The invasive fish, Amur sleeper, poses a significant and growing threat to Central European freshwater ecosystems. Despite its rapid spread, the ecological implications of its invasion have been poorly explored. Recent findings confirm its presence in various Estonian freshwater systems, raising concerns about its imminent expansion into larger lakes. To better understand its potential ecological impacts, we explored the isotopic niche of the Amur sleeper in comparison with native fish species co-existing in three Estonian freshwater ecosystems. We employed carbon and nitrogen stable isotope analyses alongside gut content analyses. Our findings show that the Amur sleeper’s diet in newly-invaded Estonian water bodies predominantly comprises benthic macroinvertebrates, although it may also include fish, confirming its role as a predator in the local food web. Notably, Amur sleeper populations exhibited clear isotopic niche partitioning in three invaded ecosystems. A logistic regression model, based on stomach content analyses, revealed an ontogenetic diet shift from benthivorous to piscivorous feeding habits from small to large specimens. Amur sleeper exhibits voracious, non-selective feeding habits, which can negatively impact native freshwater communities. The ability to occupy a distinct isotopic niche, with minimal overlap with native fish populations, may reduce interspecific competition, facilitating the spread and establishment of Amur sleeper in newly-invaded habitats. Managing the spread of this invasive species thus becomes even more critical to safeguard the integrity of native aquatic ecosystems.

Lakes play a crucial role in the nutrient cycling of Earth, despite covering only a small fraction of the planet’s surface. Their interactions with their surrounding catchment areas significantly impact ecosystems and regulatory services. The connection between a lake and its catchment, especially the drainage ratio (catchment area to lake surface area), shapes the characteristics of lakes and their response to catchment processes. Within the catchment area, geological, land cover, and land use factors influence the composition of stream water that flows into the lake. These factors play a role in transporting various substances, both organic and inorganic, to the streams. Lakes act as dynamic filters, altering the chemical composition of water that flows through them. This study aims to investigate how a large, shallow lake impacts the quality of the river water as it passes through. It builds on an analysis of nutrient (carbon, nitrogen, phosphorus, silicon) fluxes into Lake Võrtsjärv, using six years of monthly monitoring data from five main inflows and the outflow. The research explores how catchment characteristics and hydrology affect nutrient concentrations and loadings into the lake, as well as the retention or release of substances by the lake. Findings reveal that catchment characteristics, such as land use and forest cover, significantly influence water quality parameters. Different inflows showed variations in water quality, and annual variations were observed, largely correlated with precipitation and discharge. Võrtsjärv plays a critical role in retaining or releasing nutrients, with varying impacts depending on the water budget of the lake. In years with a positive water balance, the lake retains all nutrients, whereas in dry years only inflowing N and P loads exceed their outflow. Overall, this study underscores the importance of lakes as integral components of catchment ecosystems, shedding light on their complex interactions with the environment and the implications for water quality. It emphasizes the need for careful consideration of land use and hydrological factors in managing and preserving these vital aquatic systems.

We analysed long-term (1992–2020) changes in fertiliser use, wastewater treatment, and river water nutrient status in Estonia (N-E Europe) in the context of changing socio-economic situations and legislation. We hypothesised that improved regulation of fertiliser usage and wastewater treatment are reflected as declining riverine nutrient concentrations, with the largest relative improvements occurring in catchments with initially high proportions of point source loading. We analysed nutrient dynamics in 16 rivers differing by catchment land use, population and livestock densities. Data on fertiliser use and wastewater treatment originated from the Statistics Estonia database, and riverine nutrient concentrations from the State Environmental Monitoring Database. We clustered the rivers by their catchment properties and analysed trends in their nutrient status. Point source nutrient loading reductions explained most of the decline in riverine nutrient concentrations, whereas application of mineral fertilisers has increased, hindering efforts to reach water quality and nutrient load targets set by the EU Water Framework Directive and the Baltic Sea Action Plan. Highest nitrogen concentrations and strongest increasing trends were found in rivers within the Nitrate Vulnerable Zone, indicating violation of the EU Nitrates Directive. To comply with these directives, resource managers must address non-point source nutrient loading from river watersheds. Abstract Invasive species constitute a threat not only to native populations but also to the structure and functioning of entire food webs. Despite being considered as a global problem, only a small number of studies have quantitatively predicted the food web‐level consequences of invasions. Here, we use an allometric trophic network model parameterized using empirical data on species body masses and feeding interactions to predict the effects of a possible invasion of Amur sleeper (Perccottus glenii), on a well‐studied lake ecosystem. We show that the modeled establishment of Amur sleeper decreased the biomasses of top predator fishes by about 10%–19%. These reductions were largely explained by increased larval competition for food and Amur sleeper predation on fish larvae. In contrast, biomasses of less valued fish of lower trophic positions increased by about 0.4%–9% owing to reduced predation pressure by top piscivores. The predicted impact of Amur sleeper establishment on the biomasses of native fish species vastly exceeded the impacts of current‐day fishing pressures.

Lakes play a crucial role in the global biogeochemical cycles through the transport, storage, and transformation of different biogeochemical compounds. Their regulatory service appears to be disproportionately important relative to their small areal extent, necessitating continuous monitoring. This study leverages the potential of optical remote sensing sensors, specifically Sentinel-2 Multispectral Imagery (MSI), to monitor and predict water quality parameters in lakes. Optically active parameters, such as chlorophyll a (CHL), total suspended matter (TSM), and colored dissolved matter (CDOM), can be directly detected using optical remote sensing sensors. However, the challenge lies in detecting non-optically active substances, which lack direct spectral characteristics. The capabilities of artificial intelligence applications can be used in the identification of optically non-active compounds from remote sensing data. This study aims to employ a machine learning approach (combining the Genetic Algorithm (GA) and Extreme Gradient Boost (XGBoost)) and in situ and Sentinel-2 Multispectral Imagery data to construct inversion models for 16 physical and biogeochemical water quality parameters including CHL, CDOM, TSM, total nitrogen (TN), total phosphorus (TP), phosphate (PO4), sulphate, ammonium nitrogen, 5-day biochemical oxygen demand (BOD5), chemical oxygen demand (COD), and the biomasses of phytoplankton and cyanobacteria, pH, dissolved oxygen (O2), water temperature (WT) and transparency (SD). GA_XGBoost exhibited strong predictive capabilities and it was able to accurately predict 10 biogeochemical and 2 physical water quality parameters. Additionally, this study provides a practical demonstration of the developed inversion models, illustrating their applicability in estimating various water quality parameters simultaneously across multiple lakes on five different dates. The study highlights the need for ongoing research and refinement of machine learning methodologies in environmental monitoring, particularly in remote sensing applications for water quality assessment. Results emphasize the need for broader temporal scopes, longer-term datasets, and enhanced model selection strategies to improve the robustness and generalizability of these models. In general, the outcomes of this study provide the basis for a better understanding of the role of lakes in the biogeochemical cycle and will allow the formulation of reliable recommendations for various applications used in the studies of ecology, water quality, the climate, and the carbon cycle.

Disentangling causal interactions among biodiversity, ecosystem functioning and environmental factors is key to understanding how ecosystems respond to changing environment. This study presents a global scale analysis quantifying causal interactions and feedbacks among phytoplankton diversity, biomass and nutrients along environmental gradients of aquatic ecosystems.

Photoautotrophic picoplankton (0.2-2 μm) can be a major contributor to primary production and play a significant part in the ecosystem carbon flow. However, the understanding about the dynamics of both eukaryotic and prokaryotic components of picoplankton in shallow eutrophic freshwater environments is still poor. Very few studies in these ecosystems reveal the taxonomic composition of picoeukaryotes. The main objective of this study was to investigate the seasonal dynamics of phototrophic picoplankton with the emphasis on the eukaryote community composition in a large shallow, eutrophic lake of the northern temperate zone (Lake Võrtsjärv). Phytoplankton pigments were employed to determine the taxonomic composition of photoautotrophic picoplankton. We found out that photoautotrophic picoplankton constitutes an important part of the phytoplankton community in Lake Võrtsjärv and its contribution can be highly variable (from ~9.3% to ~39%) in different years. The eukaryotic photoautotrophic picoplankton was dominated by diatoms followed by chrysophytes and other minor groups. Picoeukaryotes were prevailing in low-light conditions and low temperatures as their predominance in the picoplankton community was tightly linked to the presence or absence of ice cover. Ice cover strongly suppressed the growth of picocyanobacteria. Total phosphorus, turbidity and metazooplankton abundance had a clear relationship with photoautotrophic picoplankton chlorophyll a.

The post-soviet period in Eastern Europe brought about fast changes in economy, land use, and environmental protection, whereas legacy effects of the previous era of heavy contamination continued emerging in the status of water bodies. In this paper, we analysed the post-soviet (since 1992) changes in catchment nutrient loadings and stoichiometry of nitrogen (N) and phosphorus (P) in two large non-stratified lakes in Estonia – Võrtsjärv and Peipsi. The drastic reduction in the application of P-fertilisers and P discharges with wastewaters since the early 1990s reduced P loadings and increased N/P loading ratio into both lakes. However, it was hard to find clear evidence of reduced in-lake nutrient concentrations and improved water quality. In both lakes, water transparency constantly decreased and phytoplankton biomass increased. Over the years, the difference in N/P ratio between the two lakes became smaller while the large differences in the cyanobacterial community composition remained. Although common thresholds in nutrient ratios favouring N2-fixing species could be revealed in both lakes, the phytoplankton in Võrtsjärv, strongly dominated by Limnothrix spp., remained mostly light-limited and the relationship with N/P stoichiometry was indirect. Random Forest analysis indicated an important role of light limitation in both lakes. Constantly lower levels of N in the deeper Lake Peipsi favoured N2-fixing species, which, as a paradox, became P-limited. As climate warming reinforces eutrophication phenomena in lakes by increasing internal nutrient loading and favouring bloom-forming cyanobacteria, more stringent measures would be needed to further limit nutrient loads (especially that of P) to lakes through improved wastewater treatment and increased efficiency of fertiliser application.

Cerebral dopamine neurotrophic factor (CDNF) is a neurotrophic factor that has beneficial effects on dopamine neurons in both in vitro and in vivo models of Parkinson’s disease (PD). CDNF was recently tested in phase I-II clinical trials for the treatment of PD, but the mechanisms underlying its neuroprotective properties are still poorly understood, although studies have suggested its role in the regulation of endoplasmic reticulum (ER) homeostasis and the unfolded protein response (UPR). The aim of this study was to investigate the mechanism of action of CDNF through analyzing the involvement of UPR signaling in its anti-apoptotic function. We used tunicamycin to induce ER stress in mice in vivo and used cultured primary neurons and found that CDNF expression is regulated by ER stress in vivo and that the involvement of UPR pathways is important for the neuroprotective function of CDNF. Moreover, we used AP-MS and BiFC to perform the first interactome screening for CDNF and report novel binding partners of CDNF. These findings allowed us to hypothesize that CDNF protects neurons from ER-stress-inducing agents by modulating UPR signaling towards cell survival outcomes.

Arbuscular mycorrhizal fungi (AMF) are obligate symbionts forming mutualistic relationships with most land-plants. In AM symbiosis, the plant supplies the fungal partner with products of photosynthesis and in return recieves various mineral nutrients from the soil. Because of complex interactions with both soil and plants, disturbance can dramatically decrease AMF activity in soils and in these circumstances, it could be useful to restore AMF communities using inoculations. The use of AMF inoculants has so far been minimal due to the high cost and low availability of these products. In this study, the production of simple crude inocula was tested in trap cultures and the most suitable growth substrate nutrient content determined. The effectiveness of the produced inocula was compared with two commercially available inoculants. The results of this study indicate that the best substrate for inocula production in pot-cultures is pure sand with 5% natural soil. When using roots of plants grown in this manner as inoculants, the largest biomass and root-colonisation was achieved. As one of the commercial inoculants did not contain any mycorrhizal propagules at all, the need for quality control and establishment of industry standards is paramount. This study highlights the basics of using AM inoculations in home gardens and small-scale agriculture. The inoculation of large areas is still problematic because of low-yielding inoculant production, but could become highly perspective as methodology improves.

Climate change in recent decades has been identified as a significant threat to natural environments and human wellbeing. This is because some of the contemporary changes to climate are abrupt and result in persistent changes in the state of natural systems; so called regime shifts (RS). This study aimed to detect and analyse the timing and strength of RS in Estonian climate at the half-century scale (1966-2013). We demonstrate that the extensive winter warming of the Northern Hemisphere in the late 1980s was represented in atmospheric, terrestrial, freshwater and marine systems to an extent not observed before or after the event within the studied time series. In 1989, abiotic variables displayed statistically significant regime shifts in atmospheric, river and marine systems, but not in lake and bog systems. This was followed by regime shifts in the biotic time series of bogs and marine ecosystems in 1990. However, many biotic time series lacked regime shifts, or the shifts were uncoupled from large-scale atmospheric circulation. We suggest that the latter is possibly due to complex and temporally variable interactions between abiotic and biotic elements with ecosystem properties buffering biotic responses to climate change signals, as well as being affected by concurrent anthropogenic impacts on natural environments.

Through cascading effects within lake food webs, commercial and recreational fisheries may indirectly affect the abundances of organisms at lower trophic levels, such as phytoplankton, even if they are not directly consumed. So far, interactive effects of fisheries, changing trophic state and climate upon lake ecosystems have been largely overlooked. Here we analyse case studies from five European lake basins of differing trophic states (Lake Võrtsjärv, two basins of Windermere, Lake Geneva and Lake Maggiore) with long-term limnological and fisheries data. Decreasing phosphorus concentrations (re-oligotrophication) and increasing water temperatures have been reported in all five lake basins, while phytoplankton concentration has decreased only slightly or even increased in some cases. To examine possible ecosystem-scale effects of fisheries, we analysed correlations between fish and fisheries data, and other food web components and environmental factors. Re-oligotrophication over different ranges of the trophic scale induced different fish responsesIn the deeper lakes Geneva and Maggiore, we found a stronger link between phytoplankton and planktivorous fish and thus a more important cascading top-down effect than in other lakes. This connection makes careful ecosystem-based fisheries management extremely important for maintaining high water quality in such systems. We also demonstrated that increasing water temperature might favour piscivores at low phosphorus loading, but suppresses them at high phosphorus loading and might thus either enhance or diminish the cascading top-down control over phytoplankton with strong implications for water quality.

Small-bodied cladocerans and cyclopoid copepods are becoming increasingly dominant over large crustacean zooplankton in eutrophic waters where they often coexist with cyanobacterial blooms. However, relatively little is known about their algal diet preferences. We studied grazing selectivity of small crustaceans (the cyclopoid copepods Mesocyclops leuckarti, Thermocyclops oithonoides, Cyclops kolensis, and the cladocerans Daphnia cucullata, Chydorus sphaericus, Bosmina spp.) by liquid chromatographic analyses of phytoplankton marker pigments in the shallow, highly eutrophic Lake Võrtsjärv (Estonia) during a seasonal cycle. Copepods (mainly C. kolensis) preferably consumed cryptophytes (identified by the marker pigment alloxanthin in gut contents) during colder periods, while they preferred small non-filamentous diatoms and green algae (identified mainly by diatoxanthin and lutein, respectively) from May to September. All studied cladoceran species showed highest selectivity towards colonial cyanobacteria (identified by canthaxanthin). For small C. sphaericus, commonly occuring in the pelagic zone of eutrophic lakes, colonial cyanobacteria can be their major food source, supporting their coexistence with cyanobacterial blooms. Pigments characteristic of filamentous cyanobacteria and diatoms (zeaxanthin and fucoxanthin, respectively), algae dominating in Võrtsjärv, were also found in the grazers' diet but were generally avoided by the crustaceans commonly dominating the zooplankton assemblage. Together these results suggest that the co-occurring small-bodied cyclopoid and cladoceran species have markedly different algal diets and that the cladocera represent the main trophic link transferring cyanobacterial carbon to the food web in a highly eutrophic lake.

Pigment-based chemotaxonomy and CHEMTAX software have proven to be a valuable phytoplankton monitoring tool in marine environments, but are yet underdeveloped to determine algal assemblages in freshwater ecosystems. The main objectives of this study were (1) to compare the results of direct microscopy and CHEMTAX in describing phytoplankton community composition dynamics in a large, shallow and eutrophic lake; (2) to analyze the efficiency of the pigment-based method to detect changes in phytoplankton seasonal dynamics and during rapid bloom periods; (3) to assess the suitability of specific marker pigments and available marker pigment:chlorophyll a ratios to follow seasonal changes in eutrophic freshwater environment. A 5-year (2009-2013) parallel phytoplankton assessment by direct microscopy and by CHEMTAX was conducted using published marker pigment:chlorophyll a ratios. Despite displaying some differences from microscopy results, the pigment-based method successfully described the overall pattern of phytoplankton community dynamics during seasonal cycle in a eutrophic lake. Good agreement between the methods was achieved for most phytoplankton groups - cyanobacteria, chlorophytes, diatoms and cryptophytes. The agreement was poor in case of chrysophytes and dinoflagellates. Our study shows clearly that published marker pigment:chlorophyll a ratios can be used to describe algal class abundances, but they need to be calibrated for specific freshwater environment. Broader use of this method would enable to expand monitoring networks and increase measurement frequencies of freshwater ecosystems to meet the goals of the Water Framework Directive.

The influence of functional group specific production and respiration patterns on a lake's metabolic balance remains poorly investigated to date compared to whole-system estimates of metabolism. We employed a summed component ecosystem approach for assessing lake-wide and functional group-specific metabolism (gross primary production (GPP) and respiration (R)) in shallow and eutrophic Lake Võrtsjärv in central Estonia during three years. Eleven functional groups were considered: piscivorous and benthivorous fish; phyto-, bacterio-, proto- and metazooplankton; benthic macroinvertebrates, bacteria and ciliates; macrophytes and their associated epiphytes. Metabolism of these groups was assessed by allometric equations coupled with daily records of temperature and hydrology of the lake and measurements of food web functional groups biomass. Results revealed that heterotrophy dominated most of the year, with a short autotrophic period observed in late spring. Most of the metabolism of the lake could be attributed to planktonic functional groups, with phytoplankton contributing the highest share (90% of GPP and 43% of R). A surge of protozooplankton and bacterioplankton populations forming the microbial loop caused the shift from auto- to heterotrophy in midsummer. Conversely, the benthic functional groups had overall a very small contribution to lake metabolism. We validated our ecosystem approach by comparing the GPP and R with those calculated from O2 measurements in the lake. Our findings are also in line with earlier productivity studies made with 14C or chlorophyll a (chl-a) based equations. Ideally, the ecosystem approach should be combined with diel O2 approach for investigating critical periods of metabolism shifts caused by dynamics in food-web processes.

We examined and parameterized chlorophyll-specific phytoplankton absorption coefficients (a*ph(λ)) for three turbid productive Estonian lakes on the basis of bio-optical measurements in 2005-2013. A new model parameterization was created that enables to reconstruct the spectra of a*ph(λ) for turbid productive waters with the higher reliability than previous parameterizations for ocean and coastal waters. The coefficients A(λ) and B(λ) of our model differ from those found in seas, coastal waters and other types of lakes. For any water type separately the increase of total chlorophyll concentration accompanied with the decrease of a*ph. Our results showed significant seasonal differences between the model parameters due to diversity of the phytoplankton assemblages. This suggests that season-specific models should be developed and validated. Improving the modelling of chlorophyll-specific phytoplankton absorption spectra for hypertrophic lakes is still pending on the availability of a larger dataset, which includes simultaneous measurements of chlorophyll concentrations, phytoplankton absorption coefficients and phytoplankton species composition. Our results implied that total chlorophyll concentration is not a universal predictor of the magnitude of chlorophyll-specific phytoplankton absorption coefficient. The aph(λ) models are also likely site and season dependent. Further research is needed for quantifying the role of accessory pigments and other optical constituents as well as the cell size of dominant algal species for considering their influence on the modelling outputs.

Our aim was to find out to which extent fossil phytoplankton pigments in the large shallow and turbid Lake Võrtsjärv carry information on the history of phytoplankton communities. For this purpose we examined how the changes in the pigment composition of surface sediments follow their changes in the water column. Depth-integrated lake water and surface sediment samples were collected weekly in May–October 2007. Considering cyanobacterial and diatom dominance in phytoplankton, we analysed fucoxanthin, diadinoxanthin and diatoxanthin as marker pigments for diatoms, zeaxanthin as a marker pigment for total cyanobacteria and canthaxanthin as a marker pigment for colonial cyanobacteria. Chlorophyll a and its derivative pheophytin a were applied as indicators for total phytoplankton. The dynamics of phytoplankton pigments in surface sediments generally did not follow their dynamics in the water column, possibly due to intensive resuspension and a high sedimentation rate in a large and shallow lake. It was noticed that the surface sediment carries information on pigment degradation intensity and on weight and size characteristics of phytoplankton cells, which affect their sinking and floating velocities. Higher pigment contents of sediment in spring were presumably caused by lower resuspension due to high water level and slower degradation in cold water. Pheophytin a and the marker pigments of cyanobacteria were found to be persistent against degradation in upper sediment layers, which makes them useful indicators for tracking the historical changes in phytoplankton communities also in a shallow lake. Sharp decrease in chemically unstable pigment contents between the sediment surface and deeper layers indicates that only the uppermost sediment surface is resuspended in Lake Võrtsjärv. The transformation of the diatom marker carotenoid diadinoxanthin to diatoxanthin was found to occur mainly in sediments and not in the water column, and the process is not induced by excess light.
