Objective: To evaluate anti-psoriatic activity of Coleus forskohlii in rats with imiquimod-induced psoriasis. Methods: Imiquimod was used to induce psoriasis in rats. Body weight, skin thickness, erythema, scaling, spleen weight, and histological alternations were measured to assess the effect of Coleus forskohlii. Furthermore, an emulgel formulation containing Coleus forskohlii 10% was prepared and characterized along with its ex vivo permeation studies. Results: The emulgel formulation containing Coleus forskohlii 10% had a pH of 5.40±0.36, with optimum spreadability of (31.67±2.08) g/(cms) and viscosity of (15 966.67±1 274.10) cps, and enhanced both the rate and the extent of drug permeation through psoriatic skin. In an ex vivo study, the quantity of drug permeated (19.18%), deposited (52.38%), and drug remaining in the donor compartments (28.31%) was satisfactory. Coleus forskohlii significantly alleviated imiquimod-induced psoriasis by increasing glutathione and superoxide dismutase activity, decreasing malondialdehyde and nitric oxide levels, and alleviating histological alternations in rat skin. Conclusions: Coleus forskohlii can alleviate imiquimod-induced psoriasis, which may be used as a therapeutic candidate for the treatment of psoriasis.

This study investigated the physicochemical, low glycemic index, and mineral uptake of functional noodles added with varying concentrations (0–10 %) of Lentinus edodes (also known as shiitake) powder. Noodles with 4 % mushroom powder (L3) exhibited comparable sensory attributes in comparison with control noodles. The L3 showed significantly higher protein (1.92 ± 0.035 %), decreased carbohydrate content, improved cooking characteristics. The L3 exhibited darker color with lower hardness, adhesiveness, higher mineral content and bioavailability of iron (59.22 ± 0.49 %). As well, L3 showed a significantly higher mineral transport, retention, and uptake of minerals followed by increased ferritin content (29.17 ± 0.52 ng/mg cell protein). Mushroom powder incorporation in noodles significantly decreased the area under the curve of reducing sugar release correlating with lower glycemic index and thus a potential impact on blood sugar levels. This study illustrates the efficacy of mushroom powder as a functional component in noodles, offering both augmented nutritional advantages and increased glycemic regulation.

Macro-fungi are recognized for food, medicinal, and environmental applications, therefore, the study focuses on multifaceted nutritional, techno-functional, and bioremediation-related uses of Agaricus bisporus and Ganoderma lucidum powders. Herein, A. bisporus exhibited 11.43 % significantly higher protein content, whereas, G. lucidum exhibited 63.55 % higher fiber content. For both mushrooms, spherical porous and fibrous interconnected structural morphology was observed, which led to distinguishing crystallinity and techno-functional properties. G. lucidum demonstrated 75.89 % higher iron, and 19.22 % higher zinc bioavailability while A. bisporus displayed 45.36 % higher calcium bioavailability. G. lucidum absorbed significantly higher zinc and calcium during cellular uptake, while A. bisporus revealed higher iron uptake. Both macro-fungi enhanced iron storage, with G. lucidum achieving 28.68 ± 0.61 ng ferritin/mg cell protein. A. bisporus degraded 88.69 ± 0.12 % Malachite Green while G. lucidum degraded 90.51 ± 0.19 %, maintaining efficiency in both sunlight and UV light. The research indicates that these matrices may serve as valuable nutritional sources and potential substitutes for additives.

This study aimed to evaluate the anticancer and antioxidant potential of Tridax potential and to identify its main phenolic compounds using reverse-phase HPLC. The crude extract of T. procumbens was separated into different fractions using the liquid–liquid partition method. T. procumbens crude extracts and fractions were tested on human cancer cell lines (PC-3, COLO-205, A549, A431, MDA-MB-231, MDA-MB-468, and K562) as well as a control cell line (HEK-293). The antioxidant activities of the extracts and fractions were also assessed. The majority of the extracts appeared to inhibit the progression of human cancer cell lines based on IC50 values that varied from 23.41 to 90.76 µg/mL. However, the chloroform fraction of the stem (7) displays strong activity against A431 and MDA-MB-231, with IC50 values of 23.41 g/mL and 29.45 µg/mL, respectively. The antioxidant result shows that extracts and their fractions scavenge free radicals with an IC50 value between 14.70 and 93.80 µg/mL. However, 7 showed more efficacies with an IC50 of 14.70 µg/mL in the DPPH assay and 29.90 µg/mL in the nitric oxide assay. HPLC analysis revealed that the flower part of T. procumbens contains high levels pf ferulic acid (0.55 to 2.65 mg/g) and kaempferol (1.1 to 4.95 mg/g). A strong antioxidant activity observed in DPPH and NO assay which helps to measure the extract’s effectiveness in scavenging NO free radical, neutralizing other reactive oxygen species (ROS), often correlate with significant antiproliferative effects in various cancer cell lines. This study highlights the potential of plant-based compounds in cancer treatment.

This study explores the adsorption of indigo carmine dye using bio-combusted cerium oxide nanoparticles (CeO2 NPs). CeO2 NPs were synthesized using a bio-combustion method, and then subjected to structural, morphological, and optical characterization for thorough investigation. Structural investigation was carried out using X-ray diffraction (XRD), which revealed a cubic structure with evaluated average crystallite size of 11.55 nm. Later, the same was verified by employing W–H plot (13.57 nm). UV–Vis spectroscopy revealed an effective band gap of 3 eV suited for photocatalytic applications. The metal-oxygen phonon band at 986.32 cm−1 and 871.96 cm−1 is confirmed using Infrared Spectroscopy (FTIR). The morphological analysis was done using Transmission and Scanning Electron Microscopy (TEM and SEM), which revealed well-dispersed, aggregated structure enclosing spherical nanoparticles with an average size of ∼14 nm. The early precursors were validated using EDAX analysis and SEM. Optical characteristics were investigated using photoluminescence (PL), which revealed a large charge transfer band between 360 nm and 435 nm. The dye removal efficiency of CeO2 NPs was evaluated against Indigo Carmine dye using UV light. The results showed that the significantly adsorption, with more than 70 % removed after 150 min. Kinetic experiments revealed that the depreciation occurred via a pseudo-first-order reaction process. Furthermore, the impacts of certain factors such as dye dosage, pH, reusability, and scavenger on adsorption rate were explored and shown to be effective values for the adsorption process. This study emphasizes the potential of CeO2 NPs as excellent photocatalysts for environmental remediation, especially in dye removal applications.

There are very limited analytical procedures to determine lemborexant (LMB) in dosage forms and biological materials. However, the literature does not provide any “high-performance thin-layer chromatographic (HPTLC)” assays to determine LMB. In an effort to analyze LMB in commercial pharmaceutical tablets more precisely, accurately, and sustainably than with the normal-phase-HPTLC (NP-HPTLC) approach, a sensitive and greener reverse-phase-HPTLC (RP-HPTLC) method was developed and validated. For NP-HPTLC, an acetone-petroleum ether (40:60 v/v) developing system was used. However, ethanol–water (85:15 v/v) was the developing system for RP-HPTLC. Four different techniques, including the National Environmental Method Index (NEMI), Analytical Eco-Scale (AES), ChlorTox, and Analytical GREENness (AGREE), were used to evaluate the greenness of both procedures. LMB measurement was linear in the 50–500 and 20–1000 ng/band ranges, respectively for NP and RP procedures. RP procedure was more robust (uncertainties = 0.90–0.95 %), accurate (recoveries = 98.24–101.57 %), precise (uncertainties = 0.87–1.00 %), linear (20–1000 ng/band), sensitive (LOD = 0.92 ng/band and LOQ = 2.76 ng/band), and greener over NP procedure. The results of greenness assessment using NEMI (all four circles green), AES (93), ChlorTox (0.88 g), and AGREE (0.89) demonstrated that the RP strategy was greener than NP strategy and all other reported HPLC methods. The fact that both techniques can assess LMB when its degradation products are present implies that they both have characteristics that point to stability-indicating features. 89.24 % and 98.79 %, respectively, were the assay results for LMB in pharmaceutical tablets when utilizing the NP and RP procedures. Based on all validation and greenness metrics, it was found that RP procedure was better than the NP procedure. As a result, it is possible to assess LMB in pharmaceutical products using RP procedure.

This study focuses on the solution combustion approach to examine the nanostructures of undoped and doped ZnO with different concentrations of Al (0.1 % and 0.2 %). Various physical techniques were utilized to characterize the synthesized nanoparticles. X-ray diffraction (XRD) revealed the crystalline materials, while scanning electron microscopy (SEM) with energy-dispersive X-ray (EDX) findings confirmed the products with particle size and the insertion of Al into the ZnO lattice. Fourier-transform infrared spectra (FTIR) confirmed the presence of different functional groups in the obtained material. The results indicate that Al-doped ZnO (Al–ZnO) nanoparticles show promising properties for optoelectronics and photoluminescence. Photoluminescence analysis indicated that an increase in Al3+ (0.2 %) concentration resulted in a decrease in peak intensity and an increase in the full width at half maximum. The band gap was calculated using the Taucs plot. The study also highlights the effectiveness of Zn1-xAlxO nanostructures in degrading organic pollutants, particularly in adsorbing Malachite Green (MG) dye. Among the samples, the 0.2 % Al-doped ZnO exhibited superior dye degradation efficiency due to its enhanced adsorption capacity and smaller particle size, as evidenced by multilayer adsorption capacity and chemisorption during the degradation process. This study provides valuable insights into the potential applications of Al-doped ZnO nanoparticles in various environmental and technological fields, emphasizing their significance in the degradation of organic pollutants.

The construction industry consumes significant resources, emits considerable pollutants, and generates substantial waste. Green, Lean, Six Sigma (GLS) is an emerging paradigm to control waste, carbon footprint, resource conservation, non-value-added activities, and cost. However, limited focus has been given to the risks involved in GLS construction projects (GLSCPs). This research explored risk factors (RFs) to GLSCPs based on literature review and expert judgments. Brainstorming sessions were conducted to validate the RFs and establish mutual interactions among them through experts' opinions. A 4-level structural model was extracted through Interpretive structural modeling (ISM). The Matriced Impacts Croise's Multiplication Appliqée a UN Classement (MICMAC) was integrated to assess the ‘driving’ and ‘dependence’ power of the RFs. The results show that all RFs are crucial and impact GLSCPs, but the most critical are ‘unstable inflation,’ ‘fluctuations in interest rate,’ and ‘fluctuations in exchange rate.’ This study enhances managers' and policymakers' understanding of RFs associated with GLSCPs and supports effective risk management for successful GLS implementation in construction projects.

Memory loss or dementia is a progressive disorder, and one of its common forms is Alzheimer’s disease (AD), effecting mostly middle aged and older adults. In the present study, we developed Rivastigmine (RIV) nanoparticles using poly(lactic-co-glycolic acid) (RIV-loaded PLGA NPs) and polyvinyl alcohol (PVA). The prepared RIV-PLGA nanoparticles was evaluated for the management of Alzheimer's disease (AD). The nanoparticles were prepared by the slightly modified nano-precipitation technique. The developed formulations were evaluated for particle size, zeta potential (ZP), polydispersibility index (PDI) and surface morphology and drug content. The experimental result revealed that prepared RIV-loaded PLGA NPs (F1) was optimized having particle size (61.2 ± 4.6 nm), PDI (0.292), ZP (−11.2 ± 1.2). SEM study confirms the prepared nanoparticles depicted non-aggregated as well smooth surface particles without any fracture. This formulation (F1) was further assessed for in vivo studies on animal model. Background: As the world settles down from the COVID-19 pandemic, many countries are faced with an unexpected outbreak of monkeypox infection. Monkeypox is a zoonotic disease caused by monkeypox virus (MPXV), which is an enveloped, double stranded DNA virus belonging to the Poxviridae family. Presently, we construct and analyze the phylo-geo-network and the corresponding haplogroups. Presently, we performed the haplogroup analysis with their defining mutations and phylogenetic lineage study along with geographical distributions with the aim to understand the evolutionary path of the MPXV across the world. Results: Information about 719 full length genomes of MPXV were collected from GISAID repository and the sequences extracted from NCBI. The alignment of 719 MPXV genomes and their subsequent analysis revealed a total of 1530 segregating sites of which 330 were parsimony informative (PI) sites. The variations had a positive value of Tajima’s D statistic indicating some mutations being prevalent and hence balancing selection. A total of 39 haplogroups were observed in the phylo-geo-network and their defining mutations along with the evolutionary path has been discussed. The phylo-geo-network revealed the nodal haplogroup is represented by GISAID ID 13889450, haplogroup A1, an isolate from Germany, having a total of 296 identical sequences in the study incident across 22 countries. The localized evolution is highlighted by country specific sequences and haplogroups. USA had a total of 58 genomes and 13 haplogroups as compared to Peru (89 genomes, 7 haplogroups) and Germany (26 genomes, 6 haplogroups). Conclusions: The evolution of MPXV can be happening in a localized manner and hence accumulation of variations in the MPXV genomes needs to be monitored in order to be prepared for any possible threats.

The greener “high-performance thin-layer chromatography (HPTLC)” methods for apremilast (APM) analysis in pharmaceutical products and biological fluids are not currently available in the literature. Accordingly, this study involves the development and validation of a rapid, simple, economical, and greener reversed-phase HPTLC methodology for the estimation of APM in prepared nanoparticles (NPs), nanoemulsion, and marketed tablets. The estimation of APM was conducted using “RP-18 silica gel 60 F254S HPTLC plates” as the stationary phase. The combination of ethanol/water (65:35, v/v) was used as the greener mobile phase for APM analysis. The greenness of the method was predicted using three different approaches, namely Analytical Eco-Score (AES), ChlorTox, and the Analytical GREENness (AGREE) approaches. The λmax = 238 nm was used for the APM detection. By contrasting its single band at Rf = 0.61 ± 0.01 with those of pure APM, the HPTLC peaks for APM in an prepared NP formulation, nanoemulsion, and marketed tablets were identified. In the concentration range of 100–700 ng/band, the proposed analytical methodology was linear. The values of AES, ChlorTox, and AGREE were determined to be 93, 0.66 g, and 0.89, respectively, demonstrated an outstanding greener profile for the existing method. The amount of APM in the tablet, NP formulation, and nanoemulsion was found to be 98.40, 101.60, and 99.37 %, respectively. According to the findings of validation tests and pharmaceutical analysis, the suggested analytical technique could be successfully applied for the routine examination of APM in marketed tablets and laboratory generated nanoformulations.

Herein, we report the synthesis and characterization of a novel Schiff base ligand, (Z)-5-((pyren-1-ylmethylene)amino)-2,4-dihydro-3H-pyrazol-3-one (PMDP). The characterization of ligand PMDP was carried out using ESI-MS, 1H NMR, and UV–Visible spectroscopic techniques. As a probe, PMDP displayed a detectable, colorimetric colour shift in the presence of Cu2+ and Fe2+ ions. The solution was seen to have a light brown colour and to exhibit a fluorometric “turn off” response when Cu2+ and Fe2+ ions were present in a DMSO solution (HEPES 0.01 M, pH = 7.4) at room temperature. Job’s plot revealed that the PMDP binding ratio to Cu2+ and Fe2+ ions was in 1:2 ratio. In contrast to the other metal ions (Cd2+, Mn2+, Co2+, Na+, Ni2+, Cu+, Fe3+, Hg2+, Mg2+, Zn2+, K+, and V5+), the synthesised probe showed exceptional sensitivity and selectivity for detecting Cu2+ and Fe2+ metal ions. The results indicate that the detection limits for Cu2+ and Fe2+ are 0.42 μM and 0.51 μM, respectively. Furthermore, PMDP was efficiently utilised for the quantitative analysis of Cu2+ and Fe2+ in real water samples, RGB colour values in smart phones, logic gate construction, and cell imaging in HeLa cells.

Dodonaea viscosa grows widely in Saudi Arabia, but studies evaluating its neuroprotective activity are lacking. Thus, this study aimed to isolate and identify the secondary metabolites and evaluate the neuroprotective effects of D. viscosa leaves. The isolation and identification of phytochemicals were performed using chromatographic and spectroscopic techniques. The neuroprotective potential of the extract was evaluated against focal cerebral ischaemia–reperfusion injury in rat model. Neurobehavioural deficits in the rats were evaluated, and their brains were harvested to measure infarct volume and oxidative biomarkers. Results revealed the presence of three compounds: a novel isoprenylated phenolic derivative that was elucidated as 4-hydroxy-3-(3ʹ-methyl-2ʹ-butenyl) phenyl 1-O-β-D-apiosyl-(1ʹʹʹ → 6ʹʹ)- β-D-glucopyranoside (named Viscomarfadol) and two known compounds (isorhamnetin-3-O-rutinoside and epicatechin (4–8) catechin). Pre-treatment of the rats with the extract improved neurological outcomes. It significantly reduced neurological deficits and infarct volume; significantly reduced lipid peroxidation, as evidenced by decreased malondialdehyde levels; and significantly elevated antioxidant (superoxide dismutase, catalase, and glutathione) activities. These results indicate that D. viscosa is a promising source of bioactive compounds that can improve neurological status, decrease infarct volume, and enhance antioxidant activities in rats with cerebral ischaemic injury. Thus, D. viscosa could be developed into an adjuvant therapy for ischaemic stroke and other oxidative stress-related neurodegenerative disorders. Further investigations are warranted to explore other bioactive compounds in D. viscosa and evaluate their potential neuroprotective activities.

Graphene-based nanocomposites are widely investigated as cathode materials for energy storage device. Herein, tin oxide-modified graphene (SnO2/G) and bismuth-doped SnO2/G (Bi–SnO2/G) nanocomposites with nanorod-like morphology were synthesized by the combination of electrochemical and wet chemical methods. The as-prepared nanocomposites were characterized using Raman spectroscopy, X-ray diffraction, fourier-Transform spectroscopy, and scanning electronic microscopy. Further, electrochemical impedance spectroscopy (EIS), galvanostatic charge-discharge (GCD) curves, and cyclic voltammetry (CV) were utilized to assess the ultra-capacitor properties of SnO2/G and Bi–SnO2/G nanocomposites.The electrochemical performance of SnO2/G and Bi–SnO2/G nanocomposites demonstrated exceptional specific capacitance of 487 F/g and 174 F/g, respectively, at 5 mV/s compared to as-prepared pristine graphene (108 F/g) and SnO2 nanoparticles (117 F/g). While, the GCD study confirmed the cyclic stability of these electrodes. Overall, the SnO2/G and Bi–SnO2/G nanocomposites have shown good electrochemical properties in all assays.

Background: Ischemic preconditioning (IPC) is the utmost capable design to achieve protection over ischemia-reperfusion injury (I/R), but this phenomenon gets attenuated during various pathological conditions like diabetes. Chrysin exhibits cardioprotection in various experiments however, its therapeutic potential on IPC-mediated cardioprotection via PI3K-Akt-eNOS pathway in streptozotocin (STZ) triggered diabetes-challenged rat heart is yet to be assessed. For that reason, the experiment has been planned to investigate chrysin's effect on the cardioprotective action of IPC involving the PI3K-Akt-eNOS cascade in rat hearts challenged to diabetes. Methods: The project was accomplished through means of absorbance studies for biochemical parameters, infarct size measurement (TTC stain) and coronary flow. Results: The findings of the present study revealed that STZ drastically augmented the serum glucose level and the chrysin significantly reversed the IPC-stimulated increased coronary flow, nitrite release, and reduced LDH (lactate dehydrogenase), CK-MB (creatine kinase) activities as well as infarct size in diabetes-induced rat heart. Furthermore, chrysin also reversed the IPC-induced reduction in oxidative stress in an isolated Langendorff's perfused diabetic rat heart. Moreover, four episodes of preconditioning by either PI3K or eNOS inhibitor in chrysin-pretreated diabetic rat hearts significantly abolished the protective effect of chrysin. Conclusion: Consequently, these observations suggested that chrysin increases the therapeutic efficiency of IPC in mitigating I/R injury via PI3K-Akt-eNOS signalling in diabetes-challenged rat hearts. Hence, chrysin could be a potential alternative option to IPC in diabetic rat hearts.

Objective: Neurological disorders is an increasing problem in the society. Our study aims to explore the secondary metabolites and evaluate the neuroprotective effect of leave extract of Dodonaea viscosa. Methods: The ethyl acetate extract was chromatographed and produced one new entity which was characterized by spectroscopy (IR, UV, NMR, MS) studies. This entity was named as (6S, 9R)-vomifoliol-9-β-D-glucopyranosyle (1 -3)- O-α-L-rhamnopyranoside (DVE-8) while the known entities were identified as viscosine (DVE-12), and catechin (DVE-18). The neuroprotective potential of D. viscosa leaves extract was evaluated using the transient middle cerebral artery occlusion (MCAO) method to induce focal cerebral ischemia–reperfusion injury in rats. Results: The levels of proinflammatory cytokines such as TNF-α, IL-6, and NF-κB gene expression along with the level of anti-apoptotic Bcl–2, BAX, and caspase–3 gene expression was evaluated. Abstract Porous silica particles were prepared by sol–gel method with some modification to get wide-pore particles. These particles were derivatized with N-phenylmaleimide-methylvinylisocyanate (PMI) and styrene by reversible addition fragmentation chain transfer (RAFT) polymerization to prepare N-phenylmaleimide embedded polystyrene (PMP) stationary phases. Narrow bore stainless steel column (100 × 1.8 mm i.d) was packed by slurry packing method. The chromatographic performance of PMP column was evaluated for the separation of synthetic peptides mixture composed of five peptides (Gly-Tyr, Gly-Leu-Tyr, Gly-Gly-Tyr-Arg, Tyr-Ile-Gly-Ser-Arg, Leucine enkephalin) and tryptic digest of human serum albumin (HAS) respectively. Number of theoretical plates as high as 280,000 plates/m were obtained for peptides mixture at optimum elution condition. Separation performance of the developed column was compared with commercial Ascentis Express RP-Amide column and it was observed that separation performance of PMP column was better than commercial column in terms of separation efficiency and resolution.

The present work reports studies on the effective removal of Rhodamine-B (RhB) using Aliquat-336 modified Amberlite XAD-4 resin in the fixed-bed columns in series. The effect of flow rate (Q = 2 to 6 mL·min−1), bed height (h = 3.5 to 7 cm) and initial RhB dye concentration (Cin = 10 to 20 mg·L−1) was studied. When a single column was used, 93% RhB dye was removed in 3 h at Q = 2 mL·min−1, Cin = 10 mg·L−1, and h = 7 cm. When three columns in series were used, almost 100% dye was removed until 80 h. The maximum breakthrough time (142 h) and saturation time (244 h) were found by keeping Q = 2 mL·min−1, h = 7 cm of each column and Cin = 10 mg·L−1. Mathematical modeling of the breakthrough curves was done by using Yoon-Nelson, Clark, Wolborska, and pore diffusion models. The Clark model best fitted the experimental data. The possible interaction mechanism between Aliquat-336 and RhB dye was proposed. The column was regenerated in continuous mode using 1 M HCl solution and maintaining a flow rate of 2 mL·min−1. The purpose of the current study was to prepare and evaluate a citronella oil-loaded microemulsion-based micro-emulgel for the treatment of Candida albicans. The primary objective was to use the skin to transfer hydrophobic medications into the bloodstream. The formulation included cinnamon oil as an antifungal oil and citronella oil as an active pharmaceutical ingredient, respectively. Tween 80 and PEG 200 were used as the surfactant and co-surfactant, respectively, to create phase diagrams. Carbopol 940, one of the frequently used polymers, was investigated for its ability to prepare gel formulations. The optimized (F3) batch contained the highest percentage (87.05 ± 0.03%) of drug content and, according to the statistics provided, had the highest drug release rate of around 87.05% within 4 h. The Korsmeyer–Peppas model with n value of 0.82, which is in the range 0.5–1, had the highest r2 value, indicating that release following non-Fickian/anomalous diffusion provided a better dimension for all of the formulations. The optimized (F3) formulation had stronger antifungal activity in comparison to other formulations. This leads to the conclusion that citronella oil can be made into a micro-emulgel, which may improve its release in aqueous systems while maintaining a high level of drug release at the target site.

The present work elucidates the fabrication of Barium Lanthanum Oxide nanosheets (BaLa2O4 NSs) via a simple one-pot precipitation method. The acquired results show an orthorhombic crystal system with an average crystallite size of 27 nm. The morphological studies revealed irregular-shaped sheets stacked together in a layered structure, with the confirmation of the precursor elements. The diffused reflectance studies revealed a strong absorption between 200 nm and 350 nm, from which the band-gap energy was evaluated to be 4.03 eV. Furthermore, the fluorescence spectrum was recorded for the prepared samples; the excitation spectrum shows a strong peak at 397 nm, attributed to the 4F7/2→4G11/2 transition, while the emission shows two prominent peaks at 420 nm (4G7/2→4F7/2) and 440 nm (4G5/2→4F7/2). The acquired emission results were utilized to confirm the color emission using a chromaticity plot, which found the coordinates to be at (0.1529 0.1040), and the calculated temperature was 3171 K. The as-prepared nanosheets were utilized in detecting latent fingerprints (LFPs) on various non-porous surfaces. The powder-dusting method was used to develop latent fingerprints on various non-porous surfaces, which resulted in detecting all the three ridge patterns. Furthermore, the as-synthesized nanosheets were used to degrade methyl red (MR) dye, the results of which show more than 60% degradation at the 70th minute. It was also found that there was no further degradation after 70 min. All the acquired results suggest the clear potential of the prepared BaLa2O4 NSs for use in advanced forensic and photocatalytic applications.

Entrectinib is an oral selective inhibitor of the neurotrophic T receptor kinase (NTRK). It is used in the treatment of solid tumors in NTRK gene fusion lung cancer. The study aimed to develop and validate an analytical method for quantifying entrectinib plasma by UPLC-MS/MS using quizartinib as an internal standard. The method involves liquid–liquid extraction of entrectinib from plasma using tert butyl methyl ether. The mass-to-charge transitions were 561.23 → 435.1 for entrectinib and 561.19 → 114.1 for quizartinib. The method was successfully validated according to ICH and FDA guidelines. The method has a low quantification limit of 0.5 ng/mL, and the calibration curves constructed over a wide range of 0.5–1000 ng/mL showed good linearity (≥0.997). This method exhibits a tenfold increase in sensitivity compared with the previous method. The method is also accurate, precise, and reproducible, as evidenced by the inter-day and intra-day accuracy and precision values of 82.24–93.33% and 3.64–14.78%, respectively. Principles of green analytical chemistry were considered during all analytical steps to ensure safety. The greenness of the methods was evaluated using two assessment tools. These tools are the Analytical Eco-Scale and the analytical greenness metric approach (AGREE). The results were satisfactory and compatible with the criteria of these tools for green assessment. This method is green, accurate, precise, and reproducible. The method can be used to quantitate entrectinib in plasma and its pharmacokinetics in preclinical, and therapeutic drug monitoring.

Safinamide is an orally active, selective monoamine oxidase-B inhibitor with dopaminergic and non-dopaminergic properties approved by the European Medicine Agency and US Food and Drug Administration for the treatment of mid- to late-stage fluctuating Parkinson’s disease (PD) used in combination with other PD medications such as levodopa. In this study, an analytical quality by design (AQbD) approach was applied to optimize an LC-MS/MS bioanalytical method to determine safinamide in human plasma. A full 33 factorial design was used to optimize safinamide separation conditions, with a method first screened and optimized using chromatographic responses, including peak area and retention time. The results showed that temperature had a significant indirect effect on retention time and peak area (p < 0.05), while ammonium acetate concentration had an insignificant indirect impact on peak area or retention time. However, the temperature was significantly agonistic to the effect of buffer concentration (p < 0.05). The resultant optimized chromatography conditions utilized 9.0 mM ammonium acetate buffer and acetonitrile (22.0:78.0) as mobile phases at a column temperature of 23.2 °C. The assay was linear from 0.1–1000 ng/mL, met acceptance criteria for inter- and intra-assay precision and accuracies across three quality controls, and was successfully applied to in vitro microsomal metabolic stability. The UPLC/MS/MS method was found to be adequately sensitive and suitable for routine safinamide pharmacokinetic studies.

The current study was performed to isolate keratin from chicken feathers with an intention to develop a keratin–genistein wound-healing hydrogel, along with its in vivo analysis. Pre-formulation aspects were analysed by using FTIR; SEM; HPTLC, while gel was characterized for gel strength, viscosity, spreadability, drug content, etc. Additionally, an in vivo study along with biochemical factors against pro-inflammatory factors and histopathological studies were conducted to determine possible wound-healing and anti-inflammatory effects. Pre-formulation studies revealed the presence of amide bonds with region of dense fibrous keratin and an internal porous network in extracted keratin, which corresponds with standard keratin. Evaluation of optimised keratin–genistein hydrogel indicated the development of neutral, non-sticky hydrogel which spread evenly on the skin. In vivo studies in rats indicate higher degrees of wound-healing in combined hydrogel (94.65%) for a duration of 14 days as compared to an individual hydrogel formulation with the development of the epidermis and excessive proliferation of fibrous connective tissue indicating wound repair. Furthermore, the hydrogel inhibited the overexpression of IL-6 gene along with other pro-inflammatory factors, indicating its anti-inflammatory effects. In order to find out the possibility of closure of wounds and anti-inflammatory properties of the novel product, an in vivo investigation into the healing of wounds in laboratory animals was carried out through biochemical (ELISA and qRT-PCR) analyses against inflammatory markers (IL-2, IL-6, IL-1, IL-10, and COX-2) and histopathological (liver, skin, and the kidneys) investigations. Based on the results, we conclude that keratin–genistein hydrogel is a promising therapeutic molecule for the management of wound repair.

5-Fluorouracil (5-FU) is now used in eye drops for the management of conjunctival malignant melanoma, intraepithelial neoplasia, and corneal and conjunctival squamous cell carcinoma. The previously used methods for 5-FU quantification in AqH were time-consuming and less sensitive. Herein, a highly perceptive bioanalytical UPLC–MS/MS method was developed for the quantitative determination of 5-FU in the aqueous humor (AqH) of rabbits using allopurinol as the internal standard (IS). The 5-FU and IS were well separated in an Acquity™ HILIC column. Acetonitrile and 10 mM of ammonium acetate at 95:5 (v/v) were isocratically pumped at a 0.3 mL/min flow rate with a total runtime of 2.5 min. AqH samples were processed with a liquid–liquid extraction method in ethyl acetate. The 5-FU and IS were identified in the negative mode with electrospray ionization. The parent to daughter ion transitions for the 5-FU and IS occurred at m/z 128.92→41.68 and 134.80→64.10, respectively, as quantified using the multiple reaction monitoring mode. The developed method was validated with the ICH-Harmonized Guideline for Bioanalytical Method Validation, and the parameters were within acceptable limits. The calibration curve was linear at the 10.5–2000 ng/mL concentration range, with a correlation coefficient (R2) of 0.9946, and the lower limit of detection was 3.55 ng/mL. The developed and validated method was rapid, sensitive, accurate and robustly able to quantify 5-FU in rabbit AqH. The method was effectively applied to establish the ocular pharmacokinetics of 5-FU following the topical instillation of 5-FU-containing preparations in rabbits.

Eco-friendly liquid chromatographic methods for measuring ergotamine (EGT) are scant in the published database. Accordingly, the goal of the current study was to develop a high-performance thin-layer chromatography (HPTLC) method for fluorescence detection of EGT in commercially available tablets. This approach was based on the application of ethyl alcohol–water (80:20 v/v) as the eco-friendly eluent mixture. The fluorescence detection of EGT was carried out at 322 nm. The greenness score of the present approach was evaluated by “Analytical GREENness (AGREE)” technology. The present approach for measuring EGT in the 25–1000 ng band−1 range was linear. The present assay for fluorescence detection of EGT was validated successfully by ICH guidelines for various parameters. The method was found to be rapid, sensitive, eco-friendly, and stability-indicating. The computed AGREE index for the current strategy was 0.84, displaying outstanding greenness features. The present methodology successfully separated the EGT degradation products under forced-degradation circumstances, exhibiting its stability-indicating qualities and selectivity. An amount of 99.33% of EGT was found in commercial formulations, indicating the validity of the current method for pharmaceutical analysis of EGT in commercial products. The results showed that EGT in commercial products might be regularly measured by the existing method.

Umifenovir is one of the most often prescribed antiviral medications for the prevention and treatment of COVID-19 and other viral infections. Herein, a UPLC-MS/MS method is developed through using ibrutinib as an internal standard (IS) for quantifying umifenovir in plasma samples. Both umifenovir and the IS were analytically separated on an Acquity BEH C18 column with a total run time of only 2.5 min. At a flow rate of 0.3 mLmin−1, acetonitrile:15 mM ammonium acetate (80:20) was employed as the mobile phase composition. Electrospray ionization in positive mode was used for ionization of the samples. Detection and quantification were performed in multiple reaction monitoring mode with parent-to-daughter ionization of 477.05 → 279.02 and 441.16 → 84.4 for umifenovir and the IS, respectively. The method was validated through following international guidelines for bioanalytical method validation, and all parameters were within the acceptable limits. Moreover, the eco-scale method using AGREE software was used for the evaluation of greenness, and results showed that the method is very environmentally friendly. The validated assay was successfully employed in the bioavailability assessment of a newly developed formulation of kneaded ternary umifenovir/β-cyclodextrin with 1% poloxamer 188 (KDB).

Eluxadoline (ELD), a recently approved drug, exhibits potential therapeutic effects in the management and treatment of IBS-D. However, its applications have been limited due to poor aqueous solubility, leading to a low dissolution rate and oral bioavailability. The current study’s goals are to prepare ELD-loaded eudragit (EG) nanoparticles (ENPs) and to investigate the anti-diarrheal activity on rats. The prepared ELD-loaded EG-NPs (ENP1-ENP14) were optimized with the help of Box–Behnken Design Expert software. The developed formulation (ENP2) was optimized based on the particle size (286 ± 3.67 nm), PDI (0.263 ± 0.01), and zeta potential (31.8 ± 3.18 mV). The optimized formulation (ENP2) exhibited a sustained release behavior with maximum drug release and followed the Higuchi model. The chronic restraint stress (CRS) was successfully used to develop the IBS-D rat model, which led to increased defecation frequency. The in vivo studies revealed a significant reduction in defecation frequency and disease activity index by ENP2 compared with pure ELD. Thus, the results demonstrated that the developed eudragit-based polymeric nanoparticles can act as a potential approach for the effective delivery of eluxadoline through oral administration for irritable bowel syndrome diarrhea treatment.

Foretinib, an oral multikinase inhibitor, is known to have anti-tumor effects against cancers. The doses and the levels of foretinib vary based on the type of cancer to be treated. An accurate and precise method is required to determine the level of foretinib and its pharmacokinetics. Here, we developed such a method, which was validated based on the guidelines of the FDA and EMA. Foretinib and ibrutinib (the internal standard (IS)) were extracted using tert-butyl methyl ether. Foretinib and IS were eluted in approximately 1.2 min. Thus, a linear, fast, accurate, and precise method was developed. The calibration curve was linear (r2 ˃ 0.997) in the range of 0.5–400.0 ng/mL and the lowest limit of quantitation was 0.5 ng/mL. The average recovery, accuracy, and precision were 87.9%, 88.7%, and ≤7.8%, respectively. The analyte was deemed stable using various stability tests. The validated assay was then fruitfully applied to a pharmacokinetics study in rats, which revealed that foretinib was absorbed and the maximum concentration achieved at 4.0 h after the administration of a single dose of foretinib. The present study was performed to determine the therapeutic effects of tioconazole (Tz)-loaded novel transferosome carriers (TFs) for the treatment of atopic dermatitis (AD). Method: Tioconazole transferosomes suspension (TTFs) was formulated and optimized using a 32 factorial design. After that, the optimized batch of TTFs loaded into Carbopol 934 and sodium CMC was prepared with hydrogel and noted as TTFsH. Subsequently, it was evaluated for pH, spread ability, drug content, in vitro drug release, viscosity, in vivo scratching and erythema score, skin irritation, and histopathology study. Result: The optimized batch of TTFs (B4) showed the values of vesicle size, flux, and entrapment efficiency to be 171.40 ± 9.03 nm, 48.23 ± 0.42, and 93.89 ± 2.41, respectively. All batches of TTFsH showed sustained drug release for up to 24 h. The F2 optimized batch released Tz in an amount of 94.23 ± 0.98% with a flux of 47.23 ± 0.823 and followed the Higuchi kinetic model. The in vivo studies provided evidence that the F2 batch of TTFsH was able to treat atopic dermatitis (AD) by reducing the erythema and the scratching score compared to that of the marketed formulation (Candiderm cream, Glenmark). The histopathology study supported the result of the erythema and scratching score study with intact skin structure. It showed that a formulated low dose of TTFsH was safe and biocompatible to both the dermis and the epidermis layer of skin. Conclusion: Thus, a low dose of F2-TTFsH is a promising tool that effectively targeted the skin for the topical delivery of Tz to treat atopic dermatitis symptoms.

The objective of this analysis is to establish the potential of biodegradable agro-industrial waste materials as biosorbents in the solid-phase extraction (SPE) technique for sample preparation. In this regard, waste coffee husk (CH) powder was collected, washed, treated chemically, characterized, and applied as an SPE adsorbent to extract pantoprazole from the wastewater samples. Sample detection was accomplished using the UPLC-MS/MS system. The positive mode of electrospray ionization was exploited for the ionization of the sample, and quantification of the target analyte was performed by the multiple reaction monitoring modes. The precursor to product ion transition of 384.02→1380.05 and 384.02→200.05 was used as qualifiers and quantifiers, respectively. Optimization of the particle size, adsorbent dose, and contact time were evaluated to select the best combination of features. The efficiency and regeneration capability of the CH were compared with respect to a commercially available silica-based C18 SPE adsorbent, and it was found that CH possessed comparable (~50%) extraction, as well as regeneration capacity (~95%). The developed biosorbent was applied in a wastewater sample spiked with the target analyte and recovery studies were performed, which found a range of 93.0 to 102.0% with a %RSD of 3.72 to 12.7%. Thus, CH can be exploited as a ‘greener’ replacement for the commercially available adsorbents for the extraction/retention of active pharmaceutical ingredients present in water/wastewater samples.

Recent research has shown that phytocomponents may be useful in the treatment of renal toxicity. This study was conducted to evaluate the renal disease hirsutidin in the paradigm of renal toxicity induced by cisplatin. Male Wistar rats were given cisplatin (3 mg/kg body weight/day, for 25 days, i.p.) to induce renal toxicity. Experimental rats were randomly allocated to four different groups: group I received saline, group II received cisplatin, group III received cisplatin + hirsutidin (10 mg/kg) and group IV (per se) received hirsutidin (10 m/kg) for 25 days. Various biochemical parameters were assessed, oxidative stress (superoxide dismutase (SOD), glutathione transferase (GSH), malonaldehyde (MDA) and catalase (CAT)), blood-chemistry parameters (blood urea nitrogen (BUN) and cholesterol), non-protein-nitrogenous components (uric acid, urea, and creatinine), and anti-inflammatory-tumor necrosis factor-α (TNF-α), interleukin-1β(IL-1β). IL-6 and nuclear factor-kB (NFκB) were evaluated and histopathology was conducted. Hirsutidin alleviated renal injury which was manifested by significantly diminished uric acid, urea, urine volume, creatinine, and BUN, compared to the cisplatin group. Hirsutidin restored the activities of several antioxidant enzyme parameters—MDA, CAT, GSH, and SOD. Additionally, there was a decline in the levels of inflammatory markers—TNF-α, IL-1β, IL-6, and NFκB—compared to the cisplatin group. The current research study shows that hirsutidin may act as a therapeutic agent for the treatment of nephrotoxicity induced by cisplatin.

Dasatinib (DAS), a narrow-therapeutic index drug, Bcr-Abl, and Src family kinases multitarget inhibitor have been approved for chronic myelogenous leukemia (CML) and Ph-positive acute lymphocytic leukemia (Ph+ ALL). Apigenin (APG) has a long history of human usage in food, herbs, health supplements, and traditional medicine, and it poses low risk of damage. The concomitant use of APG containing herbs/foods and traditional medicine may alter the pharmacokinetics of DAS, that probably lead to possible herb–drug interactions. The pharmacokinetic interaction of APG pretreatment with DAS in rat plasma following single and co-oral dosing was successfully deliberated using the UPLC–MS/MS method. The in vivo pharmacokinetics and protein expression of CYP3A2, Pgp-MDR1, and BCPR/ABCG2 demonstrate that APG pretreatment has potential to drastically changed the DAS pharmacokinetics where escalation in the Cmax, AUC(0–t), AUMC(0-inf_obs), T1/2, Tmax, and MRT and reduction in Kel, Vd, and Cl significantly in rats pretreated with APG 40 mg/kg, thus escalating systemic bioavailability and increasing the rate of absorption via modulation of CYP3A2, Pgp-MDR1, and BCPR/ABCG2 protein expression. Therefore, the concomitant consumption of APG containing food or traditional herb with DAS may cause serious life-threatening drug interactions and more systematic clinical study on herb–drug interactions is required, as well as adequate regulation in herbal safety and efficacy.

The fixed dose combination of valsartan (VAL) and hydrochlorothiazide (HCTZ) is the most commonly prescribed medicine for the effective treatment of hypertension. In this study, a simple sensitive and accurate liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for the simultaneous quantitation of VAL and HCTZ in human plasma by using irbesartan (IRB) and hydroflumethiazide (HFMZ) as their specific internal standards (ISs). HLB cartridge-based solid-phase extraction was used for the extraction of analytes and ISs. The chromatographic separation was achieved on Lichrocart RP Select (125 × 4 mm), 5 nm with the mobile phase composition of acetonitrile: 10 mM ammonium acetate buffer: 95:05, v/v, at flow rate of 0.5 mL/min. The turbo ion electrospray ionization in negative mode was used as ion source for the sample ionization. The precursor to product ion transitions were 434.10 > 179.10 (VAL), 295.70 > 204.90 (HCTZ), 427.10 > 192.90 (IRB), and 329.90 > 302.40 (HFMZ) for detection and quantification of analytes and their ISs. The retention times of VAL and HCTZ were 1.90 min and 2.30 min, respectively. The range for the calibration curves of VAL and HCTZ were 50.2–6018.6 ng/mL and 1.25–507.63 ng/mL, respectively, with good linearity having correlation coefficient values of ≥0.995 for both VAL and HCTZ. All validation parameter results (selectivity, precision and accuracy, matrix effects and stabilities) were within the acceptable range as per USFDA guideline for bioanalytical method validation. The intra-day and inter-day accuracy data for VAL were within the range of 105.68–114.22% and 98.41–108.16%, respectively, whereas for HCTZ they were 87.01–101.18% and 95.16–99.37%, respectively. The ion suppression effects produced for VAL and ion enhancement effects produced for HCTZ were insignificant according to the proposed sample cleanup procedure. The developed LC-MS/MS method was successfully applied to bioequivalence study on healthy volunteers.

This study investigates the development of topically applied non-invasive amino-functionalized silica nanoparticles (AMSN) and O-Carboxymethyl chitosan-coated AMSN (AMSN-CMC) for ocular delivery of 5-Fluorouracil (5-FU). Particle characterization was performed by the DLS technique (Zeta-Sizer), and structural morphology was examined by SEM and TEM. The drug encapsulation and loading were determined by the indirect method using HPLC. Physicochemical characterizations were performed by NMR, TGA, FTIR, and PXRD. In vitro release was conducted through a dialysis membrane in PBS (pH 7.4) using modified Vertical Franz diffusion cells. The mucoadhesion ability of the prepared nanoparticles was tested using the particle method by evaluating the change in zeta potential. The transcorneal permeabilities of 5-FU from AMNS-FU and AMSN-CMC-FU gel formulations were estimated through excised goat cornea and compared to that of 5-FU gel formulation. Eye irritation and ocular pharmacokinetic studies from gel formulations were evaluated in rabbit eyes. The optimum formulation of AMSN-CMC-FU was found to be nanoparticles with a particle size of 249.4 nm with a polydispersity of 0.429, encapsulation efficiency of 25.8 ± 5.8%, and drug loading capacity of 5.2 ± 1.2%. NMR spectra confirmed the coating of AMSN with the CMC layer. In addition, TGA, FTIR, and PXRD confirmed the drug loading inside the AMSN-CMC. Release profiles showed 100% of the drug was released from the 5-FU gel within 4 h, while AMSN-FU gel released 20.8% of the drug and AMSN-CMC-FU gel released around 55.6% after 4 h. AMSN-CMC-FU initially exhibited a 2.45-fold increase in transcorneal flux and apparent permeation of 5-FU compared to 5-FU gel, indicating a better corneal permeation. Higher bioavailability of AMSN-FU and AMSN-CMC-FU gel formulations was found compared to 5-FU gel in the ocular pharmacokinetic study with superior pharmacokinetics parameters of AMSN-CMC-FU gel. AMSN-CMC-FU showed 1.52- and 6.14-fold higher AUC0-inf in comparison to AMSN-FU and 5-FU gel, respectively. AMSN-CMC-FU gel and AMSN-FU gel were “minimally irritating” to rabbit eyes but showed minimal eye irritation potency in comparison to the 5 FU gel. Thus, the 5-FU loaded in AMSN-CMC gel could be used as a topical formulation for the treatment of ocular cancer.

The wound-healing process is complex and prone to interruption or failure, which can result in the development of chronic wounds that never heal. This can be overcome by seeking prompt medical attention, which will reduce the likelihood of complications and speed up the healing of the cutaneous wound. It has been established that functionalized engineered biomaterials are a possible strategy for starting skin wound care. The purpose of the current study is to develop a diosmin (DSM)-loaded nanoemulsion (NE)-based gel formulation and to investigate its wound healing and anti-inflammatory activity on rats. The DSM-loaded NEs (F1-F17) were developed and optimized with the help of Box–Behnken Design Expert. The DSM-Nes were developed using lauroglycol 90 (LG90®) as oil, Tween-80 as surfactant and transcutol-HP (THP) as co-surfactant. The optimized Nes showed globule size (41 ± 0.07 nm), polydispersity index (PDI) (0.073 ± 0.008) and percentage of entrapment efficiency (%EE) (87 ± 0.81%). This optimized DSM-loaded NEs (F1) was further evaluated and incorporated into 1% carbopol 940 gel. F1-loaded gel was then characterized for drug content, spreadability, in vitro release, wound healing, and anti-inflammatory studies. The developed gel of DSM was found to show significantly better (p < 0.05) wound-healing and anti-inflammatory activity.

Lemborexant (LEM) is a novel dual orexin receptor antagonist (DORA), recently approved for the treatment of insomnia. As with other DORAs, LEM has potential of abuse and therefore placed in Schedule IV class by the United States Drug Enforcement Administration (USDEA). In this study, a sensitive and accurate UPLC-MS/MS assay was developed for the quantification of LEM in human plasma sample using losartan as an internal standard (IS). The chromatographic separation was performed by using gradient elution of mobile phase, comprising of 10 mM ammonium acetate and acetonitrile with a flow rate of 0.3 mL/min. An Acquity UPLC BEH C18 (1.7 μm, 2.1 × 50 mm) column was used for separation of LEM and IS by maintaining the oven temperature of 40 °C. The electrospray ionization in positive mode was used for sample ionization. The precursor to product ion transition of 411.12 > 175.09 (qualifier) and 411.1 > 287.14 (quantifier) was used for detection and quantification of LEM, respectively, in multiple reaction monitoring mode. Being a drug of abuse, the assay was validated according to “Scientific Working Group for Toxicology” (SWGTOX) guidelines, including limit of detection (LOD), limit of quantification (LOQ), precision and bias, calibration model, interferences, carry-over effects, matrix effects, and stability parameters. The LOD and LOQ of the assay were 0.35 and 1.0 ng/mL, respectively. The linear range was between 1–300 ng/mL with correlation coefficient of ≥0.995. The method was also cross validated in rat plasma samples with acceptable ranges of precision and accuracy before its application for pharmacokinetic study in rats.

Two-dimensional graphene oxide (GO)-based lamellar membranes have been widely developed for desalination, water purification, gas separation, and pervaporation. However, membranes with a well-organized multilayer structure and controlled pore size remain a challenge. Herein, an easy and efficient method is used to fabricate MoO2@GO and WO3@GO nanocomposite membranes with controlled structure and interlayer spacing. Such membranes show good separation for salt and heavy metal ions due to the intensive stacking interaction and electrostatic attraction. The as-prepared composite membranes showed high rejection rates (˃70%) toward small metal ions such as sodium (Na+) and magnesium (Mg2+) ions. In addition, both membranes also showed high rejection rates ˃99% for nickel (Ni2+) and lead (Pb2+) ions with good water permeability of 275 ± 10 L m−2 h−1 bar−1. We believe that our fabricated membranes will have a bright future in next generation desalination and water purification membranes.

A wide range of analytical techniques have been reported to determine cordycepin (CDN) in various sample matrices. Nevertheless, greener analytical approaches for CDN estimation are scarce in the literature. As a result, this study was designed to develop and validate a stability-indicating greener “high-performance thin-layer chromatography (HPTLC)” technique for CDN determination in a laboratory-developed formulation. The greener eluent system for CDN detection was ethanol–water (75:25 v/v). At a wavelength of 262 nm, CDN was measured. The greenness scale of the proposed analytical technology was derived using the “Analytical GREENness (AGREE)” approach. The proposed stability-indicating HPTLC assay was linear for CDN analysis in the 50–1000 ng/band range with a determination coefficient of 0.9978. The proposed analytical technique for CDN analysis was simple, rapid, accurate, precise, robust, selective, stability-indicating, and greener. The AGREE score for the proposed stability-indicating HPTLC technique was calculated to be 0.79 using the AGREE calculator. The current protocol was able to detect CDN degradation products under various stress conditions, indicating its stability-indication characteristics and selectivity. The AGREE quantitative score indicated that the stability-indicating current protocol had outstanding greener characteristics. The amount of CDN in the laboratory-developed formulation was determined to be 98.84%, indicating the suitability of the current protocol in the assay of CDN in the formulations. These results suggested that CDN in a laboratory-developed formulation may be regularly determined using the stability-indicating greener HPTLC strategy.

In the current study, diosmin (DSM)-loaded beta-cyclodextrin (β-CD)-based nanosponges (NSPs) using diphenylcarbonate (DPC) as a cross-linker were prepared. Four different DSM-loaded NSPs (D-NSP1-NSP4) were developed by varying the molar ratio of β-CD: DCP (1:15–1:6). Based on preliminary evaluations, NSPs (D-NSP3) were optimized for size (412 ± 6.1 nm), polydispersity index (PDI) (0.259), zeta potential (ZP) (−10.8 ± 4.3 mV), and drug loading (DL) (88.7 ± 8.5%), and were further evaluated by in vitro release, scanning electron microscopy (SEM), and in vitro antioxidant studies. The NSPs (D-NSP3) exhibited improved free radical scavenging activity (85.58% at 100 g/mL) compared to pure DSM. Dissolution efficiency (%DE) was enhanced to 71.50% (D-NSP3) from plain DSM (58.59%). The D-NSP3 formulation followed the Korsmeyer–Peppas kinetic model and had an n value of 0.529 indicating a non-Fickian and controlled release by diffusion and relaxation. The D-NSP3 showed cytotoxic activity against MCF-7 breast cancer, as evidenced by caspase 3, 9, and p53 activities. According to the findings, DSM-loaded NSPs might be a promising therapy option for breast cancer.

Reduction or elimination of traditional hazardous chemicals in chromatographic separation and sample preparation is comparatively safer and can be considered as a greener approach for the determination of analytes in biological matrices. A rapid and sensitive UHPLC-MS/MS assay for determination of quetiapine in rat plasma was developed and validated using environment friendly and low toxic organic solvents as organic modifier in mobile phase and sample preparation. Quetiapine and risperidone (internal standard) were separated on Acquity BEH™ C18 column (50 × 2.1 mm, i.d. 1.7 μm) using ethanol–water-formic acid (80:20:0.1, v/v/v) as mobile phase at a flow rate of 0.3 mL/min. Detection was performed on tandem mass spectrometer using positive electrospray ionization source by multiple reaction monitoring mode. The precursor to product ion transitions of m/z 384.13 > 253.00 for quetiapine and m/z 411.18 > 191.07 for IS was used to quantify them respectively. The method was found to be linear in the concentration range of 0.45–500 ng/mL. All validation parameters were within acceptable range, as defined by guidelines of bio-analytical method validation. In conclusion, bioanalytical assay using UHPLC-MS/MS together with environment friendly and low toxic organic solvents in mobile phase and sample preparation may be considered as greener approach for quantification of analytes in plasma samples. CdS/ZnO photocatalyst were prepared by two steps via hydrothermal and photochemical method for the photodegradation of Rhodamine B (RhB) dye. The structural and morphological features of both pure ZnO nanorod and CdS/ZnO heterostructures were carefully characterized using various techniques including UV/Vis spectroscopy, XRD, SEM, EDX and EIS. The UV/Vis absorption spectra revealed that absorption performance of heterostructure is extended toward the visible light regions. Photocatalytic activity of both ZnO nanorod and CdS/ZnO heterostructures were investigated for the photodegradation of RhB dye. It was found that CdS/ZnO heterostructure prepared with 30 min light illumination shows the best photocatalytic efficiency than the one at 15 min and pure ZnO nanorod. The better and enhanced photocatalytic efficiency of CdS/ZnO heterostructure was ascribed to the high charge separation efficiency. The maximum photocatalytic efficiency of 85% was achieved within 8 h with the CdS/ZnO-30 min photocatalyst. This work revealed that composite such as CdS/ZnO heterostructure has better photocatalytic efficiency that pure ZnO nanorod. Efficient removal of organic pollutants from wastewater by using photocatalysis has become a hot research in recent days due to its ecological and environmental importance. Herein, cadmium sulfide (CdS) nanostructure with different morphologies i.e. nanoflowers and nanopetals were prepared by a hydrothermal method. The Structural properties of as-synthesized nanostructures material have been studied by X-ray diffraction (XRD), Field emission scanning electron microscopy (FESEM), Energy dispersive X-ray spectroscopy (EDX), while UV–Visible spectroscopy have been used to study their optical properties. The photocatalytic activities of the prepared nanostructures have been evaluated by the photocatalytic degradation of organic dye i.e. rhodamine B (RhB). The highest degradation of 78% in CdS nanoflower and 63% in nanopetal for RhB was observed in 4 h. The EIS measurement showed that charge transfer resistance is lower in CdS nanoflower as compare to the nanopetal. The result showed that the CdS nanoflower was more active than nanopetal for photodegradation of RB dye due to more efficient charge transfer. Due to this excellent performance of CdS nanomaterials make it an ideal candidate for various engineering applications in near future.
