these qualities. Using mathematical models for drying kinetics, this research offers practical guidance for the food processing industry. It provides an operational tool for manufacturers to optimize the drying process, reduce operational costs, and ensure the preservation of chemical qualities while extending the shelf life of M. pajang fruits. Spilanthol is a major N-alkylamide constituent of Acmella oleracea (L.) R.K. Jansen with diverse pharmacological properties. We recently showed the applicability of NADES (natural deep eutectic solvents) for the green extraction of spilanthol. However, the purification of targets from NADES poses a challenging step due to their non-volatility. A simple green method to retrieve spilanthol with minimal instrumental effort was devised, fractioning NADES (choline chloride/methylurea, choline chloride/1,2-propanediol, choline chloride/citric acid) and dry ethanolic extracts by SPE on C18 material, eluting merely with ethanolic solutions. The relative distribution of spilanthol and organic adulteration in SPE fractions were detected by HPLC-DAD, followed by scale-up, quantification and purity determination in an NMR-based approach. Isocratic elution with 52% ethanol (v/v) proved suitable in all experiments. The three purest 10 mL fractions combined yielded 12.21 mg spilanthol at 71.65% purity from NADES extract ChCl/P (choline chloride/1,2-propanediol, molar ratio 1:2, +20% m/m water). Ethanolic extract samples showed purities ranging from 77.27 to 80.27% in combined raw fractions. For all samples, purity increased by removing non-soluble substances from organic solutions. Pooled NADES extract fractions showed 89.71% in final samples, ethanolic extracts 87.25 to 91.93%. The highest purities of individual fractions per extract were 89.23 to 94.15%. This cheap and simple purification process is promising to acquire spilanthol for research purposes or as a sample preparation step before HPLC on a semi-preparative to preparative scale, as the substance is highly priced and scarcely available on the market. Exploiting by-products from the oenological industry to extract antioxidant chemicals is a shared goal that combines the need to reduce the wine sector's environmental impact with the need to improve the availability of these biomolecules, according to a circular economy approach. Natural deep eutectic solvents (NaDES) have recently captured researchers' interest as a safer and more environmentally friendly alternative to traditional solvents due to their effectiveness, low toxicity, and stability. In this work, we set out to investigate several NaDES for the extraction of phenolic chemicals from local monovarietal grape pomace resulting from different vinification procedures (including both red and rosé vinification of Negroamaro and Primitivo grapes; rosé vinification of Susumaniello grapes and white vinification of Chardonnay, Fiano and Malvasia bianca grapes), with the additional goal of generalizing the use of NaDES to extract chemicals of interest from organisms selected from the wide plant biodiversity. Three binary choline chloride-based NaDES (DES-Lac, DES-Tar, and DES-Gly, with lactic acid, tartaric acid, and glycerol as hydrogen bond donors, respectively) were compared to ethanol as a conventional solvent, and the extracts were evaluated using HPLC/MS and colorimetric techniques. The results revealed that each NaDES produces a substantially higher total phenolic yield than ethanol (up to 127.8 mg/g DW from Primitivo rosé grape pomace). DES-Lac and DES-Tar were more effective for anthocyanins extraction; the most abundant compound was malvidin 3-O-glucoside (highest extraction yield with DES-Lac from Susumaniello pomace: 29.4 mg/g DW). Regarding phenolic compounds, DES-Gly was the most effective NaDES producing results comparable to ethanol. Unexpectedly, Chardonnay pomace has the greatest content of astilbin. In most cases, grape pomace extracts obtained by rosé and white vinification provided the maximum yield. As a result, NaDES have emerged as a viable alternative to traditional organic solvent extraction techniques, allowing for higher (or equal) yields while significantly lowering costs, hazards, and environmental impact. Pea protein hydrolysate (PPH) typically exhibits a bitter taste and limited umami-enhancing properties, which impedes its use in umami seasoning products. This study aimed to increase the umami-enhancing capability of PPH by using the Maillard reaction with the integration of NADES (Natural Deep Eutectic Solvents). By comparing different Maillard reaction treatments on PPH's umami-enhancement, this study highlighted the advantages of our method and identified potential key reaction precursors, shown by changes in the main sample composition. Using sugar-derived NADES for the Maillard reaction with added xylose notably improved PPH's umami-enhancing ability by 100%. The optimal reaction conditions were heating at 55 °C for 90 min. This was the lowest reaction temperature reported to date, significantly reducing the challenges of industrial production. PPH primarily comprised small peptides and free amino acids. In the NADES system, free hydrophobic amino acids and peptides exhibited higher Maillard reactivity. With the addition of xylose, about 50% of free hydrophobic, 60% of basic, and 20% of acidic amino acids, and over half of the peptides were further consumed. PCA results and other data suggested that the Maillard reaction products of specific free amino acids and umami-potential peptides in PPH with xylose in NADES might be essential for boosting umami-enhancing ability. This study introduced a approach to selectively prepare flavor enhancers with exceptional umami-enhancing ability using PPH, while also satisfying the cost and safety requirements of industrial production. The objective of this study was to analyze the chemical profile of extracts from different fruit parts of the perennial arborescent cactus Opuntia leucotricha by applying ultra-high-performance liquid chromatography–high-resolution mass spectrometry (UHPLC-HRMS) and to evaluate their biological properties (antioxidant and enzyme inhibitory capacities). Extracts were obtained from the fruit pulp (without or with seeds), seeds, and peel by ultrasound-assisted extraction (UAE) using the conventional solvent methanol 50% and two Natural Deep Eutectic Solvents (NADESs) (glycerol:urea, 1:1 and citric acid:sorbitol, 1:2). A total of 33 compounds were identified, including phenolics, fatty acids, and others. Phenolic acids were the most abundant class of phenolics identified in all fruit parts, with the highest concentration observed in the methanol extracts from peel (593.02 µg/gDW), followed by seed-containing pulp (69.03 µg/gDW), pulp (57.83 µg/gDW), and seeds (39.97 µg/gDW). The second most effective extractant was NADES 1 (glycerol:urea, 1:1), which was also successful in extracting compounds with antioxidant capacity. Overall, the extracts demonstrated considerable enzyme inhibitory activities, with the greatest effects observed against α-amylase and α-glucosidase. The results indicate that O. leucotricha fruits could be a promising source of bioactive compounds, and NADES a viable alternative to organic solvents for their industrial exploitation. For plant metabolite extraction, natural deep eutectic solvents (NADESs) have many benefits over conventional solvents and ionic liquids. These advantages include high solubility and extraction ability, a low melting point (<100 °C), low toxicity, environmental friendliness, recyclability, and better biodegradability. This study analyses a natural deep eutectic solvent for Acmella oleracea (A. oleracea) metabolite extraction, considering the following process parameters: temperature, component ratio in the eutectic solvent, water addition, solid/liquid ratio, and extraction duration. NADESs were synthesised using a simple heating method, and the synthesis of the NADESs was verified by Fourier transform infrared spectroscopy (FTIR). In terms of total polyphenol content (TPC) and flavonoid content (TFC), the betaine/propanediol ratio in a NADES of 1:3 and S/L = 1:5 yielded the highest efficiency. A value of 8.37 mg GAE/mL was obtained for TPC by ultrasound-assisted extraction with 40% water addition, 25 °C extraction temperature, and 60 min contact time. The best result in terms of TFC was 14.50 mg QE/mL obtained through ultrasound-assisted extraction with 0% water added, 25 °C extraction temperature, and 60 min contact time. [ABSTRACT FROM AUTHOR]

Abstract This work explored the use of natural deep eutectic solvents (NADES) for extracting phenolic compounds with antioxidant activity from Uruguayan olive pomace (OP), a by-product of olive oil mills. Among nine NADES tested, lactic acid-glucose (La-Gc) was the most effective solvent for phenolic compound extraction. Further investigation focused on optimizing the extraction process using La-Gc. Response surface methodology was employed to analyze the impact of extraction temperature, water content in NADES and solid–liquid ratio on total phenols content (TPC) and antioxidant activity (FRAP and DPPH assays). Temperature and solid–liquid ratio greatly influenced TPC and antioxidant activity, while water content in NADES only showed significant influence on antioxidant activity. The optimum extraction conditions that maximized TPC and antioxidant activity were 80 $${}^{\circ }\text {C}$$ ∘ C , 68% (w/w) of water in NADES and solid–liquid ratio of 0.014 g/mL, showing a TPC of 15.56 mg GAE/g db, FRAP of 178.14 $$\mu$$ μ mol FSE/g db and DPPH of 72.75 $$\mu$$ μ mol TRE/g db, with hydroxytyrosol content of 1.24 mg/g db. These values were significantly higher than those of extracts obtained from conventional solvents under identical extraction conditions. These results highlight the suitability of the present extraction method using La-Gc as solvent to obtain phenolic compounds with antioxidant activity from OP.

The present investigation examines the antimicrobial and antifungal characteristics of natural deep eutectic solvents (NADES) and apple vinegar in relation to a diverse array of bacterial and fungal strains. The clinical bacterial strains, including gram-negative and gram-positive, and the fungal pathogen Candida albicans, were subjected to solid medium diffusion to determine the inhibitory effects of these compounds. The results show that NADES has superior antimicrobial and antifungal action compared to apple vinegar. The observed inhibitory zones for apple vinegar and NADES varied in length from 16.5 to 24.2 and 16 to 52.5 mm, respectively. The results obtained indicate that no synergy is observed for this mixture (50% AV + 50% NADES). The range of values for bactericidal concentrations (MBC) and minimal inhibitory concentrations (MIC) was 0.0125 to 0.2 and 0.0125 to 0.4 µl/ml, respectively. Antibacterial and antifungal chemicals may be found in apple vinegar and NADES, with NADES offering environmentally safe substitutes for traditional antibiotics. Additional investigation is suggested to refine these compounds for a wide range of bacteria, which could create antimicrobial solutions that are both highly effective and specifically targeted, thereby offering extensive potential in medicine and the environment. The industries are searching for greener alternatives for their productions due to the rising concern about the environment and creation of waste and by-products without industrial utility for that specific line of products. This investigation describes the development of two stable nanostructured lipid carriers (NLCs): one is the formulation of a standard NLC, and the other one is the same NLC formulation associated with a natural deep eutectic solvent (NaDES). The research presents the formulation paths of the NLCs through completeness, which encompass dynamic light scattering (DLS), zeta potential tests, and pH. Transmission electron microscopy (TEM) and confocal microscopy were performed to clarify the morphology. Cytotoxicity tests with zebrafish were realized, and the results are complementary to the in vitro outcomes reached with fibroblast L132 tests by the MTT technique and the zymography test. Infrared spectroscopy and X-ray diffractometry tests elucidated the link between the physicochemical characteristics of the formulation and its behavior and properties. Different cooling techniques were explored to prove the tailorable properties of the NLCs for any industrial applications. In conclusion, the compiled results show the successful formulation of new nanocarriers based on a sustainable, eco-friendly, and highly tailorable technology, which presents low cytotoxic potential. This was the first study to use local Southern Thai ingredients, namely, nipa palm syrup (NS) and nipa palm vinegar (NV), in the formulation of natural deep eutectic solvent (NADES) for the recovery of bioactive curcumin from turmeric. Five NADES formulations (A to E) were obtained by varying concentration of liquid media (NS, NV, and water) and characterized using FTIR. When compared to other formulations and 80% methanol, the NADES D with a NS to NV to water ratio of 1:5:5 (w/w/w) recovered the most curcumin content (p < 0.05). The extraction conditions of microwave-assisted extraction (MAE) with selected NADES were optimized using response surface methodology (RSM) to maximize curcumin recovery. NADES D achieved the highest curcumin content (43.04 mg/g) from turmeric at a solvent ratio of 1:10, microwave power of 1000 W, and extraction time of 51 s. The NADES D-based curcumin extract outperformed all antioxidant activities (DPPH∙ scavenging activity and FRAP). The NADES D–based extract is non-toxic to RAW264.7 cells at up to 62.50 µg/mL. To replace the hazardous organic solvents currently in use, one of the key objectives of green chemistry has been the development of green solvents obtained from natural sources. Natural deep eutectic solvents (NADES) are bio‐based natural compounds, namely primary metabolites like organic acids, sugar, alcohols, amines, and amino acids, which are acceptable to the environment and may eventually replace the conventionally used harsh organic solvents. This study focuses on the recent developments in the realm of these innovative materials as well as the conventional deep eutectic solvents (DES)‐based techniques. A more in‐depth examination of DES's applications in various chemical processes is covered after a description of its definition, manufacture, and unique characteristics. Grape pomace is the main by-product obtained from wine production that is still enriched in bioactive compounds. Within a framework of waste/by-product reuse through a sustainable approach, various green methods were utilized in this work to recover anthocyanins from the pomace resulting from "Sangiovese" grape vinification. Ultrasound- and Microwave-Assisted Extractions (UAE and MAE) were coupled with the use of green solvents, such as acidified water, an ethanol/water mixture, and Natural Deep Eutectic Solvents (NaDES), and their efficacy was compared with that of a conventional method based on a methanol/acidified water mixture. The Total Anthocyanin Index ranged from 36.9 to 75.2 mg/g DW for UAE, and from 54.4 to 99.6 mg/g DW for MAE, while resulting in 47.1 mg/g DW for conventional extraction. A Design of Experiments (DoE) approach was applied to MAE, the most efficient technique. Temperature, time, and the solid-to-liquid ratio were set as X variables, while malvidin-3-O-glucoside content and antioxidant activity were used as response variables, measured by High-Performance Liquid Chromatography with Diode Array Detection (HPLC-DAD) and 2,2-Diphenyl-1-picrylhydrazyl (DPPH) assay, respectively. The correlation between temperature and time and the antioxidant activity of the extract was positive, while it was found to be negative when considering malvidin-3-O-glucoside concentration as a response variable. Thus, the optimal conditions in temperature, time and solid-to-liquid ratio were different depending on the chosen variable. Introduction: The present research describes the utilization of natural deep eutectic solvent in combination with ultrasound for the extraction of milk thistle polyphenols. The extracts obtained under different conditions were evaluated for their in vitro antioxidant activities and hepatoprotective activities in Albino mice (in vivo). Methods: The extraction parameters involving liquid-to-solid ratio (S/L), ethanol-to-natural deep eutectic solvent ratio (EtOH/DES), extraction time (t), and ultrasound treatment time were investigated and optimized to enhance the recovery of bioactives, their phenolic content, Trolox equivalent antioxidant capacity (TEAC), and radical scavenging capacity (RSC). Results and discussions: The extraction of milk thistle polyphenols using glucose/citric acid (1:1)-based natural deep eutectic liquid coupled with ultrasound for 1.0min under an S/L of 6.2, EtOH/DES 9, and extraction time of 120.8min offered 35.89 ± 2.29 g/100 g of crude extracts, which were three-fold higher than that by conventional solvent extraction (CSE). Each gram of milk thistle extracts thus obtained comprised 377.93 ± 6.17mg GAE of TPC and exhibited 298.70 ± 3.06 μmol TEAC and 93.16 ± 0.58 percent inhibition of DPPH free radicals. In addition, the administration of milk thistle extracts obtained as given above at 25 mg/Kg body weight in Albino mice significantly (p ≤ 0.05) improved the liver function parameters at the end of treatment (7 days). Conclusion: Overall, it was observed that NaDES in combination with medium-level ultrasound energy (700 watt) might work as a sustainable choice to enhance the recovery of bioactive phenolics from milk thistle powder without compromising their antioxidant and hepatoprotective potential. Xanthorrhizol, an important marker of Curcuma xanthorrhiza, has been recognized for its different pharmacological activities. A green strategy for selective xanthorrhizol extraction is required. Herein, natural deep eutectic solvents (NADESs) based on glucose and organic acids (lactic acid, malic acid, and citric acid) were screened for the extraction of xanthorrhizol from Curcuma xanthorrhiza. Ultrasound-assisted extraction using glucose/lactic acid (1:3) (GluLA) gave the best yield of xanthorrhizol. The response surface methodology with a Box–Behnken Design was used to optimize the interacting variables of water content, solid-to-liquid (S/L) ratio, and extraction to optimize the extraction. The optimum conditions of 30% water content in GluLA, 1/15 g/mL (S/L), and a 20 min extraction time yielded selective xanthorrhizol extraction (17.62 mg/g) over curcuminoids (6.64 mg/g). This study indicates the protective effect of GluLA and GluLA extracts against oxidation-induced DNA damage, which was comparable with those obtained for ethanol extract. In addition, the stability of the xanthorrhizol extract over 90 days was revealed when stored at −20 and 4 °C. The FTIR and NMR spectra confirmed the hydrogen bond formation in GluLA. TheDES are formed by mixing a Hydrogen Bond Donor (HBD) and a Hydrogen Bond Acceptor (HBA) in appropriate molar ratios. These solvents have been shown to enhance drug solubility, permeability, and delivery. The main objective of the present article is to review these advantages of TheDES. TheDES show unique properties, such as low toxicity, biodegradability, improved bioavailability and enhanced drug delivery of poorly soluble active pharmaceutical ingredients. They are also biocompatible in nature which makes them a promising candidate for various therapeutic applications, including drug formulations, drug delivery and other biomedical uses. The development and utilization of TheDES shows significant advancement in pharmaceutical research, providing new opportunities for improving drug delivery. The current study was carried out by conducting a systematic literature review that identified relevant papers from indexed databases. Numerous studies and research are cited and quoted in this article to demonstrate the effectiveness of TheDES in enhancing drug solubility, permeability, and delivery. All chosen articles were selected considering their significance, quality, and approach to addressing issues. As a result, various TheDES were identified that can be formulated in different ways: one component can act as a vehicle for an API, either HBD or HBA can be an API, both HBD and HBA can be APIs, or the individual components of DES are not therapeutically active but the resulting DES possesses therapeutic activity. Natural deep eutectic solvents (NADES) have emerged as promising green solvents, particularly useful in sample treatment procedures. For the first time, different types of NADES were prepared and evaluated as potential phenolic extraction solvents from tea samples (Camellia sinensis). The NADES, composed of lactic acid:glycerol:water in a molar ratio of 1:1:3, provided the best results for the simultaneous extraction of the target phenolic compounds. This NADES was also characterized by using nuclear magnetic resonance (NMR), and the presence of nuclear Overhauser enhancement effects confirmed its supramolecular structure. The experimental conditions affecting the miniaturized ultrasonic-assisted extraction (UAE) were optimized to maximize extraction efficiency by performing a design of experiments. The proposed sample treatment was combined with ultra-high performance liquid-chromatography coupled to tandem mass spectrometry detection (UHPLC-MS/MS) to determine 21 phenolic compounds including catechin derivatives and other minor compounds. The UAE-UHPLC-MS/MS method was successfully characterized and applied to the analysis of various commercial green, black, white and red tea samples. In all cases, 18 phenolic compounds were determined with concentrations ranging from 0.3 to 29,125 mg kg−1. The proposed sample treatment is a high-throughput, easy, fast and robust alternative to conventional procedures, which reduces organic solvent consumption and costs, aligned with green sample preparation principles.

To reduce the environmental pollution caused by organic solvents and improve the extraction efficiency, a green and high-effective natural deep eutectic solvent (NADES) was screened out and prepared for the extraction of flavonoids from Fructus aurantii based on the COMOS-RS and experimental verification. The results showed the results of theoretical screening and experimental verification were consistent, and Ch-Lea was the best extraction solvent, which further proved the COMOS-RS was an efficient tool for the solvent screening. Then, the extraction conditions were investigated. The optimum extraction conditions were as follows: molar ratio (1:2), water content (30%), solid–liquid ratio (1/60 g/g) and extraction time (15 min), yet the extraction temperature and power did not significantly affect the extraction yield. Under the optimal extraction condition, the extraction yield of flavonoids was 122.68 mg/g. Compared with the traditional extraction solvents (methanol and ethanol), Ch-Lea has a higher extraction yield. And the green assessment was conducted based on Eco-Scale methodology, which results indicated the Ch-Lea was more environmentally friendly than methanol and ethanol. Meanwhile, the Ch-Lea has good reusability, and the extraction yields from four reuses were 122.68 mg/g, 125.13 mg/g, 122.56 mg/g and 105.63 mg/g, respectively. Moreover, the extraction mechanism was delved into using molecular dynamics (MD) simulation and reduced density gradient (RDG) analysis. The hydrogen bond interaction, particularly the O-H bonds, between Ch-Lea and the naringin molecule emerged as the primary force driving the extraction process. In summary, Ch-Lea is a green, economical, and highly effective solvent, making it a promising candidate for flavonoid extraction from Fructus aurantii. Its potential as an alternative to traditional extraction solvents further underscores its value.

The recently explored synergistic combination of graphene-based materials and deep eutectic solvents (DESs) is opening novel and effective avenues for developing sensing devices with optimized features. In more detail, remarkable potential in terms of simplicity, sustainability, and cost-effectiveness of this combination have been demonstrated for sensors, resulting in the creation of hybrid devices with enhanced signal-to-noise ratios, linearities, and selectivity. Therefore, this review aims to provide a comprehensive overview of the currently available scientific literature discussing investigations and applications of sensors that integrate graphene-based materials and deep eutectic solvents, with an outlook for the most promising developments of this approach. Extraction of natural compounds generally uses organic solvents, which have a high risk to human health. Green solvents based on the natural deep eutectic solvents (NADES) have been used for the extraction of active compounds from coffee cherry waste. This research has formulated eight NADES based on choline chloride by combining fructose, citric acid, glycerol, proline, glucose, xylose, propylene glycol, and lactic acid with a mole ratio of 1:2. The FTIR analysis of NADES showed that there was a hydrogen bond interaction with OH group peaks in the wave number range 3012-3357 cm-1. NADES application was used for phenolic and tannin extraction from coffee cherry waste. The extraction efficiency of phenolic was combined of choline chloride with HBD of proline>xylose>glucose>glycerol>lactic acid >fructose>propylene glycol>citric acid. The maximum phenolic content in the extract using choline chloride-proline was 11.81 mg GAE/g. The ability of NADES to extract tannins using choline chloride with the combination of glucose>proline>lactic acid>propylene glycol>glycerol>citric acid> fructose. The maximum tannin content was obtained at 1.58% using NADES based on choline chloride with glucose. The methods and solvents employed in routine extraction protocols essentially impact the composition of the resulting extracts, i.e., the relative abundances of individual biologically active metabolites and the quality and stability of the isolates. Natural deep eutectic solvents (NADESs) represent a new class of environmentally friendly solvents, which are recognized as promising extractants alternative to conventional organic liquids. However, their relative efficiencies when applied in different extraction workflows are still poorly characterized. Therefore, here, we compare the potential of three extraction methods for the extraction of biologically active natural products from Aralia elata var. mandshurica with selected natural deep eutectic solvents (NADESs) using a non-targeted metabolomics approach. The non-targeted metabolite profiling relied on reversed-phase ultra-high-performance liquid chromatography–high-resolution mass spectrometry (RP-UHPLC-HR-MS). The roots of A. elata were extracted by maceration, ultrasound-assisted extraction (UAE), and vibrocavitation-assisted extraction (VAE). Principal component analysis (PCA) revealed a clear separation of the extracts obtained with the three extraction methods employed with NADES1 (choline chloride/malic acid) and NADES2 (sorbitol/malic acid/water). Based on the results of the hierarchical clustering analysis obtained for the normalized relative abundances of individual metabolites and further statistical evaluation with the t-test, it could be concluded that NADES1 showed superior extraction efficiency for all the protocols addressed. Therefore, this NADES was selected to compare the efficiencies of the three extraction methods in more detail. PCA followed by the t-test yielded only 3 metabolites that were more efficiently extracted by maceration, whereas 46 compounds were more abundant in the extracts obtained by VAE. When VAE and UAE were compared, 108 metabolites appeared to be more abundant in the extracts obtained by VAE, whereas only 1 metabolite was more efficiently recovered by UAE. These facts clearly indicate the advantage of the VAE method over maceration and UAE. Seven of the twenty-seven metabolites tentatively identified by tandem mass spectrometry (MS/MS) were found in the roots of A. elata for the first time. The extraction of bioactive compounds from food wastes or by‐products with natural deep eutectic solvents (NADES) could be an attractive alternative to traditional hydroalcoholic extraction but needs to be optimized taking advantage of modeling. This work aims to propose, develop, and discuss a new model for the extraction of phenolic compounds from spent coffee grounds (SCGs) using NADES to overcome some gaps related to the kinetic extraction models and the surface response methodology. The model presented is based on the same assumption as the kinetic model, but improving it to be able to predict phenolic extraction changing different process parameters (e.g., water content, time of extraction, temperature, etc.) using betaine: triethylene glycol (molar ratio 1:2). The model was validated on the extraction of total phenolic compounds from SCG as a function of water content of NADES ranging from 30 to 60% v/v. There was good agreement between experimental and simulated data and even, in some cases, better than with the second‐order kinetic models. In addition, the new model needs fewer parameters to be regressed concerning the specific case investigated in this work. This model and methodology can be the basis for further modeling applicable to other bioactives, and/or other NADES and feedstocks. Practical Applications: Natural deep eutectic solvents (NADES) can be a potential sustainable solvent for the valorization of food by‐products as they can be used to extract bioactive compounds with high added value. This extraction process needs to be optimized depending on several factors like water content in NADES solution, solids ratio, extraction time, and temperature. The model validated in this work on spent coffee ground (SCG) could be used for this purpose, improving the gaps related to the kinetic extraction models and the surface response methodology (RSM). The new model enables predicting the behavior of the process as a function of the parameters previously mentioned and reduces the number of experimental trials required compared with RSM. In particular, the new model was validated on the variation of water content in NADES solution. Certainly, this model will need to be further validated, but, once further studies are conducted, this model could be used for scale‐up calculation and design optimization of the industrial process. 'Type V' natural deep eutectic solvents (TV NADES) are a novel class of solvent media with many potential 'green chemistry' applications. Here, we assess the potential of two TV NADES: menthol:carvacrol (M:C) and menthol:thymol (M:T), and a single terpenoid extraction media carvacrol (C), as media from total lipid extraction of three compositionally diverse, freeze‐dried marine tissues: hoki (Macruronus novaezelandaiae), jack mackerel (Trachurus declivis) and green‐lipped mussel (kuku, Perna canaliculus). Room temperature extraction yields into these media were benchmarked relative to lipid yields from chloroform‐methanol‐water extractions. Relative extraction yields using the M:C and M:T media were (both) 87% for jack mackerel, 85%–87% for hoki, and 43%–47% for green‐lipped mussels. Comprehensive chemical analyses of TV NADES lipid extracts suggested that neutral lipids were extracted in greater yield than phospholipids, and that phosphatidylinositol, lysophospholipids and (in the case of mussels) ceramideaminoethylphosphonates were particularly resistant to extraction into C, M:C and M:T. C—a thymol isomer that, unlike menthol and thymol is a liquid at room temperature—appeared to perform (marginally) better than M:C and M:T systems, implying there may be no advantage to using the TV NADES over a single terpenoid with similar chemistry for total lipid extraction. In the article, results of dissolution of metal oxides using a natural deep eutectic solvent (NDES or NADES) based on betaine, malic acid and ethylene glycol are presented. The obtained deep eutectic solvent was characterised by determining its physicochemical properties including density, viscosity as a function of temperature. The thermal stability of NADES was also determined. For CuO, equilibrium, kinetics, and thermodynamics of the oxide dissolution process in NADES and H 2 O were determined. Dissolution studies of selected metal oxides including ZnO, MgO, CaO, Fe 2 O 3 in NADES and water were also carried out. It was found that the solubility of the metal oxides was 2 to 100 times higher in NADES compared to the solubility in water, despite the neutral pH of NADES. The study, using XRD and SEM methods, also shows changes in the morphology of particles subjected to dissolution in water and in NADES. The results indicate the possibility of extracting metal oxides by dissolution in NADES. With a growing focus on green chemistry, the extraction of natural products with natural deep eutectic solvents (NADES), which are eutectic mixtures of hydrogen bond donors and acceptors, has become an ever-expanding field of research. However, the use of NADES for the extraction of spilanthol from Acmella oleracea (L.) R.K.Jansen has not yet been investigated. Therefore, in this study, 20 choline chloride-based NADES, and for comparison, ethanol, were used as green extraction agents for spilanthol from Acmella oleracea flower heads. The effects of time, water addition, and temperature on NADES extractions were investigated and analysed by HPLC-DAD quantification. Additionally, UHPLC-DAD-ESI-MSn results for dichloromethane extracts, as well as the isolation of spilanthol and other main constituents as reference compounds, are reported. The best green extraction results were achieved by choline chloride (ChCl) with 1,2-propanediol (P, 1:2 molar ratio, +20% water) at 244.58 µg/mL, comparable to yields with ethanol (245.93 µg/mL). Methylurea (MeU, 1:2, +20% water) also showed promising results as a hydrogen bond donor in combination with choline chloride (208.12 µg/mL). In further experiments with NADES ChCl/P (1:2) and ChCl/MeU (1:2), extraction time had the least effect on spilanthol extraction with NADES, while yield decreased with water addition over 20% and increased with extraction temperature up to 80 °C. NADES are promising extraction agents for the extraction of spilanthol, and these findings could lead to applicable extracts for medicinal purposes, due to their non-toxic constituents. Natural deep eutectic solvents (NADES) are eco-sustainable, non-toxic, non-volatile, renewable, reusable, and biodegradable, and are composed of natural compounds. NADES were developed as a new-generation solvent for extracting chitin from black soldier fly (BSF) pupae, and its effectiveness for demineralization and deproteinization was determined. Here, two promising NADES, consisting of mixtures of choline chloride-betaine-xylitol (NADES A) and choline chloride-malic acid-water (NADES B), were tested. Fourier transform infrared (FTIR) spectroscopy, thermogravimetric analysis (TGA), X-ray diffraction (XRD), and scanning electron microscopy (SEM) were used to investigate the changes in the chemical composition of the extracted chitin. α-chitin revealed at wavenumbers 1660-1500 cm-1, in the amide group and decomposed at 330-350°C. NADES A and NADES B have a crystalline index of 91.65% and 90.65%, respectively. The chitin-NADES A and chitin-NADES B surfaces reveal the repetition of square, pentagonal, and hexagonal units (250× magnification) and fibrils (25,000× magnification). This study provides a green approach for chitin production from BSF and reveals the potential of NADES for extracting bio-polymers from natural sources.

Brewer’s spent grains (BSG) offer valuable opportunities for valorization beyond its conventional use as animal feed. Among its components, lignin—a natural polymer with inherent antioxidant properties—holds significant industrial potential. This work investigates the use of microwave-assisted extraction combined with acidic natural deep eutectic solvents (NaDESs) for efficient lignin recovery, evaluating three different NaDES formulations. The results indicate that choline chloride–lactic acid (ChCl-LA), a NaDES with superior thermal stability as confirmed via thermogravimetric analysis (TGA), is an ideal solvent for lignin extraction at 150 °C and 15 min, achieving a balance of high yield and quality. ChCl-LA also demonstrated good solubility and cell disruption capabilities, while microwaves significantly reduced processing time and severity. Under optimal conditions, i.e., 150 °C, 15 min, in the presence of ChCl-LA NaDES, the extracted lignin achieved a purity of up to 79% and demonstrated an IC50 (inhibitory concentration 50%) of approximately 0.022 mg/L, indicating a relatively strong antioxidant activity. Fourier transform infrared (FTIR) and 2D-HSQC NMR (heteronuclear single quantum coherence nuclear magnetic resonance) spectroscopy confirmed the successful isolation and preservation of its structural integrity. This study highlights the potential of BSG as a valuable lignocellulosic resource and underscores the effectiveness of acidic NaDESs combined with microwave extraction for lignin recovery.

The objective of this study was to investigate the impact of nutrient starvation on the growth, biochemical, metabolomic, and biological traits of Lavandula viridis L’Hér and Thymus lotocephalus G. López and R. Morales in vitro cultures. In both species, a reduction in shoot growth and in the production of chlorophyll and carotenoids was observed in cultures grown under nutrient-deficient media (especially Fe and N) compared to those grown under control conditions. The highest levels of hydrogen peroxide and lipid peroxidation, two indicators of oxidative stress, were observed in L. viridis cultures grown under N deficiency and in T. lotocephalus under P and Fe limitation. The results demonstrated that nutrient deficiency led to a 72% and 62% increase in the quantified phenolic compounds in L. viridis and T. lotocephalus, respectively. The highest concentrations of the major compound in both species—rosmarinic acid—were observed in cultures grown under Mg-deficient (60.7 ± 1.0 mg/gDW) and Fe-deficient (50.0 ± 0.4 mg/gDW) conditions in L. viridis and T. lotocephalus, respectively. Furthermore, nutrient starvation enhanced the capacity of green extracts to inhibit three enzymes (tyrosinase, elastase, and hyaluronidase) associated with anti-aging and their antioxidant properties.

The objective of this study was to analyze the chemical profile of extracts from different fruit parts of the perennial arborescent cactus Opuntia leucotricha by applying ultra-high-performance liquid chromatography–high-resolution mass spectrometry (UHPLC-HRMS) and to evaluate their biological properties (antioxidant and enzyme inhibitory capacities). Extracts were obtained from the fruit pulp (without or with seeds), seeds, and peel by ultrasound-assisted extraction (UAE) using the conventional solvent methanol 50% and two Natural Deep Eutectic Solvents (NADESs) (glycerol:urea, 1:1 and citric acid:sorbitol, 1:2). A total of 33 compounds were identified, including phenolics, fatty acids, and others. Phenolic acids were the most abundant class of phenolics identified in all fruit parts, with the highest concentration observed in the methanol extracts from peel (593.02 µg/gDW), followed by seed-containing pulp (69.03 µg/gDW), pulp (57.83 µg/gDW), and seeds (39.97 µg/gDW). The second most effective extractant was NADES 1 (glycerol:urea, 1:1), which was also successful in extracting compounds with antioxidant capacity. Overall, the extracts demonstrated considerable enzyme inhibitory activities, with the greatest effects observed against α-amylase and α-glucosidase. The results indicate that O. leucotricha fruits could be a promising source of bioactive compounds, and NADES a viable alternative to organic solvents for their industrial exploitation.

Spilanthol is a major N-alkylamide constituent of Acmella oleracea (L.) R.K. Jansen with diverse pharmacological properties. We recently showed the applicability of NADES (natural deep eutectic solvents) for the green extraction of spilanthol. However, the purification of targets from NADES poses a challenging step due to their non-volatility. A simple green method to retrieve spilanthol with minimal instrumental effort was devised, fractioning NADES (choline chloride/methylurea, choline chloride/1,2-propanediol, choline chloride/citric acid) and dry ethanolic extracts by SPE on C18 material, eluting merely with ethanolic solutions. The relative distribution of spilanthol and organic adulteration in SPE fractions were detected by HPLC-DAD, followed by scale-up, quantification and purity determination in an NMR-based approach. Isocratic elution with 52% ethanol (v/v) proved suitable in all experiments. The three purest 10 mL fractions combined yielded 12.21 mg spilanthol at 71.65% purity from NADES extract ChCl/P (choline chloride/1,2-propanediol, molar ratio 1:2, +20% m/m water). Ethanolic extract samples showed purities ranging from 77.27 to 80.27% in combined raw fractions. For all samples, purity increased by removing non-soluble substances from organic solutions. Pooled NADES extract fractions showed 89.71% in final samples, ethanolic extracts 87.25 to 91.93%. The highest purities of individual fractions per extract were 89.23 to 94.15%. This cheap and simple purification process is promising to acquire spilanthol for research purposes or as a sample preparation step before HPLC on a semi-preparative to preparative scale, as the substance is highly priced and scarcely available on the market. Organic solvents can be reused, and preliminary scale-up possibilities are shown.

Exploiting by-products from the oenological industry to extract antioxidant chemicals is a shared goal that combines the need to reduce the wine sector’s environmental impact with the need to improve the availability of these biomolecules, according to a circular economy approach. Natural deep eutectic solvents (NaDES) have recently captured researchers’ interest as a safer and more environmentally friendly alternative to traditional solvents due to their effectiveness, low toxicity, and stability. In this work, we set out to investigate several NaDES for the extraction of phenolic chemicals from local monovarietal grape pomace resulting from different vinification procedures (including both red and rosé vinification of Negroamaro and Primitivo grapes; rosé vinification of Susumaniello grapes and white vinification of Chardonnay, Fiano and Malvasia bianca grapes), with the additional goal of generalizing the use of NaDES to extract chemicals of interest from organisms selected from the wide plant biodiversity. Three binary choline chloride-based NaDES (DES-Lac, DES-Tar, and DES-Gly, with lactic acid, tartaric acid, and glycerol as hydrogen bond donors, respectively) were compared to ethanol as a conventional solvent, and the extracts were evaluated using HPLC/MS and colorimetric techniques. The results revealed that each NaDES produces a substantially higher total phenolic yield than ethanol (up to 127.8 mg/g DW from Primitivo rosé grape pomace). DES-Lac and DES-Tar were more effective for anthocyanins extraction; the most abundant compound was malvidin 3-O-glucoside (highest extraction yield with DES-Lac from Susumaniello pomace: 29.4 mg/g DW). Regarding phenolic compounds, DES-Gly was the most effective NaDES producing results comparable to ethanol. Unexpectedly, Chardonnay pomace has the greatest content of astilbin. In most cases, grape pomace extracts obtained by rosé and white vinification provided the maximum yield. As a result, NaDES have emerged as a viable alternative to traditional organic solvent extraction techniques, allowing for higher (or equal) yields while significantly lowering costs, hazards, and environmental impact.

Green chemistry emphasizes the isolation of biologically active compounds from plants and biomass to produce renewable, bio-based products and materials through sustainability and circularity-driven innovation processes. In this work, we have investigated the extraction of rosmarinic acid (RA), a phenolic acid with several biological properties, from aromatic herbs using ultrasounds and low environmental risk natural deep eutectic solvents (NADES). Various solvent mixtures have been investigated, and the parameters influencing the process have been studied by a mixture-process experimental design to identify the optimal RA extraction conditions. The extraction yield has been calculated by HPLC-diode array analysis. The lactic acid:ethylene glycol mixture using an ultrasound-assisted process has been found to be the most versatile solvent system, giving RA yields 127–160% higher than hydroalcoholic extraction (70% ethanol). The deep eutectic solvent nature of lactic acid:ethylene glycol has been demonstrated for the first time by multi-technique characterization (1H-NMR and 13C-NMR, DSC, and W absorption properties). The aqueous raw extract has been directly incorporated into poly(vinyl alcohol) to obtain films with potential antibacterial properties for applications in the field of food and pharmaceutical packaging. Meat industry preservation methods to extend the shelf life and maintain the microbial safety of meat products. There is big shortage in research that conducting examination of the antimicrobial activity of Abutilon pannosum choline chloride based extracts (APCCBE). This study aimed to assess the efficacy of APCCBE in inhibiting the growth of common meat pathogens, investigated the potential impact of APCCBE treatment on the meat products shelf life. The polar fraction of Abutilon pannosum leaves fine powder was obtained via a solid-liquid extraction, GC-MS analysis, Antibacterial activity of fresh APCCBE was performed against four strains of microorganisms: Staphylococcus aureus, Escherichia coli, Bacillus subtilis using Muller Hinton and six antibiotics discs (50 mg/mL) as follows; "Cefoxitin (FOX), Cephalothin (KF), Cotrimoxazole (TS), Gentamicin (GM), Augmentin (AUG) and Ampicillin (AP). Determination of the storage period of meatballs using packages containing APCCBE were each 100g of meatballs were wrapped by each package film concentration, stored in dark and cool conditions and compared with meatballs wrapped by uncoated PE film package. the study has declared that the APCCBE possess efficient antimicrobial effects against the test bacterial strains. These findings provide valuable knowledge in pathogenic bacteria treatment and food preservation, in addition to its ability in extension the shelf life of the chilled meatballs to about 2 weeks instead of 5 days only in case of control meatballs before and after cooking. Hydrophilic deep eutectic solvents are actively positioned as efficient extractants for removing heterocyclic compounds from light hydrocarbon fractions. Of particular interest is the subclass of natural deep eutectic solvents (NaDESs), since they contain substances of exclusively natural origin. However, these processes have not been systematically studied to date in extraction equipment. To study the process of countercurrent extraction of pyridine, quinoline, and indole from a model solution of light hydrocarbon fractions using commercial equipment, a series of NaDESs based on citric and malic acids, xylitol, and water was used for the first time in this work. The high extraction capacity of these NaDES was demonstrated in laboratory experiments, and the extraction mechanism was determined. A detailed study of the efficiency of extraction of heterocycles with varying process conditions allowed us to move on to studying the process using extractors of the mixer–settler type. From the model solution of light hydrocarbon fractions, pyridine, quinoline, and indole were removed to concentrations <1 ppm by countercurrent extraction using a cascade of six mixer–settlers. Coriandrum sativum L. seeds are widely recognized for their traditional use in medicine. Among the most investigated components, the terpenoid linalool and monounsaturated petroselinic acid have attracted interest for their nutritional value. Instead, minor attention was paid to the polyphenolic fraction, resulting still being incomplete today. This study aimed to develop a systematic approach in which green natural deep eutectic solvents (NADES) were combined with conventional (maceration, MAC) or non-conventional (ultrasound-assisted extraction, UAE) techniques in a one-step methodology to recover polyphenols from coriander seeds. The NADES system choline chloride–citric acid (ChCl:CA, 1:1) was firstly evaluated, coupled with MAC or UAE, and then compared with ChCl–Urea (ChCl:Ur, 1:1) and ChCl–Glucose (ChCl:Glu, 1:1) under optimal conditions (20 min extraction time). The system ChCl:Ur UAE significantly improved the extraction of chlorogenic acid and its isomer (453.90 ± 4.77 and 537.42 ± 1.27 µg/g, respectively), while the system ChCl:Glu UAE improved the extraction of protocatechuic, caffeic and p-coumaric acids (131.13 ± 6.16, 269.03 ± 4.15 and 57.36 ± 0.06 µg/g, respectively). The highest levels of rutin were obtained with ChCl:CA-based NADES when the MAC technique was applied (820.31 ± 28.59 µg/g). These findings indicate that the NADES composition could be appropriately modulated to tailor extraction towards higher levels of a desirable bioactive for further applications. Natural deep eutectic solvents (NADES) are eco-sustainable, non-toxic, nonvolatile, renewable, reusable, and biodegradable, and are composed of natural compounds. NADES were developed as a new-generation solvent for extracting chitin from black soldier fly (BSF) pupae, and its effectiveness for demineralization and deproteinization was determined. Here, two promising NADES, consisting of mixtures of choline chloride-betaine-xylitol (NADES A) and choline chloride-malic acid-water (NADES B), were tested. Fourier transform infrared (FTIR) spectroscopy, thermogravimetric analysis (TGA), X-ray diffraction (XRD), and scanning electron microscopy (SEM) were used to investigate the changes in the chemical composition of the extracted chitin. a-chitin revealed at wavenumbers 1660-1500 cm-1, in the amide group and decomposed at 330-350°C. NADES A and NADES B have a crystalline index of 91.65% and 90.65%, respectively. The chitin-NADES A and chitin-NADES B surfaces reveal the repetition of square, pentagonal, and hexagonal units (250× magnification) and fibrils (25,000× magnification). IntroductionThe present research describes the utilization of natural deep eutectic solvent in combination with ultrasound for the extraction of milk thistle polyphenols. The extracts obtained under different conditions were evaluated for their in vitro antioxidant activities and hepatoprotective activities in Albino mice (in vivo).MethodsThe extraction parameters involving liquid-to-solid ratio (S/L), ethanol-to-natural deep eutectic solvent ratio (EtOH/DES), extraction time (t), and ultrasound treatment time were investigated and optimized to enhance the recovery of bioactives, their phenolic content, Trolox equivalent antioxidant capacity (TEAC), and radical scavenging capacity (RSC).Results and discussionsThe extraction of milk thistle polyphenols using glucose/citric acid (1:1)-based natural deep eutectic liquid coupled with ultrasound for 1.0 min under an S/L of 6.2, EtOH/DES 9, and extraction time of 120.8 min offered 35.89 ± 2.29 g/100 g of crude extracts, which were three-fold higher than that by conventional solvent extraction (CSE). Each gram of milk thistle extracts thus obtained comprised 377.93 ± 6.17 mg GAE of TPC and exhibited 298.70 ± 3.06 μmol TEAC and 93.16 ± 0.58 percent inhibition of DPPH free radicals. In addition, the administration of milk thistle extracts obtained as given above at 25 mg/Kg body weight in Albino mice significantly (p ≤ 0.05) improved the liver function parameters at the end of treatment (7 days).ConclusionOverall, it was observed that NaDES in combination with medium-level ultrasound energy (700 watt) might work as a sustainable choice to enhance the recovery of bioactive phenolics from milk thistle powder without compromising their antioxidant and hepatoprotective potential.

Aktivitas antioksidan dengan flavonoid memiliki hubungan yang kuat karena dipengaruhi oleh gugus fungsi yang berikatan dengan struktur utamanya. Studi ini bertujuan agar mengetahui pengaruh pelarut serta metode ekstraksi yang digunakan terhadap kadar total flavonoid serta aktivitas antioksidan dari ekstrak daun mangga gedong. Mangga gedong diekstraksi dengan metode kombinasi MAE-NADES (sodium acetate dan lactic acid 1:3 dalam air suling hingga suhu mencapai 70 ). Mangga gedong diekstraksi dalam suhu dan waktu (80 :20’, 80 :10’, 60 :20’, 60 :10’). Selanjutnya ekstrak ditentukan nilai kadar flavonoid total serta aktivitas antioksidannya. Hasil studi menyampaikan ekstrak daun mangga gedong memiliki nilai kadar total flavonoid yang tinggi, dengan nilai masing-masing 80 :20’ 6,5994%, 80 :10’ 5,5504%, 60 :20’ 5,0890%, dan 60 :10’ 3,8902%. Selain itu, ekstrak daun mangga gedong mempunyai aktivitas antioksidan dengan nilai IC50 masing-masing 80 :20’ 21,666 ppm, 80 :10’ 39,734 ppm, 60 :20’ 61,687 ppm, dan 60 :10’ 135,487 ppm. Dari hasil studi mampu disampaikan bahwa dengan metode ekstraksi MAE menggunakan pelarut NADES memiliki nilai kadar total flavonoid serta aktivitas antioksidan. Ekstrak daun mangga gedong yang terbaik adalah dengan suhu dan waktu 80 :20’. Dimana nilai kadar total flavonoid berbanding lurus dengan aktivitas antioksidan, semakin tinggi nilai senyawa flavonoid yang dihasilkan maka aktivitas antioksidan akan semakin tinggi.

Abstract Shale instability is a significant problem, accounting for approximately 70% of wellbore challenges during drilling operations. To address this issue, shale inhibitors are commonly added to drilling mud to stabilize the encountered shale formations. Ionic liquids and deep eutectic solvents have been recognized as effective shale inhibitors due to their environmental friendliness and effectiveness. However, despite their advantages, these solutions still lack full environmental sustainability, leading researchers to explore more natural alternatives like Natural Deep Eutectic Solvents (NADES). This study focuses on the synthesis and application of a NADES composed of Potassium Chloride (KCl) and glycerine, aiming to enhance the environmental profile and efficacy of shale inhibitors. The NADES was prepared with a 1:8 molar ratio at a temperature of 60 °C. Characterization analyses, including Fourier Transform Infrared and Thermogravimetric Analysis, confirmed the formation of bonds between –(OH) and Cl− and demonstrated the NADES's thermal stability up to 200 °C. Incorporating 3% NADES into water-based mud, prepared following API 13B-1 standards, resulted in significant improvements in mud rheology. Specifically, the addition of 3% NADES reduced filtrate volume by 14.2% and mud cake thickness by 19.2%. Furthermore, the 3% NADES exhibited remarkable inhibition of clay swelling by 69.23% and demonstrated a shale recovery rate of 58%. When compared with 3% KCl and 3% 1-ethyl-3-methylimidazolium chloride (EMIM-Cl), the NADES-based mud outperformed both in terms of efficacy. These findings were further supported by additional analyses, including d-spacing measurements (XRD), Zeta Potential, Surface tension, and Field Emission Scanning Electron Microscopy. Additionally, the rheological behavior of the NADES-based mud aligned with the Yield Power Law at both 25 °C and 100 °C. The study's findings contribute to the advancement of greener drilling practices and highlight the applicability of KCl-based NADES as a potential drilling fluid additive.

The industries are searching for greener alternatives for their productions due to the rising concern about the environment and creation of waste and by-products without industrial utility for that specific line of products. This investigation describes the development of two stable nanostructured lipid carriers (NLCs): one is the formulation of a standard NLC, and the other one is the same NLC formulation associated with a natural deep eutectic solvent (NaDES). The research presents the formulation paths of the NLCs through completeness, which encompass dynamic light scattering (DLS), zeta potential tests, and pH. Transmission electron microscopy (TEM) and confocal microscopy were performed to clarify the morphology. Cytotoxicity tests with zebrafish were realized, and the results are complementary to the in vitro outcomes reached with fibroblast L132 tests by the MTT technique and the zymography test. Infrared spectroscopy and X-ray diffractometry tests elucidated the link between the physicochemical characteristics of the formulation and its behavior and properties. Different cooling techniques were explored to prove the tailorable properties of the NLCs for any industrial applications. In conclusion, the compiled results show the successful formulation of new nanocarriers based on a sustainable, eco-friendly, and highly tailorable technology, which presents low cytotoxic potential.

Xanthorrhizol, an important marker of Curcuma xanthorrhiza, has been recognized for its different pharmacological activities. A green strategy for selective xanthorrhizol extraction is required. Herein, natural deep eutectic solvents (NADESs) based on glucose and organic acids (lactic acid, malic acid, and citric acid) were screened for the extraction of xanthorrhizol from Curcuma xanthorrhiza. Ultrasound-assisted extraction using glucose/lactic acid (1:3) (GluLA) gave the best yield of xanthorrhizol. The response surface methodology with a Box–Behnken Design was used to optimize the interacting variables of water content, solid-to-liquid (S/L) ratio, and extraction to optimize the extraction. The optimum conditions of 30% water content in GluLA, 1/15 g/mL (S/L), and a 20 min extraction time yielded selective xanthorrhizol extraction (17.62 mg/g) over curcuminoids (6.64 mg/g). This study indicates the protective effect of GluLA and GluLA extracts against oxidation-induced DNA damage, which was comparable with those obtained for ethanol extract. In addition, the stability of the xanthorrhizol extract over 90 days was revealed when stored at −20 and 4 °C. The FTIR and NMR spectra confirmed the hydrogen bond formation in GluLA. Our study reported, for the first time, the feasibility of using glucose/lactic acid (1:3, 30% water v/v) for the sustainable extraction of xanthorrhizol.
