Abstract

Varroa destructor (Anderson & Trueman, 2000) causes the most important parasitosis of beekeeping in the world. For this reason, prevention is needed to avoid colony death. The most typical treatments involve synthetic acaricides. However, the use of these acaricides results in the emergence of resistant populations of mites to these products and in the appearances of drug residues in products of the hives. Compounds of essential oils have emerged as an alternative to traditional acaricides; however the toxicity produced by these mixtures is currently poorly explored. The aim of this work was to assess, by means of in vitro tests with adult bees, how acaricidal action and toxic interactions in a binary mixture of essential oil compounds (Thymol, Phellandrene, Eucalyptol, Cinnamaldehyde, Myrcene, and Carvacrol) affect V. destructor. Calculations of LC50 ’s of the individual compounds on A. mellifera and V. destructor made clear that the toxic effect of each compound is different for both species. Thymol and Phellandrene turned out to be lethal for mites at lower concentrations than for bees. The binary mixture of these two substances presented a different toxicity than one produced by each pure compound, as it was highly selective for mites in bioassays at 24 hours through complete exposure to both A. mellifera and V. destructor. These results make such formulations optimal substances to be considered as alternative controls for the parasitosis.

Abstract

The European honey bee (Apis mellifera L.) is known to be affected by such stress factors as pathogen load, poor nutrition and depressed immunity. Nosema ceranae is one of the main parasites that affect colony populations. The relationship between the stress factors and honey bee-bacteria symbiosis appears as an alternative to enhance bee health. The aim of this study was to evaluate the effect of the oral administration of bacterial metabolites produced by Lactobacillus johnsonii AJ5 on nutritional parameters, the N. ceranae development and the performance of A. mellifera colonies. Laboratory assays were performed and demonstrated that the bacterial metabolites did not have a toxic effect on bees. Field trial showed an increase of colonies population over time. Also, a decreasing trend of fat bodies per bee was detected in all colonies but there were no evident changes on abdomen protein content at the end of the assay. Lastly, N. ceranae prevalence showed a tendency to reduce with the organic acids. Future studies should be performed to increase our knowledge of the physiological effects of bacterial metabolites on the health of bee colonies.

Abstract

Aim of study: To explore three isolated phytomolecules: indoleacetic acid (IAA), gibberellic acid (GA), and the secondary me-tabolite p-coumaric acid (CUM): (1) evaluating their toxicity against Apis mellifera larvae and adults under controlled conditions in the laboratory; (2) searching for antimicrobial activity against Paenibacillus larvae. Area of study: Honey bee larvae and adults were collected from the experimental apiary of the “Centro de Investigación en Abejas Sociales (CIAS)” (-37.9348798,-57.682817), Institute of the National University of Mar del Plata (UNMdP), Argentina. Material and methods: Paenibacillus larvae strains were isolated from beehives from different provinces of Argentina (Buenos Aires, Córdoba and Entre Ríos) showing clinical symptoms of the American foulbrood. All strains (S1, S2, S3, S4) were genotypi-cally identified using PL5 and PL4 primers and characterized as genotype ERIC1. Then standard essays were performed to determined toxicity of phytomolecules in honey bees and antimicrobial activity through the broth microdilution method. Main results: The diet with GA, IAA and CUM did not present toxic effects in larvae or adult bees, and only CUM showed an-timicrobial activity against P. larvae. In this study, we obtained in vitro values of MNIC (minimum non-inhibitory concentration) of 500 μg mL-1 and a MIC (minimum inhibitory concentration) of 650 μg mL-1 for CUM. Research highlights: The obtained results remark its potential as a natural alternative for the control of P. larvae, avoiding the problems generated by the use of synthetic antibiotics such as the resistance phenomena and the contamination of hive’s products. Abstract

The natural composition of nutrients present in food is a key factor determining the immune function and stress responses in honeybee (Apis mellifera). We previously demonstrated that a supplement of abscisic acid (ABA), a natural component of nectar, pollen and honey, increases honeybee colony survival overwinter. Here we further explored the role of ABA in in vitro-reared larvae exposed to low temperatures. Four-days-old larvae (L4) exposed to 25°C for 3 days showed lower survival rates and delayed development compared to individuals growing at a standard temperature (34°C). Cold-stressed larvae maintained higher levels of ABA for longer than do larvae reared at 34°C, suggesting a biological significance for ABA. Larvae fed with ABA-supplemented diet completely prevent the low survival rate due to cold stress and accelerate the adult emergence. ABA modulates the expression of genes involved in metabolic adjustments and stress responses: Hexamerin 70b, Insulin Receptor Substrate, Vitellogenin and Heat Shock Proteins 70. AmLANCL2, the honeybee ABA receptor, is also regulated by cold stress and ABA. These results support a role for ABA increasing the tolerance of honeybee's larvae to low temperatures through priming effects.

Abstract

The effects of parasites, pathogens and agrochemical exposure combined with the cold temperatures of winter in temperate-climate regions contribute to the immunosuppression of honey bees. Recently, we demonstrated that abscisic acid (ABA) dietary supplementation increased the tolerance of Apis melliferabrood to low temperatures. Here, we study the relationship between cold exposure, dietary ABA supplementation, and the expression of genes involved in the immune response of in-vitro-reared bee larvae. We found that cold exposure induced the expression of several immune-associated genes in honey bee larvae, supporting that the immune system is active during the cold stress response in A. mellifera. At the same time, the ingestion of ABA alone increased the expression levels of the Toll pathway receptor toll18Wand nitric oxide synthase(nos) genes, which were induced also by low temperatures, reinforcing the connection between honey bees’ response to cold stress and ABA.

Abstract

The honey bee Apis mellifera is affected by many parasites posing a serious threat to our ecosystem, agriculture and apiculture. The study of A. meliifera immune response represents a great complement for developing integrated disease management strategies. This work presents updated data concerning the in vitro cellular immune response of A. meliifera fifth instar larvae (L5) and newly emerged workers (W). Haemocytes from A. mellifera L5 and W clearly displayed different appearance in-vitro. The blood cells from these two developmental stages yielded behavioural differences regarding attachment and spreading. However, haemocytes extracted from L5 and W exhibited strong similarities with respect to their general (multicellular) behavioural pattern. This multi-haemolytic process, triggered after glass contact, could result from an encapsulation-like response. At the same time, an experimental system for studying A. mellifera cellular immunity in-vitro upon recognition of glass surfaces and haemolytic encapsulation response is herein developed. La abeja mellifera Apis mellifera se ve afectada por muchos parásitos que suponen una grave amenaza para nuestro ecosistema, la agricultura y la apicultura. Estudiar el sistema inmunológico de A. mellifera aportaría información de gran relevancia para el desarrollo de estrategias de manejo integrado de plagas. Hasta el momento, no contamos con datos actualizados relativos a la defensa inmune celular “hemocítica” de A. mellffera. En este trabajo estudiamos in-vitro aspectos relacionados a la inmunidad celular de larvas del quinto estadio (L5) y obreras recién emergidas (W) de A. meliifera. Los hemocitos de L5 y W mostraron claras diferencias respecto a la apariencia, adherencia y esparcimiento (fenotipo de activación inmunológica celular) in-vitro. Sin embargo los hemocitos de L5 y W exhibieron un patrón de comportamiento multicelular general asociado a una respuesta de encapsulación disparada después del contacto con una superficie de vidrio in-vitro. De esta forma, en este trabajo describimos un sistema experimental fiable para realizar experimentos in-vitro para ahondar en el estudio de la aptitud inmune celular de A. mellffera, desde las primeras etapas del reconocimiento de lo “no propio” mediado por los hemocitos hasta respuestas multicelulares relacionadas a la encapsulación. Abstract

Summary: The role of Ann Arbor staging in determining treatment intensity after achieving a negative positron emission tomography (PET) has not been established in classical Hodgkin lymphoma (cHL). Patients with stage I–IV cHL, received three cycles of ABVD (doxorubicin, bleomycin, vinblastine, dacarbazine) and an interim PET scan (PET3). PET3‐negative patients received no further therapy. PET3‐positive patients received three additional cycles of ABVD plus involved‐field radiation therapy or salvage chemotherapy, if refractory to ABVD, and were re‐evaluated by PET scan (PET6). Study endpoints were 3‐year progression‐free survival (PFS) and overall survival (OS) rates. Two hundred and thirty‐nine patients with early‐stage and 138 with advanced‐stage were evaluable. Overall, 260 patients (70%) were PET3‐negative and had higher 3‐year PFS (90% vs. 65%; P < 0·0001) and OS (98% vs. 92%; P = 0·007) rates than PET3‐positive patients. All PET3‐negative patients, regardless of disease stage at diagnosis, achieved similarly good PFS (90–91%; P = 0·76) and OS (97–99%). The only independent prognostic factor for PFS was PET3‐negativity (Hazard ratio 3·8; 95% confidence interval 2·4–6·3; P < 0·0001). This study suggests that cHL patients who achieve a negative PET3 following ABVD have an excellent outcome, regardless of stage at diagnosis. The aim of this study was to evaluate the effect of the oral administration of bacterial metabolites produced by Lactobacillus johnsonii AJ5 on nutritional parameters, the N. ceranae development and the performance of A. mellifera colonies. Laboratory assays were performed and demonstrated that the bacterial metabolites did not have a toxic effect on bees. Field trial showed an increase of colonies population over time. Also, a decreasing trend of fat bodies per bee was detected in all colonies but there were no evident changes on abdomen protein content at the end of the assay. Lastly, N. ceranae prevalence showed a tendency to reduce with the organic acids. Future studies should be performed to increase our knowledge of the physiological effects of bacterial metabolites on the health of bee colonies. [ABSTRACT FROM AUTHOR]

Abstract

The honey bee tracheal mite Acarapis woodi(Rennie) (Acari: Tarsonemidae) is an internal obligate parasite of the respiratory system of adult honey bees (Apis mellifera), which may be one of the factors causing the loss of bee colonies and the decrease in the beekeeping industry. In Argentina, the last prevalence study of A. woodiwas in 1998, and therefore there is no current data on the prevalence and impact of the mite in colonies of A. melliferain this country. For this reason, the objective of this study was to determine the prevalence of A. woodiin honey bee colonies of apiaries belonging to different geographic regions of Argentina (Northwest, Chaco, Mesopotamic, Pampean and Patagonic) using two different methods of diagnosis. One method was traditional microscopy, which is based on direct visualization of A. woodiin the prothoracic tracheas of bees, and the other technique was molecular analysis using real-time PCR with specific primers to amplify CO1 gene of A. woodi. To perform the analysis of prevalence, bees were collected during winter, because during this season, populations of A. woodiare more abundant in bee colonies. The results were negative for both methods of diagnosis, with prevalence equal to 0%. Considering that in recent years it has been reported an increased mortality in bee colonies in Argentina, we note that acarapisosis is not currently impacting on the disappearance of A. melliferacolonies in Argentina.

Abstract

SummaryThe honey bee Apis melliferais affected by many parasites posing a serious threat to our ecosystem, agriculture and apiculture. The study of A. meliiferaimmune response represents a great complement for developing integrated disease management strategies. This work presents updated data concerning the in vitrocellular immune response of A. meliiferafifth instar larvae (L5) and newly emerged workers (W). Haemocytes from A. melliferaL5 and W clearly displayed different appearance in-vitro.The blood cells from these two developmental stages yielded behavioural differences regarding attachment and spreading. However, haemocytes extracted from L5 and W exhibited strong similarities with respect to their general (multicellular) behavioural pattern. This multi-haemolytic process, triggered after glass contact, could result from an encapsulation-like response. At the same time, an experimental system for studying A. melliferacellular immunity in-vitroupon recognition of glass surfaces and haemolytic encapsulation response is herein developed.

Abstract

Introduction:The evaluation of response to imatinib in CML patients (pts) as guide to clinical management is undergoing substantial changes of criteria. The degree of reduction of total leukemia cell mass by imatinib as measured by FISH and RQ-PCR studies, correlates with progression-free survival. Therapeutic decisions, as dose scalation, is mandatory in pts with rising level of bcr/abl transcripts.
