Abstract Background The nursing industry's stability and progress are adversely affected by the high attrition rate and shortage of nurses; therefore, it is critical to investigate the variables that influence the professional stability of nurses. The sense of professional mission and career success have positive significance for reducing nurses' job burnout. The purpose of this study is to explore the potential mediating role of psychological resilience in this relationship. Methods Self-reported questionnaires were utilized by 335 intensive care unit (ICU) nurses to assess their sense of professional mission, psychological resilience, and career success in this cross-sectional study. A structural equation model was developed to validate the relationship between the variables. Results There is a correlation among sense of professional mission, psychological resilience and career success. Significant mediating effect of psychological resilience exists between sense of professional mission and career success. Conclusions In this study, psychological resilience plays an intermediary role between sense of professional mission and career success, which provides support for further understanding the mechanism between sense of professional mission and career success and bolstering the case for devising comprehensive intervention strategies for psychological resilience. Nursing managers should focus on nurses' sense of professional mission and psychological resilience, and implement strategies to enhance nurses' psychological resilience in order to boost their career success.

Background: The prediction of response to immunotherapy mostly depends on the programmed death-ligand 1 (PD-L1) immunohistochemistry (IHC) status, and the 22C3 pharmDx assay has been approved in esophageal squamous cell carcinoma (ESCC). However, the widespread use of the 22C3 pharmDx assay is limited due to its availability. Thus, alternative PD-L1 assays are needed. We aimed to investigate the analytical and clinical diagnostic performances of four PD-L1 assays and to compare their concordances with the 22C3 pharmDx assay. Methods: The PD-L1 22C3 pharmDx assay was performed on the Dako Autostainer Link 48 platform, three testing assays (PD-L1 E1L3N XP antibody [Ab], PD-L1 BP6099 Ab and PD-L1 CST E1L3N Ab) on the Leica BOND-MAX/III platform, and one testing assay (PD-L1 MXR006 Ab) on the Roche VENTANA Benchmark Ultra platform. A total of 218 ESCC cases from four centers were included in this retrospective study. Professionals from each center stained and read the IHC slides independently and determined the combined positive score (CPS) and the tumor proportion score (TPS). Results: Regarding analytical performance, the four testing assays demonstrated good correlations with the 22C3 pharmDx assay when evaluated by the TPS or CPS (ρ > 0.8 for all four assays). Regarding diagnostic performance (CPS ≥ 10 was used as the cutoff), the four testing assays showed moderate concordances with the 22C3 pharmDx assay (kappa > 0.7 for all four assays). The overall percent agreements between each testing assay and the 22C3 pharmDx assay was at least 87.2 %. Conclusion: This study provides insight into the potential interchangeability of the four PD-L1 assays with the 22C3 pharmDx assay.

Abstract Acute pancreatitis (AP) is a common systemic inflammatory disease resulting from the activation of trypsinogen by various incentives in ICU. The annual incidence rate is approximately 30 out of 100,000. Some patients may progress to severe acute pancreatitis, with a mortality rate of up to 40%. Therefore, the goal of this article is to explore the key genes for effective diagnosis and treatment of AP. The analysis data for this study were merged from two GEO datasets. 1357 DEGs were used for functional enrichment and cMAP analysis, aiming to reveal the pathogenic genes and potential mechanisms of AP, as well as potential drugs for treating AP. Importantly, the study used LASSO and SVM-RFE machine learning to screen the most likely AP occurrence biomarker for Prdx4 among numerous candidate genes. A receiver operating characteristic of Prdx4 was used to estimate the incidence of AP. The ssGSEA algorithm was employed to investigate immune cell infiltration in AP. The biomarker Prdx4 gene exhibited significant associations with a majority of immune cells and was identified as being expressed in NKT cells, macrophages, granulocytes, and B cells based on single-cell transcriptome data. Finally, we found an increase in Prdx4 expression in the pancreatic tissue of AP mice through immunohistochemistry. After treatment with recombinant Prdx4, the pathological damage to the pancreatic tissue of AP mice was relieved. In conclusion, our study identified Prdx4 as a potential AP hub gene, providing a new target for treatment.

Acute kidney injury (AKI) is associated with both kidney function loss and increased mortality. In the pathological progression of ischemia-reperfusion-induced AKI, the surge of reactive oxygen species (ROS) plays a crucial role. To combat this, mitochondrial-targeted antioxidant therapy shows great promise as mitochondria are the primary source of ROS in AKI. However, most strategies aiming to target mitochondria directly result in nanodrugs that are too large to pass through the glomerular system and reach the renal tubules, which are the main site of damage in AKI. This study focused on synthesizing a Megalin receptor-targeted polymeric prodrug, low molecular weight chitosan-thioketal-elamipretide (LMWC/TK/Ela), to mitigate excessive ROS in renal tubular epithelial cells for AKI. This soluble polymeric prodrug has the ability to successfully reach the tubular site by crossing the glomerular barrier. Once there, it can responsively release elamipretide, which possesses excellent antioxidative properties. Therefore, this research offers a novel approach to actively target renal tubular epithelial cells and intracellular mitochondria for the relief of AKI.

Acute kidney injury (AKI), a condition associated with reactive oxygen species (ROS), causes high mortality in clinics annually. Active targeted antioxidative therapy is emerging as a novel strategy for AKI treatment. In this study, we developed a polymeric prodrug that targets the highly expressed Megalin receptor on proximal tubule cells, enabling direct delivery of N-Acetylcysteine (NAC) for the treatment of ischemia reperfusion injury (IRI)-induced AKI. We conjugated NAC with low molecular weight chitosan (LMWC), a biocompatible and biodegradable polymer consisting of glucosamine and N-acetylglucosamine, to enhance its internalization by tubular epithelial cells. Moreover, we further conjugated triphenylphosphonium (TPP), a lipophilic cation with a delocalized positive charge, to low molecular weight chitosan-NAC in order to enhance the distribution of NAC in mitochondria. Our study confirmed that triphenylphosphonium-low molecular weight chitosan-NAC (TLN) exhibits remarkable therapeutic effects on IRI-AKI mice. This was evidenced by improvements in renal function, reduction in oxidative stress, mitigation of pathological progress, and decreased levels of kidney injury molecule-1. These findings suggested that the polymeric prodrug TLN holds promising potential for IRI-AKI treatment.

The riparian green space covers the inner buffer zone to maintain the ecological characteristics of the river and the outer coordination green zone to serve the adjacent urban–rural functional areas. The complex topography and landform of mountainous areas make riparian green space closely connected with the natural process and spatial function, which is the focus of urban and rural construction. However, the “out-of-context” and “one-size-fits-all” means of multi-department planning have separated the natural process maintenance and constructive utilization of riparian green space in urban–rural areas, resulting in serious conflicts between protection and development and low control effectiveness. Under the background of prioritization of ecology and the control of urban–rural spatial resources in the whole region, we draw lessons from the theories and methods of related disciplines and takes the integration of protection and utilization as the basis point. We present a new conceptual framework that follows the natural ecological characteristics of rivers and appropriately integrates the demands of urban–rural development and construction, and discusses the differentiated planning and regulation method of riparian green space in mountainous urban–rural areas based on transect zoning. This conceptual framework not only ensures the healthy operation of the ecosystem inside the riparian zone, but also improves the composite service function outside the riparian zone, and minimizes the impact of urban–rural construction on the riparian zone.

Potassium-ion batteries (PIBs) have been widely studied owing to the abundant reserves, widespread distribution, and easy extraction of potassium (K) resources. Molybdenum disulfide (MoS2) has received a great deal of attention as a key anode material for PIBs owing to its two-dimensional diffusion channels for K+ ions. However, due to its poor electronic conductivity and the huge influence of embedded K+ ions (with a large ionic radius of 3.6 Å) on MoS2 layer, MoS2 anodes exhibit a poor rate performance and easily collapsed structure. To address these issues, the common strategies are enlarging the interlayer spacing to reduce the mechanical strain and increasing the electronic conductivity by adding conductive agents. However, simultaneous implementation of the above strategies by simple methods is currently still a challenge. Herein, MoS2 anodes on reduced graphene oxide (MoS2/rGO) composite were prepared using one-step hydrothermal methods. Owing to the presence of rGO in the synthesis process, MoS2 possesses a unique scaled structure with large layer spacing, and the intrinsic conductivity of MoS2 is proved. As a result, MoS2/rGO composite anodes exhibit a larger rate performance and better cycle stability than that of anodes based on pure MoS2, and the direct mixtures of MoS2 and graphene oxide (MoS2-GO). This work suggests that the composite material of MoS2/rGO has infinite possibilities as a high-quality anode material for PIBs.

Abstract Objective This study was designed to analyze the structural characteristics of the intestinal flora of elderly Uygur patients with sarcopenia, thereby providing new ideas for clinical treatment. Methods Firstly, fecal samples were collected from 40 elderly Uygur patients with sarcopenia (Sarcopenia group) and 40 healthy people (Control group). Next, significant differences in the intestinal flora between the two groups were analyzed based on 16S rDNA high‐throughput sequencing. The linear discriminant analysis effect size (LEfSe) was used to estimate the magnitude of the effect of each component (species) abundance on the differential effect. Additionally, an analysis was also performed on the relationship between the intestinal flora and the cytokines in the peripheral blood of patients with sarcopenia. Results The results of β diversity showed that there were differences in the structure of the intestinal flora between the two groups. Besides, the phylum level of intestinal flora between the two groups was not significantly different. However, the difference was significant in the intestinal flora at the order, family, and genus levels between the two groups. Among them, Lachnoclostridium, Photobacterium, Anaerobic Bacillus, Hydrogenophilus, and Eubacterium were enriched in the Sarcopenia group; Prevotella 9, Firmicutes FCS020 group, Streptobacillus, Aggregatibacter, Corynebacterium, Clostridium Difficile, and Haloanaerobium were enriched in the Control group. The LEfSe outcomes further showed that Lachnoclostridium was highly enriched in the Sarcopenia group; Prevotella 9 and Firmicutes FCS020 group were significantly enriched in the Control group. Furthermore, the relative abundance of Lachnoclostridium and Streptobacillus were significantly different in patients with high and low IL‐6 levels. Conclusion In conclusion, Lachnoclostridium is significantly enriched in the intestines of elderly Uygur patients with sarcopenia; the increase in Lachnoclostridium abundance and the decrease in Streptobacillus abundance are associated with high levels of IL‐6. Therefore, abnormal intestinal flora is related to inflammatory reflexes in patients with sarcopenia.

This study aimed to explore the epidemic, clinical characteristics, and molecular and virulence attributes of Klebsiella pneumoniae serotype K54 (K54-Kp). A retrospective study was conducted on 328 strains of Klebsiella pneumoniae screened in a Chinese hospital from January 2016 to December 2019. The virulence genes and antibiotic resistance genes (ARGs) were detected by PCR, and a drug sensitivity test was adopted to detect drug resistance. Multilocus sequence typing (MLST) and PFGE were performed to determine the clonal correlation between isolates. Biofilm formation assay, serum complement-mediated killing, and Galleria mellonella infection were used to characterize the virulence potential. Our results showed that thirty strains of K54-Kp were screened from 328 strains of bacteria, with an annual detection rate of 2.29%. K54-Kp had a high resistance rate to antibiotics commonly used in the clinic, and patients with hepatobiliary diseases were prone to K54-Kp infection. MLST typing showed 10 sequence typing, mainly ST29 (11/30), which concentrated in the B2 cluster. Abstract Background Primary liver immature teratoma is extremely rare and only 4 cases have been reported, let alone with growing teratoma syndrome (GTS) and/or gliomatosis peritonei (GP). Case presentation Here, we report a case of a 44-year-old female presenting with progressive abdominal distension and elevated serum alpha fetal protein (AFP) level. CT/MRI scans revealed a large cystic-solid mass in the right lobe of the liver, accompanied with implant or metastasis in the abdominal cavity. Pathologic examination at biopsy suggested immature teratoma. After 4 cycles of chemotherapy, an MRI showed a slight increase in tumor size. Therefore, surgical resection of the right lobe of the liver was performed. The final histological diagnosis was a mature teratoma (tumor size 28 cm × 14 cm × 13 cm), with no residual immature component, and the diagnosis of GTS was considered. The patient continued to receive 2 courses of postoperative chemotherapy. An abdominal CT scan revealed innumerable miliary nodules in bilateral adnexal areas 2 months after surgery. Histologically, large numbers of mature glia were observed, supporting the diagnosis of GP. Conclusions We report for the first time a case of primary liver immature teratoma with GTS and GP in an adult. Longer follow-up is needed to assess definitive efficacy.

Abstract Studies have found that oxygen-rich-containing functional groups in carbon-based materials can be used as active sites for the storage performance of K+, but the basic storage mechanism is still unclear. Herein, we construct and optimize 3D honeycomb-like carbon grafted with plentiful COOH/C = O functional groups (OFGC) as anodes for potassium ion batteries. The OFGC electrode with steady structure and rich functional groups can effectively contribute to the capacity enhancement and the formation of stable solid electrolyte interphase (SEI) film, achieving a high reversible capacity of 230 mAh g−1 at 3000 mA g−1 after 10,000 cycles (almost no capacity decay) and an ultra-long cycle time over 18 months at 100 mA g−1. The study results revealed the reversible storage mechanism between K+ and COOH/C = O functional groups by forming C-O-K compounds. Meanwhile, the in situ electrochemical impedance spectroscopy proved the highly reversible and rapid de/intercalation kinetics of K+ in the OFGC electrode, and the growth process of SEI films. In particular, the full cells assembled by Prussian blue cathode exhibit a high energy density of 113 Wh kg−1 after 800 cycles (calculated by the total mass of anode and cathode), and get the light-emitting diodes lamp and ear thermometer running.

Calcineurin (CN) controls the immune response by regulating nuclear factor of activated T cells (NFAT). Inhibition of CN function is an effective treatment for immune diseases. The PVIVIT peptide is an artificial peptide based on the NFAT-PxIxIT motif, which exhibits stronger binding to CN. A bioactive peptide (named pep4) that inhibits the CN/NFAT interaction was designed. Pep4 contains a segment of A238L as the linker and the LxVP motif and PVIVIT motif as CN binding sites. Pep4 has strong binding capacity to CN and inhibits CN activity competitively. 11-arginine-modified pep4 (11 R-pep4) inhibits the nuclear translocation of NFAT and reduces the expression of IL-2. 11 R-pep4 improves the pathological characteristics of asthmatic mice to a certain extent. The above results indicated that pep4 is a high-affinity CN inhibitor. These findings will contribute to the discovery of new CN inhibitors and promising immunosuppressive drugs.

Abstract Straw incorporation is an effective measure for increasing soil organic carbon (SOC) thereby improving soil quality and crop productivity. However, quantitative assessments of the transformation and distribution of exogenous carbon (C) in soil aggregates under various field fertilization practices have been lacking. In this study, we collected topsoil samples (0–20 cm) from three fertilization treatments (no fertilization control, CK; inorganic fertilizer, IF; inorganic fertilizer plus manure, IFM) at a 29-year long-term Mollisol experiment in Northeast China. We then mixed the soil samples with 13C-labeled maize straw (δ13C = 246.9‰), referred as CKS, IFS, and IFMS, and incubated them in-situ for 360 days. Initial and incubated soil samples were separated into four aggregate fractions (> 2, 1–2, 0.25–1, and 2 mm aggregates (2.2–5.8 g kg−1), 1–2 mm aggregates (2.4–4.6 g kg−1), and 2, 1–2, 0.25–1, and 2, 1–2, 0.25–1, and 2 mm aggregates (9.4–16%). Since the rapid spread of COVID-19, our hospital has actively implemented the idea of “preventing the coronavirus from entering.” We paid close attention to details, rapidly carried out personnel and material management at all levels, and strived to do a good job in the prevention and control of the epidemic, so as to achieve zero infection. Patients' carers are an important part of quality care. Lacking of protection knowledge of infectious diseases would increase the risk of nosocomial infection. Therefore, our nursing department set up a team of epidemic prevention and control management. The team made the revision on contingency plans and care management regulations during the outbreak of COVID-19, strengthened epidemic prevention and control knowledge training, carried out the supervision inspection work, and paid attention to humanized management measures to provide an important guarantee for victory over the COVID-19.

In this paper, we mainly study the best approximation theory in Riesz space, which is not constructed by the norm, but only rely on the order structure. Based on the order structure, we propose the concept of the order best approximation in Riesz space and discuss some problems related to the order best approximation, including some sufficient and necessary conditions for satisfying the order best approximation set. Finally, we consider the order best approximation projection and its related properties.

Silicon is considered as one of the ideal anode materials for the new generation of lithium-ion batteries due to its extremely high theoretical specific capacity. Nevertheless, in the actual charging and discharging process, the Si electrode will lose its electrochemical performance due to the huge volume change of Si nanoparticles resulting in detachment from the surface of the fluid collector. The polymer binder can bond the Si nanoparticles together in a three-dimensional cross-linking network, which can thus effectively prevent the Si nanoparticles from falling off the surface of the fluid collector due to the drastic change of volume during the charging and discharging process. Therefore, this study developed a new polymer binder based on electrostatic interaction with hyperbranched polyethylenimine (HPEI) as the main body and water-soluble carboxylated polyethylene glycol (CPEG) as the cross-linker, where the degree of cross-linking can be easily optimized by adjusting the pH value. The results demonstrate that, when the density of positive and negative charges in the binder is relatively balanced at pH 7, the stability of the battery’s charge–discharge cycle is significantly improved. After 200 cycles of constant current charge–discharge test, the specific capacity retention rate is 63.3%.

BackgroundTumor-infiltrating lymphocytes (TILs) are considered a manifestation of the host immune response against cancer and tertiary lymphoid structures (TLS) may contribute to lymphocytes recruitment. Both of them have been reported as potential prognostic parameters in some human malignancies. However, the roles of TILs, TLS, and their correlation in Epstein-Barr Virus-associated gastric carcinoma (EBVaGC) and EBV-negative gastric carcinoma (EBVnGC) are largely unknown.MethodsTo observe the correlation among TILs, TLS, and clinicopathological characteristics and their prognostic significance in EBVaGC and EBVnGC, respectively. TILs and TLS were assessed by morphology and/or immunohistochemistry, and accompanied by clinicopathological analysis from 846 gastric cancer patients in multiple institutions.ResultsForty-two (5.0%) cases of EBVaGC and 804 cases of EBVnGC were identified by in situ hybridization, respectively. For EBVnGC, higher TILs grade was correlated with TLS-present. EBVnGC patients with high TILs grade and TLS-present exhibited survival benefits. TILs (P = 0.001) and TLS (P = 0.003), especially TILs & TLS (P < 0.001) were independent prognostic factors in EBVnGC. A nomogram was constructed and validated for predicting the probability of overall survival and performed well with a good calibration. No significant prognostic value was detected in EBVaGC.ConclusionTILs and TLS, especially TILs & TLS were promising prognostic indicators for overall survival in EBVnGC. TILs and TLS were highly overlapping in their extent and prognostic abilities, and may be considered as a coindicator of prognosis of gastric cancer. The evaluations of TILs and TLS are simple and can be assessed routinely in pathological diagnosis.

Stereotactic biopsies for lesions in the brainstem and deep brain are rare. This study aimed to summarize our 6-year experience in the accurate diagnosis of lesions in the brain stem and deep brain and to discuss the technical note and strategies. From December 2011 to January 2018, 72 cases of intracranial lesions in the brainstem or deep in the lobes undergoing stereotactic biopsy were retrospectively reviewed. An individualized puncture path was designed based on the lesion's location and the image characteristics. The most common biopsy targets were deep in the lobes (43 cases, 59.7%), including frontal lobe (33 cases, 45.8%), temporal lobe (4 cases, 5.6%), parietal lobe (3 cases, 4.2%), and occipital lobe (3 cases, 4.2 %). There were 12 cases (16.7%) of the brainstem, including 8 cases (11.1%) of midbrain, and 4 cases (5.6%) of pons or brachium pontis. Other targets included internal capsule (2 cases, 2.8%), thalamus (3 cases, 4.2%), and basal ganglion (12 cases, 16.7%). As for complications, one patient developed acute intracerebral hemorrhage in the biopsy area at 2 h post-operation, and one patient had delayed intracerebral hemorrhage at 7 days post-operation. The remaining patients recovered well after surgery. There was no surgery-related death. The CT-MRI-guided stereotactic biopsy of lesions in the brainstem or deep in the brain has the advantages of high safety, accurate diagnosis, and low incidence of complications. It plays a crucial role in the diagnosis of atypical, microscopic, diffuse, multiple, and refractory lesions.

Renal cell carcinoma (RCC) accounts for 2–3% of adult malignant tumors, and the incidence of RCC worldwide has increased by about 2% over the past two decades. The homeobox protein Six2 has been shown to promote the stemness of breast cancer cells and play a role in kidney development, but its involvement in RCC progression has not previously been investigated. Here, we found that six2 expression was significantly increased in RCC tissues and negatively correlated with the overall survival of patients with RCC. In addition, six2 expression exhibited a remarkably higher level relative to that in normal renal cells. Functional experiments showed that six2 knockdown attenuated the stemness of RCC cells, which was evident by decreased spheroid formation ability and stemness marker (sox2 and nanog) expression. Mechanistic studies indicated that Six2 directly bound to the enhancer of sox2, promoting sox2 expression and downstream effector expression of nanog. Furthermore, overexpression of sox2 rescued the inhibitory effects of six2 on the stemness of RCC cells. Notably, six2 expression is positively correlated with sox2 and nanog expression in RCC tissues. Collectively, our results point toward a six2/sox2 axis responsible for RCC cell stemness.

Currently, there are more Global Navigation Satellite System (GNSS) signals available for civilians. Many types of GNSS receivers have been updated and several new receivers have been developed for new signals. To know about the performance of these signals and receivers and their stochastic model for data processing, in this study, the data quality of all GNSS signals, especially the new signals are analyzed, and two modified stochastic models with observation noise statistics (STA) and post-fit residuals (RES) are formed. The results show that for all the new signals, the corresponding carrier phase noise is at the same level as other old signals. The pseudorange noise of B2a, L5, E5a, and E5b is within 4 cm and significantly smaller than other signals for receivers without a smooth algorithm, and the multipath error of these signals is about 0.1 m which is also better than other signals. For B1C, the pseudorange multipath error is about 0.4 m, which is close to L1 and E1. Stochastic models are validated for precise orbit determination (POD). Compared with the empirical stochastic model (EMP), both modified models are helpful to reduce the mean unit weight square error and obtain high accuracy orbits with reduced iteration. The 3D orbit accuracy improvement can reach 0.27 cm (7%) for the STA model, and 0.40 cm (10%) for the RES model when compared with the final products from the international GNSS service (IGS). For BDS-3 POD by using B1C and B2a observations, the improvements in the 3D orbit consistency of two adjacent three-day solutions are 0.21 cm (3%) for the STA model and 0.29 cm (4%) for the RES model. In addition, the STA model based on the observation noise of globally distributed stations is less affected by stations with problematic observations and with reduced computation burden.

Hepatic metastases were reported in up to 70% of colorectal cancer patients, among which multifocal hepatic metastasis represents one of the complications that lead to poor prognosis. The majority of the patients carrying multifocal hepatic metastases required pharmaceutical treatments to reduce the tumor size prior to surgical resection. However, the clinical responses to pharmaceutical agents were difficult to predict due to the heterogeneous nature of the multifocal tumors. Here, we report a case with multifocal hepatic metastases from colorectal cancer that was resistant to the primary chemotherapy and Bevacizumab plus chemotherapy, but responded to the combined therapy of Cetuximab and FOLFOX. Genetic tests had revealed that the tumor was highly metastatic due to the mutations of the WNT signaling pathway, and the metastatic tumors might be sensitive to Cetuximab. Consistent with the molecular characterizations, the metastatic tumors continue to emerge after chemotherapy, and rapidly relapsed in great numbers after liver resection. However, the combined therapy of Cetuximab and FOLFOX guided by the genetic tests significantly reduced the size and number of metastatic tumors. To conclude, deciphering the mutation profiles of multifocal metastatic tumors may guide the determination of treatment tactics, which may benefit the patients with non-resectable advanced carcinoma.

Severe mycological epilepsy of infancy is a catastrophic disease with preferential dysfunction of interneurons, frequent episode rate, cognitive and sudden death. The disease is mainly caused by heterozygous loss-of-function mutation of SCN1A gene encoding α subunit of the sodium channel Nav1.1. To generate mutations in normal iPSC, Transcription activator-like effector nucleases was used to introduce the epilepsy-causing mutation A5768G into the endogenous locus of SCN1A gene. The gene editing induced pluripotent stem cell line and normal iPSC were obtained from the same donor to eliminate significantly the genetic background noise.

Dravet syndrome is an epileptic encephalopathy largely due to haploinsufficiency of the voltage-gated sodium channel Nav1.1 that is expressed primarily in GABAergic neurons. In order to distinguish the different subtypes, we used gene editing to introduce tdTomato gene into the genome of iPSCs to label the GABAergic neurons in the differentiated neuronal networks. The gene-edited cell line demonstrates normal karyotype, expresses the main pluripotency markers, and shows the presence of differentiation into the three embryonic germ layers in teratomas.

Background: Previous infectious or inflammatory events may be involved in the pathogenesis of neuromyelitis optica (NMO), potentially by triggering an autoimmune response. Cytomegalovirus (CMV)-related NMO (CMV-NMO) is rarely reported. Acute hemorrhagic rectal ulcer (AHRU) is a rare disease with a largely unknown pathogenesis. Herein, we reported a co-NMO and AHRU case associated with CMV infection. In addition, we review previously reported cases of CMV-NMO and CMV-AHRU.Case presentation: A 40-year-old female diagnosed with aquaporin4 (AQP4)-IgG+ NMO and a poor response to high-dose intravenous methylprednisolone and immunoglobulin, followed by three rounds of plasma exchange was transferred to Third Affiliated Hospital of Sun Yat-sen University, Guangzhou, China. She developed repeated acute lower gastrointestinal hemorrhage from the third day of admission. Abdominal computed tomography angiography (CTA) and interventional angiography did not detect any bleeding vessel. Bedside colonoscopy revealed a large ulcer-like lesion at 10 cm above the anus. Rectal biopsy pathology confirmed a CMV infection on day 23 post-admission, and cerebrospinal fluid (CSF) pathogen gene sequencing detected CMV gene copies on day 25 post-admission. After 2 weeks of treatment with ganciclovir and sodium phosphinate, the patient's lower gastrointestinal bleeding stopped, and her limb muscle strength and visual acuity gradually improved. After 4 weeks of antiviral therapy, colonoscopy showed that the intestinal wall of the original lesion was smooth. Hematoxylin and eosin (HE) staining and immunohistochemistry (IHC) of a biopsy specimen was negative for CMV, her right eye vision was normal, and limb muscle strength had recovered. Serum AQP4-IgG was negative, and lesions on brain magnetic resonance imaging (MRI) manifested shrinkage.Conclusions: The benefits of antiviral therapy remain unclear; however, clinicians should be aware of the possibility of CMV-related NMO, if NMO was refractory to high-dose intravenous methylprednisolone, immunoglobulin, and plasma exchange. Moreover, clinicians should consider the possibility of CMV-related AHRU when recurrent acute lower gastrointestinal bleeding occurs in a patient.

ObjectiveRegional dissemination is the major cause of the widespread prevalence of a plasmid-encoding NDM-1 enzyme. We investigated the drug resistance, joint efficiency, and gene environment of a Klebsiella pneumoniae strain carrying blaNDM–1 gene.Materials and MethodsCarbapenem-non-susceptible strains were analyzed using the VITEK 2 Compact. Strains carrying blaNDM–1 were identified using polymerase chain reaction and sequencing. Antimicrobial susceptibility testing and plasmid conjugation experiments were then conducted. Strains carrying blaNDM–1 were subjected to Southern blot analysis. After the gene mapping of blaNDM–1, library construction, and sequencing, plasmids were subsequently spliced and genotyped using the software Glimmer 3.0, and then analyzed using Mauve software.ResultsAmong 1735 carbapenem-non-susceptible strains, 54 strains of blaNDM–1-positive bacteria were identified, which consisted of 44 strains of K. pneumoniae, 8 strains of Acinetobacter baumannii and 2 strains of Escherichia coli. Strains carrying blaNDM–1 had a resistance rate of more than 50% in most antibiotics. Plasmid conjugation between strains carrying blaNDM–1 and E. coli strain J53 had a success rate of 50%. Southern blot analysis indicated that each strain had multiple plasmids containing blaNDM–1. Among the five plasmids containing blaNDM–1 in K. pneumoniae for sequencing, two plasmids with complete sequences were obtained. The findings were as follows: (i) The p11106 and p12 plasmids were highly similar to pNDM-BTR; (ii) the p11106 and p12 plasmids showed differences in the 20–30 kb region (orf00032–orf00043) from the other six plasmids; and (iii) blaNDM–1 was located at orf00037, while ble was found at orf00038. Two tnpA genes were located in the upstream region, and orf00052 (tnpA) in the 36 kb region was in the downstream sequence.ConclusionblaNDM–1-containing bacteria exhibit multidrug resistance, which rapidly spreads and is transferred through efficient plasmid conjugation; the multidrug resistance of these bacteria may be determined by analyzing their drug-resistant plasmids. The presence of ble and tnpA genes suggests a possible hypothesis that blaNDM–1 originates from A. baumannii, which is retained in K. pneumoniae over a long period by transposition of mobile elements.

A density-based spatial clustering of applications with noise (DBSCAN) and three distances (TD) integrated Wi-Fi positioning algorithm was proposed, aiming to enhance the positioning accuracy and stability of fingerprinting by the dynamic selection of signal-domain distance to obtain reliable nearest reference points (RPs). Two stages were included in this algorithm. One was the offline stage, where the offline fingerprint database was constructed and the other was the online positioning stage. Three distances (Euclidean distance, Manhattan distance, and cosine distance), DBSCAN, and high-resolution distance selection principle were combined to obtain more reliable nearest RPs and optimal signal-domain distance in the online stage. Fused distance, the fusion of position-domain and signal-domain distances, was applied for DBSCAN to generate the clustering results, considering both the spatial structure and signal strength of RPs. Based on the principle that the higher resolution the distance, the more clusters will be obtained, the high-resolution distance was used to compute positioning results. The weighted K-nearest neighbor (WKNN) considering signal-domain distance selection was used to estimate positions. Two scenarios were selected as test areas; a complex-layout room (Scenario A) for post-graduates and a typical large indoor environment (Scenario B) covering 3200 m2. In both Scenarios A and B, compared with support vector machine (SVM), Gaussian process regression (GPR) and rank algorithms, the improvement rates of positioning accuracy and stability of the proposed algorithm were up to 60.44 and 60.93%, respectively. Experimental results show that the proposed algorithm has a better positioning performance in complex and large indoor environments.

The Klobuchar model has been widely used to correct the ionospheric delay in applications. However, the NVTEC (Nighttime Vertical Total Electron Content) of the Klobuchar model employs an empirical constant of 9 TECU (Total Electron Content Unit) at L1 frequency. In this paper, the rationality and reliability of the nighttime constant setting are investigated using the GIM (Global Ionosphere Map) product of the IGS (International GNSS Service) from 1998 to 2015. Our study indicates that the suitable time span of NVTEC average in nighttime should be between 20:00 and 06:00 LT (local time). The NVTEC is highly correlated with seasons, having positive extremes in spring and autumn and negative extremes in summer through the mean values in all latitudes. In addition to seasonal dependence, solar activity in the solar cycle 23 strongly influences NVTEC as well and leads to its variation within a range between 25 and 30 TECU in spring and autumn at solar maximum, which is about 1.5 times greater than that in summer and winter. The NVTEC also has a dependence on the latitude at solar maximum, with the mean value from 30 TECU in low latitudinal regions to 15 TECU in high latitudinal regions. Therefore, these results demonstrate that the nighttime VTEC has much greater deviations from the imperial constant in the Klobuchar model, and the newly estimated constant is expected to bring improvement to the predictability of the Klobuchar ionospheric delay model in nighttime. Abstract Background Splenic hamartoma is a rare benign vascular proliferative lesion composed of unorganized sinusoid-like channels lined with plump or flat endothelial cells and characterized by a CD8-positive immunophenotype of the lining cells. Scattered bizarre stromal cells can be found in some splenic hamartomas. The presence of splenic hamartoma with bizarre stromal cells is extremely rare and these bizarre cells make it possible to be regarded as a malignancy. Recognition of this rare histologic variant will help to avoid diagnostic confusion and overtreatment of this benign entity. Case presentation We report a case of a 40-year-old man with occasional left-sided waist back pain. A splenic space-occupying lesion was detected by ultrasound and magnetic resonance imaging. Microscopically bizarre large cells were scattered throughout the splenic hamartoma. The cells exhibited atypical nuclei, scarcely visible cytoplasm, and vesicular chromatin, and they did not form expansile clusters and lacked mitotic activity. An immunohistochemical panel was performed. The bizarre cells strongly expressed vimentin, and the Ki-67 index was very low. The lesion was diagnosed as a splenic hamartoma with bizarre stromal cells. Conclusions To the best of our knowledge, this is the first systematic review on a splenic hamartoma with bizarre stromal cells; only six cases have been described in the literature. Proper identification is important to secure adequate treatment.

Objective: To study the effects of regenerated tissue extracts after liver injury on the proliferation, differentiation, migration and invasion of SK-HEP1 cells. Methods: Regenerated tissue extracts after liver injury were used to induce SK-HEP1 cells after enrichment, their effects on the proliferation, differentiation, migration and invasion of SK-HEP1 cells were observed through in vitro cell culture, MTT, flow cytometry and transwell assays. Results: In response to the action of regenerated tissue extracts after liver injury, SK-HEP1 cells were blocked in G0/G1 phase, their growth rate was distinctly reduced. The number of SK-HEP1- fj colonies decreased. The migration ability of SK-HEP1 cells showed a decreased trend on day 7 and day 11 after induction. SK-HEPl’s invasion ability clearly decreased on days 7 and 11 after induction, especially on day 7. Conclusions: To a certain extent, regenerated tissue extracts after liver injury can inhibit the proliferation, differentiation, migration and invasion of hepatoma cells, showing an important potential of being a differentiating agent for the treatment of liver cancer.

The ionosphere is a significant component of the geospace environment. Storm-induced ionospheric anomalies severely affect the performance of Global Navigation Satellite System (GNSS) Positioning, Navigation, and Timing (PNT) and human space activities, e.g., the Earth observation, deep space exploration, and space weather monitoring and prediction. In this study, we present and discuss the multi-scale ionospheric anomalies monitoring over China using the GNSS observations from the Crustal Movement Observation Network of China (CMONOC) during the 2015 St. Patrick’s Day storm. Total Electron Content (TEC), Ionospheric Electron Density (IED), and the ionospheric disturbance index are used to monitor the storm-induced ionospheric anomalies. This study finally reveals the occurrence of the large-scale ionospheric storms and small-scale ionospheric scintillation during the storm. The results show that this magnetic storm was accompanied by a positive phase and a negative phase ionospheric storm. At the beginning of the main phase of the magnetic storm, both TEC and IED were significantly enhanced. There was long-duration depletion in the topside ionospheric TEC during the recovery phase of the storm. This study also reveals the response and variations in regional ionosphere scintillation. The Rate of the TEC Index (ROTI) was exploited to investigate the ionospheric scintillation and compared with the temporal dynamics of vertical TEC. The analysis of the ROTI proved these storm-induced TEC depletions, which suppressed the occurrence of the ionospheric scintillation. To improve the spatial resolution for ionospheric anomalies monitoring, the regional Three-Dimensional (3D) ionospheric model is reconstructed by the Computerized Ionospheric Tomography (CIT) technique. The spatial-temporal dynamics of ionospheric anomalies during the severe geomagnetic storm was reflected in detail. The IED varied with latitude and altitude dramatically; the maximum IED decreased, and the area where IEDs were maximum moved southward.

BeiDou global navigation satellite system (BDS) began to provide positioning, navigation, and timing (PNT) services to global users officially on 31 July, 2020. BDS constellations consist of regional (BDS-2) and global navigation satellites (BDS-3). Due to the difference of modulations and characteristics for the BDS-2 and BDS-3 default civil service signals (B1I/B3I) and the increase of new signals (B1C/B2a) for BDS-3, a systemically bias exists in the receiver-end when receiving and processing BDS-2 and BDS-3 signals, which leads to the inter-system bias (ISB) between BDS-2 and BDS-3 on the receiver side. To fully utilize BDS, the BDS-2 and BDS-3 combined precise time and frequency transfer are investigated considering the effect of the ISB. Four kinds of ISB stochastic models are presented, which are ignoring ISB (ISBNO), estimating ISB as random constant (ISBCV), random walk process (ISBRW), and white noise process (ISBWN). The results demonstrate that the datum of receiver clock offsets can be unified and the ISB deduced datum confusion can be avoided by estimating the ISB. The ISBCV and ISBRW models are superior to ISBWN. For the BDS-2 and BDS-3 combined precise time and frequency transfer using ISBNO, ISBCV, ISBRW, and ISBWN, the stability of clock differences of old signals can be enhanced by 20.18%, 23.89%, 23.96%, and 11.46% over BDS-2-only, respectively. For new signals, the enhancements are −50.77%, 20.22%, 17.53%, and −3.69%, respectively. Moreover, ISBCV and ISBRW models have the better frequency transfer stability. Consequently, we recommended the optimal ISBCV or suboptimal ISBRW model for BDS-2 and BDS-3 combined precise time and frequency transfer when processing the old as well as the new signals.

For the global ordinary users, the broadcast ephemeris plays important roles in positioning, navigation and timing (PNT) services. With the construction of a new generation of the BeiDou navigation satellite system (BDS), the development of BDS has entered the era of globalization. It is meaningful for global users to analyze and assess the BDS-2 and BDS-3 broadcast ephemeris. Therefore, the satellite orbits and clock offsets calculated by broadcast ephemeris are compared with the precise orbit and clock offset products provided by three analysis centers (i.e., Helmholtz Centre Potsdam German Research Center for Geosciences (GFZ), Wuhan University (WHU) and Shanghai Astronomical Observatory (SHA)), and the corresponding signal-in-space range error (SISRE) and the orbit-only SISRE are analyzed to assess the accuracy of BDS broadcast ephemeris. Due to the upgrade of BDS-3 satellite hardware technology and inter-satellite links payload and the development of satellite orbit determination algorithm, the accuracy of broadcast orbit and clock offsets has been greatly improved. The root mean square (RMS) of BDS-3 broadcast orbit errors is improved by 86.30%, 89.47% and 76.86%, and the standard deviation (STD) is improved by 79.41%, 77.00% and 76.78% compared with BDS-2 in the radial, along-track and cross-track directions. The corresponding RMS and STD of all BDS-3 satellite clock offsets are improved by 40.34% and 52.49% than that of BDS-2, respectively. Meanwhile, the mean RMS and STD are 1.78 m and 0.40 m for BDS-2 SISRE, 1.72 m and 0.34 m for BDS-2 orbit-only SISRE, 0.50 m and 0.14 m for BDS-3 SISRE, and 0.17 m and 0.04 m for BDS-3 orbit-only SISRE. It is noteworthy that the average broadcast-minus-precise (BMP) clock values of BDS-2 and BDS-3 are inconsistent, which can indirectly prove that the datum of broadcast clock offsets for BDS-2 and BDS-3 are inconsistent. The inconsistency of the datum of satellite clock offsets and receiver hardware delay bias between BDS-2 and BDS-3 will result in the inter-system bias (ISB) on the receiver segment. For JAVAD TRE_3 receivers, the ISB is relatively small and thus can be ignored. However, for the TRIMBLE ALLOY, SEPT POLARX5, CETC-54-GMR-4016, CETC-54-GMR-4011, GNSS-GGR and UB4B0-13478 receivers, estimating ISB can improve the positioning accuracy of single point positioning (SPP) by 20.15%, 19.81% and 12.76% in north, east and up directions, respectively.

BACKGROUND:Clonorchiasis, caused by the liver fluke Clonorchis sinensis, remains a serious public health issue in Asia, especially in China, and its relationship with cholangiocarcinoma has highlighted the importance of C. sinensis infection. Proteins containing tandem repeats (TRs) are found in a variety of parasites and, as targets of B-cell responses, are valuable for the serodiagnosis of parasite infections. Here, we identified a novel C. sinensis-specific antigen, Cs1, containing TRs, and investigated its diagnostic value, other immunological properties, and tissue distribution. METHODOLOGY/PRINCIPAL FINDINGS:A partial Cs1 cDNA sequence was cloned by screening an adult C. sinensis cDNA expression library. The full-length Cs1 cDNA was obtained by 5' rapid amplification of cDNA ends. The deduced Cs1 protein consists of a signal peptide and five TRs of 21 amino acids. The recombinant Cs1 (rCs1) was constructed and purified. rCs1 showed higher sensitivity (94.3%) and specificity (94.4%) than the C. sinensis excretory-secretory products (ESPs) according to ELISA of 114 serum samples. Native Cs1 was identified in C. sinensis ESPs and crude antigens of adult C. sinensis by western blotting using an anti-rCs1 monoclonal antibody. ELISA of recombinant peptides of different Cs1 regions demonstrated that the TR region was immunodominant in Cs1. Immunohistochemistry and confocal microscopy revealed that Cs1 is located in a granule-like structure surrounding the acetabulum of C. sinensis adults that has not previously been described. CONCLUSIONS/SIGNIFICANCE:We identified a novel C. sinensis-specific TR protein, Cs1, which is an antigen of high serological significance, compared with C. sinensis ESPs. The deduced features of Cs1 show a unique structure containing TRs and a signal peptide and the TR region is immunodominant in Cs1. This provides a basis for targeted screens of other antigens. The novel structure in which Cs1 is located also deserves further investigation.

Mitochondria synthesize select phospholipids but lack the machinery for synthesis of the most abundant mitochondrial phospholipid, phosphatidylcholine (PC). Although the phospholipid transfer protein Stard7 promotes uptake of PC by mitochondria, the importance of this pathway for mitochondrial and cellular homeostasis represents a significant knowledge gap. Haploinsufficiency for Stard7 is associated with significant exacerbation of allergic airway disease in mice, including an increase in epithelial barrier permeability. To test the hypothesis that Stard7 deficiency leads to altered barrier structure/function downstream of mitochondrial dysfunction, Stard7 expression was knocked down in a bronchiolar epithelial cell line (BEAS-2B) and specifically deleted in lung epithelial cells of mice (Stard7epi∆/∆). Stard7 deficiency was associated with altered mitochondrial size and membrane organization both in vitro and in vivo. Altered mitochondrial structure was accompanied by disruption of mitochondrial homeostasis, including decreased aerobic respiration, increased oxidant stress, and mitochondrial DNA damage that, in turn, was linked to altered barrier integrity and function. Both mitochondrial and barrier defects were largely corrected by targeting Stard7 to mitochondria or treating epithelial cells with a mitochondrial-targeted antioxidant. These studies suggest that Stard7-mediated transfer of PC is crucial for mitochondrial homeostasis and that mitochondrial dysfunction contributes to altered barrier permeability in Stard7-deficient mice.

Myocardial ischemia/reperfusion (IR) injury is a significant annual clinical threat. Reperfusion injury exacerbates this issue by causing an excessive overload of reactive oxygen species (ROS) due to heme degradation and dysfunction in mitochondrial complexes. Subsequently, this surge in ROS leads to mitochondrial damage, further compromising cardiomyocyte mitochondrial function and eventually causing cell death. Given its pivotal role, targeting ROS has emerged as a viable strategy for treating myocardial ischemic injury. One promising therapeutic intervention involves the use of nanomaterials to eliminate ROS. In this study, we synthesized a polyvinylpyrrolidone-modified ultrathin niobium carbide (Nb2C) MXene, which effectively alleviated myocardial IR injury by scavenging ROS and enhancing mitochondrial function. Nb2C demonstrated multiple enzymatic-like abilities, including catalase and superoxide dismutase, which offered effective protection against myocardial ischemic injury by neutralizing superoxide anions and hydrogen peroxide. Furthermore, in the IR mice treated with Nb2C, mitochondrial injury was significantly reduced, and mitochondrial function was notably improved. Intravenous administration of Nb2C resulted in substantial recovery of cardiac function in IR mice and a marked reduction in the infarct area. In this paper, a novel of convergence named the fuzzy u-uniform convergence is defined on the basis of the fuzzy order convergence in fuzzy Riesz spaces. The relation between the fuzzy u-uniform convergence and the fuzzy order convergence in fuzzy Archimedean Riesz spaces is studied. We also discuss the existence of the fuzzy u-uniform limit for the fuzzy u-uniform Cauchy sequence in fuzzy Archimedean Riesz Spaces. Potassium-ion batteries (PIBs) have been widely studied owing to the abundant reserves, widespread distribution, and easy extraction of potassium (K) resources. Molybdenum disulfide (MoS2) has received a great deal of attention as a key anode material for PIBs owing to its two-dimensional diffusion channels for K+ ions. However, due to its poor electronic conductivity and the huge influence of embedded K+ ions (with a large ionic radius of 3.6 Å) on MoS2 layer, MoS2 anodes exhibit a poor rate performance and easily collapsed structure. To address these issues, the common strategies are enlarging the interlayer spacing to reduce the mechanical strain and increasing the electronic conductivity by adding conductive agents. However, simultaneous implementation of the above strategies by simple methods is currently still a challenge. Herein, MoS2 anodes on reduced graphene oxide (MoS2/rGO) composite were prepared using one-step hydrothermal methods. Owing to the presence of rGO in the synthesis process, MoS2 possesses a unique scaled structure with large layer spacing, and the intrinsic conductivity of MoS2 is proved. As a result, MoS2/rGO composite anodes exhibit a larger rate performance and better cycle stability than that of anodes based on pure MoS2, and the direct mixtures of MoS2 and graphene oxide (MoS2-GO). This work suggests that the composite material of MoS2/rGO has infinite possibilities as a high-quality anode material for PIBs. [ABSTRACT FROM AUTHOR]

The electron transport behaviors through a series of molecular junctions, which consist of polyacene molecule bridging armchair CNT(5,5) electrodes, have been investigated using density functional theory combined with the non-equilibrium Green's function approach. The electronic signature is strongly impeded by the edge sites at nonvertical edges of CNT(5,5), such as the SWNT-4CH junction. This is due to the fact that the presence of edge electron injection can significantly hinder mainstream electron transport through destructive quantum interference, thereby reducing the current substantially. In contrast, when the edge sites at interfaces between molecules and electrodes are removed, edge electron injection is blocked, and an observable improvement in junction current is achieved. Substituting carbon atoms at the edge sites with B and N heteroatoms results in a nonlinear behavior for SWNT-4BH and SWNT-4NH with an "inverse Schottky" feature. This confirms that introducing B and N heteroatoms at edge sites can affect part of edge electron injection, reduce destructive quantum interference effects, and enhance junction conductivity. Based on these studies, a series of models were designed to investigate rectification characteristics. The insights gained from this study on interfaces could contribute to a better understanding of all-carbon devices for designing efficient nanoelectronic devices. [ABSTRACT FROM AUTHOR]
