Abstract Despite the identification of numerous genetic variants affecting SBP in European populations, their applicability in admixed populations remains unclear. This study evaluates the predictive efficacy of a systolic blood pressure (SBP) polygenic risk score (PRS), derived from the UK Biobank data, in two Brazilian cohorts. We analyzed 944 K genetic variants consistent across an independent UK Biobank dataset, Brazilian cohorts, and HapMap database. Results show a significant association between increased PRS and SBP, as well as hypertension, in each study groups analyzed. An increase of one standard deviation in the PRS showed a significant association with SBP (β [95% CI] (mmHg) = 5.2 [5.1–5.3], 2.8 [2.1–3.5] and 2.6 [2.2–3.0]) and hypertension (odds ratio (OR) [95% CI] = 1.56 [1.54–1.56], 1.28 [1.2–1.4] and 1.47 [1.3–1.6]) in an independent UKB dataset, Baependi, and Pelotas, respectively. The associations were weaker in the Brazilian samples and the reduced association was noticeable in the Pelotas vs. the UK comparison for hypertension stages 1 and 2 (OR [95% CI] = 2.1 [1.5–3.1] and 3.0 [1.9–4.7] vs. 2.5 [2.2–2.8] and 4.9 [4.4–5.6]), whereas the Baependi data showed no significance for stage 1 hypertension. This trend mirrors findings in homogeneous African and Asian populations with diverse genetic architecture, highlighting the limitations of European-based PRS also in admixed populations. These insights are crucial for developing tailored disease prevention and management strategies in ethnically diverse groups.

Meteorological parameters strongly affect atmospheric dispersion which makes developing a simulation to get a prediction of the odour impact significant to understand the effect of the same sources located in different sites. Each location has peculiarities depending on orography, distance from the sea and mainly wind speed and direction. This study evaluates the impact of two ideal sources, one areal on the ground and one point source at altitude, having the same odour flow.Generally, odour dispersion is affected by wind speed near the emission, so higher concentration are expected from the ground based source. The findings consistently demonstrate that this source yields the most significant odour impact, while wind speed at altitude primarily influences dispersion from the point source. Despite the areal ground-level source has a greater impact at all locations, the ratio of the mean of the two sources mean simulated impact has a linear dependence with the distance from the emission point.

La neurocisticercosis (NCC) es la enfermedad parasitaria más frecuente del sistema nervioso central causada por la forma larvaria enquistada de la Taenia solium, su diagnóstico se basa en la integración de datos clínicos, imagenológicos y serológicos. La detección de anticuerpos específicos usando el ensayo inmunoenzimático (ELISA) es una útil estrategia para confirmar el diagnóstico de la NCC. Estudios previos demuestran que la Taenia crassiceps y la Taenia solium tienen similitud estructural y antigénica; en este trabajo evaluamos antígenos de ambos parásitos frente a 68 líquidos cefalorraquídeos y 40 sueros pertenecientes a casos confirmados de NCC y a controles. Para cumplir tal objetivo enfrentamos en ELISA las muestras de los pacientes contra cada uno de tres extractos antigénicos de cada tenideo: el fluido vesicular, la membrana externa y el extracto total. El análisis estadístico demostró una alta sensibilidad y especificidad de los antígenos de la T crassiceps, especialmente el fluido vesicular; el cual dio una sensibilidad de 90,3%, una especificidad de 94,5% y valores predictivos positivos y negativos de 93,3% y 92,1% frente a los LCR. Estos atributos hacen de esta fracción parasitaria una herramienta de gran utilidad en el diagnóstico de la NCC, pudiendo sustituir los antígenos procedentes de carcasas de cerdos infectados naturalmente con T. solium.

Abstract The COVID-19 pandemic has had a devastating impact on the health and wellbeing of the global population. This paper presents the results of a longitudinal transcultural study that was begun at the peak of the pandemic (in April, 2020). An online survey was used to collect data from English-, Spanish-, and Portuguese-speaking participants. The survey collected information about sociodemographics, lifestyle activities, COVID-19-related circumstances, and drug use (with an emphasis on hallucinogenic drugs), as well as involving psychometric questionnaires. Users of hallucinogenic drugs had higher psychological well-being and lower scores on psychopathology scales, both at baseline and during follow-ups. This difference was larger when users were distinguished by frequency of use, as regular users scored higher on psychological well-being and lower on psychopathology scales. Subjects with more psychological distress had lower scores for all scales of post-traumatic growth, but if they were regular hallucinogens users, they had higher scores for post-traumatic growth. When comparing the results between cultural contexts, heterogeneous results were obtained. There were more English-speaking regular users of hallucinogenic drugs. Further research should analyse the potential role of hallucinogens in large-scale catastrophes, with a special focus on post-traumatic growth.

Abstract Cosmogenic radio-nuclei are an important source of background for low-energy neutrino experiments. In Borexino, cosmogenic $$^{11}$$ 11 C decays outnumber solar pep and CNO neutrino events by about ten to one. In order to extract the flux of these two neutrino species, a highly efficient identification of this background is mandatory. We present here the details of the most consolidated strategy, used throughout Borexino solar neutrino measurements. It hinges upon finding the space-time correlations between $$^{11}$$ 11 C decays, the preceding parent muons and the accompanying neutrons. This article describes the working principles and evaluates the performance of this Three-Fold Coincidence (TFC) technique in its two current implementations: a hard-cut and a likelihood-based approach. Both show stable performances throughout Borexino Phases II (2012–2016) and III (2016–2020) data sets, with a $$^{11}$$ 11 C tagging efficiency of $$\sim 90$$ ∼ 90 % and $$\sim $$ ∼ 63–66 % of the exposure surviving the tagging. We present also a novel technique that targets specifically $$^{11}$$ 11 C produced in high-multiplicity during major spallation events. Such $$^{11}$$ 11 C appear as a burst of events, whose space-time correlation can be exploited. Burst identification can be combined with the TFC to obtain about the same tagging efficiency of $$\sim 90\%$$ ∼ 90 % but with a higher fraction of the exposure surviving, in the range of $$\sim $$ ∼ 66–68 %.

Global warming is affecting biodiversity; however, the extent to which animal reproductive processes respond to predicted temperature increments remains largely unexplored. The thermal environment has a pronounced impact on metabolic rates of ectotherms; therefore, an interesting question to assess is whether temperature increase might affect specific reproductive mechanisms like sperm performance in ectotherms. Moreover, in many species, oviductal fluid (OF) is known to regulate and maintain sperm quality; however, the role of OF in relation to the effects of high temperature on sperm remains unclear. Our aim was to experimentally test the effect of increased temperature on sperm velocity, swimming path and percentage of motility in neutral conditions at ejaculation (without OF) and in female's reproductive tract fluid (with OF), in a social ectotherm lizard model, Tropidurus spinulosus, which has specific thermal requirements for reproduction. Our results suggest that a rising temperature associated with global warming (+4°C) affects negatively sperm dynamics and survival. However, OF ameliorated the harmful effects of high temperature. This is an important point, as this study is the first to have tested the role of OF in preserving sperm from a warmer pre-fertilization environment. These results contribute to our understanding of how thermal environment changes might affect post-copulatory reproductive mechanisms. The worldwide increase in serious infections caused by multidrug-resistant (MDR) K. pneumoniae emphasizes the urgent need of new therapeutic strategies for the control of this pathogen. There is growing interest in the use of bacteriophages (or phages) to treat K. pneumoniae infections, and newly isolated phages are needed. Here, we report the isolation and physical/biological/molecular characterization of a novel lytic phage and its efficacy in the control of MDR K. pneumoniae. The phage vB_KpnS_Uniso31, referred to hereafter as phage Kpn31, was isolated from hospital wastewater using K. pneumoniae CCCD-K001 as the host. Phage Kpn31 presents a siphovirus-like morphotype and was classified as Demerecviridae; Sugarlandvirus based on its complete genome sequence. The 113,444 bp Kpn31 genome does not encode known toxins or antimicrobial resistance genes, nor does it encode depolymerases related sequences. Phage Kpn31 showed an eclipse time of 15 min and a burst size of 9.12 PFU/host cell, allowing us to conclude it replicates well in K. pneumoniae CCCD-K001 with a latency period of 30 min. Phage Kpn31 was shown to be effective against at least six MDR K. pneumoniae clinical isolates in in vitro antibacterial activity assays. Based on its features, phage Kpn31 has potential for controlling infections caused by MDR K. pneumoniae.

Background: Recognition of emotions in facial expressions (REFE) is a key aspect of social cognition. Anxiety and mood disorders are associated with deficits in REFE, and anxiolytics and antidepressants reverse these deficits. Recent studies have shown that serotonergic hallucinogens (i.e. ayahuasca, dimethyltryptamine, psilocybin, lysergic acid diethylamide [LSD], and mescaline) have anxiolytic and antidepressant properties, but their effects on REFE are not well understood. The purpose of the study was to conduct a systematic review analyzing the effects of serotonergic hallucinogens on REFE in humans. Methods: Studies published in the PubMed, PsycINFO, and Web of Science databases until 19 October 2018 which analyzed the effects of serotonergic hallucinogens on REFE in humans were included. Results: Of the 62 studies identified, 8 studies were included. Included studies involved the administration of a single or a few doses of LSD or psilocybin, and most trials were randomized and controlled with placebo. LSD and psilocybin reduced the recognition of negative emotions in most studies and modulated amygdala activity to these stimuli, which was correlated with antidepressive effects in patients. Both drugs were well tolerated. Conclusions: Serotonergic hallucinogens reduced the recognition of negative emotions by modulating amygdala activity. Despite the small sample sizes, results suggest that serotonergic hallucinogens show promising beneficial effects on deficits in REFE.

Abstract Explaining large‐scale patterns of variation in body size has been considered a central question in ecology and evolutionary biology because several life‐history traits are directly linked to body size. For ectothermic organisms, little is known about what processes influence geographic variation in body size. Changes in body size and sexual size dimorphism (SSD) have been associated with environmental variables, particularly for Bruchinae insects, which feed exclusively on seeds during the larval stage. However, the effect of important seed traits on body size variation has rarely been investigated, and whether SSD varies substantially among populations within bruchine species is poorly known. Using the seed‐feeding beetle Acanthoscelides macrophthalmus infesting its host plant Leucaena leucocephala, we investigated whether specific seed traits (hardness, size, water content, carbon/nitrogen ratio, and phenolic content) were determinant in generating geographic variation in body size and SSD of A. macrophthalmus. We also examined the relationships between body size and SSD with latitude and altitude. The body size of both sexes combined was not related to latitude, altitude, and any of the physical and chemical seed traits. However, the female body size tended to vary more in size than the males, generating significant variation in SSD in relation to latitude and altitude. The females were the larger sex at higher latitudes and at lower altitudes, precisely where seed water content was greater. Therefore, our results suggest that water content was the most important seed trait, most severely affecting the females, promoting geographic variation in SSD of A. macrophthalmus.

Aim of this study was to investigate the activation of the IFNγ pathway in the affected liver and in the blood of patients with secondary hemophagocytic lymphohistiocytosis (sHLH). To this purpose, the mRNA expression levels of IFNG and IFNγ-inducible genes as well as Tyrosine (701)-phosphorylated signal transducer and activator of transcription 1 (STAT1) protein levels were evaluated in the liver and in peripheral blood mononuclear cells (PBMCs) of three patients with sHLH with predominant liver involvement. The mRNA expression levels of IFNG and IFNγ-inducible genes were markedly higher in patient livers compared to control livers and to one disease control liver. Conversely, slight differences in the expression levels of Type I IFN-inducible genes and other classical inflammatory cytokine genes were found. Further supporting the activation of the IFNγ pathway, higher protein levels of phosphorylated and total STAT1 were detected in patient livers compared to control livers. When the expression of the same genes analysed in liver tissues was evaluated in PBMCs collected from 2 out of 3 patients before the liver biopsy, we found that mRNA levels of IFNγ-inducible genes were markedly increased. Accordingly, high circulating levels of IFNγ-inducible CXCL9 were observed in patients. Altogether, these data demonstrate the selective and marked up-regulation of the IFNγ pathway in the liver tissue and blood of patients with active sHLH. Finally, we show that measurement of circulating CXCL9 levels and evaluation of IFNγ-inducible gene expression levels in PBMCs may represent a new valid tool to better identify patients with suspected HLH with predominant liver involvement.

The anti-inflammatory properties of sand fly saliva favor the establishment of the Leishmania infantum infection. In contrast, an antibody response against Lutzomyia longipalpis saliva is often associated with a protective cell-mediated response against canine visceral leishmaniasis. Genetic studies may demonstrate to what extent the ability to secrete anti-saliva antibodies depends on genetic or environmental factors. However, the genetic basis of canine antibody response against sand fly saliva has not been assessed. The aim of this study was to identify chromosomal regions associated with the anti-Lu. longipalpis salivary IgG response in 189 dogs resident in endemic areas in order to provide information for prophylactic strategies. Dogs were classified into five groups based on serological and parasitological diagnosis and clinical evaluation. Anti-salivary gland homogenate (SGH) IgG levels were assessed by Enzyme-Linked Immunosorbent Assay (ELISA). Genomic DNA was isolated from blood samples and genotyped using a SNP chip with 173,662 single nucleotide polymorphism (SNP) markers. The following linear regression model was fitted: IgG level = mean + origin + sex + age + use of a repellent collar, and the residuals were assumed as pseudo-phenotypes for the association test between phenotypes and genotypes (GWA). A component of variance model that takes into account polygenic and sample structure effects (EMMAX) was employed for GWA. Phenotypic findings indicated that anti-SGH IgG levels remained higher in exposed and subclinically infected dogs than in severely diseased dogs even in regression model residuals. Five associated markers were identified on chromosomes 2, 20 and 31. The mapped genes included CD180 (RP105) and MITF related to the rapid activation of B lymphocytes and differentiation into antibody-secreting plasma cells. The findings pointed to chromosomal segments useful for functional confirmation studies and a search for adjuvant molecules of the anti-saliva response.

The waste stream of second generation bioethanol production plant mainly consists of lignin - a polymer of several units of not fermentable phenylpropane - coming from lignocellulosic biomasses. This stream can be used only partly to energetically support the process of bioethanol production (about 40%), whereas the fate of the remaining 60% has to be found. Fluidized bed combustion technology can be considered as a viable option to recover thermal power from this waste stream. However, the understanding of the mechanisms of thermo-conversion and attrition of these lignin-rich particles in fluidized beds is scarce and it deserves further investigations. To this end, the combustion of lignin-rich residues were studied in lab-scale fluidized beds with the aid of different diagnostic and experimental protocols to analyze devolatilization, char burn-out and fragmentation of single fuel particles. Comparing devolatilization times with transversal mixing time of typical industrial-scale fluidized bed combustors, wet fuel particles larger than 10mm can be fed directly to the combustor chamber, whereas pre-dried fuel was extremely reactive with the risk of localized emissions of heat and micro- and macro-pollutants once fed to the fluidized bed reactors. Particles do not undergo primary fragmentation, whereas probably secondary and not percolative fragmentation occurs during the late stage of char burn-out.

We present the most recent solar neutrino results from the Borexino experiment at the Gran Sasso underground laboratory. In particular, refined measurements of all neutrinos produced in the {\it pp} fusion chain have been made. It is the first time that the same detector measures the entire range of solar neutrinos at once. These new data weakly favor a high-metallicity Sun. Prospects for measuring CNO solar neutrinos are also discussed.

Seeds of Mimosa bimucronata are heavily infested (pre-dispersal predation) by the bruchid beetle Acanthoscelides schrankiae in Brazil. In this study, firstly we set up experiments to assess seed germination under seven and six different light and temperature regimes, respectively, and then we evaluated the ability of seeds to germinate after predation. We tested the hypothesis that the non-predated seeds from infested fruits may respond differently when set for germination than those seeds of non-infested fruits. We also hypothesized that predation may increase the production of unviable seeds. Seeds under 18 hours of light presented the highest percentage of germination, and the alternating temperature 20-30 °C was considered as optimum for germination (abnormal seedlings were not considered as a successful germination). Germination of seeds from non-infested fruits was significantly higher than germination of non-predated seeds from infested fruits, and predation also caused a significant increase in the proportion of dead seeds. Our results also show a positive correlation between proportions of unviable seeds and predated seeds. These results demonstrated that seeds of M. bimucronata are strongly affected by predation because predated seeds did not germinate and non-predated seeds had their viability reduced when located in infested fruits, supporting our hypothesis

The chemical composition of monovarietal olive oils from the cultivar Colombaia was studied. Free acidity, peroxide value and UV absorbance attested to the good quality of the analyzed oils. Their fatty acid composition appeared to be quite different from the typical fatty acid profile of olive oils from Liguria but met the limits reported in the EC Regulations for olive oils. On the contrary, the amounts of Δ7-stigmastenol were often higher than the 0.5 % limit set by EC Regulations and total ß-sitosterol was below the minimum 93 % limit. The composition of polar compounds and of the volatile fraction was representative of the peculiar organoleptic character of these oils. Thus, the anomalous sterol composition of the monovarietal oils from the cultivar Colombaia calls for blending with other oils. Moreover, the use of these oils for the production of PDO oils “Riviera Ligure” must also be carefully controlled because it changes their nutritional and sensorial features
